1. By the VAN WISSELINGH color reaction and the chitosan sulfatetest it was revealed that Chlorella cells contain chitosan probablyin their cell walls. 2. By fractionating the cell material into several fractionsfollowed by their hydrolysis, it was revealed that the majorityof glucosamine was present in the residue material remaininginsoluble in ethanol-ether and perchloric acid (PCA) solution.Conceivably, this glucosamine has derived, for the most part,from the chitosan contained in the cell wall material. 3. During the course of life cycle of the algal cells, the increasein content of glucosamine occurred in three steps: first, inproportion to the growth of smaller (young) cells into largercells; second, corresponding to the formation of autosporeswithin ripened cells; and third, in parallel with the growthof newly born daughter cells. 4. Between the first and second phase mentioned above, thereoccurred an abrupt breach in the increase of glucosamine. Thisphenomenon was presumed to be closely related to the profoundchange in the permeability of cell walls occuring at this transitionalstage of cell development. The lipids extracted from Chlorella cells at different developmentalstages were separated by chromatography on silicic acid into"nonpolar" (chloroform-eluate) and "polar" (methanol-eluate)lipid fractions. The lipids were also subjected to florisilchromatography to fractionate neutral glycerides and free fattyacids. Gas-liquid chromatographic analysis of these fractions has revealeda marked difference in their fatty acid compositions which werefound to undergo characteristic changes during the course ofalgal cell cycle. It was found that the fatty acids in the "nonpolar"lipid (fat) fraction are synthesized during the growth phasein the light and consumed during the process of cellular division. 用气相色谱仪分析了鲢受精卵和仔鱼的脂肪酸组成及其变化。受精卵中共检测到19种脂肪酸;SFA含量最高,其次为PUFA,MUFA最少。SFA中以16C:0为主,MUFA中以18C:1为主,PUFA中以DHA、AA和EPA为主;n-3/n-6为2.45。初孵仔鱼中HUFA因卵膜的脱落而减少,以后逐渐积累;鲢仔鱼具有将18C PUFA转化为长链HUFA的能力;在早期发育阶段,鲢仔鱼主要消耗16C:0作为能量代谢的基质。  相似文献

Uptake of amino acids is a complex process but in cells growing with ammonia as sole nitrogen source the initial uptake rate of amino acids is a measure of the transport capacity of the uptake system (permease). In synchronous cultures of Saccharomyces cerevisiae amino acids were transported at all stages of the cell cycle. However, for any one amino acid the initial uptake rate was constant for most of the cycle and doubled during a discrete part of the cycle. Thus, for a variety of amino acids the functioning amino acid transport capacity of the membrane doubles once per cycle at a characteristic stage of the cycle. Arginine, valine, and phenylalanine exhibit periodic doubling of uptake rate at different stages of the cell cycle indicating that the transport of these amino acids is mediated by three different systems. Serine, phenylalanine, and leucine exhibit periodic doubling of the uptake rate at the same stage of the cycle. However, it is unlikely that serine and phenylalanine share the same transport system since the uptake of one is not inhibited by the other amino acid. This phenomenon is analogous to the periodic synthesis of soluble enzymes observed in S. cerevisiae.  相似文献

