Flaxseed gum solutions were extracted and dried by different methods: ethanol precipitation, freeze drying, 105 °C oven drying, 80 °C oven drying, spray drying, and vacuum drying. The effects of these drying methods on the rheological properties of flaxseed gum were investigated in present study. Ethanol precipitation increased the apparent viscosity of flaxseed gum solution, while all the other methods decreased the apparent viscosity. Most of the drying methods slightly increased the activation energy, except ethanol precipitation. In frequency sweep test, all the drying methods reduced the G′ and G″ values. In creep–recovery tests, the data were modeled by Berger’s model. The E2 and η1 values were reduced by all of the drying methods in this study. Some relationships were found between the parameters in the Power Law model of the frequency sweep test and the parameters in Berger’s model.  相似文献

Background: Acute lymphoblastic leukemia (ALL) is a complex disease with genetic background. The genetic association studies (GAS) that investigated the association between ALL and the MTHFR C677T and A1298C gene variants have produced contradictory or inconclusive results. Materials and methods: In order to decrease the uncertainty of estimated genetic risk effects, a meticulous meta-analysis of published GAS related the variants in the MTFHR gene with susceptibility to ALL was conducted. The risk effects were estimated based on the odds ratio (OR) of the allele contrast and the generalized odds ratio (ORG). Cumulative and recursive cumulative meta-analyses were also performed.ResultsThe analysis showed marginal significant association for the C677T variant, overall [OR = 0.91 (0.82–1.00) and ORG = 0.89 (0.79–1.01)], and in Whites [OR = 0.88 (0.77–0.99) and ORG = 0.85 (0.73–0.99)]. The A1298C variant produced non-significant results. For both variants, the cumulative meta-analysis did not show a trend of association as evidence accumulates and the recursive cumulative meta-analysis indicated lack of sufficient evidence for denying or claiming an association. Conclusion: The current evidence is not sufficient to draw definite conclusions regarding the association of MTHFR variants and development of ALL.  相似文献

Background: Although polymorphisms in DNA mismatch repair (MMR) gene MSH2 have been associated with risks of many cancers, little is known about their etiology role in gastric cancer (GC) and the potential interacting role with lifestyle factors known to damage DNA. Methods: A population-based study was conducted in 3 counties (Jintan, Taixing and Huaian) of Jiangsu Province, the high-risk areas of GC in China. We investigated the association of polymorphisms IVS12?6T>C and IVS10+12G>A in MSH2 gene with the risk of GC and the potential gene–lifestyle interaction. Results: The risk of GC was found to be associated with the IVS12?6C allele (CC vs TT, OR = 2.34, 95% CI: 1.17–4.71) and IVS10+12A allele (GA or AA vs GG, OR = 1.55, 95% CI: 1.14–2.21; and GA vs GG, OR = 1.51, 95% CI: 1.04–2.17). Stratified analysis indicated that an increased risk of GC also was observed in: suspected familial subjects carrying the IVS12?6T>C (OR = 1.68, 95% CI: 1.27–2.66) or IVS10+12G>A (OR = 2.57, 95% CI: 1.53–4.10); or younger subjects carrying the IVS12?6T>C (OR = 2.15, 95% CI: 1.24–3.91) or IVS10+12G>A (OR = 2.23, 95% CI: 1.20–4.33); or male subjects carrying the IVS10+12G>A (OR = 1.64; 95% CI: 1.10–2.54). Furthermore, the combined IVS12?6CC and IVS10+12AA genotypes also significantly increased the risk of GC (OR = 2.12, 95% CI: 1.22–3.66). Statistically significant interactions were observed between: IVS10+12G>A and drinking, high pickled food or fried food intake (OR = 2.32; 95% CI: 1.43–3.78, OR = 2.55; 95% CI: 1.48–4.21 and OR = 2.88; 95% CI: 1.70–4.94, respectively); and IVS12?6T>C and high pickled food intake or fried food intake (OR = 2.65; 95% CI: 1.62–4.47 and OR = 2.48; 95% CI: 1.42–4.13, respectively). Conclusion: The IVS10+12G>A and IVS12?6T>C polymorphisms in MSH2 gene appear to be associated with risk of GC in this Chinese population. Risk for GC, stratified by related genotypes, was further modified by drinking, high pickled food or fried food intake. Larger prospective studies are needed to confirm these findings.  相似文献

