The supply of Gelidium resources is dependent on collection from natural habitats. As these resources are very limited, one possible means of augmenting production is by implementation of artificial cultivation as has been done with Laminaria, Porphyra and Eucheuma. Chinese phycologists have conducted research on sporeling and field cultivation of Gelidium for many years. Trials have been made using: (1) raft cultivation based on vegetative propogation of thallus fragments, (2) cultivation based on spore-collecting and land-based tank culture to provide seedstock, (3) cultivation based on regeneration from small thallus fragments to provide seedstock. Some of these results have been promising, but development remains at the experimental stage and methods of cultivation need to be improved. This paper is an updated review of the research and development on artificial sporeling production and field cultivation of Gelidium in China.  相似文献

Currently, Gelidium and Pterocladia (Gelidiales) are collected or harvested only from the sea. Despite several attempts to develop a cultivation technology for Gelidium, no successful methodology has yet been developed. Initial steps towards developmental efforts in Portugal, Spain, South Africa and Israel have been published. More developments have probably been performed but have not been published. Two different technological concepts have been tested for Gelidium cultivation: (1) the attachment of Gelidium fragments to concrete cylinders floating in the sea, and (2) free-floating pond cultivation technology. These vegetative cultivation technologies might be partially optimized by controlling physical, chemical and biological growth factors. The pond cultivation technology is the much more controllable option. The effects of all factors are discussed in detail in this review. It seems that the main difficulty with cultivation of Gelidium is its low growth rate. The claimed yields of the two technologies are far from being economically attractive at this stage of their development. It seems that in order to introduce Gelidium into commercial cultivation, major efforts in genetic improvement through selection or genetic engineering will be required. Only high yield strains will have the potential to compete economically with the present harvesting tradition. However, accumulated experience with genetic improvement of other useful seaweed species suggests that this is possible.  相似文献

Surveys were conducted in the cold desert environment of the Lahaul valley in the northwestern Himalaya for assessing the past and present status of Kuth (Saussurea lappa) cultivation. The findings reveal that this age-old practice now is in bottleneck. Main factors responsible for this setback to the species were the lengthy cultivation cycle, small land holdings, and even fluctuating and relatively low market prices. Owing to these constraints farmers have now started replacing cultivation of this threatened herb with pea (Pisum sativum L.), potato (Solanum tuberosum L.) and hop (Humulus lupulus L.). These crops obtained popularity due to comparatively more economic returns as well as their easy adaptability to the short growth season of the cold desert environment. Kuth cultivation in this region is among the interesting examples of domesticating wild medicinal herb by some innovative farmers during the 1920s. However, in the recent past farmers have been less interested to continue this practice due to its larger cultivation cycle, more profits with cash crops like pea and potato, and permit formalities at the time of export from the valley. In addition to being the oldest cash crop in the cold desert environment, Kuth is an endangered medicinal herb that has to be conserved on a priority basis. This study attempts to find out potential measures such as regular revision of market rates, development of existing uncultivable land under medicinal plant cultivation and strengthening the marketing network through establishment of federations of farmers at village level to revive cultivation of this important species.  相似文献

通过温室盆栽试验,研究珊瑚树对土壤中镉(Cd)的耐受和富集特征。研究结果表明,珊瑚树对污染土壤中Cd具有较强的耐受能力。培养56 d内,土壤中Cd含量对珊瑚树生物量影响不明显;随着培养时间的延长(105—203 d),珊瑚树的生长明显受到土壤中Cd的抑制作用。与对照处理(土壤中Cd含量为3.6 mg/kg)相比,培养154 d后,土壤中Cd含量为24.6 mg/kg处理下珊瑚树叶片中叶绿素a、叶绿素b、类胡萝卜素和丙二醛(MDA)含量没有明显变化;培养203 d后,土壤中Cd含量为24.6mg/kg处理下珊瑚树叶片中类胡萝卜素和MDA含量无明显变化,但叶片中叶绿素a和叶绿素b含量明显受到抑制,从而导致珊瑚树叶片的生长明显受到抑制(P0.05)。培养203 d后,珊瑚树对土壤中Cd的富集系数和转运系数均大于1,表明珊瑚树对土壤中Cd具有一定的富集和转运能力。上述结果表明,珊瑚树对Cd污染土壤具有一定的生态修复潜力。  相似文献

A total of 1949 cell cultures was tested for contamination with mollicutes by cultivation on and in mycoplasma media, 25.7% of the cell cultures was positive, 243 strains of Mycoplasma hyorhinis were isolated. Furthermore, mainly M. arginini and M. orale were detected, less often Acholeplasma laidlawii, M. fermentans and M. pneumoniae. Optimal conditions for isolation were discussed. About one third of 217 hybridoma cultures and two third of 57 myeloma cultures proved to be contaminated, all with M. hyorhinis. A DNA fluorochrome staining method (DAPI-test) was compared to cultivation for testing 1039 cell cultures. The efficiency of the DAPI-test could be estimated to be about 96% that of cultivation about 89%, but cultivation is more specific. The highest assurance is obtained when both methods are applied.  相似文献