为了研究许氏平鲉(Sebastes schlegelii)雌鱼体内受精后仔鱼开口前和仔鱼开口后两个阶段氨基酸和脂肪酸的组成变化规律, 采用生化常规方法定量测定并分析了许氏平鲉发育早期的受精卵(FE)、胚胎期(ES)、初产仔鱼(PL1)、前仔鱼期(PL2)、后仔鱼期(PL3)和稚鱼期(J)6个阶段的氨基酸和脂肪酸组成特点及含量变动。结果表明: 总氨基酸含量从FE至PL1显著下降, 至PL3显著上升, 至J又显著下降(P<0.05); 游离氨基酸含量以FE最低(12.77 mg/g), 从FE到PL1显著上升(P<0.05), 并在PL1含量达到最高值(92.19 mg/g), PL1发育到J呈现先下降后上升再下降的变化趋势(P<0.05), 游离氨基酸与总氨基酸的比值范围在2.37%—19.66%。在各发育阶段干样中检出碳链长度在C14-C24的29种脂肪酸, 分别为9种饱和脂肪酸(SFA)、9种单不饱和脂肪酸(MUFA)和11种多不饱和脂肪酸为(PUFA), 受精卵中主要脂肪酸依次为C22:6n-3(DHA)、C18:ln-9c、C16:0和C20:5n-3(EPA)。胚胎期(FE-ES)的脂肪酸利用率顺序为SFA、MUFA、n-6PUFA、n-3PUFA, 主要以C18:3n-3、C18:0、C16:1n-7及C20:5n-3(EPA)作为胚胎期的能量来源, C22:6n-3(DHA)的实际利用率最低(9.71%), 被优先保存下来, C16:0的实际利用量最高(10.94mg/g); 仔鱼内源营养阶段(ES-PL1)脂肪酸利用率顺序为MUFA、n-6PUFA、SFA、n-3PUFA, 主要以C16:1n-7、C18:0、C20:4n-6(ARA)及C18:1n-9c作为开口前仔鱼的主要能量来源, 其中仔鱼对DHA实际利用量最高(18.23 mg/g)。PL1-PL3阶段DHA相对于EPA和ARA被选择性利用; PL3至J阶段ARA相对于EPA和DHA被选择性利用。研究表明: 许氏平鲉仔鱼开口前阶段总氨基酸含量与游离氨基酸含量的变化趋势截然相反, 胚胎期与仔鱼内源营养阶段脂肪酸利用率和利用量均有所不同, 仔鱼期DHA优先被利用, 过渡至稚鱼期ARA优先被利用。建议在仔鱼开口后添加富含DHA生物性饵料, 仔鱼过渡到稚鱼期在配合饵料中添加ARA营养物质, 防止苗种营养不足, 保证成活率。  相似文献

  1. Using Chlorella ellipsoidea as material, investigations weremade of the effects of ultraviolet irradiation upon variousactivities of cells at different developmental stages in theirlife cycle. Cell activities investigated were photosynthesis,respira tion, over-all growth, modes of synchronous growth andcell division as well as the formation of nucleic acids. Theu. v.- light applied was 30 µµW/cm2in intensityand 2537 Å in wavelength.
  2. The most u. v.-sensitive wasthe over-all growth activity, andin this respect the irradiationapplied at the L2-stage wasmore inhibitive than that givenat the D-stage. The next mostvulnerable was the photosyntheticactivity, the sensitivitybeing the same in the D- and L-cells.The most resistant towardu.v. was the endogenous respirationof D-cells followed by theirrespiration using exogenous glucoseas substrate. The L2-cellsappeared to be unable to use exogenousglucose as substrateof respiration, but their endogenous respirationwas considerablystronger than that of D-cells, and its u. v.-sensitivitywasthe same as that of glucose respiration of D-cells.
  3. WhenD-cells were u. v. irradiated immediately before the startofsynchronous culture, growth and cell division as well astheformation of DNA and RNA were retarded in proportion totheu. v.-dose applied. The division number (n) was normal (around4) at lower u.v.-doses (1-2 minute irradiation), but was reducedto a half (about 2) at a higher dose.
  4. When, during the synchronousculture, 1-minute u.v.- irradiationwas applied at various stagesof the ripening phase, the divisionwas retarded, but the cells,after attaining an abnormally largesize, divided into about8. If the irradiation was given atthe L4-stage, the divisionnumber was practically unmodified(n=4.5), although the divisionwas somewhat retarded comparedwith that of the control culture.When a 1-minute irradiationwas given at the L2-stage, thereoccurred an apparent stimulationof DNA- and RNA-formation,a phenomenon which corresponds tothe production of a largernumber of daughter cells than itwas the case in control cultures.
  5. Thus the cells which were moderately u.v.-irradiated at differentstages of synchronous culture were able to complete their lifecycle, but later a certain portion of irradiated cells becameunable to grow normally.

