Melatonin was found in the fresh water characeae Chara australis. The concentrations (~4 μg/g of tissue) were similar in photosynthesizing cells, independent of their position on the plant and rhizoids (roots) without chloroplasts. Exogenous melatonin, added at 10 μM to the artificial pond water, increased quantum yield of photochemistry of photosystem II by 34%. The increased efficiency appears to be due to the amount of open reaction centers of photosystem II, rather than increased efficiency of each reaction center. More open reaction centers reflect better functionality of all photosynthetic transport chain constituents. We suggest that melatonin protection against reactive oxygen species covers not only chlorophyll, but also photosynthetic proteins in general. 相似文献

Measurements of the electrical resistance and potential of thecell membranes of Chara australis have been made for over 60cells. In 17 cases the measurements extended over a period ofa few days for each cell. The most important results of theseobservations and calculations are as follows. The mean valueof the intercellular (between cells) resistance is about 1 kcm2. The transcellular (vacuole to external solution) resistanceconsists of two high-resistance layers. The mean value of theelectrical resistance of the outer layer is 7 k cm2. The capacityof the transcellular membrane equals about 2.5 µF cm2. 相似文献

Effects of cytoplasmic Ca2+ on the electrical properties ofthe plasma membrane were investigated in tonoplast-free cellsof Chara australis that had been internally perfused with media,containing either 1 mM ATP to fuel the electrogenic pump orhexokinase and glucose to deplete the ATP and stop the pump. In the presence of ATP, cytoplasmic Ca2+ up to 2.5?105M did not affect the membrane potential (about -190 mV), butmembrane resistance decreased uniformly with increasing [Ca2+]i.In the absence of ATP, the membrane potential, which was onlyabout -110 mV, was depolarized further by raising [Ca2+]i from1.4?106 to 2.5?105 M. Membrane resistance, whichwas nearly the twofold that of ATP-provided cells, decreasedmarkedly with an increase in [Ca2+]i from zero to 1.38?106M, but showed no change for further increases. Internodal cellsof Nitellopsis obtusa were more sensitive to intracellular Ca2+with respect to membrane potential than were those of Charaaustralis, reconfirming the results obtained by Mimura and Tazawa(1983). The effect of cytoplasmic Ca2+ on the ATP-dependent H+ effluxwas measured. No marked difference in H+ effluxes was detectedbetween zero and 2.5?105 M [Ca2+]i; but, at 104M the ATP-dependent H+ efflux was almost zero. Ca2+ efflux experimentswere done to investigate dependencies on [Ca2+]i and [ATP]i.The efflux was about 1 pmol cm2 s1 at all [Ca2+]iconcentrations tested (1.38?106, 2.5?105, 104M).This value is much higher than the influx reported by Hayamaet al. (1979), and this efflux was independent of [ATP]i. Thepossibility of a Ca2+-extruding pump is discussed.

The membrane potential and the ionic activity gradients of K+and Cl across the surface membrane of cytoplasmic dropletsprepared from Chara australis internodal cells, were measuredin high and low ionic strength bathing solutions using liquidion exchange microelectrodes selective for K+ and Cl.Our results indicate that K+ is close to electrochemical equilibriumwhereas Cl is not.

