Suspensions of the yeast Saccharomyces cerevisiae gave reproducible rates of O2 uptake over a period of 6 months. The relation of rate of consumption of O2 to temperature was tested over a wide range of temperatures, and the constant in the formulation of the relationship is found to be reproducible. The values of this constant (µ) have been obtained for five separate series of experiments by three methods of estimation. The variability of µ has the following magnitudes: the average deviation of a single determination expressed as per cent of the mean is ±2 per cent in the range 30–15°, and ±0.8 per cent in the range 15–3°C. This constancy of metabolic activity measured as a function of temperature can then be utilized for more precise investigations of processes controlling the velocity of oxidations of substrates, and of respiratory systems controlled by intracellular respiratory pigments. The data plotted according to the Arrhemus equation give average values of the constant µ as follows: for the range 35–30°, µ = 8,290; 30–15°, µ = 12,440 ±290; 15–3°, µ = 19,530 ±154. The critical temperatures are at 29.0° and 15.7°C. A close similarity exists between these temperature characteristics (µ) and values in the series usually obtained for respiratory activities in other organisms. This fact supports the view that a common system of processes controls the velocities of physiological activities in yeast and in other organisms.  相似文献

Young mice of a selected line of the dilute brown strain of mice exhibit over the range 15–25°C. (body temperature) a relation of frequency of breathing movements to temperature such that when fitted by the Arrhenius equation the data give a value for the constant µ of 24,000± calories or, less frequently, 28,000±. Young mice of an inbred albino strain show over the range 15–20°C. a value of µ = 34,000±, or, less frequently, 14,000±, with a critical temperature at about 20°C. and a value of µ = 14,000± above 20°C. The F1 hybrids of these two strains, and the backcross generations to either parent strain, exhibit only those four values of the temperature characteristic observed in the parent strains and none other. One may therefore speak of the inheritance of the value of the constant µ, but the inheritance shows in this instance no Mendelian behavior. Furthermore there appears to be inherited the occurrence (or absence) of a critical temperature at 20°C. These experiments indicate the "biological reality" of the temperature characteristics.  相似文献

γ-Guanidinobutyraldehyde dehydrogenase was purified 27-fold in 40% yield from extracts of Vicia faba leaves. High specificity exist only for γ-guanidinobutyraldehyde and γ-aminobutyraldehyde; the Km value was 3.4 micromolar for γ-guanidinobutyraldehyde, 25 micromolar for γ-aminobutyraldehyde, and 84 micromolar (case of γ-guanidinobutyraldehyde) for NAD, respectively. The enzyme had a molecular weight of approximately 83,000. Optimal pH and temperature for activity were 9.5 and 45°C, respectively. The enzyme was inhibited strongly by p-chloromercuribenzoate, N-ethylmaleimide, and zincon (2-carboxy-2′-hydroxy-5′-sulfoformazylbenzene).  相似文献

The temperature characteristic for respiratory production of CO2 by young seedlings of Phaseolus aureus (Roxb.) is µ = 16,500 calories, 12–21°C., even when the analyses depend upon the use of many seedlings crowded in a small respiration chamber, provided reasonable precautions are taken to avoid injury and to permit proper thermal adaptation. There is evidence of a definite critical temperature at 20–21°. These findings agree quantitatively with those obtained with other similar seedlings, and contradict the results reported by Kurbatov and Leonov (1930); the reasons for this are analysed.  相似文献

The rate of dry matter accumulation by seeds of Vicia faba L. cv. Minica increases with temperature in the range of 16 to 26°C. The duration of dry matter accumulation decreases with temperature, resulting in a decrease of final seed dry weight. In this study we test the hypothesis that a diffusion barrier for O2, located in the seed coat, inhibits seed respiration and growth. The rate of O2 uptake of intact seeds and of excised embryos and seed coats (separated seeds) was measured in air and buffer at 16, 20, and/or 26°C at various O2 concentrations and developmental stages. Oxygen uptake rates of intact seeds in buffer were only 9 to 15% of those in air. In buffer, the respiration rate of intact seeds decreased at a pO2 below air saturation (21 kilopascals), whereas separated seeds showed a decline of O2 uptake only below 80% of air saturation. In air, embryo excision had no effect on the sensitivity of seed respiration to pO2, at both 20 and 26°C. In air at 20°C, separated and intact seeds showed similar rates of O2 uptake. Oxygen uptake by intact seeds, both halfway and beyond the linear growth phase, showed a temperature coefficient Q10 of 2.3 and was insensitive to pO2 in the range of 80 to 100% of ambient. These results indicate that V. faba seed respiration in air is not limited by the diffusion of O2 into the seed.  相似文献

