Progesterone is essential for maintaining pregnancy in goats, and embryonic losses may be a consequence of the reduction in circulating progesterone levels close to the time of implantation. Some evidence exists regarding social dominance affecting the plasma progesterone levels in several species—where dominant females conceive earlier. The objective of this research was to determine whether serum progesterone levels differ in goats of different social status. A behavioural study was conducted for 10 days in a herd of 57 does and an individual success index (SI) was calculated according to the result of aggressive interactions. Goats were classified as high (SI: 1–0.67), medium (SI: 0.66–0.34) and low-ranking (SI: 0.33–0.0). Ovulation was synchronized using two injections of prostaglandin 11 days apart, and the plasma progesterone levels determined daily for a period of 20 days. The area under the plasma progesterone curve during the entire study was greater in the high than in the medium and low-ranking does (96.2 ± 5.8, 79.5 ± 5.3 and 81.3 ± 5.3 ng/ml, respectively, P < 0.05). During days 11–17 following prostaglandin synchronization, the plasma progesterone levels were higher in the high-ranking (P < 0.05), compared to the low-ranking does. Plasma progesterone levels were significantly correlated with SI at days 14 and 15 (r = 0.26; P < 0.05). Results suggest a relationship between social ranking of goats and the plasma progesterone production from the corpus luteum and other possible sources.  相似文献

We investigated whether LH secretion, estrous behavior and fertility would differ between sexually inexperienced and experienced anestrous goats exposed to the males. Male goats were rendered sexually active during the reproductive rest season by exposure to 2.5 months of artificial long days. Two groups of anovulatory sexually inexperienced and sexually experienced does were exposed to males during 15 days (n = 20 per group). LH pulsatility was determined every 15 min from 4 h before to 8 h after introducing males (Day 0). Estrous behavior was recorded twice daily. Pregnancy rates were determined on Day 50. Fertility was determined at parturition. Male sexual behavior was registered on days 1 and 2 during 1 h. Before introducing the males, the number of LH pulses did not differ between groups. After introduction of the males, all females increased their LH pulsatility, but the number of pulses did not differ between sexually inexperienced and experienced goats. The proportion of females displaying estrous behavior with a high pregnancy rate and fertility did not differ between inexperienced and experienced goats. The sexual behavior of the males did not differ significantly between those interacting with sexually inexperienced or experienced goats. We conclude that goats can show substantial endocrine and reproductive responses to males, even in the absence of previous sexual experience, when sexually active bucks are used.  相似文献

Postpartum ovarian activity, uterine involution, and the relationship with hormonal profiles were studied in non-seasonal Shiba goats, with or without nursing of their kids. After parturition, does were allocated to one of two groups, with either weaning on the day of parturition (n = 3; non-nursing group) or weaning at 7–10 weeks after parturition (n = 4; nursing group). Blood sampling (starting 7 days prior to expected day of parturition) and transrectal ultrasound evaluations (starting 2 days after parturition) were conducted every other day or daily to monitor the follicular dynamics, uterus size, and the levels of plasma progesterone, blood glucose, and insulin concentration until at 3 weeks following the first postpartum ovulation. In the nursing group, blood samples were also collected every 10 min for an 8 h period on the day before weaning, 2 and 6 days after weaning, for the analysis of the pulsatile patterns of LH release. In all animals, the blood glucose concentrations increased transiently on the day before parturition and were significantly (p < 0.05) higher than those on the day of parturition (mean of 144 ± 48.8 mg/dl vs 63 ± 11.2 mg/dl). In the non-nursing group, the first postpartum ovulation was observed 9.3 ± 3.2 days after parturition, while in the nursing group, no ovulation occurred before weaning in any of the goats. Here ovulation was observed 18.8 ± 5.0 days after weaning, which was significantly (p < 0.05) later than in the non-nursing group. After first ovulation, all animals in both groups showed early luteal regression. No significant difference in the time required for the completion of uterine involution was recorded between the non-nursing and nursing groups (18.3 ± 4.2 days vs 19.3 ± 3.6 days, respectively), nor in the plasma LH pulse frequency obtained before (1.3 ± 0.5 pulses/8 h), 2 (1.3 ± 1.0 pulses/8 h) and 6 days (2.0 ± 0.8 pulses/8 h) after parturition, in the nursing group. It can be concluded that no ovulation occurs during nursing postpartum, suggesting that nursing is a determinant of the resumption of ovulation in these non-seasonal goats. Ultrasonic observations of the ovary suggest that uterine involution was not influenced by nursing or suckling.  相似文献

