Potassium-Ammonium Uptake Interactions in Tobacco Seedlings   总被引:6,自引:0,他引:6  
Short-term (< 12 h) uptake experiments were conducted with6–7-week-old tobacco (Nicotiana tabacum L. cv. Ky 14)seedlings to determine absorption interactions between K+ andNH4+. At equal solution concentrations (0.5 mol m–3) netK+ uptake was inhibited 30–35% by NH4+ and NH4+ uptakewas decreased 9–24%. Removal of NH4+ resulted in completerecovery in K+ uptake rate, but NH4+ uptake rate did not recoverwhen K+ was removed. In both cases, inhibition of the uptakerate of one cation saturated as the concentration of the othercation was increased up to 0.5 mol m–3. The relative effectof K+-NH4+ interactions was not altered when Cl- was replacedwith SO42–, but the magnitudes of the uptake rates wereless in the absence of Cl-. The Vmax for NH4+ uptake was reducedfrom 128 to 105 µmol g–1 dry wt. h–1 in thepresence of 0.5 mol m–3 K+ and the Km for NH4+ doubledfrom 12 to 27 mmol m–3 in the presence of K+. The resultsof these K+-NH4+ experiments are interpreted as mixed-noncompetitiveinteractions. However, an enhanced efflux of K+ coupled to NH4+influx via an antiporter cannot be ruled out as contributingto the decrease in net K+ uptake. A comparison of the activity and properties of the enzyme phosphoenolpyruvatecarboxylase (PEPC) was made for plants of Sedum telephium L.grown under low (70 µmol m–2 s–1) or high(500µmol m–2 s–1) PPFD and subjected to varyingdegrees of water stress. Under well-watered conditions onlyplants grown under high PPFD accumulated titratable acidityovernight and the extractable activity of PEPC was almost 2-foldhigher in these plants than in plants grown under low PPFD.Increasing drought stress resulted in a substantial increasein the activity of PEPC extracted both during the light anddark periods and a decrease in the sensitivity to inhibitionby malic acid. The magnitude of these changes was determinedby the severity and duration of drought and by light intensity.A comparison of the kinetic properties of PEPC from severelydroughted plants revealed that plants droughted under high PPFDhad a lower Km for PEP than plants under low PPFD. Additionof 2·0 mol m–3 malate resulted in an increase inthe Km for PEP, with plants draughted under low PPFD havinga significantly higher Km in the presence of malic acid comparedto those under high PPFD. Response to the activator glc-6-P,which lowered the Km for PEP, also varied between plants grownunder the two light regimes. Under well-watered conditions PEPCextracted from plants under high PPFD was more sensitive toactivation by glc-6-P than those under low PPFD. After the severedrought treatment, however, the Km for PEP in the presence ofglc-6-P was similar for enzyme extracted from plants grown underboth light regimes. Soluble sugars and starch were depletedovernight and were both possible sources of substrate for PEPC.With increasing drought, however, the depletion of starch relativeto soluble sugars increased under both light regimes. The propertiesof PEPC and the characteristics of carbohydrate accumulation/depletionare discussed in relation to the regulation of CAM in S. telephiumgrown under different light and watering regimes. In short-term experiments sulphate influx of excised tobaccoroots {Nicotiana tabacum L. var. 'Samsun') followed monophasicMichaelis-Menten kinetics with an approximate Km of 12 ±4 µM and vmax of 657 ± 211 nmol g–1 FW h–1.An inhibition of sulphate influx, xylem loading and exudationof more than 70% was achieved with 01 mM GSH within 1 h. Cysteinewas two orders of magnitude more effective as an inhibitor thanGSH. An inhibition of more than 75% was already obtained with1.0µM cysteine. It may, therefore, be assumed that GSHis decomposed to yield cysteine concentrations that may inhibitsulphate influx, xylem loading and exudation. When BSO, a specificinhibitor of the initial step of GSH synthesis, was added, cysteine-mediatedinhibition on sulphate influx, xylem loading and exudation wasstrongly diminished. Apparently, GSH synthesis is required toobtain inhibition