尖孢镰刀菌生产蒽醌色素的液体发酵条件研究   总被引:2,自引:0,他引:2  
优化了尖孢镰刀菌液体发酵生产蒽醌类红色素的发酵条件。通过单因素实验和正交优化实验,确定最佳产色素发酵培养基为:可溶性淀粉30%,(NH4)2SO4 3%,MgSO4 0.3%,KH2PO4 4%,pH 6.0。产色素最适培养条件为:初始pH6.0,装液量20%,接种量10%,吐温-80添加量1%,温度28℃,摇床转速200r/min,发酵周期120h。此条件下,色素效价即可达到8.184U/ml,比优化前提高了1.8倍。国内首次对尖孢镰刀菌所产蒽醌色素进行研究,为其进一步应用奠定基础。  相似文献

为提高红曲色素的液态发酵水平,降低生产成本,采用Plackett-Burman试验与Box-Benhnken试验对红曲霉MY-03液态发酵生产红曲色素的工艺条件进行了优化,获得了最佳工艺条件:葡萄糖50 g/L、蛋白胨58.07 g/L、Mg SO4 2 g/L、Na NO3 2g/L、Mn SO4 0.3 g/L、Zn SO4 0.1 g/L、装瓶量50.8 m L、接种量10.0%(V/V),于150 r/min、30℃恒温培养7 d,红曲色素色价可达342.24±2.88 U/m L,比基础培养基提高了86.9%。  相似文献

经60 Coγ射线辐照处理获得的诱变菌株芽孢杆菌BS9418F ,其产酶活力比出发菌株提高 30 %以上。该菌株以大麦粉 7%、玉米粉 3%、豆粕 3%及适量无机盐为培养基最佳配比 ,其最适培养条件为 :培养基初始 pH 7.0 ,摇瓶装量 5 0mL/ 30 0mL三角瓶 ,种龄 16~ 2 0h ,接种量 2 %~ 3% ,培养温度 36~ 37℃ ,发酵周期 40h。在优化条件下 ,摇瓶发酵产 β 葡聚糖酶活力高达 5 5 0 0u/mL以上 ,比出发菌株初始发酵水平提高了 4倍以上  相似文献

洋葱假单胞菌(Pseudomonas cepacia)PCL-3产脂肪酶发酵条件研究   总被引:2,自引:0,他引:2  
研究了洋葱假单胞菌(Pseudomonas cepacia) PCL-3发酵产碱性脂肪酶培养条件的优化。采用单因子试验筛选出糊精为最适碳源,蛋白胨和尿素为复合氮源。通过Plackett-Burman设计试验,对影响产酶条件的8个相关因子进行评估并筛选出具有显著效应的三个因子:尿素、接种量以及初始pH值。用最陡爬坡实验逼近显著因子的最大响应区域后,采用响应面分析法,确定尿素、接种量以及初始pH值最优值分别为0.15%,3.05%和8.59。优化后液体发酵培养基中脂肪酶活力提高到48.88 U/mL,比初始酶活25.37U/ml提高了1.93倍。10 L 的发酵罐中,脂肪酶活力在52h达到最大,为47.69U/mL。  相似文献

[目的]构建长野芽孢杆菌普鲁兰酶突变体枯草芽孢杆菌工程菌株,优化发酵条件,筛选廉价的培养基原料生产普鲁兰酶。[方法]利用分子生物学手段,将基因pul324和表达载体p WB980连接,构建表达质粒p WB-pul324并转化Bacillus subtilis WB600;对表达产物进行SDS-PAGE分析和初始酶活的测定。进一步优化发酵条件,对不同碳源和氮源进行发酵培养基的筛选,同时研究不同金属离子的添加和培养基初始p H、接种量对发酵产酶的影响。[结果]获得基因工程菌B.subtilis WB600/p WB-pul324,SDS-PAGE电泳结果显示在89 k Da处有特异性条带,发酵初始酶活为12.34 U/ml;筛选得到玉米淀粉水解液和玉米浆干粉为培养基最适碳源和氮源,其最适浓度分别为50 g/L和30 g/L。Mn~(2+)、Fe~(3+)、Fe~(2+)和Tween-80的添加能提高发酵产酶活力。在最适初始p H 6.5,以最适5%接种量接种于优化后的培养基中,摇瓶发酵80 h普鲁兰酶的酶活达到414.48 U/ml。[结论]实现了普鲁兰酶突变体在枯草芽孢杆菌中的高效表达,筛选获得的培养基主要原料经济低廉,经过发酵条件优化后,重组菌酶活达到414.48 U/ml,是之前研究结果(20.16U/ml)的20倍。  相似文献

