In the oomycete Saprolegnia diclina, the number of oogonia formedper unit area of culture decreased as the Ca2+ concentrationin the culture medium was increased. The calcium ionophore A23187 [GenBank] decreased the number of oogonia formed and increased the numberof gemmae. In the presence of 0·08 µM Ca2+, oogoniumand oospore abortion rates were increased by A23187 [GenBank] with a peakeffect at 0·2 nM. As CaCl2 concentration in the growth medium was reduced from27.4 to 0.107 µM the range of oospore number per oogoniumin Saprolegnia terrestris increased from 1 to 8 to 1 to 25 ormore, oospore size decreased and the precision of correlationbetween oospore number per oogonium and oogonial diameter decreased,but colony d. wt was not affected. The threshold CaCl2concentrationrequired to give the lower range of oospore number per oogoniumwas independent of the concentration of other mineral nutrientsin the growth medium. The number of oogonia formed per unitarea of colony was affected by the concentration both of CaCl2and of other mineral nutrients. Oospores formed in 0.107 µMCaCl2 medium were often multinucleate but otherwise of normalappearance. The central vacuole system of oogonia of Saprolegnia terrestrisfrom Ca2+-sufficient cultures was fully enlarged prior to theformation of oosphere initials, which did not involve cleavagevesicles. In oogonia with fully-enlarged central vacuole systems,subsidiary vacuoles at the periphery of the system sometimescontained dense bodies, and dense-body profiles were sometimespresent within sections of the central vacuole system itself.As the central vacuole system enlarged, volume densities ofdense-body vesicles, peripheral vacuoles, lipid bodies and thecytoplasmic matrix decreased relative to total oogonial volume(peripheral protoplasm volume plus central vacuole volume),while the volume density of nuclei increased and that of mitochondriaremained constant. Relative to the peripheral protoplasm only,volume densities of dense-body vesicles, lipid bodies and mitochondriaincreased and volume densities of peripheral vacuoles and ofthe cytoplasmic matrix decreased, while the volume density ofnuclei increased during central vacuole enlargement but subsequentlydecreased during formation of oosphere initials. Under conditionsof Ca2+ deficiency, the volume densities of mitochondria andof the cytoplasmic matrix were significantly increased, whilethat of lipid bodies was significantly decreased, at early stagesof oogonial development; the volume densities of other organelleswere not significantly altered at any stage. Nuclear volumes and cell areas were determined for seven regionsof the meristem of roots of Zea mays. Roots were fixed in 10per cent neutral buffered formalin, in 3 per cent glutaraldehydeor in acetic acid/alcohol; they were prepared as sections oralls were teased apart. Mean volumes of interphase nuclei weresimilar in all regions of the root except the vascular tissueof the stele. Mean nuclear volumes and the overall range ofvolumes were similar in sub-populations of cells with differentproportions of G1, S and G2 cells, e.g. in row I of root capinitials, whose cells lack a G1 phase, and in quiescent centrecells, which are mainly in G1. Nuclear volume does not appearto be closely correlated with DNA content. Nuclear volumes covereda 6 to 12-fold range within a meristem and even within specificregions, in which cells are part of the same cell lineages,there was a 4- to 9-fold range. Nuclear volumes were comparedin sister cells in rows I and II of the root cap initials. In10 per cent of the pairs, sister nuclei had identical volumes;the other pain had different volumes and mean difference was68 µm3. Mechanisms by which this variability could begenerated are discussed, particularly asymmetry, at mitoses,of factors that regulate nuclear growth. Saprolegnia oliviae sp. nov. is described from litter (floating dead twigs, leaves and roots) in the Olivia River, Ushuaia Department, Tierra del Fuego Province (Argentina). The new species is illustrated and compared with other species of the genus. Distinguishing characteristics of S. oliviae are the production of smooth oogonia (with some lateral or terminal projections) and the absence of antheridial branches on the majority of the oogonia, but when present, they are mostly diclinous, at times oogonia are supplied with androgynous and monoclinous antheridial branches. The oogonial stalks are predominantly short and straight or long and bent, curved or many times coiled; oospores are distinctive subcentric, (1-) 15-50 (-70) per oogonium. Saprolegnia multispora is described from water and floating organic matter taken in the Tille River, in the Burgundian region of France. The new species is illustrated and compared with other species of the genus. Distinguishing characteristics of S. multispora are the production of smooth-walled oogonia containing many subcentric oospores which are (1-) 11-70 (-100) per oogonium. The antheridial branches supplying the oogonia are predominantly diclinous, but at times these may be monoclinous and androgynous. Morphological features of the oomycete and the sequence of the ITS region of its rDNA as well as their comparison with related species are discussed in this article.  相似文献

