Epidemic development of Septoria nodorum was examined in pure stands and mixtures of two spring wheat cultivars, Kolibri and Maris Butler. In mixtures, disease development was reduced almost to that of the more resistant pure stand (Maris Butler). Disease levels, however, were low and no significant differences in grain yield per head were demonstrated. Disease development was further investigated for the host-pathogen combinations winter wheat/S. nodorum and winter barley/Rhynchosporium secalis. With both combinations the mixed stands again reduced disease levels to almost that of the more resistant pure stands. In general, mixed stands were more effective against R. secalis, although, with S. nodorum, disease levels were low. The yield response in mixed stands differed for the host cultivars and was not significantly changed by the presence of disease. The complexity of analysing such situations and the implications of these findings for mixtures of cultivars differing in resistance to non-specialised pathogens are discussed.  相似文献

Background

Powdery mildew of barley is a wind-borne and obligate biotrophic pathogen, which ranks among the most widespread barley pathogens worldwide. However, purposeful research towards studying the structure of the barley powdery mildew populations, of their virulence and of effectiveness of certain resistance genes against the infection was not conducted in Kazakhstan till present time. This paper is the first to describe characteristics of the pathotype structure of Blumeria graminis f.sp. hordei (Bgh) population and effectiveness of resistance genes in two regions of barley cultivation in the republic.

Results

One hundred and seven isolates of Bgh were obtained from seven populations occurring on cultivated barley at two geographically locations in Kazakhstan during 2015 and 2016. Their virulence frequency was determined on 17 differential lines Pallas. All isolates were virulent on the resistance gene Mla8 and avirulent for the resistance genes Mla9, Mla1 + MlaAl2, Mla6 + Mla14, Mla13 + MlRu3, Mla7 + MlNo3, Mla10 + MlDu2, Mla13 + MlRu3 and Mlo-5. The frequencies of isolates overcoming the genes Mla3, Mla22, Mlat Mlg + MlCP and Mla12 + MlEm2 were 0.0–33.33%, and frequencies of isolates overcoming the genes Mlra, Mlk, MlLa and Mlh ranged from 10.0 to 78.6%. Based on reactions of differential lines possessing the genes Mla22, Mlra, Mlk, Mlat, MlLa and Mlh, pathotypes were identified. In total, 23 pathotypes with virulence complexity ranging from 1 to 6 were identified. During both years in all populations of South Kazakhstan and Zhambyl regions pathotypes 24 and 64 mainly prevailed.

Conclusions

Obtained data suggest that low similarity of populations Bgh in Kazakhstan to European, African, Australian and South-East Asian populations. The present study provides a foundation for future studies on the pathogenic variability within of Bgh populations in Kazakhstan and addresses the knowledge gap on the virulence structure of Bgh in Central Asia. Complete effectiveness of the resistance genes, for which no corresponding virulence was found, will allow Kazakhstanean breeders to access many modern barley cultivars that those possessing the resistance effectiveness genes.

Individual isolates (in total 4040) of Erysiphe graminis f. sp. hordei were collected between 1989 and 1991 in fields of spring barley varieties with resistance genes Mla9, Mla12 and Ml(La) in the local air spora, i.e. remote from barley fields, in Hesse, Germany. Their virulence pattern was determined on a 13-partite differential set. In the air spora. virulence complexity (the number of virulence genes per isolate) increased by 30% (4.2 vs. 5.5) between 1989 and 1991. This was mainly due to an increase of pathotypes with virulence genes Va7, Val3, Vk and V(La). In each mildew field population, frequencies of several non-corresponding virulence genes, e.g. V(La) on Mla9 variety, greatly increased during the period. Such change can be caused by hitchhiking selection, i.e. indirect selection resulting from asexual reproduction, and direct selection by non-corresponding resistance genes. To separate the two effects, the aerial barley mildew population of 1991 was sampled on cv. Pallas and near-isogenic lines of Pallas. Frequencies of non-corresponding virulence genes as observed in samples from the near-isogenic lines were compared to expected non-corresponding virulence frequencies which were obtained by subdividing the sample on Pallas into 12 subsamples, comprising all isolates virulent to line 1 through 12, respectively. Among 90 resistance/virulence gene combinations selection for non-cor-responding virulence was significant in five cases (e.g. Va9 on Mla7 host) and against non-corresponding virulence in nine cases (e.g. Vk on Ml(La) host). Hitchhiking selection was significant in eight cases. In 11 cases the two types of selection were significant in combination but not individually. It is suggested to monitor the stability of selection against non-corresponding virulence genes in race surveys and utilize it in diversified barley crops such as variety mixtures in order to retard the evolution of complex races.  相似文献