A series of symmetric and asymmetric spermine (SPM) conjugates with all-trans-retinoic acid (ATRA), acitretin (ACI), (E)-3-(trioxsalen-4′-yl)acrylic acid (TRAA) and l-DOPA, amides of ACI, l-DOPA and TRAA with 1-aminobutane, benzylamine, dopamine and 1,12-diaminobutane as well as hybrid conjugates of O,O′-dimethylcaffeic acid (DMCA) with TRAA or N-fumaroyl-indole-3-carboxanilide (FICA) and 2-(2-aminoethoxy)ethanol were synthesized and their antioxidant properties were studied. The reducing activity (RA)% of the compounds were evaluated using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay and found to be in the range 0–92(20 min)%/96(60 min)% at 100 μM, the most powerful being the conjugates l-DOPA-SPM-l-DOPA (8, RA = 89%/96%) and l-DOPA-dopamine (13, RA = 92%/92%). Conjugate DMCA-NH(CH2CH2O)2-FICA (14) was the most powerful LOX inhibitor with IC50 33.5 μM, followed by the conjugates ACI-NHCH2Ph (10, IC50 40.5 μM), ACI-SPM-TRAA (7, IC50 41.5 μM), DMCA-NH(CH2CH2O)2-TRAA (15, IC50 65 μM), 13 (IC50 81.5 μM) and ACI-dopamine (11, IC50 87 μM). The most potent inhibitors of lipid peroxidation at 100 μM were the conjugates 15 (98%) and ACI-SPM-ACI (4, 97%) whereas all other compounds showed activities comparable or lower than trolox. The most interesting compounds, namely ATRA-SPM-ATRA (3), 4, 10, 11 and 15, as well as unconjugated compounds such as ATRA and dopamine, were studied for their anti-inflammatory activity in vivo on rat paw oedema induced by Carrageenan and found to exhibit, for doses of 0.01 mmol/mL of conjugates per Kg of rat body weight, weaker anti-inflammatory activities (3.6–40%) than indomethacin (47%) with conjugate 3 being the most potent (40%) in this series of compounds. The cytocompatibility of selected compounds was evaluated by the viability of RAMEC cells in the presence of different concentrations (0.5–50 μM) of the compounds. Conjugates 3 (IC50 2.6 μM) and 4 (IC50 4.7 μM) were more cytotoxic than the corresponding unconjugated retinoids ATRA (IC50 18.3 μM) and ACI (IC50 14.6 μM), whereas conjugate 15 (IC50 12.9 μM) was less cytotoxic than either DCSP (IC50 11.3 μM) or the tert-butyl ester of TRAA (IC50 2.9 μM).  相似文献

The present work involves the use of p-tert-butylcalix[4,6,8]arene carboxylic acid derivatives (tButyl[4,6,8]CH2COOH) for selective extraction of hemoglobin. All three calixarenes extracted hemoglobin into the organic phase, exhibiting extraction parameters higher than 0.90. Evaluation of the solvent accessible positively charged amino acid side chains of hemoglobin (PDB entry 1XZ2) revealed that there are 8 arginine, 44 lysine and 30 histidine residues on the protein surface which may be involved in the interactions with the calixarene molecules. The hemoglobin–tButyl[6]CH2COOH complex had pseudoperoxidase activity which catalysed the oxidation of syringaldazine in the presence of hydrogen peroxide in organic medium containing chloroform. The effect of pH, protein and substrate concentrations on biocatalysis was investigated using the hemoglobin–tButyl[6]CH2COOH complex. This complex exhibited the highest specific activity of 9.92 × 10?2 U mg protein?1 at an initial pH of 7.5 in organic medium. Apparent kinetic parameters (Vmax, Km, kcat and kcat/Km) for the pseudoperoxidase activity were determined in organic media for different pH values from a Michaelis–Menten plot. Furthermore, the stability of the protein–calixarene complex was investigated for different initial pH values and half-life (t1/2) values were obtained in the range of 1.96 and 2.64 days. Hemoglobin–calixarene complex present in organic medium was recovered in fresh aqueous solutions at alkaline pH, with a recovery of pseudoperoxidase activity of over 100%. These results strongly suggest that the use of calixarene derivatives is an alternative technique for protein extraction and solubilisation in organic media for biocatalysis.  相似文献

