In 1983, cereal plants showing symptoms of barley yellow dwarf virus (BYDV), collected from 15 localities in Sweden, were tested for BYDV using enzyme-linked immunosorbent assay (ELISA). Antisera against two Swedish isolates of BYDV were used, a mild isolate (27/77) transmitted specifically by Sitobion avenae and a severe one (39/78) transmitted mainly by Rhopalosiphum padi. No virus was detected in 57 of 607 plants of oats and barley tested. Of the 550 plants in which virus was detected, 366 were infected with viruses similar to isolate 27/77, 116 with viruses similar to 39/78 and the remaining 68 reacted strongly with both antisera. When tested, the latter isolates were shown to be mixtures. Thirty-nine selected samples were also tested with antisera against the USA isolates RPV, RMV, MAV and PAV, and for transmission by S. avenae and R. padi. Twenty-six of these samples were transmitted specifically by S. avenae, one was transmitted only by R. padi and the remaining 12 samples were shown to be infected with a mixture of an S. avenae-specific isolate and one transmitted mainly by R. padi. Antisera against PAV and MAV each detected all isolates tested and the results were very similar to those with the antisera to the 39/78 and 27/77 isolates, respectively. None of the field isolates reacted with antisera against RMV or RPV. It was concluded that 1983 was an epidemic year for BYDV in Sweden and that isolates specifically transmitted by S. avenae predominated. Symptoms of infection by these isolates on oat plants ranged from mild to severe.  相似文献

Developing longitudinal vascular bundles of the leaf blades of maize (Zea mays L. cv. W273) were examined with the transmission electron microscope to determine the frequency of plasmodesmata between the sieve tubes and their neighboring cells. Of particular interest were the protophloem sieve tubes, the first sieve tubes to mature in importing (all large and some intermediate) bundles. The protophloem sieve tubes, most of which lack companion cells, intergrade structurally with the thin-walled metaphloem sieve tubes. Both the protophloem sieve tubes and the thin-walled metaphloem sieve tubes and their companion cells (the sieve tube-companion cell complexes) are virtually isolated symplastically from the rest of the leaf, precluding a symplastic mechanism of phloem unloading in the leaf blade of maize.  相似文献

Rhopalosiphum padi and Sitobion avenae alates were collected from colonised winter cereals and maize in N.E. Spain and fed on young wheat plants for 7–10 days in the glasshouse. Then, aphids were killed and the plants on which aphids reproduced were kept in the glasshouse for 30–40 days. ELISA of infested plants was made using polyclonal and monoclonal antisera against PAV-, RPV- and MAV-like isolates. In autumn and spring, MAV serotypes were transmitted by S. avenae and R. padi , mainly in mixed infections with PAV serotypes. This possibly explains the high frequency of MAV-like isolates and their previously recorded year-to-year stability in maize, grain and forage winter cereals and cereal volunteers. PAV-like isolates were rarely transmitted by S. avenae and its spread thus depends almost exclusively on R. padi. These results confirm the importance of forage cereals and cereal volunteers as virus sources for winter cereal infection in the autumn, and the latter as a source of BYDV for maize in spring.  相似文献

Two Swedish isolates of barley yellow dwarf virus (BYDV), one (39/78) transmitted much more efficiently by Rhopalosiphum padi than by Sitobion avenae and the other (27/77) transmitted specifically by Sitobion avenae, were purified with yields of 200–300 μg/100 g infected oat leaves. Three light scattering zones were obtained when isolate 39/78 was sedimented in 10–40% (w/w) sucrose density gradients but only one zone with the other isolate. The sedimentation coefficients of 39/78 particles were 94,106 and 150 S, respectively, whereas the 27/77 particles sedimented at 110 S. Four protein bands of mol. wts 31 900, 29 700, 28 000 and 15 000 were detected when disrupted 150 5 particles of the 39/78 isolate were electrophoresed in SDS-polyacrylamide gels. Only one major band of mol. wt 24 500 was detected when 94 or 106 S particles were electrophoresed under the same conditions. The 27/77 isolate yielded one major band of mol. wt 23 500. A weak serological relationship was found between isolates 39/78 and 27/77. In enzyme-linked immunosorbent assay, isolate 39/78 was serologically closely related to New York isolate PAY, whereas 27/77 was closely related to MAY.  相似文献

