Demodex microti n. sp. (Acari, Demodecidae) is described from the skin of the genital area of the common vole Microtus arvalis (Pallas) in Poland based on the morphology of the adult and immature stages. The new species appears most similar to D. cricetuli Hurley & Desch, 1994 from the gray dwarf hamster, Cricetulus migratorius (Pallas), but differs in the following features: the supracoxal spines are conical and located at the edge of the gnathosoma, the spines of the terminal segment of palp are three, single-tined, the opisthosomal organ is narrow and elongated in males and bubble-shaped in females, the vulva is located below the incision of the fourth pair of epimeral plates, eggs are oval. The differences also relate to body size and proportions, D. microti n. sp. being smaller and slender. The new species occurred in all of the rodents examined. A checklist of demodecid mites in cricetids world-wide is also provided.  相似文献

Hippobosca longipennis (Diptera: Hippoboscidae), the dog fly or dog louse fly, is an obligate blood‐feeding ectoparasite of wild and domestic carnivores in Africa and the Middle East. Outside its typically known geographic range, H. longipennis has been reported occasionally on mainly domestic dogs in Asia and southern Europe, and infrequently in other areas (central Europe and the U.S.A.). This paper presents the first report of H. longipennis in Romania and the second record of Lipoptena fortisetosa (Diptera: Hippoboscidae), a potentially invasive species. Hippobosca longipennis was found on domestic dogs in two regions of the country (northern Romania in Maramures and southwestern Romania in Dobrogea) and on two road‐killed wildcats in Maramures. Lipoptena fortisetosa was found on domestic dogs in Maramures. In both species identification was based on morphology and confirmed by barcoding of the cytochrome c oxidase subunit 1 gene. It is not clear for how long H. longipennis has been present in central Europe, nor if it was introduced (via the movement of domestic dogs or import of exotic carnivores) or present historically (Holocene remnants). This paper discusses the possible origins of H. longipennis in central Europe as its current distribution in the area is sparse and patchy.  相似文献

【目的】分析毛囊蠕形螨Demodex folliculorumD.f.)和皮脂蠕形螨D. brevisD.b.)基因组DNA的多态性, 对相关条带进行测序分析。【方法】采用改良小昆虫DNA提取法提取两种人体蠕形螨基因组DNA, 选择RAPD技术对其进行多态性分析, 将相关条带分别与pMD18-T载体连接, 克隆、测序后进行酶切鉴定和分析。【结果】毛囊蠕形螨共扩增15条带, 皮脂蠕形螨共扩增12条带;两种蠕形螨既有共有条带, 又有特异性条带;根据条带差异计算得到两种间的遗传距离为0.5556. 毛囊蠕形螨约800 bp处特异性条带测序结果显示, 序列片段长度为855 bp(GenBank登录号为FI277970);特异性引物扩增和酶切鉴定均为毛囊蠕形螨所特有. 序列比对显示与阿糖胞苷DNA区域结合蛋白有46%的序列相似度。两种人体蠕形螨约300 bp处共有条带序列分析显示, 碱基序列均为341 bp(GenBank登录号分别为D.f. FI520176;D.b. FI520175), 在第84和第165位点有2个碱基不同, 分别是A/G和C/T互换, 同源性高达99.4%. 但未发现有开放阅读框和相似度高的序列。 【结论】序列片段为855 bp的特异性条带为毛囊蠕形螨所特有;341 bp碱基序列为毛囊蠕形螨和皮脂蠕形螨所共有, 同源性高达99.4%. RAPD技术可用于两种人体蠕形螨基因组DNA的多态性分析和物种鉴定。  相似文献

