Surface-active amphiphiles find applications in a wide range of areas of industry such as agrochemicals, personal care, and pharmaceuticals. In many of these applications, interaction with cell membranes is a key factor for achieving their purpose. How do amphiphiles interact with lipid membranes? What are their bases for membrane specificity? Which biophysical properties of membranes are susceptible to modulation by amphiphilic membrane-effectors? What aspects of this interaction are important for performing their function? In our work on membrane biophysics over the years, questions like these have arisen and we now share some of our findings and discuss them in this review. This topic was approached focusing on the membrane properties and their alterations rather than on the amphiphile structure requirements for their interaction. Here, we do not aim to provide a comprehensive list of the modes of action of amphiphiles of biological interest but to help in understanding them.  相似文献

The ability to understand and predict the effects of environmental stress on biodiversity is becoming increasingly important in our changing environment. Antarctic marine species are some of the most stenothermal on the planet and many inhabit the waters off the Antarctic Peninsula which is one of the areas where there is rapid regional climate change. Therefore these animals are highly vulnerable to changing environmental temperatures and clearly we need to understand the complexities of their response, not just at the individual species level, but also the implications for the ecosystem as a whole. Heat shock proteins have a long history of use in studies of organism stress responses and have frequently been proposed as potential universal molecular biomarkers, especially for non-model species. In this mini-review, the heat shock response and heat shock proteins (specifically the HSP70 family) are examined in Antarctic marine species alongside their physiological capabilities and limits to answer a series of questions: do these animals have a heat shock response which includes the expression of HSP70 genes? What is the relationship between their heat shock response and physiological capabilities? Can HSP70 genes be used as molecular biomarkers for these species?  相似文献

为促进城市可持续发展,在科学合理规划城市空间格局的基础上,应突出强调保护原有特色生态环境要素,保育生态系统服务功能和原住民人居环境。作为世界文化遗产,丽江城市面貌在过去数十年发生了深刻变化,城市用地显著扩张,同时严峻的环境问题对城市可持续发展提出挑战。以丽江市为例,选取对其发展影响深远的水系元素、噪声环境因子和森林火灾3个关键生态敏感因子,提出通过分析这类生态敏感区在城市规划过程中的避让方法、程度,区划城市规划基本空间格局,以避免城市未来因生态环境问题突出而制约其发展的规划用地思路。结果表明,丽江城区周边湖泊水系、古城内部及周边原住民人口密集区都是保障城市可持续发展的生态保护重点地区,在城市用地规划中必须予以避让;该区域面积为147.2 km2,占研究区面积的11.1%。同时,明确给出了规划用地的具体空间控制范围和相应的避让原则。  相似文献

What genetic factors are responsible for tumorigenesis and cancer development? What environmental factors play a role in cancer formation and progression? In how many ways can our bodies prevent and restrict the growth of cancerous cells?How can we identify and deliver effective drugs to fight cancer? In the fight against cancer,which kills more people than any other disease,these and other questions have long interested researchers from a diverse range of fields.To answer these questions and to fight cancer more effectively,we must increase our understanding of basic cancer biology.Model organisms,including the fruit fly Drosophila melanogaster,have played instrumental roles in our understanding of this devastating disease and the search for effective cures.Drosophila and its highly effective,easy-touse,and ever-expanding genetic tools have contributed toand enriched our knowledge of cancer and tumor formation tremendously.  相似文献

Consumers increasingly demand information about the environmental impacts of their food. The French government is in the process of introducing environmental labelling for all food products. A scientific council was set up, and its main conclusions are presented in this article, through six questions: What environmental issues should be considered? What objective should be targeted? What data are needed, and for whom? What methods for assessing environmental impacts? Which environmental scores should be chosen? What label format should be proposed? By answering these questions and considering the context, the available data, the proposed methods and adjustments, and the knowledge of consumer perception of formats, the scientific council considers that a labelling scheme is feasible and relevant.

