Abscisic acid (ABA) concentration is a quantitatively inherited trait which plays a pivotal role in the response of plants to drought stress. A recent study identified 17 quantitative trait loci (QTLs) controlling bulk-leaf ABA concentration (L-ABA) in a maize (Zea mays L.) population of 80 F4 random families tested for two years under droughted field conditions. Sixteen of the QTL regions influencing L-ABA also harboured QTLs for one or more of the following traits: stomatal conductance, a drought sensitivity index, leaf temperature, leaf relative water content, anthesis-silking interval, and grain yield. The analysis of the effects of each QTL region on the investigated traits indicated that L-ABA mainly represented an indicator of the level of drought stress experienced by the plant at the time of sampling because an increase in L-ABA was most commonly associated with a decrease in both stomatal conductance and grain yield as well as an increase in leaf temperature. Opposite results were observed at one QTL region on chromosome 7 near the RFLP locus asg8. A previous study conducted on a maize (Zea mays L.) mapping population derived from Os420 × IABO78 identified a quantitative trait locus (QTL) for leaf-abscisic acid concentration (L-ABA) on chromosome 2 (bin 2.04). In order to validate this QTL, we analyzed with RFLP markers 16 F4 lines obtained by divergent selection for L-ABA from the same source. Three RFLPs mapping near bin 2.04 showed skewed allelic frequencies; the L-ABA increasing allele (+) was more frequent within the eight lines selected for high L-ABA, while the decreasing allele (–) was more frequent within the eight lines selected for low L-ABA. To characterize more accurately the direct and associated effects of this QTL, near-isogenic lines were developed by molecular marker-assisted back-crossing; four backcross-derived lines were homozygous (+/+) at the QTL and four were (–/–). A pair of near-isogenic hybrids (+/+) and (–/–) at the QTL were also produced. These materials were field tested under water-stressed and well-watered conditions. Across water regimes, the four (+/+) lines averaged a significantly higher mean value than the four (–/–) lines for L-ABA (494 vs. 396 ng ABA g–1 DW) and a significantly lower mean value for relative water content (90.6 vs. 92.0%). The (+/+) hybrid exceeded the (–/–) for L-ABA (476 vs. 325 ng g–1 DW) and was less affected by root lodging (44.6 vs. 66.1%). Our results validate the presence of a major QTL for L-ABA on bin 2.04 and indicate that the QTL also affects root traits and relative water content.  相似文献

QTL analysis of leaf morphology in tetraploid Gossypium (cotton)   总被引:13,自引:0,他引:13  
Molecular markers were used to map and characterize quantitative trait loci (QTLs) determining cotton leaf morphology and other traits, in 180 F2 plants from an interspecific cross between a Gossypium hirsutum genotype carrying four morphological mutants, and a wild-type Gossypium barbadense. The prominent effects of a single region of chromosome 15, presumably the classical ”Okra-leaf” locus, were modified by QTLs on several other chromosomes affecting leaf size and shape. For most traits, each parent contained some alleles with positive effects and others with negative effects, suggesting a large potential for adapting leaf size and shape to the needs of particular production regimes. Twenty one QTLs/loci were found for the morphological traits at LOD≥3.0 and P≤0.001, among which 14 (63.6%) mapped to D-subgenome chromosomes. Forty one more possible QTLs/loci were suggested with 2.0≤LOD<3.0 and 0.001<P≤0.01. Among all of the 62 possible QTLs (found at LOD≥2.0 and P≤0.01) for the 14 morphological traits in this study, 38 (61.3%) mapped to D-subgenome chromosomes. This reinforces the findings of several other studies in suggesting that the D-subgenome of tetraploid cotton has been subject to a relatively greater rate of evolution than the A-subgenome, subsequent to polyploid formation.  