A taxonomic study of the genus Padina from Japan, Southeast Asia, and Hawaii based on morphology and gene sequence data (rbcL and cox3) resulted in the recognition of four new species, that is, Padina macrophylla and Padina ishigakiensis from Ryukyu Islands, Japan; Padina maroensis from Hawaii; and Padina usoehtunii from Myanmar and Thailand. All species are bistratose and morphologically different from one another as well as from any known taxa by a combination of characters relating to degree of calcification; the structure, position, and arrangement of hairlines (HLs) and reproductive sori; and the presence or absence of rhizoid‐like groups of hairs and an indusium. Molecular phylogenetic analyses demonstrated a close relationship between P. ishigakiensis, P. macrophylla, P. maroensis, and Padina australis Hauck. The position of P. usoehtunii, however, was not fully resolved, being either sister to a clade comprising the other three new species and P. australis in the rbcL tree or more closely related to a clade comprising several other recently described species in the cox3 tree. The finding of the four new species demonstrates high species diversity particularly in southern Japan. The following characters were first recognized here to be useful for species delimitation: the presence or absence of small rhizoid‐like groups of hairs on the thallus surface, structure and arrangement of HLs on both surfaces either alternate or irregular, and arrangement of the alternating HLs between both surfaces in equal or unequal distance. The evolutionary trajectory of these and six other morphological characters used in species delineation was traced on the phylogenetic tree.  相似文献

sporophytes, and both male and female gametophytes of Dictyopteris undulata Holmes were collected from the Pacific coast and the Japan Sea coast of Japan. Female gametophytes are reported for the first time in the present study. All reproductive structures are aggregated in sori that form on both sides of the midrib in the upper and the middle parts of a blade. Tetraspor-angia are spherical and project from the cortical layer with a 2–4-celled stalk at the base. They are sometimes divided tetrahedrally to form four spores, but often remain undivided. Oogoniat sori also project from the cortical layer. Each oogonium is ovoid, sometimes borne on a single stalk cell at the base. Antheridial sori appear swollen above the cortex and each antheridium has a basal stalk cell. Fertile female gametophytes and te-trasporophytes are difficult to distinguish from each other in surface view because tetrasporangial and oogonial sori are similar in size and undivided tetrasporangia resemble oogonia. Oogonia are not so closely packed in small groups, unlike in Dictyota species, and they can be distinguished by the cell sizes (i.e. tetrasporangia are bigger in diameter than oogonia).  相似文献

The brown algal genus Padina (Dictyotales, Phaeophyceae) is distributed worldwide in tropical and temperate seas. Global species diversity and distribution ranges, however, remain largely unknown. Species‐level diversity was reassessed using DNA‐based, algorithmic species delineation techniques based on cox3 and rbcL sequence data from 221 specimens collected worldwide. This resulted in estimates ranging from 39 to 61 putative species (ESUs), depending on the technique as well as the locus. We discuss the merits, potential pitfalls, and evolutionary and biogeographic significance of algorithmic species delineation. We unveil patterns whereby ESUs are in all but one case restricted to either the Atlantic or Indo‐Pacific Ocean. Within ocean basins we find evidence for the vast majority of ESUs to be confined to a single marine realm. Exceptions, whereby ESUs span up to three realms, are located in the Indo‐Pacific Ocean. Patterns of range‐restricted species likely arise by repeated founder events and subsequent peripatric speciation, hypothesized to dominate speciation mechanisms for coastal marine organisms in the Indo‐Pacific. Using a three‐gene (cox3, psaA and rbcL), relaxed molecular clock phylogenetic analysis we estimated divergence times, providing a historical framework to interpret biogeographic patterns.  相似文献

