The effect of intravenous cloprostenol treatment at the time of insemination on reproductive performance was consecutively evaluated in three different subpopulations of high producing lactating dairy cows: Study (1) early postpartum synchronized and fixed-time inseminated (about 50 days in milk) cows (n = 379: 187 control and 192 treated cows); Study (2) presumed high fertility cows first inseminated between 90 and 120 days postpartum (n = 248: 124 control and 124 treated cows); and Study (3) heat stressed repeat breeder cows (n = 183: 93 control and 90 treated cows). Data were analyzed using multiple regression methods. Study 1: Parity (primiparous versus multiparous), milk production, body condition score at AI, insemination season (cool versus warm period) and treatment were included in the analysis as potential factors affecting ovulation, double ovulation, return to estrus, and pregnancy to first AI and to second AI (first AI plus return AI) rates. Logistic regression analysis indicated that the final model for ovulation rate only included the interaction (P = 0.002) between insemination season and treatment. Cloprostenol treatment at insemination led to a 4.2-fold increase in the ovulation rate in cows inseminated during the warm period. There were no significant effects of treatment, parity, milk production, body score or the insemination season on the return to estrus rate. The only variables included in the final logistic model for double ovulation and pregnancy to first AI rates were treatment and season, respectively. Treatment led to a 2.6-fold increase (P = 0.001) in the double ovulation rate, whereas cows inseminated in the warm period were 2.1 times less likely (P = 0.007) to become pregnant at first AI compared to those inseminated in the cool season. The variables included in the final logistic model for the pregnancy rate to second AI were treatment and season. Cloprostenol given at AI increased the risk of pregnancy 1.9 times (P = 0.002), and cows inseminated during the warm season were two times less likely to become pregnant (P = 0.003). No significant interactions were found among these three dependent variables (double ovulation and pregnancy to first and to second AI rates). Study 2: Logistic regression analysis of all the dependent variables: return to estrus, and pregnancy to first and to second AI (first AI plus return to AI) rates indicated no significant effects of treatment, parity, days in milk, milk production or body score at AI. No significant interactions were found. Study 3: The final model for the pregnancy rate only included the interaction between parity (primiparous versus multiparous) and treatment. Days in milk, milk production and insemination number showed no significant effect on pregnancy rate. Cloprostenol treatment at insemination increased the pregnancy rate in primiparous repeat breeder cows (odds ratio: 3.6). The treatment group and parity showed significant (P < 0.0001) interaction. This interaction suggests that cloprostenol treatment of primiparous cows at insemination might enhance pregnancy yet have no effect in multiparous cows. Our findings indicate that cloprostenol administered at insemination promotes ovulation and double ovulation in lactating dairy cows. Cloprostenol treatment showed no benefit in cows with acceptable reproductive performance, suggesting that cloprostenol treatment at AI may only be useful in cows in which stress factors affect ovulation and in repeat breeder cows.  相似文献

Lactating Holstein dairy cows (n=1,533) were allocated to one of three treatment groups, with Group I (n=514) receiving 10 mug of a GnRH analogue (buserelin) at artificial insemination (AI) and Group II (n=503) receiving 10 mug of the same analogue at both the time of AI and at 12 days post AI. Herdmates in Group III (n=516) were inseminated on the same day and served as contemporary AI controls. The trial was conducted on five large dairy farms during the spring and summer months in Saudi Arabia. Pregnancy rates were determined by palpation per rectum between 33 and 50 days following AI. The first service pregnancy rate for the control cows (42.4%) was lower (P<0.05) than that for cows treated with the GnRH analogue at AI (48.8%) or for the combined treatment at AI and at Day 12 post AI (51.5%). No additive effect on the pregnancy rate was noted from the combined analog treatment. The overall increase in pregnancy rate from the analogue treatment at AI resulted from an 11% increase in pregnancy rate in first parity cows over that of contemporary controls (P<0.05) and a 14.7% increase in pregnancy for cows mated at 40 to 59 days post partum and treated with the analogue at AI over that of the corresponding controls (P<0.05). In many epidemiological studies time to event data are clustered and the physiological relationship between (time-dependent) covariates and the log hazard is often not linear as assumed in the Cox model. Introducing frailties in the Cox model can account for the clustering of the data and smoothing splines can be used to describe nonlinear relations. These two extensions of the Cox model are introduced jointly and it is shown how penalized partial likelihood techniques can be used to fit the extended model. We demonstrate the need for such a model to study the relation between the physiological covariates milk ureum and protein concentration and the log hazard of first insemination in dairy cows, with the farms as clusters.  相似文献

