Isoparorchis hypselobagri was maintained in a non-nutrient medium containing streptomycin and penicillin to prevent bacterial growth for a period of 12 hours. The parasites were kept under aerobic conditions at a suitable temperature of 30 degrees C. The protein content in Isoparorchis hypselobagri ranged from 56.25% to 67.5% with an average of 63.5%+/-4.6 of dry seight of the tissue. The nitrogen of Isoparorchis hypselobagri excreted as ammonia, after 12 hours of starvation was 3.04% of the total body nitrogen and as uric acid 1.6% of total body nitrogen.  相似文献

Alkaline phosphatases from different trematodes occupying the same habitat have identical pH otima but different levels of enzyme activities. Isoparorchis hypselobagri, from the fish Wallago attu, shows four to six times more enzyme activity than Fasciolopsis buski, Gastrodiscoides hominis and Echinostoma malayanum, from the pig Sus scrofa, and Fasciola gigantica, Gigantocotyle explanatum, Cotylophoron cotylophorum and Gastrothylax crumenifer, from the buffalo Bubalus bubalis. At least two peaks of activity at different levels of pH were obtained for each trematode examined. Both Gastrodiscoides hominis and Isoparorchis hypselobagri enzymes had three peaks of alkaline phosphatase activity. The optimum temperature for maximum enzyme activity was 40 degrees C, above which rapid inactivation occurred. At temperatures below 40 degrees C, the enzymes of fish and mammalian trematodes did not behave similarly; I. hypselobagri enzyme being active over a wider range of temperature (20 degrees-40 degrees C. Various concentrations of KCN and arsenate proportionately inhibited enzyme activity. NaF Did not significantly influence enzyme activity, while Mg++ and Co++ acted as activators. The extent of inhibition or activation of enzyme activity of different trematodes varied, probably due to species differences. Both inhibition and activation of I. hypselobagri enzyme was higher than in the case of other trematodes.  相似文献

The swim bladder gas of Wallago attu contains O2 (22-58 mm Hg) and the presence or absence of Isoparorchis hypselobagri does not influence the O2 content of the swim bladder. Glucose increases the O2 consumption of I. hypselobagri by 50%. With the passage of in vitro culture time, the rate of O2 consumption gradually decreases to the extent of 64% on the 40th day. The optimal temperature for O2 consumption is 30 degrees C beyond which the rise in temperature is detrimental to the fish trematode as manifested by the decrease in their O2 consumption.  相似文献

对从嘉陵江北碚江段采集到的264尾瓦氏黄颡鱼的调查表明,瓦氏黄颡鱼感染鳔等睾吸虫的感染率和感染强度分别为25.70%和1.50;体长在12~18 cm的个体感染的可能性增大,而体长在14~15 cm之间的个体感染率最高,为38.00%;此外还比较了感染鱼和未感染鱼的肥满度(F)、脂肪系数(ASI )、肝系数(HSI),发现鳔等睾吸虫对瓦氏黄颡鱼的肥满度有显著影响,感染鱼的肥满度下降了16.20%,对脂肪系数、肝系数的影响不显著.感染鱼的肝脏、肠系膜等器官均有不同程度的病变.  相似文献

Oxyhaemoglobins of six digenetic trematodes,--Srivastavaia indica, Gastrothylax crumenifer, Gigantocotyle explanatum, Fasciolopsis buski, Gastrodiscoides hominis, Isoparorchis hypselobagri, and of their 3 vertebrate hosts, Bubalus bubalis, Sus scrofa, Wallago attu, were subjected to alkali denaturation at a pH of 12.4. All oxyhaemoglobins from trematodes and their hosts differ from each other in the rate and extent of alkali denaturation which may be explained due to variations in the amino acid sequences of a particular haem protein in addition to other factors.  相似文献