Some aspects of the membrane physiology of the giant-celled alga Chara corallina have been studied using the techniques of membrane noise analysis. Cellular voltage noise was measured by means of a microelectrode inserted in the vacuole of the cell. The significant feature of spectra estimated from voltage noise signals in these cells was a low-frequency component possibly associated with the active, electrogenic proton pumping which occurs in these cells. The plasmalemma action potential was found to be indispensableto the production of the tonoplast action potential. In a solutionlacking Ca2+ and containing other divalent cations such as Ba2+,Mg2+ or Mn2+, the plasmalemma excited in Nitella but did notin Chara. In Nitella, however, both the tonoplast action potentialand EC-coupling were abolished due to depletion of Ca2+ fromthe external medium. Ca2+ ions injected into the cytoplasmiclayer caused a transient change in both plasmalemma and tonoplastpotentials. These results suggest that a transient rise in Ca2+concentration during excitation of the plasmalemma may triggerthe tonoplast action potential. The regulation of ammonia uptake was investigated in internodalcells of the freshwater alga Chara australis. Ammonia uptakewas estimated by monitoring (i) its depletion from the bathingsolution, (ii) the uptake of radiolabelled methylamine, an analogueof ammonia, and (iii) depletion of ammonia in the unstirredlayer with the microelectrode ion-flux estimation technique(MIFE). Distribution of methylamine (14CH3NH3+) between thevacuole and cytoplasm was estimated with efflux analysis. Whencells were bathed continuously in solutions containing ammoniaor methylamine, the uptake rates of both amines decreased over12 to 48 h despite the continuing existence of a large electrochemicalgradient favouring influx of the NH+4 and CH3NH+4 cations. Treatmentwith 1.0 to 10.0 mM MSX, an inhibitor of glutamine synthetase,caused the internal ammonia concentration to rise and reducedthe subsequent uptake of ammonia and methylamine by up to 70%within 2 h. These results suggest that the permease facilitatingNH+4/CH3NH+4 influx is under feedback or kinetic regulationfrom either internal ammonia or an intermediate of nitrogenassimilation. Treatment with metabolic inhibitors (CCCP, azide and DCMU) andsome weak acids (DMO and butyric acid) for 30 to 60 min inhibitedmethylamine uptake, but the changes in the electrical potentialdifference across the plasma membrane could not account forthe magnitude of inhibition. The rate of cytopiasmic streaming,which is an indicator of the cellular ATP concentration in Chara,was inhibited by many of these treatments. However, under certainconditions of external pH and concentration, butyric acid couldreversibly inhibit ammonia and methylamine uptake without affectingcytoplasmic streaming, demonstrating that a decrease in cytoplasmicATP concentration was not responsible for the inhibition. Theeffect of butyric acid was rapid, causing a 60% inhibition ofuptake in 15 min. We conclude that weak acids can inhibit theNH+4/CH3NH+4 permease by acidifying the cytoplasm and suggestthat this may also explain the effects of the metabolic inhibitorson ammonia and methylamine uptake. To characterize cellular fluidity and mechanical processes, we determined the viscous properties of the cytoplasm of Chara contraria rhizoids in vivo by injecting and displacing superparamagnetic particles. After injection and a 24-h recovery period, the particles were moved to different positions within the rhizoid by an external magnet. The system was calibrated with solutions of known viscosities. The viscosity was determined based on the velocity at which individual beads moved toward the external magnet. The viscosity of the cytoplasm varied with direction of measurement (i.e., was highly anisotropic) and also varied between sites. The highest viscosity was observed near the endogenous statoliths (139 mP·s parallel and 78 mP·s perpendicular to the rhizoid axis). Depolymerization of actin filaments with latrunculin B reduced the viscosity significantly except around the nucleus but did not change the overall viscosity pattern. Microtubule depolymerization with oryzalin reduced viscosity especially between the nucleus and the statolith zone. The data indicate that F-actin but not microtubules affects statolith sedimentation and that cytoplasmic viscosity may be important for the gravisensing system. 相似文献

Voltage noise analysis is applied to plasmalemma ion transport in Chara corallina. There is a component in the noise power spectrum that is probably associated with current fluctuations within passive transport channels, and another component that may be associated either with fluctuations in the number of open channels, or with active transport. The data allow the calculation of time constants that may be attributable to molecular level events in these transport processes. 相似文献

Using permeabilized characean cells in which the ionic conditionsat the cytoplasmic side of the tonoplast are easily controlled,effects of Ca2+ ion on tonoplast potential were examined. Whenthe cell was treated with 1 µM Ca2+, the tonoplast potential(EM became positive in a complicated manner in Chara corallinawhile it simply became negative in Nitella axilliformis. Whenthe cell was treated with 9-antracenecarboxylic acid, a Cl-channelinhibitor, Em became more negative and the response of Em toCa2+ was significantly suppressed. It is suggested that Ca2+activates Cl-channel at a low concentration and inactivatesat a higher one in C. corallina while it simply inactivate Cl-channelin N. axilliformis.

Certain inhibitors have been found to affect the low frequency spectral component of the electrical noise power spectrum in Chara corallina. Application of the ATPase inhibitor N,N′-dicyclohexylcarbodiimide removed the low frequency spectral component, strengthening the case that the component is produced by active proton pumping. Cytocholasin B, which inhibits cyclosis in internodes of C. corallina, removed the low frequency spectral component in a time-dependent fashion which was correlated with the cessation of streaming. The protonophore carbonyl cyanide m-chlorophenylhydrazone did not produce consistent effects on the low frequency spectral component in these cells. 相似文献

Giant-celled Characeae (Chara australis Brown), grown for 4 months on 12/12 hr day/night cycle and summer/autumn temperatures, exhibited distinct concentration maxima in auxin (indole-3-acetic acid; IAA), melatonin and serotonin about 4 hr after subjective daybreak. These concentration peaks persisted after 3 day pretreatment in continuous darkness: confirming a circadian rhythm, rather than a response to “light on.” The plants pretreated for 3 d in continuous light exhibited several large IAA concentration maxima throughout the 24 hr. The melatonin and serotonin concentrations decreased and were less synchronized with IAA. Chara plants grown on 9/15 hr day/night cycle for 4 months and winter/spring temperatures contained much smaller concentrations of IAA, melatonin and serotonin. The IAA concentration maxima were observed in subjective dark phase. Serotonin concentration peaks were weakly correlated with those of IAA. Melatonin concentration was low and mostly independent of circadian cycle. The “dark” IAA concentration peaks persisted in plants treated for 3 d in the dark. The plants pretreated for 3 d in the light again developed more IAA concentration peaks. In this case the concentration maxima in melatonin and serotonin became more synchronous with those in IAA. The abscisic acid (ABA) and jasmonic acid (JA) concentrations were also measured in plants on winter regime. The ABA concentration did not exhibit circadian pattern, while JA concentration peaks were out of phase with those of IAA. The data are discussed in terms of crosstalk between metabolic pathways. 相似文献