An isolate of BYDV (BYDV-OC), from barley in Northwest Italy with typical symptoms of yellowing and dwarfing, was transmitted by Rhopalosiphum padi, Sitobion fragariae. S. avenae, Metopolophium festucae, R. maidis and M. dirhodum , but not by Myzus persicae or Schizaphis graminum . It reacted in DAS-ELISA with monoclonal and polyclonal antisera to PAV, but not with antibodies to MAV, RPV and RMV. A polyclonal antiserum prepared to BYDV-OC did not react with MAV-like, RPV-like, or RMV-like isolates of BYDV in ELISA or in Western blots. The concentration of BYDVOC in Avena byzantina plants decreased from weeks 1 to 10 after inoculation, but the total virus content per plant increased up to weeks 7 to 8, following the increase of plant weight.  相似文献

The presence of water-soluble compounds in sectioned plant tissue can be visualized in negative contrast by freeze-substitution in acetone, followed by embedment in Epon containing 6% Sudan B. The contents of mature sieve tubes and companion cells of bean (Phaseolus vulgaris L.) showed strong, and mostly uniform, negative staining. The degree of negative staining was measured by microspectrophotometry. Since, in sieve tubes, virtually all of the solute is the translocated sugar, the sugar concentration can be estimated by comparison with similar measurements made on sections from pith blocks which were infiltrated with sugar solutions and processed by the same procedures. Sieve tubes contained a solution of about 11.2% (w/v) sucrose; companion cells contained a similar concentration of sucrose. Populations of cereal aphids were sampled from 1985–1988 and assayed for transmission of barley yellow dwarf virus (BYDV), Rhopalosiphum padi, Rho-palosiphum maidis, Sitobion avenae, Metopolophium dirhodum, Schizaphis graminum and Macrosiphum euphorbiae collected from host plants transmitted BYDV in bioassays. Of the 1028 Diuraphis noxia collected from plants, one may have transmitted BYDV. The isolate involved resembled SGV in serological and biological characteristics, but since it was not recoverable by any of more than 800 D. noxia subsequently tested, we suspect it may have been a contaminant. Among those aphids collected during the autumn from a suction trap adapted for live collection, R. padi transmitted BYDV most frequently. Other trapped species which transmitted BYDV included: R. maidis, Rhopalosiphum insertum, Macrosiphum euphorbiae, Metopolophium dirhodum and Ceruraphis eriophori. An adapted Infectivity Index indicated that R. padi is by far the most important vector of BYDV during the autumn sowing season in southwestern Idaho. Male R. padi consistently transmitted BYDV more frequently than did females collected during the same period.  相似文献

Barley yellow dwarf virus has been detected in maize by indirect enzyme-linked immunosorbent assay (ELISA) and by immunospecific electron microscopy (ISEM). Samples of maize collected in September 1988, 1989 and 1990 showed that this crop is an important reservoir of BYDV; MAV-like isolates were the most common although PAV-like and RPV-like isolates were also present. Earlier research in Spain had shown that PAV-like isolates were predominant. Thus the evidence from this work that MAV was the main isolate, and very widely spread, is important for future research on BYDV epidemiology in Spain.  相似文献

Microautoradiographs showed that [14C]sucrose taken up in the xylem of small and intermediate (longitudinal) vascular bundles of Zea mays leaf strips was quickly accumulated by vascular parenchyma cells abutting the vessels. The first sieve tubes to exhibit 14C-labeling during the [14C]sucrose experiments were thick-walled sieve tubes contiguous to the more heavily labeled vascular parenchyma cells. (These two cell types typically have numerous plasmodesmatal connections.) With increasing [14C]sucrose feeding periods, greater proportions of thick- and thin-walled sieve tubes became labeled, but few of the labeled thin-walled sieve tubes were associated with labeled companion cells. (Only the thin-walled sieve tubes are associated with companion cells.) When portions of leaf strips were exposed to 14CO2 for 5 min, the vascular parenchyma cells-regardless of their location in relation to the vessels or sieve tubes-were the most consistently labeled cells of small and intermediate bundles, and label (14C-photosynthate) appeared in a greater proportion of thin-walled sieve tubes than thick-walled sieve tubes. After a 5-min chase with 12CO2, the thin-walled sieve tubes were more heavily labeled than any other cell type of the leaf. After a 10-min chase with 12CO2, the thin-walled sieve tubes were even more heavily labeled. The companion cells generally were less heavily labeled than their associated thin-walled sieve tubes. Although all of the thick-walled sieve tubes were labeled in portions of leaf strips fed 14CO2 for 5 min and given a 10-min 12CO2 chase, only five of 72 vascular bundles below the 14CO2-exposed portions contained labeled thick-walled sieve tubes. Moreover, the few labeled thick-walledsieve tubes of the transport region always abutted 14C-labeled vascular parenchyma cells. The results of this study indicate that (1) the vascular parenchyma cells are able to retrieve at least sucrose from the vessels and transfer it to the thick-walled sieve tubes, (2) the thick-walled sieve tubes are not involved in long-distance transport, and (3) the thin-walled sieve tubes are capable themselves of accumulating sucrose and photosynthates from the apoplast, without the companion cells serving as intermediary cells.  相似文献

