A comparative electron microscope study at magnifications ranging from about 80,000 up to 800,000 x was carried out in nine species of insects (Gryllus argentinus, Myogryllus verticalis, undetermined species of Gryllus; Blaptica dubia, Periplaneta americana, Blattella germanica; Laplatacris dispar, Aleua lineata and Omexechae servillei). Particular attention was paid: a) to the elementary components of the s. complexes and b) to the structure of their medial ribbon. - a) In all the species examined the basic element found is a curled filament some 15–20 Å thick. Filaments of this kind integrate: the 100 Å fibrils of the chromosome body, the compacts layers of the s. complexes (lateral arms) and the slender planes of the pairing space. The filaments are similar to those described in metaphase chromosomes and their kinetochores (Wettstein and Sotelo, 1965). A difference in density between the filaments of the lateral arms and those of the medial planes is sometimes noticed. - b) Three structural patterns were found in the pairing space. In crickets, the medial ribbon is composed of three parallel, longitudinal planes of filaments, interconnected and connected with the lateral arms by means of bridges. The latter are constituted by fibrils or by single filaments. In cockroaches only two longitudinal planes were found. The distribution of components in these planes follows a plan similar to the one found in crickets. In the electron-micrographs the medial component of both groups of Insecta appears as composed of three (crickets) or two (cockroaches) lines in the longitudinal frontal views, and ladder-like striated in lateral views. The latter striae correspond to filaments or groups of filaments running in antero-posterior direction. - The pattern of structure of grasshoppers differs completely from those mentioned above. Bridging between the homologues is made of regularly spaced transversal planes of filaments. Chromatin organization during the early stages of male meiotic prophase inBombyx mori was investigated by electron microscopy. The analysis of nuclei prepared by the Miller spreading procedure, suggests that chromatin fibers which are 200–300 Å in diameter undergo an orderly folding coincident with the formation of the synaptonemal complex. In very early stages the chromatin is released in linear arrays typical of interphase chromatin material. With time loops containing 5–25 of B conformation DNA, initially visualized at the periphery of early meiotic prophase nuclei, aggregate into discrete foci. These foci coalesce to form the longitudinal axis of the chromosome in conjunction with the initial appearance of the axial elements of the synaptonemal complex. At pachytene, the loops are evenly distributed along the length of the chromosome and extend radially so that in well spread preparations the chromosome has a brush-like appearance. Throughout this period nascent RNP-fibers were visualized along some of the loops.  相似文献

The light microscope showed that zygotene and pachytene were completely suppressed in pollen mother cells of an asynaptic mutant of Triticum durum; the chromosomes passed through a normal chromomeral leptotene condition and remained unpaired throughout prophase. The electron microscope confirmed the absence of synaptinemal complexes, as would be expected with no pairing. Prominent opaque axial cores were present in the chromatin from the onset of leptotene up to an indeterminate stage during prophase condensation. At an early time during condensation 150 Å particles appeared between chromatin masses. Coincident to the disappearance of axial cores from the chromatin, polycomplexes consisting of linearly associated core fragments arrayed in single layer sheets appeared between chromatin masses. The aligned fragments were invariably spaced about 625 Å from centre to centre; this is approximately half the distance between centres of the lateral elements (axial cores) of the synaptinemal complex of pachytene of synaptic sister seedlings. There was no central element between the associated fragments. The significance of these observations is discussed, as is also the essential difference between asynapsis and desynapsia.  相似文献

Whole-mounted polytene chromosomes from Drosophila melanogaster were prepared for high-voltage electron microscopy. Relative dry mass of chromosome regions was estimated by densitometry of electron microscopic negatives. Comparison of dry mass of regions of the male X chromosome with that of regions of associated autosomes established that dry mass values are proportional to DNA content. Relative dry mass values of regions of polytene chromosomes from salivary glands, fat body, and malpighian tubules were correlated with the average diameter of bands in these regions: as mass doubled, band width increased by a factor of approximately 2. To provide a standard for estimating absolute levels of polyteny, band widths were measured for chromosomes representing one major polytene class, 256n. These chromosomes were observed to have an average band width of 0.9 m — These observations provide limits to models of chromatin organization in bands. For each chromatid, this area can accommodate up to five chromatin fibers of 250 Å diameter. This value may represent the extent of folding of a chromatin fiber in an average band. Alternatively, a chromatin fiber of higher-order structure could have a maximum diameter of 560 Å in an average band.  相似文献