The production of optically pure lactic acid in a high yield from xylose or a mixture of xylose and glucose, which is a model hydrolysate of lignocellulose, is described. In a single cultivation, Enterococcus casseliflavus produced 38 g/l of lactic acid with an optical purity of 96% enantiomeric excess (ee) and 6.4 g/l of acetic acid from 50 g/l of xylose when MRS medium was used. When a mixture of 50 g/l of xylose and 100 g/l of glucose was used as the carbon source in a cultivation of E. casseliflavus alone, glucose was converted to lactic acid in the early phase of the cultivation but xylose was hardly consumed. In a co-cultivation where E. casseliflavus and Lactobacillus casei specific for glucose were simultaneously inoculated, little or no lactic acid was produced after the glucose was almost consumed. A co-cultivation with two-stage inoculation (in which E. casseliflavus was added at a cultivation time of 40 h after L. casei cells were inoculated) resulted in complete consumption of 50 g/l of xylose and 100 g/l of glucose. In the co-cultivation, 95 g/l of lactic acid with a high optical purity of 96% ee was obtained at 192 h. Such a co-cultivation using two microorganisms specific for each sugar is considered to be one promising cultivation technique for the efficient production of lactic acid from a sugar mixture derived from lignocellulose.  相似文献

The compositions of free fatty acids (FA) in the mycelia of oyster cap (Pleurotus ostreatus (Jacq. ex Fr.) Kumm.) and honey mushroom (Flammulina velutipes (Curt. ex Fr.) Sing.) and the effect of mycelium cultivation conditions on the composition and proportions of individual FA were investigated. Palmitic and linoleic acids were found to be major acids produced byP. ostreatus growing on solid agar medium and in a submerged culture with a synthetic medium. The composition of minor FA in P. ostreatuswas dependent on cultivation conditions. Surface cultivation of its mycelium yielded pentadecanoic, octadecenoic, and stearic acids. Submerged cultivation additionally yielded undecanoic, myristic, hexadecenoic, and lignoceric acids. The composition of free FA in F. velutipesshowed no significant differences from that of P. ostreatus. Variation in the C/N ratio in the cultivation medium affected both the FA composition in P. ostreatus and F. velutipes and the relationship between saturated and unsaturated FA.  相似文献

金毛狗[Cibotium barometz(L.)J.Sm.]是珍稀观赏蕨类的重要类群,为国家二级重点保护野生植物。该研究以金毛狗孢子为试验材料,探究培养密度、外源赤霉素以及光质等不同环境因子对金毛狗配子体性别分化的影响,为金毛狗人工繁育和蕨类植物配子体性别决定机制研究提供技术支持。结果表明:(1)低配子体培养密度(1个/cm2和5个/cm2)有利于颈卵器和雌配子体形成,随着配子体培养密度增加,颈卵器平均数量及雌配子体比率下降,精子器平均数量以及雄配子体和两性配子体比例增加,但配子体培养密度过高(80个/cm2)会导致大量无性配子体产生。(2)不同配子体培养密度下,随着培养时间延长,两性配子体比率均有增加,且增加幅度基本一致。(3)外源GA4显著抑制颈卵器和雌配子体形成,并显著促进精子器和雄配子体形成;外源GA3对金毛狗配子体性别分化没有显著影响。(4)白光、红光、蓝光等不同光质对金毛狗配子体性别分化未产生显著影响,但会影响配子体的发育和形态建成。  相似文献

Hermetia illucens larvae (black soldier fly larvae, BSFL) convert efficiently organic waste to high quality biomass. To gain knowledge on the specific functions of gut microbes in this process it is a prerequisite to culture members of the core gut microbiota. Two different cultivation strategies were applied here for this purpose, a dilution-to-extinction cultivation and direct plating using six different media to culture aerobic heterotrophic bacteria. A total of 341 isolates were obtained by the dilution-to-extinction cultivation and 138 isolates by direct plating from guts of BSFL reared on chicken feed. Bacterial isolates were phylogenetically identified at the genus level by 16S rRNA gene sequencing (phylotyping) and differentiated at the strain level by genomic fingerprinting (genotyping). The main proportion of isolates was assigned to Proteobacteria, Firmicutes (Bacilli), and Actinobacteria. Predominant genera discussed in literature as member of a potential BSFL core gut microbiota, Providencia, Proteus, Morganella, Enterococcus, Bacillus, and members of the family Enterobacteriaceae, were isolated. A high intra-phylotype diversity was obtained by genomic fingerprinting which was especially enhanced by the dilution-to-extinction cultivation. This study showed that the application of different cultivation strategies including a dilution-to-extinction cultivation helps to culture a higher diversity of the BSFL gut microbiota and that genomic fingerprinting gives a better picture on the genetic diversity of cultured bacteria which cannot be covered by a 16S rRNA gene sequence based identification alone.

