An apo form of cytochrome cd1 (nitrite reductase) of Paracoccus denitrificans has been detected immunologically in the periplasm of a mutant that lacks all c-type cytochromes. A method for the preparation of apo-nitrite reductase (lacking both c- and d-type haem) from the holoenzyme of wild-type cells has been developed. The apoprotein synthesized by the mutant is indistinguishable from the chemically prepared apoprotein in respect of: (i) subunit molecular weight; (ii) formation of a homodimer; (iii) properties on anion exchange chromatography. The holoenzyme has similar properties in respect of (i) and (ii) but behaves differently during anion exchange. A suggested mode of assembly of cytochrome cd1 is translocation into the periplasm of a precursor polypeptide, maturation by a signal peptidase to give an apoprotein identical to that prepared chemically from the holoenzyme, followed by insertion of c-type and d-type haem in an as yet unknown order. 相似文献

Paracoccus denitrificans cytochrome c550 is expressed as a periplasmic holo-protein in Escherichia coli; amino acid substitutions of cysteine residues in the haembinding motif (Cys-X-X-Cys-His), either together or singly, prevented covalent attachment of haem but not polypeptide translocation into the periplasm. When the three alanine residues at positions -3 to -1 in the native signal-cleavage site were deleted, or alanine at -1 was changed to glutamine, signal cleavage was at alternative sites (after only ten residues in the latter case), but haem attachment still occurred. When the same three alanines were changed to Asp-Glu-Asp, a membrane-associated apo product that had retained the complete signal sequence was detected. These and other results presented here indicate that (i) haem attachment is not required for the apo-cytochrome c550 export to the periplasm; (ii) haem cannot attach to apocytochrome c550 when attached to the cytoplasmic membrane, suggesting that signal-sequence cleavage precedes periplasmic haem attachment, which can occur at as few as six residues from the mature N-terminus; and (iii) two cysteines are required for haem attachment, possibly because a disulphide bond is an intermediate. The gene for Saccharomyces cerevisiae mitochondrial iso-1-cytochrome c was expressed as a holo-protein in E. coli when fused with the signal sequence plus the first 10 residues of the mature cytochrome c550, indicating that the E. coli cellular apparatus for the c-type cytochrome biogenesis has a broad substrate specificity. 相似文献

Abstract The gene encoding the Rhodopseudomonas viridis cytochrome c 2 (cycA) has been introduced on a broad host range vector into Paracoccus denitrificans , leading to high-level expression of the holo-cytochrome with the heme moiety covalently attached to the apoprotein. The cytochrome was demonstrated to reside in the periplasmic space of the host cell. In contrast to R. viridis , aerobic rather than anaerobic growth conditions led to higher production levels of the holo-cytochrome in P. denitrificans . This heterologous expression system provides a suitable genetic background for the functional expression and mutagenesis of polypeptides involved in bacterial photosynthesis, offering the possibility of detailed structural and functional investigation. 相似文献

Aim Numerous studies have examined the climatic factors that influence the abundance of C4 species within the grass flora (C4 relative species richness) in various regions throughout the world, but very few have examined the relative abundance of C4 vs. C3 grasses (C4 relative abundance). We sought to determine the climatic factors that influence C4 relative abundance throughout Australia. Location Australia (including Tasmania). Methods We measured C4 relative abundance at 168 locations and measured δ13C (the abundance of 13C relative to 12C) of the bone collagen of 779 kangaroos collected throughout Australia, as bone collagen δ13C was assumed to be proportional to the relative abundance of C4 grasses in the diet. Results Both C4 relative abundance and kangaroo bone collagen δ13C were found to have a strong positive relationship with seasonal water availability, i.e. the distribution of rainfall in the C4 vs. C3 growing seasons (76% and 69% of deviance explained, respectively). There was clear evidence that seasonal water availability was a better predictor of both C4 relative abundance and bone collagen δ13C than other climate variables such as mean annual temperature and January daily minimum temperature. However, seasonal water availability appeared to be a relatively poor predictor of C4 relative species richness, which was most closely related to January daily minimum temperature (90% of deviance explained). Main conclusions Our results highlight the relatively poor relationship between C4 relative abundance and C4 relative species richness, and suggest that these two variables may be related to different climatic factors. They also suggest that caution is required when using C4 relative species richness to infer the relative biomass and productivity of C4 grasses on a global scale. 相似文献

