The aim was to investigate the detection rates of periodontal bacteria (Porphyromonas gingivalis, Tannerella forsythia, Prevotella intermedia, and Aggregatibacter actinomycetemcomitans) and herpesviruses (herpes simplex virus-1 [HSV-1], cytomegalovirus [CMV], and Epstein-Barr virus [EBV]) in different forms and severity of periodontal disease, and to compare them with those in periodontally healthy subjects. One hundred and twenty-nine patients participated in the study: 39 diagnosed with periodontal abscess (PA), 33 with necrotizing ulcerative periodontitis (NUP), 27 with chronic periodontitis (CP), and 30 participants with healthy periodontal tissue represented a healthy control group. All patients with periodontal disease (PA, NUP, and CP) were also divided into two groups according to the severity of their disease: moderate and severe periodontitis. The subgingival samples were collected from the periodontitis active sites and the detection of microorganisms was performed by end-point polymerase chain reaction analyses. The results revealed significantly higher detection rates of P. gingivalis, T. forsythia, and P. intermedia in all three groups of patients with periodontitis than in healthy participants. The highest detection rate of A. actinomycetemcomitans was noticed in CP, which was significantly higher than that in PA, NUP, and healthy control. The occurrence of EBV was significantly higher in NUP than in CP and healthy participants. CMV was detected significantly more frequently in PA and NUP than in CP and healthy participants. Comparisons among healthy participants and patients with moderate and severe periodontitis showed significantly higher detection rates of EBV and CMV in patients with severe forms of periodontitis than in healthy participants and those with moderate periodontitis. 相似文献

The biosynthesis of xanthan by Xanthomonas campestris was found to be affected by the addition of citric acid in fed batch mode. Under oxygen-limiting conditions, the addition of up to 2.6 g citric acid per litre improved cell viability as well as increasing xanthan yield by up to 80%. Comparative xanthan formation profiles at different operating conditions indicate that at higher aeration (when there was no oxygen limitation), citric acid addition did not improve xanthan production. 相似文献

溶剂法直接提取的黄原胶产品含有大量菌体蛋白和色素杂质,总氮含量高、透明度差、色泽暗深。通过中性蛋白酶处理发酵液,使成品含氮量下降56.5%;通过Na2SO3漂白液,使产品色泽大为改善。实验得出最佳酶解条件:发酵液稀释1倍,温度44℃、pH7,加酶量100u/g发酵液,作用时间2.5h;最佳Na2SO3漂白条件:温度20 ̄30℃,pH5 ̄6,Na2SO3用量为1%(w/w),作用时间1 ̄1.5h。 相似文献

Aqueous NaCl solutions of dimerized Na xanthan with salt concentrations of 0.005, 0.01 and 0.1
were exposed to 80°C for different time periods t, and their viscosities were determined as a function of t. The measured relative viscosities decreased markedly with t, suggesting that Na xanthan denatured at 80°C undergoes some conformation changes or degradation. The molecular weights of the test samples recovered at different t were estimated by viscometry in cadoxen, a single-coil solvent for xanthan, and were found to decrease monotonically with t. Thus, it was concluded that the observed decreases in relative viscosity are due primarily to degradation of Na xanthan. 相似文献

The purpose of this research was to reduce the polymer concentration and to obtain reasonable viscosity at a lower concentration
of pluronic by the addition of a viscosity modifier. A 20% wt/wt pluronic gel was prepared on a weight basis using the cold
method. The effect of the amount of tetracycline and Aerosil on gel properties was studied. The gel was evaluated using different
parameters: polarizing microscopy, gelation, gel melting, bioadhesivity, viscosity, drug release, and stability of enzyme.
An in vivo study was performed to evaluate the clinical efficiency of the liquid crystalline gel. Addition of Aerosil to the
gel favored hexagonal phase formation. Viscosity and bioadhesivity increased with an increase in the concentration of Aerosil.
Release of tetracycline was sustained as the concentration of Aerosil increased. Various clinical parameters confirmed the
acceptability and efficiency of this gel system.

Several mutants of Xanthomonas campestris showing increased viscosity and/or gum production were detected after UV treatment. Xanthan solutions of the different mutants
showed different intrinsic viscosity values, and no relationship was found between pyruvate or acetate contents and viscosifying
ability of the xanthan. The best performance mutant (M-11) was obtained using halo size around colonies in starch-agar plates
as the detection criterion, and proved the usefulness of this indirect screen for improved xanthan producers. A pleiotropic
effect of this mutation on growth rate and total cell growth was observed.