This study was carried out to investigate the utility of enzymeimmunoassay (EIA) - derived progesterone profiles in the investigation of postpartum reproductive abnormalities and the effect of their use on reproductive performance in Holstein cows. Whole milk samples, collected twice weekly from parturition until confirmation of pregnancy or removal from the herd, were assayed for progesterone (P4) concentration using a commercially available microtitre plate EIA. The sensitivity, specificity, precision and accuracy were satisfactory to distinguish cyclic changes of P4 reflecting ovarian activity. The P4 profiles indicated that retained fetal membranes and anestrus were associated with increased mean intervals to first ovulation and first estrus (P < 0.05). The first luteal phase of cows with pyometra was prolonged compared with that of normal herdmates (P < 0.05). Anestrus cows had an increased number of ovulations before first service and before pregnancy (3.9 and 5.9) in comparison with normal herdmates (2.1 and 3.5; P < 0.05). Calving to pregnancy interval was increased above normal (74.8) for pyometra (125.0) and anestrus (152.7) (P < 0.05). Pregnancy rate for anestrus cows (82%) was lower than for normal cows (100%) and more anestrous cows were culled (27%) than normal cows (5%) (P < 0.5). The P4 profiles indicated that the major problem in the herd studied, anestrus (32% incidence rate), was most likely due to the failure to observe estrus rather than acyclicity.  相似文献

The objective of this study was to determine whether rheological properties of the vaginal fluid at the time of insemination could be used as an indicator of potential fertility in dairy cows. Thixotropy and flow behavior were evaluated. Vaginal samples were collected from 347 cows at the time of insemination, within a 12- to 24-h interval after the onset of a natural estrus. Logistic regression analysis showed no significant effects of thixotropy measurements on the pregnancy rate, while flow behavior and the index of consistency (K) had a significant effect. The decrease in the consistency index (K) was related to the increase in the pregnancy rate (odds ratio = 0.99; P = 0.014), and the risk of nonpregnancy was higher (odds ratio = 0.46; P = 0.024) in cows in which vaginal fluid had a Newtonian behavior. Non-Newtonian behavior was recorded in 294 samples (85%) while 53 behaved as Newtonian fluids. For samples from all cows, the mean value of the consistency index was 310 +/- 28.2 milliPascals.second(n) (x +/- SEM) and ranged from 0 to 3881 milliPascals.second(n). In Newtonian samples, the consistency index mean was 2.6 +/- 0.6 milliPascals.second(n) and 367.6 +/- 30.4 milliPascals.second(n) for non-Newtonian samples. Of the 347 cows, 131 (38%) became pregnant. For flow behavior classification, 15 (28.3%) cows in which samples had Newtonian behavior and 116 (39.5%) cows with non-Newtonian samples became pregnant. Therefore, the highest probability of pregnancy was shown by cows in which samples had non-Newtonian behavior and the lowest consistency index values. Our data suggest that flow behavior measurements of the vaginal fluid at the time of insemination may be an indicator of potential fertility in cattle.  相似文献

