Fluorescent microscopy and spectrofluorometry of biogenic amines were employed to study the peripheral catecholaminergic systems in immobilized rats which received sodium hydroxybutyrate. The content of catecholamines was measured in the adrenergic nerves of dura mater, vas deferens and chromaffin tissue of the adrenals. It was established that sodium hydroxybutyrate in a dose of 40 mg/kg intraperitoneally promoted returning to normal of the adrenergic mediator activity during alarm and resistance stages. The role of the peripheral catecholaminergic systems in the mechanism of the stress-protective effect of sodium hydroxybutyrate is discussed.  相似文献

For studying the interactions of the cholinergic and noradrenergic systems in the central nervous system rats were given muscarine or nicotine and the peripheral receptors were blocked with atropine or pempidine. After killing the animals the level of biogenic amines was determined in various parts of the brain. Evident changes were found in the levels of biogenic amines after stimulation or blockade of muscarinic or nicotinic receptors.  相似文献

Piperidine is one of biogenic amines possessing nicotine-like synaptotropic actions on the nervous systems. Since piperidine produces multiplex pharmacological actions, a role for the amine as a modulator in neuroendocrine as well as neuronal functions has been supposed. In the present study, piperidine levels in the brain and peripheral organs in rats during cold exposure were examined by use of a mass fragmentographic technique and it was found that piperidine concentrations in the brain and peripheral endocrine glands significantly increased at 180 min following exposure to 4°C. The significance of the findings is discussed with respect to the neurohormone-releasing effect of piperidine.  相似文献

Investigations were carried out on the role of biogenic amines in the development of glomerulonephritis in rabbits using the experimental model of acute and chronic serum sickness. The animals immunized with foreign-species albumin received additionally 5HT and some of them were given reserpine or PCPA for reducing the level of biogenic amines. The results of histological examinations demonstrated a clear cut inhibitory effect of PCPA and reserpine on the development of inflammatory changes in the glomeruli, and their intensification with simultaneous 5HT administration. The obtained results seem to suggest a significant role of biogenic amines in the pathogenesis of inflammatory lesions developing in renal glomeruli in acute or chronic serum sickness in rabbits.  相似文献

Ultrastructural studies of human mast cells (HMCs) and basophils (HBs) are reviewed. Sources of HMCs include biopsies of tissue sites and in situ study of excised diseased organs; isolated, partially purified samples from excised organs; and growth-factor-stimulated mast cells that develop de novo in cultures of cord blood cells. Sources of HBs for study include partially purified peripheral blood basophils, basophils in tissue biopsies, and specific growth factor-stimulated basophils arising de novo from cord blood cells. The ultrastructural studies reviewed deal with identity, secretion, vesicles, recovery, and synthesis issues related to the biology of these similar cells.  相似文献

While holding on youth may be a universal wish, aging is a natural process associated with physical and physiological impairment in living organisms. Drosophila provides useful insights into aging-related events. Hence, this study was conducted to investigate the age-related changes in muscle function and architecture in relation to the biogenic amine titers. To achieve this aim, visceral and skeletal muscles performance was tested in newly-eclosed, sexually mature and old adult flies using climbing and gut motility assays. In addition, age-related ultrastructural alterations of muscular tissue were observed using transmission electron microscopy (TEM). The titer of selected biogenic amines was measured using high-performance liquid chromatography (HPLC). The results demonstrated that old flies were dramatically slower in upward movement than either newly-eclosed or sexually mature flies. Similarly, gut contraction rate was significantly lower in old flies than the sexually mature, although it was markedly higher than that in the newly-eclosed flies. In TEM examination, there were several ultrastructural changes in the midgut epithelium, legs and thorax muscles of old flies. Regarding biogenic amine titers, the old flies had significantly lower concentrations of octopamine, dopamine and serotonin than the sexually mature. We concluded that aging has adverse effects on muscular system function and ultrastructure, synchronized with biogenic amine titers changes. Our results highlighted the need for more researches on therapeutics that may balance the levels of age-related alterations in biogenic amines.  相似文献

