Summary Water relations of the root hemiparasite Olax phyllanthi were compared with those of its major species of hosts in natural habitat in coastal heath near Denmark, SW Australia. Leaf water potentials of Olax during winter were 0.4 to 1.4 MPa lower (more negative) than those of all (29) non parasitic host species examined. During the dry summer months (January to March), shallow-rooted hosts developed water potentials up to 3 MPa lower than those of Olax, and were accordingly rated as no longer accessible as a source of water to the hemiparasite. By contrast, deep-rooted hosts, with access to the water table, showed water potentials less negative than Olax, and haustorial contacts retained with these apparently enabled continued extraction of water and nutrients throughout the summer. Three other species of root hemiparasites parasitized by Olax, but not themselves parasitizing Olax, showed leaf water potentials throughout the year very close to, and mostly slightly more negative than those of Olax. Nocturnal measurements of leaf water potential in winter (July and August) in soil at field capacity (water potential –0.006 MPa) showed maintenance of a 0.5–0.8 MPa potential difference between Olax and a range of common host species. By dawn most hosts had equilibrated with the water potential of the soil, whereas both exposed and bagged Olax plants recorded potentials of –0.8 MPa. Daytime rates of transpiration and photosynthesis of Olax were less than those of a range of common hosts, but water use efficiencies were not consistently different between hemiparasite and hosts. This was reflected in almost identical mean values for carbon isotope ratio (13C/12C) between Olax (mean value –27.0) and thirteen frequently exploited hosts ( value –27.1). The results are discussed in relation to published information on other angiosperm hemiparasites.  相似文献

Summary The transfer of nutrients between host and parasite in mistletoes has generally been considered to occur via the xylem to xylem contacts at the host-parasite interface in the haustorial organ of attachment. A few workers, however, have recently begun to question this assumption and have suggested an alternative pathway of transport involving the intervening parenchyma cells which are often abundant in the parasite at the interface. But no morphological experimental evidence has yet been forthcoming in support of an apoplastic continuum across this interface between parasite and host.Our observations on the dwarf mistletoeKorthalsella lindsayi first indicate an absence of plasmodesmata at the interface, with the conclusion that symplastic transport between the two plants is not involved. However, application of apoplastic markers, such as Calcofluor white and lanthanum and uranyl ions, to the stem of the host results in the transfer of these tracers across the interface and into the tissues of the parasite. This demonstrates the existence of an apoplastic continuum between the two plants, and a pathway that is probably used in the normal transfer of water and other nutrients from host to parasite.From the apoplastic continuum provided by the walls of the haustorial parenchyma tissue, nutrients are transferred to the symplast for eventual distribution to other parts of the plant. Evidence for the active uptake of substances from the apoplast by the protoplasts of the parenchyma cells is shown by the convoluted appearance of the plasmalemma and its differentiation often into plasmatubules.  相似文献

Heterotrophic gains of carbon from various host species by the root hemiparasitic shrub Olax phyllanthi (Labill) R.Br. were assessed using techniques based on carbon isotope discrimination (13C) on C3 and C4 hosts and C:N ratios of xylem sap and dry matter of host and parasite. Heterotrophic benefits (H) to Olax based on 13C values were 30% and 19% from two nonnative C4 hosts (Portulaca oleracea and Amaranthus caudatus respectively) compared with 13% and 15% from these hosts when computed on the basis of C:N ratios of host xylem sap and C and N increments of Olax dry matter. Nitrate was the source of N available to pot cultures of the above species and estimates based on C:N ratios assumed that all N accumulated by Olax had come from nitrate absorbed by the host. Equivalent estimates of H for Olax, grown in nitrogen-free pot culture with the native N2-fixing host Acacia littorea as its sole source of N, indicated 63% and 51% dependence on host carbon when assessed in terms of xylem sap composition of host parasite respectively. Comparisons of xylem sap solutes of Olax and a range of partner hosts indicated marked selectivity in haustorial uptake and transfer of nitrate, amino compounds, organic acids and sugars. Possible implications of variations between hosts in absolute levels of C and inorganic and organic forms of N in xylem are discussed in relation to evidence of much better growth performance of Olax on Acacia littorea and other N2-fixing legumes than on non-fixers.  相似文献

