Capsicum annuum (pepper) plants were inoculated with the arbuscular mycorrhizal (AM) fungi Glomus intraradices Smith and Schenck, an undescribed Glomus sp. (AZ 112) or a mixture of these isolates. Control plants were non-mycorrhizal. Plants were grown for 8 weeks at moderate (20.7–25.4°C) or high (32.1–38°C) temperatures. Colonization of pepper roots by G. intraradices or the Glomus isolate mixture was lower at high than at moderate temperatures, but colonization by Glomus AZ112 was somewhat increased at high temperatures. Pepper shoot and root dry weights and leaf P levels were affected by an interaction between temperature and AM fungal treatments. At moderate temperatures, shoot dry weights of plants colonized by the Glomus isolate mixture or non-AM plants were highest, while root dry weights were highest for non-AM plants. At high temperatures, plants colonized by Glomus AZ112 or the non-AM plants had the lowest shoot and root dry weights. AM plants had generally higher leaf P levels at moderate temperatures and lower P levels at high temperatures than non-AM plants. AM plants also had generally higher specific soil respiration than non-AM plants regardless of temperature treatment. At moderate temperatures, P uptake by all AM plants was enhanced relative to non-AM plants but there was no corresponding enhancement of growth, possibly because less carbon was invested in root growth or root respiratory costs increased. At high temperatures, pepper growth with the G. intraradices isolate and the Glomus isolate mixture was enhanced relative to non-AM controls, despite reduced levels of AM colonization and, therefore, apparently less fungal P transfer to the plant.  相似文献

In a greenhouse experiment, the effect of arbuscular mycorrhizal (AM) fungus (Glomus intraradices Schenck & Smith) colonization on N assimilation in maize (Zea mays L.) was examined after well-watered, drought and recovery periods. Seeds of selection cycles C0 (drought-sensitive) and C8 (drought-resistant) of the tropical maize cultivar Tuxpeño sequía were used for this study. Maize plants were exposed or not to drought stress for 3 weeks (45-65 days after sowing, DAS) followed by 3 weeks of recovery (66-86 DAS) at the preflowering stage. Root and shoot samples harvested at the end of the drought or well-watered and recovery periods were determined for key enzymes involved in N assimilation (NR, nitrate reductase; NiR, nitrite reductase; GS, glutamine synthetase; GOGAT, glutamate synthase), protein and amino acid concentrations, and total N contents. Drought stress significantly (P ≤ 0.01 or P ≤ 0.001) decreased all the enzyme activities except NiR in the roots and shoots of both cultivars. After 3 weeks of drought, the AM roots of both cultivars had higher activities of NR (C0, 45%; C8, 26%), GS (C0, 76%; C8, 33%) and GOGAT (C0, 41%; C8, 53%) than non-AM roots and were comparable to well-watered plants. These enzyme activities were also enhanced in drought-stressed AM shoots of C0 and C8. Total amino acid concentrations in AM plants of C0 were 4.6 and 1.6 times higher in roots and shoots, respectively, compared to non-AM plants. The predominant amino acids detected were Ala, Arg, Asn, Asp, Gln and Glu which constituted approximately 56 and 75% of the total pool in roots and shoots, respectively. Soluble proteins and total N contents were also higher in AM plants than non-AM plants under drought conditions. The enhancement of N-assimilating enzymes and nitrogenous compounds in maize may indicate a transfer of NO3? through the extraradical mycelium or increased N assimilation due to the AM symbiosis. Our overall results suggest that AM association plays an important role in enhancing N assimilation or N nutritional status which enables the host plant to withstand drought conditions and recover after stress is relieved.  相似文献