of these processes by cysteine. The physiologicalmechanisms that may cause the inhibition of sulphate influx,xylem loading and exudation by glutathione are discussed. In situ light measurements were used to obtain information oninherent and apparent optical properties. The average verticalattenuation coefficient Kd(ave) varied from 1.1 to 4.6 In unitsm–1 During three periods the variation in Kd(ave) correlatedwith changes in chlorophyll a concentration and specific attenuationcoefficients Ks, of 0.013, 0.014 and 0.022 m2 mg Chl a–1were calculated. Chlorophyll-specific diffuse absorption coefficients(A,) for these periods were 0.012. 0.013 and 0.017 m2 mg Chla–1 and only varied significantly from estimates of Ksin the period when scattering was intense. Absorption coefficientsa(zmid) and scattering coefficients b(zmid) calculated for themid-point of the euphotic zone ranged between 0.45 and 2.9 mand 3.5–52.0 m respectively. Chlorophyll-specific absorptioncoefficients Ka, of 0.005, 0.006 and 0.007 m2 mg Chl a–1and scattering coefficients Kb of 0.05. 0.09 and 0.191 m2 mgChl a–1 were measured during the three periods. The highKb value occurred when gas-vacuolate cyanobactena were dominant.Algal photosynthesis and light absorption were related throughthe maximum quantum yield m which varied between 0.019 and 0.11mol C Einstein–1 while average quantum yields a, variedbetween 0.006 and 0.024 with a mean of 0.013 mol C Einstein–1A comparison of changes in the mean irradiance of the mixedzone and chlorophyll concentration indicated that growth waslight limited below 0.04–0.05 Einsteins absorbed mg Chla–1 day–1.  相似文献

NADP malic enzyme (EC 1.1.1.40 [EC] ) from leaves of two C4 speciesof Cyperus (C. rotundus and C. brevifolius var leiolepis) exihibiteda low level of activity in an assay mixture that contained lowconcentrations of Cl. This low level of activity wasmarkedly enhanced by increases in the concentration of NaClup to 200 mM. Since the activity of NADP malic enzyme was inhibitedby Na2SO4 and stimulated by relatively high concentration ofTris-HCl (50–100 mM, pH 7–8), the activation ofthe enzyme by NaCl appears to be due to Cl. Variationsin the concentration of Mg2+ affected the KA (the concentrationof activator giving half-maximal activation) for Cl,which decreased from 500 mM to 80 mM with increasing concentrationsof Mg2+ from 0.5 mM to 7 mM. The Km for Mg2+ was decreased from7.7 mM to 1.3 mM with increases in the concentration of NaClfrom zero to 200 mM, although the increase of Vmax was not remarkable.NADP malic enzyme from Cyperus, being similar to that from otherC4 species, was able to utilize Mn2+. The Km for Mn2+ was 5mM, a value similar to that for Mg2+. The addition of 91 mMNaCl markedly decreased the Km for Mn2+ to 20 +M. NADP malicenzyme from Setaria glauca, which contains rather less Clthan other C4 species, was inactivated by concentrations ofNaCl above 20 mM, although slight activation of the enzyme wasobserved at low concentrations of NaCl at pH7.6. Barley (Hordeum vulgare L.) varieties differed in their raponseto [K+]0, in terms of their utilization efficiencies (UE = freshweight. concentration of [K+]1–1). At low [K+]0, Compana,an efficient-non-responder demonstrated superior utilizationof absorbed K+. On the other hand, at high [K+]0, Fergus (anefficient responder) and BT 334 (an inefficient responder) hadhigher UE values for K+ than Compana which performed poorlyat this [K+]0. Kinetic parameters for K+ activation of the enzyme pyruvatekinase from 12 barley varieties, representing a range of UEvalues, were determined. Varieties showed substantial differencesin their Vmax values (P<0·01). Compana, an efficientvariety, had the highest Vmax (31 µmol g–1 freshwt. h–1) which was about 50% higher than that of Mingo,an inefficient variety. By contrast, Km values for the enzymeswere not significantly different among varieties The mean valuesfor all varieties (3·9±0·15 mol m–3K+) is far below the estimated cytoplasmic [K+] (100-200 molm–3). It is, therefore, unlikely that differences in theutilization