The anamorph life cycle of the black yeastExophiala (Wangiella) dermatitidis is described. The fungus is dimorphic, yeast cells being the prevalent form of propagation. The fungus is strongly hydrophilic, probably completing its anamorph life cycle in submersion. Adaptation to dry conditions is slow. Types of conidiogenesis comprise annellidic, phialidic and sympodial reproduction, in addition to isotropic development. Phialoconidia fail to germinate under the conditions tested, and thus may have a function other than dispersal. Sterile, multicellular bodies resembling aCapronia teleomorph are described.  相似文献

Conidiogenous cells of both the synnematous and mononematous conidiophores in Ceratocystis ulmi and C. piceae develop by sympodial proliferation. Holoblastic conidia are produced on nodules or short denticles from the synnematous conidiogenous cells and on well-defined denticles from the mononematous conidiogenous cells. Graphium penicillioides is characterized by percurrent proliferation of the conidiogenous cells and the conidia are holoblastic and annellidic. A comparison of the type material of G. penicillioides with the lectotype specimen of C. piceae indicates that G. penicillioides is not the conidial state of C. piceae. The method of conidial development in C. ulmi and C. piceae is distinct from that of G. penicillioides, the lectotype species of Graphium; these conidial states are, therefore, placed in a new hyphomycete genus, Pesotum, as the Pesotum state of Ceratocystis ulmi and C. piceae, respectively.  相似文献

A hitherto undescribed black yeast was isolated from an extract of brown coal containing humic and fulvic acids at pH 0.6. The fungus showed morphological similarity to some members of the genus Exophiala (Chaetothyriales) and of Hortaea (Dothideales). Based on SSU rDNA sequence similarity to meristematic members of the Dothideales, the new species was accommodated in Hortaea, which presently contains only a single, halophilic species, H. werneckii.  相似文献

Conidiogenesis of the type species and two additional species of the genus Raffaelea Arx & Hennebert are described. In contrast to previous conclusions based on light microscopy that indicate sympodial production of the conidia, we found that conidium development was by annellidic percurrent proliferation in the type species Raffaelea ambrosiae, as well as in Raffaelea arxii and Raffaelea albimanens. Consequently, this mode of conidium development is similar to the conidiogenesis of anamorphs within the Ophiostomatales, but distinguished by sporodochia formed within the genus Raffaelea. These findings reduce the differences between Raffaelea and other anamorphs with annellidic percurrent proliferation of the conidiogenous cells within the Ophiostomatales to different conidiomata and conidiophores, and they support previous reports of the close phylogenetic relationship between Raffaelea and Ophiostoma.  相似文献

Analysis of ITS rDNA of the black yeast Exophiala dermatitidis revealed a close phylogenetic relationship to the meristematic fungus Sarcinomyces phaeomuriformis. As most strains ofS. phaeomuriformis have a yeast-like phenotype corresponding to the anamorph genus Exophiala, a new combination in Exophiala is proposed. On the basis of ITS sequence, M-13 fingerprint and SSU intron data, two main entities could be distinguished within E. dermatitidis. One of these (B) contained prevalently strains from environmental sources, while the other (A) mainly comprised strains from clinical sources. This may be due to a difference in virulence. All strains from severe brain and disseminated infections in East Asia clustered in group A. However, strains of group A caused a relatively mild fungemia in patients outside East Asia. Tinea nigra is a superficial mycotic infection produced by Hortaea werneckii, formerly known as Phaeoannelomyces werneckii, formerly known as Exophiala werneckii, and Stenella araguata, the latter autochthonous in Venezuela, it was first described and named as Cladosporium castellanii in 1973. The present report describes 12 cases in the period of 1972–2002, diagnosed at the Medical Mycology Section at the Tropical Institute of the Universidad Central de Venezuela, in Caracas. This mycosis is more prevalent among young people, with fair skin, from 3 to 28 years of age, who visited beaches and in whom the lesions are more evident. Out of these 12 patients, 8 (66.66%) had Phaeoannelomyces werneckii as the causal agent and 2 (16.67%) Stenella araguata was isolated. Two patients had more than one macule, and curiously in these cases, each lesion was caused by a different species of the aforementioned fungus.  相似文献

