Treatment of water for aquatic bacterial growth studies   总被引:1,自引:1,他引:0  
The composition of a water is inextricably linked to its nutrient status and hence influences the behaviour of bacteria in artificial water systems. It has not yet been possible to devise a defined liquid medium representative of the complex composition of a treated water. Instead sterilized natural or distilled waters are used to study the growth or survival of aquatic bacteria in the laboratory. This has led to conflicting data and opposing opinions when the same water has proved toxic in some laboratory studies and growth-supporting in others. These differences may be explained by the variations in chemical composition which occur when water is collected, transported and treated or stored in the laboratory. This study describes a simple, membrane filtration method of preparing a fresh sample of water collected from the environment or a building water system such that it is both sterile and chemically unaltered. The availability of such water may enhance understanding of the behaviour of bacteria in the aquatic environment.  相似文献

The interaction of water flow and nutrients on aquatic plant growth   总被引:4,自引:1,他引:3  
A long-term experiment was conducted to compare the effects of flowing and still water on growth, and the relationship between water flow and nutrients, in Aponogeton elongatus, a submerged aquatic macrophyte. A. elongatus plants were grown for 23 weeks with three levels of nutrition (0, 0.5 and 1g Osmocote Plus® fertiliser pot–1) in aquaria containing stirred or unstirred water. Fertilized plants grew much better than non-fertilized. The highest fertilizer level produced 29% wider leaves and 58% higher total dry weight in stirred water. Stirred water increased leaf area by 40% and tuber size by 81%, but only with the highest level of nutrition. These results suggest that this plant depends on its roots for mineral uptake, rather than from the open water, and the major limitation to growth in still water is the supply of dissolved inorganic carbon. It was the combined effects of nutrient availability and stirring that produced the strongest response in plant growth, morphology and composition. This study provides some explanation for the observations of others that these plants grow best in creeks or river systems with permanently flowing water.  相似文献

The availability of organic carbon is considered the key factor to regulate microbial regrowth in drinking water network. However, boreal regions (northern Europe, Russia, and North America) contain a large amount of organic carbon in forests and peatlands. Therefore, natural waters (lakes, rivers, and groundwater) in the northern hemisphere generally have a high content of organic carbon. We found that microbial growth in drinking water in Finland is highly regulated not only by organic carbon but also by the availability of phosphorus. Microbial growth increased up to a phosphate concentration of 10 micrograms of PO4-P liter-1. Inorganic elements other than phosphorus did not affect microbial growth in drinking water. This observation offers novel possibilities to restrict microbial growth in water distribution systems by developing technologies to remove phosphorus efficiently from drinking water.  相似文献

For lake microbes, water column mixing acts as a disturbance because it homogenizes thermal and chemical gradients known to define the distributions of microbial taxa. Our first objective was to isolate hypothesized drivers of lake bacterial response to water column mixing. To accomplish this, we designed an enclosure experiment with three treatments to independently test key biogeochemical changes induced by mixing: oxygen addition to the hypolimnion, nutrient addition to the epilimnion, and full water column mixing. We used molecular fingerprinting to observe bacterial community dynamics in the treatment and control enclosures, and in ambient lake water. We found that oxygen and nutrient amendments simulated the physical-chemical water column environment following mixing and resulted in similar bacterial communities to the mixing treatment, affirming that these were important drivers of community change. These results demonstrate that specific environmental changes can replicate broad disturbance effects on microbial communities. Our second objective was to characterize bacterial community stability by quantifying community resistance, recovery and resilience to an episodic disturbance. The communities in the nutrient and oxygen amendments changed quickly (had low resistance), but generally matched the control composition by the 10th day after treatment, exhibiting resilience. These results imply that aquatic bacterial assemblages are generally stable in the face of disturbance.  相似文献

Bacterial viruses, or bacteriophages, have numerous roles in marine systems. Although they are now considered important agents of mortality of bacteria, a second possible role of regulating bacterial community composition is less well known. The effect on community composition derives from the presumed species-specificity and density-dependence of infection. Although models have described the "kill the winner" hypothesis of such control, there are few observational or experimental demonstrations of this effect in complex natural communities. We report here on some experiments that demonstrate that viruses can influence community composition in natural marine communities. Although the effect is subtle over the time frame suitable for field experiments (days), the cumulative effect over months or years would be substantial. Other virus roles, such as in genetic exchange or microbial evolution, have the potential to be extremely important, but we know very little about them.  相似文献

The report presents studies on temporal and spatial variations of kinetics (Vmax and Km) of bacterial ectoenzyme activity (-glucosidase - Glc, leucine aminopeptidase - Leu-amp) in the naturally eutrophic Plusee. Glc and Leu-amp activity were positively correlated with the flux of polymeric materials (polysaccharides, proteins) in the lake. Glc activity was low when algal populations grew actively, but during the algal bloom breakdown Glc activity increased rapidly. Leu-amp displayed the highest rates of activity in the epilimnion and was tightly coupled to bacterial production. The synthesis of studied ectoenzymes was under control of a repression/derepression mechanism. The significance of ectoenzymes for the transformation and bacterial utilization of organic matter, and their role in the microbial loop in aquatic environments is discussed.  相似文献