Improved salt tolerance of mycorrhizal plants is commonly attributed to their better mineral nutrition, particularly phosphorus. However, the effect of arbuscular-mycorrhizal (AM) fungi on salt tolerance may not be limited to this mechanism. We investigated the possibility that non-nutritional effects of AM fungi, based on proline accumulation or increased photosynthesis and related parameters, can influence the tolerance of lettuce (Lactuca sativa L.) to salinity. Three levels of salt (3, 4 and 5 g NaCl kg-1 dry soil) were applied and plants were maintained under these conditions for 7 weeks. The salt-treated AM plants produced greater root and shoot dry weights than unfertilized or P-fertilized non-AM controls. With increasing salinity, both shoot and root dry weights were reduced, but this decrease was greater in uninoculated plants. In particular, shoot dry weight was not reduced in G. fasciculatum-colonized plants as a consequence of salt, whereas in uninoculated plants it was reduced by about 35% at the highest salt level. Proline accumulation was considerably lower for P-amended non-AM and for AM plants except for G. mosseae-colonized plants than was the case for unamended plants. Transpiration, carbon dioxide exchange rate (CER), stomatal conductance and water use efficiency (WUE) were higher in mycorrhizal plants. At 5 g NaCl kg-1, both photosynthesis and WUE increased by more than 100% in mycorrhizal treatment relative to uninoculated plants. The contents of phosphorus of P-fertilized non-AM plants was similar to or higher than those of G. mosseae- and G. fasciculatum-colonized plants. Plants colonized by G. deserticola had the highest P-content regardless of salt level. Hence, the effect of G. mosseae and G. fasciculatum on salt tolerance in this experiment could not be attributed to a difference in the P content. The mechanisms by which these two fungi alleviated salt stress appeared to be based on physiological processes (increased CER, transpiration, stomatal conductance and WUE) rather than on nutrient uptake (N or P).  相似文献

This study evaluates antioxidant responses and jasmonate regulation in Digitaria eriantha cv. Sudafricana plants inoculated (AM) and non-inoculated (non-AM) with Rhizophagus irregularis and subjected to drought, cold, or salinity. Stomatal conductance, photosynthetic efficiency, biomass production, hydrogen peroxide accumulation, lipid peroxidation, antioxidants enzymes activities, and jasmonate levels were determined. Stomatal conductance and photosynthetic efficiency decreased in AM and non-AM plants under all stress conditions. However, AM plants subjected to drought, salinity, or non-stress conditions showed significantly higher stomatal conductance values. AM plants subjected to drought or non-stress conditions increased their shoot/root biomass ratios, whereas salinity and cold caused a decrease in these ratios. Hydrogen peroxide accumulation, which was high in non-AM plant roots under all treatments, increased significantly in non-AM plant shoots under cold stress and in AM plants under non-stress and drought conditions. Lipid peroxidation increased in the roots of all plants under drought conditions. In shoots, although lipid peroxidation decreased in AM plants under non-stress and cold conditions, it increased under drought and salinity. AM plants consistently showed high catalase (CAT) and ascorbate peroxidase (APX) activity under all treatments. By contrast, the glutathione reductase (GR) and superoxide dismutase (SOD) activity of AM roots was lower than that of non-AM plants and increased in shoots. The endogenous levels of cis-12-oxophytodienoc acid (OPDA), jasmonic acid (JA), and 12-OH-JA showed a significant increase in AM plants as compared to non-AM plants. 11-OH-JA content only increased in AM plants subjected to drought. Results show that D. eriantha is sensitive to drought, salinity, and cold stresses and that inoculation with AM fungi regulates its physiology and performance under such conditions, with antioxidants and jasmonates being involved in this process.  相似文献

Summary Maize was grown under axenic conditions in laboratory devices, in a K+-deficient medium, where biotite was the K+ source. In different treatments plants were inoculated by symbiotic (Glomus mosseae) and/or non symbiotic microflora. In those treatments inoculated byGlomus mosseae, the percentage of roots infection after 7 weeks plant growth was 65%. Rhizospheric bacterial population was approximately 108/g (dry weight). Endomycorrhizae stimulated growth and K uptake. Non-symbiotic microflora increased also plant growth but promoted much more biotite weathering and K uptake. Endomycorrhizae and more particularly non-symbiotic microflora increased also Ca and Mg absorption by plants. Possible mechanisms involved and implications in plant growth and pedogenesis are discussed.  相似文献