Spread of Septoria nodorum from inoculated point sources was examined in pure stands and mixtures of two spring wheat cultivars Kolibri and Maris Butler. Gradients in disease were observed soon after inoculation; the presence of the more resistant cultivar (Maris Butler) in the mixtures retarded the outward spread of disease compared with the susceptible pure stand (Kolibri). Regression analysis suggested that gradients in incidence were influenced by nearness to the source whereas gradients in severity were not. Spread of disease was also examined in pure stands and mixtures for the host/pathogen combinations, winter wheat (cvs Maris Huntsman and Maris Ranger)/S. nodorum and winter barley (cvs Maris Otter and Hoppel)/Rhynchosporium secalis. In contrast to the spring wheat experiment, no gradients were observed; explanations for the uniform distribution of disease were (a) extensive spread prior to the period of assessment, (b) no physical barrier to dispersal due to the juvenile growth stage of the crop and (c) exhaustion of the point-source.  相似文献

The obligate biotrophic, soil-borne fungus Synchytrium endobioticum causes wart disease of potato (Solanum tuberosum), which is a serious problem for crop production in countries with moderate climates. S. endobioticum induces hypertrophic cell divisions in plant host tissues leading to the formation of tumor-like structures. Potato wart is a quarantine disease and chemical control is not possible. From 38 S. endobioticum pathotypes occurring in Europe, pathotypes 1, 2, 6 and 18 are the most relevant. Genetic resistance to wart is available but only few current potato varieties are resistant to all four pathotypes. The phenotypic evaluation of wart resistance is laborious, time-consuming and sometimes ambiguous, which makes breeding for resistance difficult. Molecular markers diagnostic for genes for resistance to S. endobioticum pathotypes 1, 2, 6 and 18 would greatly facilitate the selection of new, resistant cultivars. Two tetraploid half-sib families (266 individuals) segregating for resistance to S. endobioticum pathotypes 1, 2, 6 and 18 were produced by crossing a resistant genotype with two different susceptible ones. The families were scored for five different wart resistance phenotypes. The distribution of mean resistance scores was quantitative in both families. Resistance to pathotypes 2, 6 and 18 was correlated and independent from resistance to pathotype 1. DNA pools were constructed from the most resistant and most susceptible individuals and screened with genome wide simple sequence repeat (SSR), inverted simple sequence region (ISSR) and randomly amplified polymorphic DNA (RAPD) markers. Bulked segregant analysis identified three SSR markers that were linked to wart resistance loci (Sen). Sen1-XI on chromosome XI conferred partial resistance to pathotype 1, Sen18-IX on chromosome IX to pathotype 18 and Sen2/6/18-I on chromosome I to pathotypes 2,6 and 18. Additional genotyping with 191 single nucleotide polymorphism (SNP) markers confirmed the localization of the Sen loci. Thirty-three SNP markers linked to the Sen loci permitted the dissection of Sen alleles that increased or decreased resistance to wart. The alleles were inherited from both the resistant and susceptible parents.  相似文献