We describe the construction of a dog embryonic head/neck cDNA library and the isolation of the dog homolog of the Treacher Collins Syndrome gene, TCOF1. The protein shows a similar three-domain structure to that described for human TCOF1, but the dog gene lacks exon 10 and contains two exons not present in the human sequence. In addition, exon 19 is differentially spliced in the dog. How these structural differences relate to TCOF1 phosphorylation is discussed. Isolation of a genomic clone allowed the exon/intron boundaries to be characterized and the dog TCOF1 gene to be mapped to CF Chr 4q31, a region syntenic to human Chr 5. Genetic analysis of DNA of dogs from 13 different breeds identified nine DNA sequence variants, three of which gave rise to amino acid substitutions. Grouping dogs according to head type showed that a C396T variant, leading to a Pro117Ser substitution, is associated with skull/face shape in our dog panel. The numbers are small, but the association between the T allele and brachycephaly, broad skull/short face, was highly significant (p= 0.000024). The short period of time during which the domestic dog breeds have been established suggests that this mutation has arisen only once in the history of dog domestication. 【目的】探索人体毛囊蠕形螨和皮脂蠕形螨DNA的提取方法。【方法】采用液氮反复冻融研磨法破碎螨体细胞, 选用改良小昆虫DNA提取法、碱裂解法和试剂盒提取法, 分别提取冻存时间在5个月内和8~10个月的毛囊蠕形螨和皮脂蠕形螨基因组DNA, 并用随机引物PCR方法进行检测。【结果】蛋白核酸测定仪检测结果显示, 试剂盒法提取的DNA纯度较高、量较多, 明显优于改良小昆虫法和碱裂解法。随机引物扩增结果显示清晰的DNA指纹图谱, 两种人体蠕形螨DNA指纹具有明显差异。蠕形螨冻存时间影响DNA提取的量, 但对DNA提取的纯度和RAPD指纹图谱影响较小。不同DNA提取方法提取的同一种蠕形螨DNA指纹图谱基本相似, 试剂盒法和改良小昆虫法提取的DNA样本条带多而清晰, 碱裂解法提取的样本条带少而模糊。【结论】液氮反复冻融研磨法破碎蠕形螨细胞是有效的, 蠕形螨冻存时间不宜超过6个月, 试剂盒提取法是提取蠕形螨DNA的好方法。RAPD技术可以用于这两种人体蠕形螨DNA分子水平上的检测和分类。  相似文献

本文报道了大连地区人体正常皮肤蠕形螨的生态分布。共检查551人,阳性153人,定居率为27.8%,检出毛囊螨和皮脂螨共两种。用透明胶带定量计数检查法,研究蠕形螨在面部的分布,结果表明蠕形螨主要分布于颏、鼻、颊和额部位。对10例阳性者,在面部检出毛囊螨和皮脂螨共85只,其中颏31、鼻26、颊17、额11只螨,总定居度为每条胶带(6cm~2)2.1只螨,定居度以颏部位为最高,鼻、颊、额次之。  相似文献

Type 10 collagen alpha 1 (COL10A1) is a short-chain collagen of cartilage synthesized by chondrocytes during the growth of long bones. COL10A1 mutations, which frequently result in COL10A1 haploinsufficiency, have been identified in patients with Schmid metaphyseal chondrodysplasia (SMCD), a cartilage disorder characterized by short-limbed short stature and bowed legs. Similarities between SMCD and short stature in various dog breeds suggested COL10A1 as a candidate for canine skeletal dysplasia. We report the sequencing of the exons and promoter region of the COL10A1 gene in dog breeds fixed for a specific type of skeletal dysplasia known as chondrodysplasia, breeds that segregate the skeletal dysplasia phenotype, and control dogs of normal stature. Thirteen single nucleotide polymorphisms (SNPs), one insertion, and two deletions, one of which introduces a premature stop codon and likely results in nonsense-mediated decay and the degradation of the mutant allele product, were identified in the coding region. There appear to be no causal relationships between the polymorphisms identified in this study and short stature in dogs. Although COL10A1 haploinsufficiency is an important cause of SMCD in humans, it does not seem to be responsible for the skeletal dysplasia phenotype in these dog breeds. In addition, homozygosity for the nonsense allele does not result in the observed canine skeletal dysplasia phenotype. Well-informed biodiversity conservation practice can often be precluded by poor species detectability. For example, populations being missed during surveys can lead to them being omitted from species lists or area management plans. iDNA (invertebrate-derived DNA) is a recently developed set of techniques for improving the detectability of elusive vertebrates by exploiting their associated invertebrates. Parasitic and scavenging invertebrates can be readily collected, and their gut contents DNA barcoded to detect local vertebrate diversity. However, most iDNA surveys have targeted mammals and have been carried out in tropical areas and/or rainforests. We carried out iDNA surveys targeting frogs in temperate dry sclerophyll forests in south-eastern Australia. We set mosquito traps broadcasting recorded frog calls with the aim of collecting frog-biting flies, which are attracted to frog calls. We collected 156 fly specimens, although none were of frog-biting species, and no frogs were detected via iDNA, despite many being observed in the field. However, two mammal and one reptile species were detected via iDNA: the feral cat (Felis catus: Felidae), domestic dog or dingo (Canis lupus: Canidae) and the threatened Rosenberg's monitor (Varanus rosenbergi: Varanidae). Vertebrate-sampling flies are likely highly abundant in the area since they were collected apparently incidentally in traps lacking appropriate attractants; a promising result for further surveys is different attractants are employed. This study is one of the few in which an invasive species has been detected through iDNA, and highlights its potential for improved detectability of threatened species outside of the tropics and early detection of invasive species.  相似文献