Transient electric birefringence studies have been performed on heat denatured xanthan in 4 m urea. The induced birefringence was positive, the Kerr law was obeyed at low field amplitudes and the birefringence saturated at high fields. The orientation mechanism appears to be mainly induced dipolar in character and the magnitude of the induced dipole moment can be explained on the basis of counterion polarization. The molecules behave as independent rods of mean length 0.65 μm with no evidence for ‘hindered rotation’ in moderately concentrated solutions. The molecular rigidity is attributed to extension of the polyanion due to charge charge repulsions or steric hindrance due to the side chains. 相似文献

of Xanthomonas campestris, xanthan biosynthesis, kinetics and chemical composition were investigated. Both growth and xanthan production were dependent
on the acid hydrolysate concentrations and available nitrogen. Melon acid hydrolyzed waste was the best substrate for xanthan
production. Exopolysaccharide obtained throughout this study was compared to commercial xanthan, showing a very similar chemical
composition. Acid hydrolyzed wastes are proposed as a new carbon source for xanthan production.

Batch fermentation kinetics of xanthan gum production from glucose by Xanthomonas campestris at temperatures between 22 degrees C and 35 degrees C were studied to evaluate temperature effects on cell growth and xanthan formation. These batch xanthan fermentations were modeled by the logistic equation for cell growth, the Luedeking-Piret equation for xanthan production, and a modified Luedeking-Piret equation for glucose consumption. Temperature dependence of the parameters in this model was evaluated. Growth-associated rate constants increased to a maximum at approximately 30 degrees C and then decreased to zero at approximately 35 degrees C. This temperature effect can be modeled using a square-root model. On the contrary, non-growth-associated rate constants increased with increasing temperature, following the Arrhenius relationship, in the entire temperature range studied. The model developed in this work fits the experimental data very well and can be used in a simulation study. However, due to the empirical nature of the model, the parameter values need to be reevaluated if the model is to be applied to different growth conditions. 相似文献

This work concerns the activity coefficient determination for sodium and calcium ions in a semi-dilute solution of xanthan when they are mixed in various ratios, and their correlation with the conformational structures. From these values we show that the conformation can be related to the dissociation of the polymer carboxylic acid sites. Below an apparent dissociation coefficient of about 0.29, the conformation is ordered and above 0.55 the xanthan conformation is entirely disordered. The value of the counterion activity coefficient in pure solution of sodium or calcium xanthan salts agrees fairly well with the theoretical ones derived from Manning's and Lifson-Katchalsky's theories if we make the assumption that the Na form is a stretched coil and the Ca form a single fivefold helix. 相似文献

Xanthan gum, an anionic polysaccharide with an exceptionally high molecular weight, is produced by the bacterium Xanthomonas sp. It is a versatile compound that has been utilized in various industries for decades. Xanthan gum was the second exopolysaccharide to be commercially produced, following dextran. In 1969, the US Food and Drug Administration (FDA) approved xanthan gum for use in the food and pharmaceutical industries. The food industry values xanthan gum for its exceptional rheological properties, which make it a popular thickening agent in many products. Meanwhile, the cosmetics industry capitalizes on xanthan gum's ability to form stable emulsions. The industrial production process of xanthan gum involves fermenting Xanthomonas in a medium that contains glucose, sucrose, starch, etc. as a substrate and other necessary nutrients to facilitate growth. This is achieved through batch fermentation under optimal conditions. However, the increasing costs of glucose in recent years have made the production of xanthan economically unviable. Therefore, many researchers have investigated alternative, cost-effective substrates for xanthan production, using various modified and unmodified raw materials. Metastable and stable states of xanthan polyelectrolyte complexes studied by atomic force microscopy
The compaction of the semiflexible polysaccharide xanthan with selected multi- and polyvalent cations was studied. Polyelectrolyte complexes prepared at concentrations of 1-2 microg/ml were observed by tapping mode atomic force microscopy. High-molecular-weight xanthan compacted with chitosan yields a blend of mainly toroidal and metastable structures and a small fraction of rod-like species. Polyelectrolyte complexes of xanthan with polyethylenimine and trivalent chromium yielded similar structures or alternatively less well packed species. Racquet-type morphologies were identified as kinetically trapped states occurring on the folding path toward the energetically stable state of the toroids. Thermal annealing yielded a shift of the distribution of xanthan-chitosan morphologies toward this stable state. Ensembles of toroidal and rod-like morphologies of the xanthan-chitosan structures, collected using an asphericity index, were analyzed. The mean height of the toroids increased upon heating, with a selective increase in the height range above 2 nm. It is suggested that the observed metastable structures are formed from the high-molecular-weight fraction of xanthan and that these are driven toward the toroidal state, being a low-energy state, following annealing. Considered a model system for condensation of semiflexible polymers, the compaction of xanthan by chitosan captures the system at various stages in the folding toward a low-energy state and thus allows experimental analyses of these intermediates and their evolution. 相似文献