The study deals with evaluation of ligand specificities of hemoagglutinins and hemolysins of hemolymph of three species of gastropods(Planorbius corneus, Lymnaea stagnalis, andAchatina fulica) and one species of bivalve molluscs(Anodonta cygnea). The hemoagglutinin titer was estimated from hemoagglutination reaction, the hemolysin titer, spectrophotometrically, from release of hemoglobin from sheep erythrocytes. The ligand specificity of hemoagglutinins and hemolysins was evaluated by an inhibitory modification of the hemoagglutination reaction. It has been shown that hemoagglutinins and hemolysins of pulmonary gastropod molluscs have different spectra of ligand specificity to the substances of the carbohydrate nature. Hemoagglutinins and hemolysins of the bivalve molluscAnodonta cygnea have the same spectrum of ligand specificity. Among the ligands of these lectins, terminal N-acetylated sugars and monosaccharides predominate. Components of bacterial cell walls, lipopolysaccharide and phosphorylcholine, have been revealed among specific ligands of hemolymph hemoagglutinins and hemolysins.  相似文献

The islets of Langerhans of sand rats (Psammomys obesus) have been investigated ultrahistochemically in order to demonstrate biogenic amines. It could be found that amine-containing vesicles with a diameter of 600-700 nm from the type of catecholamine-granules exist in A-cells as well as in B-cells. It can be stated that the sand rat belongs to the species containing amines in A- and B-cells. The importance of the amines for the regulation of the hormone secretion of the islets of Langerhans is discussed and the connections to diabetes mellitus are referred too.  相似文献

Results of experimental studies on mice concerning the important role of cells producing normal autoantibodies (AAPC) in the reaction of the organism to specific antigens have been systematized. The number of AAPC in the blood and organs increases as early as 15-20 min after the administration of antigen and this increase is particularly intensive during reimmunization. On the other hand, the administration of heterologous antigens to the same mice causes a decrease in the number of AAPC. Transfer of blood plasma from immunized donors induces an increase in the number of AAPC in the recipients. The addition of specific antigen to blood preparations conditions a considerable increase in the number and size of AAPC in immunized humans and animals. It has been demonstrated that one and the same cell produces both auto-antibodies and specific hemolysins. A hypothesis has been proposed dealing with the stages of involvement of autoantibody-producing cells in the immune response and with a new type of intercellular interactions between erythrocytes and karyocytes.  相似文献

The content was studied of biogenic amines and their metabolites (by the method of high-effective fluid chromatography electrochemical detection) in the reticular formation of the midbrain, locus coeruleus and sensorimotor cortex in the rats of Wistar and August lines, differing in the behaviour in the open field, in conditions of immobilization stress. The dependence was revealed of the biogenic amines level on the animals genotype and individual characteristics. Most probably, the level of biogenic amines metabolism in central brain structures determines the stability of the animals against emotional stress.  相似文献

We have characterized the changes in tissue concentrations of amino acids and biogenic amines in the central nervous system (CNS) of mice with MOG35-55-induced experimental autoimmune encephalomyelitis (EAE), an animal model commonly used to study multiple sclerosis (MS). High performance liquid chromatography was used to analyse tissue samples from five regions of the CNS at the onset, peak and chronic phase of MOG35-55 EAE. Our analysis includes the evaluation of several newly examined amino acids including d-serine, and the inter-relations between the intraspinal concentration changes of different amino acids and biogenic amines during EAE. Our results confirm many of the findings from similar studies using different variants of the EAE model as well as those examining changes in amino acid and biogenic amine levels in the cerebrospinal fluid (CSF) of MS patients. However, several notable differences were observed between mice with MOG35-55-induced EAE with findings from human studies and other EAE models. In addition, our analysis has identified strong correlations between different amino acids and biogenic amines that appear to change in two distinct groups during EAE. Group I analyte concentrations are increased at EAE onset and peak but then decrease in the chronic phase with a large degree of variability. Group II is composed of amino acids and biogenic amines that change in a progressive manner during EAE. The altered levels of these amino acids and biogenic amines in the disease may represent a critical pathway leading to neurodegenerative processes that are now recognized to occur in EAE and MS.  相似文献