Summary This paper reports on the structure of rat primary oocytes, as observed with the electron microscope. Four main components are described in the cytoplasm: Golgi apparatus, centrioles, mitochondria and multivesicular bodies.The components of the Golgi apparatus are forming a single mass confined to a limited region of the cytoplasm and the centrioles were found located in a clear zone sited in the middle of this mass. Mitochondria are scattered at random in the cytoplasm. Multivesicular bodies are elements integrated by an enveloping membrane containing a varied number of tiny vesicles. They are generally found associated with a short number of small free vesicles. Only one two groups of this kind are found per oocyte. This contrast with what has been observed previously in full-grown rat oocytes, where the groups are numerous and constituted by many units.Two components were described for the oocyte nucleus: nucleoli and chromosomes. Nucleoli are constituted by a tangled thread whose elemental component is a fine fibrous material of high electron density.At the age studied on this paper, primary oocytes are undergoing meiotic prophase, chromosomes have at this time the same components observed by different authors in primary spermatocytes. These are two thick ribbon-like threads helically twisted around a thinner medial filament. Each tripartite group is attached by one end to the nuclear membrane. It was actually seen tripartite groups incompletely organized; the images recorded of such groups suggest that the medial filament is the first to appear in the nucleoplasm. The possible significance of these filaments in respect to the meiotic phase called chromosome pairing is discussed.  相似文献

On the basis of light, autoradiographic (uridine-3H incorporation) and electron microscopic investigation changes of nuclear structures were examined during the oogenesis in Chrysopa perla L. — In early meiotic prophase the oocyte nuclei were found to contain a large body of extrachromosomal DNA. In certain cases the latter splits up into several DNA clumps giving rise to a few (4–7) primary nucleoli, 3–5 in diameter. The primary nucleoli consist of densely packed fibrils 50–100 Å thick. They contain no granular component and are inactive in RNA synthesis. — At the beginning of large growth the extrachromosomal DNA bodies disappear and numerous electron-dense clumps, 0,5–1 in diameter, appear in the nucleus. Instead of the primary nucleoli, the nucleus now contains a great number of ring nucleoli about 0,5–1 in diameter with a granular component (granules are 150 Å). The space between them is filled up with nucleolar strands running from the surface of the ring nucleoli. — At the stage ring nucleoli of uridine–3 H incorporation into the oocyte nucleus begins. — During later previtellogenesis and at the beginning of vitellogenesis the ring nucleoli disappear and the nucleus is filled with the network of nucleolar strands. Among them there are specific complexes. These consist of electron dense masses, of granular clusters (granules 500 Å in diameter) and large fibrillar electron light bodies. At this stage the nucleus takes the most active part in RNA synthesis. — The process of karyosphere capsule formation was studied by electron microscopy. The capsule was found to be of fibrillar nature; its structure is very peculiar and unlike any known membrane components of the cell. On the basis of cytochemical evidences the characteristics of the capsule are given. — The development of a powerful nucleolar apparatus based on the extrachromosomal DNA and a possible role of the synaptonemal complex and extrachromosomal DNA in formation of the karyosphere capsule is discussed.  相似文献

Electron micrographs of the relatively young spermatids of the Ascidian, Boltenia villosa, reveal that its chromosomes exist in the form of 120–180 Å fibers which are easily seen to be composed of two subunits of about 50–70 Å each. Fibers of the diameter of such subunits have never been seen as single entities without a mate. Subunits in pairs seen imbedded in areas of fused chromosomes only measure 30–40 Å, probably due to the loss of some protein. It is pointed out that the chromatin goes into the sperm as chromosomes in two-chromatid state, and that each chromatid probably contains only one Watson-Crick double helix.  相似文献

This paper studies the process and features of chromosome construction in mitotic prophase cells of Allium cepa. The results showed that a prominent reorganization of chromatin occurred during G2-early prophase. The 250–400 nm thick compact chromatin threads in G2 nuclei began to disorganize into about 30, 100 and 220 nm chromatin fibres which constituted the loosely organized chromosome outlines in early prophase before chromosome condensation. In middle prophase, chromosome condensation was characterized by the formation of many condensed regions (aggregates of chromatin), which increased in size (1–1.5 m) when prophase proceeded. Meanwhile, the chromatin threads that constituted and connected the condensed regions became increasingly thicker (120–250 nm). In late prophase adjacent condensed regions fused to form cylinder-shaped chromosomes. Based on these observations, we come to the conclusion that the construction of prophase chromosomes is a two-step process, that is, the reorganization and condensation of chromatin. In addition, we report the study of silver-stained, DNA- and histone-depleted prophase chromosomes, describe morphological features of the non-histone protein (NHP) residue in early, middle and late prophase chromosomes, and discuss the roles of NHPs in chromosome construction.  相似文献