The time-course response of natural bacterial populations and isolates from lake water to various densities of the filamentous cyanobacteriaAphanizomenon flos-aquae andLyngbya birgei collected from the same lake is reported. The cyanobacteria were separated from the bacteria by dialysis membranes that allowed only dissolved cyanobacterial products to pass. Bacterial3H-thymidine incorporation and cell number were significantly (p<0.05) correlated with cyanobacterial density for both species. Estimated dissolved organic carbon (DOC) utilization, based on bacterial biomass changes over time, were usually significantly (p<0.01) correlated with cyanobacterial density and the decrease in bulk pool DOC for both species. Bacterial volume per cell increased significantly (p<0.05) in response to cyanobacterial density on day 5 of the experiments; cell volume remained unchanged on day 1. Bacterial cell numbers on outer surfaces of the tubular membrane containing the cyanobacteria (on the side exposed to the test bacteria) were significantly (p<0.01) correlated with cyanobacterial density. Statistical analysis inferred that bacteria closely associated with cyanobacteria (i.e. attached) responded more strongly to cyanobacterial products than free-living bacteria. Overall, our results indicate that cyanobacterial products have a potentially important role in regulating bacterioplankton productivity in aquatic systems.  相似文献

Summary A survey of the phycomycete flora of three types of collecting stations in Allahabad was made for a year. Twelve species were recorded, belonging to the Saprolegniales, Blastocladiales and Peronosporales. The results indicate that the rarity of many of the aquatic phycomycetes may be due not only to specificity of substratum but also to temperature and the time needed for maturation of the oospores. There is a distinct seasonal periodicity of growth which is mainly restricted to winter months.  相似文献

A portable quantum meter-spectroradiometer for use in aquatic studies   总被引:2,自引:0,他引:2  
This paper describes the design, construction and use of a portable battery-powered, submersible spectroradiometer. The distribution of energy underwater is determined by measuring successively through six sensors fitted with interference filters which partition the underwater spectrum. This system also contains a quantum sensor which detects the photosynthetically active radiation under water. The instruments’ small size and battery operation make it especially useful for studying small lakes or reservoirs from small boats. Measurements taken from Pawnee and Wagontrain, two of the Salt Valley Reservoirs in eastern Nebraska, are given as examples of the use of the instrument.  相似文献

Recent studies highlight the need to include estimates of detection/capture probability in population studies. This need is particularly important in studies where detection and/or capture probability is influenced by parasite-induced behavioral alterations. We assessed potential biases associated with sampling a population of the amphipod Gammarus lacustris in the presence of Polymorphus spp. acanthocephalan parasites shown to increase positive phototaxis in their amphipod hosts. We trapped G. lacustris at two water depths (benthic and surface) and compared number of captures and number of parasitized individuals at each depth. While we captured the greatest number of G. lacustris individuals in benthic traps, parasitized individuals were captured most often in surface traps. These results reflect the phototaxic movement of infected individuals from benthic locations to sunlit surface waters. We then explored the influence of varying infection rates on a simulated population held at a constant level of abundance. Simulations resulted in increasingly biased abundance estimates as infection rates increased. Our results highlight the need to consider parasite-induced biases when quantifying detection and/or capture probability in studies of aquatic invertebrate populations.  相似文献

The western Antarctic Peninsula is an extreme low temperature environment that is warming rapidly due to global change. Little is known, however, on the temperature sensitivity of growth of microbial communities in Antarctic soils and in the surrounding oceanic waters. This is the first study that directly compares temperature adaptation of adjacent marine and terrestrial bacteria in a polar environment. The bacterial communities in the ocean were adapted to lower temperatures than those from nearby soil, with cardinal temperatures for growth in the ocean being the lowest so far reported for microbial communities. This was reflected in lower minimum (Tmin) and optimum temperatures (Topt) for growth in water (?17 and +20°C, respectively) than in soil (?11 and +27°C), with lower sensitivity to changes in temperature (Q10; 0–10°C interval) in Antarctic water (2.7) than in soil (3.9). This is likely due to the more stable low temperature conditions of Antarctic waters than soils, and the fact that maximum in situ temperatures in water are lower than in soils, at least in summer. Importantly, the thermally stable environment of Antarctic marine water makes it feasible to create a single temperature response curve for bacterial communities. This would thus allow for calculations of temperature‐corrected growth rates, and thereby quantifying the influence of factors other than temperature on observed growth rates, as well as predicting the effects of future temperature increases on Antarctic marine bacteria.  相似文献

A range of platinum group metal complexes (PGMC) were screened for antibacterial activity against Klebsiella aerogenes and Staphylococcus aureus. The effects of the complexes were monitored by changes in the thermal and growth properties of the organisms. Active complexes caused an immediate cessation of power and biomass production which did not recover whilst the complex was present in the medium. Removal of the complex by washing the cells allowed the growth of the small viable proportion of the cells; the majority of the cells had been killed. Changes which occurred in aqueous solutions of active complexes rendered them less bactericidal; this was a possible cause of regrowth observed at sub-bactericidal levels of some complexes. Resistance to active complexes could not be achieved by serial subculture. Three palladium complexes active against K. aerogenes all had similar square planar structures, but in general it was not possible to correlate activity with structure. The unique effects of the PGMCs at low concentrations, combined with the apparent inability of the bacteria to develop resistance to them, makes the complexes a valuable addition to the antibacterial arsenal.  相似文献
