Two isomeric dibenzo-O2S2 macrocycles L1 and L2 have been synthesised and their coordination chemistry towards palladium(II) has been investigated. Two-step approaches via reactions of 1:1-type complexes, [cis-Cl2LPd] (1a: L = L1, 1b: L = L2), with different O2S2 macrocycle systems (L1 and L2) have led to the isolation of the following bis(O2S2 macrocycle) palladium(II) complexes in the solid state: [Pd(L1)2](ClO4)2 (2a) and a mixture of [Pd(L1)2](ClO4)2 (2a) + [Pd(L2)2](ClO4)2 (2b).  相似文献

Eleven prenylated C6-C3 compounds, illioliganpyranone A (1), illioliganfunone A-D (2-5), and illioliganone D-I (6-11), together with five known prenylated C6-C3 compounds (12-16), were isolated from roots of Illicium oligandrum. The structures of 1-11 were elucidated by spectroscopic methods including 1D and 2D NMR, HRESIMS, and CD experiments. Possible biosynthetic pathways to compounds 1-16 derived from a common precursor of 5-allylbenzene-1,2,4-triol were postulated. All compounds were evaluated for cytotoxic activities against five human cancer cell lines (HCT-8, Bel-7402, BGC-823, A549 and A2780). Compound 15 exhibited significant cytotoxicity against HCT-8, BGC-823, A549, and A2780 cell lines with IC50 values of 0.30-2.57 μM. Compound 16 showed moderate selective cytotoxicity against sensitive A2780 cells with IC50 value of 1.38 μM.  相似文献

We synthesized disulfide-based cyclic RGD pentapeptides bearing a near-infrared fluorescent dye (cypate), represented by cypate-c(CRGDC) (1) for integrin-targeted optical imaging. These compounds were compared with the traditional lactam-based cyclic RGD counterpart, cypate-c(RGDfK) (2). Molecular modeling suggests that the binding affinity of 2 to integrin αvβ3 is an order of magnitude higher than that of 1. This was confirmed experimentally, which further showed that substitution of Gly with Pro, Val and Tyr in 1 remarkably hampered the αvβ3 binding. Interestingly, cell microscopy with A549 cells showed that 1 exhibited higher cellular staining than 2. These results indicate that factors other than receptor binding affinity to αvβ3 dimeric proteins mediate cellular uptake. Consequently, 1 and its analogs may serve as valuable molecular probes for investigating the selectivity and specificity of integrin targeting by optical imaging.  相似文献

The reaction of 3β-hydroxy-21-hydroxymethylidenepregn-5-en-3β-ol-20-one (1) with phenylhydrazine (2a) affords two regioisomers, 17β-(1-phenyl-3-pyrazolyl)androst-3-en-3β-ol (5a) and 17β-(1-phenyl-5-pyrazolyl)androst-5-en-3β-ol (6a). The direction of the ring-closure reactions of 1 with p-substituted phenylhydrazines (2b-e) depends strongly on the electronic features of the substituents. Oppenauer oxidation of 3β-hydroxy-17β-exo-heterocyclic steroids 5a-e and 6a-e yielded the corresponding Δ4-3-ketosteroids 9a-e and 10a-e. The inhibitory effects (IC50) of these compounds on rat testicular C17,20-lyase were investigated by means of an in vitro radioligand incubation technique.  相似文献

Three novel metal-organic frameworks, [Zn(btze)]n (1), [Zn(btze)(H2O)]n (2) and [Mn(btze)(H2O)4]n·(H2O)2 (3) [btze = 1,2-bis(tetrazol-5-yl) ethane anion], were synthesized and characterized by elemental analysis, IR spectroscopy, X-ray crystallography and thermogravimetric analysis. The crystal structures study reveal that 1 displays a 3D framework, 2 displays a 2D layer structure and 3 displays a 1D polymeric chain. The luminescence properties of 1-3 were investigated at room temperature in solid state.  相似文献