of K+ by these varieties can be explained on thebasis of differential requirements for (K+) activation of theseenzymes. The kinetic properties of phosphoenolpyruvate (PEP) carboxylasehave been studied among several Flaveria species: the C3 speciesF. cronquistii, the C3–C4 species F. pubescens and F.linearis, and the C4 species F. trinervia. At either pH 7 or8, the maximum activities (in µmol.mg Chl–1.h–1)for F. pubescens and linearis (187–513) were intermediateto those of the C3 species (12–19) and the C4 species(2,182–2,627). The response curves of velocity versusPEP concentration were hyperbolic for the C3 and C3–C4species at either pH 7 or 8 while they were sigmoidal for theC4 species at pH 7 and hyperbolic at pH 8. The Km values forPEP determined from reciprocal plots were lowest in the C3 species,and of intermediate value in the C3–C4 species comparedto the K' values of the C4 species determined from Hill plotsat either pH 7 or 8. Glucose-6-phosphate (G6P) decreased theKm values for PEP at both pH 7 and 8 in the C3 and C3–C4species. In the C4 species, G6P decreased the K' values at pH8 but increased the K' values at pH 7. In all cases, G6P hadits effect by influencing the activity at limiting PEP concentrationswith little or no effect on the maximum activity. At pH 8 andlimiting concentrations of PEP the degree of stimulation ofthe activity by G6P was greatest in the C4 species, intermediatein F. linearis, a C3–C4 species, and lowest in the C3species. Millhouse, J. and Strother, S. 1987. Further characteristicsof salt-dependent bicarbonate use by the seagrass Zostera muelleri.—J.exp. Bot. 38: 1055–1068. The contribution of HCO3to photosynthetic O2 evolutionin the seagrass Zostera muelleri Irmisch ex Aschers. increasedwith increasing salinity of the bathing seawater when the inorganiccarbon concentration was kept constant. K1/2 (seawater salts)for HCO3 -dependent photosynthesis was 66% of seawatersalinity. Both short- and long-term pretreatment at low salinitiesstimulated photosynthesis in full strength seawater. Twentyfour hours pre-incubation of seagrass plants in 3·0 molm–3 NaHCO3 resulted in increased photosynthesis at allsalinities, apparently due to stimulation of HCO3 use(K1/2 (seawater salts) = 26%). Vmax (HCO3) was not affectedby low salinity pretreatment. The kinetics of HCO3 stimulationby the major seawater cations was investigated. Ca2+ was themost effective cation with the highest Vmax (HCO3) andwith K1/2(Ca2+) = 14 mol m–3. Mg2+ was also very effectiveat less than 50 mol m–3 but higher concentrations wereinhibitory. This inhibition cannot be accounted for solely byprecipitation of MgCO3. Na+ and K+ were both capable of stimulatingHCO3 use. Stimulation was in two distinct parts. Up to500 mol m–3, both citrate and chloride salts gave similarresults (K1/2(Na+) 81 mol m–3, Vmax(HCO3) 0·26µmol O2 mg–1 chl min–1), but use of citratesalts above 500 mol m–2 caused a second stimulation ofHCO3 use (K1/2(Na+) 830 mol m–3, Vmax(HCO3)0·68 µmol O2 mg–1 chl min–1). Vmax(HCO3)for the second-phase Na+ or K+ stimulation was of the same orderas for Ca2+-stimulated HCO3 use. To further characterizesalt-dependent HCO3 use, the sensitivity of photosynthesisto Tris and TES buffers was investigated. The effects of Trisappear to be due to the action of Tris+ causing stimulationof HCO3 -dependent photosynthesis in the absence of salt,but inhibition of HCO3 use in saline media. TES has noeffect on photosynthesis. External carbonic anhydrase, althoughimplicated in salt-dependent HCO3 use in Z. muelleri,could not be detected in whole leaves. Influx of nitrate into the roots of intact barley plants wasfollowed over periods of 1–15 min using nitrogen-13 asa tracer. Based on measurements taken over 15 min from a rangeof external nitrate concentrations (0·2–250 mmolm–3), the kinetic parameters of influx, Imax and Km, werecalculated. Compared with plants grown in the presence of nitrate throughout,plants that had been starved of N for 3 d showed a significantlygreater value ofImax for 13N-nitrate influx (by a factor of1·4–1·8), but