Morels (Morchella, Ascomycota), which are some of the most highly prized edible and medicinal mushrooms, are of great economic and scientific value. Morel cultivation has been a research focus worldwide for more than 100 years, and the outdoor cultivation of morels has succeeded and expanded to a large scale in China in recent years. In this study, we review the progress in recent research regarding the life cycle and reproductive systems in the genus Morchella and the current state of outdoor cultivation. Sclerotia formation and conidia production are two important phases during the life cycle. The morel species cultivated commercially in America is M. rufobrunnea based on molecular phylogenetic analysis. The species currently cultivated in China are black morels, including M. importuna, M. sextalata and M. eximia. The field cultivation of morels expanded in the majority of the provinces in China with a yield of fresh morels of 0–7620?kg per ha. The key techniques include spawn production, land preparation and spawning, the addition of exogenous nutrition, fruiting management and harvesting. The application of exogenous nutrition is the most important breakthrough in the field of morel cultivation, but the mechanism remains unclear. It was estimated that the total amount of field cultivated fresh morels was ~500 t in 2015–2016. We also discuss the potential issues remaining in the current literature and suggest directions for future studies.  相似文献

The use of lettuce brine, a by-product of the vegetable fermentation industry, as a medium for yeast cultivation was investigated. Six strains of yeast, Saccharomyces sp., Pichia sp., Rhodotorula sp., Candida sp., Kluyveromyces sp. and Trichospora sp. grew well in diluted lettuce brine under aerobic conditions. The acid brine becomes neutral after yeast cultivation. The yeast strains reached the maximum growth after the first day of cultivation. Trichosporon sp. was found to grow best in the brine with the maximum specific growth rate at 0.09 h−1 and growth yield of 67%. Effects of a fungal endophyte, Fusarium mairei, on growth and paclitaxel formation of Taxus cuspidata cells were investigated by adding fungal endophyte culture supernatant (FECS) to suspension cultures of T. cuspidata cells. The main effective chemical responsible for paclitaxel formation in FECS was an exopolysaccharide (EPS) of molecular weight ~2 kDa. FECS fractions except EPS stimulated growth of Taxus cells but had no effects on paclitaxel accumulation. Additionally, elicitation efficiency of FECS based on different culture conditions was studied. EPS content in FECS was related to FECS culture conditions. FECS with long cultivation and high-aeration cultivation contained higher EPS content and resulted in higher paclitaxel yield than that with short cultivation and low-aeration cultivation. The maximum yield of paclitaxel from Taxus cultures, elicited by FECS with 9-day cultivation, was 4.7-fold that of the control cultures.  相似文献

The aim of this study was to analyze successional changes in the bacterial community over a period of 6 months of cultivation of Aplysina aerophoba sponges under different artificial cultivation conditions by use of denaturing gradient gel electrophoresis (DGGE). The cultivation conditions varied concerning the water temperature (20 ± 2 °C and 25 ± 2 °C) of the aquaria, additional illumination of one aquarium, and feeding of the sponges. Amplicons from DGGE separation of dominant colonizing or variably appearing bacteria were sequenced and aligned for taxonomical identification. In addition, secondary metabolites typically found in A. aerophoba were analyzed to investigate changes in the natural product profile during cultivation. The cultivation of sponges under any given condition did not lead to a depletion of their bacterial community in the course of the experiment. On the contrary, the distinctive set of associated bacteria was maintained in spite of a dramatic loss of biomass and morphological degradation during the cultivation period. Generally, all sequences obtained from the DGGE gels were related to bacteria of five phyla: Actinobacteria, Cyanobacteria, α-Proteobacteria, γ-Proteobacteria, and Chloroflexi. Despite the overall stability of the bacterial community in A. aerophoba, an unambiguous variability was detected for the CyanobacteriaA. aerophoba clone TK09”. This variability was ascribed to the predominant light conditions. The analysis of the metabolic pattern revealed that the concentration of a class of characteristic-brominated compounds typically found in A. aerophoba, like aeroplysinin-1, aerophobin-1, aerophobin-2, and isofistularin-3, increased over the 6 months of cultivation.  相似文献

