The longitudinal and lateral transport of 2, 4-D-[1-14C] hasbeen studied in 6-mm horizontally disposed segments of the hypocotylof Helianthun annuus. The technique involved the asymmetricapplication of the auxin in agar donor blocks to either theupper or lower halves of the cut surface of the segments andthe measurement of 14C accumulating with time in split receiverblocks on the upper and lower halves of the cut surfaces atthe opposite ends. A highly significant effect of gravity inducinga polar migration of 2, 4-D to the lower side has been establishedand represents, under optimal conditions, about 10 per centof the total 2, 4-D in transit. This lateral polar movementshows a tendency to saturate at higher donor concentrations(5 mg/1) and is affected by temperature in precisely the sameway as the basipetal longitudinal transport. The optimal flux(intensity of transport) occurs at about 35 °C or slightlyabove, and above 40 °C both transport systems are seriouslyimpaired. There is some evidence that gravity increases thevelocity of 2, 4-D transport in the lowermost tissues of thehorizontal hypocotyl but does not affect the transport intensity.  相似文献

The polar transport of indol-3yl-acetic acid (IAA-2-14C) instem explants and decapitated shoots of tumour-prone Nicotianahybrids (2n, 3n, and 4n) was compared with that in the normal,non-tumorous parent species N. glauca and N. langsdorffii. Thetotal uptake of the auxin from donor blocks was greatest inthe hybrids and N. glauca. The velocity of the basipetal movementof IAA-14C was the same in all species tested, i.e. 8 mm/h.The transport capacity for the hormone, however, was decreasedin the three tumour-prone hybrids. Gas chromatography showedthat between 70 and 90 per cent of the transported auxin waspresent in the form of IAA, between 10 and 30 per cent in theform of indol-3yl-aldehyde (IAld). The basipetal transport exceeded the acropetal transport inyoung (third) intemodes of all plants studied, whereas in olderstem segments (tenth intenodes) the reverse was found. The polarity of auxin transport was less well expressed in thetumorous hybrids. Blocking the active transport by pre-treatment of stem cuttingswith 2,4-dinitrophenol (2,4-DNP) caused a drastic reductionin the polar IAA-14C movement; in all plants tested the auxintransport was reduced to the same low level. The accumulation of auxin at the base of cuttings was higherin N. glauca and the 2n hybrid than in N. langsdorffii, i.e.about seven times higher after 1-h and three times higher after12-h transport experiments. The release of 14C from the cuttinginto an agar receiver block, however, was markedly reduced inthe 2n hybrid, whereas in N. glauca the labelled substancesmoved more freely into the receiver blocks. Differences in the capacity for the accumulation and the releaseof IAA-14C in hybrid and N. glauca stem tissues were studiedusing decapitated greenhouse plants wounded by incision abovethe fourth internode. Accumulation of the auxin occurred onlyabove the wound-cut in hybrid plants. This observation is consistentwith the view that tumour formation on hybrid stems occurs atsites of wounding. Our data suggest an elevated auxin levelto be present during tumour initiation at these sites. These results on polar transport and accumulation of IAA-14Cin tumorous Nicotiana plants together with our previous dataon various endogenous auxins suggest that the induction of neoplasticgrowth in tobacco plants is correlated with increased auxinlevels and an accumulation of the hormone at sites of wounding.  相似文献

