This study was carried out to measure blood volume in interleukin-6 (IL-6) deficient and the wild type mice and to relate any differences to concomitant changes in body composition. Young animals (8-20 weeks of age) were used in this study. The red blood cell (RBC) and plasma volumes were measured by isotope-dilution using Cr(51)-labeled erythrocytes and I(131)-labeled human albumin injected simultaneously in each animal. Red blood cells and plasma volumes and hence total measured blood volume were significantly larger in the IL-6-deficient than in either male or female wild type mice. There were no differences in either whole body hematocrit or Fcells ratio. There were no differences in water, fat, protein and mineral content between the genotypes of either gender. Thus, IL-6-deficient mice at young age have a significantly greater blood volume than the wild type mice without concomitant changes in body composition. We suggest that the changes in blood volume may reflect a role for IL-6 in the regulation of the peripheral circulation.  相似文献

Plasma and erythrocyte cholinesterase activities were determined for 40 free-living and 12 captive common long-nosed armadillos (Dasypus novemcinctus) in order to establish normal values for monitoring pesticide exposure. Plasma cholinesterase activity ranged from 105 to 549 U/liter with no sexual or seasonal differences. Plasma values from captive animals were significantly lower than those from wild armadillos. Erythrocyte cholinesterase activity ranged from 2,915 to 15,126 U/liter with no differences detected between captive and wild animals or between sexes. However, erythrocyte cholinesterase values varied seasonally. Erythrocyte and plasma cholinesterase activities were not significantly correlated. Packed cell volume ranged from 24 to 51% and did not vary significantly between captive and wild samples, between sexes or among seasons. However, both whole blood and erythrocyte cholinesterase activities showed significant negative correlations with packed cell volume. Controlled experiments are needed to find the factors responsible for the statistically significant difference between plasma cholinesterase activities of captive and wild armadillos. The seasonal variation in erythrocyte cholinesterase activity and the negative correlation between erythrocyte cholinesterase activity and packed cell volume can be explained by an hypothesis that relates the variation in erythrocyte cholinesterase activity to variation in erythrocyte turnover rate. Future work should involve experiments to test this hypothesis.  相似文献

To determine whether expanded intravascular volumes contributeto the older athlete's higher exercise stroke volume and maximal oxygen consumption(O2 max),we measured peak upright cycle ergometry cardiac volumes(99mTc ventriculography) andplasma (125I-labeled albumin) andred cell (NaCr51) volumes in 7 endurance-trained and 12 age-matched lean sedentary men. The athleteshad ~40% higherO2 max values thandid the sedentary men and larger relative plasma (46 vs. 38 ml/kg), red cell (30 vs. 26 ml/kg), and total blood volumes (76 vs. 64 ml/kg) (allP < 0.05). Athletes hadlarger peak cycle ergometer exercise stroke volume indexes (75 vs. 57 ml/m2,P < 0.05) and 17% largerend-diastolic volume indexes. In the total group,O2 maxcorrelated with plasma, red cell, and total blood volumes(r = 0.61-0.70,P < 0.01). Peakexercise stroke volume was correlated directly with the blood volumevariables (r = 0.59-0.67,P < 0.01). Multiple regressionanalyses showed that fat-free mass and plasma or total blood volume,but not red cell volume, were independent determinants ofO2 max andpeak exercise stroke volume. Plasma and total blood volumes correlated with the stroke volume and end-diastolic volume changes from rest topeak exercise. This suggests that expanded intravascular volumes, particularly plasma and total blood volumes, contribute to the higherpeak exercise left ventricular end-diastolic volume, stroke volume, andcardiac output and hence the higherO2 max in master athletes by eliciting both chronic volume overload and increased utilization of the Frank-Starling effect during exercise.

