In experiments on outbred female rats the influence was studied and compared of two representatives of endogenous opioids beta-endorphine and the analogue of leu-enkephalin dalargin on the processes of learning and memory in normal conditions and at the change of functional state of serotoninergic system of the brain. Parallel, the influence was studied of neuropeptides on the content of serotonin (5-OT) and its metabolite--5-oxyindolacetic acid in various areas of the brain in control and at the 5-OT redundancy. Conditioned reflexes (CRs) were used of two-way avoidance and defensive CRs. It has been established that administration of neuropeptides to intact animals influences in different directions the elaboration of the CR of two-way avoidance and maze defensive CR, but also worsens their preservation. Redundancy of 5-OT in the brain modifies behavioural effects of beta-endorphine and dalargin manifested in appearance of new effect and elimination and change of direction of the effects observed in the intact animals. Redundancy of 5-OT in the brain changes metabolic effects of beta-endorphine and particularly of dalargin. The obtained data testify to a dependence of the effects of beta-endorphine and dalargin on the functional state of 5-OT-ergic system.  相似文献

Changes of antioxidant activity of dalargin in the liver after naloxone (100 micrograms/kg) administration were examined in experiment on 144 rats with cholestasis. It was found that dalargin inhibited the activity of xanthine oxidase by 32-37% in different time periods after the injection. Dalargin and naloxone, when used in combination, had no effect on the enzyme activity. Glutathione-S-transferase activity rose by 38.0% and 21.8% on hour 1 and 3 after the injection, respectively, while simultaneous injection of dalargin and naloxone induced no changes in the enzyme activity after 1 hour, though decreased it by 36.8% and 26.4% on hour 3 and 5, respectively. Dalargin inhibited lipid peroxidation by 29-35%, simultaneous injection of dalargin and naloxone raised lipid peroxidation by 109.2%, 80.7% and 25.7% after 1, 3 and 5 hours, respectively. Dalargin injection elucidated a marked tendency to lowering of blood release of the liver-specific enzymes histidase and urokaninase in line with enhancement of their activity in the liver. A combined injection of dalargin and naloxone promoted high release of histidase and urokaninase in blood and did not change histidase activity in the liver in all cases. Urokanidase activity elevated in 5 hours. It was noticed that dalargin raised leu-enkephalin levels in the liver 3.5-fold 1 h after the injection. The reduced dalargin antioxidant effect coupled with naloxone pretreatment demonstrated indirect action of the neuropeptide on the liver via neuron receptors of the liver.  相似文献

The aim of the work was to study the involvement in the process of learning of synapses between the perforant path and granular cells of the hippocampal dentate fascia for formation of conditioned reflex (CR) of active avoidance elaborated in response to electrostimulation of the perforant path. For solving this task, the property was used of the neurotoxin colchicine in a definite concentration to abolish selectively the fascia cells. One month after colchicine injection, destruction of granular cells was observed, disappearance of the dentate fascia focal evoked potentials and considerable deterioration of elaboration of active avoidance CRs. Administration of saline solution in control experiments did not elicit such changes. The obtained results show the decisive role of entorhinal input to the hippocampus in the used learning model.  相似文献

Bioavailability and metabolism of a peptide drug Dalargin with a chemical structure Tyr-D-Ala-Gly-Phe-Leu-Arg have been examined. Dalargin is applied for the treatment of gastric and duodenal ulcers. Bioavailability was estimated following intramuscular (i/m) and intranasal (i/n) routes of administration of 3H-dalargin in anesthetized dogs. The highest dalargin concentration was achieved about 10 min after i/m and i/n administration. Absolute dalargin bioavailability was 15% and 8%, while its elimination half-life was 23.2 min and 21.3 min, respectively. Tyrosine, N-terminated tetra- and pentapeptides were the main metabolites detected in the blood. The intranasal route of dalargin administration is concluded to be possible in the clinical practice.  相似文献