This study was aimed at comparing the temporal relationship between estrus, the LH peak and ovulation following estrous synchronization in goats using medroxiprogesterone acetate (MGA) or fluorogestone acetate (FGA). Twenty-four cyclic goats were randomly assigned to two treatments: MGA group (n = 12) that individually received a daily oral dose of 0.22 mg MGA for 12 days, and a FGA group (n = 12) treated for 12 days with intravaginal sponges containing 45 mg FGA. The goats from both groups were treated with a prostaglandin analogue (75 μg Cloprostenol i.m.) during the last day of their progesterone treatment. Estrous detection was carried out every 4 h after the end of the treatment, and blood samples for the determination of the serum LH concentrations collected at 2 h intervals for 24 h from the onset of estrus. Ovulation was detected with the aid of transrectal ultrasonography, performed every 4 h starting 15 h after the onset of estrus. All goats showed estrus. In the MGA group the onset of estrus occurred later and was more variable in relation to the end of treatment (86.7 ± 3.9 h), compared to the FGA group (44.4 ± 1.5 h; P < 0.01). The interval between the LH peak and ovulation was significantly (P < 0.05) longer in the MGA group (26.2 ± 1.1 h) than the FGA group (22.4 ± 0.8 h). There were no differences regarding the intervals from the onset of estrus to the LH peak (14.9 ± 1.8 and 15.3 ± 0.9 h) and to ovulation (40.1 ± 2.3 and 37.6 ± 0.5 h) for the MGA and FGA groups, respectively. The amplitude of the LH peak was not different between groups. It could be concluded that the timing of the LH peak and ovulation is not related to the end of treatment, but with the time of onset of estrus. The longer and more variable interval for estrous demonstration following MGA treatment, represents a serious limitation for its use in synchronization programs in goats, where mating is to be performed on a fixed-time basis.  相似文献

Twenty non-lactating, indigenous Small East African does and postpubertal doelings were injected 125 μg cloprostenol, PG (i.m.). Animals in oestrus were penned with males every night for 45 days. Jugular blood samples for P4 determination were collected immediately before the cloprostenol injection, 4 days later and twice a week for up to 80 days after the end of the mating season. Of the 19 animals with basal P4 concentration 4 days after PG-treatment, only 4 (20%) conceived within 14 days of the treatment. Starting from 4 days after PG-treatment, 1 (5%), 6 (30%) and 8 (40%) of the remaining 15 animals exhibited a short luteal phase, a luteal phase of normal duration or remained anoestrus for 22.2±2.2 days, respectively. The average interval from introduction of the buck to the fertile oestrus for the 19 animals that eventually became pregnant was 23.2±2.3 days (range 9–45). From conception to midgestation the mean plasma P4 concentration ranged from 2.6±1.2 to 10.8±2.4 ng/ml and between days 25 and 35 of pregnancy these values were higher (P < 0.05) in twin-than in singleton-carrying does.It is concluded that for optimal fertility in does of the Small East African goats treated with cloprostenol, mating should be postponed until the first spontaneous oestrus following such treatment.  相似文献

The present study was conducted to determine if the social status of Australian Cashmere goats affects their response to the male effect in terms of LH secretion, ovulation and expression of estrus. Australian Cashmere goats were kept isolated from the males during 5 months. The index of success (SI) of each goat was calculated to establish their social rank. In the first experiment, the ten most dominant and the 10 most subordinate goats were separated from the original herd and housed in two pens (5 dominant and 5 subordinate animals in each pen). An androgenized wether was then introduced into each pen. Luteinizing hormone (LH) was measured every 20 min from 2h before to 4h after introduction of the male in the goats of first pen and from 4 to 8h after male introduction in the second pen. In the second experiment, the remaining 50 goats were exposed in their original pen to two androgenized wethers. Their association index with the males (AI) was calculated for each of these 50 goats, and the intervals from exposure to the males to the onset of estrus and to ovulation were determined. During the first 4h after male introduction, the dominant goats had more LH pulses (0.65+/-0.06 compared with 0.3+/-0.09; P<0.05) and greater LH mean concentrations (1.79+/-0.14 ng/ml compared with 1.30+/-0.15 ng/ml, P=0.05) than the subordinate animals. Although not significantly different, the AI was 35% greater for high and medium ranking goats than for low ranking animals (0.031+/-0.004, 0.032+/-0.005 and 0.023+/-0.005, respectively, P>0.05). Although the number of goats ovulating in response to male exposure was similar between dominance groups (high: 100%, medium: 94% and low ranking: 92%), the high and medium dominance goats showed a greater incidence of expression of estrus than low-dominance goats (94.4%, 89.5% and 53.8%, respectively, P<0.05). It is concluded that the social rank of the Australian Cashmere goat influences their response to the male effect in terms of early LH secretion and expression of estrus.  相似文献