Quantitative trait loci (QTLs) contributing to salt tolerance during the vegetative stage in tomato were investigated using an interspecific backcross between a salt-sensitive Lycopersicon esculentum breeding line (NC84173, maternal and recurrent parent) and a salt-tolerant Lycopersicon pimpinellifolium accession (LA722). One hundred and nineteen BC1 individuals were genotyped for 151 RFLP markers and a linkage map was constructed. The parental lines and 119 BC1S1 families (self-pollinated progeny of the BC1 individuals) were evaluated for salt tolerance in aerated saline-solution cultures with the salt concentration gradually raised to 700 mM NaCl+70 mM CaCl2 (equivalent to an electrical conductivity of approximately 64 dS/m and a water potential of approximately −35.2 bars). The two parental lines were distinctly different in salt tolerance: 80% of the LA722 plants versus 25% of the NC84173 plants survived for at least 2 weeks after the final salt concentration was reached. The BC1S1 population exhibited a continuous variation, typical of quantitative traits, with the survival rate of the BC1S1 families ranging from 9% to 94% with a mean of 51%. Two QTL mapping techniques, interval mapping (using MAPMAKER/QTL) and single-marker analysis (using QGENE), were used to identify QTLs. The results of both methods were similar and five QTLs were identified on chromosomes 1 (two QTLs), 3, 5 and 9. Each QTL accounted for between 5.7% and 17.7%, with the combined effects (of all five QTLs) exceeding 46%, of the total phenotypic variation. All QTLs had the positive QTL alleles from the salt-tolerant parent. Across QTLs, the effects were mainly additive in nature. Digenic epistatic interactions were evident among several QTL-linked and QTL-unlinked markers. The overall results indicate that tomato salt tolerance during the vegetative stage could be improved by marker-assisted selection using interspecific variation. Genetic factors controlling quantitative inheritance of grain yield and its components have not previously been investigated by using replicated lines of an elite maize (Zea mays L.) population. The present study was conducted to identify quantitative trait loci (QTLs) associated with grain yield and grain-yield components by using restriction fragment length polymorphism (RFLP) markers. A population of 150 random F23 lines was derived from the single cross of inbreds Mo17 and H99, which are considered to belong to the Lancaster heterotic group. Trait values were measured in a replicated trial near Ames, Iowa, in 1989. QTLs were located on a linkage map constructed with one morphological and 103 RFLP loci. QTLs were found for grain yield and all yield components. Partial dominance to overdominance was the primary mode of gene action. Only one QTL, accounting for 35% of the phenotypic variation, was identified for grain yield. Two to six QTLs were identified for the other traits. Several regions with pleiotropic or linked effects on several of the yield components were detected.  相似文献

One hundred to 120 maize recombinant inbred lines at the mature fourth leaf stage derived from F-2 and Io parental lines were grown in a glasshouse and were deprived of water for 9 days in order to detect pertinent markers of the physiological response to water stress which may be used for breeding. Carbohydrate metabolism QTLs were compared to photosynthesis gas exchange QTLs. The locations of these QTLs were further compared with those of morphological trait QTLs when water availability varied. The traits ranged from three enzyme activities (invertase, sucrose-P synthase, ADP glucose pyrophosphorylase) and hexose, sucrose, starch content to CO2 uptake and stomatal conductance, water status, leaf size, root/shoot ratio, and ABA (leaf, root and xylem sap). Four main results were obtained (1) only 14 % of QTLs were common to both drought and watered treatments, confirming the existence of stress specific chromosome regions, (2) the QTLs tended to form clusters, frequently consisting of QTLs from different classes (growth, photosynthesis, water status, carbohydrate metabolism and ABA), (3) carbohydrate metabolism trait QTLs were more frequently co-located with growth trait QTLs than photosynthesis related ones, especially in control conditions, (4) one co-location was observed between the three enzyme activities implied in sucrose and starch metabolism and a corresponding structural gene, which can be considered as a candidate gene for explaining part of the variability of each enzymatic trait (invertase, sucrose-P synthase, ADPglucose pyrophosphorylase). It is concluded that, carbohydrate metabolism provides valuable traits for understanding and improving maize responses to water stress.  相似文献