In Sphagnum cuspidatum unesterified polyuronic acids form 12per cent. of the dry weight; in S. acutifolium 25 per cent,of the dry weight. A good correlation has been found for Sphagnabetween the content of unesterified polyuronic acid and thecation exchange ability, and between cation exchange abilityand height of normal habitat above the water table. Anion exchangeability in Sphagna is less than 0.0026 m.eq./g. D.W. comparedwith about 1.2 m.eq./g. D.W. for cations at pH values above7. In natural conditions the exchange sites are, however, onlypartly dissociated. The production of new plant material ina bog dependent on rainwater for nutrients can be sufficientto maintain the pH below 4.5, but on average in England onlyof the order of 2 per cent, of the monovalent cations otherthan H+ could be retained in exchangeable form. A greater proportionof polyvalent cations could be retained. The kinetics of cation exchange are consistent with a heterogeneousexchange phase containing regions of high charge density andregions with lower charge density. At equilibrium the proportionsof different cations in the exchange phase are largely explicableby a Donnan distribution, but there are notable exceptions.Two estimates based on Donnan distribution suggest that withlow external pH and/or low cation concentration the apparentconcentration of exchange sites may be 2-3 eq./l., falling withrise in pH and/or increase in cation concentration to 0-9-1-5 eq./l. The apparent dissociation coefficient also variesin these conditions from 2 x 10–2 to 1 x 10–4.  相似文献

The crop growth rates and structures of three temperate foragegrasses Lolium perenne cv. S24, L. perenne cv. Reveille andFestuca arundinacea cv. S170, were examined in the field duringa summer growth period. The growth rates of the varieties wereremarkably similar at 7 g DM m–2 day–1. The angularstructures of the varieties were different and they varied duringthe experiment. However, these differences did not seem to affectcrop growth rates. Nevertheless, a decrease in the efficiencyof light energy conversion of approximately 24 per cent wasobserved after a change to a more prostrate form of canopy dueto lodging. There appeared to be an inverse relationship betweenthe number of tillers per unit ground area and the weight ofan individual stem. There were large numbers of relatively lighttillers in S24 whereas S1 70 had fewer but heavier tillers.Furthermore, S24 had many small leaves per unit ground areacompared with SI70 which had fewer longer leaves per groundarea and a slower rate of leaf appearance. There were diurnalchanges in the rates of leaf extension for all the varieties.The mean daily extension rates declined as the canopies developed.  相似文献

Saprolegnia brachydanis is described from zebra fish (Brachydanio rerio) in Wuhan, Hubei Province, China. The species is illustrated and compared with other species of the genus. The distinctive characteristics of S. brachydanis are the production of glomerulate oogonia wrapped around by predominantly monoclinous antheridia which can be up to eight in one oogonium. The oogonial stalks are short, straight, or curved and the antheridia, twisted, can enwind one or more oogonia. The oospores cannot mature or easily abort. Morphological features of the oomycete and the ITS sequence of its rDNA as well as the comparison with related species are discussed in this article.  相似文献

The growth of lucerne var. Europe was examined in the fieldduring 1976. The annual dry matter production of unirrigatedlucerne during 1976, with no nitrogen fertilizer application,was 82.5 per cent greater than unirrigated S.24 perennial ryegrass,with a nitrogen application of 384 kg ha–1. The mean aboveground growth rate of lucerne was 7.3 g DM m–2 day–1between March and early June, of which stem material contributeda maximum of 76.5 per cent. Significant losses of leaves andstems occurred from the end of April, indicating a loss of potentialforage material. Nitrogen analyses of the above ground crop suggested that in56 days lucerne yielded 10.7 per cent more nitrogen than didS.24 annually with a nitrogen fertilizer addition of 280 kgha–1. Saprolegnia diclina andS. parasitica isolated from three sources could germinate in strong acidic conditions. Growth ability correlated with the species of fungi rather than with the sources from which they were isolated.S.diclina isolates appeared to germinate at a pH condition as low as 3.5, whereasS. parasitica isolates could not germinate at below pH 3.8.S. parasitica isolates from visceral mycoses still showed good growth at 30°C, whereas other isolates did not. Brassica oleracea pollen was applied to a basic medium of 1.5per cent agar and 15 per cent sucrose to which flavanoids wereadded at three concentrations. Two types of agar were used;with agar 1, quercetin at a concentration of 0.5 x 10–3per cent gave an increase in percentage germinating grains.With agar 2, an increase in germination occurred with kaempferoland naringin at concentrations of 0.5 x 10–3 and 0.5 x10–1 per cent respectively. Increase in pollen tube lengthoccurred with agar 2 and quercetin at a concentration of 0.5x 10–3 per cent. The stigma tissue of B. oleracea contains at least three andthe pollen at least one glycoside of quercetin. The sugars inthe glycosides were not identified. Pollen germination and pollentube extension were not stimulated exclusively by the flavanoidspresent in the stigma. The flavanoid composition of the stigmadid not vary amongst five different S-allele genotypes, indicatingthat flavanoids are probably not directly involved in the incompatibilityreaction of B. oleracea.  相似文献