A total of 585 repeat-breeder dairy cows was used to study the effect of GnRH treatment, either at or prior to insemination, on the pregnancy rate. The cows were divided into 6 treatment groups. Cows in Group 1 (n = 142) were observed in estrus, and 11 +/- 0.42 hours (mean +/- SEM) later they were given 100 ug, i.m. gonadotropin releasing hormone (GnRH) and were inseminated. Cows in Group 2 (n = 139) were observed in estrus and were inseminated 11.4 +/- 0.43 hours later. Cows in Group 3 (n = 33) were monitored for estrus with an activated heatmount detector but were not observed in estrus; they were inseminated 1.5 +/- 0.87 hours later and were given 100 ug, i.m. GnRH. Cows in Group 4 (n = 35) were not observed in estrus, but they did activate the heatmount detector and were inseminated 2.2 +/- 0.87 hours later. Cows in Group 5 (n = 107) were observed in estrus, given 100 ug, i.m. GnRH 2.0 +/- 0.40 hours later, and were inseminated 9 +/- 0.60 hours after GnRH treatment. Cows in Group 6 (n = 129) were observed in estrus and were inseminated 10 +/- 0.50 hours later. Pregnancy rates were analyzed by Chi-square. Interactions between pregnancy rate, treatment and time of insemination were evaluated using ANOVA and LSM (P < 0.05). There was no effect on pregnancy rate when GnRH was given at or prior to insemination. Cows inseminated on the basis of observed estrus had a higher pregnancy rate (P < 0.05) than cows inseminated on the observation of an activated heatmount detector. From the results of this study, it is concluded that treatment with GnRH at or prior to insemination did not improve the pregnancy rate of repeat-breeder dairy cows.  相似文献

Sixteen multiparous nonpregnant lactating Holstein cows (each weighing 662 ± 65 kg in 150.4 ±40 day of lactation) were confined to wooden metabolic cages with 12:12 h light:dark cycle during the experiment. The cows were divided into two sequences of eight cows each and exposed to electric and magnetic fields (EMF) in an exposure chamber. This chamber produced a vertical electric field of 10 kV/m and a uniform horizontal magnetic field of 30 μT at 60 Hz. One sequence was exposed for three estrous cycles of 24 to 27 days. During the first estrous cycle, the electric and magnetic fields were off; during the second estrous cycle, they were on; and during the third estrous cycle, they were off. The second sequence was also exposed for three 24 to 26 days estrous cycles, but the exposure to the fields was reversed (first estrous cycle, on; second estrous cycle, off; third estrous cycle, on). The length of each exposure period (21 to 27 days) varied according to the estrous cycle length. No differences were detected in plasma progesterone concentrations and area under the progesterone curve during estrous cycles between EMF nonexposed and exposed periods (2.28 ±0.17 and 2.25 ± 0.17; and 24.5 ± 1.9 vs. 26.4 ± 1.9 ng/ml, respectively). However, estrous cycle length, determined by the presence of a functional corpus luteum detected by concentrations of progesterone equal to or more than 1 ng/ml plasma, was shorter in nonexposed cows than when they were exposed to EMF (22.0 ± 0.9 vs. 25.3 ± 1.4 days). Six cows, 220 to 260 d pregnant, were used to study possible abortifacient effects of absorbed nitrite. Maternal and fetal bloodvessels were catheterized. Nitrite was infused intravenously (i.v.) for 30 minutes on Days 1, 3 and 5, commencing at least 5 d postcatheteration. The dose used was 7, 9.5 and 12 mg NO(2)(-)/kg bodyweight, respectively. Nitrite caused a dose-related (P < 0.05) conversion of maternal hemoglobin (Hb) into methemoglobin (MHb); a 30 to 50% decrease of mean arterial bloodpressure; an increase in heart rate, with dose-related recovery periods; and a decrease in partial oxygen tension (PO(2)) of maternal blood. Maternal partial carbon dioxide tension (PCO(2)), pH and electromyographic activity of the uterus were unchanged. Fetal changes included a small increase in MHb content, variable changes in heart rate (tachycardia and bradycardia), and decreases in fetal PO(2), with considerable differences between animals. All calves were born alive. Three cows calved early, 2 to 3 d after the highest nitrite dose. The hematological and cardiovascular data suggest that these three fetuses experienced a more serious hypoxemic stress than the other fetuses.  相似文献