The synthesis, structure and spectral and redox properties of the copper(II) complexes [Cu(pmtpm)Cl2] (1) and [Cu(pmtpm)2](ClO4)2 (6), where pmtpm is the linear tridentate ligand 2-pyridyl-N-(2′-methylthiophenyl)methyleneimine containing a thioether and two pyridine donors, are described. Also, the mixed ligand complexes [Cu(pmtpm)(diimine)](ClO4)2 (2-5), where the diimine is 2,2′-bipyridine (bpy) (2), 1,10-phenanthroline (phen) (3), 2,9-dimethyl-1,10-phenanthroline (2,9-dmp) (4) or dipyrido-[3,2-d:2′,3′-f]-quinoxaline (dpq) (5), have been isolated and studied. The X-ray crystal structures of the complexes 1, [Cu(pmtpm)(2,9-dmp)](ClO4)24 and 6 have been successfully determined. The complex 1 possesses a trigonal bipyramidal distorted square based pyramidal (TBDSBP) coordination geometry in which three corners of the square plane are occupied by two nitrogens and thioether s of pmtpm ligand and the remaining equatorial and the axial positions by two chloride ions. The complex 4 contains a CuN4S chromophore also with a TBDSBP coordination geometry in which two nitrogens and the thioether sulfur of pmtpm ligand occupy three corners of the square plane. One of the two nitrogens of 2,9-dmp ligand is equatorially coordinated and the other axially to copper. On the other hand, the complex 6 is found to possess a square based pyramidal distorted trigonal bipyramidal (SPDTBP) coordination geometry. The CuN2S trigonal plane in it is comprised of the pyridine and imine nitrogens and the thioether sulfur of the pmtpm ligand. The pyridine nitrogens of the ligand occupy the axial positions and the second thioether sulfur remains uncoordinated. On long standing in acetonitrile solution the mixed ligand complexes 2 and 3 undergo ligand disproportionation to provide the corresponding bis-complexes of bpy and phen, respectively, and 6. The electronic and EPR spectral parameters and the positive redox potential of complex 4 are consistent with the equatorial location of the thioether sulfur in the square-based coordination geometry around copper(II). On the other hand, the higher g and lower A values and lower E1/2 values for the complexes 2, 3 and 5 are consistent with the axial coordination of the thioether sulfur. Also, the Cu(II)/Cu(I) redox potentials increase with increase in number of aromatic rings of the diimine ligand. The steric and electronic effects of the bidentate diimine ligands in orienting the thioether coordination to axial or equatorial position are discussed.  相似文献

Compounds containing a quinone moiety represent an important class of biologically active molecules that are widespread in nature, displaying anticancer, antibacterial, antimalarial, and fungicidal activities. In the course of designing 2,3-disubstituted-1,4-naphthoquinones derivatives as potential cysteine protease inhibitors, two naphtho[2,3-d]isoxazole-4,9-dione-3-carboxylates, 1a and 1b, were obtained. The antiapoptotic potential of 1a and 1b was then evaluated and compared to that of naphthoquinone 4. Primary rat hepatocytes were incubated with synthesized naphthoquinone derivatives and then exposed to the apoptotic stimulus camptothecin. Our results indicate that naphtho[2,3-d]isoxazole-4,9-dione-3-carboxylates 1a and 1b exerted a potent protective role in camptothecin-induced apoptosis in primary rat hepatocytes. Both 1a and 1b significantly increased cell viability, while reducing nuclear fragmentation, caspase-3, -8 and -9 activation, and cytochrome c release induced by camptothecin. In addition, 1a and 1b were shown to up-regulate Bcl-XL, a pro-survival member of the Bcl-2 family of proteins, which modulates the mitochondrial pathway of apoptosis. Similar protective effects of quinone derivatives were seen in HuH-7 and PC12 cells incubated with distinct apoptotic stimuli, such as camptothecin, TGF-β1, or rotenone. Our results suggest that naphtho[2,3-d]isoxazole-4,9-dione-3-carboxylates 1a and 1b may act as potent, cytoprotective agents, through modulation of apoptotic pathways.  相似文献