The effects of arbuscular mycorrhizal (AM) fungus, Glomus mosseae, on growth, osmotic adjustment and antioxidant enzymes of bahia grass (Paspalum notatum) were studied in potted plants under water stress conditions. AM colonization significantly enhanced the plant height, root and shoot fresh weight, Phosphorus (P), potasium (K), manganese (Mn) contents in shoots, P, calcium (Ca), Mn contents in roots, whereas obviously decreased zinc (Zn) content in shoots, iron (Fe), boron (B), copper (Cu) contents in roots. During water stress, the relative water and chlorophyll contents were relatively stable and signifciantly higher in AM than in non-AM plants, AM inoculation notabley decreased the shoot relative conductivity and malondialdehyde (MDA) content, markedly increased shoot peroxidase (POD) activity and proline content, while AM infection did not affect the dismutase (SOD) activity of shoots. Our results suggested that AM colonization improved the protective enzyme activity (such as POD) and osmotic adjustment originating from proline P, K, Ca, resulting in the enhancement of drought tolerance.  相似文献

This study examined the uptake of nitrogen by external hyphae of an arbuscular mycorrhizal (AM) fungus (Glomus intraradices Schenck &; Smith) and its impact on physiological responses in maize plants subjected to well-watered or drought-stressed conditions. Plants were grown in compartmented boxes divided by a nylon mesh (40?μm) into a root compartment and a hyphal compartment. Maize plants (Zea mays cv. 'Tuxpeño sequia' selection cycle C0) were exposed to 2 weeks of drought 56 days after sowing. A ^[15]N tracer was applied as K^[15]NO_[3] to the hyphal compartment at a distance of 5?cm from the root compartment. Root and shoot samples were then analyzed for ^[15]N atom % excess (APE), glutamine synthetase (GS) activity, protein concentration and nutritional status. Evapotranspiration rate and stomatal resistance were monitored daily to determine the degree of drought stress. The APE values for AM shoots and roots were 32% and 33% higher than non-AM shoots and roots, respectively, under drought conditions. This provides clear evidence that the external mycelium of AM fungus transports considerable amounts of ^[15]NO_[3]^[– ]to the host plant under drought conditions. Drought-stressed AM roots had 28% higher GS activity, possibly as a consequence of higher hyphal acquisition of NO_[3]^[–] ions. Mycorrhizal colonization significantly increased the host plant P status regardless of soil moisture regime. In addition, the N status of drought-stressed AM shoots and roots was slightly higher than stressed non-AM shoots and roots. The improved nutritional status may assist AM plants to exploit available soil moisture more efficiently and to maintain higher leaf relative water content under moderate drought conditions.  相似文献

 The influence of an arbuscular mycorrhizal (AM) fungus on phosphorus (P) and iron (Fe) uptake of peanut (Arachis hypogea L.) and sorghum (Sorghum bicolor L.) plants was studied in a pot experiment under controlled environmental conditions. The plants were grown for 10 weeks in pots containing sterilised calcareous soil with two levels of Fe supply. The soil was inoculated with rhizosphere microorganisms only or with rhizosphere microorganisms together with an AM fungus (Glomus mosseae [Nicol. & Gerd.] Gerdemann & Trappe). An additional small soil compartment accessible to hyphae but not roots was added to each pot after 6 weeks of plant growth. Radiolabelled P and Fe were supplied to the hyphae compartment 2 weeks after addition of this compartment. After a further 2 weeks, plants were harvested and shoots were analysed for radiolabelled elements. In both plant species, P uptake from the labelled soil increased significantly more in shoots of mycorrhizal plants than non-mycorrhizal plants, thus confirming the well-known activity of the fungus in P uptake. Mycorrhizal inoculation had no significant influence on the concentration of labelled Fe in shoots of peanut plants. In contrast, 59Fe increased in shoots of mycorrhizal sorghum plants. The uptake of Fe from labelled soil by sorghum was particularly high under conditions producing a low Fe nutritional status of the plants. These results are preliminary evidence that hyphae of an arbuscular mycorrhizal fungus can mobilise and/or take up Fe from soil and translocate it to the plant. Accepted: 6 March 1998  相似文献