We studied the effect of synthetic analogue of opioid peptides dalargin on kinetics of cell population of corneal epithelium during stress. It was found out that dalargin introduction before stress decreased the level of pathological mitoses induced by the acute stress action in white rats early in the morning. Dalargin prevented the activation of the proliferative process after the repeated stress activation, made stress milder. The stress induced vertical migration of the cells of the corneal epithelium normalized by dalargin. We believe these results show that dalargin has a good protective effect.  相似文献

After dalargin treatment of fish eggs (at the stage of swollen egg envelopes) and on juveniles (at the stage of early meiotic oocyte appearance in gonads) DNA content is observed to raise up to 85% and 66%, respectively in yearlings and up to 20% and 23% respectively in second-year fish as compared to the control ones. Dalargin also influences the exchange of muscle RNA in yearlings and second-year rainbow-trout. Dalargin effect is higher when peptide influences at the very onset of organism differentiation.  相似文献

Pharmacokinetics of dalargin, an opioid hexapeptide, was investigated on 7 males by two approaches. Dalargin radioimmunoassay was performed using a highly specific antiserum reacting only with the whole molecule. In radioreceptor assay lyophilized rat brain membranes containing opiate receptors were used. 2-6 min after intravenous introduction of 1-10 mg dalargin, immunoreactive dalargin blood concentration was lower than 0.5 ng/ml. The results of radioreceptor assay were presented as a biexponential curve with a fast main phase of activity changes (90%, characteristic time 1.5-5.0 min) and a slow "clearance" phase (10% of the substance, characteristic time 85-200 min). Prolonged presence of receptor-active substances in the blood can be attributed to the products of dalargin degeneration, namely its N-terminal penta- and tetrapeptides.  相似文献

On the basis of the idea of the important role of neurotransmitter systems in realization of neuropeptide effects, the participation was studied of the monoaminergic systems in the mechanisms of the ACTH analogue influence on the processes of learning and memory in control animals and animals with a changed functional state of the monoaminergic systems. In parallel the influence was studied of the ACTH analogue on the content of the endogenic monoamines in various brain structures of rats. It has been shown that administration of the ACTH analogue in a dose of 10 mcg affects the elaboration and preservation of conditioned reflexes (CRs) of passive avoidance, CRs of two-side avoidance and labyrinth CRs only in conditions of changed functional state of the monoaminergic systems. Amnesia, usually elicited by 5-oxytryptophane and disulfiram is prevented by administration of the ACTH analogue. Administration of the ACTH analogue is accompanied by the intensification of serotonine metabolism in the midbrain and medulla and by an increase of noradrenaline content in the hypothlamus.  相似文献

Content was compared of noradrenaline (NA) and serotonine (5-OT) in the right and left halves of the rats brain in norm, at elaboration of defensive conditioned reflexes of two-ways avoidance (CRTWA) and at administration of neuropeptides influencing the learning and memory--dezglycilargininvasopressin (DG-AVP), ACTH4-7 pro-gli-pro and dalargin. The conducted studies showed that in control animals the content of NA in the cortex of the right hemisphere was significantly higher than in the cortex of the left one. For the content of 5-OT in symmetric brain parts no significant differences were revealed. Under the elaboration of CRTWA the asymmetry of NA content was not eliminated. Systemic administration of DG-AVP, ACTG4-7 pro-gli-pro and dalargin practically did not change the content of 5-OT, but reduced the content of NA in the cortex and the rest of the brain, and the content of NA in the right and left cortex was equalized. The obtained data point to the asymmetric character of neuropeptides action and to greater resistance of 5-OT-ergic brain system to functional load and to administration of peptides in comparison with NA-ergic system.  相似文献

The action of dalargin, synthetic analogue of leuenkephaline, on stress-induced changes in 5'-nucleotidase activity and endogenous hydrocortisone levels was investigated in mice. It was found that there is a direct relation between the activity of 5'-nucleotidase and the level of hydrocortisone in CBA mice. For C57Bl/6 mice the relation was inverse. Dalargin is able to change the dependence from direct to opposite in CBA mice.  相似文献