A tridentate NNO donor Schiff base ligand [(1Z,3E)-3-((pyridin-2-yl)methylimino)-1-phenylbut-1-en-1-ol = LH] in presence of azide ions coordinates with cobalt(II) and copper(II) ions giving rise to three new coordination complexes [Co2(L)21,1-N3)2(N3)2] (1), [Cu2(L)21,3-N3)]·ClO4 (2) and [(μ1,1-N3)2Cu5(μ-OL)21,1-N3)41,1,1-N3)2]n (3). The complexes have been characterized by elemental analysis, FT-IR, UV-Vis spectral studies, and single crystal X-ray diffraction studies. These complexes demonstrate that under different synthetic conditions the azide ions and the Schiff base ligand (LH) show different coordination modes with cobalt(II) and copper(II) ions, giving rise to unusual dinuclear and polynuclear species (1, 2 and 3) whose structural variations are discussed. Magneto-structural correlation for the very rare singly μ1,3-N3 bridged CuII-Schiff base dinuclear species (2) has been studied. In addition, the catalytic properties of 1 for alkene oxidation and the general catalase-like activity behavior of 2 have been discussed.  相似文献

A series of mononuclear manganese(III) complexes of formulae [Mn(L)(X)(H2O)] (1-13) and [Mn(L)(X)] (14-17) (X = ClO4, F, Cl, Br, I, NCS, N3), derived from the Schiff bases of 5-bromosalicylaldehyde and different types of diamine (1,2-diaminoethane, 1,2-diaminopropane, 1,3-diaminopropane and 1,4-diaminobutane), have been synthesized and characterized by the combination of IR, UV-Vis spectroscopies, cyclic voltammetry and by X-ray crystallography. The redox properties of all the manganese(III) complexes show grossly identical features consisting of a reversible or quasireversible MnIII/MnII reduction. Besides MnIII/MnII reduction, the complexes 4, 5, 10, 13 and 16 also show reversible or quasireversible MnIII/MnIV oxidation. A linear correlation has been found for the complexes 5, 7, 11 and 13 [Mn(L2)(X)(H2O)] (X = F, Cl, Br, I) when E1/2 [MnIII/MnII] is plotted against Mulliken electronegativities (χM). The effect of the flexibility of the ligand on redox potential has been studied. It has been observed that the manganese(II) state is stabilized with increasing flexibility of the ligand environment. The crystal structure of 6 shows an octahedral geometry.  相似文献

The synthesis of Al4R4 (R = C5(CH3)4H) (3) and the tetrahedral structure in the solid state are described. These results as well as the 27Al NMR spectra of 3 in solution are in line with the data obtained from DFT calculations. These calculations also support the failed observation of a monomeric AlR species in solution. Monomeric and tetrameric molecules of 3 are discussed with respect to those of (AlCp)4 (1) and (AlCp)4 (2). The increasing Al-Al bond strength from 1 to 3 and 2 from X-ray data is also supported by structural and energetic results from DFT calculations.  相似文献

