Fine structure of the fertilization membranes of sea urchin embryos   总被引:3,自引:0,他引:3  
The fine structure of the fertilization membranes from S. purpuratus embryos has been studied with the electron microscope. Isolated membranes before and after their full development and membranes formed under the influence of 10−3% cystine have been observed. The membrane structure was found to be trilamella: a middle layer about 200 Å thick, which originally was the vitelline membrane, and about 175 Å thick peripheral layers organized by the “crystalline material” from the cortical granules. These surface layers were again found to be trilaminated structure composed of a monolayer of parallel, closely packed flat fibrils, about 160 Å wide and 75 Å thick, adhering on both sides to parallel, 40–50 Å thick filaments separated from each other by about 100 Å and intersecting with the fibrils by an angle of about 75 °.  相似文献

Polymorphism of phosphatidylethanolamines from two natural sources   总被引:1,自引:0,他引:1  
Two pure phosphatidylethanolamines, isolated from porcine erythrocytes (PPE) and from larvae of Phormia regina (FPE), were analysed and found to differ only in their fatty-acyl composition. An X-ray diffraction study was made of the equilibrium structures formed by these two lipids, in water, as a function of lipid concentration and of temperature. They behave identically and the structural parameters of a widely occurring lamellar phase were determined and found to be similar to phosphatidylcholine. The lamellar phase, composed of alternating layers of lipid, of thickness d1, and water, of thickness dw, swells only to a limited extent (dw maximum is 13 Å) and d1 varies from 38 Å to 43 Å as the lipid concentration varies from 70% to 90% dry weight. The equilibrium structures formed by FPE and PPE in low pH buffers are quite different, PPE being identical to that in water but FPE forming a cubic phase. The reason for the difference is unclear, but the results suggest the importance of fatty-acyl composition and of the effect of pH on the polymorphism of these systems.  相似文献

Whole blood cultures from humans and from the New World primate, Saguinus fuscicollis, were irradiated with various doses of 250 kV X-rays. The resulting centric ring plus dicentric aberration yields were fitted to the three models, Y = a+bD, Y = a+bD+cD2, and Y = a+cD2, by least squares regression. In both instances the best fit was to the model Y = a+bD+cD2, with coefficients of the one- and two-track components for human and marmoset being: b = (0.78 ± 0.09)·10−3, c = (5.92 ± 0.31)·10−6, and b = (1.11 ± 0.36)·−3, c = (7.7 ± 1.7)·10−6, respectively.  相似文献

Macromomycin (Mr 12,000) is the apoprotein of the antitumor drug auromomycin, which inhibits DNA synthesis by causing single-strand breaks in DNA. Two orthorhombic crystal forms of macromomycin have been observed. The platelike crystals of one form belong to space group P212121, with cell dimensions a = 48·92 A?, b = 54·71 A?, c = 103·31 A?, Z = 8. The crystals of the second form are needle-shaped, and belong to space group P21212, with cell dimensions a = 46·1 A?, b = 54·4 A?, c = 41·2 A?, Z = 4. At this point in time, the platelike crystals appear the most suitable for continued crystallographic studies.  相似文献

Molecular mechanics simulations in Cerius2 have been used for modelling vermiculite intercalated with tetramethylammonium and aniline cations. The published structure data obtained for these intercalated structures from X-ray single crystal diffraction have been used to test the force fields and modelling strategy for organo-clays. The strategy of modelling was based on the nonbond host-guest interactions and on rigid silicate layers and rigid guest species. The rigidity of silicate layers requires that the cell parameters a, b and % are kept fixed during the energy minimisation. The energy term was set up using the nonbond interaction terms only and the Crystal Packer module in Cerius2 has been used for the energy minimisation. In Crystal Packer the rigid units, i.e. the silicate layers and guest species can be translated and rotated during energy minimisation and the cell parameters c, !, and # have been varied. Three sets of Van derWaals (VDW) parameters available in Crystal Packer: Tripos, Universal and Dreiding have been used in present molecular simulations. Ab initio MP2 calculations were performed to justify the application of the force field. The best agreement of molecular mechanics simulations with both: experimental and ab initio data was obtained with the Tripos VDW parameters for both intercalates. The results of modelling are in good agreement with the experimental data as to the cell parameters and the interlayer packing. The cell parameters reported by Vahedi-Faridi and Guggenheim (1997) for tetramethylammonium-vermiculite are: c = 13.616 Å, ! = 90°, # = 97.68° ; from the present modelling we obtained: c = 13.609 Å, ! = 90.19°, # = 97.56°. Tetramethylammonium-cations are arranged in one layer in the interlayer space. One C-C edge of NC4 tetrahedra is perpendicular to the silicate layers. The deep immersion of the methyl groups into the ditrigonal cavities suggested by Vahedi-Faridi and Guggenheim was not confirmed by modelling. Slade and Stone (1984) presented the measured cell parameters for aniline vermiculite: c = 14.89 Å, ! = 90°, # = 97°; present result is: c = 14.81 Å, ! = 90.72°, # = 96.70° for partially exchanged vermiculite and c = 14.84 Å, ! = 90.53°, # = 97.17° for fully exchanged vermiculite. The aniline cations are positioned over the ditrigonal cavities alternating in their anchoring to lower and upper silicate layer. The C-N bonds are perpendicular to layers.  相似文献