In a basic attempt to develop the prophylactic and therapeutic measures on intestinal giant-cystic disease of the Israel carp, Cyprinus carpio nudus, the effects of physical and chemical factors on viability or survival of the spores of Thelohanellus kitauei were checked in vitro by means of extrusion test on the polar filament. When the fresh spores suspended with 0.45% and 0.9% sodium chloride solution and distilled water were laid at 5 degrees C and 28 degrees C for short terms, the extrusion rates increased until the 3rd day, meanwhile when some of them were suspended with Tyrode's solution at -70 degrees C the rates increased gradually until the 8th day. Viabilities of the spores suspended with 0.9% saline and added antibiotics to the suspension at 5 degrees C for long terms lasted for 997 days and 1,256 days (presumed values) at maximum, respectively. The spores suspended with distilled water at 28 degrees C for long terms survived 152.4 days, but the spores suspended with Tyrode's solution at -70 degrees C for long terms showed almost the same viable pattern as early freezing stages up to 780 days. The spores suspended with Tyrode's solution, frozen at -70 degrees C and thawed at 5 degrees C, showed the highest rate of extrusion of the polar filament. In the case of frozen spores, the extrusion rates during heating tend to become higher in accordance with the increase of frozen period, and the critical points of 180 day-frozen spores to be killed were generally 78.5 hr. at 60 degrees C, 23.4 hr. at 70 degrees C, 189.1 min. at 80 degrees C or 10.5 min. at 90 degrees C. The longer the spores were frozen, the more time was needed for the death of spores after thawing; 20 days-17.4 days, 100 days-33.2 days, and 400 days-37.8 days. The longer the spores were frozen, the more time was needed for the death of spores at a conventional when they were dried air drying condition, 540 days-23.5 days, 160 days-21.0 days, and 20 days-14.4 days. On the other hand, the longer the spores were frozen, the more spores were dead rapidly when they were irradiated with 10W UV-ray; 100 days-26.0 hr, 300 days-21.9 hr, and 540 days-13.9 hr. The time needed for killing 200 days-frozen spores by various disinfectants at 1,000 ppm was 5.2 min. by calcium oxide, 10.4 min. by potassium permanganate, 27.8 min. by malachite green and 14.3 hr. by formalin.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Quantitative estimation of absolute levels and in vitro release of acetylcholinesterase (AChE) in seven species of digenetic trematodes: Isoparorchis hypselobagri from the swim bladder of catfish, Wallago attu; Srivastavaia indica and Gastrothylax crumenifer from the rumen, and Gigantocotyle explanatum from the liver of the water buffalo, Bubalus bubalis; Fasciolopsis buski, Echinostoma malayanum from the small intestine and Gastrodiscoides hominis from the caecum of the pig, Sus scrofa revealed that the enzyme is present in remarkably high quantities in species which inhibit gastrointestinal tract compared with those that parasitize liver and swim bladder. The rate of in vitro release of AChE also varies with the species which supports the view that such differential secretion probably takes place in situ as well to counteract peristalsis and it is a biochemical adaptation on the part of these trematodes.  相似文献

Adult schistosomes live in the host''s bloodstream where they import nutrients such as glucose across their body surface (the tegument). The parasite tegument is an unusual structure since it is enclosed not by the typical one but by two closely apposed lipid bilayers. Within the tegument two glucose importing proteins have been identified; these are schistosome glucose transporter (SGTP) 1 and 4. SGTP4 is present in the host interactive, apical tegumental membranes, while SGTP1 is found in the tegumental basal membrane (as well as in internal tissues). The SGTPs act by facilitated diffusion. To examine the importance of these proteins for the parasites, RNAi was employed to knock down expression of both SGTP genes in the schistosomula and adult worm life stages. Both qRT-PCR and western blotting analysis confirmed successful gene suppression. It was found that SGTP1 or SGTP4-suppressed parasites exhibit an impaired ability to import glucose compared to control worms. In addition, parasites with both SGTP1 and SGTP4 simultaneously suppressed showed a further reduction in capacity to import glucose compared to parasites with a single suppressed SGTP gene. Despite this debility, all suppressed parasites exhibited no phenotypic distinction compared to controls when cultured in rich medium. Following prolonged incubation in glucose-depleted medium however, significantly fewer SGTP-suppressed parasites survived. Finally, SGTP-suppressed parasites showed decreased viability in vivo following infection of experimental animals. These findings provide direct evidence for the importance of SGTP1 and SGTP4 for schistosomes in importing exogenous glucose and show that these proteins are important for normal parasite development in the mammalian host.  相似文献