 Most cultivars of tomato (Lycopersicon esculentum) are sensitive to salinity during seed germination and at later stages. Genetic resources for salt tolerance have been identified within the related wild species of tomato. The purpose of the present study was to identify quantitative trait loci (QTLs) for salt tolerance during germination in an inbred backcross (BC1S1) population of an interspecific cross between a salt-sensitive tomato breeding line (NC84173, maternal and recurrent parent) and a salt-tolerant Lycopersicon pimpinellifolium accession (LA722). Onehundred and nineteen BC1 individuals were genotyped for 151 restriction fragment length polymorphism (RFLP) markers and a genetic linkage map was constructed. The parental lines and 119 BC1S1 families (self-pollinated progeny of 119 BC1 individuals) were evaluated for germination at an intermediate salt-stress level (150 mM NaCl+15 mM CaCl2, water potential approximately −850 kPa). Germination was scored visually as radicle protrusion at 8-h intervals for 28 consecutive days. Germination response was analyzed by survival analysis and the time to 25, 50, and 75% germination was determined. In addition, a germination index (GI) was calculated as the weighted mean of the time from imbibition to germination for each family/line. Interval mapping, single-marker analysis and distributional extreme analysis, were used to identify QTLs and the results of all three mapping methods were generally similar. Seven chromosomal locations with significant effects on salt tolerance were identified. The L. pimpinellifolium accession had favorable QTL alleles at six locations. The percentage of phenotypic variation explained (PVE) by individual QTLs ranged from 6.5 to 15.6%. Multilocus analysis indicated that the cumulative action of all significant QTLs accounted for 44.5% of the total phenotypic variance. A total of 12 pairwise epistatic interactions were identified, including four between QTL-linked and QTL-unlinked regions and eight between QTL-unlinked regions. Transgressive phenotypes were observed in the direction of salt sensitivity. The graphical genotyping indicated a high correspondence between the phenotypes of the extreme families and their QTL genotypes. The results indicate that tomato salt tolerance during germination can be improved by marker-assisted selection using interspecific variation. Plants accumulate abscisic acid (ABA) under droughted conditions. Genetic variation in the accumulation of ABA in detached and partially dehydrated leaves of rice has previously been reported, and this was found to be associated with variation in leaf size (smaller leaves made more ABA). Correlation analysis failed to distinguish clearly between a causal relationship between the two traits and close genetic linkage between loci controlling the traits. Here we present a detailed genetic analysis of ABA accumulation in detached and partially dehydrated rice leaves, using a population of F2 plants generated from the lowland × upland cross IR20 (high-ABA) × 63-83 (low-ABA) which was mapped with RFLP and AFLP markers. Several highly significant quantitative trait loci (QTLs) for ABA accumulation and leaf weight were identified. Only one of the minor QTLs for ABA accumulation (accounting for only 4% of the phenotypic variance) was coincident with any QTLs for leaf size such that the high-ABA allele was associated with smaller leaves. This analysis, therefore, showed that the association previously found between ABA accumulation and leaf size was probably largely due to genetic linkage and not to a direct effect of leaf size on ABA accumulation or vice versa. Because of the importance of ABA accumulation in regulating responses of plants to drought stress and the effects of plant size on the rate of development of stress, QTLs for drought-induced ABA accumulation, leaf size and tiller number were compared between rice and wheat. In particular, a possible location in rice was sought for a homoeologue of the major wheat vernalization responsive gene, Vrn1, as this gene is also associated with major effects on leaf size, tiller number and ABA accumulation in wheat. The likelihood of homoeologous loci regulating ABA accumulation, leaf size and tiller number in the two crops is discussed.  相似文献

A population of 117 doubled haploid (DH) lines derived from the cross of Zhaiyeqing 8 (indica)× Jingxi 17 (japonica) was employed to map quantitative trait loci (QTL) underlying four physiological traits related to chlorophyll contents of the flag leaf. There were significantly positive correlations among chlorophyll a, chlorophyll b and chlorophyll a + b content. Chlorophyll a/b ratio was significantly negatively correlated with chlorophyll b content. These four traits were normally distributed with transgressive segregation, suggesting that they were controlled by multiple minor genes. A total of 11 QTLs were detected for the four traits and they lay on six chromosomes. Each of them explained 9.2%–19.6% of the phenotypic variations, respectively. Of these, two QTLs controlling chlorophyll a content were mapped on chromosomes 2 and 5; four QTLs underlying chlorophyll b content were mapped on chromosomes 2, 3, 5 and 9; three QTLs underlying chlorophyll a + b amount