Intracellular degradation of newly synthesized collagen is quantitated by incubating fibroblasts with [14C]proline and determining the percentage of total [14C]hydroxyproline that is present in a low molecular weight fraction. Several problems make this difficult. (1) Commercial [14C]proline is often contaminated with [14C]hydroxyproline and must be purified before use. (2) Salt and [14C]proline interfere with the determination of [14C]hydroxyproline in the low molecular weight fraction and must be removed by preparative ion-exchange chromatography. (3) Epimerization of trans- to cis-hydroxyproline during acid hydrolysis is variable and must be taken into account. (4) Loss of [14C]hydroxyproline during processing varies; [3H]hydroxyproline can be used as an internal measure of recovery, even though tritium may be lost during hydrolysis. An analytic cation-exchange resin is used for the final quantitation of [14C]hydroxyproline in the low and high molecular weight fractions. With these methods, degradation of newly synthesized collagen can be determined with a precision of ± 3%.  相似文献

Incorporation of L-[3H]fucose into glycoproteins was studied in R2, the giant neuron in the abdominal ganglion of Aplysia. [3H]fucose injected directly into the cell body of R2 was readily incorporated into glycoproteins which, as shown by autoradiography, were confined almost entirely to the injected neuron. Within 4 h after injection, 67% of the radioactivity in R2 had been incorporated into glycoproteins; at least 95% of these could be sedimented by centrifugation at 105,000 g, suggesting that they are associated with membranes. Extraction of the particulate fraction with sodium dodecyl sulfate (SDS), followed by gel filtration on Sephadex G-200 and polyacrylamide gel electrophoresis in SDS revealed the presence of only five major radioactive glycoprotein components which ranged in apparent molecular weight from 100,000 to 200,000 daltons. Similar results were obtained after intrasomatic injection of [3H]N-acetylgalactosamine. Mild acid hydrolysis of particulate fractions released all of the radioactivity in the form of fucose. When ganglia were incubated in the presence of [3H]fucose, radioactivity was preferentially incorporated into glial cells and connective tissue. In contrast to the relatively simple electrophoretic patterns obtained from cells injected with [3H]fucose, gel profiles of particulate fractions labeled with [14C]valine were much more complex.  相似文献

Polysomes were isolated from aortas of 17-day-old chick embryos, and the synthesis of the nascent polypeptide chains was completed in vitro. When a mixture of a labeled amino acids found in elastin was used, the major radioactive product obtained was of molecular weight about 70,000 and was similar to elastin by several criteria. The 70,000 molecular weight product was extractable in propanol-butanol, it was not labeled with [35S]methionine, and it was precipitated by antibodies against elastin. Polypeptides larger than 70,000 molecular weight were also synthesized but these larger polypeptides incorporated relatively small amounts of [14C]valine, and they appeared to represent proα chains of procollagen. The results suggest that the major gene product for elastin has a molecular weight of about 70,000.  相似文献

The transfer of mannose from GDP[14C]mannose to lipid and to insoluble polymer by a particulate preparation of Phaseolus aureus has been investigated. The evidence favours the lipid being a prenol phosphate mannose. Of a range of prenol phosphates tried, betulaprenol phosphate was the most effective exogenous acceptor of mannose. Most of the insoluble [14C]polymer formed was glycoprotein in nature although small quantities of 14C were associated with glucomannan and galactoglucomannan fractions. Time studies failed to reveal a typical precursor-product relationship between the lipid and polymer fractions but on incubation of [14C]mannolipid with the particulate fraction a small transfer (0·5–0·7%) of [14C] to polymer was detected. p-Hydroxymercuribenzoate inhibited (by 90%) the transfer of [14C] from GDP[14C]-mannoseto polymer and simultaneously increased (3-fold) the [14C] recovered in the lipid fraction. The effect was nullified by mercaptoethanol. Attempts to solubilize the transfer system were only partially successful. The formation of a chromatographically identical mannolipid was demonstrated in particulate fractions of Codium fragile and tomato roots.  相似文献

Pea microsomal membranes catalyze the transfer of [14C]fucose (Fuc) from GDP-[U-14C]fucose, with or without added unlabeled UDP-glucose (Glc), UDP-xylose (Xyl) or UDP-galactose (Gal), to an insoluble product with properties characteristic of xyloglucan. After digestion of the ethanol-insoluble pellet with Streptomyces griseus endocellulase, [14C] fucose residues occur exclusively in a fragment corresponding in size to the xyloglucan nonasaccharide, Glc4 Xyl3 Gal Fuc. This fragment contains a single labeled fucose residue per oligomer, α-linked in a terminal nonreducing position. By comparison, in incubations where GDP-[14C] fucose is absent and replaced by UDP-[3H]xylose, the maximum size of labeled oligosaccharide found following cellulase digestion of products is an octasaccharide. In the presence of both GDP-[14C]fucose and UDP-[3H]xylose, a nonasaccharide containing the two labels is produced. Fucose and xylose residues are transferred within a few minutes to acceptor molecules of molecular weight up to 300,000. Such products do not elongate detectably over 60 minutes of incubation. The data support the conclusion that the nonasaccharide subunit of xyloglucan may be generated in vitro by transfucosylation to preformed acceptor chains, and that its synthesis is dependent on the inclusion of exogenous GDP-fucose.  相似文献