A study of blood parasites in small wild non-flying mammals was undertaken in three areas of the Atlantic Forest in Southeastern Brazil: Serra de Itatiaia, RJ, Serra da Bocaina, SP and Serra da Fartura, SP, from June 1999 to May 2001. A total of 450 animals (15 species) were captured in traps and it was observed in 15.5% of the blood smears the presence of Haemobartonella sp. and Babesia sp. in red blood cells. There was no statistically significant difference between parasited and non-parasited specimens regarding total plasma protein, packed cell volume and body weight, which strongly suggests that these specimens might be parasite reservoirs.  相似文献

Melatonin administration to male blue foxes from August for 1 year resulted in profound changes in the testicular and furring cycles. The control animals underwent 5-fold seasonal changes in testicular volume, with maximal values in March and lowest volumes in August. In contrast, melatonin treatment allowed normal redevelopment of the testes and growth of the winter coat during the autumn but prevented testicular regression and the moult to a summer coat the following spring. At castration in August, 88% of the tubular sections in the testes of the controls contained spermatogonia as the only germinal cell type, whereas in the treated animals 56-79% of sections contained spermatids or even spermatozoa. Semen collection from a treated male in early August produced spermatozoa with normal density and motility. Measurement of plasma prolactin concentrations revealed that the spring rise in plasma prolactin values (from basal levels of 1.6-5.4 ng/ml to peak values of 4.1-18.3 ng/ml) was prevented; values in the treated animals ranged during the year from 1.8 to 6.3 ng/ml. Individual variations in plasma LH concentrations masked any seasonal variations in LH release in response to LHRH stimulation, but the testosterone response to LH release after LHRH stimulation was significantly higher after the mating season in the treated animals, indicating that testicular testosterone production was maintained longer than in the controls. The treated animals retained a winter coat, of varied quality and maturity, until the end of the study in August.  相似文献

Recombinant human erythropoietin (rHuEPO) was purified from the conditioned media of Chinese hamster ovary cells with a transfected human erythropoietin gene. We investigated the effects of the rHuEPO in rats with renal anemia induced by partial nephrectomy. Five-sixth nephrectomy resulted in renal failure with anemia. Twenty-five days after the operation plasma urea nitrogen was increased about 2.5 times, and the red blood cell count, hematocrit, and hemoglobin concentration fell to 85% of normal. The reticulocyte count and plasma erythropoietin level did not change such as they do in patients with anemia due to chronic renal failure. Both total red blood cell volume and the plasma iron turnover rate were depressed in five-sixth nephrectomized rats compared with normal rats.The five-sixth nephrectomized rats were injected with rHuEPO (60 IU/kg) intravenously every second day for a total of six injections. After three injections of rHuEPO, circulation volume of total red blood cells was increased from 9.9 ml to 14.6 ml, and the plasma iron turnover rate was increased from 1.03 mg/kg/day to 2.12 mg/kg/day, and the reticulocyte count was also increased. After six injections, a marked increase of the red blood cell count, hematocrit, and hemoglobin concentration were observed. Plasma urea nitrogen and the creatinine levels as indications for renal function did not change after rHuEPO administration in both normal and five-sixth nephrectomized rats.In conclusion rHuEPO has a potent erythropoietic action and it is possible to cure the anemia caused by renal failure.  相似文献

Plasma and red cell volumes, body density, and water balance were measured in 19 men (32-42 yr) confined to bed rest (BR). One group (n = 5) had no exercise training (NOE), another near-maximal variable-intensity isotonic exercise for 60 min/day (ITE; n = 7), and the third near-maximal intermittent isokinetic exercise for 60 min/day (IKE; n = 7). Caloric intake was 2,678-2,840 kcal/day; mean body weight (n = 19) decreased by 0.58 +/- 0.35 (SE) kg during BR due to a negative fluid balance (diuresis) on day 1. Mean energy costs for the NOE, and IKE, and ITE regimens were 83 (3.6 +/- 0.2 ml O2.min-1.kg-1), 214 (8.9 +/- 0.5 ml.min-1.kg-1), and 446 kcal/h (18.8 +/- 1.6 ml.min-1.kg-1), respectively. Body densities within groups and mean urine volumes (1,752-1,846 ml/day) between groups were unchanged during BR. Resting changes in plasma volume (ml/kg) after BR were -1.5 +/- 2.3% (NS) in ITE, -14.7 +/- 2.8% (P less than 0.05) in NOE, and -16.8 +/- 2.9% (P less than 0.05) in IKE, and mean water balances during BR were +295, -106, and +169 ml/24 h, respectively. Changes in red cell volume followed changes in plasma volume. The significant chronic decreases in plasma volume in the IKE and NOE groups and its maintenance in the ITE group could not be accounted for by water balance or by responses of the plasma osmotic, protein, vasopressin, or aldosterone concentrations or plasma renin activity. In the frog, Rana esculenta (L.), the liver can change in volume by over 25%, depending on the respiratory conditions of the animal: in well-oxygenated specimens the organ can hoard about half of the total amount of erythrocytes in its sinusoids, and release them into the bloodstream under conditions of hypoxia. This phenomenon can be observed at a temperature of 6°C by comparing the liver volumes and haematic values of chlorobutanol-anaesthetized animals exposed to the air or submerged in still water (a condition which induces hypoxia): the blood volume remains constant, at about 5 ml per 100g of body weight, but red blood cell count and haematocrit value differ by as much as 50%. At 18°C there is an increase in oxygen demand and in anaesthetized animals, which rely totally on cutaneous respiration, the compensatory liver mechanism can no longer be observed, since all the available erythrocytes are already circulating in a blood volume which, depending on respiratory conditions, can vary between about 7 and 8ml/100g. At 30°C, cutaneous respiration alone does not allow the anaesthetized animals to survive long enough to stabilize their haematic parameters.  相似文献