Objectives were to evaluate progesterone concentrations after cows had initiated estrous cycles following calving and induction of estrous cycles in postpartum anovular high-producing Holstein dairy cows treated with controlled internal drug releasing (CIDR). In experiment 1 (EXP1), 62 cows that had initiated estrous cycles received a new CIDR (NCIDR) containing 1.38 g of progesterone or a 7-d used autoclaved CIDR (UCIDR) 48h after luteolysis for 7 d. Ovaries were examined by ultrasonography, and plasma analyzed for concentrations of progesterone. In experiment 2 (EXP2), 515 cows diagnosed as anestrus were randomly assigned to untreated control, NCIDR or UCIDR for 6d. Plasma was analyzed for concentration of progesterone 12 d after CIDR removal to determine ovulation. In EXP1, milk yield and body condition did not influence progesterone concentrations. Concentration of progesterone tended to increase faster (P=0.10) in cows receiving UCIDR than NCIDR, but both treatments reached a plateau at 90min. Cows receiving the NCIDR had greater (P=0.04) concentrations of progesterone during the 7-d treatment, but they were mostly subluteal (<1.0 ng/mL) after d 2. After removal, concentrations of progesterone were greater for NCIDR than UCIDR for the first 45 min, and were similar thereafter. Multiparous cows had lesser (P=0.004) concentrations than primiparous cows throughout the study. The pattern of ovarian follicular development was not affected by treatment. In EXP2, induction of onset of estrous cycles increased (P<0.01) with progesterone treatments, but was similar between NCIDR and UCIDR. Proportion of cows experiencing shorter than typical length estrous cycles after first AI tended to be greater (P=0.09) for control cows than those receiving the CIDR, and for cows remaining anestrous than those in which onset of estrous cycles was induced. Pregnancy per AI and pregnancy loss were similar among treatments. Cows that resumed estrous cyclicity prior to first AI had greater (P=0.01) pregnancy per AI. Treatment of high-producing Holstein cows that had previously initiated onset of estrous cycles with CIDR resulted in subluteal concentrations of progesterone, but in anestrous high-producing cows increased induction of estrous cycles with no effect on fertility at first insemination.  相似文献

Congeneric invasive species reduce genetic endemism of native species through hybridization and sometimes cause extinction through genetic or demographic swamping. In the range of Magnolia stellata, a native rare subtree species, Magnolia kobus, an invasive planted/escaped tree species, can also be found. In order to examine possible natural hybridization between the two species, a reciprocal cross-pollination experiment was conducted, and fruit set, seed set, female reproductive success in seed formation (FRS; fruit set × seed set) and seed weight were estimated. With M. stellata as the maternal tree, there were no significant differences in all measurements between intra- and interspecific crosses. With M. kobus as the maternal tree, there were also no significant differences in all measurements except fruit set. These results suggest that there are no reproductive barriers between the two species and planted/escaped M. kobus near the natural habitat of M. stellata presents a threat through hybridization.  相似文献

Sexual selection theory predicts that females face contrasting selection pressures when choosing the age of their mate. On the one hand, older males have demonstrated their ability to survive and they may be more experienced than younger males. At the same time, however, younger males are expected to have accumulated fewer deleterious mutations in their germline as compared to older males. These contrasting pressures on female preference may result in a preference for intermediate‐aged males. A preference for males of a particular age can only be expressed, however, if females are able to identify males of different ages. We have previously shown that male swamp sparrows display age‐related changes in vocal quality, such that males display sharp increases in vocal quality in early adulthood, followed by gradual senescent declines thereafter. We have also shown that territorial males discriminate these within‐individual differences, giving stronger aggressive responses to songs of peak‐aged males than to those of senescent males. Here, we use a copulation solicitation assay to test whether females also discriminate these within‐signaler markers of senescence in song. Contrary to our prediction, females did not show any evidence of discriminating between songs recorded from peak‐aged males as compared to songs from the same males following song senescence. We suggest that this difference in demonstrated discrimination between males and females may be the result of the two sexes attending to different song characteristics.  相似文献

