Ranavirus in wood frogs (Rana sylvatica): potential sources of transmission within and between ponds
Members of the genus Ranavirus (family Iridoviridae) can cause catastrophic mortality of pond-breeding amphibians and are associated with an emerging infectious disease that may be contributing to amphibian declines. We conducted three experiments to examine factors that may affect transmission both within and between local breeding populations of the wood frog (Rana sylvatica). In a laboratory study, when exposed to moribund tadpoles collected during a local ranaviral die-off, uninfected tadpoles died as soon as 4 days after exposure. The onset of death was accelerated when tadpoles were allowed to scavenge on carcasses of infected tadpoles. In a mesocosm experiment that was conducted in outdoor wading pools, die-offs of tadpoles began approximately 19 days after infected tadpoles were added to pools containing uninfected tadpoles. Mass die-offs with greater than 98% mortality occurred in all pools, regardless of the initial tadpole density. In a second mesocosm experiment, the addition of water and bottom sediments that were collected from a pond during a ranaviral die-off did not result in lower tadpole survival or growth relative to controls. Only a small percentage of tadpoles appeared to be sick, and most tadpoles survived until the first individuals began metamorphosing within a pool. However, tests for ranavirus using polymerase chain reaction were positive for most pools that received contaminated sediment, suggesting that some infections were sublethal. Our results indicate that transmission within ponds is enhanced by scavenging and that spread between local ponds could occur via the transport of contaminated sediment by animals or humans. 相似文献

Whilst the Major Histocompatibility Complex (MHC) is well characterized in the anuran Xenopus, this region has not previously been studied in another popular model species, the common frog (Rana temporaria). Nor, to date, have there been any studies of MHC in wild amphibian host-pathogen systems. We characterise an MHC class I locus in the common frog, and present primers to amplify both the whole region, and specifically the antigen binding region. As no more than two expressed haplotypes were found in over 400 clones from 66 individuals, it is likely that there is a single class I locus in this species. This finding is consistent with the single class I locus in Xenopus, but contrasts with the multiple loci identified in axolotls, providing evidence that the diversification of MHC class I into multiple loci likely occurred after the Caudata/Anura divergence (approximately 350 million years ago) but before the Ranidae/Pipidae divergence (approximately 230 mya). We use this locus to compare wild populations of common frogs that have been infected with a viral pathogen (Ranavirus) with those that have no history of infection. We demonstrate that certain MHC supertypes are associated with infection status (even after accounting for shared ancestry), and that the diseased populations have more similar supertype frequencies (lower FST) than the uninfected. These patterns were not seen in a suite of putatively neutral microsatellite loci. We interpret this pattern at the MHC locus to indicate that the disease has imposed selection for particular haplotypes, and hence that common frogs may be adapting to the presence of Ranavirus, which currently kills tens of thousands of amphibians in the UK each year. 相似文献

The status of amphibians in populations subjected to anthropogenic influence of various natures has been evaluated. We studied the effect of complex anthropogenic influences as well as chemical and radiation pollution. The status of the specimens in populations was evaluated by morphological (developmental stability as the level of fluctuating asymmetry) and cytogenetic (micronucleus test) methods. Disturbed developmental stability and cytogenetic homeostasis have been observed in populations affected by anthropogenic factors, which indicates the changed status of the organism. 相似文献

We use 14 microsatellite loci to investigate the impact of a viral disease ( Ranavirus ) on the population genetic structure of wild common frogs ( Rana temporaria ). Populations with a history of Ranavirus mortalities (and 83% declines in the number of frogs) were compared with populations with no history of infection. Infected ponds showed significantly elevated F IS (homozygote excess), significantly reduced relatedness, and no detectable effect on allelic richness. We hypothesize that the elevated F IS and reduced relatedness are consequences of assortative mating, and that allelic richness is maintained by immigration from nearby populations. Simulations indicate that the elevated F IS cannot be explained by population size reductions, but can indeed be explained by assortative mating (even if a mate choice locus is unlinked to the genetic markers). While the majority of studies consider demographic outcomes following disease outbreaks, our results indicate that emerging infectious diseases could also result in behavioural changes. 相似文献