Glucose, either uniformly labelled with14C, or specificallylabelled in the I, 2, or 6 position, was added to C. vulgaris.Radio-active carbon dioxide was produced initially ten timesfaster from glucose-I-14C than from glucose-6-14C. This differencewas found with carbohydrate-starved cultures, exponentiallygrowing cultures, and cultures assimilating ammonia or nitraterapidly. A similar difference was also found with C. pyrenoidosaand Ankistrodesmus. 37 per cent. of the 14C added as glucose-1-14Cto exponentially growing cells was recovered as carbon dioxidebut generally the recovery was less than this. A long-term, steady-state 13CO2 assimilation system at a constantCO2 concentration with a constant 13C abundance was designedand applied to quantitative investigations on the allocationof photoassimilated carbon in nodulated soya bean (Glycine maxL.) plants. The CO2 concentration in the assimilation chamberand its 13C abundance were maintained constant with relativevariances of less than ±0.5 per cent during an 8-h assimilationperiod. At the termination of 8-h 13CO2 assimilation by plantsat early flowering stage, the currently assimilated carbon relativeto total tissue carbon (measured by the degree of isotopic saturation)were for young leaves (including flower buds), 13.9 per cent;mature leaves, 15.7 per cent; stems+petioles, 5.9 per cent;roots, 5.4 per cent and nodules, 6.9 per cent, 48 h after theend of the 13CO2 assimilation period, they were 12.3, 7.5, 7.4,6.8 and 6.1 per cent, respectively. The treatment with a highconcentration of nitrate in the nutrient media significantlydecreased the allocation of 13C into nodules. Experiments on13CO2 assimilation by plants at the pod-filling stage were alsoconducted. Labelling by 13C was weaker than at the early floweringstage, but an intense accumulation of 13C into reproductiveorgans was observed. Sixty-eight per cent of nuclei in the cells of the upper fourlayers of carrot slices treated with heat-killed conidia ofBotrytis cinerea for 6 h followed by inoculation with live sporesfor 18 h, migrated to the cell face nearest to the treated surface,compared with 46 per cent in cells of control slices showinga wound-healing response only. Nucleolar volumes in the surfacecell layers of control slices increased from a mean of 1.0 µm3to 3.8 µm3 over 24 h, and in ‘induced’ slicesto 7.28 µm3. Using a 40 min pulse of [5–3H]uracil,there was an increase within 15 h of slicing in the number oflabelled nuclei in cells from control slices undergoing healing.Within 8 h after treatment of slice surfaces with heat-killedconidia, there was an accelerated incorporation of label into‘nuclear’ RNA. Slices from roots cold-stored for12 months failed to show an induction response and nucleolarvolumes did not increase more than in control slices. The development of lateral root primordia has been investigatedin excised roots of Vicia faba, Pisum sativum, Zea mays andPhaseolus vulgaris cultured in White's medium supplemented with2 per cent sucrose and compared with previously published dataon such development in primaries of the corresponding intactplants (control roots). Primordia were produced in each batchof excised roots over the 6 day culture period but at a lowerrate (number day–1) than in the controls. Such primordia in cultured roots of Zea and Phaseolus completedtheir development and grew out as lateral roots over a periodsimilar in length to that found in the controls, but with acell number of only about 33 per cent of that attained at thetime of secondary emergence in the primaries of the latter roots.These lower cell numbers were at least partly a reflection ofincreases in mean cell doubling time over the period of anlagedevelopment investigated in the excised roots relative to thecorresponding values found in the controls. Primordia initiated in excised roots of Pisum and Vicia didnot complete their development in culture, i.e. no lateral rootsemerged and arrest took place with cell numbers of only 37 (Pisum)and 17 (Vicia) per cent of the numbers determined at the timeof secondary root emergence in the controls. Such arrested primordiahad few nuclei in S and none in mitosis. Moreover, at leastin Pisum, the frequency distribution of the relative DNA contentof the nuclei in the latter primordia approximated that foundin the apical meristem of primary roots following the establishmentof the stationary phase under conditions of carbohydrate starvation. It has also been demonstrated in the course of these investigationsthat lateral root primordium development in all four speciesis at least biphasic and possibly triphasic. The mineral, carbohydrate, protein, amino acid, lipid and energycontents of the seven main pollens gathered by honey-bees insouth-east Scotland were determined. Neither dry matter (mean73 per cent) nor ash (mean 3 per cent) were very variable. Thecalcium content of all pollens was relatively similar as werethe magnesium, potassium and sodium contents but manganese whichwas relatively high in ericaceae and phosphorus which was lowin Trifolium were more variable. Acer pollen contained mostprotein and Fagus least, and of the amino acids only serine,cystine and histidine were significantly different in the differentpollens. Over 30 per cent of all pollens was a mixture of fructoseand glucose, ericaceae containing most and Acer least. Hemicellulose(mean 7 per cent) was high in Ranunculus and low in Acer, cruciferaeand Fagus. ‘Cellulose’ (mean 0•5 per cent)was high in cruciferae and low in ericacea. Ether extract (mean5 per cent) was high in cruciferae and Fagus. The mean grossenergy of pollen was 23 kJ g–1. Fresh pollen is comparable with lean meat as a source of energyand protein, and is more like dormant plant seeds than herbagein its water, ash and protein content but stores energy as sugarrather than starch or lipid.  相似文献