a similar value of Km (12–14mmol m–3). Pre-treating N-starved plants with nitratefor about 5 h further increased the subsequent rate of 13N-nitrateinflux, but had little effect in the unstarved controls. Allowingfor this induction of additional nitrate transport, the differencein rates of nitrate influx in control and N-starved plants wassufficient to account for the previously-observed differencein net uptake by the two groups of plants. In barley plants grown without any exposure to nitrate, butwith ammonium as N-source, both Imax and Km for subsequent 13N-nitrateinflux were significantly decreased (by about one-half) comparedwith the corresponding nitrate-grown controls. The effects of ABA, isobutyric acid (IBA) and nicotine on K+and Cl+ efflux were studied in Arabidopsis thaliana seedlings,and the role of pHcyt, and Em in the regulation of the effluxof these ions was discussed. The data show that treatments withIBA and nicotine influenced in opposite directions the effluxof either K+ or Cl: K+ efflux was increased by nicotineand reduced in the presence of IBA, whereas Cl effluxwas stimulated by IBA and decreased by nicotine treatment. Underall the conditions tested ABA induced cytoplasmic acidificationand inhibition of K+ and Cl net efflux. Experiments aimedto estimate the individual contribution of pHcyt and Em in modulatingK efflux indicated that, within the range of acidic pHcytvalues, a regulation of K+ efflux was imposed by pHcyt on thecontrol exerted by Em, the efflux being inhibited by lower pHcytvalues. Conversely, in the alkaline side of pHcyt K+ effluxseemed linked only to the Em values. These results are consistentwith the hypothesis that the decrease in K+ efflux observedin non-stomatal tissues in the presence of ABA may be mediatedby the cytoplasmic acidification induced by the hormone. Exposure to high K+ medium transforms Chara plasmalemma into[K+]osensitive state (K+ state). The current-voltage (I/V)characteristicsunder such conditions display a negative conductance region.This feature results from the complex time and voltage dependenceof K+ channel opening At potentials more negative than a thresholdp.d. the channels are closed and the I/V characteristics becomelinear with a low slope conductance of 0.8 S m2 and only a weakdependence on [K+]o. Such behaviour is usually associated witha non-specific leak current The threshold level for K+ channelclosing depends on [K+]o. In 2.0 mol m–3 and 5.0 mol m–3K+ medium the membrane resting p.d. follows EK, but hyperpolarizesgradually if the [K+]o is lowered. The proton pump thus appearsto be non-operative, while the cell is in the K+ state, andrecovers slowly as the cell is returned to a low K+ medium.Excitation currents decline if the cells are kept in K+ statefor some hours. Memon, A. R., Saccomani, M. and Glass, A. D. M. 1985. Efficiencyof potassium utilization by barley varieties: The role of subcellularcompartmentation.?J. exp. Bot. 36: 1860–1876. The subcellulardistributions of K+ in roots of three barley (Hordeum vulgareL.) varieties, grown at 10 and 100 mmol m–3 external K+([K+]o) were estimated by compartmental analyses. In general,increased [K+]o caused a 2–3 fold increase in vacuolar[K+], but cytoplasmic [K+] increased only slightly. Nevertheless,the three varieties, which had been selected for study on thebasis of their different rates of K+ utilization, showed distinctdifferences in the allocation of K+ between cytoplasm and vacuole.At 10 mmol m–3 [K+]o var. Betzes exhibited typical K+deficiency symptoms while var. Fergus and var. Compana did not,even though Betzes had higher [K+] in shoots and roots. Theinefficient utilization of K+ in this variety appears to beassociated with a failure to mobilize vacuolar K+ into the cytoplasmiccompartment (the ratio of vacuolar: cytoplasmic K+ contentsfor Betzes was 4.1 compared to 2.7 and 2.5, respectively, forFergus and Compana). Fergus and Betzes, which demonstrate pronouncedgrowth responses to increased [K+]0 between 10 and 100 mmolm–3, showed significant increases of cytoplasmic [K+]in this