Weekly reproductive health examinations were performed on 46 multiparous Holstein cows from 14 to 100 d post partum. Sixteen cows developed 19 nonsimultaneous ovarian cysts, with a mean day of first detection at 34.3 +/- 4.5 d post partum and a mean duration of 31.0 +/- 4.3 d after first detection. Coccygeal blood was collected three times weekly, and plasma progesterone concentrations were determined by radioimmunoassay. Cysts were diagnosed by palpation per rectum or by ultrasonography and classified as follicular or luteal cysts; the cows were not treated. Cows with a mean plasma progesterone concentration of < 1 ng/ml from the first day of detection (Day 1) of a cyst until Day 10 were classified as having a follicular cyst, and those with a mean plasma progesterone concentration of >/= 1 ng/ml from Day 1 to Day 10 were classified as having a luteal cyst. According to this classification, 58% of the cysts were follicular and 42% were luteal. There was an overall 47% agreement between classification by palpation and by ultrasonography on Day 1 with progesterone concentration during Days 1 to 10 after detection of the cyst. Detailed graphs of progesterone concentrations and area of largest follicles or cysts and corpora lutea demonstrate the variability of ovarian structures and progesterone profiles in cystic cows. Detection of a cyst at any one time accompanied by simultaneous measurement of progesterone can lead to different diagnoses of cyst type depending on the method of classification, the presence and age of luteinized tissue in the cyst and undetected corpora lutea.  相似文献

The objective of this study was to evaluate changes in ruminal microorganisms and fermentation parameters due to dietary supplementation of soybean and linseed oil alone or in combination. Four dietary treatments were tested in a Latin square designed experiment using four primiparous rumen-cannulated dairy cows. Treatments were control (C, 60 : 40 forage to concentrate) or C with 4% soybean oil (S), 4% linseed oil (L) or 2% soybean oil plus 2% linseed oil (SL) in a 4 × 4 Latin square with four periods of 21 days. Forage and concentrate mixtures were fed at 0800 and 2000 h daily. Ruminal fluid was collected every 2 h over a 12-h period on day 19 of each experimental period and pH was measured immediately. Samples were prepared for analyses of concentrations of volatile fatty acids (VFA) by GLC and ammonia. Counts of total and individual bacterial groups (cellulolytic, proteolytic, amylolytic bacteria and total viable bacteria) were performed using the roll-tube technique, and protozoa counts were measured via microscopy in ruminal fluid collected at 0, 4 and 8 h after the morning feeding. Content of ruminal digesta was obtained via the rumen cannula before the morning feeding and used immediately for DNA extraction and quantity of specific bacterial species was obtained using real- time PCR. Ruminal pH did not differ but total VFA (110 v. 105 mmol/l) were lower (P < 0.05) with oil supplementation compared with C. Concentration of ruminal NH3-N (4.4 v. 5.6 mmol/l) was greater (P < 0.05) due to oil compared with C. Compared with C, oil supplementation resulted in lower (P < 0.05) cellulolytic bacteria (3.25 × 108 v. 4.66 × 108 colony-forming units (CFU)/ml) and protozoa (9.04 × 104 v. 12.92 × 104 cell/ml) colony counts. Proteolytic bacteria (7.01 × 108 v. 6.08 × 108 CFU/ml) counts, however, were greater in response to oil compared with C (P < 0.05). Among oil treatments, the amount of Butyrivibrio fibrisolvens, Fibrobacter succinogenes and Ruminococcus flavefaciens in ruminal fluid was substantially lower (P < 0.05) when L was included. Compared to C, the amount of Ruminococcus albus decreased by an average of 40% regardless of oil level or type. Overall, the results indicate that some ruminal microorganisms, except proteolytic bacteria, are highly susceptible to dietary unsaturated fatty acids supplementation, particularly when linolenic acid rich oils were fed. Dietary oil effects on ruminal fermentation parameters seemed associated with the profile of ruminal microorganisms.  相似文献