The amino acid compositions of the root xylem saps of Olax phyllanthi and a range of its common hosts were examined in native coastal heath in Western Australia and in pot cultures of Olax reliant on single hosts. When hosts specializing in the xylem transport of one major solute (asparagine, glutamine, histidine, arginine or proline) were exploited, the endophytic tissue of haustoria and the xylem sap of Olax showed much lower proportions of this than of other solutes, suggesting pronounced metabolic transformation prior to xylem loading by the parasite. However, the xylem sap of Olax did partly reflect the compositions of its hosts; for example, djenkolic acid and pipecolic acid were present when Olax was parasitic on species of Acacia, and levels of citrulline and aspartic acid were higher than normal when it exploited hosts transporting large amounts of these compounds. Back-flow of S-ethenyl cysteine, a novel amino acid specific to Olax, was observed to another root hemiparasite (Exocarpos sparteus) in native habitat and to certain non-parasitic hosts in water-stressed pot cultures. Haustoria exhibited high levels of glutamine synthetase but showed appreciable in vivo nitrate reductase activity only when on hosts with high xylem levels of nitrate.  相似文献

Nutlets of Hemigenia R.Br. and Microcorys R.Br. were examined using SEM. Significant variation, mainly useful at the infrageneric level, was found in nutlet shape, nature of the attachment scar, nature of surface sculpturing, exocarp cell shape and sculpturing, and nature of the indumentum. Typical nutlets are ovoidal, strongly reticulate or rugose. The exocarp cells are isodiametric and convex to papillate. Also common are cylindrical nutlets, often with longitudinal ridging and papillate exocarp cells. Surface pitting and concave exocarp cells are rare. A cladistic analysis of nutlet characters suggests both Hemigenia and Microcorys are polyphyletic, and Microcorys paraphyletic with respect to Westringia Sm. Notwithstanding that, the infrageneric classification of Hemigenia was largely supported, while in Microcorys, there was support for sect. Hemigenioides, but sects Anisandra and Microcorys were not resolved as distinct.  相似文献

TEM observations were carried out on 40 taxa of the familyVerbenaceae and 35 taxa of the familyOleaceae, in order to ascertain distribution, ultrastructure and morphology of the intranuclear proteinic inclusions in the mesophyll parenchymatic cells. The investigated genera amount to some 25% and 60% respectively of the genera of the two families. Inside theVerbenaceae, lamellar inclusions (L-type) occur in 6 out of 23 investigated genera: they are mostly present inside the tribesCitharexyleae andVerbeneae (both belonging toVerbenoideae), while they are absent in other subfamilies. All of the investigatedOleaceae genera show intranuclear crystalline inclusions (C 1-type) of three different shapes. Among theAsteridae this is a character peculiar toOleaceae. They appear to be a well defined natural group, including the controversial genusNyctanthes.  相似文献

The ultrastructure of dikaryotic haustoria of sevenPucciniastrum species,Thekopsora galii, Naohidemyces vaccinii, andCalyptospora goeppertiana was investigated.Pucciniastrum actinidiae, P. agrimoniae, P. pyrolae, andCalyptospora goeppertiana revealed haustoria whose necks were wrapped by a fold of the extrahaustorial matrix. The matrix-fold ofCalyptospora goeppertiana was characteristically shaped.Pucciniastrum circaeae, P. epilobii, P. hikosanense, P. styracinum, Thekopsora galii, andNaohidemyces vaccinii showed typical haustorial necks which were not sheathed by a matrix-fold. Haustorial necks which were wrapped by a fold of the extrahaustorial matrix were designated velopedunculate, and those which were naked gymnopedunculate. The leaf oils of both subspecies of Atherosperma moschatum have been investigated to determine if there were any chemical differences between the two subspecies. While both subspecies of A. moschatum contained methyl eugenol (55–87%), safrole (5–20%) and linalool (1–6%), camphor (5–10%) was found to be only in the oils of A. moschatum subsp. moschatum and to be either absent or, at most, less than 0.5% in the oils of A. moschatum subsp. integrifolium. There was also a range of monoterpene hydrocarbons and 1,8-cineole, all in the range of 0.1–5%, that were present in subspecies moschatum but virtually absent in subspecies integrifolium.  相似文献