Energy storage is an important component in the life history of species that directly influences survival and reproduction. The energetic demands of amphibian reproduction can differ between the sexes, with environmental conditions, reproductive pattern or process of the species, and depending upon the timing of breeding, and the reproductive season for a species. Surprisingly, comparative studies of pre-hibernation energy storage for anuran populations from different latitudes are relatively few in Asia, especially in China. Here we investigated the patterns of pre-hibernation energy storage of Heilongjiang brown frogs Rana amurensis, based on five populations along a finely latitudinal gradient in north China(40.7–43.7°N). We found that pre-hibernation energy storage of the frogs did not show a clear latitudinal cline, but differed strongly between the sexes, with males depositing more energy reserves into the muscle and liver, whereas females accumulate more energy in the gonads. The sexual differences in energy storage may result from differential timing of energy allocation for reproduction. 相似文献

Amphibians are often exposed to a wide variety of perturbations. Two of these, pesticides and pathogens, are linked to declines in both amphibian health and population viability. Many studies have examined the separate effects of such perturbations; however, few have examined the effects of simultaneous exposure of both to amphibians. In this study, we exposed larval southern leopard frog tadpoles (Lithobates sphenocephalus) to the chytrid fungus Batrachochytrium dendrobatidis and the fungicide thiophanate-methyl (TM) at 0.6 mg/L under laboratory conditions. The experiment was continued until all larvae completed metamorphosis or died. Overall, TM facilitated increases in tadpole mass and length. Additionally, individuals exposed to both TM and Bd were heavier and larger, compared to all other treatments. TM also cleared Bd in infected larvae. We conclude that TM affects larval anurans to facilitate growth and development while clearing Bd infection. Our findings highlight the need for more research into multiple perturbations, specifically pesticides and disease, to further promote amphibian heath. 相似文献

The pathogen Batrachochytrium dendrobatidis (Bd) has been associated with amphibian declines in multiple continents, including western North America. We investigated Bd prevalence in Oregon spotted frog (Rana pretiosa), a species that has declined across its range in the Pacific Northwest. Polymerase chain reaction analysis of skin swabs indicated that Bd was prevalent within populations (420 of 617 juvenile and adults) and widespread among populations (36 of 36 sites) where we sampled R. pretiosa in Oregon and Washington. We rarely detected Bd in R. pretiosa larvae (2 of 72). Prevalence of Bd in postmetamorphic R. pretiosa was inversely related to frog size. We found support for an interactive effect of elevation and sampling date on Bd: prevalence of Bd generally increased with date, but this effect was more pronounced at lower elevations. We also found evidence that the body condition of juvenile R. pretiosa with Bd decreased after their first winter. Our data indicate that some Oregon spotted frog populations are currently persisting with relatively high Bd prevalence, but the risk posed by Bd is unknown. 相似文献

It has long been speculated that urea accumulated during seasonal dormancy contributes to metabolic depression. Recent work
suggests urea can indeed act as a metabolic depressant during dormancy in a number of taxonomically diverse species of ectotherms.
The mechanisms by which urea exerts its hypometabolic effect are unknown, but potentially stem from inhibition of mitochondrial
respiration. We isolated mitochondria from Rana sylvatica skeletal muscle, an organ that is metabolically responsive to urea, and measured respiration rates in the absence or presence
of 80 mmol l−1 urea in the respiration medium. Because the effect of urea may be influenced by the intracellular milieu, in these experiments
we varied substrate (pyruvate or palmitoylcarnitine), temperature (4, 10, or 15°C), and pH (6.8 or 7.4). Oxygen consumption
of control and urea-treated mitochondria was sensitive to each of these variables, but neither state 3 nor state 4 respiration
was reduced by urea treatment and, to the contrary, urea treatment slightly increased state 4 respiration at higher test temperatures.
Although we did not test the efficacy of other incubation times or urea concentrations, the outcome of our experiment intimates
that the urea-induced hypometabolism observed in hibernating R. sylvatica results from inhibition of energy-utilizing processes elsewhere in the cell, rather than a direct inhibition of mitochondrial
respiration. Future investigation into urea’s effects on non-mitochondrial metabolic pathways is necessary to uncover the