The objectives of this study were to evaluate the effect of a single subcutaneous injection of melatonin, on estrous induction and conception rate during the non-breeding season at different times of the year. In Experiment 1 the melatonin powder was dissolved in an oily vitamin A, D, E solution and injected subcutaneously randomly to goats in two dose treatment groups of 20 mg (MLT-20; n = 20) and 40 mg (MLT-40; n = 20) in January (winter). Twenty does were injected with 1 ml vitamin A and served as the control. In the MLT-20 treated goats 70% of the does were in estrus within 20.0 ± 2.0 days, whereas in the MLT-40 group 85% of the does were in estrus within 15.5 ± 2.5 days and 100% and 80% of the does, respectively, conceived on mating with the bucks. Only 10% of the untreated control does exhibited estrus, but none conceived. The breeding season was thus initiated earlier by 1–1.5 months in the treated goats. In Experiment 2, goats were treated with similar MLT-20 and MLT-40 treatments in May (spring), with 20 goats in each treatment group and 20 control goats. The proportion of goats that responded to the melatonin treatments was 80% and 90% in the MLT-20 and MLT-40 treatments, respectively, with no significant differences recorded regarding the estrous response. However, in the MLT-20 treatment group the estrous induction interval was significantly longer (P < 0.05), compared to the MLT-40 treatment. The pregnancy rates were not significantly different for the MLT-20 and MLT-40 treatments, with melatonin resulting in significantly higher pregnancy rates than in the control (88.4% versus 33.3%) and the breeding season initiated 2 months earlier. It could be concluded that a single subcutaneous injection of melatonin can initiate the breeding season (irrespective of the season of the year) earlier by 1–2 months in goats and this could be advantageous when using accelerated breeding systems.  相似文献

IntroductionHypermetabolism is universal in the severely burned and is characterized by catabolism of lean mass and body fat with associated insulin resistance. Adipokines are likely to play a role in these changes but have not been identified to date in burn patients.MethodsFrom a single burn ICU, 17 burn patients with an expected stay >14 days were studied. Study period began within 14 days of admission. Over 7 days, plasma samples were collected for measurement of leptin, adiponectin, resistin, ghrelin, insulin, and cortisol by ELISA. For comparison, samples from 15 healthy controls of similar age, BMI, and blood glucose were obtained.ResultsMean age was 33 ± 17 years and BMI 26 ± 3.4. Average burn size was 45 ± 20% TBSA and ISS 32 ± 10 with 72% having inhalation injury; in-hospital mortality was 29%. Estimated energy needs were 3626 ± 710 kcal, of which 84 ± 21% were met by enteral feeding with intensive insulin treatment (glucose 80–110 mg/ml). Using the homeostasis model assessment of insulin resistance, burned subjects were more resistant than controls (17 ± 11.3 and 8 ± 10.0). Insulin levels were elevated (57 ± 35.6 μU/ml in burned subject vs. 26 ± 31.1 μU/ml in controls), and cortisol concentrations increased (50 ± 41.2 μg/dl vs. 12 ± 3.9 μg/dl). These traditional hormone changes were associated with increased resistin (16.6 ± 5.5 ng/ml vs. 3.8 ± 0.9 ng/ml) and decreased leptin (8.8 ± 8.9 ng/ml vs. 19.4 ± 23.5 ng/ml), adiponectin (9 ± 3.5 ng/ml vs. 17 ± 10.2 ng/ml), and ghrelin (0.37 ± 0.14 ng/ml vs.0.56 ± 0.26 ng/ml).ConclusionPatients with burns, who are characteristically hypermetabolic with hypercortisolism and insulin resistant, have significant changes in adipokine levels that appear independent of the magnitude of initial injury or metabolic derangement. In addition, suppression of ghrelin in the presence of decreased leptin and adiponectin levels in combination with increased insulin and resistin levels represent unexpected changes in the metabolic milieu of the injured patient possibly due to dramatic activation of inflammatory pathways, indicating strategies for treatment.  相似文献

The use of exotic dairy goats in breeding programmes for smallholder production systems is popular in Eastern Africa. However, information on the performance of exotic breeds within these systems is scarce. This paper presents information on performance of Toggenburg dairy goats under smallholder production systems in a medium to high agricultural potential environment in Kenya under a community-based set-up as part of the characterization of its adaptive and productive attributes. Environmental factors affecting growth of 646 goats born in the environment, and fertility and milk production of 160 does from 1997 to 2005 were evaluated. Genetic parameters were also estimated for early growth traits. The average birth weight (BW) for 607 kids was 3.27 kg. Corrected weaning weights (CW), and average daily gain to weaning (ADG) for 646 kids were 19.12 kg and 136 g/day, respectively. The results indicated that the smallholder farmers were able to maintain comparably high levels of milk production in the first three parities (LMY), with yields of 475 ± 9 l in 201 days for 160 does in the first parity, 507 ± 9 l in 264 days from 130 does in the second parity and 513 ± 13 l in 296 days for 82 does in the third parity. Kidding intervals though initially long decreased with time to reasonable levels (302 ± 117 days). The heritability estimates obtained were low (0.23 ± 0.13 for BW, 0.18 ± 0.11 for CW and 0.14 ± 0.11 for ADG). Genetic correlations between the traits were also low. Genetic and phenotypic trends indicated little change in BW, CW and LMY over the years. The phenotypic trend in the kidding interval showed a reducing interval over time. The results demonstrated that the Toggenburg goats were able to perform and thrive reasonably well under the low-input farming conditions.  相似文献