This study was performed to clarify the levels of growth hormone (GH) and insulin (INS) secretions and the glucose response to INS during lactation in a representative beef breed in Japan, Japanese Black cows, and to compare them with their counterparts in a dairy breed, Holstein cows. Six Japanese Black and seven Holstein primiparous cows received a single intravenous injection of GH-releasing factor (GRF; 0.25 microg/kg), glucose (112.5 mg/kg), or INS (0.2 U/kg) from late pregnancy (2 weeks antepartum) to mid-lactation (6 months postpartum). Japanese Black cows had one-tenth of the total milk yield of Holstein cows during lactation, and significantly lower GRF-induced GH and higher glucose-induced INS secretions than Holstein cows at all stages. In Japanese Black cows, even with lactation, these secretions remained essentially unchanged, whilst Holstein cows showed higher GH and lower INS secretions after the onset of lactation as compared with cows in late pregnancy. Both breeds had similar glucose response to INS at the respective stages. These results suggest that, during lactation, Japanese Black cows may minimize the catabolic effects of GH and sustain the anabolic effects of INS, in contrast with Holstein cows, but have similar ability to inhibit INS-mediated glucose utilization in peripheral tissues to Holstein cows.  相似文献

Introduction

Physiological adaptations in the energy metabolism of dairy cows during the periparturient period are partly mediated by insulin resistance (IR), which may subsequently induce metabolic disorders postpartum. The molecular mechanisms underlying IR in dairy cows are largely unknown.

Objective

This study aimed to find a novel insight into the molecular mechanisms underlying IR in dairy cows during the periparturient period by analyzing the effects of prepartal overfeeding on the lipidomic profiles in the liver and adipose tissue (AT).

Methods

Sixteen cows were allocated to controlled-energy and high-energy feeding groups. Lipidomic profiling was conducted on liver and adipose tissue samples collected at 8 days prior to the predicted parturition, and 1 day (only AT) and 9 days after the actual parturition.

Results

Five ceramides (Cers) were identified to be significantly increased by prepartal overfeeding in AT in the analysis of the variance between groups within time points. Principal component-linear discriminant analysis showed that lipidomic profiles between the feeding groups were mainly characterized by phosphatidylcholines (PC), phosphatidylethanolamines (PE), lysophophosphatidylcholines (LysoPC), and lysophosphatidylethanolamines (LysoPE) in the liver, and by Cer, PE, and phosphatidylinositols (PI) in AT. Lipid class levels indicated that prepartal overfeeding elevated the concentration of PE, PI, LysoPC, LysoPE, and sphingomyelin in the liver, and increased the concentration of Cer in AT during the periparturient period.

Conclusion

Prepartal overfeeding significantly altered the concentrations of various sphingolipids, phospholipids, and lysophospholipids in the liver and AT of dairy cows during the periparturient period.