The effect of morphactin (methyl 2-chloro-9-hydroxyfluorene-9-carboxylate)on basipetal transport of auxin (Indol-3-ylacetic acid-2-14C)was studied in bean (Phaseolus vulgaris) hypocotyl with thedonor-receiver block method. Morphactin (5 x 10–6m) reduced IAA (5 x 10–6m) transportintensity by an average of 83 per cent and auxin transport capacityby 90 per cent, but transport velocity was not affected. Morphactin did not inhibit uptake of IAA into hypocotyl tissue,but it did prevent transfer of IAA from the tissue into receiverblocks. Chromatographic analysis of the tissue after 4 h IAA-2-14Ctransport showed that 54 per cent of the total activity wasin the form of IAA in the control and 42 per cent in the morphactintreated tissue. No difference was found in the rate of decarboxylationof IAA-1-14C between control and morphactin treated tissue sections.Nor could any difference between control and morphactin be shownin the radioactivity associated with a TCA ppt fraction. Ina study of the transportable auxin pool, morphactin decreasedthe size of the pool and increased the half-life of decay ofauxin transport from 1•22 h to 3•85 h. In a kineticanalysis of the reversal of morphactin (5 x 10–6m) inhibitionby increasing concentration of IAA-2-14C (5 x 10–6m to2 x 10–5m), it was shown that IAA transport resemblesMichaelis-Menten enzyme reaction kinetics, and that inhibitionby morphactin fitted a ‘mixed type’ model. IAA hada dissociation constant of 8•5 x 10–6m and morphactinthat of 4•3 x 10–7m with a Km for the transport processof 8•5 x 10–6m.  相似文献

Abscisic acid (ABA) at low concentrations brings about the formationof turions (dormant fronds) in Lemna polyrhiza within 3 to 5days after application. The incorporation of 3H-thymidine intoDNA, separated by polyacrylamide-gel electrophoresis, is inhibitedby 80 to 90 per cent within 1 to 3 h of ABA application. Theincorporation of 14C-orotate and 32P into RNA is not inhibiteduntil 3 to 9 h after ABA application, but 70 per cent inhibitionis reached after 24 h on 10–5 M ABA. There is little inhibitionof 14C-leucine incorporation into protein until 2 to 3 daysafter application of ABA. The capacity of nitrate to inducenitrate reductase in cultures previously grown on nitrate-freemedium is not affected by ABA even up to 3 days after application.The results are discussed in relation to the mode of actionof ABA.  相似文献

Azetidine-2-carboxylic acid (AZC), which occurs naturally inLiliaceous plants, is reported to be a proline (pro) analoguePlant cell walls contain ‘extensin’, which is richin hydroxyproline (hyp). Peptidyl hyp arises through hydroxylationof peptidyl pro followed by glycosylation (arabinose attachment)of hyp Because AZC replaces peptidyl prolyl residues, it maybe a useful tool for evaluating the significance of hyp-o-arabinoselinkages in cell elongation. Therefore, we determined the effectof AZC on [14C]pro uptake, incorporation and conversion to wall-bound[14C]hyp in relation to elongation of lily pollen tubes whosewalls consist, in part, of hyp-containing glycopeptides TheAZC suppressed pollen germination 9–42 per cent (1–10mM) and subsequent tube elongation 40–54 per cent (0·1–1mM without affecting respiration In contrast, similar hyp concentrationswere without effect on tube elongation Whereas uptake of [14C]prowas 16·5–6·2 per cent of the control at0·1–1 mM AZC, [14C]leucine uptake was 85–25per cent of the control. Light microscope radioautography revealedfewer silver grains over tubes elongated in 0·1–1mM AZC than in its absence. Incorporation of [14C]pro into tnchloroaceticacid (TCA)-precipitable cytoplasm was reduced by only 10 percent at 0·01–1 mM but 43 per cent at 10 mM AZCGel filtration of cytoplasm from pollen germinated without AZCbut with [14C]pro resulted in labelled void volume (V) and threeretarded peaks (RI–III) Incorporation into V and RI wasinhibited at both 0·01 and 1 mM AZC These AZC concentrationsreduced conversion of [14C]pro to wall-bound hyp by 20 percent However, total incorporation of [14C]pro into salt-water-purifiedwall fractions was suppressed 47–53 per cent (0·1–1mM AZC). Guttation was used as a non-destructive way to study the flowof water and mineral ions from the roots and compared with parallelmeasurements of root exudation. Guttation of the leaves of barley seedlings depends on age andon the culture solution. Best rates of guttation were obtainedwith the primary leaves of 6- to 7-day-old seedlings grown onfull mineral nutrient solution. The growing leaf tissue becomessaturated with K+ below 1.5 mM K+ in the medium, whereas