The non-autonomous logistic equation

is studied under conditions that include an environment which is completely deteriorating. In this setting, when the population's growth rate, r, is large on the average, solutions track the environment with a consequent extinction of the population. However, when both r and rK?1 are small in the sense that they are in L1[0,∞) then an asymptotic equivalence, where all solutions tend to positive limits as t approaches infinity, results and the population is persistent, independent of initial density. The asymptotic equivalence produces an unreasonable overshoot of carrying capacity which leads to concern about employing the logistic equation in the above form as a population model when growth rates are close to zero.A re-interpretation of the parameters of the logistic equation leads to the alternative logistic formulation

. A biological interpretation of the parameters is presented and this equation is compared with the classical logistic model in the case where the parameters are constant. If the alternative logistic model is applied in a situation with time-varying parameters, then a deteriorating environment always leads to extinction of the population regardless of the behavior of r. Similarly, a growth rate which is small on the average results in extinction regardless of the behavior of B. Furthermore, r and B have limiting values as t approaches infinity then so does x and the terminal value of x is equal to the terminal value of the carrying capacity of the population. In general, the alternative formulation seems to be the more reasonable model in situations where perturbations lead to severe decreases in environmental quality and growth rates.  相似文献

pH Dependences of steady-state kinetic parameters of cytochrome chains of submitochondrial particles have been studied. It has been shown that the lifetimes of activated states (τ) of the pairs of cytochromes bc1 and aa3 have different pH dependences; those for the c1c and ca cytochrome pairs being similar. The rate constants for the non-activated state of the respiratory chains decreased for the bc1 pair and increased for the aa3 pair when the pH value was increased.The values of pK calculated from these dependences for the pairs bc1 and aa3 were 7.2 and 8.9, respectively. It has been supposed that the ratio of activated to non-activated electron carriers may be controlled by the local pH value in the mitochondrial membrane, the latter being dependent upon the rate of electron transfer. The kinetic model based on this assumption allows one to explain the experimental dependences on pH of the rate constants for cytochromes bc, and aa3.The values of the diffusion rate constants for H+ and OH? ions in the mitochondrial membrane estimated from these kinetic data obtained in this study weree 104–105 s?1 and 102–103 s?1, respectively.  相似文献

Cassia leiandra is an Amazonian plant species that is used popularly for the treatment of mycoses. Recently, a protease inhibitor, named ClTI, with insecticidal activity against Aedes aegypti, was purified from the mature seeds of C. leiandra. In this work, we show that ClTI has antifungal activity against Candida species and describe its mode of action towards Candida albicans. This study is relevant because the nosocomial infections caused by Candida species are a global public health problem that, together with the growing resistance to current drugs, has increased the urgency of the search for new antifungal compounds. ClTI inhibited the growth of Candida albicans, Candida tropicalis, Candida parapsilosis, and Candida krusei. However, ClTI was more potent against C. albicans. The candidicidal mode of action of ClTI on C. albicans involves enhanced cell permeabilization, alteration of the plasma membrane proton-pumping ATPase function (H+ -ATPase), induction of oxidative stress, and DNA damage. ClTI also exhibited antibiofilm activity and non-cytotoxicity to mammalian cells. These results indicate that ClTI is a promising candidate for the future development of a new, natural, and safe agent for the treatment of infections caused by C. albicans.  相似文献