As scientists, we are at least as excited about the open questions—the things we do not know—as the discoveries. Here, we asked 15 experts to describe the most compelling open questions in plant cell biology. These are their questions: How are organelle identity, domains, and boundaries maintained under the continuous flux of vesicle trafficking and membrane remodeling? Is the plant cortical microtubule cytoskeleton a mechanosensory apparatus? How are the cellular pathways of cell wall synthesis, assembly, modification, and integrity sensing linked in plants? Why do plasmodesmata open and close? Is there retrograde signaling from vacuoles to the nucleus? How do root cells accommodate fungal endosymbionts? What is the role of cell edges in plant morphogenesis? How is the cell division site determined? What are the emergent effects of polyploidy on the biology of the cell, and how are any such “rules” conditioned by cell type? Can mechanical forces trigger new cell fates in plants? How does a single differentiated somatic cell reprogram and gain pluripotency? How does polarity develop de-novo in isolated plant cells? What is the spectrum of cellular functions for membraneless organelles and intrinsically disordered proteins? How do plants deal with internal noise? How does order emerge in cells and propagate to organs and organisms from complex dynamical processes? We hope you find the discussions of these questions thought provoking and inspiring.

Many types of data are best analyzed by fitting a curve using nonlinear regression, and computer programs that perform these calculations are readily available. Like every scientific technique, however, a nonlinear regression program can produce misleading results when used inappropriately. This article reviews the use of nonlinear regression in a practical and nonmathematical manner to answer the following questions: Why is nonlinear regression superior to linear regression of transformed data? How does nonlinear regression differ from polynomial regression and cubic spline? How do nonlinear regression programs work? What choices must an investigator make before performing nonlinear regression? What do the final results mean? How can two sets of data or two fits to one set of data be compared? What problems can cause the results to be wrong? This review is designed to demystify nonlinear regression so that both its power and its limitations will be appreciated.  相似文献

The environmental effects of e‐commerce may be described in terms of first‐, second‐, and third‐order effects. Data for these effects are scarce, partly because research on environmental effects of e‐commerce and e‐business is still in its infancy, although it is evolving very rapidly. Until now, positive environmental consequences of e‐commerce have generally been coincidental. Two crucial questions that must be addressed are (1) How do we improve our understanding and management of the environmental effects of e‐commerce? and (2) Which approaches are best suited to the development of sustainable e‐solutions? Three approaches to developing sustainable e‐commerce solutions are discussed: the extension of environmental performance measurement and management to e‐commerce activities, the use of new cooperative forms of innovation management, and the provision of customer choice. Finally, an outlook on future research demands is presented. The technology itself (information and communication technologies, Internet) does not determine sustainability, but rather its design, use, and regulation does.  相似文献

Many intriguing relationships between allergy and cancer continue to pose important unanswered questions. Is there a correlation between IgE and atopy, and cancer? Which are the most important IgE effector cells and do they inhibit or promote tumor growth? Will IgE-mediated antigen uptake and cross-presentation by DCs activate CTLs or Tregs? What animal models are useful in this context and should we design specific anti-tumor immunotherapies, active or passive, based on IgE? Recombinant IgE has already been generated against the tumor-associated antigens folate-binding receptor, HER-2 and EGFR-how to exclude the risk of anaphylaxis in passive immunotherapy with IgE? How can we deal with hypersensitivity to antineoplastic drugs? What is the role of chemotherapeutics and biologicals in the modulation of IgE responses and the impact of the aberrant expression of mutated enzyme AID in cancer tissue? In an effort to survey the state-of-the-art in this area and to answer some of these questions, we invited leaders in the field to participate in a "Symposium-in-Writing". This represents a collection of peer-reviewed papers covering each group's specialist area of expertise. In this way, we aim to target the most important topics in these areas and to provide a comprehensive view of the state-of-the-art in the emerging multi-disciplinary field of "AllergoOncology".  相似文献

More than fifteen years after the first identification of a class II isoform of phosphoinositide 3-kinase (PI3K) in Drosophila melanoǵaster this subfamily remains the most enigmatic among all PI3Ks. What are the functions of these enzymes? What are their mechanisms of activation? Which downstream effectors are specifically regulated by these isoforms? Are class I and class II PI3Ks redundant or do they control different intracellular processes? And, more important, do class II PI3Ks have a role in human diseases? The recent increased interest on class II PI3Ks has started providing some answers to these questions but still a lot needs to be done to completely uncover the contribution of these enzymes to physiological processes and possibly to pathological conditions. Here we will summarise the recent findings on the alpha isoform of mammalian class II PI3Ks (PI3K-C2α ) and we will discuss the potential involvement of this enzyme in human diseases.  相似文献