Concerning the accumulation of S042‐ in cells, three types of species are known in the Dictyotales: (i) acidic type (high H2S04 accumulation); (ii) high MgS04 accumulation type; and (iii) nonacidic type (low S042‐ accumulation). Seasonal changes of intracellular pH and concentrations of inorganic ions were examined in six dictyotalean species. In acidic species (Dictyopteris prolifera (Okamura) Okamura and Spatoglossum eras‐sum J. Tanaka), intracellular concentrations of S042‐ and H+ estimated by pH were high through all seasons. In high MgS04 accumulating species (Dictyotasp. and Padina arborescens Holmes), intracellular concentrations of S042‐ and Mg2+ were high through all seasons. In nonacidic species (Dictyota dichotoma (Hudson) Lamouroux and Dictyopteris undulata Holmes), intracellular concentration of sulfuric acid ion was low all year round.  相似文献

Considering the lack of knowledge on genetic variation on members of the freshwater red algal of the order Batrachospermales in tropical regions, phylogeographic patterns in Sirodotia populations were investigated using two mitochondrial regions: the cox2‐3 spacer and partial cox1 gene (barcode). Individuals identified as Sirodotia delicatula were analyzed from 14 stream segments across its distribution in Brazil. Phylogenetic analyses based on the ribulose‐1,5‐bisphosphate carboxylase/oxygenase large sub‐unit gene showed three clades, one representing S. delicatula, from all locations in southeastern Brazil and other regions from Brazil. The remaining samples formed two clades, which were highly divergent and distantly positioned from those of S. delicatula: 2.5–2.7% and 3.4–3.7%. This level of variation would warrant the species split of these taxa from mid‐western Brazil. A total of eight cox2‐3 spacer and nine cox1 haplotypes were observed among the 122 individuals studied. One location had two cox2‐3 haplotypes and three locations had two cox1 haplotypes; all others had a single dominant haplotype each. The existence of high intraspecific genetic variation among individuals of distinct locations (several haplotypes), but little variation within a location seems to be a pattern for the Batrachospermales. Haplotype networks showed low variation among the haplotypes from southeastern Brazil (10 locations with divergence of 0.3–1.1% for cox2‐3, 0.1–0.3% for cox1) and high variation among the haplotypes from the mid‐west region (four locations, 4.0–9.3% for cox2‐3, 6.2–8.4% for cox1). Thus, the present data clearly suggest the existence of cryptic species in Sirodotia in Brazil.  相似文献

Lobophora is a common tropical to temperate genus of brown algae found in a plethora of habitats including shallow and deep‐water coral reefs, rocky shores, mangroves, seagrass beds, and rhodoliths beds. Recent molecular studies have revealed that Lobophora species diversity has been severely underestimated. Current estimates of the species numbers range from 100 to 140 species with a suggested center of diversity in the Central Indo‐Pacific. This study used three molecular markers (cox3, rbcL, psbA), different single‐marker species delimitation methods (GMYC, ABGD, PTP), and morphological evidence to evaluate Lobophora species diversity in the Western Atlantic and the Eastern Pacific oceans. Cox3 provided the greatest number of primary species hypotheses(PSH), followed by rbcL and then psbA. GMYC species delimitation analysis was the most conservative across all three markers, followed by PTP, and then ABGD. The most informative diagnostic morphological characters were thallus thickness and number of cell layers in both the medulla and the dorsal/ventral cortices. Following a consensus approach, 14 distinct Lobophora species were identified in the Western Atlantic and five in the Eastern Pacific. Eight new species from these two oceans were herein described: L. adpressa sp. nov., L. cocoensis sp. nov., L. colombiana sp. nov., L. crispata sp. nov., L. delicata sp. nov., L. dispersa sp. nov., L. panamensis sp. nov., and L. tortugensis sp. nov. This study showed that the best approach to confidently identify Lobophora species is to analyze DNA sequences (preferably cox3 and rbcL) followed by comparative morphological and geographical assessment.  相似文献