  1. Starting with uniformly 32P-labeled Chlorella cells, a synchronousculture was run in a medium containing non-labeled phosphate.During the synchronous growth and division of the algal cells,the changes in amount of total and labeled P in various phosphatecompounds were followed.
  2. Characteristic changes were observedwith (acid-soluble) polyphosphate"A", nucleotidic labile phosphates,(acid-insoluble) polyphosphate"C", DNA-P and protein-P. Thelabeled phosphorus of polyphosphate"C" showed a decrease duringthe earlier phase of experiment,although a considerable uptakeof non-labeled P from the culturemedium into this compoundwas observed throughout the experiment.In parallel with theloss of labeled phosphorus in this compound,the increase oflabeled phosphorus occurred in polyphosphate"A", in the nucleotidiclabile-P compounds, and in DNA, suggestingthat these substancesreceived P from polyphosphate "C". Thelabeled P in polyphosphate"A" and in the nucleotidic labile-Pcompounds increased graduallywith the progress of culture,attained their maximum levelsat the stage of ripening, anddecreased markedly during theprocess of "post-ripening" anddivision of cells, indicatingthat these compounds were in activeturnover and playing someimportant roles in the process ofcell maturation and division.
  3. The total amounts of inorganic P, RNA-P and lipid-P increasedcontinuously throughout the experiment and showed no significantchange in the content of labeled P.

The cellular content of carbon, nitrogen, amino acids, polysaccharides, phosphorus and adenosine trtphosphate (ATP) was determined at several stages during the life cycle of the dinoflagellate Scrippsiella trochoidea (Stein) Loeblich. Carbon per cell decreased slightly between exponential and stationary phase growth in vegetative cells whereas nitrogen per cell did not change. Both of these cellular components increased markedly on encystment and then decreased to vegetative cell levels during dormancy and germination. C/N ratios increased gradually during cyst dormancy and activation, reflecting a more rapid decrease in N than in C pools, even though both decreased through time. Amino acid composition was relatively constant during the vegetative cell stages; glutamic acid was the dominant component. Arginine was notably higher in cysts than in vegetative cells but decreased significantly during germination, suggesting a role in nitrogen storage. The ratio of neutral ammo acids to total ammo acids (NAA/TAA) decreased as cysts were formed and then gradually increased during storage and germination. The ratio of basic ammo acids to total ammo acids (BAA/TAA) changed in the opposite direction of NAA/TAA, whereas the ratio of acidic acids to total amino adds (AAA/TAA) was generally invariant. Ammo acid pools were not static during the resting slate in the cysts: there was degradation or biosynthesis of certain, but not all, classes of these compounds. The monosacchande composition of cold and hot water extracted polysaccharides was quite different between cells and cysts. A high percentage of glucose in cysts suggests that the storage carbohydrate is probably in the form of glucan. Total cellular phosphorus was higher in all cyst stages than in vegetative cells. However, ATP-cell?1 decreased as vegetative cells entered stationary phase and encysted, and continued to decrease in cysts during dark cold storage. ATP increased only as the cysts were activated at warm temperatures in the light and began to germinate. The above data demonstrate that dormancy and quiescence are not periods of inactive metabolism but instead are times when numerous biochemical transformations are occurring that permit prolonged survival in a resting state.  相似文献

  1. Chlorella ellipsoidea was grown synchronously and the changesin content of various vitamins during the algal life cycle werefollowed either by chemical or microbiological assay methods.
  2. In terms of µg per gram of cell dry weight, the contentof some vitamins (niacin, biotin, inositol and choline) remainedalmost constant throughout the algal life cycle, while thatof others (vitamin B6-complex, pantothenic acid, folic acid,thiamine and riboflavin) was found to decrease more or lessmarkedly during the "growing phase" and increase at later phasesof "ripening". The content of p-aminobenzoic acid increasedonly at an early stage of "ripening", and that of ascorbic acidincreased only at the stages in which photosynthesis occurredmost actively.
  3. These results were discussed in an attemptto interprete theirrelationship with the previously reportedobservations pertainingto the physiological and biochemicalevents occurring in thelife cycle of the alga.