The distribution of plastoquinones A 45, B and C was studied in representatives from 34 different plant families beginning with liverworts and mosses to higher plants. All of these species, including many monocots and dicots, contained significant amounts of the 3 quinones. Two species of Aesculus contained plastoquinone A 20 in addition to plastoquinone A 45, B, and C. Many dicots, such as Aesculus, watermelon, tobacco and tomato accumulated increasing quantities of plastoquinones A and C1-C4 during the growing season. The concentrations of plastoquinones B and C5-C6 tended to remain at a constant low level during the season (<0.01 μmole per mg chlorophyll). Preliminary studies with bean plants (Vicia faba and Phaseolus sp.) indicate that the levels of quinones varied little under different growth conditions (day length and temp.) although Vicia faba tended to have higher PQ A values with increased temperature.  相似文献

The temperature characteristic for the rate of O2 consumption by Chlorella pyrenoidosa suspended in Knop solution containing 1 per cent glucose was studied between 1° and 27°C. with the Warburg technic. The value of µ was found to be about 19,000 ±1,000 cal. There is some indication of a critical temperature at 20°C., with shift to a lower µ above this temperature. The effect of sudden changes in temperature on the rate of respiration and the variation of the latter with time at constant temperatures are discussed. It is concluded that the "normal" respiration (in absence of external glucose) does not appear in the determination of this temperature characteristic.  相似文献

The speed of translatory movement of Beggiatoa alba is governed by temperature in such a way that between 5° and 33° the temperature characteristics µ = 16,100 and µ = 8,400 respectively obtain for the temperature ranges 5° to 16.5° and 16.5° to 33°. The "break" at 16°–17° is emphasized by the occurrence of a wider latitude of variation in speed above this temperature. Above 16° the progression of Thiothrix yields µ = 8,300. The possible relation of these values to that previously obtained for similar movement in (photosynthetic) Oscillatoria is commented upon.  相似文献

Stomata are microscopic pores in leaf epidermis that regulate gas exchange between plants and the environment. Being natural openings on the leaf surface, stomata also serve as ports for the invasion of foliar pathogenic bacteria. Each stomatal pore is enclosed by a pair of guard cells that are able to sense a wide spectrum of biotic and abiotic stresses and respond by precisely adjusting the pore width. However, it is not clear whether stomatal responses to simultaneously imposed biotic and abiotic signals are mutually dependent on each other. Here we show that a genetically engineered Escherichia coli strain DH5α could trigger stomatal closure in Vicia faba, an innate immune response that might depend on NADPH oxidase-mediated ROS burst. DH5α-induced stomatal closure could be abolished or disguised under certain environmental conditions like low [CO2], darkness, and drought, etc. Foliar spraying of high concentrations of ABA could reduce stomatal aperture in high humidity-treated faba bean plants. Consistently, the aggressive multiplication of DH5α bacteria in Vicia faba leaves under high humidity could be alleviated by exogenous application of ABA. Our data suggest that a successful colonization of bacteria on the leaf surface is correlated with stomatal aperture regulation by a specific set of environmental factors.  相似文献

The observation was made previously that the reduction in radiosensitivity in Vicia faba (as measured by postirradiation root growth) by prolonging the exposure time from about 10 minutes to 24 hours is much less marked at 3°C. than at 19°C. If chromosome damage is mainly responsible for the reduced root growth, this observation might be explained by a smaller drop in the "two-hit" aberration component, resulting from an increased time for which breaks are available for rejoining at 3°C. This hypothesis was tested by comparing chromatid aberration frequencies in root meristem cells produced by 105 rads of 60Co γ rays, given at dose rates of 19.4 and 0.073 rads per minute. Beans were maintained in aerated water at 2°C. prior to and during irradiation, and at this temperature the rate of development of cells was such that the two different exposure times both occupied a period during which the cell sensitivity was approximately constant. Immediately subsequent to irradiation, the roots were returned to 19°C. and examined cytologically. All chromatid aberrations were less frequent after low dose rate treatment, but only the chromatid interchange reduction was significant. The average time for which breaks are available for reunion, calculated from Lea's G function, was found to be 12 hours (95 per cent C.L. 6 to 24 hours).  相似文献