When an exogenous protein, bovine serum albumin, was introduced into the vacuole of a Chara australis internodal cell, it was degraded with time. This degradation proceeded only in the vacuole as far as could be observed by sodium dodecylsulfate-polyacrylamide gel electrophoresis. Degradation was inhibited by protease inhibitors such as antipain and leupeptin. Endogenous proteins introduced into the vacuole were also degraded there. Furthermore, intravacuolar cytoplasmic drops, which were often formed by cell ligation, seemed to be degraded in the vacuole. However, bovine serum albumin degradation did not proceed when mixed with isolated vacuolar sap. These results show that the vacuole in the Chara internodal cell has the capacity to degrade cellular proteins, but that cytoplasmic support is needed for this degrading activity to be maintained. 相似文献

Both parts of the actin-myosin complex involved in cytoplasmic streaming could be regulated by mineral ions. The main goal of this study was to find a relationship between cyclosis and ion transport across the cell wall and plasma membrane. The transport of K(+) and Ca(2+) along pH bands in Chara branchlet internodal cells was characterized by using the MIFE system for non-invasive microelectrode measurement of ion fluxes. Branchlets formed acidic and alkaline bands with the pH ranging from 5 to 8. Different pH patterns were observed for different sides of the branchlets. Sides with cyclosis streaming acropetally generally showed greater variation in the profiles of pH and H(+) fluxes. Although a high correlation was not found between pH bands and Ca(2+) or K(+) fluxes, there was a positive correlation between Ca(2+) and K(+) fluxes themselves for both sides of the branchlets. Application of cytochalasin D, an inhibitor of cyclosis, had no immediate effect on pH and ion fluxes, however, the time of cyclosis cessation corresponded with a dramatic change in Ca(2+) and K(+) fluxes; pH profiles and H(+) fluxes were affected within 2 h. The evidence suggests that, in Chara branchlets, pH band formation and Gd(3+)-insensitive Ca(2+) transport systems are linked to the cyclosis machinery: (i) the pH band amplitude for the acropetally streaming side was larger than that for the basipetally streaming side; (ii) cessation of cytoplasmic streaming after cytochalasin D application resulted in changed pH banding profiles and H(+), Ca(2+) and K(+) fluxes; and (iii) the application of GdCl(3) or incubation in GdCl(3) solutions did not lead to the cessation of cytoplasmic streaming, although external Ca(2+) fluxes changed. 相似文献

Aminopeptidase II, one of the two major aminopeptidases in the giant alga Chara australis, was partially purified. Its molecular weight was estimated to be about 80,000 by gel permeation chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis showed that it is composed of a single polypeptide with a molecular weight of about 85,000. Aminopeptidase II hydrolyzed alanine-2-naphthylamide more efficiently than the naphthylamides of lysine and proline, and only weakly hydrolyzed the naphthylamides of arginine, phenylalanine, valine, and leucine. The optimal pH for the hydrolysis of alanine-2-naphthylamide was near 7.0. The activity of aminopeptidase II was inhibited by the SH-reagents p-chloromercuribenzoic acid and N-ethylmaleimide and by the metal chelator 1,10-phenanthroline. 相似文献

The internodal cell of Chara australis was made tonoplast-freeby internal perfusion with a medium containing a Ca2+-chelatorEGTA and the net H+ efflux across the plasma membrane was estimatedeither by titration of the external medium or by measuring thechange in pH in the external medium. The use of the low frequency (0.11.0 Hz) power spectraldensity function of the vacuolar resting potential is here appliedto the examination of a number of conditions which might beexpected to influence the active transport of ions. Once again,spectra of the form A/fµif are encountered. µifand p, the total power in the band, are significantly reducedby cyanide and by cyanide + salicylhydroxamic acid, but notby O2 deprivation, µif and p are powerfully reduced byimidazole and reduced to a lesser extent by 1, 10-phenanthroleneand DCMU. The effects of darkness and deprivation of O2 and/orCO2 could not be told from the effects of darkness alone. Theresults of varying the external pH were not unambiguous butdo suggest that µif is maximal near pH 7.35 and that itfalls off for more acidic or more basic values. A polyacrylate chamber with three pools was developed to measurethe changes in electrical membrane resistance during generationof receptor potentials upon mechanical stimulation in Charainternodal cells. The membrane resistance decreased at the peakof the receptor potentials. The present results support thehypothesis that the receptor potential is generated by activationof ion channel(s).