以同步核内有丝分裂的多头绒泡菌(Physarum polycephalum)原质团为材料,在有丝分裂周期中连续取材,按常规方法制备超薄切片,在电镜下研究了染色体形态构建的整个过程。有丝分裂前期,首先是G_2期凝集的染色质块逐渐解集缩成为松散状,染色质在松散的同时逐渐改组成直径为80~150nm的松散染色线结构。接着是在松散的染色线上形成一些电子密度高的集缩区,随着集缩区的增多和扩展,染色线缩短变粗,最后形成直径300~350nm的染色体。上述两个过程各需30min左右。与上述过程同时发生的是,核仁由中央位置逐渐移向边缘,前期50min左右时在近核膜处呈团块状解体。染色体形态构建的整个过程约需1h,可分为染色质的松散改组和集缩两个连续的步骤,25~30nm染色质纤维是这一过程中能分辨的最细的形态单位。  相似文献

An ultrastructural study has been made of spermatogenesis in two species of primitive spiders having holocentric chromosomes (Dysdera crocata, XO and Segestria florentia X1X2O). Analysis of the meiotic prophase shows a scarcity or absence of typical leptotene to pachytene stages. Only in D. crocata have synaptonemal complex (SC) remnants been seen, and these occurred in nuclei with an extreme chromatin decondensation. In both species typical early prophase stages have been replaced by nuclei lacking SC and with their chromatin almost completely decondensed, constituting a long and well-defined diffuse stage. Only nucleoli and the condensed sex chromosomes can be identified. — In S. florentina paired non-homologous sex chromosomes lack a junction lamina and thus clearly differ from the sex chromosomes of more evolved spiders with an X1X2O male sex determination mechanism. In the same species, sex chromosomes can be recognized during metaphase I due to their special structural details, while in D. crocata the X chromosome is not distinguishable from the autosomes at this stage. — The diffuse stage and particularly the structural characteristics of the sex chromosomes during meiotic prophase are reviewed and discussed in relation to the meiotic process in other arachnid groups.  相似文献

Male meiosis in D. melanogaster cytologically follows the usual pattern, whereas in D. melanogaster and in D. virilis oocytes the chromosomes clump into a karyosphere at early meiotic prophase and remain so up to metaphase I.Male meiosis in D. virilis spermatocytes has an intermediate character: a part of the chromatin clumps together in a karyosphere at early prophase, whereas the other part of the chromatin remains diffuse all through prophase. At the end of prophase, the diffuse chromatin becomes integrated into the karyosphere before metaphase I. During the meiotic divisions the chromosomes have the same clumped aspect as those in Drosophila oocytes and thus differ strikingly from the dividing chromosomes in D. melanogaster spermatocytes.In D. virilis spermatocytes the nucleolus exhibits changes during the meiotic prophase that may be related to synthetical activities. The DNA specific staining with the fluorochrome DAPI reveals the existence of extrachromosomal DNA in the later prophase. Other striking differences in meiotic events between the two Drosophila species concern the centrioles and spermiogenesis.  相似文献

Chromatin elimination in the hypotrichous ciliate Stylonychia mytilus was studied by means of electron microscopy using a microspreading procedure. In the polytene chromosomes of the macronuclear anlagen three organization patterns are observed: Bands of various size composed of 300 Å chromatin fibers, large blocks of 300 Å nucleofilaments which probably represent the heterochromatic regions of the chromosome and axial 120 Å filaments. Those DNA sequences which become eliminated belong to the 300 Å fiber type. The eliminated chromatin occurs in the form of rings of variable size corresponding to a DNA content between 18 and 160 Kb while the axial 120 Å filaments appear to be preserved.  相似文献

Cockroaches are major pests, vectors of pathogenic bacteria and induce allergies. Current control methods use chemical pesticides, but they can be ineffective and costly and there are reports of resistance in the field; hence new control methods are needed. There are conflicting reports about the susceptibility of cockroaches to entomopathogenic nematodes (EPNs); so we investigated if EPNs could kill several diverse cockroach species, including the Madagascan hissing roach (Gromphadorhina portentosa), the Lobster roach (Nauphoeta cinerea) and Blaptica dubia. Female adult cockroaches were exposed to either commercial products containing Steinernema kraussei or a combination of Heterorhabditis spp. and Steinernema spp. at 50 and 150 nematodes per cm2 for 21 days. We also monitored feeding and the numbers of infective juveniles that were produced from each cockroach corpse. We found that S. kraussei were harmless to all cockroach species (at both doses) but when exposed to a mixture of Heterorhabditis spp. and Steinernema spp. B. dubia died after 6 days and its feeding was strongly inhibited. We also found that the mixture of Heterorhabditis spp. and Steinernema spp. could proliferate in the cadavers of B. dubia whilst S. kraussei could only reproduce in G. portentosa and B. dubia but not N. cinerea. In conclusion, S. kraussei was harmless to all three cockroach species but B. dubia was killed when exposed to Heterorhabditis spp. and Steinernema spp., highlighting the differences in the host range of EPNs.  相似文献