目的探讨亲缘供者外周血红细胞参数对COBE Spectra血细胞分离机的自动外周血干细胞采集程序(AutoPBSC程序)与单个核细胞采集程序(MNC程序)的影响及经验分析。 方法选取河北燕达陆道培医院2019年6月至2021年2月小红细胞亲缘供者31例45次采集为小红细胞组,选取同期非小红细胞亲缘供者51例60次采集为非小红细胞组,分别应用AutoPBSC程序和MNC程序,比较两组采集情况及采集产品相关指标。采用独立样本t检验和Mann-Whitney U检验分析2组计量资料的差异。 结果与小红细胞AutoPBSC程序组比较,小红细胞MNC程序组血小板(PLT)降低率[(25.88±15.83)﹪比(36.64±10.22)﹪]、采集效率[32.65﹪(23.60﹪,73.82﹪)比63.74﹪ (59.83﹪,68.55﹪)]、采集物体积[(158.83±34.39)比(222.91±63.9)mL]、MNC总数[(218.04±117.57)×108/L比(350.24±127.64)×108/L]、CD34+细胞总数[113.83×106/L (79.25×106/L,154.10×106/L)比233.26×106/L (177.18×106/L,392.51×106/L)]、MNC计数[(4.04±2.61)×108/kg比(5.54±2.22)×108/ kg]、CD34+计数[1.84×106/kg (1.16×106/kg,4.41×106/kg)比3.64×106/kg (2.49×106/kg,6.37×106/kg)]均升高,差异有统计学意义(P < 0.05);与非小红细胞AutoPBSC程序组比较,非小红细胞组MNC程序组采集物体积[(162.83±51.74)比(242.56±43.25)mL]升高,差异有统计学意义(P < 0.05)。 结论对造血干细胞移植供者红细胞体积偏小时,应用MNC程序采集外周血造血干细胞,比应用AutoPBSC程序更有优势。  相似文献

Abstract

Changes in the circulating plasma volume were monitored for twelve consecutive months in five white‐tailed deer (Odocoileus virginianus) maintained outdoors in a 2.4‐ha enclosure in northern New York stale. The mean annual relative plasma volume of the two male white‐tailed deer (43.6 ± 1.0 ml/kg) was not significantly different from that of three females (45.7 ± 0.7 ml/kg). Mean annual absolute plasma volume, however, was significantly higher in males (2588 ± 90.7 ml) than in females (2092 ± 79.1 ml), mainly because of the greater body weights of males in late summer and early fall. Both sexes had a marked seasonality in the level of relative and absolute plasma volume regulation. Relative plasma volumes were lowest in February‐March and highest in May‐June and again in October, while absolute plasma volumes were lowest in late winter to early spring, when body weights in the annual cycle were lowest, and volumes were highest in mid‐fall, at the annual peak of body condition. Males had a greater excursion about their annual mean relative plasma volumes (‐16 to 13%) and absolute plasma volumes (‐23 to 32%) than did females (respectively, ‐11 to 9% and ‐13 to 19%). Variations in the level of plasma volume regulated are related to the annual cycle of environmental conditions and changes in body and physiological condition of white‐tailed deer.  相似文献

A method is described for frequent sequential blood volume estimation in baboons using 32P for red cell volume measurements and 125I-albumin for simultaneous plasma volume measurements. Values for red cell, plasma, and total blood volumes are reported. Close correlations of the volumes to bodyweight were demonstrated. Circulatory half-lives of the isotopes, determined from disappearance curves, confirmed their suitability for serial measurements in these baboons.  相似文献

