Successful implementation of homogeneous slice-selective RF excitation in the human brain at 9.4T using 16-channel parallel transmission (pTX) is demonstrated. A novel three-step pulse design method incorporating fast real-time measurement of eddy current induced B0 variations as well as correction of resulting phase errors during excitation is described. To demonstrate the utility of the proposed method, phantom and in-vivo experiments targeting a uniform excitation in an axial slice were conducted using two-spoke pTX pulses. Even with the pre-emphasis activated, eddy current induced B0 variations with peak-to-peak values greater than 4 kHz were observed on our system during the rapid switches of slice selective gradients. This large B0 variation, when not corrected, resulted in drastically degraded excitation fidelity with the coefficient of variation (CV) of the flip angle calculated for the region of interest being large (∼12% in the phantom and ∼35% in the brain). By comparison, excitation fidelity was effectively restored, and satisfactory flip angle uniformity was achieved when using the proposed method, with the CV value reduced to ∼3% in the phantom and ∼8% in the brain. Additionally, experimental results were in good agreement with the numerical predictions obtained from Bloch simulations. Slice-selective flip angle homogenization in the human brain at 9.4T using 16-channel 3D spoke pTX pulses is achievable despite of large eddy current induced excitation phase errors; correcting for the latter was critical in this success.  相似文献

If it is assumed that the kinetics of the process of excitation in nerve is given by dp/dt = KI – kp, I being the actual exciting component of the current, p the state of excitation, and K and k constants, it is necessary to postulate that on application of a rectangular stimulus of voltage, V, the current, I, undergoes a transient exponential variation, usually a decrease, in order that the integral of the differential equation (above) may fit the strength-duration data in V and t. This hypothesis is substantiated by data by Sakamoto on single fibers of the sciatic nerve of the frog. The time constant of the postulated current transient is of the order of 10–4 sec. for single fibers and of the order of 10–5 sec. or less in the sciatic nerve trunk. The latter value is about the same as that found by Cole in the same tissue by purely physical measurements. Some criticisms by Rushton (1934) are discussed.  相似文献

Experimental data elucidating mechanisms for halo formation in θ-pinch discharges are presented and discussed. The experiments were performed with different gases (H2, D2, He, and Ar) in a theta-pinch device with a porcelain vacuum chamber and an excitation coil 15 cm in diameter and 30 cm in length. The stored energy, the current in the excitation coil, and the current half-period were W = 10 kJ, I = 400 kA, and T/2 = 14 μs, respectively. It is found that the plasma rings (halos) surrounding the pinch core arise as a result of coaxial pinch stratification due to both the excitation of closed currents (inductons) inside the pinch and the radial convergence of the plasma current sheaths produced after the explosion of T-layers formed near the wall in the initial stage of the discharge. It is concluded that halo structures observed in pinches, tokamaks, and other high-current devices used in controlled fusion research have the same nature.  相似文献

Eccentric cells of Limulus respond with repetitive firing to sustained depolarizing currents. Following stimulation with a step of current, latency is shorter than first interval and later intervals increase progressively. A shock of intensity twice threshold can evoke firing 25 msec. after an impulse. But in the same cell, a current step twice rheobase evokes a second impulse more than 50 msec. after the first, and current intensity must be raised to over five times rheobase to obtain a first interval of about 25 msec. Repetitive firing was evoked by means of trains of shocks. With stimuli of moderate intensity, firing was evoked by only some of the shocks and intervals between successive impulses increased with time. This is ascribed to accumulation of refractoriness with successive impulses. Higher frequencies of firing are obtained with shocks of intensity n x threshold than with constant currents of intensity n x rheobase. It is concluded that prolonged currents depress the processes leading to excitation and that (in the cells studied) repetitive firing is controlled both by the after-effects of firing (refractoriness) and by the depressant effects of sustained stimuli (accommodation). Development of subthreshold "graded activity" is an important process leading to excitation of eccentric cells, but is not the principal factor determining frequency of firing in response to constant currents.  相似文献

Accommodation and excitation in space-clamped squid axons were studied with the double sucrose gap technique, using linear current ramps, short (50 µsec) square wave pulses, and rheobasic square wave pulses as stimuli. The temperature was varied from 5° to 35°C. Experimental results showed a Q10 for accommodation which was 44% higher than that for excitation. Yet calculations on the basis of the Hodgkin-Huxley equations predict equal Q10's for excitation and accommodation. Although the Hodgkin-Huxley equations are spectacularly successful for so many nerve phenomena, the differences between calculations of accommodation and these experiments, which were designed to test the equations, show that the equations need modification in this area.  相似文献