目的:探索和了解发酵生产葡甘聚糖酶的最佳条件。方法:在 5L发酵罐上测定不同温度、pH和接种量对发酵的影响,并用正交试验筛选最佳的产酶条件配伍。结果:该菌产生葡甘聚糖酶的条件为接种量 1 0 %,通气量 2 0L h,发酵的前 1 6h温度为 40℃,搅拌速度 2 0 0r min,pH 7 0左右,之后温度调至 5 0℃,搅拌速度 1 0 0r min,pH调至 6. 0左右,添加 0 . 2 5 %的豆油做消泡剂。在这个条件下发酵获得的酶比活力可达 5 0 0 9U mg,总活力达到 1 0 81 9U ml。  相似文献

从2株芽胞杆菌中通过筛选获得了一株产α-葡萄糖苷酶活力较高的嗜热脂肪芽孢杆菌U2,以嗜热芽孢杆菌U2为菌种,优化发酵培养基后,在温度45℃、初始pH6.8、转速200r/min和10%接种量条件下,发酵20h,菌株U2产酶水平可达到2.62U/mL,比出发菌提高了4倍。  相似文献

对生物柴油废液作简单处理,利用红曲茵发酵生物柴油废液中副产物甘油生产红曲色素。通过响应面方法确定最佳发酵培养基为:甘油48.49g/L,蛋白胨3.12g/L,K2HPO4·3H202.01g/L,MgSO4 0.48g/L,ZnSO4·7H2O 0.04g/L,MnSO4·H2O 0.03g/L,玉米浆13mL/L,植物油10mL/L,起始pH为6。发酵结果表明:在接种量6%(v/v),转速140r/min,35℃的条件下发酵培养6d,红曲色素最高产量到达204U/mL。说明用生物柴油废液中的粗甘油为原料生产红曲色素是基本可行的。可望为生物柴油废液的资源化提供一条环境友好型的途径。  相似文献

对通过紫外线和60Co两次育种得到的桔青霉W48高产菌株培养基的单组分和发酵条件进行了优化,最后又用正交实验对培养基的各组分的浓度进行了优化。得到了产酶量最高的培养基组分(质量分数)是:KH2PO40.05%,K2HPO40.03%,酵母膏+蛋白胨0.7%,CaCl20.02%,MgSO40.04%,ZnSO40.03%,葡萄糖5%,pH5.5。最佳发酵条件是:接种量10%,装液量50 mL,摇床转速180 r/min,温度30℃,发酵时间66 h。用最佳培养基和最佳发酵条件发酵生产核酸酶的酶活力为758.10 U/mL,原始菌种产核酸酶的酶活力为272.26 U/mL,提高了2.78倍。  相似文献

采用黑曲霉 (Aspergillusniger)GD 6液体发酵生产纤维素酶 ,研究了碳源、氮源、培养基起始 pH值、接种量、摇床转速、通气量对该菌株产纤维素酶活力的影响。结果表明 ,GD 6的最适发酵温度为 2 8~ 3 0℃ ,产酶pH为 5 .5~ 6.0 ,摇床最适转速为 1 5 0r/min ,最佳接种量为 1 0 %。在以 6.0 %稻草粉为碳源、1 %豆饼粉为氮源时产酶活力最高。在最适培养条件下 ,发酵周期为 1 2 0h,发酵液中CMC酶活为 1 88.6U/mL ,FP酶活为 2 7.0U/mL。  相似文献

