Following 10 days of infestation by the aphid Myzus persicae there was an increase in the amount of growth inhibiting substances and a decrease in cytokinins, gibberellins and auxins in infested as compared with similar uninfested radish seedlings. Even after previously infested seedlings have been freed of aphids for 10 days, differences in the hormone balance remained. The possible relationships between the changes in hormonal balance and the effect of the aphid infestation on growth, translocation and wilting are discussed.  相似文献

After 10 days of infestation by the aphid Myzus persicae growth of radish seedlings was reduced by 38 %, and the volume of sap bleeding from the stumps of cut stems of infested seedlings was 4% of that exuding from the stumps of uninfested seedlings of the same age. Diminished bleeding could not be accounted for by the 52 % reduction in the weight of the roots of infested seedlings. After 14 days of infestation, necrotic patches developed along the veins of the cotyledons which yellowed noticeably. If freed of aphids the seedlings subsequently resumed growth and their was an increase in the volume of bleeding sap produced by their cut stems. Symptoms associated with aphid infestation cannot be attributed either to virus disease or initially to exhaustion of the seedling's carbohydrate and nitrogen reserves. Possible causes of the symptoms described are discussed.  相似文献

Proteomics in Myzus persicae: effect of aphid host plant switch   总被引:2,自引:0,他引:2  
Chemical ecology is the study of how particular chemicals are involved in interactions of organisms with each other and with their surroundings. In order to reduce insect attack, plants have evolved a variety of defence mechanisms, both constitutive and inducible, while insects have evolved strategies to overcome these plant defences (such as detoxification enzymes). A major determinant of the influence of evolutionary arms races is the strategy of the insect: generalist insect herbivores, such as Myzus persicae aphid, need more complex adaptive mechanisms since they need to respond to a large array of different plant defensive chemicals. Here we studied the chemical ecology of M. persicae associated with different plant species, from Brassicaceae and Solanaceae families. To identify the involved adaptation systems to cope with the plant secondary substances and to assess the differential expression of these systems, a proteomic approach was developed. A non-restrictive approach was developed to identify all the potential adaptation systems toward the secondary metabolites from host plants. The complex protein mixtures were separated by two-dimensional electrophoresis methods and the related spots of proteins significantly varying were selected and identified by mass spectrometry (ESI MS/MS) coupled with data bank investigations. Fourteen aphid proteins were found to vary according to host plant switch; ten of them were down regulated (proteins involved in glycolysis, TCA cycle, protein and lipid synthesis) while four others were overexpressed (mainly related to the cytoskeleton). These techniques are very reliable to describe the proteome from organisms such as insects in response to particular environmental change such as host plant species of herbivores.  相似文献

The ecological consequences of genetic variability in host plant exploitation of Myzus persicae (Hemiptera, Aphididae) were studied in several agro-ecosystems, differing in crop plant heterogeneity. An analysis of aphid populations sampled from sugar beet, potatoes, and lettuce revealed changes in the frequency distribution of aphid clones according to their respective host plant adaptations. The spatial unit of differentiation was below an average field size of 3–5 hectares and a differentiation could be found in the centre of potato and sugar beet fields but not at the edge of the fields. On a temporal scale, the differentiation of populations is transitory even on a large-scale basis of highly specialized cropping areas. The ecological parameters of differentiation are discussed with special reference to agro-ecosystems and integrated pest management. Increasing the genetic heterogeneity of crop fields may help to reduce population outbreaks.
Zusammenfassung Die ökologischen Konsequenzen genetischer Variabilität in der Wirtspflanzennutzung von Myzus persicae (Hemiptera, Aphididae) wurden in mehreren Agro-Ökosystemen untersucht, die sich in der Kulturpflanzenheterogenität unterscheiden. Eine Analyse von auf Rüben, Kartoffeln und Salat siedelnden Populationen zeigte eine genetische Differenzierung in Wirtspflanzen-angepaßte Feld-populationen. Die räumliche Einheit solcher Differenzierungen liegt unterhalb einer Feldgröße von 3–5 ha. Eine Differenzierung konnte hier in der Mitte der Felder aber nicht am Rand festgestellt werden. Aus zeitlicher Sicht ist die Differenzierung selbst in großräumigen, spezialisierten Anbaugebieten nur vorübergehend feststellbar. Die ökologischen Parameter der Differenzierung werden besonders in Hinsicht auf die integrierte Schädlingsbekämpfung in Agro-ökosystemen diskutiert. Eine Erhöhung der genetischen Heterogenität von Kulturpflanzenbeständen kann möglicherweise zu einer Verringerung von Blattlaus-Massenvermehrungen beitragen.