The consequences of incorporating non-complementary residues into the poly (I) · poly (C) helix have been investigated. Complexes of poly (I) and copolymers of C with different mole-ratios of I, A and U residues have been prepared and denatured in a variety of solvents. The results of both denaturation and analysis of the stoichiometry of the reactions suggest that in poly (I)· poly (C, Ix) complexes, the I residues are excluded from the helix matrix, whereas in the poly (I) · poly (C, Ux) and poly (I) · poly (C, Ax) systems the minor component bases are retained. Preliminaries to a quantitative analysis of the transition data are presented, permitting rough estimates of the difference in stability between poly (I) · poly (C) and poly (I) · poly (U) or poly (I) · poly (A) pairs in these complexes—the results being 1.7 kcal./mole and 1.3 kcal./mole, respectively. The differences in behavior of poly (I) · poly (C, I) complexes are found to be most evident in the presence of 8 m-urea.  相似文献

The relative reactivity of 3 naphthoquinones, which are feeding inhibitors for Periplaneta americana and Scolytus multistriatus, with each of 7 amino acids was measured by ultraviolet difference spectroscopy. Juglone (5-hydroxy-1,4-naphthoquinone), menadione(2-methyl-1,4-naphthoquinone) or 1,4-naphthoquinone produced difference spectra immediately upon mixing with cysteine, but not with valine, serine, glutamic acid, arginine, tryptophan or proline in phosphate buffer (pH 7.0). The Ks values for the reactions indicated that the affinities of 1,4-naphthoquinone (Ks = 4.4 · 10−4M) and juglone (Ks = 8.3 · 10−4M) for cysteine were comparable, but were both approx. 10 times greater than that for menadione (Ks = 3.2 · 10−3M). The extinction coefficient of the complex formed by cysteine with juglone (A = 3.448 · 10−1M) was approx. 2 times greater than that of 1,4-naphthoquinone (A = 1.290 · 10−1M) or menadione (A = 1.176 · 10−1 M). The importance of these results to explaining the mechanism of chemoreception in P. americana and S. multistriatus is discussed.  相似文献

A hexagonal crystal form (P6322, a = b = 34.0 A?, c = 113.5 A?) and a monoclinic form (P21, a = 37.1 A?, b = 32.2 A?, c = 32.4 A?, β = 110 °) of neutrophil cationic protein NP2, isolated from rabbit leukocytes, have been characterized. The monoclinic form, containing two promoters (Mr = 3844) per asymmetric unit, diffracts to at least 1.8 Å and is suitable for high-resolution structural studies.  相似文献

Levansucrase, an exocellular enzyme, has been isolated from a high producer mutant, the BS5C4 constitutive strain, of Bacillus subtilis. Three crystalline forms have been obtained, all three belonging to the orthorhombic space group P212121. The most suitable form for a three-dimensional structure investigation has cell dimensions, a = 68 A?, b = 125 A?, c = 54 A?. There is one molecule in the asymmetric unit.  相似文献

A recombinant thermostable l-fucose isomerase from Dictyoglomus turgidum was purified with a specific activity of 93 U/mg by heat treatment and His-trap affinity chromatography. The native enzyme existed as a 410 kDa hexamer. The maximum activity for l-fucose isomerization was observed at pH 7.0 and 80 °C with a half-life of 5 h in the presence of 1 mM Mn2+ that was present one molecular per monomer. The isomerization activity of the enzyme with aldose substrates was highest for l-fucose (with a kcat of 15,500 min−1 and a Km of 72 mM), followed by d-arabinose, d-altrose, and l-galactose. The 15 putative active-site residues within 5 Å of the substrate l-fucose in the homology model were individually replaced with other amino acids. The analysis of metal-binding capacities of these alanine-substituted variants revealed that Glu349, Asp373, and His539 were metal-binding residues, and His539 was the most influential residue for metal binding. The activities of all variants at 349 and 373 positions except for a dramatically decreased kcat of D373A were completely abolished, suggesting that Glu349 and Asp373 were catalytic residues. Alanine substitutions at Val131, Met197, Ile199, Gln314, Ser405, Tyr451, and Asn538 resulted in substantial increases in Km, suggesting that these amino acids are substrate-binding residues. Alanine substitutions at Arg30, Trp102, Asn404, Phe452, and Trp510 resulted in decreases in kcat, but had little effect on Km.  相似文献