BackgroundThe cerebellum has never been mechanically characterised, despite its physiological importance in the control of motion and the clinical prevalence of cerebellar pathologies. The aim of this study was to measure the linear viscoelastic properties of the cerebellum in human volunteers using Magnetic Resonance Elastography (MRE).MethodsCoronal plane brain 3D MRE data was performed on eight healthy adult volunteers, at 80 Hz, to compare the properties of cerebral and cerebellar tissues. The linear viscoelastic storage (G′) and loss moduli (G) were estimated from the MRE wave images by solving the wave equation for propagation through an isotropic linear viscoelastic solid. Contributions of the compressional wave were removed via application of the curl-operator.ResultsThe storage modulus for the cerebellum was found to be significantly lower than that for the cerebrum, for both white and grey matter. Cerebrum: white matter (mean±SD) G′=2.41±0.23 kPa, grey matter G′=2.34±0.22 kPa; cerebellum: white matter, G′=1.85±0.18 kPa, grey matter G′=1.77±0.24 kPa; cerebrum vs cerebellum, p<0.001. For the viscous behaviour, there were differences in between regions and also by tissue type, with the white matter being more viscous than grey matter and the cerebrum more viscous than the cerebellum. Cerebrum: white matter G″=1.21±0.21 kPa, grey matter G″=1.11±0.03 kPa; cerebellum: white matter G″=1.1±0.23 kPa, grey matter G″=0.94±0.17 kPa.DiscussionThese data represent the first available data on the viscoelastic properties of cerebellum, which suggest that the cerebellum is less physically stiff than the cerebrum, possibly leading to a different response to mechanical loading. ObjectiveTo investigate nutritive microvascular function in young nonobese females with polycystic ovary syndrome (PCOS) and to correlate microvascular reactivity with sex steroids, inflammatory markers, and metabolic variables.MethodsFourteen nonobese females with PCOS (24.6 ± 2.7 years, body mass index [BMI] 23.7 ± 3.1 kg/ m2) and 13 age- and BMI-matched controls (22.8 ± 2.3 years, 22.5 ± 3.4kg/m2) underwent anthropometric, hormonal, and microvascular evaluations. The main outcome measures were capillary density, red blood cell velocity (RBCV) at resting and peak during postocclusive reactive hyperemia (RBCVmax), and time taken to reach RBCVmax (TRBCVmax).ResultsSubjects with PCOS had lower RBCV and higher TRBCVmax compared to controls, respectively (0.237 [0.220-0.324] vs. 0.362 [0.297-0.382] mm/s, F < .01) and (5 [5-6] vs. 4 [3-5] s, P < .05]. The free androgen index (FAI) and sex hormone-binding globulin (SHBG) level were different between groups. FAI correlated to RBCVmax (ρ = -0.49, P < .05) and to TRBCVmax (ρ = 0.41, P < .05). SHBG correlated with RBCVmax (ρ = 0.52, P < .01) while estradiol (E2) levels correlated with RBCV (ρ = 0.80, P < .001) and RBCVmax (ρ = 0.46, P < .05).ConclusionMicrovascular dysfunction characterized by reduced RBCVmaxand prolonged TRBCVmaxwas present in young, nonobese PCOS subjects. An accumulation of evidence suggests that the force–velocity relationship (FVR) of skeletal muscle plays a major role in limiting maximum human sprinting speed. However, most of the theories on this limiting role have been non-specific as to how the FVR limits speed. The FVR is characterized by three parameters that each have a different effect on its shape, and could thus limit sprinting speed in different ways: the maximum shortening velocity Vmax, the shape parameter AR, and the eccentric plateau Cecc. In this study, we sought to determine how specifically the FVR limits sprinting speed using forward dynamics simulations of human locomotion to examine the sensitivity of maximum speed to these three FVR parameters. Simulations were generated by optimizing the model's muscle excitations to maximize the average horizontal speed. The simulation's speed, temporal stride parameters, joint angles, GRF, and muscle activity in general compared well to data from human subjects sprinting at maximum effort. Simulations were then repeated with incremental and isolated adjustments in Vmax, AR, and Cecc across a physiological range. The range of speeds (5.22–6.91 m s?1) was most sensitive when Vmax was varied, but the fastest speed of 7.17 m s?1 was attained when AR was set to its maximum value, which corresponded to all muscles having entirely fast-twitch fibers. This result was explained by the muscle shortening velocities, which tended to be moderate and within the range where AR had its greatest effect on the shape of the FVR. Speed was less sensitive to adjustments in Cecc, with a range of 6.23–6.70 m s?1. Increases in speed with parameter changes were due to increases in stride length more so than stride frequency. The results suggest that the shape parameter AR, which primarily determines the amount of muscle force that can be produced at moderate shortening velocities, plays a major role in limiting the maximum sprinting speed. Analysis of muscle force sensitivity indicated support for previous theories on the time to generate support forces in stance (Weyand et al., 2000, Journal of Applied Physiology, 89, 1991–1999) and energy management of the leg in swing (Chapman &; Caldwell, 1983, Journal of Biomechanics 16, 79–83) as important factors in limiting maximum speed. However, the ability of the knee flexors to slow the rotational velocity of the leg in preparation for footstrike did not appear to play a major role in limiting speed.  相似文献

