Time course, up to 100d, of uptake and metabolism of aldrin and dieldrin added at subculture to suspension cultures from Phaseolus vulgaris (French bean) root and shoot, and Solanum tuberosum (potato) tuber comparable, with rapid dieldrin production and delayed appearance of other metabolites. When aldrin and dieldrin not added to Phaseolus cultures until 10 or 20d after subculture usual extent of conversion of aldrin to dieldrin, but reduced production of other metabolites, and growth inhibition negated. Increasing volumes of 2-methoxyethanol had detrimental effect on growth and uptake and metabolism. Dieldrin production maximal during rapid growth phase and probably independent of other conversions.  相似文献

An extensive clinical and epidemiological study of workers engaged in the manufacturing and formulation of aldrin and dieldrin, the Pernis study, provides occupational hygiene and biological monitoring data on individual exposures over the years of employment and provides the opportunity to investigate dose response relationships for these chemicals. The human epide miological mortality data on these workers, who were exposed to fairly substan tial lifetime average daily doses of aldrin and dieldrin, suggest that low dose exposures do not significantly increase human mortality and may even de crease the human mortality hazard rate. While hormesis from low dose expo sure to aldrin and dieldrin is not statistically significant, it is observed in the raw data and in the best fitting dose response models. The decrease in risk suggests increased survival time at low doses of aldrin and dieldrin. Using an upper bound on cancer potency based on mouse liver tumors, the U.S. Environmental Protection Agency (EPA) estimated that lifetime average daily doses (LADDs) of 0.0000625 and 0.00625?µg/kg body weight/day would correspond to increased cancer risks of 0.000001 and 0.0001, respectively. However, the best estimate from the Pernis epidemiological data is that LADDs of 0.0000625 and 0.00625?µg/kg body weight/day correspond to no increase in cancer risk and a decrease in the probability of mortality from all causes by the age of 70 years. At low doses of aldrin and dieldrin, the estimated decrease in mortality in a reference period of 70 years is more than 1000 times larger than the U.S. EPA's upper bound on the increase in the lifetime probability of cancer.  相似文献

Laboratory techniques are described for the estimation of the stomach poison, direct and residual film contact poison and fumigant poison effects of chemicals to adult worker honey-bees.
The toxicity of eleven chemicals used in plant protection has been investigated by these methods. The order of effectiveness as stomach and contact poisons was: parathion, TEPP, γ-BHC, dieldrin, aldrin, chlordane, o , o -diethyl- o -ethylmercaptoethyl thiophosphate (constituent of Systox), bisdimethylamino fluorophosphine oxide, toxaphene and the sodium salts of 2:4-D and MCPA: as residual films, dieldrin, aldrin, γ-BHC, parathion, chlordane, and o , o -diethyl- o -ethylmercapto-ethyl thiophosphate (constituent of Systox); toxaphene, TEPP and bisdimethylamino fluorophosphine oxide had no measurable effect; as fumigants, dieldrin, γ-BHC, aldrin, parathion, and chlordane; the remainder had no measurable effect.  相似文献

Twenty-two strains of soil bacteria, including representatives of the genera Bacillus, Micromonospora, Mycobacterium, Nocardia, Streptomyces, Thermoactinomyces, and Pseudomonas and 10 unidentified gram-negative, motile, rod-shaped bacteria, were shown to degrade aldrin to its epoxide dieldrin. Twenty microbial cultures which had been shown to degrade dieldrin were tested to determine their ability to degrade endrin, aldrin, DDT, gamma isomers of benzenehexachloride (gamma-BHC), and Baygon. All isolates were able to degrade DDT and endrin, whereas 13 degraded aldrin. However, none of them was able to degrade Baygon or gamma-BHC.  相似文献