Fritillaria pallidiflora Schrenk has been treasured in traditional classic medicine as an antitussive, antiasthmatic and expectorant for hundreds of years. With gradually decreasing wild F. pallidiflora resources, the herb can no longer satisfy the demand. Artificial cultivation is one of the most effective ways to solve the contradiction between supply and demand in the medicinal material market. During the growth of Rhizomes medicinal plants, root biomass and active ingredient content showed dynamic accumulated variation with increasing cultivation years. Up to now, hardly any attempts have been made to investigate the relationship between quality and cultivation years of F. pallidiflora. Therefore, in this paper, we determined the optimum harvesting time by comparing biomass and biological characteristics of F. pallidiflora at different cultivation times. High-performance liquid chromatography with evaporative light scattering detection and phenol-sulfuric acid visible spectrophotometry was performed to determine imperialine and polysaccharide content of F. pallidiflora bulbs. From year 1 to 6 of cultivation, we observed an upward trend in plant height, diameter and dry weight of F. pallidiflora, while water content decreased. Plant height and dry weight increased remarkably during the fourth year of cultivation. The content of imperialine and polysaccharide of F. pallidiflora bulbs, on the other hand, showed an upward trend from year 1 to 3, after which it decreased from year 3 to 6. By comparing plant growth, biomass development and the accumulation of imperialine and polysaccharide, the best harvesting time of F. pallidiflora was determined to be after 4 years of cultivation. Our results showed that it is possible to establish a “safe, effective, stable and controllable” production process, which could play an important role in achieving sustainable utilization of F. pallidiflora resources.  相似文献

To determine the stability and specificity of microbes associated with the marine cold-water sponge Geodia barretti during cultivation, we compared the microbial community of freshly retrieved specimens to that of cultivated explants by fluorescence in situ hybridization (FISH). G. barretti hosts a specific homogeneous microbial community in its mesohyl, which is maintained during a cultivation period of 8 months. In 10-day-old explants, bright colonies of unusually large bacterial cells, located predominantly at canal walls, were observed in addition to the common bacteria. Bacteria of the aberrant type included both lineages present in whole sponges and foreign ones, notably numerous genera of sulfate-reducing bacteria. We assume that these represent infectious bacteria that eluded the innate immune system of the sponge. Explants that resist these microbial attacks during the critical phase of cultivation eliminate infectious bacteria. The intrinsic microbial community of G. barretti is not affected by these infections and remains persistent over a cultivation period of at least several months.  相似文献

The yield and the quality of carrageenan depend, among other things, on the cultivar or strain and on the cultivation and processing techniques. This work presents carrageenan yields and some properties of Kappaphycus alvarezii under selected cultivation conditions i.e. cultivation period, depth and planting density. Growth rates (GR) ranged from 5.2–7.2% day−1, with the highest GR at 28 days, at 0–0.5 m depth, and planting density of 12 and 8.4 plants m−2. Highest productivity was observed in samples after 44 and 59 day cultivation period, which were grown at 0–0.5 m depth, and a planting density of 24 plants m−2. Carrageenan yields, iota fraction, viscosity, molecular weight and gel strength were measured. A cultivation period of 28 days during the winter had a significant higher carrageenan yield, while samples from 59 days showed a significantly higher iota fraction. Carrageenan also presented an increasing molecular weight under longer cultivation periods. A similar trend was observed for viscosity and gel strength. All samples cultivated in Brazil gave higher values when compared to a K. alvarezii commercial reference sample, with the exception of carrageenan yield values, which were lower in this study. Taking into account all parameters, the culture condition which provided the best carrageenan from a commercial perspective were 45 days of cultivation, growing at the surface, with a planting density of 12 plants m−2. Considering that this study was performed in the least favorable season (winter), these results indicate that the site is suitable for the implementation of commercial cultivation.  相似文献

An increase ofK L a, of a cultivation device achieved by increasing the specific output of mechanical energy for air dispersion and mixing may lead to cell damage and to changes in the cell cycle. Abstract

For a feasible microalgae biodiesel, increasing lipid productivity is a key parameter. An important cultivation parameter is light wavelength (λ). It can affect microalgal growth, lipid yield, and fatty acid composition. In the current study, the mixture design was used as an alternative to model the influence of the λ on the Dunaliella salina lipid productivity. The illumination was considered to be the mixture of different λ (the light colors blue, red, and green). All experiments were performed with and without sodium acetate (4?g/L), as carbon source, allowing the identification of the impact of the cultivation regimen (autotrophic or mixotrophic). Without sodium acetate, the highest lipid productivity was obtained using blue and red light. The use of mixotrophic cultivations significantly enhanced the results. The optimum obtained result was mixotrophic cultivation under 65% blue and 35% green light, resulting in biomass productivity of 105.06 mgL?1day?1, a lipid productivity of 53.47 mgL?1day?1, and lipid content of 50.89%. The main fatty acids of the oil obtained in this cultivation were oleic acid (36.52%) and palmitic acid (18.31%).  相似文献