The effects of two anion/Cl? channel inhibitors, Zn2+ and niflumic acid (NA), on seedling photosynthetic and fluorescent parameters of two Glycine soja populations (salt-tolerant BB52; salt-sensitive N23227) and Glycine max cultivar (salt-tolerant Lee68) were studied and compared under salt stress. Treatments with Zn2+ and NA only (10, 20 μmol L?1) were also imposed for comparisons. Results showed that, there were non-toxic and non-nutritional effects of Zn2+ and NA treatments alone on seed germination and seedling growth of soybeans. Under 150 mmol L?1 NaCl for 6 d, leaf chlorophyll and carotenoid contents, net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci), transpiration rate (Tr), and the maximum photochemical efficiency of photosystem II (PS II) (Fv/Fm) except the stomatal limitation (Ls) significantly decreased in three kinds of soybean seedlings when compared with their control plants. The NaCl stress plus additional 20 μmol L?1 Zn showed an obvious enhancement of leaf chlorophyll and carotenoid contents, Pn, Gs, Ci and Tr, especially for the G. max cultivar Lee68, but the supplementation of 20 μmol L?1 NA showed the reverse effects.  相似文献

Alanine aminotransferase (ALT), which catalyzes the reversible conversion between l-glutamic acid (l-Glu) and l-alanine (l-Ala), is one of the most active aminotransferases in the clinical diagnosis of liver diseases. This work displays a microanalytical method for evaluating ALT enzyme kinetics using a microchip electrophoresis laser-induced fluorescence system. Four groups of amino acid (AA) mixtures, including the substrates of ALT (l-Glu and l-Ala), were effectively separated. Under the optimized conditions, the quantitative analysis of l-Glu and l-Ala was conducted and limits of detection (signal/noise = 3) for l-Glu and l-Ala were 4.0 × 10?7 and 2.0 × 10?7 M, respectively. In the reaction catalyzed by ALT, enzyme kinetic constants were determined for both the forward and reverse reactions by monitoring the concentration decrease of substrate AAs (l-Ala and l-Glu), and the Km and Vmax values were 10.12 mM and 0.48 mM/min for forward reaction and 3.22 mM and 0.22 mM/min for reverse reaction, respectively. Furthermore, the applicability of this assay was assessed by analysis of real serum samples. The results demonstrated that the proposed method could be used for kinetic study of ALT and shows great potential in the real application.  相似文献