The huge number of neurons in the human brain are connected to form functionally specialized assemblies. The brain's amazing processing capabilities rest on local communication within and long-range communication between these assemblies. Even simple sensory, motor and cognitive tasks depend on the precise coordination of many brain areas. Recent improvements in the methods of studying long-range communication have allowed us to address several important questions. What are the common mechanisms that govern local and long-range communication and how do they relate to the structure of the brain? How does oscillatory synchronization subserve neural communication? We have approached the problem of nerve excitability through three questions: (a) What is the diagram for a channel? That is, what conformational states can the protein assume, and what transitions between these conformations are permitted? (b) What is the channel conductance associated with each conformation the channel can assume? (c) How do the rates for conformational transition depend upon membrane potential? These three questions arise from a standard statistical mechanical treatment of a nerve membrane containing several classes of identical, independent channels. Gating of channels, in this view, is associated with conformational changes of the channel protein, and it is assumed these conformations are distinct. The precise formulation of these questions is presented in terms of the theoretical treatment, and the approaches we have taken to answer the questions are indicated. Our present results indicate: Transition rates should depend exponentially on membrane potential over a limited voltage range, but probably will show a more complex dependence for extremes of the range; channels probably can take on only two conductances, open and shut, but more complicated situations are not entirely excluded; the diagram for a channel cannot be determined from standard voltage clamp data alone, but by studying gating currents and conductance fluctuations, it should be possible to select between alternative plausible physical mechanisms.  相似文献

近10年来,可持续性科学蓬勃发展,已成为21世纪全球普遍关注的一个重要的新科学领域.然而,在大力提倡可持续发展和生态文明建设的中国,可持续性科学尚未引起科学家和实践者的足够重视.为促进可持续性科学在中国的发展,2014年邬建国等曾撰文介绍什么是可持续性科学.本文进一步探讨了这一问题,并补充阐述了可持续性科学与可持续发展研究的关系、可持续性科学的科学范式及其8个基本论题.基于对可持续性科学发展动态的分析,作者认为,一方面,可持续性科学已进入系统推进的成熟发展期;另一方面,虽然我国可持续发展研究、实践与教育的热情高涨,但在可持续性科学领域起步较晚,落后于主要发达国家和南非.为此,本文在文献综述的基础上,提出促进中国发展可持续性科学的“三位一体”策略:一是“请进来”以服务中国实践;二是“走出去”以贡献中国智慧;三是“中西医结合”以引领学科发展.  相似文献

In this paper, we ask if the use of a voluntary standard assessment system for environmental reporting could help mitigate the damage caused by oil and gas companies to developing nations. What level of data reporting by these companies is needed to allow the assessment of environmental practices? To answer these questions, we evaluate the environmental practices of eight oil and gas companies against the Sustainability Reporting Guidelines issued in 2006 by the Global Reporting Initiative (GRI). Content analysis of their environmental reports 2009 indicates that they made reasonable efforts to disclose their environmental performance in accordance with the GRI Sustainability Reporting Guidelines. These guidelines appear to provide a robust and readily available tool for reporting comprehensive progress concerning all aspects of environmental activities. The voluntary adoption of the guidelines by a vast majority of the oil and gas companies increases transparency, credibility and comparability in sustainability reporting. In addition, five of the eight oil and gas companies disclosed that they had obtained third-party assurance of their sustainability reports.  相似文献

Gladstone, Queensland, is one of Australia’s largest coal ports and the site of many industries. Few of the 65,000 residents of the region are socially and economically independent of the fossil fuel industry. Environmental engagement is fragmented. The city epitomises, in a striking way, the double bind of growth and sustainability. The Australian environmental movement is generally seen as distant and irrelevant to local concerns. The article analyses conditions for, forms of and implications of environmental engagement in a city where coal, gas and industry are taken for granted as necessary conditions for prosperity and economic security. To what extent are ecological relevant to the residents; and how can they be dealt with within the parameters that define their lifeworlds? What can the Gladstone experience teach us about possible solutions to the double bind of growth and sustainability?  相似文献

