Electron microscope studies were made of retrogradely horseradish peroxidase-labeled pyramidal neurons forming transcallosal projections in layer III of the cat primary auditory cortex (Al). These showed a significant proportion of the somatic membrane to be covered with processes of astroglia, while synapses occupy 20% of the synaptic surface on average. Between 4 and 10 axosomatic synapses were identified on the profiles of callosal cell somata. All these were formed by axonal terminals containing small, flattened synaptic vesicles and had symmetrical contacts. Average length of these synaptic contacts equaled 1.6 µm. Numerous anterogradely horseradish peroxidase-labeled axonal terminals of callosal fibers were found in cortical area Al in amongst retrogradely HP-labeled neurons. Electron microscope research was conducted on the synaptic apparatus of the feline primary auditory cortex (Al). A total of 2096 profiles of axonal terminals (AT) were found over a total area of 8230 µm2 of ultrathin slices at different layers of this cortical layer — an average of 255 profiles per 1000 µm2 of the surface area on these slices. The AT profiles occupied about 8.9% of the surface of these cross-sections. It was found that 52% of the AT containing synaptic vesicles formed asymmetrical or symmetrical synaptic contacts (83.9% and 16.1% respectively) and that AT had no contacts which could be considered synaptic junctions on 48% of slices. It was also observed that 45.3% of the AT forming contacts synapsed on spines, 48.5% on dendrites, and 6.2% on neuronal somata. Finally, 95.4% and 4.6% of axo-spinal synapses contained rounded and flattened vesicles respectively; equivalent figures for axodendritic synapses were 79.4% and 20.6% respectively and 19.8 and 80.2% for axosomatic synapses. Calculations revealed an average of 322.8 × 106 AT over 1 mm3 of cat auditory cortex. Organizational aspects of synaptic apparatus at different layers of area A1 were ascertained.A. A. Ultrastructural features of thalamic afferent fibers were studied in the cat auditory cortex in the early stages (on the 4th day) of experimental degeneration produced by destruction of the medial geniculate body. A coordinate grid was used in conjunction with an electron micro-scope to study the topography of the degenerating elements over wide areas of sections, so that the density of degeneration could be determined quantitatively in different layers of the cortex. Degenerating axons were found in all layers. Most of the large (5–7 µ) degenerating axons are located in layer VI; their diameters were smaller in the upper layers of the cortex. Degenerative changes affecting synaptic terminals of thalamo-cortical afferents were of the "dark" type. Fibers of the geniculo-cortical tract were shown to terminate mainly in cortical layer IV. A few degenerating synapses were found in the molecular layer. Terminals with sperical synaptic vesicles are found mainly on the spines of dendrites where they form "asymmetrical" contacts. A few degenerating axo-somatic synapses were observed on stellate neurons in layer IV. The results are discussed in connection with electrophysiological investigations of the cat auditory cortex during stimulation of specific afferent fibers.A. A. Numbers of intact and degenerating axonal terminals (AT) were determined in area 5b of the cat association cortex two, four, and 30 days after electrolytic destruction of the posterolateral thalamic nucleus (PL). The proportion of degenerating AT in area 5b two and four days after PL lesion was found to be 7.4 and 7.3%, respectively. Intact AT in the same field accounted for 83.6% of the total in intact animals four days after PL lesion. A 16.4% reduction occurred, consisting of AT with round vesicles and asymmetrical synaptic contacts (i.e., primarily excitatory AT); 40% of the damaged AT formed synapses on dendrites and 60% on spines. It follows that neurons of cortical association area 5b receive direct excitatory and mainly axonospinal afferents from the thalamic PL. Degenerating AT accounted for 4.6% 30 days after PL lesion, as compared with a decline of 31.2% and a rise of 7.0% in axospinal and axodendritic AT, respectively. No change occurred in axosomatic AT. The total number of AT had declined by 10.0%. Aspects of reinnervation of area 5b neurons following degeneration of the neuronal synapses induced by PL lesion are discussed.A. A. The effects were studied of ethimizol, a substance activating memory processes, on features of synaptic transmission during experiments on frog cutaneous pectoris muscle. It was found that the presynaptic action of ethimizol consists of raising the