The role of thyroid hormone (TH) in anuran metamorphosis has been documented from a variety of approaches, but the sequence of morpho-histological development of the thyroid glands that produce the secretion of the hormone was assumed invariant from studies of relatively few species even when the effects of environmental influences on larval development and metamorphosis have been largely documented. There are anurans in which developmental and growth rates diverge, and the resulting heterochrony in growth and development produces giant/miniature tadpoles, and or rapid/delayed metamorphosis suggesting changes of the activity of the thyroid glands during larval development. Herein, we analyze the morpho-histological variation of the thyroid glands in larval series of Ceratophrys cranwelli, Chacophrys pierottii, Lepidobatrachus laevis and L. llanensis that share breeding sites along semiarid environments of the Chaco in South America, belong to a monophyletic lineage, and present accelerated patterns in growth and development in order to have a morphological evidence about a possible shift of TH physiology. We describe gross morphology and histology of the thyroid glands and find features shared by all studied species such as the presence of supernumerary heterotopic follicles; changes in the volume and number of follicles towards the metamorphic climax, and cuboidal epithelia with occasional intra-cellular vacuoles as signs of low glandular activity without a manifest peak at the climax as it was assumed for anurans. We discuss different lines of evidence to interpret sources of extra supplement of TH to support the rapid metamorphosis. These interpretations highlight the necessity to design a research program to investigate the endocrine variation during development of ceratophryids taking in account their morphology, physiology and ecology in order to learn more about the effects of environmental and developmental interactions involved in the anuran evolution. 相似文献

Environmental unpredictability can influence strategies of maternal investment among eggs within a clutch. Models predict that breeding females should adopt a diversified bet-hedging strategy in unpredictable environments, but empirical field evidence from Asia is scarce. Here we tested this hypothesis by exploring spatial patterns in egg size along an altitudinal gradient in a frog species(Rana kukunoris) inhabiting the Tibetan Plateau. Within-clutch variability in egg size increased as the environment became variable(e.g., lower mean monthly temperature and mean monthly rainfall at higher altitudes), and populations in environments with more unpredictable rainfall produced eggs that were smaller and more variable in size. We provide support for a diversified bet-hedging strategy in high-altitude environments, which experience dynamic weather patterns and therefore are of unpredictable environmental quality. This strategy may be an adaptive response to lower environmental quality and higher unpredictable environmental variance. Such a strategy should increase the likelihood of breeding success and maximize maternal lifetime fitness by producing offspring that are adapted to current environmental conditions. We speculate that in high-altitude environments prone to physical disturbance, breeding females are unable to consistently produce the optimal egg size due to physiological constraints imposed by environmental conditions(e.g., duration of the active season, food availability). Species and populations whose breeding strategies are adapted to cope with uncertain environmental conditions by adjusting offspring size and therefore quality show a remarkable degree of ability to cope with future climatic changes. 相似文献

The nitrogen content, distribution, and amino acid composition of protein material were determined in wood and lignin of Fagus sylvatica. The data indicated that the nitrogen originated from hydroxyproline-rich cell wall glycoprotein, about half of which may be bound to the lignin polymer. Experiments were carried out to investigate whether Iberian green frog tadpoles Pelophylax perezi (formerly Rana perezi) are able of using the geomagnetic field for y‐axis orientation (i.e. orientation toward and away from shore). Tadpoles were trained outdoor for 5 d, in two different training configurations: (i) a training tank aligned along the magnetic north–south axis, with shore facing south, and (ii) a training tank aligned along the magnetic east–west axis, with shore located east, and similar to the shore–deep water axis (‘y‐axis’) found in their home stream, which flows from south to north. After training, tadpoles were individually tested for magnetic orientation in a water‐filled circular outdoor arena surrounded by a pair of orthogonally aligned cube‐surface‐coils used to alter the alignment of the earth's magnetic field. Tadpoles held in the east–west training tank oriented towards shore, indicating that they were able to distinguish between the shoreward and waterward direction along the y‐axis. Tadpoles trained in the tank that was aligned along the north–south axis showed bimodal magnetic compass orientation along the shore–deep water magnetic axis. These findings provide evidence for the use of magnetic compass cues for y‐axis orientation by P. perezi tadpoles. 相似文献