were mapped on chromosomes 3, 5 and 9; two QTLs underlying chlorophyll a/b ratio were mapped on chromosomes 6 and 11. A strong photosynthetic performance and rapid leaf development, are important indicators of vigorous early growth. The aim of this study was to (1) evaluate the tropical maize (Zea mays L.) inbred lines CML444 and SC-Malawi for their photosynthetic performance at different growth stages and (2) assess quantitative trait loci (QTL) of photosynthesis-related traits in their 236 recombinant inbred lines at the heterotrophic growth stage. CML444 had a higher leaf chlorophyll (SPAD) content than SC-Malawi. Ten QTLs were found for the quantum efficiency of photosystem II (ΦPSII; four), SPAD (three) and the specific leaf area (SLA; three). The relevance of seedling QTLs for ΦPSII, SPAD and SLA for yield formation is emphasized by seven collocations (bins 5.01, 7.03, 8.05) with QTLs for kernel number and grain yield under field conditions. QTLs for SPAD at the V2 and at the reproductive stage did not collocate, indicating differences in the genetic control of SPAD at different growth stages. Knowing which loci affect SLA, SPAD and ΦPSII simultaneously and which do not will help to optimize light harvest by the canopy.  相似文献

 An amplified fragment length polymorphism (AFLP) map covering 965 cM was constructed using 94 recombinant inbred lines of a cross between the spring barley varieties Prisma and Apex. This map was employed to identify quantitative trait loci (QTLs) controlling plant height, yield and yield-determining physiological characters using an approximate multiple-QTL model, the MQM method. The seven physiological traits were parameters used in a process-based crop-growth model that predicts barley biomass production as affected by daily temperature and radiation. The traits were measured in experiments conducted over 2 years. Except for the relative growth rate of leaf area, all traits examined had at least one QTL in each year. QTLs and their effects were found to vary with developmental stages for one trait, the fraction of shoot biomass partitioned to leaves, that was measured at several stages. Most of the traits were associated, though to different extents, with the denso dwarfing gene (the height-reducing allele in Prisma) located on the long arm of chromosome 3. Some of the QTLs were mapped to similar positions in both years. The results in relation to effects of the dwarfing gene, the physiological basis for QTL×environment interaction, and the relative importance of the parameter traits with respect to yield, are discussed. Construction of an RFLP linkage map for cultivated sunflower   总被引:5,自引:0,他引:5  
 An RFLP linkage map was constructed for cultivated sunflower Helianthus annuus L., based on 271 loci detected by 232 cDNA probes. Ninety-three F2 plants of a cross between inbred lines RHA 271 and HA 234 were used as the mapping population. These genetic markers plus a fertility restoration gene, Rf 1, defined 20 linkage groups, covering 1164 cM of the sunflower genome. Of the 71 loci 202 had codominant genotypic segregation, with the rest showing dominant segregation. Thirty-two of the 232 probes gave multiple locus segregation. There were 39 clusters of tightly linked markers with 0 cM distance among loci. This map has an average marker-to-marker distance of 4.6 cM, with 11 markerless regions exceeding 20 cM.  We constructed a genetic map of a cross between the Swiss winter wheat (Triticum aestivum L.) variety Forno and the Swiss winter spelt (Triticum spelta L.) variety Oberkulmer. For the linkage analysis,176 polymorphic RFLP probes and nine microsatellites were tested on 204 F5 recombinant inbred lines (RILs) of Forno×Oberkulmer revealing 242 segregating marker loci. Thirty five percent of these loci showed significant (P>0.05) deviation from a 1 : 1 segregation, and the percentage of Forno alleles ranged from 21% to 83% for individual marker loci. Linkage analysis was performed with the program MAPMAKER using the Haldane mapping function. Using a LOD threshold of 10, we obtained 37 linkage groups. After finding the best order of marker loci within linkage groups by multi-point analysis we assembled the linkage groups into 23 larger units by lowering the LOD threshold. All except one of the 23 new linkage groups could be assigned to physical chromosomes or chromosome arms according to hybridisation patterns of nulli-tetrasomic lines of Chinese Spring and published wheat maps. This resulted in a genetic map comprising 230 marker loci and spanning 2469 cM. Since the analysed population is segregating for a wide range of agronomically important traits, this genetic