Summary Developing oocytes of the newt Triturus cristatus were studied in order to clarify the role played by the Golgi apparatus in the formation of yolk. The cytochemical method used for this purpose was that of Maillet (1968) which employs an Osmium Zinc Iodide (OZI) complex.Previtellogenic oocytes reveal a pattern of OZI staining only after hormonal (HCG) stimulation, following which both the Golgi apparatus and the multivesicular bodies are stained.Vitellogenic oocytes taken from non-hormonally stimulated females reveal OZI deposits in a number of vesicles peripheral to the Golgi apparatus as well as within the superficial layer of the forming yolk platelets. Following hormone stimulation, many of the Golgi apparatus located in the central ooplasm of vitellogenic oocytes have all their cisternae blackened by the OZI deposits; other apparatuses, more peripherally located, remain essentially unchanged in their staining pattern. Further, a large number of OZI stained vesicles becomes visible in the vicinity of the Golgi apparatus and within the superficial layer of the forming yolk platelets.The present findings are interpreted as indicating the occurrence of fusion between Golgi derived vesicles and forming yolk platelets. A macrocytic hypocromic anemia was observed in newts after toluene inhalation, with 2 concentrations, for 6 weeks. Anemia resulted associated with the used dose of toluene. Erythroblast counts, performed on blood smears and on spleen and heart imprints, showed an erythropoietic stimulus during the first weeks of treatment followed by a depressed erythrostimulating action. Differential count of erythroblasts indicated a slow maturation induced by toluene on erythroblasts. An aplasia of the spleen was suggested on the basis of the results from 6 week-treated animals. A reduction in white blood cell count, associated with increased neutrophils and lymphocytopenia, occurred only in the animals treated with the higher concentration of toluene, after 5 and 6 weeks of treatment. A decrease of phagocytic activity of neutrophils, obtained with the in vitro assays, appeared the most relevant finding at the level of lekocyte compartment.  相似文献

Adult newts, Triturus viridescens, were treated with from 1.0–10.0 μg/g body weight of actinomycin D one day before amputation of both forelimbs. Mean survival times ranged from over 50 days in newts treated with 1.0 μg/g to 13.2 days in animals given 10.0 μg/g body weight of actinomycin. Low doses little altered the course of regeneration, but animals treated with over 2.0 μg/g never formed blastemas. In another series, animals were given doses of 2.5 μg/g body weight of actinomycin D at intervals from 14 days before to 30 days after amputation. It was found that certain signs of toxicity (loss of equilibrium) are related to the time of administration of the drug whereas others (hemorrhage into the limb stumps) are restricted to a definite phase of the regenerative process. Early administration of actinomycin completely inhibits regeneration whereas later treatment results in a considerably lessened effect. The postamputational stages which are basically destructive in nature are not noticeably affected by actinomycin D, but the phases of dedifferentiation, blastema formation and redifferentiation are strongly inhibited.  相似文献

Summary 1)As in mammals, the juxtaglomerular apparatus of the Newt (Triturus cristatus) is composed by cells of the media of the afferent glomerular arteriole and by cells of the intermediary tubule. 2) The cells of the media of the glomerular arteriole are of two different types: granular and agranular cells. 3) The intermediary tubule is formed by dark and light cells. 4) Part of interrenal body is located close to glomerular arteriole and intermediary tubule.This work was supported by grant of Consiglio Nazionale delle Ricerche of Italy (C.N.R.) The mitotic and labelling incidence of intestine, liver, spleen and pancreas cells of Triturus cristatus carnifex adults kept at 15°C, 20°C, 25°C and 30°C were examined. Intestine mitotic and labelling incidences were highest at 25°C and lowest at 30°C. There was no significant difference between 15°C and 20°C. No such relationship could be shown for liver, spleen or pancreas, which had very much lower mitotic and labelling incidences. In culture, intestine mitotic and labelling incidences fell significantly within the first four hours, and maintained these low levels for the next five days. In contrast, liver mitotic and labelling incidences rose for 9–11 days, and then began to fall, while pancreas mitotic and labelling incidences reached peak values at day 5, and were kept in good condition for up to 14 days.  相似文献