Pectin rich wastes and waste dump yard soils were screened and eighty pectinolytic fungal isolates were obtained by enrichment culturing and ruthenium red plate assay. Eight isolates with higher zones of pectin hydrolysis were selected and tested for polygalacturonase production. One isolate identified as Aspergillus awamori MTCC 9166 with highest polygalacturonase activity was tested for utilization of raw pectins for enzyme production. Polygalacturonase production was high in raw pectin sources like Orange peel (16.8 U/ml) Jack fruit rind (38 U/ml) Carrot peel (36U/ml) and Beet root peel (24U/ml). Selected Aspergillus awamori MTCC 9166 was found to be having good polygalacturonase, xylanase, cellulase and weak amylase and protease activities. This isolate with multi-enzyme production could have application for enzymes production and degradation of fruit and vegetable waste in the process of urban waste disposal.  相似文献

The aim of the study was to investigate the effect of Garcinia cambogia extract on serum calcium (Ca), phosphorus (P), magnesium (Mg), iron (Fe), zinc (Zn) and copper (Cu) concentrations in rats fed with the normal or the high-lipid and -cholesterol diet. Thirty 1-year-old female Sprague-Dawley rats (pathogen-free), weighing an average of 229 g, were randomly assigned to three experimental groups of ten animals each. Diets and tap water were given ad libitum for 75 days. Group 1 (control group) was fed with basal diet (2 % liquid vegetable oil, 0 % cholesterol), while the diets of groups 2 and 3 contained vegetable oil (2 % liquid vegetable oil and 5 % hydrogenated vegetable oil) and cholesterol (3 %) in high levels. 4,5 % G. cambogia extract containing 65 % HCA was added to the diet of group 3 as from day 45. Blood samples were withdrawn on days 0, 45 and 75. Serum mineral levels were analyzed using standard enzymatic colorimetric methods with a spectrophotometer. All significant differences were p<0.05. Serum Ca levels were not significantly different between all groups on days 45 and 75. Serum P level was significantly higher in the group fed with high-lipid diet and G. cambogia extract than in the control group on day 45. Serum Mg level was significantly higher in group 2 than in the control group on day 45. Serum Fe levels were significantly lower in the control group than in the other groups on days 45 and 75. Serum Zn level of the group fed with high-lipid diet and G. cambogia extract was significantly higher than in the control group on day 75. Serum Cu levels were significantly higher in group 2 than in the control group, and in group 3 than in group 2 on day 75. In conclusion, a diet containing the high fat amounts may lead to the increase in circular levels of some minerals due to the short-chain fatty acid production lowering the luminal pH which increases mineral solubility, or serving as a fuel for mucosal cells and stimulating cell proliferation in the large intestine. G. cambogia extract may be used in the P and Cu deficiencies due to increases resulting in the present P and Cu amounts in G. cambogia extract, or the use of phytate P in diet. It was hoped that with further evidence-based study this product will enter to mainstream medicines.  相似文献

Fatty acid composition and stability of vegetable oils have taken more attention as an essential source of biologically active compounds in a good balanced diet. The purpose of the study was to determine peroxide value, free fatty acids, unsaponifiable matter, total carotenoid content, iodine value and fatty acid composition of sunflower, rapeseed, mustard, peanut and olive oils. Rapeseed and peanut oils had the highest peroxide values, while sunflower oil had the lowest peroxide values. The free fatty acid value of the tested oils varied between 0.43 and 1.36% oleic. The peanut oil had the highest free acid value and the mustard oil had the lowest one. Total carotenoid contents of mustard and rape seed oil were higher than those of the other oils tested. Palmitic acid (C16:0), oleic acid (C18:1) and stearic acid (C18:0) were the common main fatty acid components of the vegetable oils tested. Followed by linoleic acid, the amount of oleic acid was the highest among other fatty acid components. Mustard oil had the highest erucic acid (C22:1) with the amount of 11.38%, indicating that it cannot be used for human consumption. Among the oils investigated, sunflower and mustard oils were more stable than rapeseed, peanut and olive oils.  相似文献