K+concentration in the guttated fluid still increases furtheras K+ concentration in the medium is raised. At 3 mM K+ averagevalues of guttation were 1.4–2.4 mm3 h–1 per plantwith a K+ concentration of 10–20 mM; for exuding plantsthe flow was 4.2–7.6 mm3 h–1 per plant and K+ concentration35–55 mM. Abscisic acid (ABA) at 10–6 to 10–4 M 0–2h after addition to the root medium increased volume flow ofguttation and exudation and the amount of K+ exported. Threeh after addition of ABA both volume and amount of K+ were reduced.There was an ABA-dependent increase in water permeability (Lp)of exuding roots shortly after ABA addition. Later Lp was decreasedby 35 per cent and salt export by 60 per cent suggesting aneffect of ABA on salt transport to the xylem apart from itseffect on Lp. Benzyladenine (5 x 10–8 to 10–5 M)and kinetin (5 x 10–6 M) progressively reduced volumeflow and K+ export in guttation and exudation and reduced Lp. Guttation showed a qualitatively similar response to phytohormonesas found here and elsewhere using exuding roots. Activity curves are determined for gibberellins A1 to A0 bythe Avena first-leaf bioassay method. Gibberellins A1, A4 andA5 can be detected at 10-11 or 10-10 g/ml and give optimum activityof approximately 230 per cent elongation (water controls = 100per cent). Gibberellins A2A3, and A9 can be detected at 10-3g/mland give optimum activity of approximately 200 per cent. GibberellinsA6 and A7 can be detected at 10-5g/ml; GA7 gives optimum activityof around 190 per cent. All the gibberellins except GA8 canbe detected by this bioassay method after chromatography inn-butanol: 1.5 N ammonia (3: 1) and benzene: acetic acid: water(4: 2: 1) when applied to the paper at concentrations from O.Ito µg. Embryogenic cell suspension cultures were established from calliderived from young leaves of sugarcane (Saccharum officinarumL.) by placing them in liquid medium containing 5 per cent coconutwater (CW), 2–3 mg 1–1 2, 4-D and 500 mg 1–1casein hydrolysate (CH). The cultures were maintained by transferring2.5–5.0 ml of the suspension to 35 ml of fresh mediumevery 4–5 days. Organized structures resembling the earlystages of embryogeny were formed when 2, 4-D in the medium waslowered (0.1–1.0 mg 1–1) but these did not developbeyond the globular or early scutellar stages. High levels ofsucrose (6–10 per cent) promoted the formation of proembryoids.Plating of the suspension on MS agar medium supplemented with0.25–2.0 mg 1–1 2, 4-D, 5 per cent CW, 500 mg 1–1CH, with or without activated charcoal, resulted in the formationof embryogenic calli. A large number of embryoids were formedin media containing lower 2, 4-D concentrations. Transfer ofembryoids to half-strength MS medium with 6 per cent sucroseestablished plantlets which were successfully transferred tosoil. A long-term, steady-state 13CO2 assimilation system at a constantCO2 concentration with a constant 13C abundance was designedand applied to quantitative investigations on the allocationof photoassimilated carbon in nodulated soya bean (Glycine maxL.) plants. The CO2 concentration in the assimilation chamberand its 13C abundance were maintained constant with relativevariances of less than ±0.5 per cent during an 8-h assimilationperiod. At the termination of 8-h 13CO2 assimilation by plantsat early flowering stage, the currently assimilated carbon relativeto total tissue carbon (measured by the degree of isotopic saturation)were for young leaves (including flower buds), 13.9 per cent;mature leaves, 15.7 per cent; stems+petioles, 5.9 per cent;roots, 5.4 per cent and nodules, 6.9 per cent, 48 h after theend of the 13CO2 assimilation period, they were 12.3, 7.5, 7.4,6.8 and 6.1 per cent, respectively. The treatment with a highconcentration of nitrate in the nutrient media significantlydecreased the allocation of 13C into nodules. Experiments on13CO2 assimilation by plants at the pod-filling stage were alsoconducted. Labelling by 13C was weaker than at the early floweringstage, but an intense accumulation of 13C into reproductiveorgans was observed. Brassica