Ten species of Sargassum (Sargassaceae, Phaeophyceae) were found along the Gulf of Thailand. Morphological characteristics of Sargassum baccularia (Mertens) C.A. Agardh, S. binderi Sonder, S. cinereum J.G. Agardh, S.crassifolium J.G. Agardh, S. longifructum Tseng et Lu, S. oligocystum Montagne, S. polycystum C.A. Agardh, S. siliquosum J.G. Agardh, S. swartzii (Turner) C.A. Agardh and one unidentified species were examined and are described in detail. The most common species were S. polycystum distributed widely in almost all the study sites, S. crassifolium restricted to Prachuap Khirikhan Province, S. longifructum restricted to Chumphon Province, S. siliquosum restricted to Surat Thani Province and one unidentified species restricted to Songkhla Province. Three species (S. cinereum, S. longifructum and S. swartzii) are new records for the algal flora of Thailand. Five species (S. baccularia, S. cinereum, S. longifructum, S. polycystum and the unidentified species) belong to the section Zygocarpicae (J.G. Agardh) Setchell.  相似文献

Colpomenia borea sp. nov. is described from Hokkaido, Japan and Magadan, Far East Russia based on morphological observations and molecular analyses using mitochondrial cox3 and chloroplast rbcL genes. This new species is distinguished from other Colpomenia by its small globular to ovoidal thallus up to 5 cm in diameter, and thin thallus membrane composed of a cortex of one to two-cell layers and a colorless medulla of up to three layers. This species is epiphytic on the brown alga Stephanocystis in areas protected against waves. The life history in culture of C. borea was investigated and the observed life history pattern was similar to those reported in C. peregrina and C. sinuosa. Our phylogenetic analyses supported that C. borea is a distinct species, yet it is more related to C. peregrina than C. sinuosa.  相似文献

Scytosiphon lomentaria (Scytosiphonaceae, Ectocarpales) is believed to include some cryptic species, particularly in the Pacific. We attempted to delimit these species in Japan using mitochondrial cox1 and cox3 and nuclear ITS2 and the second intron of the centrin gene (cetn‐int2). Fifty‐three cox1+cox3 mitotypes, 26 ITS2 ribotypes and 45 cetn‐int2 haplotypes were found in 107 samples collected from 33 localities in Japan. Based on phylogenetic analyses, similar sequence types were grouped into ten mitogroups, eight ribogroups and six cetn‐int2 haplogroups (sequence‐type groups). From the molecular trees and combinations of the mito‐, ribo‐ and haplogroups, three cryptic species were apparent (Groups I–III). Group I, widely distributed on Pacific coasts, was highly supported by all molecular trees, whereas Groups II (North Pacific) and III (Northwestern Pacific and Australasia) were more closely related to each other. However, sequence‐type‐group combinations that would be characteristic of hybrids between Groups II and III were not detected, suggesting no gene flow between the two Groups. Further investigations of an additional 127 sympatrically growing plants supported the absence of gene flow between Groups II and III. Four samples did not belong to any of the Groups I–III and possibly represent additional species.  相似文献

The brown algal genus, Padina, has a worldwide distribution in tropical and subtropical climate zones. Padina individuals are common and sometimes dominant in both the intertidal and shallow subtidal regions associated with coral reefs. We investigated the population structure and recruitment of two populations of Padina boryana at an exposed shore in Sirinart National Park (SNP) and a sheltered shore of Tang Khen Bay (TKB), Phuket Province, Thailand. From September 2005 to August 2006, the number of released spores and the height and radius of fronds were measured monthly. New recruits were recorded and monitored on manipulated permanent plots 0.25 m2. The experiment was carried out monthly over a year. We found Padina recruits 1 month after the plots were cleared at both sites. There was a significant difference in percentage cover by new individuals between the two locations (P < 0.05) and also at the shore levels within the sites. At SNP, the highest recruitment was found in the uppermost zone, while at TKB high recruitment occurred at all shore levels except at 80-100 m from the shore. The factors influencing recruitment of P. boryana include wave motion, competition with other earlier successional species and sediment. The high availability of reproductive spores throughout the year, the successful recruitment and the Dictyerpa stage promote the successful establishment of Padina populations.  相似文献