Using a phosphorus based Mannich condensation reaction the new pyridylphosphines {5-Ph2PCH2N(H)}C5H3(2-Cl)N (1-Cl) and {2-Ph2PCH2N(H)}C5H3(5-Br)N (1-Br) have been synthesised in good yields (60% and 88%, respectively) from Ph2PCH2OH and the appropriate aminopyridine. The ligands 1-Cl and 1-Br display variable coordination modes depending on the choice of late transition-metal complex used. Hence P-monodentate coordination has been observed for the mononuclear complexes AuCl(1-Cl) (2), AuCl(1-Br) (3), RuCl2(p-cymene)(1-Cl) (4), RuCl2(p-cymene)(1-Br) (5), RhCl2(Cp)(1-Cl) (6), RhCl2(Cp)(1-Br) (7), IrCl2(Cp)(1-Cl) (8), IrCl2(Cp)(1′-Cl) (8′), IrCl2(Cp)(1-Br) (9), cis-/trans-PdCl2(1-Cl)2 (10), cis-/trans-PdCl2(1-Br)2 (11), cis-PtCl2(1-Cl)2 (12) and cis-PtCl2(1-Br)2 (13). Reaction of Pd(Me)Cl(cod) (cod = cycloocta-1,5-diene) with either 1 equiv. of 1-Br or the known pyridylphosphines 1′-Cl, 1-OH or 1-H gave the P/N-chelate complexes Pd(Me)Cl(1-Br-1-H) (14)-(17). All new compounds have been fully characterised by spectroscopic and analytical methods. Furthermore the structures of 4, 5, 10 and 16 · (CH3)2SO have been elucidated by single crystal X-ray crystallography. A crystal structure of the dinuclear metallocycle trans,trans-[PdCl2{μ-P/N-{Ph2PCH2N(H)}C5H4N}]2 · CHCl3, 18 · CHCl3, has also been determined. Here 1-H bridges, using both P and pyridyl N donors, two dichloropalladium centres affording a 12-membered ring with the PdCl2 units adopting a head-to-tail arrangement.  相似文献

Our series of competitive antagonists against the G-protein coupled receptor P2Y14 were found to be highly shifted in the presence of serum (>99% protein bound). A binding assay using 2% human serum albumin (HSA) was developed to guide further SAR studies and led to the identification of the zwitterion 2, which is substantially less shifted (18-fold) than our previous lead compound 1 (323-fold). However, as the bioavailability of 2 was low, a library of ester pro-drugs was prepared (7a-7j) and assessed in vitro. The most interesting candidates were then profiled in vivo and led to the identification of the pro-drug 7j, which possesses a substantially improved pharmacokinetic profile.  相似文献

In order to develop a fluorinated radiotracer for imaging of ??1 receptors in the central nervous system a series of (2-fluoroethyl) substituted spirocyclic piperidines 3 has been prepared. In the key step of the synthesis 2-bromocinnamaldehyde acetal 5 was added to piperidones 6 with various substituents at the N-atom. Unexpectedly, this reaction led to 2-benzoxepines 8, which were contracted with acid to afford the spirocyclic 2-benzofuranacetaldehydes 9. The best yields were obtained, when the transformations up to the alcohols 10 were performed without isolation of intermediates. Generally the (2-fluoroethyl) derivatives 3 have higher ??1 affinity and ??1/??2 selectivity than the corresponding (3-fluoropropyl) derivatives 2. The most promising candidate for the development as radiotracer is the (2-fluoroethyl) derivative 3a (WMS-1828, fluspidine, 1′-benzyl-3-(2-fluoroethyl)-3H-spiro[[2]benzofuran-1,4′-piperidine]), which shows subnanomolar ??1 affinity (Ki = 0.59 nM) and excellent selectivity over the ??2 subtype (1331-fold) as well as some other receptor systems. The novel synthetic strategy also allows the systematic pharmacological evaluation of intermediate alcohols 10. Despite their high ??1 affinity (Ki = 6-32 nM) and selectivity the alcohols 10 are 10-30-fold less potent than the bioisosteric fluoro derivatives 3.  相似文献