The shell length, height, and width, live body weight, and edible tissue weight of Manila clam of 1, 2, and 3 years of age were measured, and their correlation coefficients were calculated. The shell morphological traits were used as independent variables, and live body weight or edible tissue weigh used as a dependent variable for calculating the path coefficients, correlation index and determination coefficients. The results showed that the correlation coefficients between each shell morphological trait and the live body weight or edible tissue weight were all highly significant (P < 0. 01). The shell height at 1-year old clams was highly correlated with the live body weight and edible tissue weight. The shell width of 2- to 3-year-old clams was strongly associated with the live body weight, while the shell length was closely linked to the edible tissue weight. The results of coefficients of determination for the morphological traits against weight traits agreed well with the results of path analysis. The correlation indices for all morphological traits against weight traits were approximately the same as determination coefficients regardless of clam age. The correlation indices (R2) of morphological traits against the live body weight of clams of all ages and edible tissue weight of 1-year-old clams were larger than 0.85, but R2 of morphological traits against the edible tissue weight of 2- and 3-year-old clams was smaller than 0.85, indicating that some other factors might be associated with the edible tissue weight of 2- and 3-year-old clams. In the present study, a novel, fast, sensitive and robust method to quantify budesonide in human plasma using 3-keto-desogestrel as the internal standard (IS) is described. The analyte and the IS were extracted from human plasma by liquid–liquid extraction (LLE) using ether. Extracted samples were analyzed by high performance liquid chromatography coupled to Atmospheric pressure photoionization tandem mass spectrometry (HPLC–APPI-MS/MS). Chromatography was performed isocratically on a C18, 5 μm analytical column. The temperature of the autosampler was kept at 6 °C and the run time was 4.00 min. A linear calibration curve over the range 7.5–1000 pg ml?1 was obtained and the lowest concentration quantified was 7.5 pg ml?1, demonstrating acceptable accuracy and precision. This analytical method was applied in a relative bioavailability study in order to compare a test budesonide 64 μg/dose nasal spray formulation vs. a reference 64 μg/dose nasal spray formulation (Budecort Aqua) in 48 volunteers of both sexes. The study was conducted in an open randomized two-period crossover design and with a one-week washout period. Plasma samples were obtained over a 14 h interval. Since the 90% CI for both Cmax, AUClast and AUC0-inf were within the 80–125% interval proposed by the Food and Drug Administration and ANVISA, it was concluded that budesonide 64 μg/dose nasal spray was bioequivalent to Budecort Acqua® 64 μg/dose nasal spray, according to both the rate and extent of absorption.  相似文献

Human serum albumin (HSA) displays esterase activity reflecting multiple irreversible chemical modifications rather than turnover. Here, kinetics of the pseudo-enzymatic hydrolysis of 4-nitrophenyl acetate (NphOAc) are reported. Under conditions where [HSA] ? 5×[NphOAc] and [NphOAc] ? 5×[HSA], the HSA-catalyzed hydrolysis of NphOAc is a first-order process for more than 95% of its course. From the dependence of the apparent rate constants kapp and kobs on [HSA] and [NphOAc], respectively, values of Ks, k+2, and k+2/Ks were determined. Values of Ks, k+2, and k+2/Ks obtained at [HSA] ? 5×[NphOAc] and [NphOAc] ? 5×[HSA] are in good agreement, the deacylation step being rate limiting in catalysis. The pH-dependence of k+2/Ks, k+2, and Ks reflects the acidic pKa shift of the Tyr411 catalytic residue from 9.0 ± 0.1 in the substrate-free HSA to 8.1 ± 0.1 in the HSA:NphOAc complex. Accordingly, diazepam inhibits competitively the HSA-catalyzed hydrolysis of NphOAc by binding to Tyr411.  相似文献