The aim of these studies was to determine the effect of levels of dry matter (DM) and metabolisable energy (ME) intakes on clearance rate of progesterone (P4) in dairy cows. Thirty-two lactating Holstein-Friesian cows were selected for the study and were fed indoors in individual stalls for a period of 5 weeks. They were individually offered a diet of combinations of pasture, hay and pelleted cereal grain to achieve two different levels of DM and ME. In the first trial, 16 cows were allocated to two groups: (i) high DM (HDM), and (ii) low DM (LDM) intakes, while the amount of ME intake was constant. In the second trial, 16 cows were allocated to two groups: (i) high ME, and (ii) low ME intakes with similar amount of DM intake. A GnRH-agonist (deslorelin) was initially implanted in the ear of each cow to block endogenous P4 secretion. Then 3 weeks later, a CIDR device was inserted into the vagina of each cow and left in place for 11 days. Chromic oxide (Cr(2)O(3)) capsules were administered to allow daily faecal output (FO) to be estimated. Daily blood, faecal and milk samples were taken during the period of the experiment for P4 and faecal P4 metabolites analyses. Trial 1: The average milk yield was similar among cows in high and LDM intake groups (26.7 versus 25.0 l per day, P = 0.2). The average daily FO was 7.8 kg DM in the HDM and 5.7 in the LDM cows (P < 0.0001). Average daily DM intakes were 17.3 kg and 15.4 kg in the HDM and LDM groups, respectively (P < 0.0001). The average plasma P4 concentrations were similar between the two groups (1.56 versus 1.60 ng/ml, P = 0.7) but milk P4 concentrations were higher in LDM cows (4.6 versus 3.6 ng/ml, P = 0.02). The average daily excretion rate of P4 into the milk was higher in LDM cows (122.3 versus 88.5 microg, P = 0.002). The concentrations of faecal P4 metabolites (FP4M) were not influenced by the level of daily DM intake (2.85 versus 2.90 microg/g, P = 0.6). The average daily yields of FP4M were higher among cows in the HDM group (23.2 versus 16.3mg, P = 0.01).Trial 2: The average milk yield was 31.2l per day in HME cows compared to 25.0l per day in LME cows (P < 0.0001). The average daily FO was 7.8 kg DM in LME and 5.8 kg DM in HME cows (P < 0.0001), and the average DM content of faeces was higher in LME cows (15.8 versus 12.7%, P = 0.01). The average daily ME intake was 213MJ per day in HME group compared to 183MJ per day in LME group (P<0.0001). The average plasma and milk P4 concentrations were similar between the two groups (plasma P4 = 1.54 versus 1.56 ng/ml, P = 0.4; milk P4: 3.7 versus 3.6 ng/ml, P = 0.6). The average daily excretion rate of P4 into the milk was higher in HME cows (114 versus 88.5 microg, P = 0.03). Concentrations of FP4M were not influenced by the level of daily ME intake (2.5 versus 2.85 micro g/g, P = 0.08). However, daily yields of FP4M were greater in the LME group (23.2 versus 14.4 mg, P = 0.01).In conclusion, this study was unable to establish a relationship between the level of DM and ME in the diet with the excretion rates of FP4M metabolites and plasma P4 concentrations.  相似文献

This study was designed to compare two timed insemination protocols, in which progesterone, GnRH and PGF2alpha were combined, with the Ovsynch protocol in presynchronized, early postpartum dairy cows. Reproductive performance was also evaluated according to whether cows showed high or low plasma progesterone concentration, at the onset of treatment. One hundred and six early postpartum dairy cows were presynchronized with two cloprostenol treatments given 14 days apart, and then assigned to one of the three treatment groups. Treatments for the synchronization of estrus in all three groups started 7 days after the second cloprostenol injection, which was considered Day 0 of the actual treatment regime. Cows in the control group (Ovsynch, n=30) were treated with GnRH on Day 0, PGF2alpha on Day 7, and were given a second dose of GnRH 32 h later. Cows in group PRID (n=45) were fitted with a progesterone releasing intravaginal device (PRID) for 9 days, and were given GnRH at the time of PRID insertion and PGF2alpha on Day 7. In group PRID/GnRH (n=31), cows received the same treatment as in the PRID group, but were given an additional GnRH injection 36 h after PRID removal. Cows were inseminated 16-20 h after the administration of the second GnRH dose in the Ovsynch group, and 56 h after PRID removal in the PRID and PRID/GnRH groups. Ovulation rate was determined on Day 11 postinsemination by detecting the presence of a corpus luteum in the ovaries. Lactation number, milk production, body condition at the onset of treatment and treatment regime were included as potential factors influencing ovulation and pregnancy after synchronization. Logistic regression analysis for cows with high and low progesterone concentration on treatment Day 0 revealed that none of the factors included in the models, except the interaction between progesterone and treatment regime, influenced the risk of ovulation and pregnancy significantly. In cows with high progesterone concentration at treatment onset, Ovsynch treatment resulted in a significantly improved pregnancy rate over values obtained following PRID or PRID/GnRH treatment. In cows with low progesterone concentration, PRID or PRID/GnRH treatment led to markedly increased ovulation and pregnancy rates with respect to Ovsynch treatment. These findings suggest the importance of establishing ovarian status in early postpartum dairy cows before starting a timed AI protocol, in terms of luteal activity assessed by blood progesterone.  相似文献