The current generally accepted structure of light-harvesting LH2 complexes from purple phototrophic bacteria conflicts with the observation of singlet-triplet carotenoid excitation fission in these complexes. In LH2 complexes from the purple bacterium Allochromatium minutissimum, a drop in the efficiency of carotenoid triplet generation is demonstrated, which correlates with the extent of selective photooxidation of bacteriochlorophylls absorbing at ~850 nm. We conclude that singlet-triplet fission of carotenoid excitation proceeds with participation of these excitonically coupled bacteriochlorophylls. In the framework of the proposed mechanism, the contradiction between LH2 structure and photophysical properties of carotenoids is eliminated. The possibility of singlet-triplet excitation fission involving a third mediator molecule was not considered earlier.  相似文献

Results of active experiments on electron beam injection from the Intercosmos-25 satellite into the ionospheric plasma are presented. A quasistatic magnetic field and the VLF-wave magnetic component are excited when an unmodulated electron beam with a current of I be ?0.1 A and energy of ? be =mv 2/2?10 keV is injected into the ambient plasma. The magnetic field excitation is attributed to the onset of plasma gradient instabilities.  相似文献

1. Pelagia noctiluca responds to galvanic stimulation by a luminescent glow at the anode. If placed near the cathode a secondary glow occurs also on the cathodal side. 2. The luminescent slime of Pelagia when subjected to the galvanic current glows around the cathode. This is referred partly to the movement of hydrogen bubbles, but in the main to the alkali formed at the cathode. 3. The cause of galvanic stimulation in Pelagia is ionic. (1) Anodal stimulation is referred to the blocking of positive ions by the tissue on that side. (2) Cathodal stimulation, when the animal lies near the cathode, is due to the diffusion of alkali outward from a region of high concentration (the cathode). 4. Only the margin of the bell is excited to luminescence by the galvanic current. It is therefore concluded that nervous elements are the seat of excitation. 5. Luminescence is not a result of muscular contraction, since K ion causes relaxation of musculature but a continuous luminescent glow in Pelagia. The galvanic current causes pulsations of the bell (contraction and relaxation of the musculature) but a continuous glow.  相似文献

Results are presented from experimental studies of the glow dynamics of active nitrogen in the stage of its excitation by a current pulse and during the discharge afterglow. The mechanism is proposed for the generation of a light splash in a highly activated nitrogen after the end of its pulsed excitation. The key role in the generation of this splash is played by the D-V processes, by which the dissociation energy is transferred to the vibrational degrees of freedom in the course of recombination of nitrogen atoms, and the V-E processes, by which the vibrational energy of highly excited molecules N2(X, v ≥ 25–27) is transferred to the emitting electronic states N2(B, v) after the V-V delay. Results of simulations based on the mechanism proposed are also presented.  相似文献

Upon irradiation in the ultraviolet region aromatic chromophores may transfer energy to a nearby Tb3+, which in turn emits a green phosphorescence. This paper reports the characterization of the ultraviolet excitation spectra of aromatic chromophores capable of transferring energy to Tb3+ by monitoring of the green Tb3+ emission in the 540-550 nm region. Results are included for complexes containing phenyl, hydroxyphenyl, indole. and catechol chromophores. Characteristic excitation spectra are presented for the aromatic chromophores occurring as side chains in proteins. Though it is preferable to compare entire excitation spectra, the ratio of intensities at 292 to 276 nm, R, is suggested as a useful diagnostic criterion. Numerical R values are indicative of the following aromatic side chains as the energy donor to Tb3+: R <0.2, unionized tyrosine; R = 0.5 to 1.0, tryptophan; and R > 1.8. ionized tyrosine. Tlie phenylalanyl chromophore displays a definitive excitation spectrum at shorter wavelengths. For ovotransferrin R = 0.9 and comparison of the full excitation spectra suggests that it contains comparable contributions from both ionized tyrosine and tryptophan side chains. Some difficulties in obtaining reliable excitation spectra are described. An analysis of inner-filtering of incident light reveals that for an absorbance less than 0.8 the excitation spectrum is broadened and flattened compared to the absorption spectrum. At maximum absorbances greater than 0.8 false maxima may appear to both sides of a real maximum. Two spurious maxima in an excitation spectrum were generated in a Tb3+ complex and compared to the correct excitation spectrum of the same complex obtained at lower absorbance.  相似文献