The pygmy loris (Nycticebus pygmaeus) is a small prosimian living in Vietnam, Laos, eastern Cambodia and the south part of China. In China it is only found in Pingbian, Hekou, Jinping, Luchun of Yunnan. As N. pygmaeus is seriously threatened by hunting, trade and habitat destruction, it is listed in Appendix II of CITES, and in 2006 the IUCN classified it as “vulnerable”. In order to understand the characteristics of energy metabolism and thermoregulation of N. pygmaeus, the resting metabolic rate (RMR) and body temperature (Tb) at different ambient temperature (Ta) of pygmy lorises, as well as body mass, energy intake, digestable energy intake, digestability and the thermal conductance were measured in captivity. The results obtained mainly are as follows: (1) Pygmy loris feed dry food averaged 12.90 ± 1.02 g/d. They could gain 214.87 ± 16.65 kJ/d from food intake, and earned 200.15 ± 16.36 kJ digestable energy intake per day with 90.13 ± 1.34% of the digestability. (2) The Tb at room temperatures was a little low (35.23 ± 0.16 °C) and varied with Ta from 25 °C to 35 °C. There was a positive relationship between Tb and Ta, which was described as: Tb = 27.22 + 0.34Ta (r = 0.880). (3) The resting metabolic rate (RMR) of the pygmy loris was 0.3844 ± 0.0162 mlO2/g/h, which was 51.91 ± 1.90% of the previous predicted rate by Kleiber (1961) [21]. (4) The average thermal conductance of the pygmy loris (N. pygmaeus) was 0.0449 ± 0.0031 mlO2/g/h/°C. These characteristics of energy metabolism and thermoregulation of N. pygmaeus in Yunnan Daweishan Nature Reserve might be considered as the adaptive characteristics to their environment in tropical semi-evergreen forests and secondary forests.  相似文献

The objective of this study was to develop a novel liposomal formulation, containing phosphatidylserine (PS), of buparvaquone (BPQ) and to evaluate its in vivo effectiveness in Leishmania (L.) infantum chagasi-infected hamsters. The activity of BPQ was evaluated against both the promastigote forms of different Leishmania species and the intracellular amastigotes of L. (L.) infantum chagasi. Buparvaquone was entrapped in PS-liposomes (BPQ–PS-LP), and the drug was quantified by ultra-high-performance liquid chromatography. The treatment was quantified by detecting the RNA of the living amastigotes in the spleen and the liver by real-time PCR. In vitro assays with L. (L.) infantum chagasi intracellular amastigotes were performed in peritoneal macrophages for the evaluation of the 50% inhibitory concentration (IC50). BPQ–PS-LP at 0.33 mg/kg/day for eight consecutive days reduced the number of amastigotes by 89.4% (P < 0.05) in the spleen and by 67.2% (P > 0.05) in the liver, compared to 84.3% (P < 0.05) and 99.7% (P < 0.05), respectively, following Glucantime® treatment at 50 mg/kg/day. Free BPQ at 20 mg/kg/day failed to treat the hamsters when compared to the untreated group. BPQ was significantly (P < 0.05) selective against L. (L.) infantum chagasi intracellular amastigotes, with an IC50 value of 1.5 μM; no in vitro mammalian cytotoxicity could be detected. Other cutaneous species were also susceptible to BPQ, with IC50 values in the range 1–4 μM. BPQ–PS-LP caused a significant reduction in the parasite burden at a 60-fold lower dose than did the free BPQ. These results show the potential of PS-liposome formulations for the successful targeted delivery of BPQ in visceral leishmaniasis.  相似文献