This study was done to verify if the lack of increase of milk fat content observed in Alpine dairy goats under once daily milking (ODM) compared to twice daily milking (TDM), results from disturbances of animals and/or milk ejection reflex. In this respect, we determined the milk yield and composition in the same 12 multiparous Alpine dairy goats when they were managed first under TDM (period 1: P1) and then under ODM (period 2: P2). Furthermore, oxytocin (OT) and cortisol (CORT) releases and milk emission kinetic of these goats were measured at morning milking 2 and 4 times in P1 and P2, respectively.ODM compared to TDM, caused 18 and 23% reductions, respectively in daily milk yield and milk fat content without milk protein content modification.Although baseline concentration of blood OT was lower under ODM than TDM management (7.0 pg/mL vs 17.8 ± 2.0 pg/mL, respectively), ODM did not modify the total amount of OT released during milking (15,484 pg/mL/32 min vs 18,996 ± 2865.3 pg/mL/32 min, respectively), the peak concentration of OT (57.2 pg/mL vs 73.6 ± 10.5 pg/mL, respectively) or the time to reach it (3.0 min vs 2.0 ± 0.5 min, respectively) by comparison to TDM.ODM compared to TDM, never modified the baseline concentrations of CORT (6.1 ng/mL vs 7.1 ± 1.1 ng/mL, respectively), the total amount of CORT released (11,727 ng/mL/32 min vs 10,073 ± 1522.2 ng/mL/32 min, respectively), the peak concentrations of CORT (16.1 ng/mL vs 15.1 ± 2.1 ng/mL, respectively) and the time to reach it (13.1 min vs 11.0 ± 1.5 min).During ODM, milk flow latency was reduced (−61%) while the mean flow rate was increased (+28%) by comparison to TDM. ODM compared to TDM management, did not modify the maximum flow rate (1.6 L/min vs 1.6 ± 0.2 L/min, respectively) or the time to reach this maximum (103.0 s vs 95.0 ± 10.0 s, respectively). The total milking duration at morning milking was not different (250.0 s vs 221.0 ± 22.0 s, respectively) although the morning milk yield was significantly higher under ODM management (2.63 kg/d vs 1.87 ± 0.1 kg/d, respectively).This results show that milk emission is improved under ODM management in Alpine goats without inhibition or disturbance of neuro-hypophyseal OT release pattern and lack of activation of the hypothalamic–pituitary–adrenal axis and plasma cortisol increase, disproving our initial hypotheses of a disturbance of animals and/or incomplete milk ejection to explain the low fat content of milk under ODM in this breed.  相似文献

Progesterone concentration was measured in specific brain regions of the ram and anestrous ewe to provide reference values for future studies to investigate if local CNS lesions resulting from neurodegenerative diseases of sheep are associated with progesterone loss. Using radioimmunoassay, plasma progesterone was recorded throughout all brain regions assayed. No significant differences were found between the ewe and ram for any brain regions. There were however, significant differences between the different regions of an individual brain (P < 0.05). Plasma progesterone concentration for the highest to the lowest value recorded was as follows: 2.93 ± 0.85 and 2.77 ± 0.51 ng/g in the frontal cortex; 2.33 ± 0.67 and 2.25 ± 0.48 ng/g in the parietal cortex; 1.32 ± 0.36 and 1.29 ± 0.35 ng/g in the temporal cortex; 1.25 ± 0.32 and 1.25 ± 0.31 ng/g in the occipital cortex; 1.24 ± 0.30 and 1.23 ± 0.31 ng/g in the corpus callosum; 1.16 ± 0.30 and 1.21 ± 0.38 ng/g in the cerebellum; 1.09 ± 0.30 and 1.12 ± 0.39 ng/g in the medulla oblongata of the ewe and ram, respectively. Plasma progesterone concentration in the ewe (0.28 ± 0.06 ng/ml) was significantly higher than that in the ram (0.10 ± 0.03 ng/ml) (P < 0.001). Furthermore, plasma progesterone concentration in all sheep was several times lower than that of any regions of the brain. The results indicate that the sheep brain accumulates progesterone in significant levels, which may be independent of the circulating progesterone. The brain progesterone concentration in CNS regions assayed was similar for the ram and anestrous ewe. Neurodegenerative processes in visna, border disease and enzootic ataxia should be questioned in further studies if they are associated with local progesterone loss.  相似文献