  1. Using intact cells of Chlorella, the effects of CO2 on thelevelsof oxidized and reduced forms of DPN and TPN in the lightandin the dark were investigated.
  2. It was found that the light-inducedchanges of the DPNH-levelwere not affected by the presenceor absence of CO2. On theother hand, the light-induced increaseof TPNH was suppressedin the presence of CO2 and the levelof TPNH which was raisedon illumination in the absence of CO2was lowered by the provisionof CO2.
  3. On the basis of thesefindings, it was concluded that TPNH,but not DPNH, is participating,in some way, in the mechanismof photosynthesis.
  4. Discussionswere made on the difference in the sites of participationofTPNH and of the photogenic reducing agent (R) in the pathofcarbon in photosynthesis.

  1. Previous work has demonstrated that when cells of Chlorellaprotothecoides are grown mixotrophically under illuminationin a medium rich in nitrogen source (urea) and poor in glucose,normal green cells are obtained, while in a medium rich in glucoseand poor in the nitrogen source, strongly bleached cells containingapparently no discernible chloroplast structures — called"glucose-bleached" cells — are produced either in thelight or in darkness. When the green cells are incubated ina glucose-enriched mineral medium without added nitrogen source,they are fairly rapidly bleached with concomitant degenerationof chloroplast structures (" bleaching "). When, on the otherhand, the "glucose-bleached" cells are transferred in a nitrogen-enrichedmedium without added glucose under illumination, they turn greenwith regeneration of chloroplasts (" greening "). In the presentstudy changes in contents of carbohydrate and fatty acid inalgal cells were followed during these processes of "bleaching"and "greening.".
  2. During the process of "bleaching", the quantityof glucose existingin the insoluble carbohydrate fraction ofalgal cells increasedrapidly and markedly. A considerable increasewas also observedin the contents of cells in oleic, linoleicand palmitic acids.It was noted, however, that linolenic aciddecreased in quantityduring the most active phase of cell bleaching.
  3. During the process of "greening", the glucose in the insolublecarbohydrate fraction rapidly decreased, suggesting that itis utilized, as carbon and energy sources, for the chloroplastregeneration. Linolenic acid was found to be synthesized inparallel with formation of chlorophyll. A peculiar pattern ofchange in contents was observed with oleic and palmitic acids,which was interpreted as being related with the process of cellulardivision occurring incidentally during the process of greening.

  1. Using the technique of synchronous culture, investigationsweremade of the effects of temperature and light-intensityon cellularlife cycle of Chlorella ellipsoidea. Some improvementsin theculture technique for obtaining a good synchrony of algalgrowthwere described.
  2. By following the changes of averagecell volume and cell numberoccurring during culturing, therates of the following processesof life cycle were determined:(i) "growth" (or the increasein cell mass) occurring from thestage of smaller cells (Da)to the stage of ripened cell (L3),(ii) "ripening" (or processofformation of "nuclear substances"as estimated from the averagenumber of daughter cells formedfrom single mother cell), and(iii) " maturing and division" which leads to the full maturationof mother cells (L-cells)and their division into separate daughtercells (D-cells).
  3. "Growth"and "ripening" were found to be dependent in light,"maturingand division" light-independent. The time requiredfor "growth"and "ripening" (C) is dependent on temperaturebut independentof light intensity, the onset of "maturing anddivision" occurringat the same time (D) of culturing undervaried light intensities.The average cell volume at this stage(L3),however, was foundto be markedly modified by light intensity;larger with highertemperatures (see Fig. 4).
  4. Changes in incubation temperature(under the condition of saturatinglight intensities) were foundto affect the life cycle in thefollowing way: (i) The timeof onset of "maturing and division"(D), varies markedly withculturing temperature; earlier athigher temperatures, (ii)The average cell volume at this stagealso depends on temperature; smaller at higher temperatures.
  5. The average number of daughtercells (n) emerging from singlemother cells, was found to beuninfluenced by culturing temperature;(4.0–4.1 underthe conditions of the present study). Itwas found that thedivision number n is remarkably varied bychanging the lightintensity in the "growth" and "ripening"phases; 2.0 at 1 kilolux,3.7 at 5 kilolux, 4.2 at saturatinglight intensities (10 and25 kilolux). This finding was explainedby assuming a light-dependentformation of "nuclear substances"during the "growth" and "ripening"phases, the quantity of thesubstances in the cell at L3 stagedeterminig the division number.
  6. The experimental data wereanalyzed reaction kinetically, therate constants and othercharacteristics of the reactions constitutingthe processesof life cycle were determined, and values forthe apparent activationenergy for each reaction were computed.The reactions were discussedwith special reference to theirrelationship with photosyntheticprocess was discussed.