Summary The ammoniacal silver reaction was used for localization of histones in meristematic root tip cells of Allium cepa. Electron microscopic observations showed that yellow or brown colour of interphase and prophase nuclei and brown nucleolar colour produced in the reaction coincides with the appearance of silver grains, about 400 Å in diameter, in the interphase and prophase chromatin and nucleoli. This together with the complete absence of staining reaction and silver grains in the cytoplasm could mean quite a specific reaction with histones and might suggest also that in these cells the site of histone synthesis is in the nucleolus.  相似文献

Summary The shape, relationships, relative DNA content, and nucleolar activity of the short arm of acrocentric bivalents were studied in human oocytes from early diplotene to dictyotene. At the beginning of diplotene, the short arms of the previously paired chromosomes were again separated and displayed the same morphological features as in mitotic prophase chromosomes. They were connected only with the nucleolus. In situ hybridization and silver staining showed that the nucleolar organizer regions (NORs) were located in the peripheral region of the nucleolus. Tritiated-uridine incorporation was active. At birth, the relationships of the acrocentric short arms showed increasing complexity. The chromosomes ended in nucleolus-associated chromatin blocks of irregular shape, containing large quantities of DNA as demonstrated by intense binding of3H-actinomycin D. The number of chromosomes converging on these chromatin blocks exceeded the number of acrocentrics, suggesting that heterochromatic regions of other chromosomes were associated with the short arm of acrocentrics. In the electron microscope, the NORs were represented by fibrillar centers located on the periphery of the nucleolus and consistently connected with the blocks of dense chromatin. These relationships remained unchanged in the primordial oocyte in the adult ovary. Persistence of3H-uridine uptake showed that the oocyte was not at a resting stage. The possible cytogenetic consequences of these observations are discussed.  相似文献

The chromatin organization in developing germ cells of Drosophila hydei males was studied with the highly sensitive DNA stain DAPI (4, 6-diamidino-2-phenylindole dichloride). The prophase of meiosis I is characterized by decondensed chromosomes and only late during this stage do they condense rapidly. The sex chromosomes show allocycly. During postmeiotic development the final condensation of chromatin is preceded by a cycle of condensation and subsequent decondensation. Meiotic chromosomes were studied in more detail after orcein staining. Pairing sites of the sex chromosomes could be localized in the distal end of the heterochromatic arm of the X chromosome and distally in both arms of the Y chromosome. The various heterochromatic parts of the genome condense differentially in meiosis. Chromatin reorganization was studied cytochemically with antibodies raised against histones H1 and H2A of D. melanogaster. The core histone H2A is present in spermatid nuclei until the late elongation stage. However, histone H1 is not found in the chromatin later than the early primary spermatocyte stage. Thus, chromatin reorganization during spermatogenesis in D. hydei is complex. The process is discussed with regard to possible functions.  相似文献

The expansion of the worldwide pet trade has increased the invasion risks from not only exotic pet animals, but also live feeder organisms for predatory pet animals. Here, we report the first field record of a tropical live feeder cockroach, Blaptica dubia (Serville, 1838), in the central part of Japan. This widely traded feeder insect may be able to colonize temperate zones because they were estimated to survive through two winters in Japan at temperatures close to the upper limits of their critical thermal minimum. Further field observations are needed to understand the behavior of this species in the wild and predict the risk of invasion in temperate zones.  相似文献