Pedal progression of the slug Limax maximus was studied to obtain relations between wave velocity on the sole of the foot, wave frequency, the advance due to a single wave, and the velocity of vertically upward creeping. Each of the first three quantities is directly proportional to the simultaneous velocity of progression. Under comparable conditions, that is when work is done at a constant rate, the frequency of pedal waves is influenced by the temperature according to the equation of Arrhenius, with µ = 10,700 (Q 10 for 11° to 21° = 2.1). The velocity of a single wave must have very nearly the same "temperature characteristic," which is found also in another case of nerve net transmission (in Renilla).  相似文献

Rates of development of stem nematode (Ditylenchus dipsaci) in white clover (Trifolium repens) seedlings were found to be linearly related to temperature. Basal developmental temperature (Tb) was 3 °C, and the thermal constant (S) for development of gravid adult females from freshly laid eggs was 270 accumulated day-degrees above the Tb. Only 12% at 20 °C and 4% at 4 °C of the gravid female nematodes inoculated into seedling axils successfully penetrated seedling epidermis. These nematodes slowly migrated within the seedling and after a lag of 5 days at 20 °C started to lay eggs. The maximal rate of egg production was temperature-dependent, being 0.8 and 3.1 eggs female⁻¹ day⁻¹ at 10 and 20 °C, respectively. Nematodes emigrated rapidly from infested stolons when they were immersed in water, with rates being highest at 25 °C and lowest at 4 °C. The sensitivity to temperature of many of the parameters that govern nematode population dynamics indicates that climatic changes will have a marked effect upon this host-parasite system. A study of infested stolons from the field indicated that nematode numbers increased up to 3,000 or more before tissue senesence, triggered by nematode damage, caused a mass emigration of nematodes from the stolon.  相似文献

Temperature effects on nocturnal carbon gain and nocturnal acid accumulation were studied in three species of plants exhibiting Crassulacean acid metabolism: Mamillaria woodsii, Opuntia vulgaris, and Kalanchoë daigremontiana. Under conditions of high soil moisture, nocturnal CO2 gain and acid accumulation had temperature optima at 15 to 20°C. Between 5 and 15°C, uptake of atmospheric CO2 largely accounted for acid accumulation. At higher tissue temperatures, acid accumulation exceeded net carbon gain indicating that acid synthesis was partly due to recycling of respiratory CO2. When plants were kept in CO2-free air, acid accumulation based on respiratory CO2 was highest at 25 to 35°C. Net acid synthesis occurred up to 45°C, although the nocturnal carbon balance became largely negative above 25 to 35°C. Under conditions of water stress, net CO2 exchange and nocturnal acid accumulation were reduced. Acid accumulation was proportionally more decreased at low than at high temperatures. Acid accumulation was either similar over the whole temperature range (5-45°C) or showed an optimum at high temperatures, although net carbon balance became very negative with increasing tissue temperatures. Conservation of carbon by recycling respiratory CO2 was temperature dependent. At 30°C, about 80% of the dark respiratory CO2 was conserved by dark CO2 fixation, in both well irrigated and water stressed plants.  相似文献

1. The relation of temperature to the pedal rhythm of Balanus balanoides L. has been studied under otherwise constant conditions. 2. The frequency of movement increases with temperature, showing three groups of thermal increments and three critical temperatures. Five animals yielded µ = 5,700 above 14.5° C. and 12,100 below; 3 gave µ = 7,800 above 9.3° and 22,500 below; while 9 showed µ = 9,500 above 8.1° and 22,100 below. 3. The upper critical temperatures, above which different effects appeared in different animals were 23.4°, 26.0°, and 27.0°. Above 27.0° none of the valves remained open. 4. Excepting the values 5,700 and 9,500, the increments are similar to those previously found to be associated with respiratory and with neuromuscular activities. 5. Dilution of the sea water with from 3 to 4 per cent fresh water decreases the rate without altering the increments. More than 4 per cent dilution causes irregularity.  相似文献

1. The sulfonium salt H·2TDG is formed when H is mixed with even dilute solutions of TDG. Crystalline H·2TDG was isolated from such a reaction mixture. A simple method of preparation of this salt is outlined. 2. A material which differs from H·2TDG in that it hydrolyzes faster, is formed when H hydrolyzes in water. This material is probably H·1TDG but it was not isolated. Approximately 5 to 8 per cent of the original H is converted to this sulfonium salt. 3. The hydrolysis constant of M/100 H·2TDG has been determined at 20°, 25.5°, 37°, 75°, and 100°C., a temperature coefficient, Q 10, of 3–4 was obtained. The effect of temperature is in agreement with that predicted by the Arrhenius equation. An activation energy of 26,000 calories was calculated.  相似文献