range of [K+]o. By contrast, cytoplasmic [K+] in Compana,a variety whose growth is not stimulated by increased [K+]0(from 10 to 100 mmol m–3) showed virtually no increase.It is suggested that the efficiency of K+ utilization and thegrowth response to [K+]0 in these varieties are functions ofthe subcellular distribution of this ion between cytoplasm andvacuole. Net accumulation of Cl by intact barley plants was virtuallyeliminated in roots and reduced by 40% in shoots when externalmedia (0.5 mol m–3 CaSO4 plus 0–5 mol m–3KCI) were supplemented with 0.25 mol m Ca(NO3)2. Plasmalemma36Cl influx (oc) was shown to be insensitive to externalNO3- in plants which had previously been grown in solutionslacking –3, but oc became sensitive to NO3-after a lagperiod of 3–6 h. Kinetic analyses revealed that the inhibitionof 36C1 influx by external NO3- was complex. At 0.25mol m–3 NO3- the Vmax for Cl influx was reducedby greater than 50%, with insignificant effects upon Km. At0.5 mol m–3 NO3- there was no further effect upon Vmaxbut Km for influx increased from 38±5 mmol m–3to 116±26 mmol m–3. By contrast, Cl effluxwas found to be insensitive to external NO3-. A model for theregulation of Cl influx is proposed which involves bothnegative feedback effects from vacuolar NO3- +Cl) concentrationand (external) NO3- inhibition of Cl influx at the plasmalemma.These combined effects serve to discriminate against Claccumulation, favouring NO3- accumulation, when the latter ionis available. Such observations are inconsistent with recentproposals for the existence of bona fide homeostats for chlorideaccumulation in higher plants. The use of chlorate as an analogue for NO3 during nitrateuptake into Chara corallina cells has been investigated. NO3inhibits 36C1O3 influx into Chara over the concentrationrange 0–1000 mmol m–3. Lineweaver-Burke plots ofthe data are characteristic of competitive inhibition by NO–3in the low concentration range (0–300 mmol m–3 ClO3)and apparent KINO3 is 140 mmol m–3 which is of a similarorder of magnitude as apparent KmCIO3- 180 mmol m–3. Athigher substrate concentrations the inhibition by NO3was not characteristic of competitive or uncompetitive inhibition. 36C1O3/NO3 influx was dependent on K+ and Ca2+in the external medium and inhibited by FCCP. NO3 pretreatmentor N starvation increased subsequent 36C1O3/NO3influx into Chara. A comparison between rates of net NO3uptake and 36C1O3/NO3 influx supported the previoushypothesis that NO3 efflux is an important componentin the determination of overall uptake rates. When young plants of Macroptilium atropurpureum, cv. Siratrowere deprived of external sulphate (-S plants) growth of shootsand roots continued at rates comparable to those in plants wellsupplied with sulphate (control) for 3 d and 5 d respectively.Dilution of internal sulphur therefore took place and redistributionof sulphur occurred between inorganic and organic forms andbetween roots and younger leaves. Even when S-deficiency limitedgrowth, plants contained 16% of their total sulphur as sulphate,but most of this was retained in old leaves and redistributedslowly to growing zones. The capacity for sulphate uptake increased in roots of –Splants very soon after they were deprived of external sulphate;within 24 h the absorption from 0.25 mol m–3 SO42–was more than five times that of control roots. Maximum increasedcapacity was reached after 2–3 d stress when the Vmaxof system 1 was 1948 nmol h–1g–1root fr. wt. in–S plants and 337 nmol h–1g–1root fr. wt.in controls. The Kmfor system 1 did not change significantlywith S-stress being between 5–8 µM in both setsof plants. Absorption of L-cysteine was not stimulated by S-stress. There was a close, positive relationship between plant growthrate and the rate at which sulphate uptake capacity was enhancedby withholding sulphate from culture solutions. When –S plants were replaced in sulphate-containing solutiontheir capacity for SO42– declined to the control levelwithin 24 h. Very marked repression of capacity was also foundwhen –S plants were treated with L-cysteine, but