Burgos cheese was manufactured from pasteurized ewes milk inoculated with Staphylococcus aureus strains FRI 137 and FRI 361, at levels of ca 103 and 105 cfu/ml and stored at 4°, 10° and 15°C and at room temperature (10°-15°C). Populations of Staph. aureus and mesophilic aerobes, pH, and production of thermonuclease and enterotoxins C1 and C2 were investigated. Aerobic counts increased during cheese-making and storage. With both test strains, important growth was observed only during the storage period, the larger levels corresponding to the higher temperatures. Although Staph. aureus strains attained populations of over 108 cfu/g, no enterotoxin was detected. Strain FRI 361 reached 107 cfu/g without production of a detectable amount of thermonuclease. With strain FRI 137, the minimal population associated with enzyme activity was influenced by the inoculum size. Staphylococcus aureus counts are better indicators of staphylococcal growth in Burgos cheese than the thermonuclease test. 相似文献

In order to establish a model system for comparative studies of C3 and C3–C4 intermediate photosynthesis, the development of efficient transformation systems and the monitoring of transgene behaviour and stability were carried out in two closely related Moricandia species (Brassicaceae): the C3–C4 photosynthetic intermediate species M. arvensis and the C3 species M. moricandioides. In this study the green fluorescent protein (gfp) reporter gene was used as a vital marker gene while the use of the β‐glucuronidase (gusA) gene was based on the highly sensitive detection of its activity. For Agrobacterium‐mediated transformation of leaf explants, a cauliflower mosaic virus 35S promoter‐driven, modified version of gfp, the mgfp5‐ER gene and the gusA gene, respectively, were introduced into the new dual binary transformation vector system pGreen/pSoup (Hellens et al. 2000, Plant Mol Bio 42: 819–832). GFP5 produced bright‐green fluorescence in transformed tissues that was distinctly detected 5–12 days following transformation in developing calli of the two species. Visual screening, combined with antibiotic selection, enabled early and easy identification of transformation events and contributed to improvements in the transformation strategies. Transgene integration studies demonstrated that mgfp5‐ER was inserted with low copy number in the M. arvensis plant lines and the transgene was transmitted in a Mendelian fashion to T1 and T2 progenies. GFP5 expression levels in a population of 100 independent primary transformed M. arvensis plant lines (T0) showed great variation between transformation events (coefficient of variation of 108%). The mgfp5‐ER or gusA reporter genes were expressed in 90–95% of the kanamycin‐resistant M. arvensis plant lines and in up to 98% of the independent M. moricandioides plant lines. 相似文献

Bradyrhizobium japonicum cytochrome c 550 , encoded by cycA , has been previously suggested to play a role in denitrification, the respiratory reduction of nitrate to dinitrogen. However, the exact role of this cytochrome in the denitrification process is unknown. This study shows that cytochrome c 550 is involved in electron transfer to the copper-containing nitrite reductase of B. japonicum , as revealed by the inability of a cycA mutant strain to consume nitrite and, consequently, to grow under denitrifying conditions with nitrite as the electron acceptor. Mutation of cycA had no apparent effect on methylviologen-dependent nitrite reductase activity. However, succinate-dependent nitrite reduction was largely inhibited, suggesting that c 550 is the in vivo electron donor to copper-containing nitrite reductase. In addition, this study demonstrates that a cytochrome c 550 mutation has a negative effect on expression of the periplasmic nitrate reductase. This phenotype can be rescued by extending the growth period of the cells. A model is proposed whereby a mutation in cycA reduces expression of the cbb 3 -type oxidase, affecting oxygen consumption rate by the cells and consequently preventing maximal expression of the periplasmic nitrate reductase during the first days of the growth period. 相似文献