The rhodophyte seaweed Asparagopsis armata Harvey is distributed in the northern and southern temperate zones, and its congener Asparagopsis taxiformis (Delile) Trevisan abounds throughout the tropics and subtropics. Here, we determine intraspecific phylogeographic patterns to compare potential causes of the disjunctions in the distributions of both species. We obtained specimens throughout their ranges and inferred phylogenies from the hypervariable domains D1-D3 of the nuclear rDNA LSU, the plastid spacer between the large and small subunits of RuBisCo and the mitochondrial cox 2-3 intergenic spacer. The cox spacer acquired base changes the fastest and the RuBisCo spacer the slowest. Median-joining networks inferred from the sequences revealed the absence of phylogeographic structure in the introduced range of A. armata, corroborating the species' reported recent introduction. A. taxiformis consisted of three nuclear, three plastid and four mitochondrial genetically distinct, lineages (1-4). Mitochondrial lineage 3 is found in the western Atlantic, the Canary Islands and the eastern Mediterranean. Mitochondrial lineages 1, 2, and 4 occur in the Indo-Pacific, but one of them (lineage 2) is also found in the central Mediterranean and southern Portugal. Phylogeographic results suggest separation of Atlantic and Indo-Pacific lineages resulted from the emergence of the Isthmus of Panama, as well as from dispersal events postdating the closure event, such as the invasion of the Mediterranean Sea by mitochondrial lineages 2 and 3. Molecular clock estimates using the Panama closure event as a calibration for the split of lineages 3 and 4 suggest that A. taxiformis diverged into two main cryptic species (1 + 2 and 3 + 4) about 3.2-5.5 million years ago (Ma), and that the separation of the mitochondrial lineages 1 and 2 occurred 1-2.3 Ma.  相似文献

The use of cholinesterase inhibitors and antioxidants is currently considered as an effective therapeutic strategy for the treatment of Alzheimer’s disease (AD). There is also a growing trend to use nutraceuticals for cognitive impairment. Since natural product-based drugs offer better hope for the treatment of AD, the present study was aimed at evaluating the nutritional profile of the brown seaweed Padina gymnospora and investigating the antioxidant and inhibitory effect of different solvent extracts of P. gymnospora on acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) enzymes. The nutritional profile of P. gymnospora includes large amounts of carbohydrate, protein, lipid, proline, chlorophyll, fibre, minerals, fatty acids and amino acids, along with antioxidants such as vitamins C and E. Antioxidant activities of various solvent extracts of P. gymnospora were assessed by the DPPH scavenging assay, nitric oxide scavenging assay, reducing power assay and FRAP assay. Acetone extracts showed significant DPPH radical and nitric oxide scavenging activity with IC50 values of 402 ± 9.12 and 441 ± 48.16 µg ml–1 respectively and aqueous extracts had better reducing potential. Among the different solvent extracts, the acetone extract showed the highest inhibitory activity with IC50 values <150 µg ml–1 for both AChE and BuChE. Qualitative phytochemical screening of P. gymnospora revealed the presence of flavonoids and cardiac glycosides. Overall the results suggest that the seaweed Padina gymnospora may be a good nutraceutical candidate for discovering drugs against AD.  相似文献

In depth genetic comparisons of populations of Cutleria multifida (Tilopteridales, Phaeophyceae) collected from Europe, the northwestern Pacific Ocean, Australia and New Zealand using the DNA sequences of four gene regions (the mitochondrial cox2 and cox3 genes, the intergeneric spacer region adjacent to cox3, and the open reading frame) suggested that the northwestern European and Japanese populations were considerably greater in terms of their genetic divergence than Mediterranean, Australian or New Zealand populations. The haplotypes of the populations in northwestern European (distribution range including the type locality, seven haplotypes) and Japanese populations (seven haplotypes) were unique except for one shared haplotype. There were weak but positive correlations between the geographical distance and the genetic divergence among northwestern European and Japanese populations. Moreover, both female and male gametophytes occurred in eight of the nine Japanese localities, suggesting Japanese populations showed normal sexual heteromorphic life history of the species. In light of these results, it appears that Japanese populations were native to the area despite earlier hypothesis. In contrast, Australian and New Zealand populations were composed of only one haplotype that is very close to those found in northwestern Europe and Japan, suggesting a recent introduction history from Europe (or from northeastern Asia via Europe) by ship transport to Australia and New Zealand. The Mediterranean populations included two haplotypes identical to those found in northwestern Europe and Japan, and it is suggestive of transoceanic introductions of some populations between Mediterranean and Japanese coasts.  相似文献