It is becoming generally accepted in recent literature that the Warburg effect in cancer depends on inhibition of M2PYK, the pyruvate kinase isozyme most commonly expressed in tumors. We remain skeptical. There continues to be a general lack of solid experimental evidence for the underlying idea that a bottle neck in aerobic glycolysis at the level of M2PYK results in an expanded pool of glycolytic intermediates (which are thought to serve as building blocks necessary for proliferation and growth of cancer cells). If a bottle neck at M2PYK exists, then the remarkable increase in lactate production by cancer cells is a paradox, particularly since a high percentage of the carbons of lactate originate from glucose. The finding that pyruvate kinase activity is invariantly increased rather than decreased in cancer undermines the logic of the M2PYK bottle neck, but is consistent with high lactate production. The “inactive” state of M2PYK in cancer is often described as a dimer (with reduced substrate affinity) that has dissociated from an active tetramer of M2PYK. Although M2PYK clearly dissociates easier than other isozymes of pyruvate kinase, it is not clear that dissociation of the tetramer occurs in vivo when ligands are present that promote tetramer formation. Furthermore, it is also not clear whether the dissociated dimer retains any activity at all. A number of non-canonical functions for M2PYK have been proposed, all of which can be challenged by the finding that not all cancer cell types are dependent on M2PYK expression. Additional in-depth studies of the Warburg effect and specifically of the possible regulatory role of M2PYK in the Warburg effect are needed.  相似文献

Culex salinarius is susceptible to Plasmodium hermani, a malarial parasite of wild turkeys in Florida. The sporogonous cycle was completed and mosquitoes with infected salivary glands transmitted the parasite by bites. Transmission was also achieved by intraperitoneal and intravenous injections of whole body slurry. This is the third species found to be susceptible to turkey malaria in Florida. A comparison of C. salinarius with two other susceptible Florida mosquitoes, Culex nigripalpus and Wyeomyia vanduzeei, revealed that C. salinarius was more susceptible to P. hermani based on oocyst counts. C. nigripalpus has previously been demonstrated as an experimental and a natural vector of P. hermani, whereas W. vanduzeei has been designated as an experimental host only. In W. vanduzeei at least 30% of the oocysts were melanized (“black bodies”) and this mosquito did not transmit the parasite via bites. Additional detailed comparisons of comparative susceptibility and transmission potentials of these three species to turkey malaria, P. hermani, have been made.  相似文献

Microiontophoretic injection of calcium ions or of adenosine 3′:5′-cyclic monophosphoric acid (3′:5′-cAMP) causes fertilized eggs of Ilyanassa obsoleta to form a large lobe-like protuberance near the micropipet tip within 15–30 sec. A protuberance can be induced to form anywhere on the egg surface, i.e., animal hemisphere as well as vegetal hemisphere. Injections of comparable amounts of Na+, K+, Mg2+, Hepes buffer, seawater, guanosine 2′:3′-cyclic monophosphoric acid (2′:3′-cGMP), or guanosine 3′:5′-cyclic monophosphoric acid (3′:5′-cGMP) have no effect on cell shape. Injection of 2′:3′-cAMP causes slight changes in cell shape. Injection of Ca2+ generates a shape change in spherical eggs, as well as during all phases of normal polar lobe formation, but not when polar lobes are being resorbed. Injection of Ca2+ elicits a shape change only when injection currents exceed 120 nA and only when Ca2+ also is present in the exogenous bath solution. Cell shape changes causes by injection of 3′:5′-cAMP also are dependent upon a minimum current (approximately 300 nA) and upon the presence of exogenous Ca2+. These shape changes may depend upon exogenous Ca2+ either because the injections trigger a change in membrane permeability, or because exposure of eggs to Ca2+-free seawater lowers intracellular [Ca2+] to such an extent that threshold levels of Ca2+ are not attained during injection.  相似文献

In this study, the leaves extract of Cordia myxa, has been used for the first time to synthesize zinc oxide (ZnO) hierarchical microstructures. The solution combustion method was employed as a self-sustaining reaction between zinc nitrate and the leaves extract. The surface properties of leaves mediated ZnO microstructures were determined by UV–Visible spectral analysis, Fourier transform infrared (FT-IR), Cold field emission-scanning electron microscopy (CFE-SEM), Energy dispersive X-ray (EDX), X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). In addition, the effect of the leaves extract concentration on ZnO structures, size and surface properties was also studied. ZnO structures synthesized employing C. myxa were found to be hexagonal, triangular and round in shape which was determined using CFE-SEM. X-ray diffraction (XRD) analysis confirmed the crystalline nature of compounds. Furthermore, C. myxa mediated ZnO microstructures shows good bactericidal activity against Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria.  相似文献