Myzus persicae showed a reduced relative growth rate on plants given soil drenches of 1% CCC (a plant growth retardant), and a greater reduction when CCC was applied as a foliar spray. The use of the relative growth rate of aphids as a convenient measure of their performance in relation to plant resistance is discussed.
Zusammenfassung Die Resistenz von Kohlpflanzen, die mit 1% CCC (einem Regulator des Pflanzenwachstums) behandelt worden waren, gegen Myzus persicae wurde mit Hilfe des Wachstumsquotienten der Blattläuse (g/g/Tag) gemessen. Diese Methode arbeitet rasch und ergibt eine ziemlich geringe Variabilität. Der Maßstab scheint sinnvoll und ziemlich empfindlich zu sein.Mit CCC gedüngte Pflanzen zeigten stets eine ziemlich starke Verminderung des Wachstumsquotienten von Myzus persicae. Wurde CCC auf die Pflanzen aufgesprüht, so ergab sich ein noch geringerer Wachstumsquotient der Blattläuse.

Female adults of the aphidopagous gall midge, Aphidoletes aphidimyza (Rondani) (Diptera: Cecidomyiidae), showed an olfactory response to honeydew excreted by the green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae) under laboratory conditions. The response was only elicited by treatments with honeydew, whereas aphids, aphids with pepper plants or leaves, and pepper plants or leaves were not attractive to the midges. Dose‐dependent responses were observed from whole honeydew, honeydew volatiles extracted in pentane, and honeydew volatiles captured on Porapak Q®. When honeydew was eluted with three sequential pentane washes, a positive response was only observed from the midges for the first wash. Female midges laid more eggs on pepper plants infested with higher densities of M. persicae. The olfactory response of midges to honeydew is discussed with respect to prey location.  相似文献

Three isofemale lines of Myzus persicae (Sulzer), two lines collected from and reared on a brassicaceous host, and one line collected from and reared on a malvaceous host, were evaluated for their efficiency of transmitting Zucchini yellow mosaic virus (family Potyviridae, genus Potyvirus, ZYMV). In the first experiment, the transmission efficiencies of two clones from Brassicaceae (B1 and B2) were 52.0 and 60.8%, respectively, and these transmissions were not significantly different. In a second experiment, the transmission efficiencies of the clone on Malvaceae (M1) and clone B2 were significantly different at 35.6 and 55.7%, respectively. Further experiments evaluated host-related mechanisms that may have contributed to the differential transmissions observed between clones M1 and B2. Studies on short-term feeding showed that aphids continuously reared on okra, Abelmoschus esculentus (L.) Moench (malvaceous host), and those that were reared on okra and allowed a 24-h preacquisition feeding period on mustard, Brassica juncea (L.) Czern (brassicaceous host), had significantly lower transmission than aphids continuously maintained on mustard. Aphids reared on mustard and allowed a 24-h preacquisition feeding period on okra had intermediate transmission efficiency. In long-term host association studies, we found that aphids reared on mustard had significantly higher transmission efficiency than those reared on okra, and aphids reared first on okra and then switched to mustard had a transmission efficiency that was intermediate and not significantly different from the other two treatments. Our study reveals the existence of intraspecific variation in the transmission of ZYMV by M. persicae, and it suggests that to accurately assess the transmission capability of ZYMV by this species, multiple clones should be examined. Furthermore, the host plant on which the aphid is reared as well as the host plant on which it feeds just before virus acquisition contribute to ZYMV transmission efficiency of M. persicae.  相似文献

