Actin and myosin are components of the plant cell cytoskeleton that extend from cell to cell through plasmodesmata (PD), but it is unclear how they are organized within the cytoplasmic sleeve or how they might behave as regulatory elements. Early work used antibodies to locate actin and myosin to PD, at the electron microscope level, or to pitfields (aggregations of PD in the cell wall), using immunofluorescence techniques. More recently, a green fluorescent protein (GFP)-tagged plant myosin VIII was located specifically at PD-rich pitfields in cell walls. Application of actin or myosin disrupters may modify the conformation of PD and alter rates of cell-cell transport, providing evidence for a role in regulating PD permeability. Intriguingly, there is now evidence of differentiation between types of PD, some of which open in response to both actin and myosin disrupters, and others which are unaffected by actin disrupters or which close in response to myosin inhibitors. Viruses also interact with elements of the cytoskeleton for both intracellular and intercellular transport. The precise function of the cytoskeleton in PD may change during cell development, and may not be identical in all tissue types, or even in all PD within a single cell. Nevertheless, it is likely that actin- and myosin-associated proteins play a key role in regulating cell-cell transport, by interacting with cargo and loading it into PD, and may underlie the capacity for one-way transport across particular cell and tissue boundaries.  相似文献

Atypical neuropathological and molecular phenotypes of bovine spongiform encephalopathy (BSE) have recently been identified in different countries. One of these phenotypes, named bovine "amyloidotic" spongiform encephalopathy (BASE), differs from classical BSE for the occurrence of a distinct type of the disease-associated prion protein (PrP), termed PrP(Sc), and the presence of PrP amyloid plaques. Here, we show that the agents responsible for BSE and BASE possess different biological properties upon transmission to transgenic mice expressing bovine PrP and inbred lines of nontransgenic mice. Strikingly, serial passages of the BASE strain to nontransgenic mice induced a neuropathological and molecular disease phenotype indistinguishable from that of BSE-infected mice. The existence of more than one agent associated with prion disease in cattle and the ability of the BASE strain to convert into the BSE strain may have important implications with respect to the origin of BSE and spongiform encephalopathies in other species, including humans.  相似文献

High levels of ROS have been linked to programmed cell death pathways and to ageing. In the recent decade, our view on the organization of the bacterial cell has been revolutionized by the identification of cytoskeletal elements. Most bacterial species have structural homologs of actin and tubulin that assemble into dynamic, filamentous structures at precisely defined sub-cellular locations. The essential cell division protein FtsZ forms a dynamic ring at mid-cell and is similar in its structure to tubulin. Proteins of the MreB family, which are structural homologs of actin, assemble into helical or straight filaments in the bacterial cytoplasm. As in eukaryotic cells, the bacterial cytoskeleton drives essential cellular processes such as cell division, cell wall growth, DNA movement, protein targeting, and alignment of organelles. Different high-throughput assays have been developed to search for inhibitors of components of the bacterial cytoskeleton. Cell-based assays for the detection of cell division inhibitors as well as FtsZ GTPase assays led to the identification of several compounds that inhibit the polymerization of FtsZ, by this blocking bacterial cell division. Such inhibitors might not only be valuable tools for basic research, but might also lead to novel therapeutic agents against pathogenic bacteria. For example, the polyphenol dichamanetin, the 2-alkoxycarbonylaminopyridine SRI-3072, and the benzophenanthridine alkaloid sanguinarine inhibit the GTPase activity of FtsZ and exhibit antimicrobial activity.  相似文献

The river domain: why are there more species halfway up the river?   总被引:2,自引:0,他引:2  
Biologists have long noted higher levels of species diversity in the longitudinal middle‐courses of river systems and have proposed many explanations. As a new explanation for this widespread pattern, we suggest that many middle‐course peaks in richness may be, at least in part, a consequence of geometric constraints on the location of species’ ranges along river courses, considering river headwaters and mouths as boundaries for the taxa considered. We demonstrate this extension of the mid‐domain effect (MDE) to river systems for riparian plants along two rivers in Sweden, where a previous study found a middle‐course peak in richness of natural (non‐ruderal) species. We compare patterns of empirical richness of these species to null model predictions of species richness along the two river systems and to spatial patterns for six environmental variables (channel width, substrate fineness, substrate heterogeneity, ice scour, bank height, and bank area). In addition, we examine the independent prediction of mid‐domain effects models that species with large ranges, because the location of their ranges is more constrained, are more likely to produce a mid‐domain peak in richness than are species with small ranges. Species richness patterns of riparian plants were best predicted by models including both null model predictions and environmental variables. When species were divided into large‐ranged and small‐ranged groups, the mid‐domain effect was more prominent and the null model predictions were a better fit to the empirical richness patterns of large‐ranged species than those of small‐ranged species. Our results suggest that the peak in riparian plant species richness in the middle courses of the rivers studied can be explained by an underlying mid‐domain effect (driven by geometric constraints on large‐ranged species), together with environmental effects on richness patterns (particularly on small‐ranged species). We suggest that the mid‐domain effect may help to explain similar middle‐course richness peaks along other rivers.  相似文献

