Spontaneous chromosome mutations in Truxaline grasshoppers   总被引:2,自引:0,他引:2  
Three distinct mutant conditions are described in single male individuals from three species of short horn grasshopper. Of these, one is an entire germ line mutant of Myrmeleotettix maculatus, heterozygous for a centric fusion between single M4 and M5 telocentric chromosomes. In contrast, the remaining two mutants are present in mosaic form. One is heterozygous for an L1-M4 interchange in Omocestus viridulus, the other tetrasomic for the M4 chromosome in Chorthippus parallelus which in addition is characterised by the inclusion of a supernumerary heterochromatic segment on one S8 homologue. Centric fusion in Myrmehotettix maculatus has neither disturbed the chiasma potential of the elements constituting the fusion multiple nor, has it apparently influenced the production of balanced gametes. The pattern of chiasma formation in the L1-M4 interchange multiple lends support to the contention that the process of chiasma formation originates near the distal end of chromosome arms in Omocestus viridulus. There is no interaction between the two mutant conditions of tetrasomy and the presence of supernumerary segments in Chorthippus parallelus. Moreover, because of the precocious nature of two of the four M4 homologues there is little tendency to form multivalents. The two M4 bivalents share a similar mean chiasma frequency.  相似文献

Four populations of Astyanax scabripinnis (Pisces, Characidae) were analyzed for B chromosome features. This species contains a metacentric macrochromosome (BM), similar in size to the first chromosome of the karyotype, besides two variant forms, a large submetacentric (BSM) and a small metacentric (Bm), both showing a reduced frequency. These variant forms were observed only in the populations from the Campos do Jordão region (São Paulo State, Brazil), although not present in all the populations sampled. The three B chromosome forms are heterochromatic and at least the BM and BSM are also GC-rich, as evidenced by C-banding and chromomycin A3 staining. In spite of the morphological similarity between the BM form and the first chromosome pair, they differ in the G-banding pattern, which does not favor a possible relationship among these chromosomes. FISH with a telomeric DNA probe evidenced signals on the terminal region of all chromosomes, including the Bs. Interstitial telomeric bands, indicative of some chromosomal rearrangements such as translocation or tandem fusion in the origin of the B chromosomes, were not observed. BSM and Bm are probable derivative B chromosome forms from an ancestral BM chromosome, showing a restricted occurrence and low frequency in the populations.  相似文献

The male chromosome complement of Psophus stridulus (L.) (Orthoptera: Acrididae) has been analyzed by using orcein staining, C-banding and silver impregnation. During spermatogenesis only one pair of autosomes (M9) shows an active nucleolar organizer region located in a C-banded constriction. There are other chromosome pairs with constrictions but these do not show nucleolar activity. The relationship between these constrictions and the C-banding pattern exhibited by this species is analyzed.In a sample of 83 males from five populations, two different supernumerary chromosomes were observed. Four males had a metacentric B-chromosome (Bm) similar in size to the sex chromosome and mitotically stable. Its meiotic behaviour indicates that it is an isochromosome. An additional small B-chromosome (B8) was also found in a single follicle of one individual carrying the Bm.A high rate of abnormal spermatids (macrospermatids) was scored in the individuals carrying B's. This proportion is notably higher in the follicle containing both the Bm and the B8.  相似文献

A complex and parallel pattern of polymorphism for heterochromatic supernumerary segments in the M7 and S8 chromosomes has been found in 14 populations of the meadow grasshopperChorthippus parallelus. Nine distinct karyotype classes for these two chromosome pairs occur though they are not equally represented in different populations. Populations differ also with respect to the frequency of supernumerary segments they contain. In all populations the presence of supernumerary segments leads to a significant elevation of mean cell chiasma frequency compared to individuals from the same population lacking such segments. The extent of the effect appears to differ in different populations. The observed frequencies of S8 karyotypes conform to the expectations of a Hardy-Weinberg distribution. Those of the M7, however, do not, and in all but one of the 14 populations there is a significant excess of homokaryotypes. In the Ashurst population 26% of the individuals sampled were characterised by germ-line polysomy for the M4 chromosome, either in the form of entire tetrasomics or more usually as mosaics ranging from tri- to hepta-somics. In all these polysomics the M4 chromosomes in excess of two were regularly heteropycnotic at first meiotic prophase from zygotene to diakinesis. As a consequence of this multivalents are rare. Extra M4 chromosomes do not modify the chiasma characteristics of the other chromosomes in the complement. Nor do they modify the action of the supernumerary segments in any way.  相似文献