The pineal body of white normal rats, 1.5 to 3 months old, was studied under the electron microscope. A single type of parenchymal cell—the pinealocyte—is recognized as the main component of the tissue, and some of the structural characteristics of the nucleus and cytoplasm are described. The main morphological characteristic of the pinealocytes is represented by club-shaped perivascular expansions connected to the cell by thin pedicles. They are found lying in a large, clear space surrounding the blood capillaries. The name plurivesicular secretory processes is proposed, to emphasize the main structural feature and the probable function of these cellular expansions. A tubulofibrillar component is mainly found in the pedicle, and within the expansion there are numerous small mitochondria and densily packed vesicles of about 425 A. Two types of vesicles, one with a homogeneous content and another with a very dense osmium deposit, are described. Between the two types there are intermediary forms. In these processes, mitochondria show profound changes which may lead to complete vacuolization. The significance of this plurivesicular secretory component is discussed in the light of recent work on the biogenic amines of the pineal body and preliminary experiments showing the release of the vesicles containing dense granules after treatment with reserpine. These vesicles are interpreted as the site of storage of some of the biogenic amines. Bundles of unmyelinated nerve fibers and endings on large blood vessels which also contain a plurivesicular content are described and tentatively interpreted as adrenergic nerve terminals.  相似文献

Biogenic amines are an important class of primary messengers in the central (CNS) and peripheral nervous systems and in peripheral organs. These substances regulate and modulate many physiological and behavioral processes. Various inactivation mechanisms for these substances exist to terminate biogenic amine-mediated signal transduction. In vertebrates, the enzymes monoamine oxidase and/or catechol-O-methyl-transferase are involved in these processes. In insects, however, in which both enzymes are low in abundance or absent, biogenic amines are inactivated mainly by N-acetylation or O-sulphation. In Drosophila, beta-alanyl conjugation mediated by the Ebony protein has recently been shown to be a novel and alternative pathway for biogenic amine inactivation. Here, we report the cloning of ebony cDNA (Peaebony) from a brain-specific cDNA library of the cockroach Periplaneta americana. The open reading frame encodes a protein of 860 amino acid residues (PeaEbony). The PeaEbony polypeptide shares homology to Ebony sequences from Anopheles gambiae, Apis mellifera, and Drosophila melanogaster. In addition, PeaEbony exhibits sequence similarity to a family of microbial non-ribosomal peptide synthetases. The mRNA encoding PeaEbony is highly expressed in the cockroach brain and to a lesser extent in the salivary glands. PeaEbony is, therefore, probably involved in the inactivation of various biogenic amines through beta-alanyl conjugation in the cockroach CNS. Since the salivary glands in Periplaneta are innervated by dopaminergic and serotonergic neurons, PeaEbony probably also biochemically modifies dopamine and serotonin in these acinar glands.  相似文献

A procedure for the separation, detection, and quantification of picomole levels of dansyl derivatives of the biogenic amines, dopamine, norepinephrine, octopamine, and serotonin, has been developed using high-performance thin-layer chromatography. The detection limit is 1 to 2 pmol. Each of the amine derivatives has been detected in insect brain tissue and a solvent system has been developed for the separation and quantification of octopamine in insect tissue samples.  相似文献

Mast cells in the thyroid glands of sand rats with different disturbances of the carbon hydrate tolerance (reference-, impaired-glucose-tolerance-, and diabetic group) are quantitative-histologically investigated. In the IGT-group (impaired glucose tolerance), mast cells are significant increased. However, the number of these cells in the diabetic group decreased significantly. This investigations demonstrate that the mellitus-syndrome is not only a complex neuroendocrine but also a disturbance of biogenic amines and the organ of the connective tissue.  相似文献