Two experiments were performed in vitro with human sperm from a panel of 11 donors. In both experiments, washed sperm from each donor were divided into four equivolume samples, three of which were resuspended in various formulations of Tyrode's solution and the fourth was resuspended in its own seminal plasma. Each of the four samples was then stored at 37 or 3 degrees C for fixed intervals of 0, 2, 6, 12, and 24 h. Motility was assessed at 37 degrees C for all samples at the end of each interval. The results indicate that sperm survival in vitro at 3 degrees C was significantly enhanced by 20 mM K+ in Tyrode's solution relative to Tyrode's with less K+. A metabolizable sugar such as glucose was essential to maintaining sperm viability in K(+)-free media. The addition of raffinose to media containing glucose improved motility of sperm stored at 3 degrees C for 6 h.  相似文献

Comparisons were made between glucose, sucrose, and dimethyl sulfoxide (DMSO) as cryoprotective agents for the hemoprotozoan parasite, Babesia rodhaini, using infectivity for mice as the criterion of survival. Concentrations of the cryoprotectants tested were from 0.1 to 0.5 M for the sugars, and 1.5 to 2.5 M for DMSO. Glucose and sucrose were comparable as cryoprotectants, although glucose reduced infectivity of the parasites slightly more than did sucrose at above-freezing temperatures. When sucrose and DMSO were compared for cryoprotection during cooling to ?196 °C at nominal rates of 5, 100, and 500 °C/min, parasite survival varied with the type and concentration of cryoprotectant, but was higher in blood containing DMSO at all three cooling rates. The percentages of parasites that survived cooling at 100 °C/min and frozen storage in the presence of DMSO ranged from 20 to 36%.  相似文献

Metacercariae of Clinostomum marginatum obtained from naturally infected Perca flavescens were cultured by 5 methods. In vitro cultivation at 37 C in twice daily changes of Tyrode's, Na-poor Tyrode's, and Eagle's media failed to produce ovigerous adults after 4-5 days. Metacercariae placed on the chorioallantois of week-old chick embryos at 37 C migrated to the albumen. Only 8 of 123 worms recovered were ovigerous after 4-7 days in ovo. Best success occurred with metacercariae injected in groups of 4 or 6 into the abdominal cavities of male A/J mice. Despite liver lesions and strong inflammatory responses in the host, all 174 worms recovered were ovigerous after 4-8 days. None of 41 mice died prematurely from the procedure or the parasites.  相似文献

Sperm-freezing extenders supplemented with sugar or a combination of different sugars are widely used for the cryopreservation of nonhuman primate spermatozoa. Understanding which sugar or combination of sugars offers the highest level of cryoprotection would be beneficial for the development of sperm-freezing extenders. In the present study we aimed to investigate the effect of glucose, lactose, and raffinose separately or in combination on the cryosurvival of rhesus monkey spermatozoa. Toward that end, we prepared eight extenders by adding various types of sugars to a basic medium (BM): G-BM (0.3 M glucose), L-BM (0.3 M lactose), R-BM (0.3 M raffinose), LG-BM (0.15 M lactose+0.15 M glucose), RG-BM (0.15 M raffinose+0.15 M glucose), LR-BM (0.15 M lactose+0.15 M raffinose), and LRG-BM (0.1 M lactose+0.1 M raffinsoe+0.1 M glucose). A saline control (0.157 M sodium chloride) was also used. The results showed no significant difference in post-thaw motility when spermatozoa were frozen with G-BM, L-BM, LG-BM, RG-BM, and LRG-BM, but the post-thaw motility was significantly lower when it was frozen with R-BM, LR-BM, and the saline control. The highest plasma membrane integrity was achieved when spermatozoa were frozen with G-BM, L-BM, LG-BM, RG-BM, and LRG-BM, and the highest acrosome integrity was achieved with G-BM, L-BM, LG-BM, RG-BM, LRG-BM, and the saline control. The results indicate that the various sugars offered different protective effects. For the cryopreservation of rhesus monkey spermatozoa, glucose (monosaccharide) and lactose (disaccharide) were shown to be more suitable than raffinose (trisaccharide) for preserving spermatozoal motility, plasma membrane, and acrosome. Specifically, raffinose was detrimental to sperm acrosome integrity.  相似文献