The genus Piophila Fallén (Diptera: Piophilidae) is known from only two species: Piophila casei (L.) is a major pest, a cosmopolitan species and is commonly used as a forensic indicator, whereas Piophila megastigmata McAlpine has until now only been recorded in a natural environment in South Africa. The present work reports the first occurrence of P. megastigmata in the Palaearctic region from specimens collected by carrion‐baited traps throughout different natural habitats of central Spain. Furthermore, the species was also collected with P. casei on corpses of domestic pigs used in a carrion succession study in a periurban habitat in central Spain. Both species occurred on carrion in different seasons, but P. megastigmata was more abundant than P. casei in autumn, arriving earlier at the carcasses and persisting for a longer period. The contrary pattern was observed in spring. The presence of P. megastigmata in different localities in central Spain and its coexistence with P. casei in a periurban habitat make it a potentially useful new tool for legal medicine in Europe; thus this species must be considered in forensic entomology studies.  相似文献

Reliable population estimation and species inventories are important for wildlife conservation, but such estimations are often difficult due to unreliable identification of the species in question. Furthermore, for predator conflict resolution, it is essential to be able to reliably identify the predator. This study presents a new method to quantitatively distinguish African wild dog (Lycaon pictus) footprints from feral domestic dog (Canis lupus familiaris) footprints. Footprint photographs were digitally processed using Photoshop and the NIH image processing software ImageJ, and total pad area and angles between the centroids of the backpad and the digits of the paw were measured. Pad angles showed statistically significant differences between the two species and, with the exception that there was no significant difference in pad area between African wild dog females and domestic dog males, total pad areas were also diagnostic. Consequently, the combination of total pad area and the angle between backpad and digits are useful discriminators to reliably identify the species from an unknown footprint.  相似文献

We tested the hypothesis that black-tailed prairie dogs (Cynomys ludovicianus) influence avian community structure on the shortgrass prairie. We surveyed 36 prairie dog towns and 36 paired sites without prairie dogs during summer and fall of 1997, 1998, and 1999 in the Oklahoma Panhandle. Our surveys totaled 9,040 individual observations for 73 avian species. Significantly distinct avian communities were present on prairie dog towns when compared to sites within four different macrohabitats of the surrounding landscape: open rangeland, scrub/sandsage (Artemisia filifolia) habitats, Conservation Reserve Program (CRP) plots, and fallow crop fields. Relative densities of all bird species combined was higher on prairie dog towns versus paired sites in summer and fall. Mean species richness of birds was significantly higher on prairie dog towns than paired sites during summer, but there were no significant differences in fall. Open rangeland had the highest mean species richness in fall. Assemblages of avian communities differed significantly between prairie dog towns and the four macrohabitat types during summer. Burrowing owls (Athene cunicularia), killdeer (Charadrius vociferous), horned larks (Eremophila alpestris), and meadowlarks (Sturnella spp.) were positively and significantly associated with prairie dog towns during summer, while horned larks and ferruginous hawks (Buteo regalis) were significantly associated with prairie dog towns during fall. Even in their current remnant state, black-tailed prairie dogs continue to play a significant role in the assembly of ecological communities across the Great Plains. Conservation of prairie dogs goes well beyond a single species, and is an important strategy for the preservation of the prairie ecosystem as a whole.  相似文献

Demodex folliculorum and D. brevis are cosmopolitan, obligatory parasites. Epidemiological studies have established a clear association between these species and various facial diseases in humans. However, not much is known of the ecology of these mites, also because it has proven difficult to culture them. Here we report a laboratory study on the (combined) effects of temperature and culture medium on the viability of both species. Results showed significant differences in the survival time of D. folliculorum and D. brevis among different temperatures and media. Pair-wise comparisons revealed that both species survived longest in serum medium, at a temperature of 16–22°C. Therefore, we conclude that these conditions allow preservation and maintenance of both Demodex mites.  相似文献