frequency of miniature potentials, when used at a concentration of 0.5–10 mM, and modulating quantal content of synaptic transmission due to changes in binomial quantal release parameters p and n when 0.5–2 mM ethimizol was used. This substance facilitated transmission at synapses with a low initial level of transmitter release. This substance facilitated transmission at synapses with a low initial level of transmitter release. Ethimizol was also found to have a postsynaptic action, consisting of reducing amplitude at a concentration of 5–10 mM and prolonging synaptic currents and potentials when concentrations of 0.5–10 mM were used. The latter effect produced a considerable increase in the time integral of endplate potentials. The postsynaptic action of ethimizol is perhaps seen in its effects on features of postsynaptic ionic channels. An electron microscope study of retrogradely labeled pyramidal neurons in layer VI of the primary auditory cortex (AI) after injecting horseradish peroxidase (HP) into the medial geniculate body was carried out in cats. Not less than 57.8±1.9% on average of the perimeter of perikaryon profiles of corticogeniculate neurons labeled with HP were found to be covered with astroglia processes. Between three and eight synapses occupying an average of 10.8±1.0% of the perimeter length were found on the perikaryon profiles of these neurons. Nearly all synapses (a total of 98.7%) at the soma of corticogeniculate neurons had symmetrical active zones, being made up of axonal terminals with flattened synaptic vesicles. Anterogradely HP-labeled axonal terminals of geniculocortical fibers were also found in the neuropil of layer VI in area AI, in addition to retrogradely labeled neurons. They contained large round synaptic vesicles and formed asymmetrical synapses. The potential role of axosomatic synapses in the shaping of corticogeniculate neuronal activity is discussed.A. A. Summary Nerve terminals forming typical synapses with adrenal chromaffin tissues have been examined in the goldfish, frog (Rana pipiens), hamster and rat. Presumptive secretory inclusions present in the terminals are of two distinct types. Electron-lucent synaptic vesicles 30–50 nm in diameter are densely clustered adjacent to membrane thickenings and presumably discharge their contents into the synaptic clefts. Secretory granules (i.e. large dense-cored vesicles) 60–100 nm in diameter are more abundant in other parts of the terminals. Sites of granule exocytosis have been observed in each of the animals investigated. They are usually encountered within apparently undifferentiated areas of plasmalemma and only rarely occur within synaptic thickenings. Granule exocytosis from within synaptic terminals and chromaffin gland cells is most readily observed in specimens exposed, prior to fixation, to saline solutions containing both tannic acid, and 4-aminopyridine and/or elevated levels of K+. These findings show that the pattern of secretory discharge, involving both synaptic and non-synaptic release, which is widespread in invertebrate central nervous systems, is also characteristic of vertebrate, peripheral cholinergic terminals.  相似文献

The ultrastructure of layer I in the middle ectosylvian gyrus (area 22) of the cat's cerebral cortex was investigated. Beneath the subpial astrocytic layer most of the neuropil in layer I was shown to be occupied by nerve fibers and their terminals, terminal branches, dendritic spines, and astrocytic processes surrounding them. More than 90% of the presynaptic terminals contained spherical synaptic vesicles. The predominant types of interneuronal junctions are axo-spinous and axo-dendritic synapses of asymmetrical type. Presynaptic terminals, which contain flattened and pleomorphic synaptic vesicles, take part in the formation of all symmetrical junctions, accounting for 6% of the total number of synapses. Large polymorphic outgrowths filled with vacuoles — so-called multivacuolar sacs — are described. These structures were invaginated into varicose expansion of the terminal branches of apical dendrites of pyramidal neurons. They are shown to be outgrowths of presynaptic terminals. Area 5 of the cat cortex was studied by Nissl's method and by Golgi's chromate-silver impregnation method. Its typical six-layered structure with well-developed layers of pyramidal cells was revealed. The characteristic features of area 5 are: predominance of pyramidal cells in layers II–III and the presence of large forms (40×26 µ) among them (in layer III); giant pyramidal neurons (70×23 µ) arranged singly or nidally in layer V; large (diameter 25–30 µ) and giant (diameter 40–45 µ) stellate cells with