We isolated a novel aerobic dieldrin-degrading bacterium from an enrichment culture in a soil–charcoal perfusion system. Enrichment culture using a soil–charcoal perfusion system was an effective way to obtain microorganisms that degrade recalcitrant compounds. The soil–charcoal perfusion was performed using aldrin trans-diol, which was a metabolite of dieldrin. Aldrin trans-diol had higher bioavailability (2.5 mg/l) than dieldrin (0.1–0.25 mg/l), therefore it is possible for microorganisms to utilize it as a substrate in soil. After 100 days of circulation and three exchanges of the medium, the enriched charcoal was harvested and a bacterium isolated. The isolate was designated as strain KSF27 and was found to be closely related to Pseudonocardia spp. as determined by 16S rRNA sequencing analysis. Strain KSF27 degraded aldrin trans-diol by 0.05 μmol/l from an initial concentration of 25.5 μmol/l. The metabolite of aldrin trans-diol was detected by HPLC/MS and determined to be aldrindicarboxylic acid based on retention time and the MS fragment. Moreover, strain KSF27 degraded dieldrin from 14.06 μmol/l to 2.01 μmol/l over a 10-day incubation at 30 °C. This strain degraded dieldrin and other persistent organochlorine pesticides, such as α-endosulfan, β-endosulfan, endosulfan sulfate, heptachlor, heptachlor epoxide and chlordecone.  相似文献

The primary objective of this study was to determine specific cytochrome P450 isozyme(s) involved in the metabolism of aldrin to its toxic metabolite dieldrin in flathead mullet (Mugil cephalus) liver microsomes. To identify the cytochrome P450 isozyme responsible for the aldrin metabolism in mullet liver, the effects of mammalian‐specific cytochrome P450 inhibitors and substrates were determined in the epoxidation reaction of aldrin. CYP3A‐related inhibitors, ketoconazole, SKF‐525A, and cimetidine, inhibited the metabolism of aldrin. The contribution of CYP1A to the aldrin metabolism was shown by the inhibition of 7‐ethoxyresorufin‐O‐deethylase activity in the presence of aldrin. The results indicate that CY1A and CYP3A are the cytochrome P450s involved in aldrin epoxidase activity in mullet. In addition, the suitability of aldrin epoxidase activity for monitoring of environmental pollution was also assessed in the fish samples caught from four different locations of the West Black Sea coast of Turkey.  相似文献

During 1963-89, 627 Barn Owl Tyto alba carcasses were received for autopsy and chemical analysis. Much larger numbers were received per month outside the breeding season than within it, with peaks in autumn (mainly juveniles) and late winter (adults and juveniles).
The main causes of recorded deaths were collisions (mostly with road traffic) and starvation. No great seasonal variation occurred in the main causes of recorded deaths and starved juveniles were reported even in summer. Most starved males weighed less than 240 g, and most starved females less than 250 g.
Another important cause of mortality in eastern arable counties, at least to 1977, was poisoning by organochlorine pesticides, especially aldrin/dieldrin. Levels of HEOD (the metabolized product of aldrin/dieldrin) in the livers of birds that had apparently died of aldrin/ dieldrin poisoning were in the range 6–44 ppm (geometric mean 14 ppm). Pesticide victims formed up to 40% of all dead Barn Owls obtained from some eastern counties during 1963-77. By 1987-89, HEOD levels in Barn Owls in eastern counties had fallen to less than 1.6 ppm, and no deaths from organochlorine poisoning were recorded.
Organochlorine pesticides almost certainly contributed to population decline in eastern England evident in the 1950s and 1960s, and reductions in the use of these chemicals may have allowed a subsequent increase, apparent over the last 10–15 years.  相似文献

Residues of chlorinated hydrocarbon insectidies and some of their metabolites, including p,p'-DDT, p,p'-DDD, p,p'-DDE, gamma-BHC, aldrin, dieldrin, heptachlor and heptachlor epoxide, were examined in the soils collected from 21 tea-gardens distributed in 15 countries of 6 prefectures in 1977, and from 100 locations distributed in 46 counties of 10 prefectures in 1984 in Taiwan. Although the use of these chlorinated hydrocarbon insecticides has been banned for many years, minor amounts of residues in the soils have still been detected. From 1977 to 1984, average residues of the chlorinated hydrocarbon insecticides and their metabolites in the soils of tea-gardens decreased from 5.64 to 0.06 ppb for gamma-BHC, 97 54 to 1.10 ppb for aldrin, 35.91 to 1.00 ppb for p,p'-DDE, 17.41 to 0.28 ppb for dieldrin, and 30.17 to 3.42 ppb for p,p'-DDT. But the number of positive cases increased for all chlorinated hydrocarbon insecticides tested, the residual levels were not significantly different among soils with different physiochemical properties or among soil layers. No significant amount of these insecticides was found in the processed tea.  相似文献