The problem of singlet excitation kinetics and dynamics, especially at high excitation intensities, among a small number of chromophores of a given system has been addressed. A specific scheme for the kinetics is suggested and applied to CPII, a small chlorophyll (Chl)a/b antenna complex the fluorescence lifetime of which has been reported to be independent of excitation intensity over a wide intensity range of picosecond pulses. We have modeled the kinetics from the point of view that Chla molecules in CPII are Förster coupled so that a second excitation received by the group of Chla's either creates a state with two localized excitons or raises the first one to a doubly excited state. The data on CPII can be understood on the basis of a kinetic model that does not exclude exciton annihilation during the excitation pulse. The implied annihilation rate is consistent with our theoretical estimates of that rate obtained by applying excitation transfer theory to pairs of molecules both initially excited.  相似文献

Alternating current threshold excitation of space-clamped squid giant axons was measured as a function of frequency, external calcium concentration, temperature (from 10° to 35°C), and hyper- and depolarizing steps. In normal axons there is usually an optimum frequency at about 120 Hz, at which the threshold is a minimum. The threshold rises at both lower and higher frequencies to give a resonance curve. Low calcium causes an increase in optimum frequency, a decrease in current threshold, and an increase in sharpness of tuning in both real axons and axons computed according to the Hodgkin-Huxley formulation; high calcium causes opposite effects. An increase in temperature causes an increase of optimum frequency, an increase in sharpness of tuning, and an increase in threshold current in both real and computed axons. The Q10 for the effect of temperature upon optimum frequency is 1.8 in real and computed axons at moderate temperatures. Hyperpolarization causes (a) a decrease in optimum frequency, (b) a decrease in sharpness of tuning, and (c) an increase in threshold. Depolarization causes opposite effects.  相似文献

Blair''s recent theory of excitation is analysed with the following conclusions: 1. The theory is inapplicable to currents of long duration; i.e., slowly increasing currents and the opening excitation. 2. The theory is a modification of the condenser theory of excitation but the modification is to be rejected on three grounds: (a) The modification has no obvious physical significance. (b) It does not in fact remedy the divergence between calculation and observation. (c) It leads to certain conclusions of a surprising kind which are contrary to observed fact. 3. The qualitative value of the condenser theory is demonstrated by the fairly close agreement between calculation and observation over a considerable field of enquiry.  相似文献

Results are presented from experimental studies of variations in the plasma parameters during the excitation of a multiaxis magnetic configuration by the induction current (up to 17 kA) in the basic magnetic configuration of the L-2M stellarator in the regime of ECR heating at a microwave power of ~200 kW (~1 MW m?3) and an average plasma density of (1–2) × 1019 m?3. The current direction was chosen to reduce the net rotational transform (the so-called “negative“ current). The current was high enough for the rotational transform to change its sign inside the plasma column. Computer simulations of the L-2M magnetic structure showed that the surface with a zero rotational transform is topologically unstable and gives rise to magnetic islands, i.e., to a multiaxis magnetic configuration. Magnetic measurements showed that, at negative currents above 10 kA, intense bursts of MHD oscillations with a clearly defined toroidal mode number n = 0 were observed in the frequency range of several kilohertz. Unfortunately, the experimental data are insufficient to draw the final conclusion on the transverse structure of these oscillations. The radial temperature profiles along the stellarator major radius in the equatorial plane were studied. It is found that the electron temperature decreases by a factor of 1.3 in the plasma core (r/a ≤ 0.6) and that the temperature jump is retained near the boundary. A change in turbulent fluctuations of the plasma density during the excitation of a negative current was studied using wave scattering diagnostics. It is found that the probability density function of the increments of fluctuations in the plasma core differs from a Gaussian distribution. The measured distribution is heavy-tailed and broadens in the presence of the current. It is found that the spectrum of turbulent fluctuations and their Doppler shift near the plasma boundary are nonuniform in the radial direction. This may be attributed to the shear of the poloidal velocity. The experimental results indicate that the formation of regions with a zero rotational transform in the plasma core somewhat intensifies plasma transport.  相似文献