Genetic relationships among Undaria species and among populations of each species were studied based on DNA sequences of the mitochondrial cox3 gene. Although three Undaria species, U. peterseniana (Kjellman) Okamura, U. pinnatifida (Harvey) Suringar and U. undarioides (Yendo) Okamura, have been described based mostly on blade morphology, plants with intermediate morphologies have also been found. Multiple plants from several populations in Japan were collected. Morphological characters could identify most of the samples unambiguously. A few samples with intermediate morphologies were also collected. Mitochondrial haplotypes found in each population were different for each identified species, and each species had multiple haplotypes. In the cox3 haplotype network analysis, the numbers of steps between haplotypes within and between species were similar, and haplotypes of each species did not group together. The close genetic relationships among species strongly suggest that these species are conspecific. Alternatively, recent speciation could be possible with maintenance of ancestral polymorphisms within the species (i.e. incomplete lineage sorting). Haplotypes of samples with intermediate morphologies were different for each sample and the same as ones found in the local population, suggesting interspecific hybridizations among species.  相似文献

Cutleria cylindrica Okamura was described from Japan in 1902 and has been reported only from northwestern Asia until its relatively recent discovery in California, USA, and Baja California, Mexico. To clarify the genetic relationships within and among the disjunct populations, we carried out a molecular phylogenetic study, as well as the examination of sex ratio and the life‐history patterns, of populations in Japan, Korea, and California. Based on the DNA sequences of mitochondrial genes cox2, cox3, the open reading frame (ORF) region, and the spacer between cox3 and ORF, a total of 23 haplotypes were detected in the 85 individuals from 20 localities in Japan, Korea, and California. All localities in Japan and Korea included multiple haplotypes, but only a single haplotype was found in California. There was a positive relationship between distance and genetic divergence in Japan and Korea. The single haplotype found in California was the same as one occurring in Japan (Aomori Pref. and Fukuoka Pref.) and Korea (Daedaepo, Pusan). Both male and female gametophytes were distributed in most northeastern Asian populations. Only female gametophytes, developing parthenogenetically from female gametes, were found in California and Aomori Pref., Japan. On the basis of these results, we conclude that the disjunct population of C. cylindrica in California originated from a relatively recent introduction from Japan and shares its origin with the parthenogenetic population in the Tsugaru Strait.  相似文献

The taxonomic and phylogenetic concepts within the Sargassum C. Agardh (Sargassaceae) species complex were evaluated through molecular phylogenetic analyses using portions of the chloroplast encoded rbcLS Operon. According to more conservative sequences (rbcL), Turbinaria (Turner) J. Agardh is a close and well‐supported sister lineage to the Sargassum species complex and an appropriate external outgroup for analyses of subgenera and subsections within Sargassum. Both rbcL and more rapidly evolving rbcLS spacer sequences indicated that the East Asiatic genus Myagropsis (Mertens et Turner) Fensholt, along with Sargassum sinicola Setchell et Gardner, represent the closest lineage to Sargassum and form appropriate internal outgroups. The rbcLS spacer region supported three of four subgeneric designations by J. Agardh and sectional levels within the subgenus Sargassum. However, some aspects of Agardh's system were not supported: many of the subsectional ranks or the phyletic concepts; Phyllotrichia was not monophyletic as a subgenus, and its species were also not the most ancestral of Sargassum; and subgenus Sargassum was not the most derived subgenus within the genus. This modern phylogeny suggests a deep evolutionary history for subgenus Sargassum with rapid speciation in closely related subsections and series, and a sister relationship between subgenera Arthrophycus and Bactrophycus.  相似文献