Summary The ependymal cells bordering the median eminence to the third ventricle are characterised by many microvillus-like projections and bulbous cell processes of the luminal plasma membrane. The latter contain many vesicles 500–1,000 Å in diameter. Cilia with 9+2 fibrillar pattern are seen occasionally. Adhesive devices in the from of zonula adhaerens and zonula occludens are found in the apical part of the intercellular junction. Unmyelinated nerve fibres with a mean diameter of 1 and containing many electron dense granules of 830–1,330 Å are often seen between the ependymal cells.Two types of glial cells are found in the median eminence. One is characterised by a nucleus with dense blods of chromatin and dense cytoplasm, and it is associated chiefly with the nerve fibres in the region of the hypothalamo-hypophysial tract. The other type of glial cell is characterised by fine, uniformly distributed chromatin in the nucleus and a relatively pale cytoplasm and branched processes which terminate perivascularly in the base of the median eminence.Myelinated nerve fibres are seen only in the region of the hypothalamo-hypophysial tract. Only a part of them contain electron dense granules 1,330–2,330 Å in diameter.Three types of unmyelinated nerve fibres can be distinguished in the median eminence according to the size of the electron dense granules they contain: 1. Nerve fibres containing granules 1,330–2,330 Å in diameter. They are seen primarily in the hypothalamo-hypophysial tract, but also in the zona externa; 2. those containing granules with a mean diameter of 1,330 Å; and 3. those containing granules with a mean diameter of 1,000 Å. The last two types are both encountered in the hypothalamo-hypophysial tract, the zona externa and the perivascular region of the base of the median eminence. Under high magnification, the membrane of the granules show evidence of a trilaminar structure and the content of the granules with a low electron density appeares to consist of small microvesicles or globular components. Besides granules, these nerve fibres contain vesicles mostly 420 Å in diameter whose relative number increases towards the perivascular nerve endings. 53 per cent of the inclusions in the hypothalamo-hypophysial tract are granules and 47 per cent vesicles, while the corresponding percentages for the zona externa are 40 and 60 and for the perivascular nerve endings 20 and 80.The mean width of the pericapillary space is 1 , but it varies greatly. It containes many collagen fibrils and fibroblasts. The capillary endothelium is frequently fenestrated and contains many vesicles of various sizes.Two types of granules-containing cells are found in the pars tuberalis depending on the size of the electron dense granules: 1. cells containing granules with a mean diameter of 1,330 Å: and 2. cells containing granules with a mean diameter of 2,000 Å. 【背景】杜比亚蟑螂(Blaptica dubia)可用于活体饲料、化妆品和医药保健品的生产,其肠道菌的研究对杜比亚蟑螂的饲养和肠道菌资源的开发与利用都十分重要。【目的】揭示杜比亚蟑螂肠道可培养菌的种类,筛选具有产消化酶功能的菌株,为理解肠道菌对宿主的影响机理及功能菌株的利用提供科学依据和研究材料。【方法】采用体外培养法获得杜比亚蟑螂肠道菌,结合形态学和分子生物学方法进行鉴定;用水解圈法分别筛选产纤维素酶、蛋白酶、脂肪酶和淀粉酶菌株。【结果】在杜比亚蟑螂肠道中共分离出4属7种细菌,其中芽孢杆菌属(Bacillus)2种,沙雷氏菌属(Serratia)和柠檬酸杆菌属(Citrobacter)各2种,肠球菌属(Enterococcus)1种。从获得的20个菌株中筛选出10个具有产消化酶功能的菌株。其中,芽孢杆菌属的菌株D6、D12和D20具有产纤维素酶、蛋白酶、淀粉酶及脂肪酶4种消化酶的功能;沙雷氏菌属的菌株D3、D7、D9、D11和D15具有产纤维素酶、蛋白酶和脂肪酶3种消化酶的能力;柠檬酸杆菌属的菌株D5具有产纤维素酶的功能;肠球菌属的菌株D17具有产蛋白酶的能力。【结论】杜比亚蟑螂肠道多种细菌具有产消化酶帮助降解大分子营养物质的功能,可通过协助食物消化影响宿主健康。菌株D12、D7和D11分别具有最强产纤维素酶、蛋白酶和脂肪酶的能力,是可进一步开发利用的肠道功能菌株资源。  相似文献

Young spermatids of the prosobranch mollusc Goniobasis have spherical nuclei with diffuse centers and condensation of the chromatin only at their peripheries. At later stages, this condensation becomes uniform for the entire nucleus, and has a lamellar appearance in sectioned material. In specimens prepared using freeze-etch techniques, these lamellae are resolvable into chromatin strands, each of which has a helical substructure, with a period of ca. 100 Å and a pitch angle of 35°. The strands are ca. 130–140 Å in diameter (comparable to measurements made on micrographs of sectioned material). Most of the strands are oriented in the long axis of the elongating nucleus, but a few are oblique, suggesting that they might serve some sort of binding function. In the mature spermatozoon the chromatin is very compact and dense, and all evidence of the lamellar or stranded appearance is absent, in both sectioned and freeze-etch preparations.  相似文献