The effect of habitat fragmentation on finescale population structure of wood frogs (Rana sylvatica)
We examined the impact of recent anthropogenic habitat fragmentation on the genetic structure of wood frog (Rana sylvatica) breeding sites in Wellington County of Ontario, Canada. In addition to geographic distance (average pairwise distance ~22 km, greatest distance ~50.22 km), four landscape features hypothesized to contribute to genetic differentiation between breeding sites were considered: road density, a major highway (highway 401), canopy cover, and watershed discontinuity. Analysis of data from 396 samples across nine breeding sites using eight microsatellite DNA loci, revealed a small degree of significant genetic structure between breeding sites. The presence of highway 401 and road density were correlated with small but statistically significant structure observed between several groups of sites. One outlier breeding site outside of Wellington County located within the city of Toronto, had significantly lower allelic richness and much larger population differentiation with the Wellington sites. Our data suggest that recent fragmentation has had an effect on wood frog population structure and also demonstrate the importance of dispersal for this species in maintaining levels of genetic diversity. 相似文献

In many oviparous vertebrates, hatchling phenotypes are influenced by egg incubation temperature. Many of those phenotypic traits can also acclimate to long-term thermal conditions of juveniles and adults, yet the interactive effects of prehatching and posthatching temperatures on phenotypes have not been studied. To address such interaction, we incubated eggs of wood frogs (Rana sylvatica) at two temperatures and subsequently reared larvae at three temperatures in a fully factorial design. We measured body size, size-independent morphology, and burst swimming speed at one developmental stage. Body size was independent of egg temperature but decreased significantly with increasing larval temperature. Size-independent morphology depended in complex ways on both temperature treatments directly and on their interaction. Burst speed was not influenced directly by egg temperature but was influenced by larval temperature and by the interactions among egg temperature, larval temperature, and test temperature. Our results indicate pervasive effects of egg temperature even late in the larval period and show that prehatching and posthatching temperatures can interact to affect various phenotypic traits. Tadpoles may be able to alter the long-term effects of incubation temperature by choosing particular larval developmental temperatures. Thus, the importance of incubation temperature in oviparous vertebrates should be evaluated by considering the effects of posthatching temperatures. 相似文献

Summary We investigated kin recognition by larval wood frogs (Rana sylvatica) in blind laboratory experiments using spatial affinity as a recognition assay. Tadpoles reared with full-sibs displayed a significant preference for familiar full-sibs over unfamiliar non-kin, but failed to discriminate between unfamiliar full-sibs and unfamiliar paternal half-sibs. Tadpoles reared in social isolation (with or without maternal egg jelly) from the two-celled embryonic stage displayed a significant preference for unfamiliar full-sibs over unfamiliar non-kin. Tadpoles reared on a meat diet with their full-sibs: 1) exhibited a significant preference for unfamiliar full-sibs fed meat over unfamiliar non-kin fed meat, 2) failed to discriminate between unfamiliar full-sibs fed lettuce and unfamiliar non-kin fed meat, 3) exhibited a significant preference for unfamiliar non-kin fed meat over unfamiliar non-lin fed lettuce, 4) failed to discriminate between unfamiliar full-sibs fed meat and unfamiliar full-sibs fed lettuce, and 5) displayed a significant spatial preference for odors associated with meat (a familiar food) over odors associated with lettuce (an unfamiliar food). Our results, together with those of Cornell et al. (1989), indicate that the recognition cue of larval R. sylvatica has both genetic and environmental (dietary) components. Our findings establish that previous exposure to maternal egg jelly, kin, or conspecifics is not necessary for the development of kin recognition ability in larval R. sylvatica. Our results are more consistent with the self-learning of recognition cues (a form of phenotype matching) than with a recognition mechanism that involves a genetically fixed recognition template. Finally, our results indicate that increasing similarity between the recognition template and perceived cue does not necessarily result in increasing spatial affinity for kin. 相似文献