The architecture and action of quantitative trait loci (QTL) contributing to plant resistance mechanisms against aphids, the largest group of phloem-feeding insects, are not well understood. Comparative mapping of several components of resistance to the green peach aphid (Myzus persicae) was undertaken in Prunus davidiana, a wild species related to peach. An interspecific F(1) population of Prunus persica var. Summergrand × P. davidiana clone P1908 was scored for resistance (aphid colony development and foliar damage) and 17 aphid feeding behaviour traits monitored by means of the electrical penetration graph technique. Seven resistance QTLs were detected, individually explaining 6.1-43.1% of the phenotypic variation. Consistency was shown over several trials. Nine QTLs affecting aphid feeding behaviour were identified. All resistance QTLs except one co-located with QTLs underlying aphid feeding behaviour. A P. davidiana resistance allele at the major QTL was associated with drastic reductions in phloem sap ingestion by aphids, suggesting a phloem-based resistance mechanism. Resistance was also positively correlated with aphid salivation into sieve elements, suggesting an insect response to restore the appropriate conditions for ingestion after phloem occlusion. No significant QTL was found for traits characterising aphid mouthpart activity in plant tissues other than phloem vessels. Two QTLs with effects on aphid feeding behaviour but without effect on resistance were identified. SSR markers linked to the main QTLs involved in resistance are of potential use in marker-assisted selection for aphid resistance. Linking our results with the recent sequencing of the peach genome may help clarify the physiological resistance mechanisms.  相似文献

ABSTRACT. Compensation in mean relative growth rate of M. persicae occurred when aphids normally kept on susceptible brussels sprout plants were starved for only 2 h daily or kept on 'resistant' plants (given low nitrogen:high potassium fertilization) for 48 h. More adverse conditions, e.g. 4 or 8 h daily starvation, 48 h on leaf discs or 60 h on 20% sucrose, resulted in a low aphid growth rate being maintained on transfer to susceptible plants. The low overall growth rate could not be explained in terms of a weight loss by the aphid at any stage and is assumed to be a behavioural phenomenon. It is concluded that it is unwise, in the study of aphid—plant relationships, to rely on pre-conditioning aphids in poor conditions with the hope that this might magnify differences in subsequent aphid response.  相似文献

The biology of the aphid hyperparasitoid Alloxysta victrix was investigated with respect to its interaction with the primary parasitoid Aphidius colemani and the host aphid Myzus persicae. Laboratory investigations of host selection behaviour indicated that naive A. victrix females could rapidly distinguish between unparasitized aphids and those parasitized by A. colemani, but showed little tendency to discriminate between aphids parasitized by the primary parasitoid three, five, seven, or ten days earlier (even though the latter were already mummified). Host suitability studies, in contrast, indicated that aphids parasitized by A. colemani became progressively less suitable as the primary parasitoid developed. When aphids were exposed to A. victrix more than seven days after being parasitized by A. colemani, survivorship of both primary and secondary parasitoid species decreased, with a significantly higher proportion of mummies producing no viable adult parasitoid of either species. The developmental period (egg to adult eclosion) of surviving hyperparasitoids was also longer in older host stages, and females developed significantly more slowly than males (14.2 days versus 13.6 days, respectively). The sex ratio of hyperparasitoid broods emerging from all host stages was consistently male-biased (31–46% female), but adult longevity was higher for females than for males (37.1 days versus 23.6 days).  相似文献