A microbial sensor consisting of immobilized living whole cells of Brevibacterium lactofermentum and an oxygen electrode was prepared for continuous determination of total assimilable sugars (glucose, fructose and sucrose) in a fermentation broth for glutamic acid production. Total assimilable sugars were evaluated from oxygen consumption by the immobilized microorganisms. When a sample solution containing glucose was applied to the sensor system, increased consumption of oxygen by the microorganisms caused a decrease in the dissolved oxygen around the Teflon membrane of the oxygen electrode and the current of the electrode decreased markedly with time until steady state was reached. The response time was ≈ 10 min by the steady state method and 1 min by the pulse method. A linear relationship was found between the decrease in current and the concentration of glucose (<1 mM), fructose (<1 mM) and sucrose (<0.8 mM). The ratio of the sensitivity of the microbial sensor to glucose, fructose and sucrose was 1.00:0.80:0.92. The decrease in current was reproducible to within 2% of the relative standard deviation when a sample solution containing glucose (0.8 mM) was employed for experiments. The selectivity of the microbial sensor for assimilable sugars was satisfactory for use in the fermentation process. The additivity of the response of the microbial sensor for glucose, fructose and sucrose was examined. The difference between the observed and calculated values was within 8%. The microbial sensor was applied to a fermentation broth for glutamic acid production. Total assimilable sugars can be determined by the microbial sensor which can be used for more than 10 days and 960 assays.  相似文献

Clonorchis sinensis is a biological carcinogen inducing human cholangiocarcinoma, and clonorchiasis is one of the important endemic infectious diseases in East Asia. The present study investigated survival longevity of C. sinensis adult worms in various in vitro conditions to find the best way of keeping the worms longer. The worms were maintained in 0.85% NaCl, 1×PBS, 1×Locke''s solution, RPMI-1640, DMEM, and IMDM media, and in 1×Locke''s solution with different supplements. All of the worms died within 3 and 7 days in 0.85% NaCl and 1×PBS, respectively, but survived up to 57 days in 1×Locke''s solution. The worms lived for 106 days in DMEM, and 114 days in both RPMI-1640 and IMDM media. The survival rate in RPMI-1640 medium was the highest (50%) compared to that in DMEM (20±10%) and in IMDM (33.3±25.2%) after 3 months. The 1×Locke''s solution with 0.005% bovine bile supplement showed increased duration of maximum survival from 42 days to 70 days. Higher concentration of bile supplements than 0.005% or addition of glucose were disadvantageous for the worm survival. The worms died rapidly in solutions containing L-aspartic acid, L-glutamic acid, and adenine compared to L-arginine, L-serine, and L-tryptophan. In conclusion, the 1×Locke''s solution best supports the worms alive among inorganic solutions for 57 days, and the RPMI-1640 medium maintains living C. sinensis adults better and longer up to 114 days in vitro than other media.  相似文献

To understand the nature and amount of excretory products of Isoparorchis hypselobagri, a digenetic trematode inhabiting the swim bladder of Wallago attu the parasites were kept in PBS media without glucose (control) and with glucose (treated) successfully up to 300 h and estimated different excretory products in the media after every alternate 12 h. The quantitative estimation of urea excreted by I. hypselobagri is done. It is lower when the flukes were kept in vitro in the control incubation media than in the treated incubation media. The highest amount of urea excreted in control media was 18.563 mg % at 276 h, and in treated condition 29.759 mg % after 216 h of incubation, while the lowest amount of urea excreted, 6.08 mg %, in control media and 10.343 mg % in treated media respectively after 12 h of incubation. The results were highly significant at 5 % level. The rate of change of excretion of urea in treated condition after every 12 h of incubation time interval was also studied. In the swim bladder washings of host urea is also present in significant amount and the amount excreted depends on the number of parasites harbor. From the results presence of both ammonotelic and ureotelic conditions are suggested.  相似文献