Summary Pairs of susceptible and resistant, near-isogenic cultivars ofHordeum vulgare which differ for the Mla, Mlk and Mlp genes for resistance toErysiphe graminis f. sp.hordei were inoculated with race 3 of this pathogen and patterns of protein synthesis associated with primary infection mapped using pulse-labelling with L-[35S]methionine and 2-dimensional electrophoresis. Extraction of proteins with buffer containing detergent revealed the enhanced synthesis of 5 and 8 polypeptides at 25 and 30 h respectively after inoculation of barley carrying the Mla gene (cvMla). The enhanced synthesis of these same polypeptides together with 11 additional polypeptides was observed at 48 h and 72 h after inoculation of barley carrying either the Mlp (cvMlp) or Mlk (cvMlk) genes. The labelling of several major constitutive polypeptides was suppressed in cvMla at 24 h after inoculation; the labelling of six of these polypeptides was also suppressed in both cvMlp and cvMlk but not until 48 and 72 h after inoculation. These results indicate that changes occur in the synthesis of some common polypeptides following infection of cultivars carrying different resistance genes but the timing and extent of these changes varies with the resistance gene in the host.  相似文献

Colonies of Diplocarpon rosae derived from single conidia were isolated on malt extract agar, multiplied (at 23°C) and stored (at ?20°C) on surface‐sterilised leaf discs of a universally susceptible rose, ‘Frensham’. The resistance of 16 species and cultivars of Rosa to different isolates of D. rosae was assessed using surface‐sterilised leaf discs. Four pathotypes of D. rosae were distinguished on the basis of host range. One species and one hybrid were resistant to all pathotypes. Two species and two cultivars were susceptible to all pathotypes. Four species and six cultivars were interpreted as having vertical resistance because they were strongly resistant to some but not all pathotypes. Only species and hybrids of the section Cinnamomeae were resistant to the pathotype identified as CW1 whereas only roses of other origins were resistant to the pathotype DA2.  相似文献

Globodera pallida is a parasitic root cyst nematode of potato, which causes reduction of crop yield and quality in infested fields. Field populations of G. pallida containing mixtures of pathotypes Pa2 and Pa3 (Pa2/3) are currently most relevant for potato cultivation in middle Europe. Genes for resistance to G. pallida have been introgressed into the cultivated potato gene pool from the wild, tuber bearing Solanum species S. spegazzinii and S. vernei. Selection of resistant genotypes in breeding programs is hampered by the fact that the phenotypic evaluation of resistance to G. pallida is time consuming, costly and often ambiguous. DNA-based markers diagnostic for resistance to G. pallida would facilitate the development of resistant varieties. A tetraploid F1 hybrid family SR-Gpa segregating for quantitative resistance to G.␣pallida was developed and evaluated for resistance to G. pallida population ‘Chavornay’. Two subpopulations of 30 highly resistant and 30 susceptible individuals were selected and genotyped for 96 single nucleotide polymorphism (SNP) markers tagging 12 genomic regions on 10 potato chromosomes. Seven SNPs were found significantly linked to the nematode resistance, which were all located within a resistance ‘hotspot’ on potato chromosome V. A haplotype model for these seven SNPs was deduced from the SNP patterns observed in the SR-Gpa family. A PCR assay ‘HC’ was developed, which specifically detected the SNP haplotype c that was linked with high levels of nematode resistance. The HC marker was only found in accessions of S.␣vernei. Screening with the HC marker 34 potato varieties resistant to G. pallida pathotypes Pa2 and/or Pa3 and 22 susceptible varieties demonstrated that the HC marker was highly diagnostic for presence of high levels of resistance to G. pallida pathotype Pa2/Pa3.Amirali Sattarzadeh and Ute Achenbach contributed equally to the work  相似文献