Synchronized populations of Chinese hamster ovary (CHO) cells in confluent culture have been examined by scanning electron microscopy and their surface changes noted as the cells progress through the cycle. During G1 it is characteristic for cells to show large numbers of microvilli, blebs, and ruffles. Except for the ruffles, these tend to diminish in prominence during S and the cells become relatively smooth as they spread thinly over the substrate. During G2 microvilli increase in number and the cells thicken in anticipation of rounding up for mitosis. It appears that the changes observed here reflect the changing capacity of CHO cells during the cycle to respond to contact with other cells in the population, because, as noted in the succeeding paper (Rubin and Everhart), CHO cells in sparse nonconfluent cultures do not show the same wide range of changes during the cell cycle. Normal, nontransformed cells of equivalent type in confluent culture are essentially devoid of microvilli, blebs, and ruffles. The relation of these surface configurations to the internal structure of the cell is discussed.  相似文献

本文分析了我国豆科黄华族3属7种植物种子中的游离氨基酸组成,并初步讨论了其系统学意义。发现均含有较高的游离氨基酸,其总量的变化范围在2.792—12.51毫克/克干重之间,其中除谷氨酸和天门冬氨酸的含量均很高外,还发现含有很高的精氨酸,尤其是新疆黄华中精氨酸含量高达8.62毫克/克干重;属间、种间的游离氨基酸组成有着较大的差异,但不能完全反映出种属间的系统关系。  相似文献

There have been no studies in which a significant number of isozymes have been investigated during a synchronous growth cycle of any organism. The present study was designed to obtain information on the fluctuations of a broad spectrum of enzymes during a synchronous growth cycle of Chlorella pyrenoidosa. Of the thirty-two enzymic activities investigated, seventeen could be localized on starch gels from a high temperature strain of Chlorella pyrenoidosa. Nine of these activities were found to possess more than one band of activity by starch gel electrophoresis. Seven of these activities were localized on starch gels throughout a synchronous growth cycle of C. pyrenoidosa grown in continuous light. Assay techniques are described. The fluctuations in enzyme activity are discussed with relation to concurrent metabolic and cytological changes during cell maturation in C. pyrenoidosa.  相似文献

Teliospores were aerated and agitated in a mineral salts medium and their free amino acid contents were analyzed at eight different times, from shortly after imbibition of water until just before germ tube emergence. In addition to the common amino acids, eight unidentified ninhydrin-positive components were detected. About 50 % or more of nearly each of the amino acids diffused out of the spores during the initial phase of germination. These released amino acids were actively taken up by the spores during the latter stages of germination. The free amino acids in largest amounts in the dormant spores of T. caries were arginine 15.0, glutamic acid 6.3, and alanine 3.7 μmoles per g dry spores. Together these three amino acids accounted for about 71 % of the total free amino acids in dormant spores of T. caries and T. controversa. The total amounts of free amino acids in spores of common bunt were much higher than in spores of dwarf bunt.  相似文献