Variable factors affecting the enzymatic isolation of mesophyll protoplasts from Triticum aestivum (wheat), a C3 gras, and mesophyll protoplasts and bundle sheath strands from Digitaria sanguinalis (crabgrass), a C4 grass, have been examined with respect to yields and also photosynthetic capacity after isolation. Preparations with high yields and high photosynthetic capacity were obtained when small transverse leaf segments were incubated in enzyme medium in the light at 30°C, without mechanical shaking and without prior vacuum infiltration. Best results were obtained with an enzyme medium that included 0.5 M sorbitol, 1 mM MgCl2, 1 mM KH2PO4, 2% cellulase and 0.1% pectinase at pH 5.5. In gerneral, leaf age and leaf segment size were important factors, with highest yields and photosynthetic capacities obtained from young leaves cut into segments less than 0.8 mm. To facilitate the cutting of such small segments, a mechanical leaf cutter is described that uniformly (± 0.05 mm) cuts leaf tissue into transverse segments of variable size (0.4–2 mm). Isolations that required more than roughly 4 h gave poor yields with reduced photosynthetic capacity; however, using the optimum conditions described, functional preparations could be roughly 2 h. High rates of light dependent CO2 fixation by the C4 mesophyll protoplasts required the addition of pyruvate and low levels of oxalacetate, while isolated bundle sheath strands and C3 mesophyll protoplasts supported CO2 fixation without added substrates. Rates of CO2 fixation by isolated wheat protoplasts generally exceeded the reported rates of whole leaf photosynthesis. Wheat mesophyll protoplasts and crabgrass bundle sheath strands were stable when stored at 4°C while C4 mesophyll protoplasts were stable when stored at 25°C. 相似文献

In this study, the response of N2 fixation to elevated CO2 was measured in Scirpus olneyi, a C3 sedge, and Spartina patens, a C4 grass, using acetylene reduction assay and 15N2 gas feeding. Field plants grown in PVC tubes (25 cm long, 10 cm internal diameter) were used. Exposure to elevated CO2 significantly (P < 0·05) caused a 35% increase in nitrogenase activity and 73% increase in 15N incorporated by Scirpus olneyi. In Spartina patens, elevated CO2 (660 ± 1 μ mol mol − 1) increased nitrogenase activity and 15N incorporation by 13 and 23%, respectively. Estimates showed that the rate of N2 fixation in Scirpus olneyi under elevated CO2 was 611 ± 75 ng 15N fixed plant − 1 h − 1 compared with 367 ± 46 ng 15N fixed plant − 1 h − 1 in ambient CO2 plants. In Spartina patens, however, the rate of N2 fixation was 12·5 ± 1·1 versus 9·8 ± 1·3 ng 15N fixed plant − 1 h − 1 for elevated and ambient CO2, respectively. Heterotrophic non-symbiotic N2 fixation in plant-free marsh sediment also increased significantly (P < 0·05) with elevated CO2. The proportional increase in 15N2 fixation correlated with the relative stimulation of photosynthesis, in that N2 fixation was high in the C3 plant in which photosynthesis was also high, and lower in the C4 plant in which photosynthesis was relatively less stimulated by growth in elevated CO2. These results are consistent with the hypothesis that carbon fixation in C3 species, stimulated by rising CO2, is likely to provide additional carbon to endophytic and below-ground microbial processes. 相似文献

C4 photosynthesis had a mid‐Tertiary origin that was tied to declining atmospheric CO2, but C4‐dominated grasslands did not appear until late Tertiary. According to the ‘CO2‐threshold’ model, these C4 grasslands owe their origin to a further late Miocene decline in CO2 that gave C4 grasses a photosynthetic advantage. This model is most appropriate for explaining replacement of C3 grasslands by C4 grasslands, however, fossil evidence shows C4 grasslands replaced woodlands. An additional weakness in the threshold model is that recent estimates do not support a late Miocene drop in pCO2. We hypothesize that late Miocene climate changes created a fire climate capable of replacing woodlands with C4 grasslands. Critical elements were seasonality that sustained high biomass production part of year, followed by a dry season that greatly reduced fuel moisture, coupled with a monsoon climate that generated abundant lightning‐igniting fires. As woodlands became more open from burning, the high light conditions favoured C4 grasses over C3 grasses, and in a feedback process, the elevated productivity of C4 grasses increased highly combustible fuel loads that further increased fire activity. This hypothesis is supported by paleosol data that indicate the late Miocene expansion of C4 grasslands was the result of grassland expansion into more mesic environments and by charcoal sediment profiles that parallel the late Miocene expansion of C4 grasslands. Many contemporary C4 grasslands are fire dependent and are invaded by woodlands upon cessation of burning. Thus, we maintain that the factors driving the late Miocene expansion of C4 were the same as those responsible for maintenance of C4 grasslands today. 相似文献