Four PAV-like isolates of barley yellow dwarf virus (BYDV) were identified as causing very severe (RG), severe (2t), moderately severe (3b) and mild symptoms (13t) in barley (Hordeum vulgare) cultivar Plaisant in a growth chamber at 25 days after inoculation. These isolates had different effects on a range of barley genotypes. Cultivar Vixen, which contains the Yd2 resistance gene, and 80-81 BQCB10 were not affected by any isolate. Five other genotypes were significantly affected by at, least one of the isolates. Line Ea52 (which is a mutant of the Japanese cultivar Chikurine Ibaraki) was more susceptible to BYDV-PAV than Chikurin Ibaraki 1. No serological differences were detected between the four isolates using monoclonal or polyclonal antibodies. Virus antigen concentration, estimated by enzyme-linked immunosorbent assay (ELISA), was correlated with the decrease in the shoot fresh weight for all isolates and all genotypes except for Vixen and 80-81BQCB10. In field tests, the severity of symptoms induced by the BYDV-PAV isolates was in accordance with that estimated in the growth chamber. However isolate 2t was more severe on cultivar Vixen and overcame the partial resistance of Chikurin Ibaraki 1 to the three other isolates. The results show that virus antigen concentration not only contributes to characterizing the resistance levels of barley genotypes but also the severity of BYDV-PAV isolates.  相似文献

Small and intermediate vascular bundles and contiguous tissues of the leaf blade ofThemeda triandra var.imberbis (Retz.) A. Camus were examined with transmission and scanning electron microscopes to determine the distribution and frequency of plasmodesmata between various cell types. Plasmodesmata are most abundant at the mesophyll/bundle-sheath cell and bundle-sheath/vascular parenchyma cell interfaces, and their numbers decrease with increasing proximity to both thick- and thin-walled sieve tubes. Among cells of the vascular bundles, the greatest frequency of plasmodesmata occurs between vascular parenchyma cells, followed by that of plasmodesmata between vascular parenchyma cells and companion cells, and then by the pore-plasmodesmata connections between companion cells and thin-walled sieve tubes (sieve tube-companion cell complexes). The sieve tube-companion cell complexes of theT. triandra leaf are not isolated symplastically from the rest of the leaf and, in this respect, differ from their counterparts in theZea mays leaf. In this brief review an attempt has been made to discuss some of the important features of the vascular anatomy of angiospermous leaves, especially those related to assimilate transport. Accordingly, emphasis has been placed on the small or minor veins, which are closely related spatially to the mesophyll, and which play a major role in the uptake and subsequent transport of photosynthates from the leaf. The small veins are enclosed by bundle sheaths that intervene between the mesophyll and vascular tissues and greatly increase the area for contact with mesophyll cells. In the minor veins of dicotyledonous leaves, parenchymatic cells having organelle-rich protoplasts and numerous cytoplasmic connections with sieve elements dominate quantitatively. It is these so-called intermediary cells that apparently are directly involved with the loading of assimilates into the sieve elements. In the maize leaf the small and intermediate bundles have two types of sieve tubes, relatively thin-walled ones that have numerous cytoplasmic connections with companion cells, and thick-walled ones that lack companion cells but have numerous connections with vascular parenchyma cells. The companion cell-sieve tube complexes are virtually isolated symplastically from other cells of the vascular bundle and from the bundle sheath. Thick-walled sieve tubes similar to those in the maize leaf have been recorded in the leaves of other grasses.  相似文献