oleracea pollen was applied to a basic medium of 1.5per cent agar and 15 per cent sucrose to which flavanoids wereadded at three concentrations. Two types of agar were used;with agar 1, quercetin at a concentration of 0.5 x 10–3per cent gave an increase in percentage germinating grains.With agar 2, an increase in germination occurred with kaempferoland naringin at concentrations of 0.5 x 10–3 and 0.5 x10–1 per cent respectively. Increase in pollen tube lengthoccurred with agar 2 and quercetin at a concentration of 0.5x 10–3 per cent. The stigma tissue of B. oleracea contains at least three andthe pollen at least one glycoside of quercetin. The sugars inthe glycosides were not identified. Pollen germination and pollentube extension were not stimulated exclusively by the flavanoidspresent in the stigma. The flavanoid composition of the stigmadid not vary amongst five different S-allele genotypes, indicatingthat flavanoids are probably not directly involved in the incompatibilityreaction of B. oleracea.  相似文献

Aqueous foliar sprays of N-dimethylaminosuccinamic acid (daminozide)at 2000 p.p.m. and gibberellic acid (GA) at 100 p.p.m. wereapplied 45, 59, 82 and 100 days after sowing to Chantenay carrotswith population densities of 244, 495 and 883 plants m–2.The plants were harvested on ten approximately weekly occasions;fresh weights were determined and d. wt estimates were obtainedfor the separated shoots (s) and roots (r). Allometric linearregressions of the logarithm of s on that of r at each harvestseparately, clearly showed that GA always increased shoot: rootratio and reduced root yield (by approximately 35 per cent)but could sometimes also increase whole-plant weight. Daminozideincreased root yield (by approximately 7 per cent from 80 tonnesha–1) and tended to have effects opposite to those ofGA. Glucose, either uniformly labelled with14C, or specificallylabelled in the I, 2, or 6 position, was added to C. vulgaris.Radio-active carbon dioxide was produced initially ten timesfaster from glucose-I-14C than from glucose-6-14C. This differencewas found with carbohydrate-starved cultures, exponentiallygrowing cultures, and cultures assimilating ammonia or nitraterapidly. A similar difference was also found with C. pyrenoidosaand Ankistrodesmus. 37 per cent. of the 14C added as glucose-1-14Cto exponentially growing cells was recovered as carbon dioxidebut generally the recovery was less than this. Only 5 per cent.of 14C added as glucose-6-14C was recovered as carbon dioxide.The specific activity of the carbon dioxide produced was considerablylower than that of the carbon in the added glucose.  相似文献

The effects of some amino acid analogues (o-, m- and p-fluorophenylalanineand azetidine-2-carboxylic acid) on uptake of42K and 45Ca intothe roots and transport to the shoots of whole wheat and mungbean seedlings were measured. The effect of each analogue oneither K+ or Ca2+ movement could be placed into one of fourcategories: (1) No effect on either ion uptake or transport;(2) No effect on ion uptake, but a reduction in transport; (3)Similar reductions in ion uptake and transport; (4) A relativelygreater reduction in ion transport than in uptake. At leasttwo independent sites of protein involvement in ion movementwere required to account for all four types of analogue effectobserved; one site of protein involvement was probably at theplasmalemma of root cortex cells and the second site, involvinga protein that turned over more quickly, was within the stele.Some evidence was found that Ca2+ transport is a passive process.Light did not stimulate uptake. Incorporation of labelled mevalonate into abscisic acid (ABA)has been demonstrated in the cotyledons of mature avocado seeds,embryos and endosperms of developing wheat seeds, and avocadostems. The increase in ABA concentration on wilting parallelsthe increased incorporation of [2–14C)mevalonate intoABA in avocado leaves and stems, suggesting that the increasein ABA content occurs by synthesis rather than by release froma stored precursor. Incorporation of [2–14C]mevalonateby avocado mesocarp segments is unaffected by an 18 per centwater loss. The ABA content of roots was hardly affected bya 30 per cent water loss, indicating that the wilt-activatedmechanism is not fully operative in these tissues. Submerged Ceratophyllum plants and submerged parts of Callitricheshoots show a twofold increase in ABA content on wilting whereasthe aerial rosettes of the latter plant show a sixfold increase.This suggests that the occurrence of the wilt-induced mechanismis affected by previous growth conditions as well as by themorphology of the tissue.  相似文献