The aim of this study was to evaluate the effect of delaying ovulation subsequent to superstimulation of follicular growth in beef cows (Bos indicus) on embryo recovery rates and the capacity of embryos to establish pregnancies. Ovulation was delayed by three treatments using either progesterone (CIDR-B) or a GnRH agonist (deslorelin). Multiparous Nelore cows (n = 24) received three of four superstimulation treatments in an incomplete block design (n = 18 per group). Cows in Groups CTRL, P48 and P60 were treated with a CIDR-B device plus estradiol benzoate (EB, 4 mg, i.m.) on Day-5, while cows in Group D60 were implanted with deslorelin on Day-7. Cows were superstimulated with FSH (Folltropin-V, 200 mg), from Day 0 to 3, using twice daily injections in decreasing amounts. All cows were treated with a luteolytic dose of prostaglandin on Day 2 (08:00 h). CIDR-B devices were removed as follows: Group CTRL, Day 2 (20:00 h); Group P48, Day 4 (08:00 h); Group P60, Day 4 (20:00 h). Cows in Group CTRL were inseminated at 10, 20 and 30 h after first detected estrus. Ovulation was induced for cows in Group P48 (Day 4, 08:00 h) and Groups P60 and D60 (Day 4, 20:00 h) by injection of LH (Lutropin, 25 mg, i.m.), and these cows were inseminated 10 and 20 h after treatment with LH. Embryos were recovered on Days 11 or 12, graded and transferred to synchronized recipients. Pregnancies were determined by ultrasonography around Day 100. Data were analyzed by mixed procedure, Kruskal-Wallis and Chi-square tests. The number of ova/embryos, transferable embryos (mean +/- SEM) and pregnancy rates (%) were as follows, respectively: Group CTRL (10.8+/-1.8, 6.1+/-1.3, 51.5), P48 (12.6+/-1.9, 7.1+/-1.0, 52.3), P60 (10.5+/-1.6, 5.7+/-1.3, 40.0) and D60 (10.3+/-1.7, 5.0+/-1.2, 50.0). There were no significant differences among the groups (P > 0.05). It was concluded that fixed time AI in association with induced ovulation did not influence embryo recovery. Furthermore, pregnancy rates in embryos recovered from cows with delayed ovulation were similar to those in embryos obtained from cows treated with a conventional superstimulation protocol.  相似文献

This study investigated the relationships amongst personality traits and attitudes of 311 dairy stockpeople and the milk yield they obtained. A questionnaire pack consisting of a big-five measure of personality (which includes the traits of extraversion, agreeableness, conscientiousness, emotional stability and intellect), a four-factor attitude questionnaire and associated demographic and production questions was posted out to Northern Ireland dairy farmers. Pearson correlations were used to assess the relationship between personality and attitudes and partial correlations were calculated between milk yield and these psychometric measures. The personality traits of agreeableness and conscientiousness were most strongly correlated to positive attitudes towards working with dairy cows. None of the stockpeople's personality traits were significantly correlated with the milk yield they obtained. Three of the attitude scales, however, were significantly correlated with milk yield; milk yield was related to higher levels of empathy and job satisfaction and lower levels of negative beliefs. These findings, along with previous research, suggest stockperson attitudes may be important in relation to dairy cow welfare and production.  相似文献