This study was designed to investigate any seasonal (spring, summer, autumn and winter) changes in haematological parameters in the blood of Tinca tinca measuring the number of red blood cells (RBC), haematocrit, white blood cells, and total plasma proteins.The results show significant changes in RBC and haematocrit in males comparing spring and summer with autumn and winter, whereas in females the RBC remained constant for all 4 seasons but the haematocrit decreased in autumn and winter compared to spring and summer. The white blood cells of male and female animals were significantly lower in spring and winter compared to summer and autumn. In male fish total protein contents significantly decreased in autumn and winter compared to spring and summer, whereas in females protein output significantly decreased in winter compared to the other seasons. The results indicate marked seasonal variation in the blood of male and female Tinca tinca. This variation may play a important protective role for the survival of the animals.  相似文献

Blood volume, estradiol 17 beta and estrone concentrations, and plasma renin activity were determined weekly in 26 chronically catheterized nonanesthetized rabbits during pregnancy and after delivery. We determined blood volume by 99technetium, and plasma volume by the microhaematocrit method. The values for whole blood volume were 52.7 (+/- 2.3) and 63 (+/- 2.2) ml X kg-1 for nonpregnant and pregnant animals, respectively. During the course of gestation plasma renin activity increased about 300%, but estradiol 17 beta and estrone concentrations did not change significantly. The increase in whole blood and plasma volumes during pregnancy correlated highly with the increase in plasma renin activity, r = 0.53 and 0.59, respectively.  相似文献

Use of either urine or saliva samples to estimate extracellular water volume was investigated in 10 men using nonradioactive bromide (Br) and in seven newborn piglets using radioactive Br (82Br) and chloride (36Cl). The relation to Br to Cl concentrations in urine enabled an estimation of Br dilution volume from human urine (267 +/- 42 ml/kg, mean +/- SD) that was not significantly different (P = 1.0) from the Br dilution volume calculated from plasma Br concentration (268 +/- 20 ml/kg). Although the Br dilution volume estimated from saliva was not different from that of plasma, the error in the estimates of Br dilution volume from saliva was too large (mean difference, -36 +/- 64 ml/kg) to make its use practical. The data from piglets showed good agreement between 82Br and 36Cl dilution volumes calculated from 4-hr plasma samples (356 +/- 14 ml/kg and 347 +2- 12 ml/kg; P greater than 0.1) and between 82Br dilution volumes calculated from urine 82Br:36Cl and plasma 82Br (360 +/- 31 ml/kg and 356 +/- 14 ml/kg; P greater than 0.1). Extracellular water volume can be estimated in both adult and young animals using the Br dilution volume calculated from urine samples. It requires (i) two urine collections: one before and one 4 to 8 hr after administration of Br; (ii) a measurement or estimate of plasma Cl concentration; and (iii) a correction factor that describes the relationship of the ratio of Br to Cl in urine to that ratio in plasma.  相似文献

The reproductive cycle of wild rabbits (Oryctolagus cuniculus) living in Zembra Island (North Tunisia) is dependent on an external factor, the photoperiod: the gonads are inhibited by long days and stimulated by short days or melatonin implants. Here we studied the role of an internal factor, thyroid hormones and the possible thyroid-gonadal interrelationships, in animals captured on Zembra Island and maintained in natural conditions of photoperiod and temperature. We determined the seasonal profile of the thyroid and testis cycles and investigated the effects of castration and thyroidectomy on the seasonal testosterone and thyroxine cycles. Plasma thyroxine and testosterone levels followed similar, parallel seasonal patterns, with a peak in autumn (October) and low values from January to August. In thyroidectomized animals, plasma testosterone levels, although significantly higher than those in controls (P < 0.001), remained low throughout the 13 mo of the experiment, and no testicular reactivation was observed in the fall. In castrated animals, despite the increase in thyroxine concentration in the 3 mo following castration (P < 0.01), plasma thyroxine levels remained low during the 2 yr of the study. We then investigated the combined effects of long days (16L:8D) and moderately high temperature (25 degrees C) on these two endocrine axes. In constant gonado-inhibiting conditions (16L:8D), whether the temperature was kept constantly high or allowed to fluctuate naturally, no reactivation of the thyroid and testicular axes was observed in the fall. In control animals, the peaks of testosterone and thyroxine concentrations observed in September were larger (P < 0.001) than those in animals subjected to the same natural photoperiod conditions but with constantly high temperature. The lower level of autumnal testis stimulation (P < 0.001) in animals maintained in conditions of constant high temperature (25 degrees C) may be attributed to the low thyroxine levels induced by high temperature. These results clearly confirm that the thyroid and testicular cycles display similar seasonal variations and show that the thyroid and gonadal axes are strictly interdependent. This study provides the first demonstration, for a given species, that the seasonal reactivation of gonad activity is controlled by the thyroid, and thyroid activity is controlled by the gonads.  相似文献