In Paracoccusdenitrificans the aa3-type cytochrome c oxidase and the bb3-type quinol oxidase have previously been characterized in detail, both biochemically and genetically. Here we report on the isolation of a genomic locus that harbours the gene cluster ccoNOQP, and demonstrate that it encodes an alternative cbb3-type cytochrome c oxidase. This oxidase has previously been shown to be specifically induced at low oxygen tensions, suggesting that its expression is controlled by an oxygen-sensing mechanism. This view is corroborated by the observation that the ccoNOQP gene cluster is preceded by a gene that encodes an FNR homologue and that its promoter region contains an FNR-binding motif. Biochemical and physiological analyses of a set of oxidase mutants revealed that, at least under the conditions tested, cytochromes aa3, bb3. and cbb3 make up the complete set of terminal oxidases in P. denitrificans. Proton-translocation measurements of these oxidase mutants indicate that all three oxidase types have the capacity to pump protons. Abstract Chlorate-resistant mutants were generated by random insertion of the transposon Tn5 into genomic DNA of Pseudomonas stutzeri ZoBell strain and selected for loss of nitrate respiration (Nar phenotype). The mutants were differentiated by restriction-fragment analysis, by assaying for nitrate assimilation and for molybdenum co-factor activity, and by the amount of respiratory nitrate reductase. Two mutants, lacking both nitrate respiration and nitrate assimilation, over-produced an inactive nitrate reductase but synthesized in the presence of nitrate only a reduced amount of respiratory nitrite reductase (cytochrome cd 1 ). Expression of cytochrome cd 1 in these mutants was specifically induced by nitrate, suggesting a sensor system for this substrate. 相似文献

It has been suggested previously that Japanese larch ( Larix kaempferi ) exhibits characteristics of C4 photosynthesis. To further evaluate this suggestion, stable carbon isotope ratios were determined for leaf and bark tissue of Larix gmelini, L. kaempferi, L. laricina, L. Iyallii, L. occidentalis , and L. sibirica. All δ13 C values were more negative than –22‰. Short-term labeling with 14 CO2 showed that phosphoglyceric acid and other phosphorylated compounds were the first products of photosynthesis in L. sibirica. Both of these results strongly suggest that the initial fixation of atmospheric CO2 in these six Larix species is accomplished solely via the C3 photosynthetic pathway. 相似文献

Abstract. Germination modes of lower seeds of cocklebur (Xanthium pennsylvanicum Wallr.) under different water stresses, prepared with mannitol solution, were examined in relation to gaseous factors. As the concentration of mannitol increased, germination was increasingly inhibited at a mode which was drawn by two straight lines having different slopes and meeting at an angle. One is a sharp line occurring at the lower concentrations of mannitol; the other is a gentle line occurring at higher concentrations of mannitol. The former reflected the growth response of axial tissues to mild water stress, whereas the latter reflected the growth response of cotyledonary tissues to severe water stress. The germination potential of cocklebur seeds increased with increasing temperature. Thus, the seeds were more resistant to water stress at higher than al lower temperatures. This increased germination potential under water stress resulted from the greater growth potential of axial tissues, but not cotyledonary tissues, at higher temperature. Increased O2 levels improved both the reduced axial and cotyledonary growth under water stress. Carbon dioxide predominantly enhanced axial growth under water stress, whereas C2H4 exclusively enhanced cotyledonary growth. Thus, these gases were effective in potentiating germination under water stress. When combined with each other, these gases caused more pronounced growth of the axial and cotyledonary tissues, leading to germination under more severe water stresses. Maximal axial and cotyledonary growth under water stress occurred in the simultaneous presence of CO2, C2H4 and O2, which allowed the germination at higher mannitol concentrations above 0.6 kmol m?3 From these results, it was suggested that cocklebur seeds would override water stress by depending upon both the Corresponding axial growth and the C2H4-responding cotyledonary growth. 相似文献