Although both an active form of the vitamin D metabolite, 1,25(OH)2D3, and the vitamin D analogue, ED71 have been used to treat osteoporosis, anti-bone resorbing activity is reportedly seen only in ED71- but not in 1,25(OH)2D3 -treated patients. In addition, how ED71 inhibits osteoclast activity in patients has not been fully characterized. Recently, HIF1α expression in osteoclasts was demonstrated to be required for development of post-menopausal osteoporosis. Here we show that ED71 but not 1,25(OH)2D3, suppress HIF1α protein expression in osteoclasts in vitro. We found that 1,25(OH)2D3 or ED71 function in osteoclasts requires the vitamin D receptor (VDR). ED71 was significantly less effective in inhibiting M-CSF and RANKL-stimulated osteoclastogenesis than was 1,25(OH)2D3 in vitro. Downregulation of c-Fos protein and induction of Ifnβ mRNA in osteoclasts, both of which reportedly block osteoclastogenesis induced by 1,25(OH)2D3 in vitro, were both significantly higher following treatment with 1,25(OH)2D3 than with ED71. Thus, suppression of HIF1α protein activity in osteoclasts in vitro, which is more efficiently achieved by ED71 rather than by 1,25(OH)2D3, could be a reliable read-out in either developing or screening reagents targeting osteoporosis.  相似文献

Single crystals of an l-arabinose-binding protein from Escherichia coli B/r suitable for a high-resolution structural analysis have been obtained. An X-ray examination of the orthorhombic crystals shows the space group is P212121, with unit cell dimensions: a = 55.3 A?, b = 71.4 A? and c = 77.5 A?. Ten gibberellins (GAs) have been identified by Kovats retention indices and full mass spectra from GC-MS analysis of purified extracts of sporophytes of the tree-fern, Cyathea australis. These include the known GA1, GA4, GA9, GA15, GA24, GA35, and GA58 and three new GAs, 12β-hydroxyGA9 (GA69), 12α-hydroxyGA9 (GA70) and 12β-hydroxyGA4 (GA71). The structure of GA71 was established by the preparation and characterization of its methyl ester (as a metabolite of GA4 methyl ester in a culture of prothallia of Lygodium japonicum).  相似文献

Crystals of the complex between porcine pancreatic secretory trypsin inhibitor (Kazal PSTI) and trypsinogen have been grown from magnesium sulphate solution at pH 6.7. The crystals belong to space group P212121, with unit cell edges a = 67.1 A?, b = 75.5 A?, c = 66.9 A?, and the asymmetric unit contains one complex moiety. The diversity of hemoglobin phenotypes observed among Malaysian Macaca nemestrina (pig-tailed macaques) has been attributed, in part, to the presence of duplicated α-chain loci in some members of this species. To date, evidence in support of this view has been indirect, consisting of variation in proportions of αI (Asp71, Gln78) and αII (Asp71, His78) chains among presumptive heterozygotes. However, the discovery that erythrocytes from some M. nemestrina contain αI and αII chains in company with a third, or αIII, chain (Gly71, Gln78) provides direct evidence of duplicated α-chain loci.  相似文献

A rapid surface adhesion-based immunofluorescence technique was used to detect Listeria monocytogenes from inoculated culture systems. The effect of culture type (pure, mixed and meat), pH (7·00, 6·40, 4·76 and 3·13), acids (citric and HCl) and temperature (25°, 30° and 37°C) on the adhesion of Listeria to the polycarbonate membrane used in this technique was determined. It was found that pH had a significant effect ( P < 0·05) with higher numbers of Listeria adhering at low pH values (4·76). Culture type was also important with significantly higher numbers of Listeria ( P < 0·05) adhering to membranes immersed in meat cultures than in pure or mixed cultures. This effect was seen at 30°C but not at 25° or 37°C. The total viable count (TVC) on the membrane was unaffected by pH but temperature had an influence with optimum adhesion occurring at 25°C. The reasons for observed differences and their implications for the surface adhesion immunofluorescent rapid method are discussed.  相似文献

Single crystals of the regular 1:1 complex of T and R-state tetramers of l -lactate dehydrogenase from Bifidobacterium longum have been grown from polyethylene glycol 6000 solution in the presence of NADH, fructose 1,6-bisphosphate and oxamate. The crystals belong to space group F222 with unit cell dimensions of a = 148·4 Å, b = 295·9 Å and c = 71·0 Å. The crystals are suitable for X-ray analysis and diffract to beyond 2·5 Å spacing. A crystallographic study showed that the asymmetric unit contains two subunits and that one of them belongs to the T-state tetramer and the other to the R-state tetramer.  相似文献