Oxylipins derived from lipoxygenase (LOX) activity play important roles in plant growth, development and stress response. In a recent study, we provided evidence that infestation of Arabidopsis thaliana foliage by the green peach aphid (GPA; Myzus persicae), a phloem sap-consuming insect, was promoted by plant LOX5-derived oxylipins. In comparison to the wild-type (WT) plant, GPA population was smaller on the Arabidopsis lox5 mutant. The insect spent less time feeding from the sieve element and xylem of the lox5 mutant compared with the WT plant. In addition, compared with insects feeding on the WT plant, when on the lox5 mutant, the GPA was unable to suppress an antibiotic activity that is present in Arabidopsis vascular sap. Roots are the critical source of a LOX5-derived oxylipin(s) that promotes colonization of the foliage by GPA. Here we show that the 9-hydoxy-10E, 12Z-octadecadienoic acid (9-HOD), a LOX5-derived oxylipin, accumulated in GPA that were reared on the WT, but not the lox5 mutant plant. However, 9-HOD accumulated in insects reared on lox5 mutant plants that were irrigated with 9-HOD, thus indicating that the insect ingests oxylipins from the host plant. We further demonstrate that the host plant requires LOX5 function to promote expression of the defense regulatory gene PHYTOALEXIN-DEFICIENT4 in the foliage. Taken together, our previous observations and results presented here indicate that while the host plant utilizes LOX5-dependent factors for promoting defense mechanisms, GPA has evolved to utilize plant 9-LOX-derived oxylipins as cues to facilitate infestation, thus suggesting a complex involvement of oxylipins in Arabidopsis interaction with GPA.  相似文献

The peach-potato aphid Myzus persicae (Sulzer) can resist a wide range of insecticides, but until recently (1990) the only mechanism identified was the increased production of carboxylesterases (E4 or FE4), which cause enhanced degradation and sequestration of insecticidal esters. We have now identified two forms of target-site resistance involving changes in the acetylcholinesterase (AChE) and sodium channel (kdr) genes. Biochemical and DNA diagnostic methods can be used to identify all three mechanisms in individual aphids, and thereby establish their spatial distributions and temporal dynamics. Amplified genes underlie the increased production of esterases but their expression is modulated by DNA methylation. Amplification of the E4 gene is in strong linkage disequilibrium with the kdr mechanism. This may reflect strong insecticidal selection favouring aphids with multiple mechanisms, tight chromosomal linkage and/or the prominence of parthenogenesis in many M. persicae populations. The decreased fitness of resistant aphids under winter conditions may be a consequence of the altered sodium-channel gene affecting behaviour and/or the perception of external stimuli.  相似文献

研究白蝴蝶(Syngonium podophyllum )乙醇提取物、苍耳(Xanthium sibiricum )乙醇提取物、机油乳剂和0.3%印楝素乳油对桃蚜(Myzus persicae)和萝卜蚜(Lipaphis erysimi)的控制效果.室内四因子(1/2实施)二次正交回归旋转组合设计测试对有翅蚜的忌避作用,结果表明,对有翅桃蚜的主要忌避作用物为苍耳乙醇提取物,当苍耳提取物与白蝴蝶乙醇提取物混用,以及机油乳剂和印楝素混用时,对桃蚜有翅蚜的忌避效果提高;而对萝卜蚜有翅蚜的忌避作用主要受苍耳提取物和白蝴蝶提取物的影响.这些干扰作用均是非线性的.田间试验结果,单独使用白蝴蝶提取物对有翅成蚜有较强的驱避作用;对萝卜蚜自然种群的干扰控制作用以4种植物提取物和药剂混配效果最好,达95.7%;对桃蚜自然种群干扰控制效果最好的则是白蝴蝶提取物,控制效果达87%;同时,白蝴蝶提取物与印楝素乳油混配以及苍耳提取物与印楝素乳油混配,对两种蚜虫的控制效果均达80%以上.  相似文献