Complexation in the H+-Si(OH)4-tropolone (HL) system was studied in 0.6 M (Na)Cl medium at 25° C. Speciation and formation constants were determined from potentiometric (glass electrode) and 29Si-NMR data. Experimental data cover the ranges 1.5 ? - log[H+] ? 8.4, 0.002 ? B ? 0.012 M, and 0 ? C ? 0.060 M (B and C stand for the total concentration of Si and tropolone, respectively). In acid solutions (-log[H+] ? 3) a hexacoordinated cationic complex, SiL3+, is formed with log K(Si(OH)4 + 3HL + H+ XXX SiL3+ + 4H2O) = 7.08 ± 0.03. Furthermore, the formation of a disilicic acid was established from 29Si-NMR data. The dimerization constant of Si(OH)4 was found to be 10 exp (1.2 ± 0.1). In model calculations the solubility of quartz and amorphous SiO2 in the presence of tropolone is demonstrated. Data were analyzed using the least-squares computer program LETAGROPVRID.  相似文献

Rhizobia nodulating native Astragalus and Oxytropis spp. in Northern Europe are not well-studied. In this study, we isolated bacteria from nodules of four Astragalus spp. and two Oxytropis spp. from the arctic and subarctic regions of Sweden and Russia. The phylogenetic analyses were performed by using sequences of three housekeeping genes (16S rRNA, rpoB and recA) and two accessory genes (nodC and nifH). The results of our multilocus sequence analysis (MLSA) of the three housekeeping genes tree showed that all the 13 isolates belonged to the genus Mesorhizobium and were positioned in six clades. Our concatenated housekeeping gene tree also suggested that the isolates nodulating Astragalus inopinatus, Astragalus frigidus, Astragalus alpinus ssp. alpinus and Oxytropis revoluta might be designated as four new Mesorhizobium species. The 13 isolates were grouped in three clades in the nodC and nifH trees. 15N analysis suggested that the legumes in association with these isolates were actively fixing nitrogen.  相似文献

The final step in the erythrocyte methemoglobin reduction pathway, the transfer of an electron from cytochrome b5, to methemoglobin, has been studied using magnetic circular dichroism spectroscopy. Spectral analysis allowed us to determine accurately the concentration of each redox species in mixtures of the two heme-proteins and to follow simultaneously the kinetics of the appearance or disappearance of each of these species during reduction reactions. Our analysis detected a substantial increase in the high-spin hydroxymethemoglobin species in the partially reduced bovine hemoglobin tetramer. This species was sensitive to the degree of reduction and pH, and was spectrally similar to fluoride methemoglobin. At pH 7.8. 100% of the hydroxide component of methemoglobin was in the high-spin form when two or more subunits were in the ferrous form. Kinetic analysis of bovine methemoglobin reduction yielded values for the apparent first-order rates for the tetrameric species possessing four, three, two, and one ferric subunit. Further analysis showed that the reduction kinetics can also be described by an equilibrium state, pure competitive inhibition model for enzyme catalysis in which ferrous and ferric subunits of hemoglobin compete for cytochrome b5 This analysis generated a KD that depends on ionic strength and hemoglobin tetramer conformation, a Vmax that was independent of these factors, and an inhibition constant that was equal to Kd. This model is consistent with the hypothesis that the reduction of methemoglobin can be separated into two steps, the ionic interaction between cytochrome b5 and hemoglobin and the electron transfer.  相似文献