The sensitivity of catfish, Ictalurus punctatus, brain ATPase activities to cyclodiene compounds was investigated. The ATPase system showed differences in sensitivity to aldrin, dieldrin and photodieldrin. However, aldrin-transdiol (a more terminal metabolite of dieldrin and reported as a more potent neurotoxin than dieldrin) had no effect on any ATPase activity from fish brain homogenates. Mitochondrial Mg2+ ATPase was the most sensitive ATPase to the cyclodiene compounds tested. The possibility that the neurotoxic effects of these compounds is a secondary response resulting from mitochondrial Mg2+ ATPase inhibition is discussed.  相似文献

Reproducibility of metabolism of aldrin and dieldrin in cell suspension cultures from Phaseolus vulgaris (French Bean) root, and effects of variations in hormonal supplementation and light, studied. Aldrin always converted to dieldrin, aldrin-transdihydrodiol, and other unidentified more polar metabolites, but not to dihydrochlordene-dicarboxylic acid. Dieldrin always converted to photodieldrin and more polar metabolites, but not to aldrin-transdihydrodiol. Metabolism reasonably consistent from one subculture to next, basically unaffected by alterations in medium tested, or by presence or absence of light. Variations observed quantitative rather than qualitative supporting validity of use of tissue culture in metabolic studies on xenobiotics. Photodieldrin shown to be a direct product of dieldrin metabolism and not the result of microbial or environmental interference.  相似文献

A discharge of aldrin and dieldrin caused mortality of salmonid fry in a hatchery. Pesticide residues in fish flesh and resulting management action are described. Differences in wild fish stock densities could be due to pesticides but other possibilities are explored. Dieldrin was widely distributed, even at control sites, at levels considered potentially dangerous to piscivores. The effect of the cyclodiene organochlorine pesticides aldrin, dieldrin and endosulfan was assessed on CHO-K1 cultures at fractions of their lethal doses, determined by the neutral red (NRI) incorporation assay (NRI6.25, NRI12.5 and NRI25). Glutathione peroxidase, reductase and S-transferase, and total and oxidised glutathione were evaluated along the standard growth curve of the cultures. After a 24-h incubation with each insecticide, glutathione peroxidase incurred a large increase, while glutathione reductase and S-transferase activities were slightly higher than untreated controls. Unlike oxidised glutathione, the content of total glutathione declined significantly after exposure to cyclodiene insecticides. Changes in cell membrane integrity were assessed by the lactate dehydrogenase (LDH) release assay and lipid peroxidation for a wide range of pesticide concentrations. Membrane leakage and peroxide production were significantly enhanced at concentrations of aldrin and as low as 12.5 μg/ml, whereas dieldrin and endosulfan increased membrane fragility at much higher concentrations.  相似文献

The membrane system which epoxidizes aldrin to dieldrin in pea roots was separated from polyphenolic inhibitors and soluble peroxidase by rapid anaerobic gel filtration. Epoxidase activity was strongly inhibited by cytochrome-c, cyanide, electron acceptors and polyphenols and was enhanced by chelating and solubilizing agents which inhibit cytochrome P450 systems.  相似文献

Food consumption is one of the main routes of human exposure to organochlorine pesticide residues (OCPRs). To assess the potential health risks associated with OCPRs contaminants due to freshwater organism consumption, a number of vegetables, prawn, snail, and fish were collected from Khlong 7 (canal), Rangsit agricultural area, Pathum-Thani Province, Thailand. The samples were extracted using a multiresidue extraction method and then analyzed by gas chromatography with microelectron capture detector (μ -ECD). The results show that low concentrations of OCPRs were detected in parts per billion (ppb) levels. Based on a plausible worst-case scenario, the local population could be at risk for cancer due to consumption of fish contaminated by α -, β -HCH, heptachlor, heptachlor epoxide, aldrin, dieldrin, DDD, DDE, and DDT. Likewise, individuals may be at risk from consumption of Lanchester's freshwater prawn Macrobrachium lanchesteri, freshwater snail Filopaludina mertensi, swamp morning-glory Ipomomea aquatica, neptunia Neptunia oleracea, and water lily Nymphaea lotus because these species all contained elevated concentrations of α -, β -HCH, heptachlor, heptachlor epoxide, aldrin, and dieldrin.  相似文献