Influence was studied of dalargin--synthetic analogue of enkephalin administered in early postnatal period to young rats with intrauterine ethanol injection, on their behaviour and conditioned activity. Parameters were established of spontaneous motor activity, behaviour in the open field, acquisition and preservation of conditioned reflexes of passive and active avoidance in experimental and intact rats, injected with dalargin. A reduction of sensitivity to corasol convulsive effect was observed under the influence of dalargin.  相似文献

Long-term influence was studied of the acute hypoxic hypoxia seance on rats behaviour in situation of elaboration of the conditioned reaction of active avoidance of electric shocks in shuttle chamber. It was found that in 2.5-3 months after the hypoxia seance, the experimental animals significantly differed from the intact controls by dynamics of CR elaboration (rats which had hypoxia were ahead of the control ones) and by distribution of the conditioned reactions latencies (for experimental animals this distribution was shifted to minor values). The character of these behavioural shifts coincided with that observed in the group of rats with local unilateral hippocampus lesion. The obtained results and numerous data presented in literature on the influence of the hippocampus lesion on animals shuttle avoidance learning, allow to conclude that the seance of hypoxic hypoxia leads to the disturbance of the hippocampus function. This conclusion conforms to the data on diffusive death of the hippocampal and neocortical neurones as a result of hypoxia action.  相似文献

The influence of scopolamine on elaboration and maintenance of conditioned reflexes of two-way avoidance was studied in rats under conditions of excess and deficit of serotonin in the brain. Administration of scopolamine to intact rats accelerated conditioning and did not prevent fixation of the reflex. Administration of scopolamine to animals with a lowered level of serotonin in the brain (by means of para-chlorophenylalanine) impaired conditioning and induced amnesia. In animals with ablated raphe nuclei, the same dose of scopolamine did not prevent elaboration and maintenance of conditioned reflexes. Accumulation of scopolamine in the brain by means of 5-oxytryptophan abolished acceleration of conditioning, which is specific for scopolamine, and affected the preservation of the reflexes. Against the background of the action of iprozid, scopolamine impaired the conditioning and tended to deteriorate the maintenance of conditioned reflexes. It is assumed that the serotoninergic system exerts a modulating influence on the activity of the brain cholinergic and cholinoreactive mechanisms.  相似文献

In experiments on three dogs there was shown that testing electrostimulation of the lateral hypothalamus reproduced the motor reaction which is a signal stimulus at elaboration of classic alimentary conditioned reflexes (CRs) and did not reproduce it at elaboration of classic defensive CR. Testing electrostimulation of medial parts of the hypothalamus reproduced, as LH electrostimulation the "signal" motor reaction, but in less percentage of cases, during elaboration of classic alimentary CRs and did not reproduce it at elaboration of classic defensive CRs. The reproduction of the signal motor reaction at LH electrostimulation is connected with activation of backward conditioned connection from motivation structures of the hypothalamus to representation of the signal stimulus in the motor cortex.  相似文献

Effect of dalargin, an opioid peptide (a synthetic analogue of Leu-enkephalin), on proliferation and intensity of DNA synthesis of C6 glioma cells was studied. Specific conditions of cultivation were selected, with a low control value of proliferation, which permitted to assess growth-stimulating effect of the peptide. Growth curves were plotted to assess peptide activity, which demonstrated that reaction was a many-phase process: a significant increase in cell number under peptide effect was observed only at the beginning of the logarithm phase and at the beginning of the prestationary phase of the growth curve. Cell number increased on average by 25-27% in the presence of dalargin as compared to control. Reaction of glioma DNA synthesis to dalargin also demonstrates complexity of the process: the peptide changes DNA synthesis, but as a rule, this process has a three-phase character and is not directly associated with the duration of cultivation in the presence of dalargin. Effect of naloxone, an opiate receptor blocker, was analysed to assess the receptor mechanism. It was found that reaction for naloxone and for combined effect of naloxone and dalargin was not the same.  相似文献