A series of 1-(1-benzyl-2-methyl-5-((1-phenyl-1H-1,2,3-triazol-4-yl)methoxy)-1H-indol-3-yl)ethanone and ethyl 1-benzyl-2-methyl-5-((1-phenyl-1H-1,2,3-triazol-4-yl)methoxy)-1H-indole-3-carboxylate derivatives were designed based on bioisosteric replacement of previously reported antitubercular agent (IND-07). Twenty ligands were successfully synthesized and some of them were found to have good in vitro activity (MIC?<?10?μM) against the H37Rv strain of Mycobacterium tuberculosis. Among these compounds, KC-08 and KC-11 inhibited Mtb-DHFR with 4- and 18-fold selectivity for Mtb-DHFR over h-DHFR, respectively. Compound KC-11 display acceptable ADME, and better pharmacokinetic profiles than IND-07. Docking studies were performed to predict the binding mode of the compounds within the active site of Mtb-DHFR and h-DHFR. The results of our study suggest that compound KC-11 may serve as a valuable lead for the design and development of selective inhibitors of Mtb-DHFR with potential therapeutic application in tuberculosis.  相似文献

The crossover or nearest neighbor interchange metric has been proposed for use in numerical taxonomy to obtain a quantitative measure of distance between classifications that are modeled as unrooted binary trees with labeled leaves. This metric seems difficult to compute and its properties are poorly understood. A variant called the closest partition distance measure has also been proposed, but no efficient algorithm for its computation has yet appeared and its relationship to the nearest neighbor interchange metric is incompletely understood. I investigate four conjectures concerning the nearest neighbor interchange and closest partition distance measures and establish their validity for trees with as many as seven labeled vertices. For trees in this size range the two distance measures are identical. If a certain decomposition property holds for the nearest neighbor interchange metric, then the two distance measures are also identical at small distances for trees of any size.  相似文献

6-Chloromethylbenzo[a]pyrene (6-CMBP) labeled with 13C in the chloromethyl group was used as a model for those carcinogens which form essentially free carbocations. Using 13C-NMR to identify products, the selectivity with which this electrophile modifies nucleosides was investigated. At pH 7, guanosine and deoxyguanosine are the most nucleophilic nucleosides toward the carbocation generated by solvolysis of 6-CMBP. Attack at N-7 predominates over attack at N-2. At higher pH, the nucleophilicity of guanosine and deoxyguanosines increases markedly. In addition, the site of modification changes to N-1 with secondary modification at O-6. The pH dependence of the rate of this reaction implicates a group with pK-value approx. 8.7 which was assigned to the hydrogen on N-1. The presence of a methyl group on the N-7 position of guanosine lowers this pK-value to approx. 7.2. Consequently, N7-methylguanosine shows the high nucleophilicity at physiological pH that guanosine has at high pH. These observations lead to the suggestion of a one base: two-site model for chemical carcinogenesis.  相似文献

Dihydroorotate dehydrogenase in rat brain mitochondria is capable of producing superoxide. The presence of a superoxide dismutase activity in brain mitochondria, similar to that found in mitochondria from chicken liver, suggests that production of superoxide may occur in vivo. Formation of superoxide is not dependent upon reduction of cytochrome b, rather, superoxide production is competitive with cytochrome b reduction. Phenazine methosulfate apparently competes with both oxygen (superoxide production) and cytochrome b as an electron carrier but does not enhance reduction of dichlorophenolindophenol or cytochrome c.  相似文献