This study aimed to evaluate the effects of betacarotene (BC) supplementation on ovulation rate (OR) and luteinizing hormone (LH) secretion in adult goats during the breeding season. Additionally, total ovarian activity (TOA) comprising the total number of ultrasonographically detectable antral follicles (AF) and corpora lutea (OR) was also assessed. In early October, adult goats [n = 22, 3.5 years of age, 7/8 Sannen-Alpine; 26°N, 103°W at 1117 m.a.s.l.] were randomly assigned to: (i) BC group (BCG), orally supplemented with 50 mg of BC/goat/day [n = 10; live weight (LW) = 45.9 ± 2.0 kg, body condition score (BCS; range: 0-emaciated to 5-obese) = 3.0 ± 0.1], and (ii) control group (CONT) [n = 12; LW = 46.2 ± 2.0 kg, BCS = 3.0 ± 0.1]. All animals received a basal diet of alfalfa hay, corn silage and corn grain, with free access to water and mineral salts. The whole experimental period spanned 34 days before and 17 days after ovulation. On day 23 of the experiment, estrus was synchronized with progestin-releasing intravaginal sponges; 36 h prior to estrus, an intensive blood sampling (every 15 min for 6 h) was performed to determine mean LH concentrations, pulsatility (LH-PULSE) and area under the curve (LH-AUC) for serial LH concentrations. Afterwards, by the end of the luteal phase (i.e., 17 days after the onset of estrus), an ultrasonographic scanning was performed to evaluate OR and TOA [AF + OR]. The average LW and BCS did not differ (p > 0.05) during the experimental period. BC-supplemented goats showed an increase in OR (3.4 ± 0.2 versus 2.8 ± 0.2; p < 0.05) and exhibited lower (p < 0.05) serum LH concentrations, LH-AUC and LH-PULSE compared to CONT. A positive correlation was recorded between OR and LW (r2 = 0.42, p < 0.05) and BCS (r2 = 0.47, p < 0.05). In addition, AF (5.0 ± 0.6 versus 3.4 ± 0.6) and TOA (8.4 ± 0.6 versus 6.2 ± 0.6) were greater (p < 0.05) in the BC-supplemented group than CONT. Supplementation with BC enhanced ovarian follicular development and ovulation rate in adult female goats under decreased photoperiods through LHRH-independant pathways or direct effects of BC on ovarian function.  相似文献

The objective of this study was to investigate the regulation of water balance and dry matter (DM) intake of male Ethiopian Somali goats. Twenty-eight goats were grouped into four treatments; goats watered every day (W1), every 2nd day (W2), every 3rd day (W3) and every 4th day (W4) for a total period of 72 days. Plasma sodium and potassium were analyzed by flame photometry, plasma osmolality by freezing point depression, plasma protein concentration by TS refractometry, and the plasma vasopressin concentration by radioimmunoassay. Goats were fed grass hay ad libitum and were given 200 g of concentrates daily. The calculated daily water intake (ml/kg BW0.75) of W1, W2, W3 and W4 was 128 ± 4, 86 ± 4, 88 ± 4 and 78 ± 4, respectively, (P < 0.001, W1 versus the other groups). Calculated total concentrates intake did not change, whereas Group W2 and W4 consumed less hay on days of water deprivation (W1 versus W2, P < 0.01; W1 versus W4, P < 0.001). Plasma sodium concentration of W1 was 143 ± 1 mmol/l. Intermittent watering increased plasma sodium concentration from 140 or 141 ± 1 mmol/l to 145 or 146 ± 1 mmol/l every 2nd day in group W2. In W3, it increased from 139 ± 1 to 146 ± 1 mmol/l and in W4 from 137 ± 1 to 149 ± 1 mmol/l (P < 0.001 for all) on days after watering compared to last day of water deprivation, respectively. Corresponding changes were observed in plasma osmolality. Total plasma protein concentration increased only in W3 from 65 ± 1 to 67 ± 1 mmol/l on day 3 (P < 0.01) and in W4 from 65 ± 1 to 67 and 68 ± 1 mmol/l on days 3 and 4, respectively (P < 0.05). The plasma vasopressin concentration of W1 was close to 0.80 ± 0.28 pmol/l on all days, but it increased on dehydration days in W2 (P < 0.05). In W3, the vasopressin concentration increased from 0.95 or 1.19 pmol/l the day after watering to 3.11 ± 0.28 pmol/l on day 3 (P < 0.01). In W4, it was 0.82 ± 0.31 pmol/l the day after watering and reached 6.06 ± 0.31 pmol/l on day 4 (P < 0.001). We conclude that male Ethiopian Somali goats can be watered once every 3rd days, and that longer interval is not recommended because it demands marked mobilization of water saving mechanisms and causes decreased DM intake.  相似文献