The germination responsiveness of an F2 population derived from the cross Lycopersicon esculentum (UCT5) x L. pennellii (LA716) was evaluated for salt tolerance at two stress levels, 150 mM NaCl + 15 mM CaCl2 and 200 mM NaCl + 20 mM CaCl2. Individuals were selected at both tails of the response distribution. The salt-tolerant and salt-sensitive individuals were genotyped at 16 isozyme loci located on 9 of the 12 tomato chromosomes. In addition, an unselected (control) F2 population was genotyped at the same marker loci, and gene frequencies were estimated in both selected and unselected populations. Trait-based marker analysis was effective in identifying genomic locations (quantitative trait loci, QTLs) affecting salt tolerance in the tomato. Three genomic locations marked by Est-3 on chromosome 1, Prx-7 on chromosome 3, and 6Pgdh-2 and Pgi-1 on chromosome 12 showed significant positive effects, while 2 locations associated with Got-2 on chromosome 7 and Aps-2 on chromosome 8 showed significant negative effects. The identification of genomic locations with both positive and negative effects on this trait suggests the likelihood of recovering transgressive segregants in progeny derived from these parental lines. Similar genomic locations were identified when selection was made either for salt tolerance or salt sensitivity and at both salt-stress treatments. Comparable results were obtained in uni- and bidirectional selection experiments. However, when marker allele gene frequencies in a control population are unknown, bidirectional selection may be more efficient than unidirectional selection in identifying marker-QTL associations. Results from this study are discussed in relationship to the use of molecular markers in developing salt-tolerant tomatoes.  相似文献

Heterochromatic supernumerary segments, present in either or both the M7 and the S8 chromosomes of the Ashurst (AS) population of Chorthippus parallelus, lead to a significant elevation in mean cell chiasma frequency. This effect appears to be dosage independent within both the S8 and the M7 karyomorphs when these are considered separately. Neither is there any marked interaction between the two systems when they are combined with one another. The selective factors controlling the distribution of S8 karyomorphs within the population appear, however, to act quite independently of those which regulate the M7 karyomorph pattern. This is revealed in two ways. First, whereas the frequency of S8 karyomorphs conform to the expectations of a Hardy-Weinberg distribution those of the M7 do not. Secondly, though the pattern of distribution of S8 types is uniform throughout the AS population that of the M7 is not. This heterogeneity of M7 karyomorphs implies that the population is in some way internally stratified in terms of its breeding structure.  相似文献

The mode of inheritance of supernumerary segments located on three different chromosome pairs was investigated in controlled crosses with specimens of the grasshopper Chorthippus jacobsi. While extra segments located on chromosomes M5 and M6 showed Mendelian inheritance, that on S8 did not. Thus, the two supernumerary heterochromatic chromosome segments located distally on the S8 chromosome accumulated through non-Mendelian transmission through both sexes. The observed transmission patterns may be explained by gametic selection for spermatozoa carrying segmented S8 chromosomes, in addition to meiotic drive for segmented S8 chromosomes in heterozygous females. Natural populations of Dichroplus elongatus often exhibit diverse forms of supernumerary heterochromatin. In Tafi Viejo, Tucumán (Argentina), parallel polymorphisms for extra segments on pairs M6, S9 (proximal) and S10 (distal) and a mitotically unstable B chromosome were detected. The segments produce intrachromosome effects excluding chiasmata from their neigbourhood and displacing them to regions distal to the extra heterochromatin. The individuals bearing simultaneously the B and the segment on M6 show a decrease in the frequency of interstitial chismata (Xi) while those carrying only segments on M6 or S10 have an increase of this variable when compared with standard males. The between cell variance of Xi is also higher in M6 segment carriers. B individuals present an increase of abnormal spermatid frequency which may be explained by the simultaneous occurrence of two mechanisms: 1) lagging of supernumeraries and 2) physiological effects of B's affecting the meiosis even in cells lacking them. The intra and interchromosome effects may be important for the plasticity of populations allowing recombination to explore new chromosome regions. The B polymorphism seems to be stable which points to an advantage of B carriers and/or the occurrence of accumulation mechanisms counterbalancing their negative effects on fertility.  相似文献