radial dendrites, arranged singly or in groups in layers V–VI; infrequent efferent fusiform neurons (40×20 µ) in layers V–VI. Stellate cells connecting pyramidal neurons in the same or in different layers were found in layers II–VI. Some stellate cells in layers II–III form long horizontal connections within area 5. Interneuronal connections are effected by axosomatic and axodendritic terminals, the latter being more numerous; Dendrodendritic and axoaxonal synapses are less common.A. A. An electron-microscopic study was made of 4520 synapses in different layers of the cat auditory cortex. Of the total number of synapses 53% were located on dendritic spines, 37% on dendrites, and 10% on neuron bodies; 91% of the synapses belonged to Gray's type I, 9% to type II. Most of the type I synapses were located on dendrites and dendritic spines, whereas the type II synapses were distributed on neuron bodies, axon hillocks, and large dendrites. Signs of degeneration were discovered 60 h after complete neuronal isolation of an area of the auditory cortex in 22.8% of synapses. No degenerating type II synapses were found. This indicates that they are formed by axons of intracortical neurons. The quantitative and qualitative composition of the synapses were shown to differ in different layers of the auditory cortex.A. Involvement of the adenylate cyclase system in cholinergic modulation of synaptic transmission was investigated in area CA1 in rat hippocampal slices. Microiontophoretic application of acetylcholine as well as addition of carbachol to the superfusate or of tolbutamide (a cAMP-dependent protein kinase inhibitor) depressed transmission at synapses formed by Schaffer collaterals and commissural fibers with dendrites of pyramidal cells belonging to hippocampal area CA1. Both numbers of free quanta of neurotransmitter and the likelihood of transmitter release decreased following carbachol action. Atropine suppressed the inhibitory action of carbachol on synaptic transmission. Dibutyryl cAMP and forskolin increased the amplitude of synaptic potentials and suppressed, either partially or in full, the inhibitory effects of cholinomimetics on synaptic potentials. It was concluded that cholinomimetics and activators of the adenylate cyclase system exert opposing effects on neurotransmission at synapses formed between Schaffer collaterals/commissural fibers and dendrites of pyramidal neurons belonging to hippocampal area CA1.Institute of Biophysics, Academy of Sciences of the USSR, Pushchino. Summary The threedimensional ultrastructure of presynaptic dense bars was examined by serial section electron microscopy in the excitatory neuromuscular synapses of the accessory flexor muscle in the limbs of larval, juvenile, and adult lobsters. The cross-sectional profile of the dense bar resembles an asymmetric hourglass, the part contacting the presynaptic membrane being larger than that projecting into the terminal. The bar has a height of 55–65 nm and varies in length from 75–600 nm. In its dimensions it resembles the dense projections in the synapses of the CNS of insects and vertebrates. The usual location of these dense bars is at well defined synapses, though a few are found at extrasynaptic sites either in the axon or terminal. In the latter case the bars are close to synapse-bearing regions, particularly in the larval terminals, suggesting that the extrasynaptic bars denote early events in synapse formation. In all cases the bars are intimately associated with electron lucent, synaptic vesicles located on either side, in the indentation of its hourglass-shaped cross sectional profile. The vesicles occur along the length of the bar and contact the presynaptic membrane. Consequently the dense bar may serve to align the vesicles at the presynaptic membrane prior to exocytosis. A similar role has been suggested for the presynaptic dense bodies at the neuromuscular junction of the frog, where synaptic vesicles form a row on either side of this structure.Supported by Muscular Dystrophy Association of Canada and NSERCC. An electron microscopy study was made of synaptic organization in the cat association cortex, area 5b. A total of 1635 axonal terminals were discovered over 6215 µm2 (240 electronic imagings of slices of different association cortex layers); i.e., an average of 263±16 terminals per 1000 µm2 expanse. It was found that 75.5% of axon terminals contained synaptic vesicles and formed either one- or two-sided contact with postsynaptic structures; 24.5% of axonal terminals contained synaptic vesicles but formed no distinct synaptic contacts with nearby neurons; 84.9% of terminals contained round-shaped or slightly