Summary Urastoma cyprinae (Graff) is a microturbellarian which has been recorded both as a free-living organism by Westblad (1955) and Marcus (1951) and as a commensal in various lamellibranch molluscs (see Burt & Drinnan 1968). The material used in this study came from oysters, Crassostroea virginica, collected off the coast of Prince Edward Island, in which hosts it occurs in large numbers especially during the summer months when the oysters are spawning (Fleming et al. 1981). When U. cyprinae is exposed to light as happens, for example, when an oyster is opened, it shows a marked negative phototactic response.Preliminary work on the fine structure of the photoreceptors in U. cyprinae shows that the two eyes each consists of: (1) a single cup cell full of relatively large, electron-dense pigment granules; (2) a tripartite conical lens system; and (3) what appear to be two photosensitive rhabdomes. The pigment cup cell has a single, well defined nucleus situated basally and close to the membrane of the pigment cell furthest away from the rhabdomeres. The lens system consists of a cone made up of three, separate but equal, parts. Each part has two, flat inner surfaces which join at an angle of 120°, an outer rounded surface, and a rounded upper surface. When these three parts fit together, the cone-shaped lens is formed with the apex of the lens within the cup of the pigment cell and the rounded, convex, broad end of the cone lying more or less at the same level as the top of the pigment cup and below the epidermis layer. The rhabdomeres lie between the electron dense lenses and the inside of the pigment cup. They show connections to the visual cells which are bipolar: one extension joining the rhabdomeres; the other constituting the axon which extends into the centrally situated brain or into the longitudinal, lateral nerves. The axons that enter the brain, form connections with other axons from the other eye. The axons that extend posteriorly in a lateral position, presumably play a role in facilitating the avoidance reaction.The chemical nature of the unusual lens has not yet been determined. This is presently under investigation and will be reported later at which time our work will be discussed in relation to other types of rhabdomeric eyes in the Turbellaria.  相似文献

Four different pearl millet breeding lines were transformed and led to the regeneration of fertile transgenic plants. Scutellar tissue was bombarded with two plasmids containing the bar selectable marker and the -glucuronidase reporter gene (gus or uidA) under control of the constitutive CaMV 35S promoter or the maize Ubiquitin1 promoter (the CaMV 35S is not a maize promoter). For the delivery of the DNA-coated microprojectiles, either the particle gun PDS 1000/He or the particle inflow gun was used. The calli and regenerants were selected for their resistance to the herbicide Basta (glufosinate ammonium) mediated by the bar gene. Putative transformants were screened for enzyme activity by painting selected leaves or spraying whole plants with an aqueous solution of the herbicide Basta and by the histochemical GUS assay using cut leaf segments. PCR and Southern blot analysis of genomic DNA indicated the presence of introduced foreign genes in the genomic DNA of the transformants. Five regenerated plants represent independent transformation events and have been grown to maturity and set seed. The integration of the bar selectable and the gus reporter gene was confirmed by genomic Southern blot analysis in all five plants. All five plants had multiple integrations of both marker genes. To date, the T1 progeny of three out of four lines generated by the PDS particle gun shows co-segregating marker genes, indicating an integration of the bar and the gus gene at the same locus in the genome.  相似文献