Water contents of various body compartments were estimated within 9 h of birth in eight baboon neonates and at a mean of 29 days (27-32 days) in seven baboons. All animals were normally grown and delivered spontaneously at term. There was no difference in mean antipyrine space, corrected bromide space, intracellular water, interstitial water, and plasma and blood volumes observed on day 1 and day 29. Mean estimate of red cell volume was lower on day 29 than on day 1.  相似文献

In many species, territoriality is expressed only during the breeding season, when plasma testosterone (T) is elevated. In contrast, in song sparrows (Melospiza melodia morphna), males are highly territorial during the breeding (spring) and nonbreeding (autumn) seasons, but not during molt (late summer). In autumn, plasma sex steroids are basal, and castration has no effect on aggression. However, inhibition of aromatase reduces nonbreeding aggression, suggesting that neural steroid metabolism may regulate aggressive behavior. In wild male song sparrows, we examined the neural distribution of aromatase mRNA and seasonal changes in the activities of aromatase, 5 alpha-, and 5 beta-reductase, enzymes that convert T to 17 beta-estradiol, 5 alpha-dihydrotestosterone (5 alpha-DHT, a potent androgen), or 5 beta-DHT (an inactive metabolite), respectively. Enzyme activities were measured in the diencephalon, ventromedial telencephalon (vmTEL, which includes avian amygdala), caudomedial neostriatum (NCM), and the hippocampus of birds captured during spring, molt, or autumn. Aromatase and 5 beta-reductase changed seasonally in a region-specific manner. Aromatase in the diencephalon was higher in spring than in molt and autumn, similar to seasonal changes in male sexual behavior. Aromatase activity in the vmTEL was high in both spring and autumn but significantly reduced at molt, similar to seasonal changes in aggression. 5 beta-Reductase was not elevated during molt, suggesting that low aggression during molt is not a result of increased inactivation of androgens. These data highlight the relevance of neural steroid metabolism to the expression of natural behaviors by free-living animals.  相似文献

Levels of peripheral plasma testosterone and LH were studied in 4 bulls hourly during a 12 hr. sampling period at 5 times of the year. The average plasma testosterone levels were significantly lower in October (1.8 ng/ml, Ρ < 0.001) and December (2.5 ng/ml, Ρ < 0.05) than in February, June and August (3.5, 3.7 and 3.7 ng/ml respectively). LH showed a slight fluctuation during the day, with values ranging between 0.8 and 3.8 ng/ml, but underwent no significant seasonal variation. A significant increase in average plasma testosterone was observed 1 hr. after the LH peaks (P < 0.001).  相似文献

In an attempt to explore the acute maternal responses to exercise we measured oxygen consumption, uterine blood flow, and blood volume in 13 chronically catheterized pregnant sheep at rest and while exercising on a treadmill. With maximal exercise O2 consumption increased 5.6 times, from a resting value of 5.8 +/- 0.3 (SE) to 32.1 +/- 2.8 ml X min -1 X kg -1, cardiac output increased 2.7 times, from 149 +/- 8 to 404 +/- 32 ml X min -1 X kg -1, and arteriovenous oxygen content difference increased 2.1 times, from 3.9 +/- 0.2 to 8.0 +/- 0.4 ml X dl -1. Total uterine blood flow decreased from a mean resting value of 292 +/- 6 to 222 +/- 19 ml X min -1 X kg fetus -1 near exhaustion during prolonged (40 min) exercise at 70% maximal oxygen consumption. Maternal blood volume decreased 14% (P less than 0.01) from 67.5 +/- 3.7 to 57.8 +/- 3.6 ml X kg -1 during this exercise period, with a 20% decrease in plasma volume without a change in red cell volume. We conclude that uterine blood flow decreases during maternal exercise. However, hemoconcentration helps to maintain a relatively constant oxygen delivery to the uterus.  相似文献