We investigated the phloem loading pathway in barley, by determining plasmodesmatal frequencies at the electron microscope level for both intermediate and small blade bundles of mature barley leaves. Lucifer yellow was injected intercellularly into bundle sheath, vascular parenchyma, and thin-walled sieve tubes. Passage of this symplastically transported dye was monitored with an epifluorescence microscope under blue light. Low plasmodesmatal frequencies endarch to the bundle sheath cells are relatively low for most interfaces terminating at the thin- and thick-walled sieve tubes within this C3 species. Lack of connections between vascular parenchyma and sieve tubes, and low frequencies (0.5% plasmodesmata per μm cell wall interface) of connections between vascular parenchyma and companion cells, as well as the very low frequency of pore-plasmodesmatal connections between companion cells and sieve tubes in small bundles (0.2% plasmodesmata per μm cell wall interface), suggest that the companion cell-sieve tube complex is symplastically isolated from other vascular parenchyma cells in small bundles. The degree of cellular connectivity and the potential isolation of the companion cell-sieve tube complex was determined electrophysiologically, using an electrometer coupled to microcapillary electrodes. The less negative cell potential (average –52 mV) from mesophyll to the vascular parenchyma cells contrasted sharply with the more negative potential (–122.5 mV) recorded for the companion cell-thin-walled sieve tube complex. Although intercellular injection of lucifer yellow clearly demonstrated rapid (0.75 μm s-1) longitudinal and radial transport in the bundle sheath-vascular parenchyma complex, as well as from the bundle sheath through transverse veins to adjacent longitudinal veins, we were neither able to detect nor present unequivocal evidence in support of the symplastic connectivity of the sieve tubes to the vascular parenchyma. Injection of the companion cell-sieve tube complex, did not demonstrate backward connectivity to the bundle sheath. We conclude that the low plasmodesmatal frequencies, coupled with a two-domain electropotential zonation configuration, and the negative transport experiments using lucifer yellow, precludes symplastic phloem loading in barley leaves.  相似文献

Small and intermediate (longitudinal) vascular bundles of the Zea mays leaf are surrounded by chlorenchymatous bundle sheaths and consist of one or two vessels, variable numbers of vascular parenchyma cells, and two or more sieve tubes some of which are associated with companion cells. Sieve tubes not associated with companion cells have relatively thick walls and commonly are in direct contact with the vessels. The thick-walled sieve tubes have abundant cytoplasmic connections with contiguous vascular parenchyma cells; in contrast, connections between vascular parenchyma cells and thin-walled sieve tubes are rare. Connections are abundant, however, between the thin-walled sieve tubes and their companion cells; the latter have few connections with the vascular parenchyma cells. Plasmolytic studies on leaves of plants taken directly from lighted growth chambers gave osmotic potential values of about-18 bars for the companion cells and thin-walled sieve tubes (the companion cell-sieve tube complexes) and about-11 bars for the vascular parenchyma cells. Judging from the distribution of connections between various cell types of the vascular bundles and from the osmotic potential values of those cell types, it appears that sugar is actively accumulated from the apoplast by the companion cell-sieve tube complex, probably across the plasmalemma of the companion cell. The thick-walled sieve tubes, with their close spatial association with the vessels and possession of plasmalemma tubules, may play a role in retrieval of solutes entering the leaf apoplast in the transpiration stream. The transverse veins have chlorenchymatous bundle sheaths and commonly contain a single vessel and sieve tube. Parenchymatic elements may or may not be present. Like the thick-walled sieve tubes of the longitudinal bundles, the sieve tubes of the transverse veins have plasmalemma tubules, indicating that they too may play a role in retrieval of solutes entering the leaf apoplast in the transpiration stream.  相似文献

Winged individuals of Rhopalosiphum padi and Sitobion avenae transmitted the PAV-like and MAV-like isolates of barley yellow dwarf virus respectively. Success of transmission after inoculation access periods of 2, 6, 12, 24, 48 and 72 h were examined and survival, reproduction and movement of the aphid vectors were recorded at these times. The experiment was done at four different temperatures: 6oC, 12oC, 18oC and 23oC. For both isolates the inoculation efficacy did not increase after a 24 h inoculation access period and there was no difference in inoculation efficiency at the three highest temperatures, that at 6oC being significantly lower than at 12oC to 23oC. The results suggest that autumn temperature is a critical factor for BYDV epidemiology in Britain with a small increase in autumn temperature leading to greatly increased infection rates.  相似文献