Microsomal membranes from elongating regions of etiolated Pisum sativum stems were separated by rate-zonal centrifugation on Renografin gradients. The transfer of labeled fucose and xylose from GDP-[14C] fucose and UDP-[14C]xylose to xyloglucan occurred mainly in dictyosomeenriched fractions. No transferase activity was detected in secretory vesicle fractions. Pulse-chase experiments using pea stem slices incubated with [3H]fucose suggest that xyloglucan chains are fucosylated and their structure completed within the dictyosomes, before being transported to the cell wall by secretory vesicles.  相似文献

Phospholipase A2 modification of lipid-protein interactions of normal O,Rh(D) positive erythrocyte membranes increased the fluorescence intensity of the membrane bound probe, 1-anilinonaphthalene-8-sulfonate (ANS) and increased the N-1-[14C]-ethyl maleimide ([14C]-NEM) labeling of sulfhydryl groups in two proteins of molecular weight >200,000. In marked contrast, phospholipase A2 modification of the rare phenotype O,Rhnull membranes resulted in no significant increase in ANS fluorescence or labeling of sulfhydryl groups by [14C] NEM. Since the O,Rhnull erythrocytes demonstrated an increased osmotic fragility and decreased survival time, the fluorescence and sulfhydryl labeling data support the conclusion that hydrophobic bonding between β-fatty acid side chains and non-polar regions of asymmetric proteins is necessary for maintaining the native structure of the O,Rh(D) positive membrane. Comparative studies with phospholipase C or D implied that ionic bonding played a similar though less important structural role in both membranes.  相似文献

Radioactive polysaccharide was synthesized when uridine 5′-(α-d-[U-14C]apio-d-furanosyl pyrophosphate) (containing some uridine 5′-(α-d-[U-14C]xylopyranosyl pyrophosphate)) was incubated with a particulate enzyme preparation from Lemna minor. Characterization experiments established that the product: (i) was insoluble in methanol and water, (ii) contained d-[U-14C]apiose (75%) and d-[U-14C]xylose (25%), and (iii) was soluble in 1% ammonium oxalate. The material solubilized by ammonium oxalate (solubilized product): (i) was separated into five fractions by column chromatography with diethylaminoethyl-Sephadex (DEAE-Sephadex), (ii) contained [U-14C]apiobiose side chains that were removed by hydrolysis at pH 4, and (iii) was degraded by fungal pectinase. Both d-[U-14C]apiose residues of the [U-14C]apiobiose side chains were synthesized in vivo since radioactivity was distributed equally between the two residues. The presence of uridine 5′-(α-d-galactopyranosyluronic acid pyrophosphate) during synthesis of radioactive polysaccharide resulted in: (i) an increase in the incorporation of radioactive d-[U-14C]apiose into solubilized product, (ii) an increase in the ratio of d-[U-14C]apiose to d-[U-14C]xylose present in solubilized product, (iii) an increase in the amount of [U-14C]apiobiose plus d-[U-14C]apiose released from the solubilized product by hydrolysis at pH 4, and (iv) a tighter binding of the solubilized product to DEAE-Sephadex. These results show that apiogalacturonans similar to or the same as those synthesized by the intact plant were synthesized in the particulate enzyme preparation isolated from L. minor. [14C]Apiogalacturonans completely free of d-[U-l4C]xylose were not isolated. The [14C]apiogalacturonan with the least d-[U-14C]xylose still had 4.8% of its radioactivity present in d-[U-14C]xylose. The possibility remains that d-xylose is a normal constituent of the apiogalacturonans of the cell wall of L. minor.  相似文献