Oculocutaneous albinism type 4 (OCA4) in humans and similar phenotypes in many animal species are caused by variants in the SLC45A2 gene, encoding a putative sugar transporter. In dog, two independent SLC45A2 variants are known that cause oculocutaneous albinism in Doberman Pinschers and several small dog breeds respectively. For the present study, we investigated a Bullmastiff with oculocutaneous albinism. The affected dog was highly inbred and resulted from the mating of a sire to its own grandmother. We obtained whole genome sequence data from the affected dog and searched specifically for variants in candidate genes known to cause albinism. We detected a single base deletion in exon 6 of the SLC45A2 gene (NM_001037947.1:c.1287delC) that has not been reported thus far. This deletion is predicted to result in an early premature stop codon. It was confirmed by Sanger sequencing and perfectly co‐segregated with the phenotype in the available family members. We genotyped 174 unrelated dogs from diverse breeds, all of which were homozygous wildtype. We therefore suggest that SLC45A2:c.1287delC causes the observed oculocutaneous albinism in the affected Bullmastiff.  相似文献

Equine piroplasmosis (EP), caused by two parasitic organisms, Theileria equi and Babesia caballi, is a tick‐borne disease of recent concern in horses in the U.S.A. Outbreaks of EP have been detected in Florida, Missouri, Kansas and Texas. In 2009, EP transmission in Texas occurred through the adults of two tick species, Amblyomma mixtum [formerly known as Amblyomma cajennense (Fabricius, 1787)] Koch (Ixodida: Ixodidae) and Dermacentor variabilis (Say) (Ixodida: Ixodidae), the American dog tick (ADT). In this study, we developed a continent‐scale map for the distribution of the EP vector species D. variabilis, using a presence‐only modelling approach to assess the habitat preferences of this tick. We used identification records from our tick geodatabase of locations in which the presence of the ADT had been noted. The potential distribution of the ADT in the U.S.A. was estimated from environmental factors using the maximum entropy approach based on localities in which there is a high probability of occurrence according to habitat suitability. Elevation and temperature were found to be biologically significant environmental variables influencing the presence of this tick species. Properly designed and constructed probability surfaces using maximum entropy offer one useful approach to the mapping of distribution ranges of tick species based on suitable habitat in the U.S.A.  相似文献

Members of the rare microbiome can be important components of complex microbial communities. For example, pet dog ownership is a known risk factor for human campylobacteriosis, and Campylobacter is commonly detected in dog feces by targeted assays. However, these organisms have not been detected by metagenomic methods. The goal of this study was to characterize fecal microbiota from healthy and diarrheic pet dogs using two different levels of molecular detection. PCR amplification and pyrosequencing of the universal cpn60 gene target was used to obtain microbial profiles from each dog. To investigate the relatively rare epsilon-proteobacteria component of the microbiome, a molecular enrichment was carried out using a PCR that first amplified the cpn10–cpn60 region from epsilon-proteobacteria, followed by universal cpn60 target amplification and pyrosequencing. From the non-enriched survey, the major finding was a significantly higher proportion of Bacteroidetes, notably Bacteroides vulgatus, in healthy dogs compared to diarrheic dogs. Epsilon-proteobacteria from the genera Helicobacter and Campylobacter were also detected at a low level in the non-enriched profiles of some dogs. Molecular enrichment increased the proportion of epsilon-proteobacteria sequences detected from each dog, as well as identified novel, presumably rare sequences not seen in the non-enriched profiles. Enriched profiles contained known species of Arcobacter, Campylobacter, Flexispira, and Helicobacter and identified two possibly novel species. These findings add to our understanding of the canine fecal microbiome in general, the epsilon-proteobacteria component specifically, and present a novel modification to traditional metagenomic approaches for study of the rare microbiome.  相似文献