We describe a model that enables us to trace the traffic of allograft-responding inflammatory leukocytes to and from the graft without handling of these cells in vitro. At different times after transplantation, the kidney transplant pedicle—including the artery, vein, and draining lymphatics—is clamped. The allograft-responding leukocytes are labeled by a [3H]thymidine pulse either in situ or in the systemic lymphoid organs of the recipient. Fifteen minutes later the pulse is chased with a 1000-fold excess of cold thymidine, and the clamp is opened. The animals are sacrificed 18 hr later, when a balance between the synthesis of new labeled leukocytes from the originally labeled ones and dilution of intracellular label has been achieved. This model was used to analyze the allograft-responding inflammatory cell traffic to and from a renal transplant performed across the major histocompatibility complex in the rat. A sizable traffic was observed to both directions: After systemic injection of label only 0.008 × 106 labeled cells × hr?1 were found to emigrate into a kidney allograft (control). Already on the third day after transplantation—when the in situ inflammatory response is still at its beginning—more than 0.3 × 106 labeled cells × hr?1 migrated from the host to the allograft and 1.6 × 106 labeled cells × hr?1 left the allograft to the recipient spleen. The first figure is several-fold higher than any previous estimate. The findings emphasize the systemic nature of the antiallograft inflammatory response.  相似文献

Kluyveromyces marxianus CCT 7735 has been used to produce ethanol, aromatic compounds, enzymes and heterologous proteins besides assimilates lactose as carbon source. Its genome has 10.7 Mb and encodes 4787 genes distributed in 8 nuclear chromosomes and one mitochondrial. Contrary to Kluyveromyces lactis, which has a unique LAC12 gene (encodes lactose permease), K. marxianus possesses four. The presence of degenerated copies and Solo-LTRs related to retrotransposon TKM close to the LAC12 genes in K. marxianus indicates ectopic recombinations. The Lac12 permeases of K. marxianus and K. lactis are conserved, however the conservation is higher between the copy of the left side of the chromosome three and the unique copy of K. lactis, indicating that this copy is the ancestor. The expression of the four LAC12 genes occurred in aerobiosis and hypoxia. Notably, the high lactose consumption in hypoxia seems to be related to the high expression of the LAC12 genes.  相似文献

The Notch signaling pathway has been identified as a therapeutic target for cancers. γ-Secretase inhibitors (GSIs) have been progressively recognized as potential anticancer drugs. The present study aimed to investigate the effects of anti-delta like legend 4 (anti-DLL4) treatment on the anticancer efficacy of GSIs in gastric cancer. SGC-7901-GFP human gastric cancer cells were tested for DLL4 expression by rosette formation test and immunofluorescence, and then were treated with anti-DLL4 antibody N-[N-(3,5-difluorophenacetyl)-L-ananyl]-S-phenylglycine t-butyl ester (DAPT, a type of GSI), or a combination of anti-DLL4 antibody and DAPT. The effects of in vitro treatments on cell apoptosis, cell cycle, and cell invasion were analyzed. For in vivo study, an orthotopic mouse model of gastric cancer was established with green fluorescence expressing SGC-7901. Ultrasound targeted microbubble destruction was used to treat tumor-bearing mice with anti-DLL4 antibody conjugated microbubbles, DAPT, and a combination of the two. Real-time fluorescence imaging was performed to assess tumor cell inhibition in each group. Following in vivo treatments, apoptosis of tumor cells and the expression of apoptosis-related genes BAX, Bcl-2, and P53 were detected by TUNEL and immunohistochemical staining. In vivo combined treatment of anti-DLL4 and DAPT led to a higher rate of cell apoptosis and greater inhibition of cell invasion than that observed with DAPT treatment alone. DAPT and anti-DLL4 combination therapy resulted in decreased cell distribution at G1 phase and increased cell distribution at S phase, compared to the untreated control group (P < .01). In vivo combined therapy with anti-DLL4 and DAPT significantly increased tumor growth inhibition and tumor cell apoptosis when compared to DAPT therapy alone (P < .05). In addition, combined treatment significantly increased expression of BAX and P53 and reduced Bcl-2 expression (P < .05). Conversely, treatment with DAPT alone only increased expression of BAX and P53 (P < .05), suggesting that the reduction of Bcl-2 expression may play an important role in the synergetic antitumor and proapoptosis effects of the combined treatment. Concurrent treatment with anti-DLL4 enhances the antitumor and proapoptotic efficacy of the γ-secretase inhibitor in gastric cancer both in vitro and in vivo.  相似文献