 Seedlings of papaya (Carica papaya L. var. Solo) were transplanted to pots with or without an arbuscular mycorrhizal (AM) fungus (Gigaspora margarita Becker and Hall). After 3 months, half the plants were subjected to water stress by withdrawing irrigation. The leaf water potential (LWP) was measured during 20 days of water-stress treatment and then the plants were harvested. Root ethylene and 1-aminocyclopropane-1-carboxylic acid (ACC) concentrations were measured and plant fresh weight determined. The LWP decreased during the water-stress treatment and this decrease was more severe in the non-AM plants. Plant fresh weight was higher for AM than non-AM plants under both conditions. Under well-irrigated conditions, the ethylene concentration in the roots was increased by the presence of AM, although there was no significant difference between AM and non-AM roots in ACC levels. ACC increased in both AM and non-AM roots under water-stress conditions. The water-stress treatment resulted in a marked increase in ethylene concentration in non-AM roots but the concentration in AM roots was slightly lower than under normal conditions. This study investigated several aspects related to drought tolerance in arbuscular mycorrhizal (AM) soybean plants. The investigation included both shoot and root tissues in order to reveal the preferred target tissue for AM effects against drought stress. Non-AM and AM soybean plants were grown under well-watered or drought-stressed conditions, and leaf water status, solute accumulation, oxidative damage to lipids, and other parameters were determined. Results showed that AM plants were protected against drought, as shown by their significantly higher shoot-biomass production. The leaf water potential was also higher in stressed AM plants (-1.9 MPa) than in non-AM plants (-2.5 MPa). The AM roots had accumulated more proline than non-AM roots, while the opposite was observed in shoots. Lipid peroxides were 55% lower in shoots of droughted AM plants than in droughted non-AM plants. Since there was no correlation between the lower oxidative damage to lipids in AM plants and the activity of antioxidant enzymes, it seems that first the AM symbiosis enhanced osmotic adjustment in roots, which could contribute to maintaining a water potential gradient favourable to the water entrance from soil into the roots. This enabled higher leaf water potential in AM plants during drought and kept the plants protected against oxidative stress, and these cumulative effects increased the plant tolerance to drought.  相似文献

In order to understand the salt-tolerance mechanism of alkali grass (Puccinellia tenuiflora) compared with wheat (Triticum aestivum L.), [K(+)] and [Na(+)] in roots and shoots in response to salt treatments were examined with ion element analysis and X-ray microanalysis. Both the rapid K(+) and Na(+) influx in response to different NaCl and KCl treatments, and the accumulation of K(+) and Na(+) as the plants acclimated to long-term stress were studied in culture- solution experiments. A higher K(+) uptake under normal and saline conditions was evident in alkali grass compared with that in wheat, and electrophysiological analyses indicated that the different uptake probably resulted from the higher K(+)/Na(+) selectivity of the plasma membrane. When external [K(+)] was high, K(+) uptake and transport from roots to shoots were inhibited by exogenous Cs(+), while TEA (tetraethylammonium) only inhibited K(+) transport from the root to the shoot. K(+) uptake was not influenced by Cs(+) when plants were K(+) starved. It was shown by X-ray microanalysis that high [K(+)] and low [Na(+)] existed in the endodermal cells of alkali grass roots, suggesting this to be the tissue where Cs(+) inhibition occurs. These results suggest that the K(+)/Na(+) selectivity of potassium channels and the existence of an apoplastic barrier, the Casparian bands of the endodermis, lead to the lateral gradient of K(+) and Na(+) across root tissue, resulting not only in high levels of [K(+)] in the shoot but also a large [Na(+)] gradient between the root and the shoot.  相似文献

This greenhouse study aimed to determine the effect of colonization by the arbuscular mycorrhizal (AM) fungus (Glomus intraradices Schenck & Smith) on the “wild” tobacco (Nicotiana rustica L. var. Azteca), under soil–zinc (Zn) conditions. Plants of N. rustica were grown in AM or non-AM inoculated substrate and subjected to four soil–[Zn] concentrations (0, 50, 100, and 250 mg Zn kg−1 dry soil). The AM root colonization increased markedly from 14 to 81% with the increasing soil–[Zn] and the mycorrhizal structures were significantly more abundant at the highest soil–[Zn], suggesting that Zn may be involved directly or indirectly in AM root colonization. In addition, total Zn content or Zn concentrations in shoots and roots were shown to increase as soil–[Zn] increased in both AM and non-AM plants. As for the growth parameters studied, there were no significant differences between treatments despite the increase in Zn content or concentration. The AM roots subjected to the highest soil–[Zn] had a significant reduction by about 50% of total Zn content and Zn concentration compared to non-AM roots. Still, the relative extracted Zn percentage decreased dramatically as soil–[Zn] increased. Soil pH was significantly lower in non-AM than AM treatments at the highest soil–[Zn]. In summary, AM plants (particularly roots) showed lower Zn content and concentration than non-AM plants. In this regard, the AM fungi have a protective role for the host plant, thus playing an important role in soil-contaminant immobilization processes; and, therefore, are of value in phytoremediation, especially when heavy metals approach toxic levels in the soil.  相似文献