map is an ideal basis for the identification of quantitative trait loci (QTLs) for these traits. A segregating F2 population of Arabidopsis thaliana derived from a cross between the late-flowering ecotype Hannover/Münden (HM) and the early-flowering ecotype Wassilewskija (WS) was analyzed for flowering time and other morphological traits. Two unlinked quantitative trait loci (QTLs) affecting days to first flower (DFF-a and DFF-b) mapped to chromosome 5. QTLs which affect node number (NN), leaf length at flowering (LLF), and leaf length at 35 days (LL35) also mapped to chromosome 5; LLF-a, LL35-a, NN-a map to the same region of chromosome 5 as DFF-a; LLF-b and LL35-bmap to the same region of chromosome 5 as DFF-b. Another QTL affecting leaf length at flowering (LLF-c) maps to chromosome 3. The proximity of DFF-a, LLF-a, LL35-a and NN-a, as well as the similarity in gene action among these QTLs (additivity), suggest that they may be pleiotropic consequences of a single gene at this locus. Similarly, LL35-b and LLF-b map near each other and both display recessive gene action, again suggesting the possibility of pleiotropy. DFF-b, which also maps near LL35-b and LLF-b, displays largely additive gene action (although recessive gene action could not be ruled out). This suggests that DFF-b may represent a different gene from LL35-b and/or LLF-b. DFF-a maps near two previously identified mutants: co (which also affects flowering time and displays gene action consistent with additivity) and flc. Similar map locations and gene actions of QTLs affecting the correlated traits DFF, LLF, LL35 and NN suggest that these genomic regions harbor naturally occurring allelic variants involved in the general transition of the plant from vegetative to reproductive growth.  相似文献

To capture diverse alleles at a set of loci associated with disease resistance in maize, heterogeneous inbred family (HIF) analysis was applied for targeted QTL mapping and near-isogenic line (NIL) development. Tropical maize lines CML52 and DK888 were chosen as donors of alleles based on their known resistance to multiple diseases. Chromosomal regions (“bins”; n = 39) associated with multiple disease resistance (MDR) were targeted based on a consensus map of disease QTLs in maize. We generated HIFs segregating for the targeted loci but isogenic at ~97% of the genome. To test the hypothesis that CML52 and DK888 alleles at MDR hotspots condition broad-spectrum resistance, HIFs and derived NILs were tested for resistance to northern leaf blight (NLB), southern leaf blight (SLB), gray leaf spot (GLS), anthracnose leaf blight (ALB), anthracnose stalk rot (ASR), common rust, common smut, and Stewart’s wilt. Four NLB QTLs, two ASR QTLs, and one Stewart’s wilt QTL were identified. In parallel, a population of 196 recombinant inbred lines (RILs) derived from B73 × CML52 was evaluated for resistance to NLB, GLS, SLB, and ASR. The QTLs mapped (four for NLB, five for SLB, two for GLS, and two for ASR) mostly corresponded to those found using the NILs. Combining HIF- and RIL-based analyses, we discovered two disease QTLs at which CML52 alleles were favorable for more than one disease. A QTL in bin 1.06–1.07 conferred resistance to NLB and Stewart’s wilt, and a QTL in 6.05 conferred resistance to NLB and ASR.  相似文献

The natural variation of many traits is controlled by multiple genes, individually referred to as quantitative trait loci (QTL), that interact with the environment to determine the ultimate phenotype of any individual. A QTL has yet to be described molecularly, in part because strategies to systematically identify them are underdeveloped and because the subtle nature of QTLs prevents the application of standard methods of gene identification. Therefore, it will be necessary to develop a systematic approach(es) for the identification of QTLs based upon the numerous positional data now being accumulated through molecular marker analyses. We have characterized a QTL by the following three-step approach: (1) identification of a QTL in complex populations, (2) isolation and genetic mapping of this QTL in near-isogenic lines, and (3) identification of a candidate gene using map position and physiological criteria. Using this approach we have characterized a plant height QTL in maize that maps to chromosome 9 near the centromere. Both map position and physiological criteria suggest the gibberillin biosynthesis gene dwarf3 as a candidate gene for this QTL.  相似文献