Two new species of the genus Corollospora, namely, C. anglusa sp. nov. with its anamorph Varicosporina anglusa sp. nov. and C. portsaidica sp. nov., which were isolated from the coast of the Mediterranean Sea in Egypt, are described in this article based on morphological and molecular evidence. The two new species have one-septate ascospores. Corollospora anglusa resembles C. gracilis by having narrow one-septate hyaline ascospores; however, they differ in ascomata and ascospore dimensions and in pure culture characteristics. Single-ascospore culture of C. anglusa produces the conidia of its anamorph, whereas an anamorph has not been reported for C. gracilis. Varicosporina anglusa differs from the other two known Varicosporina species by having conidial branches that are filamentous, rectangularly branched, hypha like, and disarticulated into two- or one-celled fragments. Corollospora portsaidica is morphologically similar to C. cinnamomea, but the two species differ in the dimensions, shape, and ornamentation of the ascospores. The new Corollospora species were confirmed to be divergent from other similar Corollospora species based on phylogenetic analyses of partial sequences of the LSU rDNA region.  相似文献

Nutritional physiological and tolerance tests were performed for all type strains of species currently classified in the black yeast generaExophiala andPhaeococcomyces, including some additional type strains of taxa recently reidentified asExophiala species. Most describedExophiala species can be distinguished by physiological characters.Exophiala jeanselmei with its varieties, andE. castellanii should all be retained as separate taxa. The pairs of strainsMycotorula schawii/Exophiala dermatitidis, Hormodendrum negronii/Exophiala jeanselmei var.lecaniicorni andSporotrichum gougerotii/Torula bergeri were found to be conspecific. Phenetic analyses of physiological data support the identity ofPhaeococcomyces exophialae as a yeast-like synanamorph ofExophiala spinifera. 72 Strains belonging to 44 species of ascomycetous black yeasts were analyzed for their coenzyme Q systems. Prevalent were Q-10 and dihydrogenated Q-10 systems. Members of the Dothidealean suborder Dothideineae have Q-10 (H2), while those belonging to the suborder Pseudosphaeriineae mostly have Q-10. The anamorph genus Exophiala Carmichael and the teleomorph genus Capronia Sacc. seem to be heterogenous.  相似文献

Deep infections by melanized fungi deserve special attention because of a potentially fatal, cerebral or disseminated course of disease in otherwise healthy patients. Timely diagnostics are a major problem with these infections. Rolling circle amplification (RCA) is a sensitive, specific and reproducible isothermal DNA amplification technique for rapid molecular identification of microorganisms. RCA-based diagnostics are characterized by good reproducibility, with few amplification errors compared to PCR. The method is applied here to species of Exophiala known to cause systemic infections in humans. The ITS rDNA region of five Exophiala species (E. dermatitidis, E. oligosperma, E. spinifera, E. xenobiotica, and E. jeanselmei) was sequenced and aligned in view of designing specific padlock probes to be used for the detection of single nucleotide polymorphisms (SNPs) of the Exophiala species concerned. The assay proved to successfully amplify DNA of the target fungi at the level of species; while no cross-reactivity was observed. Amplification products were visualized on 1% agarose gels to verify the specificity of probe-template binding. Amounts of reagents were minimized to avoid the generation of false positive results. The sensitivity of RCA may help to improve early diagnostics of these difficult to diagnose infections.  相似文献

This paper deals mainly with the conidium ontogenesis and phylogenesis of black yeasts such as E. jeanselmei, E. gougerotii, E. dermatitidis and E. spinifera. The conidium ontogenesis of E. jeanselmei, E. gougerotii and E. dermatitidis was almost the same. One to five annellated tips were observed through a scanning microscope at the apices of conidiogenous cells, which were bottle- or jar-shaped. Annellations on the tips looked like fringes and the conidiogenous cells of these three species were annellides. Annellated projections occurred on hyphae and annelloconidia were also produced from them. Occasionally, secondary annellides occurred from primary ones. They looked like moniliform hyphae. Daughter conidia sometimes budded directly from mother cells. The shapes and sizes of the conidia of these species were very similar to each other. The conidium ontogenesis of E. spinifera was annellidic as well. However, a single annellated tip usually occurred on an annellide. The annellated tips of the fungus were long and more than 20 annellations were observed on their walls. The conidiogenesis of the four species of Exophiala is only annellidic. There were no differences in the biological examinations except KNO3 assimilation among these four species. The growth of E. jeanselmei and E. gougerotii was poor at 37 degrees C. The GC contents of E. jeanselmei 1171, E. gougerotii B-1800, E. dermatitidis MM-7 and E. spinifera DU-3342 were 54.6, 54.6, 56.6 and 59.7%, respectively.  相似文献