The influence of Glomus etunicatum colonization on plant growth and drought tolerance of 3-month-old Pistacia vera seedlings in potted culture was studied in two different water treatments. The arbuscular mycorrhiza (AM) inoculation and plant growth (including plant shoot and root weight, leaf area, and total chlorophyll) were higher for well-watered than for water-stressed plants. The growth of AM-treated seedlings was higher than non-AM-treatment regardless of water status. P, K, Zn and Cu contents in AM-treated shoots were greater than those in non-AM shoots under well-watered conditions and drought stress. N and Ca content were higher under drought stress, while AM symbiosis did not affect the Mg content. The contents of soluble sugars, proteins, flavonoid and proline were higher in mycorrhizal than non-mycorrhizal-treated plants under the whole water regime. AM colonization increased the activities of peroxidase enzyme in treatments, but did not affect the catalase activity in shoots and roots under well-watered conditions and drought stress. We conclude that AM colonization improved the drought tolerance of P. vera seedlings by increasing the accumulation of osmotic adjustment compounds, nutritional and antioxidant enzyme activity. It appears that AM formation enhanced the drought tolerance of pistachio plants, which increased host biomass and plant growth.  相似文献

This greenhouse study aimed to examine the contribution of arbuscular mycorrhizal (AM) colonization on the uptake of and tolerance to nickel (Ni) in sunflower (Helianthus annuus L.). We hypothesized that AM colonization increases Ni content and tolerance in sunflower grown under varying soil Ni concentrations. The combined effect of AM colonization and soil Ni input on the assimilation of nitrogen, in particular the activity of glutamine synthetase (GS), in sunflower plants was also investigated. A factorial experimental design was performed with sunflower cv. Lemon Queen, with or without the AM fungus, Glomus intraradices Schenck & Smith, and treated with 0, 100, 200, or 400 mg Ni kg−1 dry soil (DS). The AM colonization significantly enhanced plant growth and Ni content, especially at the lower soil Ni treatments. Furthermore, the AM plants exposed to the highest soil Ni level of 400 mg Ni kg−1 DS had a significantly higher shoot Ni extracted percentage than non-AM plants, suggesting that the AM symbiosis contributed to Ni uptake, then its translocation from roots to shoots. The AM colonization also significantly increased the GS activity in roots, this being likely an indicator of an enhanced Ni tolerance. These findings support the hypothesis that AM symbiosis contributes to an enhanced Ni plant uptake and tolerance and should be considered as part of phytoremediation strategies.  相似文献

This study investigated the accumulation of osmotic solutes in citrus (Poncirus trifoliata) seedlings colonized by Glomus versiforme subjected to drought stress or kept well watered. Development of mycorrhizae was higher under well watered than under drought-stressed conditions. Arbuscular mycorrhizal (AM) seedlings accumulated more soluble sugars, soluble starch and total non-structural carbohydrates in leaves and roots than corresponding non-AM seedlings regardless of soil-water status. Glucose and sucrose contents of well-watered and drought-stressed roots, fructose contents of well-watered roots and sucrose contents of drought-stressed leaves were notably higher in AM than in non-AM seedlings. K+ and Ca2+ levels in AM leaves and roots were greater than those in non-AM leaves and roots, while AM symbiosis did not affect the Mg2+ level. AM seedlings accumulated less proline than non-AM seedlings. AM symbiosis altered both the allocation of carbohydrate to roots and the net osmotic solute accumulations in response to drought stress. It is concluded that AM colonization enhances osmotic solute accumulation of trifoliate orange seedlings, thus providing better osmotic adjustment in AM seedlings, which did not correlate with proline but with K+, Ca2+, Mg2+, glucose, fructose and sucrose accumulation.  相似文献