Activity of 144 neurones of the dorsal part of the rabbits hippocamp was recorded during elaboration of motor conditioned reflex to time. Chronic amphetamine intoxication lowered the ability of hippocampal neurones to form conditioned reactions in response to pairings of sound stimuli with electrocutaneous reinforcement and fully suppressed mechanisms of reproduction by cells of engrams of previous pairings in series of their omissions Single administration of haloperidol to intact animals somewhat increased the number of neurones reacting to the pairing and their omissions in conditioned reflex to time without significantly influencing the intensity and dynamics of reproduction of endogenous cellular reactions in the series of consecutive omissions of pairing. Haloperidol administration during amphetamine intoxication elicited shifts towards normalization of conditioned activity of neurones, eliminating the suppressing action of amphetamine on mechanisms of reproduction of engrams of combined stimuli. Such "therapeutic" effect of haloperidol in many cases did not depend on the character of its psychotropic action. The properties of amphetamine and haloperidol action on the cells of the hippocamp are discussed as compared to their action on the neurones of other brain structures, previously studied in an analogous experimental situation.  相似文献

Intraperitoneal injection of cadmium chloride (4 mg/kg) to rats before elaboration or reproduction of two-way avoidance conditioned reflex (TACR) disturbs both these processes. Deterioration of elaboration does not affect connections fixation and their subsequent reproduction. Injection of the substance before the elaboration of passive avoidance conditioned reflex (PACR) depresses elaboration and consolidation. Injection of cadmium chloride before testing of PACR preservation does not influence the processes of engrams reproduction. The effect was studied of arginine-vasopressin (AVP) intraperitoneal administration in a doze of 0.001 mg/kg on elaboration of conditioned reaction (CR) of active avoidance in rats. Estimation of reaction dynamics by dynamic delta index allowed to increase the informational value of the experimental data with taking into account the direction of reaction shift and individual characteristic of the animal. AVP improved the elaboration of active avoidance CR, lowered variability of individual learning ability and did not influence the change of the number of intersignal reactions and short-latency escapes in the process of learning. It is suggested that the main site of AVP effect is the animal's reaction to conditioned sound stimulus.  相似文献

In experiments on 250 Wistar female rats the influence was studied of a single maximum and submaximum physical loads on the emotional behaviour and the state of conditioned activity. It has been established, that as a result of maximum possible run of the animals depression takes place of orienting investigatory activity in conditions of the "open field" and disturbance of elaboration and reproduction of the conditioned reflex of two-way avoidance. Normalization of these parameters took place in 48 hours after the maximum load action. Analogous effect was observed in the period of aftereffect of the submaximum physical load, characterized by lowering of the level of behavioural activity in the "open field", disturbance of the elaboration of two-way avoidance as a result of presumable increase of functional tonus of the brain inhibitory systems.  相似文献

As a result of intravenous administration of 100 mg/kg of 5-oxytryptophane (5-OTP) to mice, the serotonin content in the brain rises in ten minutes by 35 to 37 per cent, and in an hour, by 58 to 60 per cent, and declines in a day to its initial level. In case of elaboration of passive avoidance reflexes (PAR) an hour after 5-OTP injection, their preservation considerably decreases in a day's time, while 5-OTP administration immediately following PARs elaboration does not affect their preservation. However, 5-OTP injected after PARs elaboration to animals subjected to preliminary cooling, sharply deteriorates their preservation. These facts in conjunction with the previously obtained date attest that excess of serotonin in the brain has a negative effect on the earliest stages of consolidation. When the animals are cooled and the brain temperature is reduced, the consolidation processes are retarded; as a result, the temporary connections prove to be still vulnerable to the action of serotonin excess set up after PAR elaboration.  相似文献