Polysomes were isolated from Aspergillus niger and were characterized on sucrose gradients in several ways. First, they were found to be susceptible to degradation by treatment with RNase or EDTA. Second, they were labeled after treating mycelia with short pulses of [3H]uridine or [3H]leucine prior to polysome isolation. Third, they were capable of stimulating incorporation of [3H]leucine into trichloroacetic acid-precipitable material in a chick reticulocyte cell-free protein-synthesizing system. When isolated [3H]leucine pulse-labeled polysomes were treated with either EDTA-RNase or puromycin, 80–90% of the radioactivity was released, indicating that only the nascent polypeptide chains were labeled. After exposing mycelia for 1 min to [14C]mannose, the polysomes were exclusively labeled, indicating that initial glycosylation takes place on nascent polypeptide chains. Preincubation of mycelia with 2-deoxyglucose followed by pulse-labeling with [3H]leucine and [14C]mannose showed that 2-deoxy-d-glucose inhibits both protein synthesis and glycosylation. However, similar preincubation with tunicamycin caused an 80% drop in [14C]mannose label in the polysomes, but only a 10–20% drop of [3H]leucine label, suggesting that glycosylation of nascent chains in A. niger involves an oligosaccharide-lipid intermediate, since it has been shown that tunicamycin inhibits the synthesis of such an intermediate. When isolated polysomes were placed into an in vitro glycosylating mixture containing Mn2+, GDP-[14C]mannose, and smooth membranes from A. niger nascent chains were labeled. This reaction was shown to be dependent on addition of polysomes to the mixture and was not inhibited by 2-deoxy-d-glucose or tunicamycin. Both in vivo and in vitro glycosylated nascent chains were found to have about the same size range, and so it is suggested that in vitro no new oligosaccharide chains were synthesized, but preexisting chains were extended.  相似文献

The distribution of nuclear non-histone proteins and their labelling with [14C]amino acids or [32P]O4 in rat ventral prostate cells undergoing hypertrophy (cell growth) or hyperplasia (cell division) were compared by SDS polyacrylamide gel electrophoresis. Marked quantitative changes in these proteins accompanied castration and replacement of hormone, but no absolute qualitative differences in their staining patterns were established. Preparations from cells undergoing hyperplasia were augmented in high molecular weight proteins and contained fewer proteins of <20000 D. The increased amounts of nuclear non-histone proteins from hormone-treated castrated rats were highly labelled by [14C]amino acids, but the molecular weights of radioactive proteins from cells undergoing hypertrophy were less diverse than those from replicating cells, Phosphorylation of nuclear non-histone proteins from short-term and long-term castrates, treated with testosterone propionate, was 170 and 60% greater than their controls. Proteins from 20–45 × 103 D were actively phosphorylated. Nuclear extracts from dividing cells contained additional radioactive high molecular weight proteins and fewer phosphorylated lower molecular weight components. The distribution of phosphorylated proteins and newly synthesized proteins was dissimilar. Quantitative and possible qualitative differences in staining of nuclear proteins isolated in N-ethylmalleamideurea-phosphate buffer from normal or hormone-treated castrated rats were accentuated when they were separated by charge at pH 2.8. In replicating cells, a more generalized synthesis of acidic nuclear proteins from all molecular weight classes occurred, which were not as highly phosphorylated as less heterogeneous nuclear proteins from cells undergoing hypertrophy. Examination of the immediate and the subsequent events following androgen-induced cellular hypertrophy or hyperplasia in the ventral prostate permits their comparison in the same tissue.  相似文献

A glucose receptor with high affinity for phlorizin from isolated brush border of rat kidney was labelled specifically withN-[14C]ethylmaleimide and then extracted from the membranes.After the solubilization of the brush borders with sodium dodecyl sulphate theN-[14C]ethylmaleimide-labelled receptor protein was isolated and was found to have a molecular weight of approximately 30 000 as determined by sodium dodecyl sulphate-polyacrylamide gel disc electrophoresis. The receptor protein eluted from the sodium dodecyl sulphate-containing gels migrates as a single band on sodium dodecyl sulphate-free polyacrylamide gels.The receptor protein can also be released from the brush borders with low concentrations of sodium deoxycholate. Under these conditions the molecular weight of theN-[14C]ethylmaleimide-labelled receptor protein is approximately 60 000 in contrast to the protein component solubilized with sodium dodecyl sulphate. Since this detergent is known to dissociate the brush border membrane into its protein components, our results suggest that the phlorizin- sensitive glucose receptor protein has a molecular weight of about 30 000.  相似文献

Membrane glycoprotein biosynthesis of ascites hepatoma cells is followed by [14C]glucosamine and [3H]leucine incorporation into cells in culture. The rate of incorporation is strongly increased by the addition of Robinia lectin in culture medium. Labeled glycoproteins are released from lectin stimulated and non-stimulated ceils by trypsin digestion. Studies of labeled trypsinates on sodium dodecyl sulfate gel electrophoresis and Sephadex G-200 filtration exhibit two fractions both labeled with [14C]glucosamine and [3H]leucine and having different molecular weights, one over 200 000 and the other about 2000. Identical results are obtained when external membrane glycoproteins are solubilized by sodium deoxycholate. Comparison of surface glycoproteins isolated by trypsinization from control cells labeled with [3H]glucosamine and from lectin stimulated cells labeled with [14C]glucosamine displays no significant qualitative differences between glycoprotein fractions released from both cell groups.  相似文献