We have developed four 99mTc(CO)3-labeled lipophilic tracers as potential radiolabeling agents for cells based on a hexadecyl tail. 99mTc(CO)3-hexadecylamino-N,N′-diacetic acid (negatively charged), 99mTc(CO)3-hexadecylamino-N-α-picolyl-N′-acetic acid (uncharged), 99mTc(CO)3-N,N′-dipicolylhexadecylamine (positively charged), 99mTc(CO)3-N-hexadecylaminoethyl-N′-aminoethylamine (positively charged) were prepared in a radiolabeling yield: >90%. Preliminary cell uptake studies were performed in mixed blood cells with or without plasma and were compared with 99mTc-d,l-HMPAO and [18F]FDG. In plasma-free blood cells, maximum uptake (78%) was obtained for 99mTc(CO)3-N-hexadecylaminoethyl-N′-aminoethylamine after 60 min incubation (compared to 55% and 23% for 99mTc-d,l-HMPAO and [18F]FDG, respectively) while in plasma-rich medium, 99mTc(CO)3-N,N′-dipicolylhexadecylamine was best bound (54%, similar to the binding of 99mTc-d,l-HMPAO). Biodistribution in normal mice showed mainly hepatobiliary clearance of the agents and initial high lung uptake. The radiolabeled compounds showed good blood clearance with maximally 7.9% injected dose per gram at 60 min post injection. While the least lipophilic agent (99mTc(CO)3-N,N′-dipicolylhexadecylamine, log P = 1.3) showed the best cell uptake, there appears to be no direct correlation between lipophilicity and tracer uptake in mixed blood cells. In view of its comparable cell uptake to well known cell labeling agent 99mTc-d,l-HMPAO, 99mTc(CO)3-N,N′-dipicolylhexadecylamine merits further evaluation as a potential cell labeling agent.  相似文献

The validity of the Sorensen test as a measure for back muscle endurance is controversial due to a possible impact of hip extensor muscles. The aim of this study was to investigate the criterion validity of an alternative test (Ito test) compared to the Sorensen test. Both procedures were performed by 29 healthy subjects (11 women) for 5 s and until exhaustion (randomized order). EMG activity was measured from 3 lumbar back and 3 hip extensor muscles. Muscular involvement in test positions was calculated as percentage of maximal voluntary contraction (MVC). Muscle fatigue was determined by the normalized regression coefficient of the median frequencies of the EMG power spectrum (NMFslope). Prediction of holding time by NMFslope values was investigated using regression analysis. In the test positions, the hamstring muscles were activated to a higher MVC percentage in the Sorensen than in the Ito test, while the iliocostalis muscle was less activated. Similarly, the iliocostalis (p = 0.006) and the multifidi muscles (p = 0.03) significantly contributed to predict holding time in the Ito test, whereas the multifidi muscles (p = 0.001) and the semitendinosus muscle (p = 0.046) did so in the Sorensen test. The results of this study indicate that the Ito test might present a valuable alternative for testing back muscle endurance in LBP patients.  相似文献

Purpose. – To evaluate the value of 18F-fluoro-2-désoxyglucose (FDG) PET-CT and maximal standardized uptake value (SUVmax) of the primary tumor for lymph node staging in cervical cancer.Materials and methods. – This retrospective study involved a series of 18 consecutive patients who had benefited from PET-CT and MRI at initial staging for a stage IB or higher cervical carcinoma. The SUVmax of each primary tumor was measured retrospectively. All patients had been previously treated by radiochemotherapy. Lymph node status was obtained in 12 of 18 cases.Results. – The sensitivity and specificity for determining lymph node status was 80 and 86%, respectively, for PET-CT, and 80 and 71% for MRI. In 16.6% of cases, PET-CT revealed unknown sus-diaphragmatic lesions. SUVmax of the primary tumor was significantly higher in the N+ than in the N− group (15.6 ± 1.6 vs 8.5 ± 3.9, p < 0.01). The optimal threshold was determined to be 10.8 from ROC analysis.Conclusion. – When staging with FDG PET-CT, SUVmax of a primary cervical cancer seems to be a good predictor of lymph node status. This could lead to an intensification of treatment for patients whose SUVmax is higher than 10.8. A prospective study would allow to assess a potential benefit of treatment intensification for patients with SUVmax higher than 10.8.  相似文献