Twenty-four Boer goat does were used to compare three superovulation protocols, with 8 does allocated per treatment during the natural breeding season. In Group 1 (Day 0 protocol), the oestrous cycles of does were synchronised for 7 days with CIDR's and injected PGF2α at CIDR insertion. Does were then superovulated with pFSH in 7 dosages at 12 h intervals, starting 88 h following CIDR removal. Concurrently with the 6th dosage, does were injected PGF2α. Cervical inseminations were performed 24 h and 36 h following the last superovulatory treatment. For Groups 2 and 3, the oestrous cycles of the does were also synchronised for 17 days using CIDR's. On day 14 of CIDR insertion, Group 2 does were injected with PGF2α. A superovulation treatment similar to Group 1 was administered in Groups 2 and 3, starting 48 h before CIDR removal. All does in these groups were also cervically inseminated with fresh undiluted Boer goat semen 24 h and 36 h following CIDR withdrawal. Embryos from all 3 treatment groups were flushed on day 6 following AI. Does in Group 1 responded to the short oestrous synchronisation protocol before the administration of a superovulation treatment (71.4% response), with time to onset of oestrus of 37.2 ± 0.7 h and duration of an induced oestrous period of 36.4 ± 0.5 h being recorded. Following superovulation only two does exhibited signs of oestrus in Group 1, while Groups 2 and 3 exhibited a 100% oestrous response. Groups 1 and 2 recorded similar intervals to the onset and durations of the induced oestrous period. The number of ovulations per donor was significantly lower in Group 1 (4.0 ± 0.7), compared to Groups 2 and 3 (14.5 ± 0.6 and 16.5 ± 0.8, respectively), with no significant difference between Groups 2 and 3. The Day 0 protocol (Group 1) also resulted into a significantly lower total number of structures recovered, compared to Group 3. Groups 2 and 3 recorded a relatively similar number of structures recovered. The number of embryos recovered was significantly lower (P < 0.01) in Group 1 (0.2 ± 0.1) than in Group 2 (13.2 ± 0.5) and Group 3 (11.5 ± 1.1), with the mean number of unfertilised ova and degenerated embryos being similar for all 3 treatment groups. Groups 2 and 3 also produced a similar number of transferable embryos. The blood progesterone concentrations followed a similar trend in the 3 treated groups, from CIDR insertion to embryo flushing. However, the mean serum progesterone concentration was significantly lower on day 4 in the Day 0 group, compared to Groups 2 and 3. The inclusion of PGF2α treatment in the superovulation protocol for Boer goats had no beneficial effect, while the Day 0 protocol engaged in this trial, resulted in a lower superovulation response. Further research is warranted, focusing on synchronisation, time when initiating superovulatory treatment and AI to improve the embryo yield in goats.  相似文献

Sulfur mustard (SM) is an incapacitating chemical warfare agent that was used against Iranian soldiers during the period from 1983 to 1988. We have investigated serum cytokines profiles of Khorasan veterans who were exposed to SM >23 years earlier. Forty-four male Iranian veterans who had >40% disabilities due to delayed complications of SM poisoning and had disabilities were investigated. A total of 30 healthy male volunteers (relatives of the veterans) were selected as the control group. Cytokine levels were measured in the serum of case and control subjects using commercial ELISA kits. Hematologic parameters (white/red blood cell counts, hemoglobin levels, immune cell differentials) were also performed on blood samples from the study subjects. The results indicated that serum levels of ICAM-1 were significantly higher in the samples from SM-exposed veterans (772.8 [±15.1] ng/ml [p = 0.014] vs. control values of 710.2 [±20.0] ng/ml). On the other hand, serum IL-1β, IL-8 levels and TNFα, were significantly lower for the veterans than the controls (IL-1β: 3.8 [±0.1] vs. 4.3 [±0.2] pg/ml, p = 0.037; IL-8: 21.0 [±6.1] vs. 84.6 [±20.3] pg/ml, p = 0.002; TNFα: 4.5 [±0.1] vs. 5.5 [± 0.1] pg/ml, p = 0.027). Levels of other assayed cytokines, e.g., IL-2, -4, -5, -6, -10, and -12, IFNγ, TNFβ, and sVCAM-1 were not significantly different between the study populations. None of the assayed hematologic parameters appeared to differ as well. It seems possible that dysfunctions could have been induced in the innate immune functions of the SM-exposed veterans as a result of these changes in cytokine expression and that these, in turn, may have contributed to the increased incidence of a myriad of diseases that have been documented in these veterans, including cancers. Future studies must focus on examining the significance of these changes in circulating cytokines and their potential contribution to the development of different diseases in veterans exposed to SM.  相似文献