The synlophes of Mazamastrongylus odocoilei (Dikmans, 1931) and M. pursglovei (Davidson & Prestwood, 1979) from Odocoileus virginianus were described to provide characteristics for their differentiation from related species in domestic and wild ruminants. The synlophes of M. odocoilei and M. pursglovei were identical. Both had single continuous dorsal, ventral and lateral ridges. Other ridges in the lateral fields in the anterior third of the body angled posteriorly ending adjacent to the single lateral ridge. The ridge system in Mazamastrongylus spp. appears to be unique in lacking the continuous subventral and subdorsal ridges present in all previously studied medium stomach worms from domestic ruminants. Only two characters, spicule length and structure, are known to be useful for differentiating males of M. odocoilei and M. pursglovei, and females cannot be differentiated. Evidence that M. odocoilei and M. pursglovei are separate species include: (1) the two species are frequently found together in the same individual host, but each is also found in the absence of the other; (2) no overlap in spicule size or intergrades in shape have been found although numerous populations have been sampled; and (3) the two species have overlapping, but different, geographical ranges in eastern North America. No differences in the synlophe were found between these two species. The importance of this finding is that the absence of differences in the synlophe is not an unequivocal indicator that nematodes are conspecific.  相似文献

Among the CD1 proteins that present lipid antigens to T cells, CD1d is the only one that stimulates a population of T cells with an invariant T‐cell receptor known as NKT cells. Sequencing of a 722 nucleotide gap in the dog (Canis lupus familiaris) genome revealed that the canine CD1D gene lacks a sequence homologous to exon 2 of human CD1D, coding for the start codon and signal peptide. Also, the canine CD1D gene contains three different short tandem repeats that disrupt the expected gene structure. Because canine CD1D cDNA lacks sequences homologous to human exon 2 and 3, the functionality of canine CD1d protein may be affected, and this could have consequences for the development and activation of canine NKT cells.  相似文献

Alpine plants of the eastern Qinghai-Tibetan plateau (Sichuan, China) are developed under long-lasting grazing by wild and domestic yaks. Among morphological features of plants, life forms may reflect their adaptation to grazing. We studied life-form composition of four typical communities within the alpine belt (3930–3960 m a.s.l.) subjected to grazing of various intensity: alpine fen (heavily grazed), alpine shrub meadow (heavily grazed), Spiraea alpina thicket (grazed), and Rhododendron thicket (practically not grazed). The following morphological traits were studied: (1) life form according to Raunkiaer, (2) life form according to Serebryakov, (3) canopy structure, and (4) rate of lateral spreading. We derived life-form spectra based on (1) the number of species per life form and (2) the cumulative abundance of species which have the same life form. One-way ANOVA and nonparametric ANOVA were run to test for significance of differences between spectra. The studied communities differed significantly by the proportion of different life forms. The main life forms are caudex and short rhizome hemicryptophytes, nonclonal species, or species with a low rate of lateral spreading. Therophytes made up 10–11% of the communities except in Rhododendron thickets, where such were absent. These life forms can indicate grazing in the study area.  相似文献

The raccoon (Nyctereutes procyonoides) is described as a host of the dog biting louse, Trichodectes canis (De Geer, 1778). The present note reports the occurrence of this species of louse on two raccoon dogs from Moravia and Bohemia. The specimens of T. canis collected on this host differed from T. canis originating from the domestic dog by body size only.  相似文献

Rabies virus (RV) and canine distemper virus (CDV) can cause significant mortality in wild carnivore populations, and RV threatens human lives. We investigated serological patterns of exposure to CDV and RV in domestic dogs (Canis familiaris), African wild dogs (Lycaon pictus), black-backed jackals (Canis mesomelas), spotted hyenas (Crocuta crocuta), striped hyenas (Hyaena hyaena) and African lions (Panthera leo), over a 10-year period, in a Kenyan rangeland to assess the role domestic dogs may play in the transmission dynamics of these two important canid pathogens. Observed patterns of RV exposure suggested that repeated introduction, rather than maintenance, occurred in the wild carnivore species studied. However, RV appeared to have been maintained in domestic dogs: exposure was more likely in domestic dogs than in the wild carnivores; was detected consistently over time without variation among years; and was detected in juveniles (≤1-year-old) as well as adults (>1-year-old). We conclude that this domestic dog population could be a RV reservoir. By contrast, the absence of evidence of CDV exposure for each carnivore species examined in the study area, for specific years, suggested repeated introduction, rather than maintenance, and that CDV may require a larger reservoir population than RV. This reservoir could be a larger domestic dog population; another wildlife species; or a “metareservoir” consisting of multiple interconnected carnivore populations. Our findings suggest that RV risks to people and wild carnivores might be controlled by domestic dog vaccination, but that CDV control, if required, would need to target the species of concern.  相似文献