The scolytid ambrosia beetles Xyleborus monographus and X. dryographus were investigated to identify their nutritional ambrosia fungi. The examination of the oral mycetangia of the beetles, the specialized organs for fungal transport, revealed the dominant occurrence of Raffaelea montetyi, a fungus that was also predominant in the beetle tunnels in the immediate vicinity of the feeding larvae. R. montetyi was previously known only as the ambrosia fungus of the platypodid ambrosia beetle, Platypus cylindrus. These beetle species inhabit the same habitat, mainly trunks of oaks in the Western Palaeartic. The possibility of an exchange of the symbiotic fungus between the ambrosia beetles within their common breeding place is discussed. Consequently, the previous hypothesis of a species-specific association of a single ambrosia fungus with a single beetle species is questioned. A phylogenetic analysis based on DNA sequences classified R. montetyi within the Ophiostomatales of the ascomycetes. The investigation of conidiogenesis of R. montetyi by SEM supported this taxonomic placement and showed the development of the conidia by annellidic percurrent proliferation, identical to the conidiogenesis reported for many anamorph states of the Ophiostomatales.  相似文献

Samples of bathwater from 14 homes and 22 public bathhouses and sludge in drainpipes from 19 house-hold bathrooms were plated out onto potato dextrose agar supplemented with chloramphenicol. Several media were used to study colony morphology of the isolates and the thermotolerance and alkaline tolerance of each isolate were examined.Eleven sludge samples produced 12 isolates of Exophiala jeanselmei, 2 of E. dermatitidis and 1 of E. moniliae. Five household bathwater samples produced 2 isolates of E. jeanselmei, 4 of E. dermatitidis and 1 of E. alcalophila. One isolate of E. jeanselmei, 2 of E. dermatitidis, 3 of E. moniliae and 2 of unidentified Exophiala species were recovered from 6 samples of the bathwater dissolving Chinese medicine in the bathtubs of public bathhouses. One isolate of E. jeanselmei was recovered from the 15 samples of bathwater from public bathhouses. Bathwater and sludge in bathroom drainpipes may be an important habitat of Exophiala species.  相似文献

A clade where the most halotolerant fungus in the world – Hortaea werneckii, belongs (hereafter referred to as Hortaea werneckii lineage) includes five species: Hortaea werneckii, H. thailandica, Stenella araguata, Eupenidiella venezuelensis, and Magnuscella marina, of which the first species attracts increasing attention of mycologists. The species diversity and phylogenetic relationships within this lineage are weakly known. In this study two moderately halophilic black yeast strains were isolated from brine of graduation tower in Poland. Molecular phylogenetic analyses based on the rDNA ITS1-5.8S-ITS2 (=ITS), rDNA 28S D1–D2 (=LSU), and RNA polymerase II (rpb2) sequences showed that the two strains belong to Hortaea werneckii lineage but cannot be assigned to any described taxa. Accordingly, a new genus and species, Salinomyces and Salinomyces polonicus, are described for this fungus. Furthermore, molecular phylogenetic analyses have revealed that Hortaea thailandica is more closely related to S. polonicus than to H. werneckii. A new combination Salinomyces thailandicus is proposed for this fungus.  相似文献

Molecular studies on halophilic adaptations have focused on prokaryotic microorganisms due to a lack of known appropriate eukaryotic halophilic microorganisms. However, the black yeast Hortaea werneckii has been identified as the dominant fungal species in hypersaline waters on three continents. It represents a new model organism for studying the mechanisms of salt tolerance in eukaryotes. Ultrastructural studies of the H. werneckii cell wall have shown that it synthesizes dihydroxynaphthalene (DHN) melanin under both saline and non-saline growth conditions. However, melanin granules in the cell walls are organized in a salt-dependent way, implying the potential osmoprotectant role of melanin. At the level of membrane structure, H. werneckii maintains a sterol-to-phospholipid ratio significantly lower than the salt-sensitive Saccharomyces cerevisiae. Accordingly, membranes of H. werneckii are more fluid over a wide range of NaCl concentrations, indicating high intrinsic salt stress tolerance. Even H. werneckii grown in high NaCl concentrations maintains very low intracellular amounts of potassium and sodium, demonstrating the sodium-excluder character of this organism. The salt-dependent expressions of two HwENA genes suggest roles for them in the adaptation to changing salt concentrations. The high similarity of these ENA ATPases to other fungal ENA ATPases involved in Na+/K+ transport indicates their potential importance in H. werneckii ion homeostasis. Glycerol is the main compatible solute which accumulates in the cytoplasm of H. werneckii at high salinity, although it seems that mycosporines may also act as supplementary compatible solutes. Salt dependent increase in glycerol synthesis is supported by the identification of two copies of a gene putatively coding for glycerol-3-phosphate-dehydrogenase. Expression of only one of these genes is salt dependent.  相似文献