The purpose of this study was to determine the allelic state of the resistance gene H1 against the Ro1 and Ro4 pathotypes of the golden potato cyst nematode (Globodera rostochiensis) among Ukrainian and world potato (Solanum tuberosum ssp. tuberosum) cultivars. The allelic state of the TG689 marker was determined by PCR with DNA samples isolated from potato tubers and primers, one pair of which flanked the allele-specific region and the other one was used to control the DNA quality. Among 77 potato cultivars analyzed, the allele of the marker associated with the H1-type resistance was found in 74% of Ukrainian and 90% of foreign cultivars, although some of them proved to be susceptible to the potato cyst nematode in the field. The obtained data confirm the presence of H1 resistance against golden nematode pathotypes Ro1 and Ro4 among Ukrainian potato cultivars and the efficiency of the used marker within the accuracy that has been declared by its authors.  相似文献

Phytophthora root rot is one of the most serious diseases of soybeans in Argentina. Surveys of commercial fields and trial plots of soybean were conducted throughout the northern Pampeana subregion (Argentina) between 1998 and 2004. A total of 193 isolates of Phytophthora sojae were collected and classified into races or virulence formulae. Among the 173 isolates tested on 8 differentials, 42 different pathotypes were detected, including 18 described races. Races 1, 4, 5, 7, 9, 13, 23 and 24 were found in both plants and soils, whereas races 2, 3, 6, 8, 11, 14, 15, 17, 43 and 44 were only isolated from plants. An additional 19 pathotypes were described from 20 isolates tested in Canada on the expanded set of 14 differential cultivars. Currently, all Rps genes/alleles associated with resistance have been defeated, indicating an increased complexity of virulence within the P. sojae populations in the region. The great increase in virulence complexity found in this study is most likely a result of a long period of continuous production of soybean cultivars with Rps genes and the extensive adoption of the no‐tillage system.  相似文献

A model describing disease progress of non-specialised fungal pathogens in intraspecific mixed stands of cereal cultivars (Jeger, Griffiths & Jones, 1981b) has been extended beyond the logarithmic phase of disease development. The extended. logistic model indicates that whatever the outcome in the early stages (disease less than or more than the mean of disease in the pure stands), disease will eventually level off at a value equal to the mean of disease asymptotes in the pure stands. The time scale involved in such levelling-off, however, means that substantial benefit may still be expected from deployment of such mixtures. Analytical solutions for the model are obtained for the special case where the ratio of disease asymptote to initial disease is the same for each pure stand.  相似文献

Two pathotypes of the cereal cyst-eelworm (Heterodera avenae), occurred commonly in England and Wales and a third pathotype was apparently present in low numbers in a few areas. Pathotype 1, which can breed on most cultivars of wheat, barley and oats, but not on Drost barley nor barley cultivars containing the gene for resistance from barley No. 191, was most common in the south west of England. Pathotype 2, which can breed on most cereals, including Drost barley but not No. 191, was common throughout England and Wales and mixtures of these pathotypes occurred in many places. Pathotype 3, which can breed on barley No. 191 and probably on Drost and most cultivars of cereals, was rare and was found only in low numbers together with either or both Pathotypes 1 and 2. In the initial tests it was present or suspected in only nine fields (6%), but when these soils were retested in more detail the following year, Pathotype 3 was confirmed at only one site.  相似文献

Introgressive hybridization of Senecio hercynicus and S. ovatus (Compositae, Senecioneae) was studied in a hybrid zone on the southern slopes of Mt Brocken (Harz Mountains, Germany). A total of 415 plants representing 10 stands along an altitudinal gradient were investigated using multivariate statistical analyses of morphological characters and molecular markers (random amplified polymorphic DNA[RAPD]). Both types of traits detected pure S. hercynicus stands on the summit plateau, pure S. ovatus stands at the lowest elevations, and hybrid swarms at intermediate elevations. While morphological and molecular patterns coincided, some individuals in hybrid stands combined morphological patterns typical of S. ovatus with RAPD patterns typical of S. hercynicus, and vice versa. In general, introgression was symmetrical within stands, though one stand combined S. ovatus characters with the glandular hair typical for S. hercynicus, and two stands combined a S. hercynicus typical RAPD genotype with morphological characters shifted towards S. ovatus. Because pure stands of S. hercynicus occurred only on the summit plateau of Mt Brocken, and markers typical for S. ovatus were detectable in stands up to 1040 m a.s.l., future fusion or assimilation of the rare form, S. hercynicus, by the more widespread S. ovatus appears possible at Mt Brocken.  相似文献