Citrus limon has a wet stigma which can be divided in two zones: a glandular superficial one formed by papillae, and a non-glandular one formed by parenchymatic cells. The stigmatic exudate is produced by the papillae after the latter have reached their ultimate size. The papillae of the mature pistil are of varying size and composition. Both the unicellular and multicellular ones are present. The cells at the base of the papillae are rich in cytoplasm, whereas the tip cells are vacuolated. Histochemical analysis has shown that the exudate of Citrus is composed of lipids, polysaccharides, and proteins. Our results indicate that the lipidic component is produced and secreted first, followed by production and secretion of the polysaccharidic component. The lipidic component of the exudate is produced in the basal papillae cells and accumulates as droplets in dilated parts of the smooth endoplasmic reticulum (SER). Subsequently the lipid droplets are transported to the plasma membrane, and transferred by the latter into the cell walls. Then the exudate component is accumulated in the intercellular spaces and in the middle lamellar regions of the walls. At maturity, spermatozoids of the green algaChara vulgaris are biflagellated, contain little cytoplasm, and coil for approximately 2 1/2 gyres within the mother cell wall. The anterior of the cell contains an ovoid headpiece anchoring two slightly staggered basal bodies that are positioned above and directly in front of approximately 30 linearly arranged mitochondria. An elongated stellate pattern occupies the transition zone between the BBs and axonemes. Flagella emerge from the cell just in front of the nucleus and encircle the full length of the spermatozoid. The spline comprises a maximum of 38 microtubules surrounding the anterior mitochondria and gradually decreases posteriorly to a minimum of 11. The dense nucleus is narrow, cylindrical, and occupies the central revolution of the cell. Six starch-laden plastids and associated mitochondria are linearly arranged at the cell posterior. Phylogenetic analyses of charalean taxa and archegoniates based on spermatogenesis strongly support the orderCharales, withNitella as the sister group toChara. Diagnostic features ofChara spermatozoids include absence of a lamellar strip and axonemes embedded in the cell for almost the entire length of the anterior mitochondria. Potential relationships amongCharales, Coleochaetales and archegoniates are evaluated in regards to the probable course of evolution of streamlined biflagellated gametes.  相似文献

A procedure is outlined for the establishment of a proliferating cell suspension culture and subsequent plant regeneration of the latex-producing plant,Calotropis gigantea (Linn.) R. Br. Friable calluses were obtained by culturing hypocotyl explants on modified Murashige and Skoog medium with 2.69 μM α-naphthaleneacetic acid and 4.44 μM 6-benzyl-aminopurine. Friable calluses were transferred to modified Murashige and Skoog liquid medium containing 500 mg l−1 casein hydrolysate, 5% (v/v) coconut water and 5% (w/v) sucrose to initiate suspension cultures. Suspensions were subcultured every 10–12 days and supplemented with 13.56 μM 2,4-dichlorophenoxyacetic acid (2,4-D). After 3–4 subcultures, suspensions consisted of small, highly cytoplasmic cell clumps and large vacuolate cells. Plating the suspension clumps on medium containing 4.52 μM 2,4-dichlorophenoxyacetic acid and culturing in darkness produced macroscopic calluses, which subsequently produced a high number of shoots when placed in light and supplemented with 2.22 μM 6-benzyl-aminopurine and 0.45 μM 2,4-dichlorophenoxyacetic acid. Shoots were rooted using Bonner's solution containing 0.49 μM indole-3-butyric acid, and the plants successfully transferred to soil. Interaction between arbuscular mycorrhizal fungus Glomus mosseae and plant growth promoting fungus Phoma sp. was studied for its effect on their root colonization and plant growth of cucumber. Two isolates of Phoma sp. (GS8-2 and GS8-3) were tested with G. mosseae. The percent root length colonized by G. mosseae was not adversely affected by the presence of Phoma isolates. In contrast, the root colonization of both isolates GS8-2 and GS8-3 in 4-week-old plants was significantly reduced (80.7% and 84.3%, respectively) by added G. mosseae. Inoculating plants with each Phoma isolate significantly increased the shoot dry weight. However, dual inoculation of each Phoma isolate with G. mosseae had no significant effect on growth enhancement.  相似文献

The diet of brown trout (Salmo trutta L.) and rainbow trout (S. gairdneri Richardson) in Rutland Water were compared during the first two fishing seasons (April–October 1977 and 1978).Fortnightly samples of approximately forty stomachs were obtained from boat and bank, rod-and-line caught trout giving a total of 1046 stomachs over the two seasons.During 1977 seasonal changes in the diet were divided into two phases; the first being a period of abundant drowned terrestrial food until June. This was followed by a period of more stable water level from July onwards when chironomid larvae and pupae were consistently the most important food items and the diversity of food also increased.In 1978 the proportion of chironomid pupae and larvae declined and they were replaced in the diet by Gammarus and Asellus.  相似文献