Channelrhodopsins, such as the algal phototaxis receptor Platymonas subcordiformis channelrhodopsin-2 (PsChR2), are light-gated cation channels used as optogenetic tools for photocontrol of membrane potential in living cells. Channelrhodopsin (ChR)-mediated photocurrent responses are complex and poorly understood, exhibiting alterations in peak current amplitude, extents and kinetics of inactivation, and kinetics of the recovery of the prestimulus dark current that are sensitive to duration and frequency of photostimuli. From the analysis of time-resolved optical absorption data, presented in the accompanying article, we derived a two-cycle model that describes the photocycles of PsChR2. Here, we applied the model to evaluate the transient currents produced by PsChR2 expressed in HEK293 cells under both fast laser excitation and step-like continuous illumination. Interpretation of the photocurrents in terms of the photocycle kinetics indicates that the O states in both cycles are responsible for the channel current and fit the current transients under the different illumination regimes. The peak and plateau currents in response to a single light step, a train of light pulses, and a light step superimposed on a continuous light background observed for ChR2 proteins are explained in terms of contributions from the two parallel photocycles. The analysis shows that the peak current desensitization and recovery phenomena are inherent properties of the photocycles. The light dependence of desensitization is reproduced and explained by the time evolution of the concentration transients in response to step-like illumination. Our data show that photocycle kinetic parameters are sufficient to explain the complex dependence of photocurrent responses to photostimuli.  相似文献

We have measured the singlet-singlet quenching of the bacteriochlorophyll (BChl) fluorescence yield as a function of excitation intensity in a number of antenna complexes isolated from photosynthetic bacteria. Our results show that the lithium dodecyl sulfate (LDS)-B875, LDS-B800 – 850 and lauryldimethylamine N-oxide complexes of Rhodopseudomonas sphaeroides contain 8, greater than 25 and greater than 600 BChl a molecules, respectively. The size of the Rhodopspirillum rubrum B880 complex is greater than 70 BChl a and that of the water-soluble BChl a complex from Prosthecochloris aestuarii about 20–25 BChl a. These results are discussed in relation to current models of the arrangement of antenna complexes within the photosynthetic membranes.  相似文献

We present a method for in situ monitoring of phytoplankton composition changes in a marine environment. The method is based on delayed fluorescence excitation spectra analyzed with CHEMTAX software, which is generally used for determination of phytoplankton communities with HPLC pigment data. Delayed fluorescence (DF) is a photosynthetic parameter that can only be measured in living cells. Algal DF excitation spectra are group-specific, based on their composition of photosynthetic pigments.DF excitation spectra of 14 marine algal species from different families were measured with a delayed fluorescence spectrometer. Mixtures were prepared from northern Adriatic algal species representing six taxonomic groups: dinoflagellates (Prorocentrum minimum), diatoms (Skeletonema costatum), cyanobacteria (Synechococcus sp.), prasinophytes (Micromonas sp.), cryptophytes (Teleaulax sp.), and prymnesiophytes (Isochrysis galbana). The DF excitation spectra (DFS) and HPLC pigment compositions of the mixtures were analyzed with CHEMTAX software. The prediction power of DFS–CHEMTAX method was comparable to HPLC–CHEMTAX.  相似文献

Picosecond absorbance difference spectra at a number of delay times after a 35 ps excitation pulse and kinetics of absorbance changes were measured in chromatophores of the photosynthetic purple bacterium Rhodospirillum rubrum after chemical oxidation of the primary electron donor P-875. Kinetics and spectra were measured of the excited singlet states of carotenoid and bacteriochlorophyll a and also of the triplet state of the carotenoid. The excited singlet state of carotenoid, produced by direct excitation at 532 nm, is characterized by a bleaching of the ground state absorption bands in the region 450–490 nm and by an absorbance increase with a maximum near 570 nm. Its lifetime was calculated to be 0.6 ± 0.1 ps in vitro and less than 1 ps in vivo. The triplet state of carotenoid in vivo is formed within 100 ps after direct carotenoid excitation via a pathway that does not involve excited states of bacteriochlorophyll. Singlet excitation of a bacteriochlorophyll a molecule causes the bleaching of its Qx and Qy absorption bands, and is probably associated with blue shifts of the Qy absorption band of about six neighboring bacteriochlorophyll molecules. Upon increasing the excitation density, the average lifetime of the singlet excitations on bacteriochlorophyll decreased from about 350 ps to about 10 ps or less. The results are in quantitative agreement with the known effect of singlet-singlet annihilation upon the fluorescence yield, and furthermore show that no bacteriochlorophyll or carotenoid triplet formation is associated with this annihilation.  相似文献