Uricase is an important medical enzyme which can be used to determine urate in clinical analysis, to therapy gout, hyperuricemia, and tumor lysis syndrome. Uricase of Candida utilis was successfully expressed in Hansenula polymorpha under the control of methanol oxidase promoter using Saccharomyces cerevisiae alpha-factor signal peptide as the secretory sequence. Recombinant H. polymorpha MU200 with the highest extracellular uricase production was characterized with three copies of expression cassette and selected for process optimization for the production of recombinant enzyme. Among the parameters investigated in shaking flask cultures, the pH value of medium and inoculum size had great influence on the recombinant uricase production. The maximum extracellular uricase yield of 2.6 U/ml was obtained in shaking flask culture. The yield of recombinant uricase was significantly improved by the combined use of a high cell-density cultivation technique and a pH control strategy of switching culture pH from 5.5 to 6.5 in the induction phase. After induction for 58 h, the production of recombinant uricase reached 52.3 U/ml (about 2.1 g/l of protein) extracellularly and 60.3 U/ml (about 2.4 g/l) intracellularly in fed-batch fermentation, which are much higher than those expressed in other expression systems. To our knowledge, this is the first report about the heterologous expression of uricase in H. polymorpha.  相似文献

Lipases are important cuticle-degrading enzymes that hydrolyze the ester bonds of waxes, fats and lipoproteins during the infection of insects by the fungus Nomuraea rileyi. Lipase production by the N. rileyi strain MJ was optimized by varying environmental and nutritional conditions in culture medium containing different vegetable oils at various concentrations with shaking at 150 rpm for 8 days at 25°C. The maximum lipase production was obtained using castor oil (30.5±0.6 U mL?1), followed in order by coconut oil (20.8±0.4 U mL?1), olive oil (20.8±0.4 U mL?1) and cottonseed oil (20.6±0.4 U mL?1). The highest lipase activity (37.7±0.4 U mL?1) was obtained when castor oil was used at a concentration of 4% (v/v) of basal medium. When the surfactant Tween 80 was added at the fourth day rather than at the beginning of incubation, a maximum lipase activity of 44.9±3.5 U mL?1 was obtained. The optimal temperature and pH for lipase production were 25°C and pH 8.0, respectively. This is the first report on lipase production by the biocontrol fungus N. rileyi.  相似文献

Comparisons were made for phytase production using wheat bran (WB) and oilcakes as substrates in solid-state fermentation (SSF) by Mucor racemosus NRRL 1994. WB was also used as mixed substrate with oil cakes. Sesame oil cake (SOC) served as the best carbon source for phytase synthesis by the fungal strain as it gave the highest enzyme titres (30.6 U/gds). Groundnut oil cake (GOC) also produced a reasonably good quantity of enzyme (24.3 U/gds). Enzyme production on WB was surprisingly much less (almost 3.5 times less in comparison to SOC). Mixing WB with SOC (1:1 ratio) resulted in better phytase activity (32.2 U/gds). Optimization of various process parameters such as incubation time, initial moisture content and inoculum concentration was carried out using the single variable mode optimization technique. Under optimized conditions, the production of phytase reached 44.5 U/gds, which was almost 1.5-fold higher than the highest yield obtained with any individual substrate used in this study and was more than 4-fold higher than that obtained from WB.  相似文献

