Acetoin production by Bacillus amyloliquefaciens was used as a model of product feedback to develop a strategy to enhance the production of acetoin. To enhance the resistance of B. amyloliquefaciens to acetoin, an acetoin-tolerant mutant E-11 was screened by using adaptive evolution with acetoin stress as the selection pressure. When compared with the parent FMME044, the mutant E-11 exhibited superior fermentation performance as follows: (1) the mutant E-11 exhibited increased tolerance to high concentration of acetoin, and the specific growth rate was 265.2% higher than that of the parent FMME044 in medium containing 80 g/L acetoin; (2) acetoin production by the mutant E-11 reached 71.5 g/L at 44 h when cultured in a 7-L fermentor with 173 g/L glucose, and the acetoin concentration and productivity of the mutant E-11 were 39.6% and 14.4% higher than those of the parent FMME044, respectively; (3) the unsaturated fatty acid contents in the mutant E-11 were 64.8%, 37.8%, and 18.4% higher than those in the parent FMME044 when cultured in 0, 40, and 60 g/L acetoin, whereas the saturated fatty acid contents in the mutant E-11 were 9.5%, 13.9%, and 14.1% lower than those in the parent FMME044, respectively.  相似文献

Data about Bacillus cereus different enterotoxins including hemolysin (HBL), nonhemolytic enterotoxin (NHE), enterotoxin (T), and emesis-inducing thermostable enterotoxin (ETE) are summarized in the article. Information about synthesis of different diarrhea-inducing and emesis-inducing enterotoxins, methods of their purification, structure, functions, and mechanisms of action are presented. One hundred and thirty-seven strains of Bacillus thuringiensis and 35 strains of Bacillus cereus were tested for the presence or absence of 99 traits. An analysis of these data indicated that strains of B. thuringiensis were indistinguishable from B. cereus, except for their ability to produce parasporal crystals. This conclusion was based on a comparison of the phenotypic properties of B. thuringiensis and B. cereus, as well as on the results of numerical analyses of the data which grouped strains into clusters on the basis of phenotypic similarity. In the resulting dendrograms, strains of B. thuringiensis and B. cereus were interspersed, exhibiting no tendency to segregate. In addition, with the exception of serovar israelensis, strains on B. thuringiensis belonging to the same flagellar serovar showed little or no tendency to group in different clusters. A comparison of the phenotypic differences between serovars indicated that the greater the number of strains in the serovars, the fewer, if any, phenotypic traits separating them. This suggests that the properties reported to differentiate serovars can be attributed to the internal phenotypic diversity of the species. Characterization of 10 mosquitocidal strains of Bacillus sphaericus indicated that the traits employed in this study readily distinguished these highly related organisms from strains of B. thuringiensis and B. cereus.  相似文献

Polyethyleneglycol induced fusion of Bacillus cereus protoplasts and its genetic consequences have been investigated. The technique used allows the transfer of a small plasmid pBC16 between Bacillus cereus cells. Fusion has resulted in isolation of hybrid cells having acquired TcR phenotype (harbouring pBC16) with high frequencies (10(-2)-10(-3)). However, the genetic instability of hybrid cells and segregation effects have influenced dramatically the final results of plasmid transfer. Nevertheless, the fusion of protoplasts proves to be useful in construction of BAcillus cereus strains inheriting plasmid determinants.  相似文献

3-羟基丁酮(Acetoin)作为一种重要的食用香料和平台化合物被广泛应用于医药、食品等领域。为改善解淀粉芽孢杆菌Bacillus amyloliquefaciens的3-羟基丁酮生产能力,采用常压室温等离子体(ARTP)和~(60)Coγ射线进行复合诱变,以3-羟基丁酮产量为分析指标,筛选获得最优突变株B.amyloliquefaciens H-5,3-羟基丁酮产量为68.2 g/L。为进一步实现3-羟基丁酮的高效生产,对此突变株进行5 L发酵罐水平的培养条件优化,并于30 L发酵罐上进行放大培养,最终3-羟基丁酮产量达85.2 g/L,较出发菌株B.amyloliquefaciens FMME088提高了26.8%。上述研究结果表明,ARTP和~(60)Coγ射线复合诱变能够有效获得高产菌株,该突变株具有较高的工业化微生物发酵生产3-羟基丁酮的潜能。  相似文献

Aims: To compare physical properties of spores that were produced in broth sporulation media at greater than 108 spores ml−1. Methods and Results: Bacillus atrophaeus reproducibly sporulated in nutrient broth (NB) and sporulation salts. Microscopy measurements showed that the spores were 0·68 ± 0·11 μm wide and 1·21 ± 0·18 μm long. Coulter Multisizer (CM3) measurements revealed the spore volumes and volume-equivalent spherical diameters, which were 0·48 ± 0·38 μm3 and 0·97 ± 0·07 μm, respectively. Bacillus cereus reproducibly sporulated in NB, sporulation salts, 200 mmol l−1 glutamate and antifoam. Spores were 0·95 ± 0·11 μm wide and 1·31 ± 0·17 μm long. Spore volumes were 0·78 ± 0·61 μm3 and volume-equivalent spherical diameters were 1·14 ± 0·11 μm. Bacillus atrophaeus spores were hydrophilic and B. cereus spores were hydrophobic. However, spore hydrophobicity was significantly altered after treatment with pH-adjusted bleach. Conclusions: The utility of a CM3 for both quantifying Bacillus spores and measuring spore sizes was demonstrated, although the volume between spore exosporium and spore coat was not measured. This study showed fundamental differences between spores from a Bacillus subtilis- and B. cereus-group species. Significance and Impact of the Study: This is useful for developing standard methods for broth spore production and physical characterization of both living and decontaminated spores.  相似文献

For the enumeration of vegetative cells and spores of Bacillus cereus in foods, a mannitol-egg yolk-phenol red-agar has been developed which exploits the failure of B. cereus to dissimilate mannitol, and the ability of most strains to produce phospholipase C. When a high degree of selectivity was required, polymyxin B sulfate in a concentration of 10 ppm appeared to be the most effective selective additive. Useful characteristics for the identification of presumptive isolates of B. cereus were found to be: morphology, dissimilation of glucose mostly to acetyl methyl carbinol under anaerobic conditions, hydrolysis of starch and gelatin, reduction of nitrate, and growth on 0.25% chloral hydrate agar.  相似文献