CO2 exchange and water relations of selected lichen species were investigated in the field and also in the laboratory, at a height of 3106 m above sea level in the Austrian Alps, during the short snowless summer period from middle of July to the end of August. In the course of the field investigations, clear summer days were quite rare. Altogether 14 diurnal courses of CO2 exchange were measured spanning a time of 255 h of measurements.The air temperatures measured close to the ground ranged between −0.7 and 17.1 °C and their daily fluctuation was lower than 10.7 °C. Fog was present for more than one-third of the measuring period and relative humidity (RH) exceeded 90% in almost half of the time. Temperature optimum of net photosynthesis (NP) of Xanthoria elegans and Brodoa atrofusca determined in the laboratory increased with increasing photosynthetic photon flux density (PPFD) from 1.5 to 11.3 °C and the maximal CO2 uptake was found to be at 10 °C. In the field the lichens were metabolically active at air temperatures between −0.7 and 12.8 °C. The light compensation points (LCP) of both lichen species ranged in the laboratory between 50 and 200 μmol m−2 s−1 PPFD (0–20 °C) and in the field between 22 and 56 μmol m−2 s−1 PPFD (3–8 °C). At 30 °C the NP of X. elegans surpassed the LCP, whereas B. atrofusca remained below the LCP. NP in X. elegans did not reach light saturation at 1500 μmol m−2 s−1 PPFD. NP in B. atrofusca reached light saturation at low temperatures (−5 to +5 °C). At higher temperatures light saturation was almost detectable. On sunny days the lichens in the field were metabolically active only for 3 h during the early morning. In this time they reached the maximal values or values close to their maximal CO2 uptake in situ. Under dry weather conditions the lichens dried out to a minimal water content (WC) of 5–12% which is below the moisture compensation point (MCP) of 34–25%. The optimal WC was between 90% and 120% dry weight (DW) in B. atrofusca and Umbilicaria cylindrica, in X. elegans between 140% and 180% DW. Species specific differences in water-holding capacity, desiccation intensity and in the compensation points of temperature, light and moisture are responsible for differences in metabolic activity. The lichens were active during less than half of the observation time. Total time of NP of X. elegans was 24% of the measuring period, for U. cylindrica 22% and for B. atrofusca 16%.  相似文献

Studies on disclosing characteristics and endangered mechanisms of Castanopsis kawakamii population ecology have already become an urgent task of protecting C. kawakamii population. The establishment of the standard life table is one of an important work study on C. kawakamii population ecology, and determining individual ages of the plant is necessary for studying age structures and population dynamics of C. kawakamii. There are three main methods of determining individual age of forest population: (1) by annual ring of tree, (2) by individual growth phase, and (3) by DBH and height of tree. However, the three methods have their shortcomings, such as low precision, worse serviceability, high difficulty for operation and so on. In this paper, a new method for determining plant individual ages more accurately is presented on the basis of the method aboUt “annual ring–time series”. Based on the stem analysis, the multidimensional time series model of diameter growth at breast height in C. kawakamii population was established by Utilizing the analYtical method of multidimensional time series: Yt = 1.325034Yt-1 ? 0.4711007Yt-2 ? 284.5648Ut + 569.4783Ut?1 ? 284.8745Ut?2, where Yt, Yt-1, Yt-2 represent diameter growth in C. kawakamii population at t, t ? 10 and t ? 20 years respectively, and Ut, Ut?1, Ut?2 represent individual age in C. kawakamii population at t, t ? 10 and t ? 20 years respectively, the model coefficient correlation is 0.9994. Based on this model CAR(2), the total increment of individual DBH in C. kawakamii population are simulated and regressively verified at different ages. The mean simulating precision of this model was 98.84%, the maximum relative error was 2.56%, bUt the next was 2.47% and the minimum relative error was 0.07%, showing that this model was suitable for estimating breast-height diameter of C. kawakamii plant. Using the multidimensional time series model, diameter growth of C. kawakamii population for longer time series was estimated in order to gain data for establishing the relationship model of individual age, diameter growth and to increase its precision in determining individual age is by tree ring analysis. A combination method of determining individual age of C. kawakamii population by integrating annual ring data with its diameter using multidimensional time series model, which can improve precision of individual ages in C. kawakami, was produced: A = 9.966671944 + 1.146011591D + 0.041059628D2 ? 0.000211907D3, where A and D represent individual age and diameter at breast height respectively in C. kawakamii population, the model coefficient correlation is 0.9998. The combination model, which shows that the regression relationship is significant and the model can exactly predict the individual age of population, is a valuable tool for determining individual ages in endangered plants.  相似文献