A difference weighing method applied to sachets of diet on which groups of 40–50 larval Myzus persicae (Sulzer) were allowed to feed for 24 hours gave information on the absolute and relative uptake of different diets in terms of the fluid volume of diet and the amounts of nutrients taken up per aphid per day.The diets tested were in a sucrose concentration series ranging from 0–40% with a 2.4% amino acid level common to the series, and in an amino acid series ranging from 0–4.8% total amino acids with 15% sucrose common to the series. Uptake was found to be markedly affected both by the level of sucrose and by the overall level of the 20 dietary amino acids.Fluid uptake was poor or non-existent on diets having low sucrose (<5%) or low total amino acid (<1%) concentrations. For sucrose, an optimal range of 10–20% was indicated with a progressive decline in fluid uptake with concentrations above 20%. For amino acids, fluid uptake increased with dietary concentrations up to 3% and then decreased slightly.The results are related to growth rates of M. persicae larvae on diets of similar composition, and the ecological significance of the findings is discussed.
Zusammenfassung Durch Gewichtsverlustbestimmung von Nährlösungssäckchen, an denen Gruppen von 40–50 Myzus persicae-Larven 24 Stunden lang saugen konnten, ließ sich die absolute und relative Aufnahme verschiedener Diäten feststellen, ausgedrückt in dem Betrag des Flüssigkeitsvolumens der Nahrung und damit der Menge an Nährstoffen, die pro Tag und Blattlaus aufgenommen wurden.Die geprüften Diäten enthielten in der einen Serie bei gleichbleibendem Aminosäurespiegel von 2,4% Rohrzuckerkonzentrationen zwischen 0 und 40%, in einer anderen Serie bei konstantem Rohrzuckergehalt von 15% Gesamtaminosäurekonzentrationen von 0 bis 4,8%. Die Nahrungsaufnahme erwies sich als merklich beeinflußt sowohl von der Rohrzuckerkonzentration wie von der Gesamtkonzentration der 20 diätischen Aminosäuren.Die Flüssigkeitsaufnahme war gering oder unterblieb völlig bei Diäten mit niedrigen Rohrzucker-(<5%) oder Gesamtaminosäurekonzentrationen (<1%). Für Rohrzucker zeigte sich ein Optimalgehalt von 10–20% und ein fortschreitendes Absinken der Flüssigkeitsaufnahme bei Konzentrationen über 20%. Bei den Aminosäuren stieg die Flüssigkeitsaufnahme mit der Konzentration an Nährstoffen bis zu 3% und verringerte sich dann schwach.Die Ergebnisse werden in Beziehung gesetzt zu Wachstumsraten von M. persicae-Larven auf Diäten ähnlicher Zusammensetzung. Die ökologische Bedeutung der Befunde wird erörtert.

Few Myzus persicae settled on polygodial (1 g litre-1) or dodecanoic acid (5 g litre-1)-treated leaves and few nymphs were deposited. Polygodial decreased acquisition of the semi-persistent beet yellows virus and the non-persistent potato virus Y and dodecanoic acid that of the persistent potato leafroll and beet mild yellowing viruses, of beet yellows virus and of the bimodally-transmitted cauliflower mosaic virus. Abstract. 1. Many aphids form mixed populations of cyclical and obligate parthenogens. This is puzzling, because all else being equal, obligate parthenogens should outcompete cyclical parthenogens due to the two‐fold cost of sex. Yet cyclical parthenogens produce frost‐resistant, diapausing eggs in autumn, while obligate parthenogens spend the winter as active stages. Frost resistance thus represents a short‐term advantage to sexual reproduction mediated by winter temperatures, which may promote this coexistence. 2. Because obligate parthenogens overwinter as active stages, there may be selection for increased cold tolerance compared to cyclical parthenogens. This has the potential to gradually erode the advantage of sexually producing eggs. 3. Four obligately and four cyclically parthenogenetic lines of Myzus persicae (Sulzer) (Hemiptera: Aphididae) were collected from each of two areas differing in winter severity, and their survival after exposure to a severe experimental frost (14 h at ?9 °C), as well as their reproductive performance at a low (10 °C) and a high (20 °C) temperature were compared. 4. There was significant variation among lines in survival after the experimental frost, but this variation was neither related to their reproductive mode, nor to their area of origin. Similarly, neither reproductive mode nor origin had a significant effect on reproductive performance, independent of temperature. The average slope of the response to variation in temperature was also similar for both reproductive modes, despite the fact that slopes differed significantly among lines. 5. Within the limits of extrapolating from laboratory experiments, it is concluded that in M. persicae, the active stages of obligate parthenogens are not better adapted to cold temperatures than those of cyclical parthenogens.  相似文献