In our efforts to investigate the factors that affect the formation of coordination architectures, such as secondary coordination donors and pendant skeletons of the carboxylic acid ligands, as well as H-bonding and other weak interactions, two kinds of ligands: (a) 3-(2-pyridyl)pyrazole (L1) with a non-coordinated N atom as a H-bonding donor, a 2,2′-bipyridyl-like chelating ligand, and (b) four carboxylic ligands with different secondary coordination donors and/or pendant skeletons, 1,4-benzenedicarboxylic acid (H2L2), 4-sulfobenzoic acid (H2L3), quinoline-4-carboxylic acid (HL4) and fumaric acid (H2L5), have been selected to react with Mn(II) salts, and five new complexes, [Mn(L1)2(SO4)]2 (1), [Mn(L1)2(L2)] (2), [Mn(L1)(HL3)2] (3), Mn(L1)2(L4)2 (4), and [Mn(L1)2(L5)] (5), have been obtained and structurally characterized. The structural differences of 1-5 can be attributed to the introduction of the different carboxylic acid ligands (H2L2, H2L3, HL4, and H2L5) with different secondary coordination donors and pendant skeletons, respectively. This result also reveals that the typical H-bonding (i.e. N-H?O and O-H?O) and some other intra- or inter-molecular weak interactions, such as C-H?O weak H-bonding and π?π interactions, often play important roles in the formation of supramolecular aggregates, especially in the aspect of linking the multi-nuclear discrete subunits or low-dimensional entities into high-dimensional supramolecular networks.  相似文献

The organotin complex [Ph3SnS(CH2)3SSnPh3] (1) was synthesized by PdCl2 catalyzed reaction between Ph3SnCl and disodium-1,3-propanedithiolate which in turn was prepared from 1,2-propanedithiol and sodium in refluxing THF. Reaction of 1 with Ru3(CO)12 in refluxing THF affords the mononuclear complex trans-[Ru(CO)4(SnPh3)2] (2) and the dinuclear complex [Ru2(CO)6(μ-κ2-SCH2CH2CH2S)] (3) in 20 and 11% yields, respectively, formed by cleavage of Sn-S bond of the ligand and Ru-Ru bonds of the cluster. Treatment of pymSSnPPh3 (pymS = pyrimidine-2-thiolate) with Ru3(CO)12 at 55-60 °C also gives 2 in 38% yield. Both 1 and 2 have been characterized by a combination of spectroscopic data and single crystal X-ray diffraction analysis.  相似文献

Reactions of 2-(3,5-dimethylpyrazol-1-ylmethyl)pyridine (L1), 2-(3,5-diphenylpyrazol-1-ylmethyl)pyridine (L2), 2-(3,5-di-tert-butylpyrazol-1-ylmethyl)pyridine (L3) and 2-(3-p-tolylpyrazol-1-ylmethyl)pyridine (L4) with K2[PtCl4] in a mixture of ethanol and water formed the dichloro platinum complexes [PtCl2(L1)] (1), [PtCl2(L2)] (2), [PtCl2(L3)] (3) and [PtCl2(L4)] (4). Complex 1, [PtCl2(L1)], could also be prepared in a mixture of acetone and water. Performing the reactions of L2 and L3 in a mixture of acetone and water, however, led to C-H activation of acetone under mild conditions to form the neutral acetonyl complexes [Pt(CH2COCH3)Cl(L2)] (2a) and [Pt(CH2COCH3)Cl(L3)] (3a). The same ligands reacted with HAuCl4 · 4H2O in a mixture of ethanol and water to form the gold salts [AuCl2(L1)][AuCl4] (5) [AuCl2(L2)][Cl] (6) [AuCl2(L3)][Cl] (7) and [AuCl2(L4)][AuCl4] (8); however, with the pyrazolyl unit in the para position of the pyridinyl ring in 4-(3,5-dimethylpyrazol-1-ylmethyl)pyridine (L5), 4-(3,5-diphenylpyrazol-1-ylmethyl)pyridine (L6) neutral gold complexes [AuCl3(L5)] (9) and [AuCl2(L6)] (10) were formed; signifying the role the position of the pyrazolyl group plays in product formation in the gold reactions. X-ray crystallographic structural determination of L6, 2, 33a, 8 and 10 were very important in confirming the structures of these compounds; particularly for 3a and 8 where the presence of the acetonyl group confirmed C-H activation and for 8 where the counter ion is . Cytotoxicity studies of L2, L4 and complexes 1-10 against HeLa cells showed the Au complexes were much less active than the Pt complexes.  相似文献