Lithium was used as a non-radioactive tracer to investigate the root activity of two cereals (wheat and barley), and of two contrasting cultivars of pea (leafy and semi-leafless), both in pure stands and in mixtures. The mixtures included combinations of each cereal with each pea cultivar in single rows, alternative rows and cross-drilled. Total lithium uptake (mg m-2) was higher for wheat than for barley, and higher for semi-leafless pea than for leafy peas. Growing cereals with peas reduced the total lithium uptake by peas, compared with pure stands, especially in alternate-row mixtures. Growing peas with cereals only reduced the total Li uptake by cereals when they were cross-drilled. The Li uptake by wheat, barley and peas generally decreased with soil depth in a similar manner; however, semi-leafless peas absorbed proportionately more Li from close to the soil surface than did leafy peas. Both pea cultivars absorbed more Li at 10–20 cm depth when grown in intimate mixtures with cereals, compared with less intimate mixtures or pure stands. The potential of lithium as a non-radioactive tracer in mixed-cropping studies is briefly discussed.  相似文献

The hatching activity of partially resistant Solanum tuberosum×S. vernei clones and susceptible cultivars was investigated in a series of tests with G. pallida conducted in pots in the glasshouse. Differences in hatching were apparent, with the partially resistant clones generally stimulating a lower total hatch than the S. tuberosum cultivars. It is concluded that reduced hatching activity can make a small contribution to overall resistance and perhaps also to the tolerance of a clone to G. pallida.  相似文献

Mildew from susceptible genotypes (SI and S2) of Hordeum bulbosum was found to be avirulent on all H. vulgare genotypes tested, including such cultivars as Proctor with no known genes for resistance to mildew. The H. bulbosum genotype SI (2n =14) proved resistant to all isolates of mildew from H. vulgare. The mildew isolates used for these tests possessed most of the common virulence factors which attack the current ‘vulgare’ cultivars in Western Europe. Some H. bulbosum genotypes were resistant to the ‘bulbosum’ mildew. Attempts at combining resistance from both species are presented and discussed.  相似文献

Daylily rust, caused by Puccinia hemerocallidis, has been present in the United States since 2000. In 2003, inoculations with a single isolate of P. hemerocallidis identified daylily cultivars with high levels of resistance to the fungus. The present study was carried out to determine if pathotypes of P. hemerocallidis are present in the south eastern United States. Sixteen isolates of P. hemerocallidis were each inoculated onto leaf segments from 19 daylily cultivars and the resulting disease phenotype assessed. A significant effect of rust isolate on host reaction phenotype was observed for nine of the 19 daylily cultivars. Five of the nine cultivars displayed reaction phenotypes with different isolates of P. hemerocallidis that included at least one susceptible or moderately susceptible and also resistant phenotypes. These results indicate that different pathotypes of the fungus are present in the south east United States. Daylily hybridizers interested in screening for host resistance to P. hemerocallidis will need to include multiple isolates of the fungus to allow for this host specialization.  相似文献

Sorghum anthracnose is one of the most important and destructive diseases of sorghum. Genetic resistance has been the most efficient strategy to control the disease, but the high variability of the pathogen population in Brazil has resulted in only modest efficacy. Accordingly, we investigated the variability of Colletotrichum sublineolum in response to sorghum populations with three levels of genetic diversity: pure stand, three‐way hybrids and physical mixtures of three‐way hybrids. Six plots of each treatment were planted in different areas and at different dates. A total of 480 isolates, that is 40 single‐conidium isolates per plot, were collected from the field experiment to characterize the variability of the pathogen in each host population. Isolates were inoculated in a greenhouse on a differential line set composed of eight sorghum inbred lines. Our results reveal that the pathogen populations derived from three‐way combinations had higher pathotype diversity than did those derived from pure stand host populations. More complexly, virulent phenotypes were also developed in genetically diverse stands compared to pure stand host populations. The diversification of the host population limits pathogen adaptation, thus resulting in a significantly higher number of pathotypes. The results of this study will improve the management of sorghum anthracnose in the field by helping sorghum breeders maintain disease resistance.  相似文献