Myzus persicae develop well on Brassica species, particularly those that grow rapidly, such as mustard and turnip, but much less well on sugar beet, spinach and lettuce. They usually develop better on young, rapidly growing leaves than on mature leaves. Two clones of M. persicae behaved similarly on the host plants tested.
Zusammenfassung Die Entwicklung von Myzus persicae-Alienicolen auf den Sekundärwirten Kohlrüben, Kopfkohl, Chinakohl, Brokkoli, Blumenkohl, Senf, Raps, Grünkohl sowie Zuckerrübe, Spinat und Salat wurde durch wiederholte Erfassung der durchschnittlichen und totalen Larvenproducktion einzelner Jungfern sowie ihrer Mortalität in Gewächshausversuchen verfolgt und mit Auszählung des Befalls von Freilandpflanzen verglichen. Myzus persicae entwickelte sich gut an Brassica-Arten, besonders an raschwüchsigen wie Senf und Kohlrüben, aber viel weniger gut an Zuckerrübe, Spinat and Salat. Sie entwickelt sich gewöhnlich an jungen schnell wachsenden Blättern besser als an ausgereiften. Zwei Klone von Myzus persicae verhielten sich an den geprüften Wirtspflanzen ähnlich.

The antennal sensilla of alate Myzus persicae were mapped using transmission electron microscopy and the ultrastructure of sensilla trichoidea, coeloconica, and placoidea are described. Trichoid sensilla, located on the tip of the antennae, are innervated by 2–4 neurons, with some outer dendrites reaching the distal end of the hair. Coeloconic sensilla in primary rhinaria are of two morphological types, both equipped with two dendrites. Dendrites of Type II coeloconic sensilla are enveloped in the dendrite sheath, containing the sensillum lymph. In sensilla coeloconica of Type I, instead, dendrites are enclosed by an electron opaque solid cuticle, with no space left for the sensillum lymph. The ultrastructure of big placoid sensillum reveals the presence of three groups of neurons, with 2–3 dendrites in each neuron group, while both small placoid sensilla are equipped with a single group of neurons, consisting of three dendrites. Both large and small placoid sensilla bear multiple pores on the outer cuticle. The function of these sensilla is also discussed. J. The green peach aphid (Myzus persicae) is a phloem-feeding insect that causes economic damage on a wide array of crops. Using a luminol-based assay, a superoxide-responsive reporter gene (Zat12::luciferase), and a probe specific to hydrogen peroxide (HyPer), we demonstrated that this aphid induces accumulation of reactive oxygen species (ROS) in Arabidopsis thaliana. Similar to the apoplastic oxidative burst induced by pathogens, this response to aphids was rapid and transient, with two peaks occurring within 1 and 4 hr after infestation. Aphid infestation also induced an oxidative response in the cytosol and peroxisomes, as measured using a redox-sensitive variant of green fluorescent protein (roGFP2). This intracellular response began within minutes of infestation but persisted 20 hr or more after inoculation, and the response of the peroxisomes appeared stronger than the response in the cytosol. Our results suggest that the oxidative response to aphids involves both apoplastic and intracellular sources of ROS, including ROS generation in the peroxisomes, and these different sources of ROS may potentially differ in their impacts on host suitability for aphids.  相似文献

The results presented in this paper suggest that sugar beet became less suitable as a host for aphids after late June and early July, when the plants were at the 10–12 leaf stage. This was indicated by a faster rate of aphid mortality and greater incidence of dark stomach deposits after this time. Increased aphid mortality coincided with a change in physiology of sugar beet leaves from being net sinks to sources of assimilate. Aphids which fed on older leaves, irrespective of plant age, suffered greater mortality than those feeding on young heart leaves. The incidence of dark deposits in aphid stomachs associated with this mortality was greater on outer than on inner leaves of old, mature and young plants. It is suggested that the incidence of the dark deposit is an indicator of declining plant quality. The consequences of this for the spread of beet yellowing viruses are discussed.  相似文献