A microsomal preparation from chick embryo epiphyseal cartilage was incubated with UDP-[14C]glucuronic acid and UDP-N-acetylgalactosamine to form [14C] chondroitin-labeled proteoglycan. Two [14C]proteoglycan populations were obtained which differed in size, [14C]glycosaminoglycan content, and susceptibility to alkali. One population of [14C]proteoglycan appeared near the void volume on Sepharose 2B, while the other population was smaller, similar in size to monomer proteoglycan. The larger [14C]proteoglycan contained long [14C]chondroitin chains added to short primers; these chains were in part resistant to alkali cleavage from protein. The smaller [14C]proteoglycan contained mainly [14C]chondroitin chains of intermediate length added to endogenous chondroitin sulfate; these chains were all susceptible to alkali cleavage from protein. The larger [14C]proteoglycan may represent a precursor proteoglycan present at the site of glycosaminoglycan chain synthesis.  相似文献

Three-day-old suspension cultured cells of Lycopersicon esculentum transferred to a Pi-depleted medium had 2.7 times the excreted acid phosphatase (Apase) activity of cells transferred to a Pi-sufficient medium. Cell growth during this time period was identical for the two treatments. Excreted Apase activity was resolved into two fractions on a Sephadex G-150 column. Most of the phosphate starvation inducible (psi) enhancement in activity was in the lower molecular weight fraction. These two fractions exhibited different substrate versus pH activity profiles. With a native polyacrylamide gel electrophoresis assay, the lower molecular weight fraction resolved into two bands of activity. Both column fractions resolved into the same single band of activity with sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The apparent molecular weight of this enzyme was 57 kilodalton. These data indicate that L. esculentum has at least two isozymes of the psi-excreted Apase and that these isozymes may associate to form high molecular weight aggregates. Labeling studies using [35S]methionine show that the psi response in tomato cells is complex and involves changes in the steady state levels of several excreted proteins.  相似文献

In developing rats, a fucose-labeled glycoprotein (mGP) which is closely associated with myelin has a higher apparent molecular weight in immature myelin in comparison with mature myelin. In order to see whether this developmental difference occurred in other species, we investigated different rodents of the suborder Myomorpha. Glycoproteins were labeled in vivo by injection of [3H]fucose of [14C]fucose, and purified myelin fractions were prepared. Proteins were extracted with SDS and separated by polyacrylamide gel electrophoresis. Using double-labeling techniques, the 3H- and 14C-labeled mGP were compared in different samples. We found the shift in the apparent molecular weight of mGP during development in rats, gerbils, and hamsters, but we were unable to demonstrate it in mice and prairie deer mice. In these latter species, the mGP of smaller apparent molecular weight was already present at the earliest ages at which it was possible to isolate myelin. Although some rodents do not exhibit the developmental change, all showed a common feature: the presence of the same small molecular weight mGP in mature myelin. It may be that the glycoprotein of smaller size is a prerequisite for the normal compaction and maturation of myelin.  相似文献

Cells were isolated from the aortae of 17-day old chick embryos by digestion of the vessels with a combination of trypsin and collagenase. When these cells were incubated in suspension culture in Krebs-Ringer media containing pancreatic trypsin inhibitor and radioactive amino acids, they synthesized and secreted labeled proteins into the media. Polyacrylamide gel electrophoresis in sodium dodecyl sulfate of the secreted proteins labeled with [14C]proline revealed two major components. The larger component with an approximate molecular weight of 125,000 had a [14C]hydroxyproline content consistent with a form of procollagen. The molecular weight of 70,000 and [14C]hydroxyproline content of the second component was consistent with that previously reported for tropoelastin extracted from chick aortae. By following the kinetics and secretion of tropoelastin labeled with [3H]valine, we have estimated that 17 minutes are required to synthesize and secrete the molecule under these experimental conditions.  相似文献

A particulate enzyme preparation isolated from Chrysochromulina chiton catalysed the transfer of [U-14C]-glucose from UDP [U-14C]-Glc to a water-soluble small molecular weight material. Chemical and enzymic analysis of this material showed that it was a phenolic compound to which are attached two β(1–3) glucosides. Properties of the UDP glucose: glucosyltransferase involved in the synthesis of this material have been studied. The UDP glucose glucosyl-transferase was found to be associated with the rough endoplasmic reticulum. A possible function of this phenolic compound in the orientation of membranes for the synthesis of scales in C. chiton has been discussed.  相似文献

The metabolism of d-gluconate-[1-14C] and -[6-14C] by segments from etiolated hypocotyls of Phaseolus mungo has been studied. The release of 14CO2 from gluconate-[1-14C] was greater than that from gluconate-[6-14C] in all parts of hypocotyls examined. Incorporation of the radioactivity from gluconate-[6-14C] into RNA, lignin and aromatic amino acid fractions was greater in the upper (younger) part of the hypocotyls. Incorporation into sugars was greater in the lower (more mature) parts.  相似文献