The vitamin B12 -producing capacity of micro-organisms isolated from baboon faeces and gastric contents was measured using Lactobacillus leichmannii . The animals were fed either a diet deficient in or supplemented with vitamin B12 (controls). Samples of gastric and small intestinal contents, obtained at laparotomy from two young vitamin B12 -deprived baboons, contained varying quantities of vitamin B12 . Many of the organisms isolated from these aspirates produced vitamin B12 in vitro . The highest levels of vitamin B12 were produced by anaerobic organisms. Gastric juice samples from vitamin B12 deprived and control baboons contained similar types of organisms with like vitamin B12 -producing capacity. The vitamin B12 content, pH and total bacterial counts of gastric juice samples aspirated after a 6 h fast from vitamin B12 deprived baboons were not significantly different from those of the control animals. The pH values of gastric juice samples aspirated 18 h after feeding, however, were significantly lower than those of 6 h fasting samples in both groups. The mean vitamin B12 levels in the total volumes of gastric juice aspirated after each fasting period were similar. The possible involvement of the gastrointestinal flora in the vitamin B12 status of the baboon is discussed. 相似文献

Abstract. The photosynthetic responses to temperature in C3, C3-C4 intermediate, and C4 species in the genus Flaveria were examined in an effort to identify whether the reduced photorespiration rates characteristic of C3-C4 intermediate photosynthesis result in adaptive advantages at warm leaf temperatures. Reduced photorespiration rates were reflected in lower CO2 compensation points at all temperatures examined in the C3-C4 intermediate, Flaveria floridana, compared to the C3 species, F. cronquistii. The C3-C4 intermediate, F. floridana, exhibited a C3-like photosynthetic temperature dependence, except for relatively higher photosynthesis rates at warm leaf temperatures compared to the C3 species, F. cronquistii. Using models of C3 and C3-C4 intermediate photosynthesis, it was predicted that by recycling photorespired CO2 in bundle-sheath cells, as occurs in many C3-C4 intermediates, photosynthesis rates at 35°C could be increased by 28%, compared to a C3 plant. Without recycling photorespired CO2, it was calculated that in order to improve photosynthesis rates at 35°C by this amount in C3 plants, (1) intercellular CO2 partial pressures would have to be increased from 25 to 31 Pa, resulting in a 57% decrease in water-use efficiency, or (2) the activity of RuBP carboxylase would have to be increased by 32%, resulting in a 22% decrease in nitrogen-use efficiency. In addition to the recycling of photorespired CO2, leaves of F. floridana appear to effectively concentrate CO2 at the active site of RuBP carboxylase, increasing the apparent carboxylation efficiency per unit of in vitro RuBP carboxylase activity. The CO2-concentrating activity also appears to reduce the temperature sensitivity of the carboxylation efficiency in F. floridana compared to F. cronquistii. The carboxylation efficiency per unit of RuBP carboxylase activity decreased by only 38% in F. floridana, compared to 50% in F. cronquistii, as leaf temperature was raised from 25 to 35°C. The C3-C4 intermediate, F. ramosissima, exhibited a photosynthetic temperature temperature response curve that was more similar to the C4 species, F. trinervia, than the C3 species, F. cronquistii. The C4-like pattern is probably related to the advanced nature of C4-like biochemical traits in F. ramosissima The results demonstrate that reductions in photorespiration rates in C3-C4 intermediate plants create photosynthetic advantages at warm leaf temperatures that in C3 plants could only be achieved through substantial costs to water-use efficiency and/or nitrogen-use efficiency. 相似文献