This study investigated the effect of dietary supplementation with calcium salts of long chain fatty acids with or without of l-carnitine on ovarian activity using 24 Rahmani ewes randomly allocated to four treatments. Control animals (n = 6) were fed a basal diet of hay (64.2%) and barley grain (35.0%) plus minerals and vitamins (0.8%). Ewes on the three treatments received the same basal diet supplemented with calcium salts of long chain fatty acids (CSFA) at 3% of the basal diet dry matter intake (1.4 kg/ewe/d); 250 ppm l-carnitine (LC); or both these supplements (CSFA + LC). All use exhibited natural estrus on one or two occasions and were weighed at the start and the end of the study as well as body condition score was assessed at the end of study. All ewes were then synchronised for estrus using intravaginal sponges for 12 d prior to the start of the nutritional treatments and three weeks after the nutritional treatments began. The nutritional treatments were imposed for a total of 8 weeks. Blood samples were collected prior to the start of treatments and every two weeks thereafter except after sponge removal of first and second synchronisation where the blood samples were collected daily for progesterone assay. The results revealed that Rahmani ewes received basal diet (control) and l-carnitine had significantly decrease final body weight and body condition score (36.3 ± 0.4; 36.8 ± 0.3; 2.2 ± 0.04; 2.1 ± 0.05; p < 0.05, respectively) than those on CSFA and CSFA + LC (38.6 ± 0.9; 39.5 ± 0.6; 3.3 ± 0.07; 3.4 ± 0.06; respectively). At the second ultrasound examination, the control animals had significantly fewer total follicles (7.3 ± 0.8; p < 0.05) than those on the CSFA (8.4 ± 0.8), l-carnitine (8.7 ± 1.5) and CSFA + LC (8.0 ± 0.6) treatments. The increased numbers occurred in the medium and large categories of follicles. In addition, the ovulation rates were significantly lower (p < 0.05) for control (1.3 ± 0.2) and l-carnitine (1.5 ± 0.00) than for CSFA (2.5 ± 0.3) and CSFA + LC (2.3 ± 0.2). Furthermore, serum progesterone concentrations had risen and were significantly higher (p < 0.05) for CSFA (2.5 ± 0.3 ng/ml) and CSFA + LC (2.7 ± 0.1 ng/ml) than for control (1.1 ± 0.7 ng/ml) and l-carnitine (1.5 ± 0.4 ng/ml). It was concluded that supplementation of the basal diet with l-carnitine alone did not improve performance of ewes or the ovarian response. However, the addition of calcium salts of long chain fatty acids to the basal diet alone or in combination with l-carnitine significantly improved the number and size of ovarian preovulatory follicles, and the ovulation rate of Rahmani ewes. Further evidence was required to study their influence on follicular atresia.  相似文献

Twenty-four Boer × Spanish does (3 years of age, having kidded once previously and with an initial BW of 42.7 ± 1.2 kg) were used to determine the efficiency of ME utilization for pregnancy (kpreg). Six does were nonpregnant and, based on ultrasound determination on day 45 of gestation, six had a litter size (LS) of 1, 2, and 3. However, only 10 of the pregnant does delivered the expected number of kids (3, 4, and 3 with LS of 1, 2, and 3, respectively). Does were fed a diet of approximately 50% concentrate in accordance with assumed maintenance plus pregnancy energy requirements based on estimated nonpregnancy tissue BW and LS. Recovered energy (RE) was determined by subtraction of energy expenditure (EE; respiration calorimetry) near days 80, 100, 120, and 140 of gestation from ME intake (MEI). RE was assumed attributable to pregnancy tissues (fetus, fetal fluids and membranes, uterus, and mammary gland), and ME used for pregnancy (MEpreg) was estimated by subtracting MEm determined with nonpregnant goats from MEI by those pregnant. For does with actual LS equal to that expected, the no-intercept equation for the regression of RE against MEpreg was: RE = MEpreg × 0.252 (S.E. = 0.030; R2 = 0.64), indicating a kpreg of 25%. A regression including LS (1 versus 2 or 3) suggested greater kpreg for LS of 1 (40.2 ± 5.6%) versus 2 or 3 (20.5 ± 3.2%). Regressions for goats with LS different from expected suggested positive effects of use of energy mobilized from nonpregnancy tissues on kpreg and of use of dietary ME for energy accretion in nonpregnancy tissues on the efficiency of whole body ME utilization. In conclusion, the average efficiency of ME use for pregnancy regardless of LS in goats was near 25%, which when considering the expected proportion of all pregnancy tissues attributable to fetal or conceptus tissues implies an energy requirement for pregnancy of goats similar to common recommendations for sheep and cattle.  相似文献