为了提高黄原胶的速溶性和粘度,将黄原胶进行改性处理。将黄原胶与马来酸酐进行酯化反应,探讨了黄原胶与马来酸酐摩尔比、反应时间和反应温度等因素的影响,以取代度为指标,利用响应面方法确定,该酯化反应的最优条件为:黄原胶与马来酸酐摩尔比1∶11.5、反应时间24.4 h、反应温度66℃。对改性黄原胶进行红外光谱、光散射和X-射线衍射等结构表征,表明酯化改性成功,且进一步解释了速溶性和粘度提高的原因。改性黄原胶细胞毒性实验,显示无毒性。结果表明,改性黄原胶的速溶性和粘度有很大提高,0.2%改性黄原胶的速溶性和粘度较对照提高了近3倍,在食品、药品等领域具有潜在的应用价值。 相似文献

The Ca2+-induced conformational changes of the extracellular microbial polysaccharide xanthan, and of some derivatives, have been investigated by circular dichroism and isothermal microcalorimetric methods. The four polymers studied are native xanthan (NX), acetyl-free xanthan (AFX), pyruvic-free xanthan (PFX) and acetyl-and-pyruvic-free xanthan (APFX), all of about the same molecular weight. Convergent evidence from both techniques indicates that the acetyl group stabilizes the ordered conformation of xanthan, which can be induced by specifically increasing the ionic strength of the dilute aqueous solution by the addition of calcium ions. Pyruvate groups have been shown to have a strong destabilizing effect on the ordered conformation, which is likely to be ascribed to an unfavourable electrostatic contribution. The relative order of stability of the ordered forms was found to be PFX > NX > APFX > AFX; PFX is largely present in the ordered conformation at 25°C even in the calcium-free aqueous solution. 相似文献

AbstractThis work aims to enhance the bioproduction of xanthan gum by screening a hyper-yield producer from the wild-type Xanthomonas campestris during a long-term continuous subculture. We reported a cell-wall deficient mutant, which performed a shift of cell morphology from rod-shaped to round-shaped. Both the yield of xanthan gum and the conversion rate of feedstock were assessed using sucrose as a carbon source with the supplement of yeast extract powder, l-glutamic acid, and other raw materials. After 96?h aerobic fermentation, the yield of xanthan gum of the mutant reached up to 32?g/L, which was 3.4 times of that of the wild-type strain. The conversion rate of feedstock in the mutant was up to 92.1%, which was 3 times of that of the wild-type (31.2%). Furthermore, pigments generated were determined and compared. As a result, the fermentation broth of the wild-type performed an OD560nm of 0.296, which was 5.8 times of that (OD560nm?=?0.051) of the mutant. Microscopy analysis showed that the percentage of free-living cells in broth affected the color of the final product. Moreover, the robustness of the fermentation performance of the cell-wall deficient mutant at a pilot scale showed potential for industrial application. 相似文献

It is demonstrated using gamma-radiolytic and photolytic techniques that, when O2?- is produced in the presence of catalytic amounts of iron(II) and iron(III) complexes depolymerization of xanthan in aqueous solution occurs. The use of these techniques also allows a quantitative measurement of the efficiency of conversion of O2?- to ?OH in these systems to be made, with the assumption that ?OH is the most likely depolymerizing species present. Using pulse radiolysis and oxidative-reductive depolymerization, the effects of some common anions on the degradation process have been correlated with the reactivities of their related free radicals with xanthan. This study aims to evaluate the in vitro antimicrobial and immunomodulatory activities and cytotoxicity of chlorhexidine (CHX) and synoeca-MP peptide alone or in combination against Pseudomonas aeruginosa. The antimicrobial property was evaluated by the determination of minimal inhibitory concentration, minimum bactericidal concentration, and planktonic bacteria and biofilm inhibition. Immunomodulatory activity was determined by enzyme-linked immunosorbent assay and nitric oxide production by the Griess reaction method. According to the results, synoeca-MP combined with CHX demonstrated antimicrobial effectiveness compared with its isolated use, in addition to immunomodulatory activity (upregulating MPC-1 and tumor necrosis factor-α and downregulating nitric oxide and interleukin-10). In this context, it is expected that the substances, together, could be capable of controlling bacterial infection and dissemination, besides potentiating macrophages’ immune response against the studied microorganism. Moreover, reducing the CHX concentration by the addition of synoeca-MP peptide may, in a beneficial way, minimize the undesirable effects of both, CHX and synoeca-MP in a clinical setting. 相似文献