 The partial resistance to leaf rust in barley is a quantitative resistance that is not based on hypersensitivity. To map the quantitative trait loci (QTLs) for partial resistance to leaf rust, we obtained 103 recombinant inbred lines (RILs) by single-seed descent from a cross between the susceptible parent L94 and the partially resistant parent Vada. These RILs were evaluated at the seedling and adult plant stages in the greenhouse for the latent period (LP) of the rust fungus, and in the field for the level of infection, measured as area under the disease progress curve (AUDPC). A dense genetic map based on 561 AFLP markers had been generated previously for this set of RILs. QTLs for partial resistance to leaf rust were mapped using the “Multiple Interval Mapping” method with the putative QTL markers as cofactors. Six QTLs for partial resistance were identified in this population. Three QTLs, Rphq1, Rphq2 and Rphq3, were effective at the seedling stage and contributed approximately 55% to the phenotypic variance. Five QTLs, Rph2, Rphq3, Rphq4, Rphq5, and/or Rphq6 contributed approximtely. 60% of the phenotypic variance and were effective at the adult plant stage. Therefore, only the QTLs Rphq2 and Rhpq3 were not plant-stage dependent. The identified QTLs showed mainly additive effects and only one significant interaction was detected, i.e. between Rphq1 and Rphq2. The map positions of these QTLs did not coincide with those of the race-specific resistance genes, suggesting that genes for partial resistance and genes for hypersensitive resistance represent entirely different gene families. Also, three QTLs for days to heading, of which two were also involved in plant height, were identified in the present recombinant inbred population. These QTLs had been mapped previously on the same positions in different populations. The perspectives of these results for breeding for durable resistance to leaf rust are discussed. The loci explaining the variability of quantitative traits related to starch content and composition (amylose, amylopectin and water soluble fraction) were searched for in maize kernels. Multifactorial genetic methods were used to detect and locate QTLs (quantitative trait loci) on a genetic map consisting mainly of RFLP markers for genes with known function. The genetic material was recombinant inbred lines originating from parents differing in starch structure (dent vs. flint). Kernels were harvested from field grown plants for two successive years and under two pollination systems. Main effect and epistasis QTLs were detected using two methods, composite interval mapping (MQTL) and ANOVA. Despite large year-to-year differences, physiologically meaningful co-locations were observed between trait QTLs. Moreover, the number of expressed sequences on our map allowed the search for co-locations between QTLs and genes involved in carbohydrate metabolism. The main co-location was between an amylose QTL and Shrunken 2 (SH2) locus, on chromosome 3 (SH2 encoding for the large subunit of ADPglucose pyrophosphorylase). The importance of this locus as a candidate gene for a starch QTL is in agreement with previous studies based either on QTL co-locations or on revertant analysis. Other co-locations were observed between amylose and amylopectin QTLs and the two loci of IVR1 invertase genes on chromosomes 2 and 10. Further comparison with previously detected QTLs for carbohydrate metabolism in maize leaves showed consistent co-location in map regions devoid of candidate genes, such as near chromosome 1S telomere. The possible contribution of regulatory genes in this region is discussed.  相似文献

 The amplified fragment length polymorphism (AFLP) technique combined with selective genotyping was used to map quantitative trait loci (QTLs) associated with tolerance for phosphorus (P) deficiency in rice. P deficiency tolerant cultivar IR20 was crossed to IR55178-3B-9-3 (sensitive to P-deficiency) and 285 recombinant inbred lines (RILs) were produced by single-seed descent. The RILs were phenotyped for the trait by growing them in P-sufficient (10.0 mg/l) and P-deficient (0.5 mg/l) nutrient solution and determining their relative tillering ability at 28 days after seeding, and relative shoot dry weight and relative root dry weight at 42 days after seeding. Forty two of each of the extreme RILs (sensitive and tolerant) and the parents were subjected to AFLP analysis. A map consisting of 217 AFLP markers was constructed. Its length was 1371.8 cM with an average interval size of 7.62 cM. To assign linkage groups to chromosomes, 30 AFLP and 26 RFLP markers distributed over the 12 chromosomes were employed as anchor markers. Based on the constructed map, a major QTL for P-deficiency tolerance, designated PHO, was located on chromosome 12 and confirmed by RFLP markers RG9 and RG241 on the same chromosome.