Of these, only [14C]lindane and [14C]chlordane underwent extensive biodegradation, as evidenced by the fact that 9.4 to 23.4% of these compounds were degraded to 14CO2 in 30 days in liquid cultures and 60 days in soil-corncob cultures inoculated with P. chrysosporium. Although [14C]aldrin, [14C]dieldrin, [14C]heptachlor, and [14D]mirex were poorly mineralized, substantial bioconversion occurred, as determined by substrate disappearance and metabolite formation. Ninety-three wild-type isolates identified as actinomycetes were tested against 11 organochlorine pesticides (OPs): aldrin, chlordane, DDD, DDE, DDT, dieldrin, heptachlor, and heptachlor epoxides, lindane, and methoxychlor. Qualitative screening agar assays displayed 62-78% tolerance of strains to OPs. Four strains designed M4, M7, M9 and M15 were selected based on multi-OP-tolerance, and identified as members of the streptomycetes group. Different growth profiles were observed in cultures of the four selected streptomycetes cultured in synthetic medium containing 5-50 microg x l(-1) aldrin or chlordane or lindane. Increase of aldrin removal by the selected microorganisms was concomitant with the 4.8-36.0 microg x l(-1) pesticide concentration range. After 72 h of streptomycete M7 growth in synthetic medium containing 48.0 microg x l(-1) aldrin, the remaining OP concentration in the supernatant was approximately 10% of the initial concentration. Also, in stationary growth phase less than 2.5 microg x l(-1) aldrin residual concentration was detected in the medium.  相似文献

Field experiments on the effect of residues of aldrin and dieldrin in the soil on the beetle predators of the cabbage root fly, showed that populations of adult carabid and staphylinid predators were altered by aldrin/dieldrin residues at concentrations of 0.06 p.p.m. and above. The lower insecticidal concentrations were aged residues which had become intimately mixed with the soil. Although the residues reduced the numbers of trapped Aleochara spp., concentrations below 0.5 p.p.m. increased the numbers of Bembidion lampros, probably because their movement was stimulated by the insecticide. Harpalus spp. and Feronia spp. were unaffected by residues below the concentration toxic to cabbage root-fly eggs and larvae. The reductions of the predators implied by the trapping records increased the survival of the cabbage root-fly eggs and larvae, thus leading to augmented damage to cauliflowers, cabbage and Brussels sprouts, which resulted in reductions in yield of up to 70%. Larval and pupal parasitism by Aleochara spp. and Idiomorpha rapae were also reduced by the insecticide residues, although concentrations of 0.2 p.p.m. and above were required to decrease parasitism by I. rapae.  相似文献

The current study was conducted to establish the levels of organochlorine pesticides (OCPs), its degradation products, and indicatory polychlorinated biphenyls (PCBs) in water and sediments from Lake Victoria in Tanzania. Seven indicator PCBs were identified in sediments, and five were identified in water. The PCBs loading ranged from lower than the limit of detection (LOD) to 10.28 µg kg?1 DW and 0.95 to 2.24 µg l?1 in sediments and water, respectively. PCBs, CB 138 and CB 153 dominated the load, because of their chemical stability and high degree of chlorination. In addition, ten OCPs were identified in sediments, and seven OCPs were identified in water. The levels of organochlorine compounds in sediments were higher than in water samples, implying that sediments may serve as a sink for these compounds. The presence of these compounds is probably as a result of either historical use, long-range environmental transport or environmental persistence. The levels of organochlorine compounds in the lake water and sediments are below the maximum residue limit set by EU and FAO, except for aldrin and dieldrin. Based on the Threshold Effect Concentration (TEC) for freshwater ecosystems, aldrin and dieldrin are the only OCPs that seem to be a threat to the lake environment.  相似文献