therewas no immediate effect with methionine. Roots of this species appear to have a very active system fordegrading L-cysteine to sulphate, 30% of the label in 35S-cysteineabsorbed by roots was recovered in 35SO42– after 20 minor 2 h incubation. By contrast, roots had a very weak abilityto reduce sulphate. When part of the root system was in solution lacking sulphatethere was enhanced uptake of sulphate by other parts which themselveswere amply supplied with sulphate. This is seen as an exampleof compensatory absorption. The response to S-stress is specific and there were no positiveinteractions between S-stress and the absorption of phosphate,or P-stress and the uptake of sulphate. The results are discussed in relation to the close control ofsulphate uptake by internal sulphate concentration, redistributionof forms of sulphur during stress and mobility of sulphate inthe phloem. The N-linked oligosaccharides of frog (Rana pipiens) rhodopsinwere analysed by sequential exoglycosidase digestion and gelfiltration chromatography, following reductive tritiation. Inaddition, selected tryptic glycopeptides obtained from frogretinal rod outer segment membranes were examined by electrospraymass spectrometry (ES-MS), fast atom bombardment mass spectrometry(FAB-MS), amino acid sequence and composition analysis, andcarbohydrate composition analysis. The amino acid sequence datademonstrated that the glycopeptides were derived from rhodopsinand confirmed the presence of twoN-glycosylation sites, at residuesAsn2 and Asn15. The predominant glycan (60% of total) had thestructure GlcNAcß1–2Man1–3(Man1–6)Manß1–4GlcNAcß1–4GlcNAc-(Asn),with the remaining structures containing 1–3 additionalhexose residues, as reported previously for bovine rhodopsin.Unlike bovine rhodopsin, however, a sizable fraction of thetotal giycans of frog rhodopsin also contained sialic acid (NeuAc),with the sialylated oligosaccharides being present exclusivelyat the Asn2 site. FAB-MS analysis of oligosaccharides releasedfrom the Asn2 site gave, among other signals, an abundant quasimolecularion corresponding to a glycan of composition NeuAc1Hex6HexNAc3(where Hex is hexose and HexNAc is N-acetylhexosamine), consistentwith a hybrid structure. Guard cell protoplasts (GCP) were prepared from leaves of Commelinacommunis L. and phosphoenolpyruvate carboxylase (PEPc) activityrecorded after injection of the protoplasts directly into theassay medium. The GCP were lysed immediately by the presenceof Triton X-100 and a lowered osmotic concentration in the assaycuvette enabling PEPc activity to be measured with ‘nascent’enzyme. There was no light activation of the enzyme with KmPEP (about 3.7 mol m–3) and Vmax being similar in light-ordark-treated protoplasts. Illumination of the GCP in the presenceof CO2-free air and KCI, a treatment which is known to swellGCP, did not change the kinetics. PEPc activity at saturating PEP was very sensitive to malateinhibition, 20 mmol m–3 (the I50 value) inhibiting activityby about 50%. Inhibition was similar in light- or dark-treatedprotoplasts. Malate inhibition was, however, much less (I50= 500 mmol m–3) if the enzyme source was a protoplastextract kept in the absence of glycerol. Inclusion of 20% glycerolin the extraction medium maintained the enzyme in the malate-sensitiveform as occurred in the in vivo assays. The high apparent KmPEP and the high sensitivity to malate inhibition of GCP PEPcare features unlike those observed with PEPc from leaf tissuesof C4 and CAM plants and from GCP extracts. PEPc activity increased slightly in the presence of KCI in theassay medium up to about 10 mol m–3 and thereafter activityslowly declined as KCI concentrations increased further. In situ growth and development of Neocalanus flemingeri/plumchrusstage C1–C4 copepodites were estimated by both the artificial-cohortand the single-stage incubation methods in March, April andMay of 2001–2005 at 5–6°C. Results from thesetwo methods were comparable and consistent. In the field, C1–C4stage durations ranged from 7 to >100 days, dependent ontemperature