Nitrogen (N) fertiliser and clover cultivar choice affect competition and productivity in grass-clover mixtures. Pure stands and mixtures of perennial ryegrass and white clover cultivars with contrasting growth habits were examined. The aim of this work was to study the effect of repetitive nitrogen (N) application and cultivar combination on competition and productivity, N yield in the harvested herbage, N2 fixation in mixtures and pure stands, and transfer of N from clover to the companion grass. Large-leaved white clover cultivar Alice and small-leaved cv. Gwenda and perennial ryegrass cvs. Barlet (erect) and Heraut (prostrate) were sown in pure stands and as four binary grass-clover mixtures on a sandy soil in 1995. In the mixtures, two levels of N fertiliser were applied: 0 (-N) and 150 and 180 kg ha-1 y-1 N (+N) in 1996 and 1997, respectively, while the grass monocultures received three N levels (0, 140/180 and 280/360 kg ha-1) in 1996 and 1997, respectively. No N was applied to pure clover. The plots were cut five times during 1996 and six times during 1997. Fertiliser N was applied in early spring and after every harvest. The treatments were continued until the summer of 1999. In pure grass, the applied N was effectively recovered. In mixtures, N application affected competition by enhancing grass growth and the overall effect of N application was 17 kg DM per kg N applied in 1996. However, there was no yield response to N fertilizer in 1997, because this was compensated for by a higher clover production in unfertilised mixtures. In 1997, -N mixtures yielded more N than +N mixtures, owing to the higher clover content and N2 fixation. Large-leaved clover cv. Alice was better able to withstand the negative effect of repetitive N application on clover production in mixtures and increased its proportion during the growing season of the second harvest year. In 1997, mixtures with Alice yielded more N than mixtures with Gwenda, but in pure clover swards, there was no cultivar effect on N yield. Also, during the autumn of 1998 and the spring of 1999, the clover content was highest in mixtures with Alice. Harvested N and apparent N2 fixation were almost twice as high in 1997 as in 1996. N yield and apparent N2 fixation were higher in pure clover than in mixtures. In mixtures, the apparent N2 fixation in 1996 was 142 kg N ha-1, irrespective of cultivar or N treatment. In 1997, it was on average 337 kg N ha-1, and higher in -N mixtures and in mixtures with Alice. For each tonne of clover DM in the harvested herbage, 65 and 57 kg N was harvested in 1996 and 1997 in -N mixtures, respectively. The apparent transfer of clover-derived N to grass was on average 29 and 70 kg N ha-1 yr-1 in 1996 and 1997, respectively. It was highest in +N mixtures and highest in mixtures with Gwenda in 1997. In contrast to clover, the grass cultivars were very similar in their productivity and seasonal patterns, despite their contrasting growth habits. Seasonal trends in N yield, N transfer and N recovery are discussed in relation to fertilizer application regimes and variation in production patterns in mixtures and pure stands. Summary The complex structure of the multigene family at the Mla locus conferring powdery mildew resistance in barley was studied by making diallel crosses between several near-isogenic lines carrying different Mla alleles. The mode of inheritance of the Mla alleles investigated was determined to be dominant for Mla1, Mla6, Mla7 and Mla13 and semidominant for Mla3, Mla12 and Mla20. F1 plants were backcrossed to the susceptible recurrent parent in order to identify susceptible and double-resistant recombinants in the BC1F1 generation. Out of 17605 progenies tested in the BC1F1 generation, two susceptible recombinants, one between Mla1 and Mla12 and one between Mla13 and Mla20 were confirmed. The former was also verified by RFLP analysis.  相似文献
