Affinity chromatography on immobilised concanavalin A revealed that transcortin and thyroxine-binding globulin isolated from human postpartum serum contained ~10% of molecular variants that did not occur in these glycoproteins isolated from normal donor serum (both male and female). The chromatographic behaviour of the pregnancy-associated glycoprotein variants, their monosaccharide compositions, and the results of methylation analysis indicated that these variants contained only triantennary oligosaccharide chains of the N-acetyl-lactosamine type.  相似文献

Affinity labelling of human transcortin   总被引:1,自引:0,他引:1  
The binding site of transcortin has been studied by using bromoacetyltestosterone and bromoacetylated derivatives of progesterone which were monohydroxylated at different positions of the steroid nucleus. Specificity of affinity labelling was demonstrated by the displad cortisol analog was added to a [3H]cortisol-transcortin complex solution. The binding site crevice was found to be very narrow in the vicinity of the A and B rings of steroid since 2alpha-hydroxyprogesterone, 6alpha- or 6beta-bromoacetoxyprogesterone and dexamethasone could not displace bound cortisol. A specific affinity labelling was obtained with 11alpha-bromoacetoxyprogesterone, 16alpha-bromoacetoxyprogesterone and 17beta-bromoacetyltestosterone. The results of the affinity labelling by these hormone analogs suggested that one methionine and one histidine residues were located within the active site:methionine might interact with the 11beta-hydroxyl group and histidine with the 20 keto group of cortisol.  相似文献

Human transcortin was purified to apparent homogeneity from plasma by a two-step procedure involving affinity and hydroxyapatite chromatography. The affinity gel incorporated denatured bovine serum albumin as the spacer and cortisol hemisuccinate as the ligand. Although isolated transcortin showed a propensity for spontaneous polymerization according to a geometric progression (1, 3, 9) only one band was observed on sodium dodecyl sulphate/polyacrylamide-gel electrophoresis. Cortisol-binding activity of the isolated protein gave an apparent association constant of 2.5 X 10(8) M-1 at 4 degree C in equilibrium dialysis. Isoelectric focusing of purified native transcortin showed six discrete bands, five between pH 3.75 and 4.15 and another, possibly desialylated, at pH 6.15. Desialylated transcortin also gave six bands on isoelectric focusing, with pI values ranging from 4.90 to 6.30.  相似文献

Serum protein bound progestins, androgens, estrogens, and cortexolone, were fractionated on a number of chromatographic systems. Contrary to earlier suggestions of a homogeneous unit by competition binding and Scatchard analysis, polymorphism and heterogeneity in the molecular nature of the corticosteroid binding globulin (CBG) were evident with progesterone on Sephadex A-25 columns. Components in the 55 000 and 67 000 molecular weight regions were obtained with cortexolone, estradiol, progesterone and testosterone on Ultrogel columns. Separation on DEAE-cellulose-52 resin revealed a fraction in the low ionic prewash followed by another, highly charged entity eluted only with 0.06 M phosphate buffers. Thus, under these conditions, serum protein bound sex steroids eluted in the same position as transcortin glucocorticoid complexes. These results are presented as a caution against the universal use of association dissociation assays to study steroid ligand binding and biological response. The techniques here detailed may fruitfully be employed to explore the hydrodynamic characteristics of protein ligand interactions. In addition, they support the model, earlier proposed with tissue specific steroid receptors, that calls for the various subunits of the ligand as a prerequisite for the expression of all grades of agonist and antagonist activity of a given hormone.  相似文献

While spacer arms have been shown to play an important role in affinity chromatography, no systematic investigations of spacer arms in the purification of transcortin have been reported. Among the five cortisol-agaroses, cortisol-21-succinyl-1,6-hexanediamidoagarose achieved the highest extraction efficiency of transcortin from plasma. The optimal length of the spacer arm for extraction is ca. 12–13 Å. Cortisol-succinyl-agaroses having hydrophobic spacer arms extract transcortin better then those having hydrophilic arms of approximately equal length. Affinity supports are usually synthesized sequentially; cortisol-agaroses thus prepared were found to complicate the purification of transcortin. The problems of nonspecificity and instability associated with these agaroses were eliminated by using reverse addition. A complete ligand-spacer arm, synthesized in a single step by displacing the tosyl group from cortisol-21-tosylate with 1,6-hexanediamine, was coupled with cyanogen bromide-activated agarose. Although the 21-deoxy-21-(ω-amidohexyl) aminocortisol-agarose ranked second in extraction efficiency, its superior stability and low nonspecific adsorption of other proteins make it the prime choice for affinity chromatography of transcortin.  相似文献

The molecular weight of human transcortin, calculated from the sedimentation coefficient, was found to be 49,500, thus slightly lower than previously reported values. After purification, human transcortin trended to polymerize rapidly, with participation of both non covalent bonds and one disulfide bridge per dimer. The physicochemical parameters, the amino-acid and carbohydrate composition were determined; its stability was studied under different conditions. Preliminary structural studies showed that the N-terminal sequence of the polypeptide chain was: Met-Asp-Pro-Asn-Ala-Ala-Tyr-Val and that the C-terminal amino acid was leucine.  相似文献

The hepatotoxic effect of 1,1 bis (p-chlorophenyl) 2,2,2 trichloroethane (DDT) treatment for 10 consecutive days has been examined in Wistar rats. DDT exposure increased relative liver weight, dose dependently, with a marked decrease of glycogen content and profound histological changes including cytoplasmic vacuolization, signs of necrosis and nuclear enlargement. The hepatomegaly induced by DDT (50 and 100 mg/kg body weight day-1) appeared not to be accompanied by a significant alteration of the hepatic glucocorticoid receptor concentration and affinity while, serum corticosteroid binding globulin level increased slightly with the lower dose of the pesticide. It is concluded that a short-term exposure to DDT did not lead to a status stress and, therefore, the hepatotoxic effect of organochlorine seemed not to be mediated by endogenous glucocorticoids.  相似文献

Seven carboxy modified and four amino modified derivatives of human serum albumin have been prepared and studied by optical rotatory dispersion measurements, gel filtration, immunoelectrophoresis, immunodiffusion and by use of an ammonium sulphate technique. It is found that carboxy groups are of major importance for the maintenance of the structure and function of human serum albumin, and that carboxy modification has a much more profound effect than amino modification has to the same relative extent.  相似文献

It was demonstrated that the physico-chemical properties of human transcortin, i.e., electrophoretic, hydrodynamic and immunochemical characteristics, amino acid composition, steroid binding parameters, do not depend on the source of the glycoprotein (male or female blood, retroplacental blood). Conversely, the retroplacental blood serum was shown to contain a transcortin form whose carbohydrate component is structurally different from that of the normal donor blood transcortin. It was found that this form interacts with the sites of specific binding of transcortin in liver cell plasma membranes in a weaker degree than the donor blood transcortin.  相似文献