This review deals with aspects of the cellular and molecular biology of the sieve element/companion cell complex, the functional unit of sieve tubes in angiosperms. It includes the following issues: (a) evolution of the sieve elements; (b) the specific structural outfit of sieve elements and its functional significance; (c) modes of cellular and molecular interaction between sieve element and companion cell; (d) plasmodesmal trafficking between sieve element and companion cell as the basis for macromolecular long‐distance signalling in the phloem; (e) diversity of sieve element/companion cell complexes in the respective phloem zones (collection phloem, transport phloem, release phloem); (f) deployment of carriers, pumps and channels on the plasma membrane of sieve element/companion cell complexes in various phloem zones; and (g) implications of the molecular‐cellular equipment of sieve element/companion cells complexes for mass flow of water and solutes in a whole‐plant frame.  相似文献

Interactions between barley yellow dwarf luteovirus (BYDV) and the aphid parasitoid, Aphidius ervi Haliday (Hymenoptera: Aphidiidae), were investigated while sharing the vector/host, Sitobion avenae (F.) (Homoptera: Aphididae). Aphids, which were parasitized during their second larval stadium, had access to virus-infected plants before, immediately after, or several days after parasitoid attack. The larval development of A. ervi in S. avenae was significantly delayed when virus acquisition took place before or shortly after the parasitoid had hatched, but not when the parasitoid was at the second larval stage during virus acquisition. Similarly, the presence of BYDV led to a significantly higher aphid mortality when they acquired virus up to and including the time that A. ervi was at the first larval stage. Adult female parasitoids deposited fewer eggs in viruliferous aphids. Virus transmission was not reduced by parasitization, and in some experiments aphids which were subjected to parasitoid attack transmitted BYDV more efficiently than unattacked insects.  相似文献

The Yd2 gene for "resistance" to barley yellow dwarf virus (BYDV) has been widely used in barley ( Hordeum vulgare ). We have tested Australian isolates of BYDV of varying severity against barley genotypes with and without the Yd2 gene and report here a positive relationship between symptoms and virus levels determined by ELISA. Cultivar Shannon is the result of backcrossing the resistant line CI 3208 to cultivar Proctor, a susceptible line. It appears to be intermediate in reaction to BYDV between Proctor and CI 3208, although it carries the major gene, Yd2. Unlike the whole plant studies, no significant differences were observed with regard to the ability of protoplasts derived from these various genotypes to support BYDV replication. It is therefore demonstrated for the first time that the Yd2 gene is not among the small number of resistance genes which are effective against virus replication in isolated protoplasts.  相似文献

Large, intermediate, and small bundles and contiguous tissues of the leaf blade of Hordeum tvulgare L. ‘Morex’ were examined with the transmission electron microscope to determine their cellular composition and the distribution and frequency of the plasmodesmata between the various cell combinations. Plasmodesmata are abundant at the mesophyll/parenchymatous bundle sheath, parenchymatous bundle sheath/mestome sheath, and mestome sheath/vascular parenchyma cell interfaces. Within the bundles, plasmodesmata are also abundant between vascular parenchyma cells, which occupy most of the interface between the sieve tube-companion cell complexes and the mestome sheath. Other vascular parenchyma cells commonly separate the thick-walled sieve tubes from the sieve tube-companion cell complexes. Plasmodesmatal frequencies between all remaining cell combinations of the vascular tissues are very low, even between the thin-walled sieve tubes and their associated companion cells. Both the sieve tube-companion cell complexes and the thick-walled sieve tubes, which lack companion cells, are virtually isolated symplastically from the rest of the leaf. Data on plamodesmatal frequency between protophloem sieve tubes and other cell types in intermediate and large bundles indicate that they (and their associated companion cells, when present) are also isolated symplastically from the rest of the leaf. Collectively, these data indicate that both phloem loading and unloading in the barley leaf involve apoplastic mechanisms.  相似文献