Extensive tropical forest loss and degradation have stimulated increasing awareness at the international policy level of the need to undertake large‐scale forest landscape restoration (FLR). Natural regeneration offers a cost‐effective way to achieve large‐scale FLR, but is often overlooked in favor of tree plantations. The studies presented in this special issue show how natural regeneration can become an important part of FLR and highlight the ecological, environmental, and social factors that must be considered to effectively do so. They also identify major knowledge gaps and outline a research agenda to support the use of natural regeneration in FLR. Six central questions emerge from these studies: (1) What are the ecological, economic, and livelihood outcomes of active and passive restoration interventions?; (2) What are the tradeoffs and synergies among ecological, economic, and livelihood outcomes of natural regeneration, restoration and productive land uses, and how do they evolve in the face of market and climate shocks?; (3) What diagnostic tools are needed to identify and map target areas for natural regeneration?; (4) How should spatial prioritization frameworks incorporate natural regeneration into FLR?; (5) What legal frameworks and governance structures are best suited to encourage natural regeneration and how do they change across regions and landscapes?; (6) What financial mechanisms can foster low‐cost natural regeneration? Natural regeneration is not a panacea to solve tensions and conflicts over land use, but it can be advantageous under some circumstances. Identifying under what conditions this is the case is an important avenue for future research.  相似文献

Stochastic modeling of phylogenies raises five questions that have received varying levels of attention from quantitatively inclined biologists. 1) How large do we expect (from the model) the ratio of maximum historical diversity to current diversity to be? 2) From a correct phylogeny of the extant species of a clade, what can we deduce about past speciation and extinction rates? 3) What proportion of extant species are in fact descendants of still-extant ancestral species, and how does this compare with predictions of models? 4) When one moves from trees on species to trees on sets of species (whether traditional higher order taxa or clades within PhyloCode), does one expect trees to become more unbalanced as a purely logical consequence of tree structure, without signifying any real biological phenomenon? 5) How do we expect that fluctuation rates for counts of higher order taxa should compare with fluctuation rates for number of species? We present a mathematician's view based on an oversimplified modeling framework in which all these questions can be studied coherently.  相似文献

Plants that have evolved to survive on metal‐rich soils—metallophytes—have key values that must drive research of their unique properties and ultimately their conservation. The ability of metallophytes to tolerate extreme metal concentrations commends them for revegetation of mines and metal‐contaminated sites. Metallophytes can also be exploited in environmental technologies, for example, phytostabilization, phytoremediation, and phytomining. Actions towards conserving metallophyte species are imperative, as metallophytes are increasingly under threat of extinction from mining activity. Although many hundreds of papers describe both the biology and applications of metallophytes, few have investigated the urgent need to conserve these unique species. This paper identifies the current state of metallophyte research, and advocates future research needs for the conservation of metallophyte biodiversity and the sustainable uses of metallophyte species in restoration, rehabilitation, contaminated site remediation, and other nascent phytotechnologies. Six fundamental questions are addressed: (1) Is enough known about the global status of metallophytes to ensure their conservation? (2) Are metallophytes threatened by the activities of the minerals industry, and can their potential for the restoration or rehabilitation of mined and disturbed land be realized? (3) What problems exist in gaining prior informed consent to access metallophyte genetic resources and how can the benefits arising from their uses be equitably shared? (4) What potential do metallophytes offer as a resource base for phytotechnologies? (5) Can genetic modification be used to “design” metallophytes to use in the remediation of contaminated land? (6) Does the prospect of using metallophytes in site remediation and restoration raise ethical issues?  相似文献

A continuing fundamental trend in the area of quality assurance in analytical laboratories are measures that enable trace-ability of analytical results to internationally renowned units, like e.g. the mole. Especially certified reference materials (CRMs) are important tools for achieving this goal. What are CRMs? Why do we need them in the field of mycotoxin analysis? Which kind of CRMs are currently available for the determination of mycotoxins and how should they be used? These are questions, which are increasingly being asked.