At fixed flash frequency (F = 20, F = 55) and with constant light time fraction (50 per cent) in the flash cycle, the critical illumination I for response of Anax nymphs to visual flicker falls continuously as the temperature rises. The temperature characteristic µ for the measure of excitability (1/I) increases continuously with elevation of temperature. The form of the F - log I curve does not change except at quite high temperature (35.8°), and then only slightly (near F = 55); Fmax. is not altered. The very unusual form of the 1/I curve as a function of temperature is quantitatively accounted for if two processes, with respectively µ = 19,200 and µ = 3,400, contribute independently and simultaneously to the control of the speed of the reaction governing the excitability; the velocities of these two processes are equal at 15.9°.  相似文献

The translational efficiency of wheat ribosomes was studied as a function of an in vivo temperature pretreatment of wheat seedlings (Triticum aestivum L.). Ribosomes were isolated from heat-pretreated (36°C) and reference (4°C, 20°C) wheat seedlings. The efficiency of the ribosomes in translating polyuridylic acid was assayed. Ribosomes from heat-pretreated seedlings exhibit a threefold enhanced incorporation rate of phenylalanine as compared to ribosomes from wheat seedlings adapted to 20 or 4°C. This difference develops within 24 hours after onset of the heat treatment of seedlings following a 3 hour lag phase. The temperature induced changes can be traced back to the cytoplasmic ribosomes, since cycloheximide inhibits translation almost completely. Thermal inactivation of ribosomes occurs at 45°C, irrespective of the temperature pretreatment of the wheat seedlings. Specific differences in the yield of ribosomes, in the polyribosomal profiles, and in the apparent Arrhenius' activation energy of protein synthesis were observed depending on the age and the temperature pretreatments. The results presented here are considered an important molecular correlation to phenotypical temperature adaptation of in vivo protein synthesis in wheat (M Weidner, C Mathée, FK Schmitz 1982 Plant Physiol 69: 1281-1288).  相似文献

In an isometric tetanus in frog's sartorius muscle tension approaches the plateau exponentially with rate constant α. α a depends on sarcomere length, s, and temperature, T, according to the Arrhenius equation See PDF for Equation for temperatures between 1 and 20°C and for sarcomere lengths 2.0–2.8 µm. The energy of activation, E, does not vary significantly with s; E = 13.9 ± 2.4 kcal/mole. A(s) decreases monotonically with s; A(2.1 µm) is about three times greater than A(2.8 µm). Late in relaxation active tension approaches zero exponentially with rate constant r. r decreases exponentially with increasing duration of tetanus, D, from r0 in a twitch to r for large D. The rate constant for decrease of r with D increases with s and with T. r0 and r obey the Arrhenius equation and decrease with increasing s.  相似文献

A spring wheat (Triticum aestivum) and an obligate winter wheat (Triticum compactum) variety were each grown for 5 weeks in controlled environments at 2° and 25°. The threshold for flower induction in the winter wheat was 4 to 5 weeks at 2°, whereas the spring wheat had no low temperature requirement for flowering. Changes in the levels of carbohydrate and nitrogen fractions in the wheat leaves were determined during their growth in the cold and warm environments. There was an enhanced accumulation of the 5 carbohydrate fractions in both wheat varieties grown at 2° compared to 25°. Highly significant differences in the levels of sucrose, oligosaccharides, and starch were found between the spring and winter varieties grown at 2°. The winter wheat seedlings grown at 2° accumulated much more of these carbohydrates than the corresponding spring wheat. The carbohydrate patterns in both varieties grown at 25° were nearly identical except for the final 2 weeks of growth.

The level of nitrogenous substances in the tissues grown at 2° was much higher than in the corresponding tissues grown at 25°. The only significant difference between the spring and winter varieties was in the soluble protein fraction. This fraction rose nearly 3-fold in the winter variety grown at 2°, whereas it remained nearly constant in the similarly grown spring wheat. Most of the changing chemical patterns observed in relation to the vernalization treatment appear to be metabolic alterations associated with low temperature rather than alterations directly related with the vernalization response.