Solar ultraviolet radiation (UV) is a major environmental factor that dramatically alters the homeostasis of the skin as an organ by affecting the survival, proliferation and differentiation of various cutaneous cell types. The effects of UV on the skin include direct damage to DNA, apoptosis, growth arrest, and stimulation of melanogenesis. Long-term effects of UV include photoaging and photocarcinogenesis. Epidermal melanocytes synthesize two main types of melanin: eumelanin and pheomelanin. Melanin, particularly eumelanin, represents the major photoprotective mechanism in the skin. Melanin limits the extent of UV penetration through the epidermal layers, and scavenges reactive oxygen radicals that may lead to oxidative DNA damage. The extent of UV-induced DNA damage and the incidence of skin cancer are inversely correlated with total melanin content of the skin. Given the importance of the melanocyte in guarding against the adverse effects of UV and the fact that the melanocyte has a low self-renewal capacity, it is critical to maintain its survival and genomic integrity in order to prevent malignant transformation to melanoma, the most fatal form of skin cancer. Melanocyte transformation to melanoma involves the activation of certain oncogenes and the inactivation of specific tumor suppressor genes. This review summarizes the current state of knowledge about the role of melanin and the melanocyte in photoprotection, the responses of melanocytes to UV, the signaling pathways that mediate the biological effects of UV on melanocytes, and the most common genetic alterations that lead to melanoma.  相似文献

The term apoptosis describes the predictable structural changes associated with many forms of programmed cell death. One of the first visible events of apoptosis is the collapse of the nucleus. Nuclear degradation is manifested by digestion of chromatin into nucleosome-sized fragments or multiples of these. This digestion of DNA is enzymatic, and several attempts have been made to characterize apoptosis-specific endodeoxyribonucleases. Although there are strong candidates for such enzymes, direct evidence for their role in apoptosis is yet to be provided.  相似文献

As a result of these efforts, we are gaining an appreciation of the varied and often surprising strategies whereby roots adjust to and condition their soil environment for optimal growth and development. This article summarizes current knowledge of the often complex interactions between roots and biotic and abiotic factors within the soil. Rabbit haemorrhagic disease virus emerged in China in 1984, and has killed hundreds of millions of wild rabbits in Australia and Europe. In the UK there appears to be an endemic non-pathogenic strain, with high levels of seroprevalence being recorded, in the absence of associated mortality. Using a seasonal, age-structured model we examine the hypothesis that differences in rabbit population demography differentially affect the basic reproductive rates (R(0)) of the pathogenic and non-pathogenic strains, leading to each dominating in some populations and not others. The strain with the higher R(0) excluded the other, with the dynamics depending upon the ratio of the two R(0) values. When the non-pathogenic strain dominated, the pathogenic strain caused only transient mortality, although this could be significant when the two R(0) values were similar. When the pathogenic strain dominated, repeated epidemics led to host eradication. Seroprevalence data suggest that the non-pathogenic strain may be protecting some, but not all UK populations, with half being 'at risk' from invasion by the pathogenic strain and a fifth prone to significant transient mortality. We identify key questions for empirical research to test this prediction.  相似文献

Microtubules are fibers of the cytoskeleton involved in mitosis, intracellular transport, motility and other functions. They contain microtubule-associated proteins (MAPs) bound to their surface which stabilize microtubules and promote their assembly. There has been a debate on additional functions of MAPs, e.g. whether MAPs crosslink microtubules and thus increase their rigidity, or whether they act as spacers between them. We have studied the packing of microtubules in the presence of MAPs by solution X-ray scattering using synchrotron radiation. Microtubules free in solution produce a scattering pattern typical of an isolated hollow cylinder, whereas tightly packed microtubules generate a pattern dominated by interparticle interference. The interference patterns are interpreted in terms of the Hosemann paracrystal concept, adapted for arrays of parallel fibers with hexagonal arrangement in the plane perpendicular to the fiber axes (Briki et al., 1998). Microtubules without MAPs can rapidly and efficiently be compressed by centrifugation, as judged by the transition from a "free microtubule" to a "packed microtubule" X-ray scattering pattern. MAPs make the microtubule array highly resistant to packing, even at high centrifugal forces. This emphasizes the role of MAPs as spacers of microtubules rather than crosslinkers. A possible function is to keep the microtubule tracks free for the approach of motor proteins carrying vesicle or organelle cargoes along microtubules.  相似文献