and chlorophyll a (Chl a) concentration. Averagestage durations were 12.4–14.1 days, yielding an averageof 56 days to reach C5, but under optimal conditions stage durationswere closer to 10 days, shortening the time to reach C5 (fromC1) to 46 days. Generally, growth rates decreased with increasingstage, ranging from 0.28 day–1 to close to zero but weretypically between 0.20 and 0.05 day–1, averaging 0.110± 0.006 day–1 (mean ± SE) for single-stageand 0.107 ± 0.005 day–1 (mean ± SE) forartificial-cohort methods. Growth was well described by equationsof Michaelis–Menten form, with maximum growth rates (Gmax)of 0.17–0.18 day–1 and half saturation Chl a concentrations(Kchl) of 0.45–0.46 mg m–3 for combined C1–3,while Gmax dropped to 0.08–0.09 day–1 but Kchl remainedat 0.38–0.93 mg m–3 for C4. In this study, in situgrowth of N. flemingeri/plumchrus was frequently food limitedto some degree, particularly during March. A comparison withglobal models of copepod growth rates suggests that these modelsstill require considerable refinement. We suggest that the artificial-cohortmethod is the most practical approach to generating the multispeciesdata required to address these deficiencies.  相似文献

To study possible changes in the transport metabolites betweenchloroplasts and cytoplasm during CAM induction of Mesembryanthemumcrystallinum, we compared substrate specificity of P11 translocator(s)in isolated chloroplasts from the C3 and CAM-induced plants.The [14C]glu-cose 6-phosphate (G6P) transport activity was significantonly in the chloroplasts of CAM-mode plants and not detectablein those of C3-mode, while a similar high rate of [32P]Pi uptakewas observed with both types of chloroplasts. Kinetic analysisof G6P uptake in the CAM chloroplasts showed a high Vmax [10.6µmol (mg Chl)–1 h–1] and a comparatively lowKm value (0.41 mM); the latter was similar to Ki values of Pi,3-phosphoglycerate and phospho-enolpyruvate, 0.30, 0.34 and0.47 mM, respectively. On the other hand, [32P]Pi uptake inthe CAM chloroplasts was inhibited competitively by G6P witha Ki value (8.4 mM) 20-fold higher than the Km value for G6Puptake, while that in C3 chloroplasts was not inhibited at all.These results suggest that a new G6P/Pi, counterexchange mechanismis induced in the chloroplast envelope of CAM-induced M. crystallinumin addition to the ordinary type of P, translocator, that cannottransport G6P, already present in the C3-type chloroplasts. Effects of diffusion and upwelling on the formation of red tides   总被引:4,自引:0,他引:4  
In this paper, records on the timing and location of specificred tides monitored once or twice a week in Mikawa Bay, Japan,are related to horizontal and vertical mixing rates determinedfrom a numerical model. Horizontal (Kh) and vertical (Kz) diffusioncoefficients, and upwelling velocities, were estimated usinga box model analysis. In the wind-mixed period and in the upperlayer during the stratified period, Kh was estimated to be ofthe order of 102 m2 s–1. During the stratified period,Kz was estimated to be of the order of 10–5 m2 s–1.The upwelling velocity was calculated to be in the range 0.35–5.1m day–1 with an average of 1.5 m day–1. Comparisonbetween the literature values of the specific growth rate (µ)of the red tide-forming diatoms and calculated Kh values duringthe red tides show that diatoms which have a low µ cannotform red tides in a strongly diffusive environment, while specieshaving a high µ can form red tides even in a strong diffusiveenvironment. On the other hand, no clear relationship was foundbetween µ of the flagellate group and Kh, although theflagellate group formed red tides even in severe diffusive conditions.From the comparison between the literature values of sinkingrate and swimming speed and the physical parameters associatedwith vertical processes, it was concluded that flagellates willform red tides, even in severe diffusive conditions, by usingtheir swimming ability, while diatoms form red tides by theirhigh growth rates with the aid of vertical diffusion and theupwelling movement of water.  相似文献