The continuous evaluation and genetic improvement of fleece traits in Angora goats are of major importance to the Angora goat industry. The objective of this study was to estimate variance components and genetic parameters for both physical and quality traits of mohair in South African Angora goats. The data used for this study were collected on between 898 and 6211 kids (depending on the trait measured) born between 2000 and 2006 in 11 different Angora goat studs. Variance components and genetic parameters were estimated with the ASREML programme for fleece weight, fibre diameter, coefficient of variation of fibre diameter, standard deviation of fibre diameter, comfort factor, spinning/effective fineness and standard deviation of fibre diameter along the length of the staple. Most of these traits (excluding fleece weight) are measured using OFDA technology exclusively, and this study was the first attempt to calculate genetic parameters for these traits in South Africa. A model including the direct additive genetic effects only, were the most appropriate for estimation of all parameters. Heritability estimates were 0.24 ± 0.03 for fleece weight, 0.45 ± 0.03 for fibre diameter, 0.37 ± 0.10 for coefficient of variation of fibre diameter, 0.32 ± 0.11 for standard deviation of fibre diameter, 0.63 ± 0.10 for comfort factor, 0.61 ± 0.10 for spinning/effective fineness and 0.14 ± 0.08 for standard deviation of fibre diameter along the length of the staple. Results from this study can now be applied for the estimation of breeding values for fleece production of South African Angora goats and in incorporation into selection programs.  相似文献

High yielding dairy cows experience a negative energy balance (NEB) early post-partum and it was hypothesized that this may be aggravated under summer heat stress (HS) conditions. In this study, which was performed in Egypt, 20 Holstein cows were followed during summer (n = 10) and winter (n = 10) seasons. All cows were multiparous and kept at the same herd. Blood was sampled from each cow starting 1 week before the expected calving date and then at 1-week intervals until week 6 post-partum. From week 2 to 6 post-partum follicular fluid was collected through transvaginal follicular fluid aspiration at 6 days intervals. Ambient air temperature (AT) and relative humidity (RH) were recorded and temperature–humidity index (THI) was calculated as well. Respiration rate (RR), rectal temperature (RT), and body condition score (BCS) were recorded for each cow at the time of blood sampling. Concentrations of glucose, insulin like growth factor-1 (IGF-1), non-esterified fatty acids (NEFA), urea and total cholesterol (TC) were measured in each blood and follicular fluid sample. All the cows showed a significantly higher RR and RT in summer (95.5 ± 1.1 and 39.88 ± 0.06, respectively) than in winter (43.89 ± 0.61 and 38.94 ± 0.07, respectively) (P < 0.001). Body condition score loss during the early post-partum period was higher in summer than in winter (1.1 ± 0.07 vs. 0.85 ± 0.06 point, respectively) (P < 0.001). The average dominant follicle diameter was significantly lower in summer than in winter during the period of negative energy balance (11.6 ± 0.7 mm vs. 15.3 ± 1.2 mm, respectively) (P < 0.01). Under summer heat stress, the concentrations of glucose (2.98 ± 0.07 and 2.19 ± 0.04 mmol/L), IGF-1 (106.7 ± 2.9 and 99.0 ± 3.4 ng/ml) and TC (137.3 ± 5.3 and 62.2 ± 5.1 mg/dl) in blood and FF, respectively, were significantly lower than winter concentrations by (0.17 ± 0.03 mmol/L, P < 0.001 and 0.26 ± 0.06 mmol/L, P < 0.001), (12.3 ± 3.6 ng/ml, P < 0.001 and 9.0 ± 2.7 ng/ml, P < 0.001) and (20.7 ± 1.8 mg/dl, P < 0.001 and 7.3 ± 1.1 mg/dl, P < 0.01), respectively. However, the concentrations of NEFA (0.68 ± 0.14 and 0.22 ± 0.02 mmol/L) and urea (9.27 ± 0.34 and 9.96 ± 0.25 mmol/L) in blood and FF, respectively, were significantly higher in summer compared to winter (0.50 ± 0.08 mmol/L, P < 0.001 and 0.20 ± 0.02 mmol/L, P < 0.001) and (8.77 ± 0.23 mmol/L, P < 0.05 and 8.96 ± 0.29 mmol/L, P < 0.001), respectively, throughout the experimental period. The results of the present study indicate that heat stress early post-partum aggravates NEB in high yielding dairy cows, reduces BCS, dominant follicle diameter and alters the biochemical concentrations in the follicular fluid of the dominant follicle which may result in inferior oocyte and granulosa cell quality and hence poorer fertility.  相似文献