The biogenic amines are widespread regulators of physiological processes, and play an important role in regulating heart rate in diverse organisms. Here, we present the first pharmacological evidence for a role of the biogenic amines in the regulation of dorsal blood vessel pulse rate in an aquatic oligochaete, Lumbriculus variegatus (Müller, 1774). Bath application of octopamine to intact worms resulted in an acceleration of pulse rate, but not when co-applied with the adenylyl cyclase inhibitor MDL-12,330a. The phosphodiesterase inhibitor theophylline mimicked the effects of OA, but the polar adenosine receptor antagonist 8(p-sulphophenyl)theophylline was significantly less potent than theophylline. Pharmacologically blocking synaptic reuptake of the biogenic amines using the selective 5-HT reuptake blocker fluoxetine or various tricyclic antidepressants also accelerated heart rate. Depletion of the biogenic amines by treatment with the monoamine vesicular transporter blocker reserpine dramatically depressed pulse rate. Pulse rate was partially restored in amine-depleted worms after treatment with octopamine or dopamine, but fully restored following treatment with serotonin. This effect of 5-HT was weakly mimicked by 5-methoxytryptamine, but not by α-methylserotonin; it was completely blocked by clozapine and partially blocked by cyproheptadine. Because they are known to orchestrate a variety of adaptive behaviors in invertebrates, the biogenic amines may coordinate blood flow with behavioral state in L. variegatus.  相似文献

Spontaneously hypertensive rats (SHR) were administered either 2.4 g/kg ethanol or an isocaloric glucose daily for 4 weeks and the levels of norepinephrine (NE), epinephrine (EP), dopamine (DA), serotonin (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) in different brain regions were determined. Results indicated a 3-fold increase in NE level in brain stem and hypothalamus and more than 2-fold increase in DA in corpus striatum in alcohol-treated rats as compared to controls. There was a significant increase in the level of DA in the corpus striatum but the levels in cerebral cortex, brain stem and hippocampus were decreased instead. Decreases in 5-HT levels were found in hypothalamus, brain stem, cortex and cerebellum of alcohol-treated brain as compared to untreated controls. These results indicate alterations of the biogenic amine contents in different regions of the SHR brain after chronic ethanol ingestion. Since stimulated release of biogenic amines in the SHR brain has been implicated in the regulation of blood pressure, changes due to ethanol ingestion may be a risk factor in hypertensive patients.  相似文献

AIDS patients often contain HIV-1-infected mast cells (MCs)/basophils in their peripheral blood, and in vivo-differentiated MCs/basophils have been isolated from the blood of asthma patients that are HIV-1 susceptible ex vivo due to their surface expression of CD4 and varied chemokine receptors. Because IL-16 is a ligand for CD4 and/or an undefined CD4-associated protein, the ability of this multifunctional cytokine to regulate the development of human MCs/basophils from nongranulated progenitors residing in cord or peripheral blood was evaluated. After 3 wk of culture in the presence of c-kit ligand, IL-16 induced the progenitors residing in the blood of normal individuals to increase their expression of chymase and tryptase about 20-fold. As assessed immunohistochemically, >80% of these tryptase(+) and/or chymase(+) cells expressed CD4. The resulting cells responded to IL-16 in an in vitro chemotaxis assay, and this biologic response could be blocked by anti-IL-16 and anti-CD4 Abs as well as by a competitive peptide inhibitor corresponding to a sequence in the C-terminal domain of IL-16. The additional finding that IL-16 induces calcium mobilization in the HMC-1 cell line indicates that IL-16 acts directly on MCs and their committed progenitors. IL-16-treated MCs/basophils also are less susceptible to infection by an M/R5-tropic strain of HIV-1. Thus, IL-16 regulates MCs/basophils at a number of levels, including their vulnerability to retroviral infection.  相似文献

The aim of this study was to evaluate the influence of an extremely low frequency sinusoidal magnetic field (ELF MF) with frequency of 10 Hz and intensity of 1.8-3.8 mT on the levels of the biogenic amines dopamine (DA), 3,4-dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA), 3-methoxytyramine (3-MT), 5-hydroxytryptamine (5-HT), 5-hydroxyindolacetic acid (5-HIAA), and noradrenaline (NA), as well as on DA and 5-HT turnover in corpus striatum and frontal cortex of adult male Wistar rats. We found that ELF MF exposure for 14 days, 1 h daily, did not influence the level of the examined biogenic amines and metabolites, but increased the rate of synthesis (turnover) of DA and 5-HT in rat frontal cortex as compared to control, sham exposed rats. On the basis of the present results and our previous findings, extremely low frequency magnetic field (ELF MF) exposure has been found to alter both turnover and receptor reactivity of monoaminergic systems, as well as some behaviors induced by these systems or their agonists and antagonists.  相似文献