oval synaptic vesicles; 7.8% had both rounded and elongated shapes, and vesicles were very elongated in the remaining 7.3%. Of the axonal terminals having synaptic contacts, axo(dendritic)-spinal terminals accounted for 46.6%, and axodendritic and axosomatic endings amounted to 50.0% and 3.4% respectively (in all 77% of axosomatic terminals contained elongated vesicles and maintained symmetrical contact, while 23% had round-shaped vesicles and formed asymmetrical contact). Calculations show that for each 1 mm3 an average of 258 million axonal terminals are found forming synaptic contacts in the cat association cortex as well as 84 million terminals containing synaptic vesicles but not forming contact.A. A morphometric electron microscope study was carried out on the ultrastructure of 140 presynaptic terminals (PT) in the dorsal horn of the cat spinal cord. Some spatial characteristics of mitochondria and active zones (AZ) for these PT were examined using statistical stereological analysis techniques. The distribution of 3-dimensional mitochondrial radii was determined, together with average mitochondrial volume, mean area of the external membrane, and average numbers of the test population falling within the PT. Distribution of diameters and estimates of mean area of AZ were obtained, as well as of mean value of synaptic clefts. Statistical parameters characterizing the structure of the vesicular apparatus of presynaptic endings (PE) were determined from the findings of ultrastructural morphometric analysis of 135 synapses in the dorsal horn of the cat spinal cord. Quantitative estimates of vesicles in the PE (averaging about 470) were obtained, based on stereometric principles. The bimodal pattern of distribution of distances from the center of each vesicle to the nearest portion of the active zone was demonstrated — viewed as the structural correlate of the two-pool model of transmitter storage. The possibility of classifying PE according to the sign of vesicle spatial distribution is discussed as well as the relationship between this distribution and parameters of transmitter mobilization and synaptic release.Dnepropetrovsk State University. In the olfactory bulb (OB) of the pike, phenomena of long posttetanic potentiation (LPTP) and long posttetanic depression (LPTD) are reproduced. Probability of induction of LPTP and level of its expression are shown to depend direct on parameters of density of axo-dendritic synapses in the glomerular neuropil and size of its active zones (AZ). In pikes with the high level of these parameters (the synapse density 5.2 per 20 µm2, the mean length of AZ cross-section 293 nm), potentiation appear in 95% of cases, while in pikes with low values (the density 3.1, the AZ length 239 nm) in as low as 30%, with LPTD developing rather often instead of LPTP. The LPTP in the phase of its maximal development (60 min after tetanus) corresponds to an increase of the mean length of AZ cross-section of axodendritic synapses by 13.6%, while the LPTD—to its decrease by 16.5% as compared with control. The LPTP induction phase (5–15 min after tetanus) is not associated with any ultrastructural transformations of postsynaptic AZ of axo-dendritic synapses. By the morphometric method, it has been established that LPTP in the pike OB is of the homosynaptic nature, i.e., is due to an enhancement of synaptic efficiency of the excitatory afferent input just subjected to tetanization. Summary The fine structure of synapses and of cellular relations was examined in the somatic efferent portion of the oculomotor nucleus of the adult rhesus monkey. Axosomatic and axodendritic synapses are characterized by distinct synaptic clefts which usually measure 20–30 nm between pre- and postsynaptic membranes. Cytoplasmic thickenings of pre- and postsynaptic membranes are often observed. Subjunctional bodies are present at both axosomatic and axodendritic synapses. Somatic and dendritic spine synapses are present. Serial synapses are also found, suggesting the operation of presynaptic inhibition in this nucleus. At some synapses the extracellular gap between pre- and postsynaptic membranes is reduced to 5–9 nm. However, junctions similar to the latter are also present between neurons and glia, and at the junctions between adjacent glial elements. An electron-microscopic study was made of the normal structure of the medial part of the ventral horn (Rexed's laminae VII and VIII) in the cervical portion of the cat's spinal cord, the region where fibers of reticulospinal and vestibulospinal tracts terminate. Neurons of this region can be divided on the basis of the density of their cytoplasmic matrix into "light" and "dark," the dark being much more numerous in this area (26% of the total number counted) than in other parts of the gray matter of the spinal cord. The mean diameter of the soma of the dark cells is smaller than that of the light cells, and it usually is 15–20 µ. Dendrites of the neurons can also be subdivided into "light" and "dark" respectively. The surface of the former is comparatively simple in shape with a small number of appendages and spine-like structures. On the surface of the dark dendrites there are many projections and irregularly shaped lacunae. The glial cells and their processes often completely cover the surface of the soma of the small neurons, and synaptic endings are found on it only where the dendrites leave the soma. Analysis of 1000 randomly chosen synaptic endings showed that 76.1% of them form axo-dendritic synapses, 14.2% axo-somatic, and 9.7% axo-axonal synapses. Of the total number of endings 50.9% contain spherical and 40.9% flattened synaptic vesicles. Some synaptic endings contain special structures under the postsynaptic membrane and have osmiophilic synaptic vesicles. Nigrothalamic neurons were identified into thesubstantia nigra by their retrograde labelling with horseradish peroxidase. Axon terminals that contain glutamate (the excitatory transmitter) were revealed immunocytochemically with an immunogold electron microscopic technique. Ultrastructural parameters (the large and small diameters of axon terminals, area of their profiles, coefficient of form of profiles, large and small diameters of synaptic vesicles) were analyzed in all 240 synapses under study. Synaptic contacts localized on both nigrothalamic and unidentified neurons belonged to three morphologically specific groups. Synapses of the groups I and III, according to classification by Rinvik and Grofova, were characterized by a symmetric type of synaptic contact and contained polymorphic synaptic vesicles. Contacts in group-II synapses were asymmetric, and respective terminals contained round vesicles. Among the studied synapses, 65.8% were classified as group-I contacts, 25.0% belonged to group II, and 9.2% belonged to group III. Glutamate-positive axon terminals formed predominantly group-II synapses; such connections constituted 70% of this group's synapses. Sixty percent of glutamate-positive synapses were localized on the distal dendrites and 23% on the proximal dendrites, while 17% of such synapses were distributed on the somata of nigral neurons. Such a pattern of distribution of glutamate-positive synapses was observed on both nigrothalamic and unidentified nigral neurons. About 7% of glutamate-positive synapses were formed by very large axon terminals containing round synaptic vesicles; yet, the contacts of these terminals were of a symmetric type. In acute experiments on cats, the effects of injections of nitric oxide (NO) donors and an inhibitor of its synthesis into the sympathoexcitatory neuronal structures in the ventrolateral medulla (VLM) were studied to examine their effects on the peripheral mechanisms of the cardiovascular control. Unilateral injections of NO donors, nitroglycerine (1.3–5.2 nmol) or sodium nitroprusside (1.1–4.6 nmol) into the sites of the sympathoexcitatory neurons residing in the VLM induced the lowering of the systemic arterial pressure (SAP) in a dose-depended fashion. Two types of the hypotensive responses have been distinguished. In the first type responses, lowering of the SAP level was mainly due to a decrease in the peripheral vascular resistance (PVR), while the heart rate (HR) and stroke volume (SV) were only slightly reduced. In the second type responses, the drop in SAP level resulted mainly from a decrease in the HR and myocardial contractivity. These effects were induced by the limitation of the descending excitatory influences to the heart and vessels from the VLM sympathoexcitatory systems. An increase in the NO concentrations in the neuronal structures located 2.5–4.5 mm caudally to the trapezold bodies resulted in the first type responses, while that in the sites immediately adjacent to the caudal sympathoinhibitory area (0.5–1.5 mm rostrally to the XIIth cranial nerve roots) was associated with the second type of reactions. Stimulation of the endogenous NO release from the neurons after injections of L-arginine induced the same cardiovascular shifts as exogenic NO did, and attenuation of NO synthesis following injections of NO antagonist L-NMMA into the VLM neuronal structures evoked hemodynamic shifts of a reverse direction. Injections of NO donors inhibited the reflex responses induced by the activation of the carotid sinus receptors.