The phototransformation of protochlorophyll(ide) (Pchl(ide)) to chlorophyll(ide) (Chl(ide)) can be demonstrated in a proplastid fraction from Euglena gracilis Klebs var. bacillaris Cori if appropriate conditions are employed. Pigments were measured fluorometrically in acetone extracts of cell or organelles. Pchl(ide) and the phototransformation to Chl(ide) are at their highest levels in cells grown in darkness on normal or low vitamin B12-containing medium (pH 3.5) to the late exponential phase (1.2–1.4 × 106 cells ml?1). Late exponential cells on low B12 medium yield a proplastid fraction that contains Pchl(ide) which is phototransformed to Chl(ide) when illuminated with red light (5.6 W m?2 for 4 min) in the presence of 10 mM Hepes, 20 mM TES, 0.5 mM potassium phosphate (pH 7.4), 70 mM sorbitol, 5 mM DTT, 5 mM ATP, 5 mM fructose-1, 6-bisphosphate, 10 mM malate and 2 mM MgCl2; intact organelles appear to be involved since deletion of osmoticum gives a lower activity, and addition of NAD(P)H is without effect. Phototransformation of Pchl(ide) to Chl(ide) in red light shows Bunsen-Roscoe reciprocity between fluence rate and duration of illumination. Although mitochondria are present, they do not appear to be involved since inhibitors of respiration and uncouplers of oxidative phosphorylation fail to block the phototransformation. The percentage phototransformation of Pchl(ide) to Chl(ide) in late exponential normal B12 cells is 61 ± 10, and is 52 ± 3 in low B12 cells. About 67% of the activity in low B12 cells is recovered in the proplastid fraction incubated with the complete incubation mixture in saturating light. In both types of cells and in the proplastid fraction, the stoichiometry of conversion of Pchl(ide) to Chl(ide) is about 1:1 (mol/mol).  相似文献

Structural changes involving fragmentation and the deletion of chromosome material have resulted in the production of morphological variants of the standard B chromosomes of Myrmeleotettix maculatus. Three distinct types are described — a considerably reduced Bst chromosome with a sub-terminal centromere and a large and small telocentric fragment. In addition, the Bst chromosome can itself give rise to a small telocentric fragment. The fact that such telocentric fragments have been found only in mosaic germ lines implies a lack of stability and an inability to perpetuate themselves between generations. The non-reciprocal translocation of one such fragment onto the short arm of a metacentric autosome does little to improve the efficiency of its transmission from one generation to another. The behaviour of autosomal fragments, discovered in both the M6 and S8 chromosomes of this species, parallels that of B fragments. Only the Bst chromosomes appear both mitotically and meiotically stable.  相似文献

Summary Rhizosphere population characteristics of two cultivars of spring wheat (Triticum aestivum L. emend Thell.), Rescue (R) and Cadet (C), and the related chromosome substitution lines, C-R5 B and C-R5 D, were investigated. Replacement of the chromosome pair, 5 B, in Cadet with 5 B from Rescue made many of the rhizosphere microbial characteristics of C-R5 B similar to or the same as those in the rhizosphere of the donor parent, Rescue. In contrast, substitution of the functionally related chromosome pair, 5 D, did not cause marked changes in the rhizosphere microbial population, demonstrating the specificity of the plant's control over factors governing the rhizosphere microbial environment.  相似文献