Chemical Composition of Bleeding Xylem Sap from Kiwifruit Vines   总被引:5,自引:0,他引:5  
A study of the chemical composition and charge balance was madeof bleeding xylem sap collected from excised one-year-old extensionshoots of healthy, Mn-deficient, Mn-toxic and Zn-deficient kiwifruitvines (Actinidia deliciosa (A. Chev.) C. F. Liang et A. R. Ferguson)immediately prior to leafburst. The exudates were analysed formacronutrient cations and anions, trace elements, amino acids,organic acids and sugars. Major charged species measured wereCa (13.3 mM), K (8.9 mM), Mg (5.6 mM), malate (12.5 mM) andphosphate (5.8 mM). Glutamine (12 mM) was the predominant Ncarrier identified, accounting for 58 per cent of the totalN followed by NO2-N (4.5 per cent), NH4+-N (3.5 per cent)and arginine-N (2.9 per cent). Approximately 22 per cent ofthe N was in a hydrolysable proteinaceous fraction comprisingmainly glutamine and glutamate. Eighteen free proteinaceousamino acids were idetified in sap, the most abundant being glutamine,glutamic acid, valine, isoleucine and phenylalanine. Computersimulation of the chemical composition predicted that in additionto hydrated cations, ion pairs formed between inorganic components(SO42–, HPO42–, H2PO4) and cations (Ca2+,Mg2+, Mn2+), plus metal-organic ligand complexes (Ca Malate,Zn Malate, FeCit, CuHis, CuGln) are important species involvedin translocation. The solubility product of hydroxyapatite wasexceeded in all exudates although in vitro precipitation wasnot observed. To achieve electroneutrality with the componentsmeasured, however, formation of precipitate precursors (hydroxyapatitenuclei) had to be assumed. Irregularities in Mn nutrition (butnot Zn) were clearly indicated by the elemental compositionof exudate suggesting the use of sap analysis as a possiblepre-season indicator of nutritional status for this species. Selenate and selenite transport through tomato root systemswere followed for periods up to 4 h after removal of the planttops, using 75Se as a tracer. With selenate, 75Se concentrations in the xylem exudate were6 to 13 times higher than in the external solution, and chromatographicanalysis showed that the selenium was transported as inorganicselenate ( ). With selenite, 75Se concentrations in the exudate were alwayslower than in the external solution. Analyses of exudate samplesshowed that negligible amounts of selenium were transportedas inorganic selenite ( except at very high external selenite concentrations (500 ?M), when up to 7 percent was transported as selenite. Most of the selenium transportin selenite-fed plants was as selenate or as an unknown seleniumcompound, the relative proportions of these two forms varyingboth with time and with external selenite concentration. Additionof a 5-fold excess of sulphate over selenite had no detectableeffect on the concentrations of selenate in the exudate, butcaused substantial decreases in the maximum concentrations ofboth total selenium (c. 47 per cent decrease) and the unknownselenium compound (c. 69 per cent decrease). Addition of a 5-foldexcess of sulphite decreased the concentration of the unknown(c. 39 per cent) but caused a large (2.7-fold) increase in themaximum total selenium concentration in the exudate and a 7.9-foldincrease in the maximum concentration of selenate. The resultssuggest metabolic involvement in the uptake and long distancetransport of solenium supplied as selenite, despite lower 75Seconcentrations in the xylem exudate than in the external solution.An attempt is made to incorporate the new and existing informationinto a selenium transport model.  相似文献