Plants of Hordeum vulgare (barley) were grown initially in a solution containing 150 M phosphate and then transferred on day 6 to solutions with (+P) and without (-P) phosphate supplied. After various times plants from these treatments were supplied with labelled phosphate. Analysis of plant growth and rates of labelled phosphate uptake showed that a general enhancement of uptake and translocation was found, in plants which had been in the-P solution, several days before the rate of dry matter accumulation was affected. Subsequently a detailed analysis of phosphate uptake by segments of intact root axes showed that the enhancement of phosphate uptake by P-stress occurred first in the old and mature parts of the seminal root axis and last in the young zones 1 cm from the root apex. During this transition period there were profound changes in the pattern of P absorption along the length of the root. Most of the additional P absorbed in response to P-stress was translocated to the shoot, particularly in older zones of the axis. Enhancement of phosphate uptake in young zones of nodal axes occurred at an earlier stage than in seminal axes. The results are related to the P-status of shoots and root zones and discussed in relation to the general control by the shoot of phosphate transport in the root.  相似文献

The epidermis and associated structures of adult and embryonic Paravortex cardii and Paravortex karlingi, internal parasites of Cerastoderma edule, have been examined using scanning and transmission electron microscopy. The cellular epidermis of adult Paravortex bears cilia and microvilli which differ in number and distribution between P. karlingi and P. cardii. Cellular organelles include mitochondria, lipid bodies, Golgi bodies, and ultrarhabdites. Epidermal nuclei are located in the proximal portion of the cells. The development of the tegument of embryo Paravortex has been described and a possible origin for the embryo capsule is suggested. Summary Seasonal and diurnal gas exchange and water relations of Amyema linophyllum and its host Casuarina obesa were studied at Gingin Western Australia. As recorded elsewhere for other species of mistletoe, stomatal conductances and transpiration rates were consistently higher in parasite than host, but assimilation rates did not differ significantly between partners, and water use efficiency was accordingly substantially lower in the parasite. Parallel responses of the species to environmental conditions suggested closely coordinated stomatal behaviour. However, sunlit and artifically shaded clumps of Amyema maintained high leaf conductances even when foliage fell below turgor loss point, yet their tissue capacitance values indicated maintenance of greater tissue water reserves during stress than in the host. Pressure-volume relationships indicated that differences in tissue water relations were unlikely to contribute significantly to the observed gradient in leaf water potential between partners. An experiment measuring changes in water potential of freshly detached host: parasite systems cut with the host shoot end immersed in water indicated that the haustorial junction was the principal site of resistance to transpiration-driven water flow into the parasite. A parallel experiment on intact attached shoots with mistletoe clumps enclosed and darkened just before dawn, demonstrated that, once the host commenced rapid transpiration, the water potential gradient between partners became reversed.  相似文献

Summary Genetic analysis was conducted on the qualitative and quantitative traits of sexual progeny derived from embryogenic cultures of two inbred lines of Pennisetum glaucum (L.) R. Br. (pearl millet). These lines included a genetically stable inbred of Tift 23 BE and a genetic marker line, derived from Tift 23BE, which bore qualitative genetic markers for a dominant purple plant trait (P) and two recessive traits, early flowering (e1) and yellow stripe (ys). Tissue culture regenerant populations (R0) and progeny populations (R1) produced from these plants by selfing showed no qualitative genetic variation when derived from the genetically stable inbred Tift 23BE. In contrast, stably inherited qualitative variation for a number of genetic markers was observed in R0, R1, and R2 progeny of the genetic marker line. In a population of 1,911 plants regenerated over a 12-month period, 0.02% of the population lost or showed reduced expression of the purple plant trait and 92% of plants were chlorophyll deficient. Plants showing reduction or loss of anthocyanin synthesis also flowered later. None of the purple plants showed any significant variation in flowering time. The incidence of chlorophyll deficiency increased with time in culture, 51 % of the progeny regenerated after 1 month were chlorophyll deficient, while 100% of the plants regnerated after 12 months were chlorophyll deficient. Qualitative variation was also observed in control populations of the genetic marker line where 1 plant in a total of 1,010 lacked purple pigmentation and a total of 6% showed chlorophyll variation in the first generation (S0). The presence of qualitative variation in controls suggests that the inherent variation present in the original explant was expressed and perpetuated in vitro. Quantitative variation was observed for a number of traits in the first sexual cycle (R1) of the marker line but did not occur in a subsequent generation, suggesting that this variation was epigenetic.  相似文献