The impact of different defoliation intensities on the ability of Lotus tenuis plants to regrowth, mobilise nutrients and to associate with native AM fungi and Rhizobium in a saline‐sodic soil was investigated. After 70 days, plants were subjected to 0, 25, 50, 75 and 100% defoliation and shoot regrowth was assessed at the end of subsequent 35 days. Compared to non‐defoliated plants, low or moderate defoliation up to 75% did not affect shoot regrowth. However, 100% treatment affected shoot regrowth and the clipped plants were not able to compensate the growth attained by non‐defoliated plants. Root growth was more affected by defoliation than shoot growth. P and N concentrations in shoots and roots increased with increasing defoliation while Na+ concentration in shoots of non‐defoliated and moderately defoliated plants was similar. Non‐defoliated and moderately defoliated plants prevented increases of Na+ concentration in shoots through both reducing Na+ uptake and Na+ transport to shoots by accumulating Na+ in roots. At high defoliation, the salinity tolerance mechanism is altered and Na+ concentration in shoots was higher than in roots. Reduction in the photosynthetic capacity induced by defoliation neither changed the root length colonised by AM fungi nor arbuscular colonisation but decreased the vesicular colonisation. Spore density did not change, but hyphal density and Rhizobium nodules increased with defoliation. The strategy of the AM symbiont consists in investing most of the C resources to preferentially retain arbuscular colonisation as well as inoculum density in the soil.  相似文献

The aim of this study is to investigate the effects of arbuscular mycorrhizal fungi (AMF) on garlic plants growth and the uptake of selenium (Se). Garlic plants were grown in the pots inoculated with Glomus fasciculatum and G. mosseae and maintained in a greenhouse. Three weeks after planting, the pots had received different concentrations of Se (5, 10, 15, 20, 25 mg kg?1 of soil) in the form of selenium dioxide (SeO2) at 3 weeks intervals up to 12 weeks. For physiological and biochemical analysis, the samples were randomly collected from five plants of each experiment. Maximum AM infection, spore population and plant biomass were observed in the roots of mycorrhizal-mediated plants without Se, and they were gradually declined in both mycorrhizal and non-mycorrhizal (NM) plants with increasing concentrations of Se. Among the two Glomus species tested, G. fasciculatum-mediated plants showed higher AM infection, spore population and plant biomass than G. mosseae. No differences were observed for the uptake of Se in mycorrhizal plants and NM plants. However, NM plants uptake more Se than mycorrhizal plants. Higher contents of total chlorophyll and sugars were observed in plants inoculated with G. fasciculatum without Se and they were decreased in the presence of Se. In contrast, increased amount of glutathione peroxidase was observed at increasing concentrations of Se up to 20 mg kg?1. High-performance liquid chromatography data revealed that SeO2 converted to organic form of Se as γ-glutamyl-Se-methylselenocysteine. These results are basis for further investigations on the role of AMF on plant growth and uptake of Se in crop plants.  相似文献

Three arbuscular mycorrhizal (AM) fungi (Glomus mosseae, Glomus claroideum, and Glomus intraradices) were compared for their root colonizing ability and activity in the root of Astragalus sinicus L. under salt-stressed soil conditions. Mycorrhizal formation, activity of fungal succinate dehydrogenase, and alkaline phosphatase, as well as plant biomass, were evaluated after 7 weeks of plant growth. Increasing the concentration of NaCl in soil generally decreased the dry weight of shoots and roots. Inoculation with AM fungi significantly alleviated inhibitory effect of salt stress. G. intraradices was the most efficient AM fungus compared with the other two fungi in terms of root colonization and enzyme activity. Nested PCR revealed that in root system of plants inoculated with a mix of the three AM fungi and grown under salt stress, the majority of mycorrhizal root fragments were colonized by one or two AM fungi, and some roots were colonized by all the three. Compared to inoculation alone, the frequency of G. mosseae in roots increased in the presence of the other two fungal species and highest level of NaCl, suggesting a synergistic interaction between these fungi under salt stress.  相似文献