Nuclear volumes and cell areas were determined for seven regionsof the meristem of roots of Zea mays. Roots were fixed in 10per cent neutral buffered formalin, in 3 per cent glutaraldehydeor in acetic acid/alcohol; they were prepared as sections oralls were teased apart. Mean volumes of interphase nuclei weresimilar in all regions of the root except the vascular tissueof the stele. Mean nuclear volumes and the overall range ofvolumes were similar in sub-populations of cells with differentproportions of G1, S and G2 cells, e.g. in row I of root capinitials, whose cells lack a G1 phase, and in quiescent centrecells, which are mainly in G1. Nuclear volume does not appearto be closely correlated with DNA content. Nuclear volumes covereda 6 to 12-fold range within a meristem and even within specificregions, in which cells are part of the same cell lineages,there was a 4- to 9-fold range. Nuclear volumes were comparedin sister cells in rows I and II of the root cap initials. In10 per cent of the pairs, sister nuclei had identical volumes;the other pain had different volumes and mean difference was68 µm3. Mechanisms by which this variability could begenerated are discussed, particularly asymmetry, at mitoses,of factors that regulate nuclear growth. Indol-3yl-acetic acid (IAA) applied to sterns of Phaseolus vulgarisseedlings, decapitated above primary leaves, enhanced the mobilizationof 14C-metabolites to the treated stumps and this effect wasapparent within 3–6 h of applying the hormone. More than90 per cent of the total 14C-activity transported to the stumpswas detected in the alcohol-soluble extracts. In all treatments,less than 5 per cent of the 14C-photosynthate exported fromthe primary leaves was translocated upwards. Accumulation of14C-activity was also increased when the IAA was applied laterallyto intact internodes. This effect was obtained when 14C wassupplied either above or below the point of hormone application.By selective heat girdling, it was shown that the auxin affected14C transport when either the root ‘sink’ was removedor transpiratory flow of water through the treated internodewas maintained. Decapitated stems treated with plain lanolinfor 3 d were found to retain their responsiveness to auxin interms of enhanced metabolite transport. Heat-girdling experimentsand estimates of 14C transport velocity suggested that mostof the 14C movement was restricted to the phloem of treatedstumps. Similar effects of IAA on a transport in excised stemsegments of Phaseolus vulgaris were observed.  相似文献

The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 7–14-dayperiod of vegetative growth varied between 10 and 17 mg CO2g–1 (dry weight) h–1. This mean value consistedof two components: a specific root respiration rate of 6–9mg CO2 g–1 h–1 and a specific nodule respirationrate of 22–46 mg CO2 g–1 h–1. Nodule respirationaccounted for 42–70 per cent of nodulated root respiration;nodule weight accounted for 12–40 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 20–30per cent greater than the equivalent rates of roots from nodulatedplants. The measured respiratory effluxes are discussed in thecontext of nitrogen nitrogen fixation, nitrate assimilation. The growth of lucerne var. Europe was examined in the fieldduring 1976. The annual dry matter production of unirrigatedlucerne during 1976, with no nitrogen fertilizer application,was 82.5 per cent greater than unirrigated S.24 perennial ryegrass,with a nitrogen application of 384 kg ha–1. The mean aboveground growth rate of lucerne was 7.3 g DM m–2 day–1between March and early June, of which stem material contributeda maximum of 76.5 per cent. Significant losses of leaves andstems occurred from the end of April, indicating a loss of potentialforage material. Nitrogen analyses of the above ground crop suggested that in56 days lucerne yielded 10.7 per cent more nitrogen than didS.24 annually with a nitrogen fertilizer addition of 280 kgha–1.