Effects of feeding different available nitrogen sources from 80 h in erythromycin biosynthesis phase on the erythromycin A (Er-A) production were investigated in 50 l fermenter. Feeding corn steep liquor and yeast extract, the Er-A production was enhanced, while the biotransformation from erythromycin C (Er-C) to Er-A had no increase. When ammonium sulphate was fed at high feeding rate, the maximal Er-A production and ratio of Er-A to Er-C were 7953 U/ml and 98.18:1 at 184 h, respectively, which were higher than that of the control (6742 U/ml and 5.47:1). The feeding ammonium sulphate process was successfully scaled up from 50 l to 25 m3 fermenter. The maximal Er-A production reached 7938 U/ml at 203 h, which was enhanced by 22.1% compared with the control (6501 U/ml at 192 h). The ratio of Er-A to Er-C was 24.05:1, which was higher than that of the control (4.77:1).  相似文献

The addition of penicillin G to combat microbial contamination in continuous fuel alcohol fermentations was performed using both continuous and pulsed addition regimes. In continuous fermentations where both Saccharomyces cerevisiae and Lactobacillus paracasei were present, the mode of addition of penicillin G determined final numbers of viable L. paracasei. When the same overall average concentration of penicillin G was added in both pulsed and continuous modes, the initial viable number of L. paracasei (8.0 x 10(9) cfu ml(-1)) decreased to a greater degree (1.02 x 10(5) cfu ml(-1) L. paracasei) when penicillin G was pulsed at 6 h frequencies at an overall average concentration of 2,475 U/l than when penicillin G was added continuously at 2,475 U/l (2.77 x 10(5) cfu ml(-1) L. paracasei). Pulsed additions over longer frequencies at 2,475 U/l were not as effective in reducing viable bacteria. Viable yeasts increased during both treatment conditions by more than 2-fold. The two addition regimes also eliminated the 40% decrease in ethanol concentration caused by the intentional bacterial infection. Although there was 3 times more bacterial death with 6 h pulsed additions compared to continuous additions of penicillin G at 2,475 U/l, there was, by that point, no practical difference in either final ethanol concentration or relative ethanol recovery.  相似文献

One hundred consecutive isolates of N. gonorrhoeae were tested for susceptibility to penicillin, ampicillin, tetracycline, erythromycin, kanamycin, cephaloridine and cephalexin by an agar dilution method. Relative resistance to penicillin was frequent. For 39% of isolates the minimum inhibitory concentration (MIC) of penicillin was 0.05 U./ml. or less; in 55% the MIC was 0.5 to 2.0 U./ml. Ampicillin was slightly more active than penicillin G: all isolates were inhibited by 0.5μg./ml. or less. Resistance to tetracycline and erythromycin was frequent with MIC of 1 μg./ml. or greater observed in 32 and 24% of isolates respectively. The MIC of kanamycin for all gonococci was 8 μg./ml. or greater. Cephalexin was slightly more active than cephaloridine, though each drug exhibited a wide range of MIC values. Gonococcus isolates resistant to penicillin (MIC of 1.0 U./ml. or greater) tended to be resistant to the other antibiotics tested.  相似文献

To improve the yield of cellulose production in bacteria, we investigated the stimulatory effects of six different alcohols during fermentation of Acetobacter xylinum 186. Our study showed that after static fermentation at 30°C for 6 days, bacterial culture with 1.0% (v/v) of methanol added in the medium produced the highest bacterial cellulose (BC) yield at 103.5 mg/100 ml, which was 21.8% higher than the control group. Addition of 0.5% of ethylene glycol in the culture yielded 105.5 mg/100 ml BC, 24.1% higher than the control group. Adding 0.5% of n-propanol yielded 96.4 mg/100 ml BC, 13.4% higher; 3.0% of glycerol yielded 108.3 mg/100 ml BC, 27.4% higher; 0.5% of n-butanol yielded 132.6 mg/100 ml BC, 56.0% higher; and 4.0% of mannitol in the culture yielded 125.2 mg/100 ml BC, 47.3% higher, respectively. The rate of bacterial cellulose production increased with the growth rate of the bacteria. The stimulatory effects of these alcohols that we observed were significant in the later stage of fermentation, which was considered to be important for the biosynthesis of bacterial cellulose.  相似文献