The chromosome complement of a local population of Astyanax scabripinnis in Brazil was investigated with emphasis on the study of the heterochromatin attached to the A-chromosomes and present in the macro B-chromosome. Analysis after C-banding, silver and CMA3 staining, incorporation of 5-bromo-2′-deoxyuridine and chromosome digestion with nine restriction endonucleases revealed that the heterochromatin in the B-chromosomes was different from that found in the A-chromosomes. A polymorphism due to the presence of a supernumerary heterochromatic chromosome segment was observed in the population investigated. Some aspects related to the origin of the heterochromatin polymorphism in Astyanax scabripinnis are discussed. Summary A class of mosaic endospersm involving the marker Su I was observed among the progeny of individuals hyperploid for the chromosome B 4 and genetically analyzed. The exceptional individuals showing mosaic endosperm were found when the hyperploid material was used as pollen source. While in some cases mosaicism was limited to the endosperm tissue, with no apparent consequences in the embryo, in others the mosaicism was transmitted to the progeny, which showed changes in the structure of the B 4 chromosome, with the formation of unstable chromosomes whose genetic behaviour was similar to that of ring chromosomes. This interpretation was cytologically confirmed. In other cases the B 4 chromosome analyzed in mosaic endosperm individuals underwent altered transmission frequencies or loss, suggesting that its original structure had been modified by breakage-fusion-bridge cycles. The changes in this chromosome revealed by the mosaic phenotype are discussed in relation to the original structure of the B chromosome and the B 4 hyperploid condition.The author dedicates the present paper to Prof. Marcus M. In the laboratory we examined the effect of pH (5–10 with one interval) on survival, reproduction, egg viability and growth rate (intrinsic growth rate—r m and population growth rate—r) of five Brachionus rotifer species (Bcalyciflorus, Bquadridentatus, Burceolaris, B. patulus and Bangularis). The pH was shown to exert a major influence on egg viability and growth rate (r m and r) for each species. The age-specific survivorship curves within a species were not significantly different at pH 6–10. The optimal pH for each species is near-neutral pH (pH 6–8), and the fecundity decreased as the pH deviated from these values. For each Brachionus species, there was no significant difference between age-specific fecundity curves at pH 7 and 8. At acid pH (pH 5 or 6) higher egg mortality was observed for each species. The r m and population r of the five Brachionus species incubated at different pHs were significantly influenced by pH. The pH supporting the highest r m or r was obtained at pH 6–8, but varied due to species. In this study Burceolaris and Bpatulus could tolerate a broad range of pH, while the populations of Bcalyciflorus, Bquadridentatus and Bangulari declined at acid conditions.  相似文献

Salmon eggs and unfed fry were planted in reaches (total length 2.8 km, mean width 4 m) of a Scottish stream between 1971 and 1977 and their subsequent progress was studied by sampling 16 sections (areas 38–126 m2) of the stream. Brown trout are the only fish which spawn in the stream, waterfalls and a dam near its mouth preventing adult salmon and sea-trout passing upstream. There were no restraints on the downstream movement of fish except in 1977, when a fry trap was operated. In 1971 and 1974 boxes each containing 300 eggs were buried in groups of 3–6. In other years fry were evenly distributed at 3.6–29.3 m?2. At the end of the first growing season, salmon occurred at decreasing population densities for a distance of 600 m below the plantings, but after two growing seasons there was little remaining indication of their pattern of dispersion when planted. Rates of survival between planting and the end of the growing season were 9.4–31%. Survival when eggs were planted (11.1–14.8%) was not affected by the numbers planted together at one point (900–1800) or the distance apart of groups of boxes (10–85 m). When fry were planted the instantaneous mortality rate (M) of the 0+ salmon during their first growing season was related to the initial stocking density (Dp) by the formula M= 0.00637 + 0.00444 log10Dp. Twenty-two to 88% of 0+ salmon present at the end of the growing season were still surviving in the stream as 1+ fish one year later. In 1973–1976 only a small number of 2+ salmon occurred, the majority having migrated between the end of the second growing season and the following spring. There were more 2+ salmon in 1977 and 1978 resulting from higher stocking densities in 1975 and 1976 and slower growth. Trout of several age classes were present but their population densities were never high (<0.6 m?2). Salmon reached a greater size than trout by the end of the first growing season. Their mean weight (Wo, g) at this time was inversely related to their population density (Do No. m?2) and the biomass (B1, g m?2) of 1+ salmon present, giving the relationship log10wo= 0.6584–0.0558 D0-0.0352B1. The mean weight of 1+ salmon tended to be highest in sections where the 0+ salmon had reached a relatively large size the previous year. When a reach of the stream was planted twice (11 and 30 May 1977) with salmon fry (total 13.9 m?2) at the same stage of development, M during the first growing season was 0.0099 per day. This was less than that of fry in a control (M= 0.0107) where the stocking density was lower (6.8 m?2) and also less than in previous years when single planting rates of approximately 14 m?2 were used (M=0.0115). The double planting resulted in a wide range of lengths of 0+ salmon in September and the highest biomass values encountered during all experiments.  相似文献