The three-substituted dipyridyl ligand bis(3-pyridylmethyl)sulfide (L1) was prepared by the reaction of 3-(chloromethyl)pyridine hydrochloride with thioacetamide under basic conditions. L1 was reacted with CuI to give complexes with 1:2 and 1:1 molar ratios. Crystal structures of [(CuI)2(L1)] (1) and [CuI(L1)] (2) were determined. In complex 1 the CuI species formed a one-dimensional staircase polymer to which L1 was bound in a side-by-side fashion with π-π interactions between the ligands on each side. Complex 2 consisted of a one-dimensional ribbon polymer of metallomacrocycles formed from two L1 ligands bridging Cu2I2 dimers which were fused within the macrocyclic ring. The analogous disulfide ligand bis(3-pyridylmethyl)disulfide (L2) was prepared by oxidation of the corresponding thiol 3-(sulfanylmethyl)pyridine. L2 was reacted with CuI in 1:2 and 1:1 molar ratios and products isolated but only the 1:1 product was able to be crystallised. The crystal structure of [CuI(L2)] (3) consisted of a one-dimensional ribbon polymer of metallomacrocycles formed from two L2 ligands linked through Cu2I2 dimers. The difference in the metallomacrocycle linking between the related structures 2 and 3 was attributed to the difference in ligand conformation.  相似文献

Two bis(pyridyl) ligands: N,N′-bis(4-pyridyl)-1,4-benzenedicarboxamide (bpba) and N,N′-bis(3-pyridylformyl)imidazolidine-2-thione (bpit) have been designed and synthesized. Self-assembly of the linear ligand bpba and angular ligand bpit with HgI2 results in a one-dimensional zigzag polymer {[HgI2(bpba)] · 1.5CH3OH}n (1) and a binuclear metallamacrocycle [HgI2(bpit)]2 · 3H2O (2), respectively. Single crystal X-ray diffraction analyses reveal that in the crystal structure of 1 the linear ligand bpba is in transoid conformation, while in the crystal structure of 2 the angular ligand bpit coordinates with HgI2 in cisoid conformation, the different geometries of the two ligands attribute to forming the dissimilar frameworks of two HgI2 adducts. The determination of third-order nonlinear optical (NLO) properties of 1 and 2 in DMF solution shows that adducts 1 and 2 possess strong NLO self-focusing effect. The hyperpolarizability γ values of 1 and 2 are calculated to be 2.10 × 10−29 and 3.24 × 10−29 esu, respectively, which are comparable to those of some NLO materials. And the result indicates that the heavier atoms Hg and I together with the polymeric aggregation play an important role in determining their NLO properties.  相似文献

Two novel phosphino-phosphaferrocenes [η5-C5H4(CH2)nPPh2]Fe(η5-PC4H2-2,5-Cy2) (PP1: n=1; PP2: n=2) have been designed and prepared in order to clarify weak chelate effect in the previously reported (η5-C5H4CH2PPh2)Fe[η5-PC4H2-2,5-((-)-menthyl)2] (1). 31P NMR studies of reactions of PP1 with PdCl2(cod) (6) revealed that PP1 showed stronger tendency to coordinate to the PdII center in bidentate fashion compared to 1. On the other hand, chelate effect in PP2 was negligibly weak and a reaction of PP2 with 6 in a PP2/6 = 2/1 molar ratio gave a complex PdCl2(PP2)2 (10) cleanly in which PP2 coordinated to the palladium center at the PPh2 moiety as a monodentate ligand. X-ray crystal structure studies of chelate complexes PdCl2(PP1) (7) and PdCl2(PP2) (9) showed that 9 had deviations from an idealized geometry in the square planar complex which could be attributed to a larger chelate ring of PP2, while PP1 in 7 constructed nearly ideal geometry for the square planar complex.From comparison of the coordination behavior between 1, PP1, and PP2, it is concluded that steric bulk of (-)-menthyl groups in 1 is the main factor of the weak chelate coordination of 1.  相似文献