The osmotic pressure equation for nonideal, associating systems of the type nA +mB ? AnBm, has been derived, by using the assumption yA nB m/yA nyB m = 1. This treatment can also be applied to related associations such as nA + mB ? AB + AB2 + A2B + …. From osmotic pressure experiments on the pure reactants it is possible to obtain the molecular weights (MA and MB) of the reactants and also the virial coefficients (BAA and BBB) of the reactants. The osmotic pressure of a nonreacting mixture of A and B can be calculated from these measurements. It can be used along with osmotic pressure measurements on equilibrium mixtures of A and B to obtain expressions containing the equilibrium constant (or constants) and the cross-virial coefficients (BAB and BBA). Several procedures are described for the evaluation of the equilibrium constant (or constants) and the BAB or BBA terms. It appears that this procedure is a general one which is applicable to associations of the type nA + mB ? AB + A2B + AB2 + …. By correcting for nonideal behavior, one should then be able to apply it to any method available for analyzing ideal associations of the types considered here. In addition it is possible, subject to certain restrictions, to analyze associations of the type 3A + B ? A2 + AB.  相似文献

The kinetics of the inhibition of photophosphorylation in chloroplasts from spinach (Spinacia oleracea) was investigated with 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) in small concentration intervals, starting at 10-7M. Plots of the reciprocal of photophosphorylation against concentration of DCMU gave essentially the same straight line with 2 mM nicotinamide adenine dinucleotide phosphate (NADP) together with saturating amounts of ferredoxin or with 4 mM K3Fe(CN)6 as the final acceptors for electrons. Practically complete inhibition was obtained at 3 x 10-6M DCMU. With 0.1 mM flavin mononucleotide (FMN) and ferredoxin, the inhibition between 10-7M and 10-6M DCMU was a little slower than in the other two cases. At 10-6M DCMU a break occurred to a new straight line in the plots, indicating that another reaction was inhibited. Total photophosphorylation without DCMU was about 77 μmol ATP per mg chlorophyll and hour. At the breaking point 20% remained, and inhibition was not complete even at 8 x 10-6M DCMU. The inhibitor constant for the high-DCMU reaction was in the order of 2 x 10-5M; for the low-DCMU reaction some complication made the “constant” appear negative. With phenazine methosulfate (PMS) added, DCMU was without effect on photophosphorylation. – As earlier shown by us, titration curves for intact cells of the microalga Scenedesmus show the break at 10-6M DCMU; and above 6 x 10-6M photophosphorylation in the algae is not further decreased by DCMU. The data are compared and their possible significance is discussed.  相似文献

A study has been made on the genetic basis of meiotic drive at the Distorter (D) locus which, in coupling with the male-determining gene (or region) M on the Y chromosome, causes production of excess male progeny. Its effect is regulated by the sensitivity/resistance of the X chromosome. This study demonstrates that there are two major loci controlling resistance/sensitivity to MD: (1) the m gene (or region) on the X chromosome (allelic with M) which may be either m R or m S (resistant or sensitive), (2) the t (tolerance) gene (or genes) which recombines with m and, if present, largely counteracts the effect of m S . There is also evidence that MD itself is capable of limited adaptation.The conclusions were derived from using MD males of the T30 or ACCRA strains (from Trinidad and Ghana respectively). The work involved the use of the CHIPEI and RED strains with sensitive X chromosomes, the latter also carrying the t (tolerance) gene which is linked to re (red eye) and m (the sex-determining locus or region) but recombines with both. The implications of these findings for using MD as a method of population control are discussed.  相似文献

In a sample of 85 males of the grasshopper Melanoplus femur-rubrum, three different supernumerary chromosomes (B-chromosomes) were observed: a medium-sized metacentric, BM, present in 12.9% of the males and two small telocentrics, BT and Bt, present in 8.2% and 3.5% of the males, respectively. The BM element is mitotically stable, while the BT and Bt elements are mitotically unstable, exhibiting numerical variation among testicular follicles. The mean number of B's per male (testis) is 1.00, 1.62, and 1.38 for the BM, BT, and Bt elements, respectively. A study of chiasma frequency among A-chromosomes revealed that there are no significant differences among the zero B and various B classes either for chiasma frequency or for the variation in chiasma frequency between individuals. There was, however, evidence for a higher within-individual variation in B-containing males when these were pooled. Cells with two Bt-chromosomes were regularly missing the smallest pair of A-chromosomes, suggesting that the Bt element induces non-disjunction of a pair of A-chromosomes and, possibly, that the Bt-chromosome is derived from the missing A-chromosome.Portions of this study were submitted by R.
