Objectives were to evaluate progesterone concentrations after cows had initiated estrous cycles following calving and induction of estrous cycles in postpartum anovular high-producing Holstein dairy cows treated with controlled internal drug releasing (CIDR). In experiment 1 (EXP1), 62 cows that had initiated estrous cycles received a new CIDR (NCIDR) containing 1.38 g of progesterone or a 7-d used autoclaved CIDR (UCIDR) 48h after luteolysis for 7 d. Ovaries were examined by ultrasonography, and plasma analyzed for concentrations of progesterone. In experiment 2 (EXP2), 515 cows diagnosed as anestrus were randomly assigned to untreated control, NCIDR or UCIDR for 6d. Plasma was analyzed for concentration of progesterone 12 d after CIDR removal to determine ovulation. In EXP1, milk yield and body condition did not influence progesterone concentrations. Concentration of progesterone tended to increase faster (P=0.10) in cows receiving UCIDR than NCIDR, but both treatments reached a plateau at 90min. Cows receiving the NCIDR had greater (P=0.04) concentrations of progesterone during the 7-d treatment, but they were mostly subluteal (<1.0 ng/mL) after d 2. After removal, concentrations of progesterone were greater for NCIDR than UCIDR for the first 45 min, and were similar thereafter. Multiparous cows had lesser (P=0.004) concentrations than primiparous cows throughout the study. The pattern of ovarian follicular development was not affected by treatment. In EXP2, induction of onset of estrous cycles increased (P<0.01) with progesterone treatments, but was similar between NCIDR and UCIDR. Proportion of cows experiencing shorter than typical length estrous cycles after first AI tended to be greater (P=0.09) for control cows than those receiving the CIDR, and for cows remaining anestrous than those in which onset of estrous cycles was induced. Pregnancy per AI and pregnancy loss were similar among treatments. Cows that resumed estrous cyclicity prior to first AI had greater (P=0.01) pregnancy per AI. Treatment of high-producing Holstein cows that had previously initiated onset of estrous cycles with CIDR resulted in subluteal concentrations of progesterone, but in anestrous high-producing cows increased induction of estrous cycles with no effect on fertility at first insemination. 相似文献

In a previous study we showed that estrus synchronization with 2 treatments of PGF2 alpha 13 d apart reduced conception rate at the synchronized estrus and that this reduction occurred mainly in cows in the early luteal phase at the second PGF2 alpha treatment. The objective of the present study was to determine the efficacy of a synchronization regimen in which PGF2 alpha was administered during the mid- to late-luteal phase to cows that had previously been synchronized with progesterone. Spring-calving cows from 6 dairy herds were used in this study. On Day -32 (Day 1 = the start of the breeding season), cows that had calved 2 or more weeks ago were randomly assigned to a synchronization (S, n = 732) or control (C, n = 731) group. Cows in Group S were treated with an intravaginal progesterone device (CIDR) for 12 d from Day -32 to Day -20, while those in Group C were left untreated. Similar percentages of cows in Group S (80.6%) and C (82.9%) had cycled by Day -7. The CIDR treatment synchronized the onset of estrus, resulting in 92.9% of cows in estrus being detected within 7 d after CIDR removal. Cows in Group S that had cycled by Day -7 were treated with PGF2 alpha (25 mg, i.m., Lutalyse) on Day -2. Cows in both groups that were anestrous on Day -7 were treated with a combination of progesterone and estradiol benzoate (EB) to induce estrus and ovulation (CIDR and a 10 mg EB capsule on Day -7, CIDR removal on Day -2, and injection of 1 mg EB 48 h after CIDR removal). The PGF2 alpha treatment synchronized the onset of estrus in 87.5% of the cows. Group S and C cows had similar conception rates to first (61.0 vs 58.3%) and second (58.4 vs 60.9%) AI; similar pregnancy rates over the AI period (82.8 vs 79.2%) and over the whole breeding season (91.9 vs 90.6%); and required a similar number of services per pregnancy to AI (1.7 vs 1.8). The interval from the start of the breeding season to conception for cows conceiving to AI or to combined AI and natural mating was shorter (P < 0.001) by 5.7 and 6.2 d, respectively, for the Group S cows. It is concluded that the treatment regimen tested in the present study achieved satisfactory estrus synchronization, had no detrimental effect on fertility at the synchronized estrus, and shortened the interval from start of the breeding season to conception. 相似文献

Two experiments were conducted to examine circulating concentrations of progesterone (P4) in cows with ovarian follicular cysts (OFCs) and to relate differing levels of P4 to subsequent follicular events. In experiment 1, peripheral concentrations of P4 were determined in cows diagnosed with OFCs. Nonpregnant, lactating Holstein and Jersey cows (n = 32) were diagnosed as having OFCs by rectal palpation. Ovarian follicular cysts were then examined by transrectal ultrasonography to confirm the presence of OFCs (follicle diameter, >/=17 mm; absence of luteal tissue). At confirmation, a blood sample was collected for quantification of P4. The concentration of P4 at confirmation was classified as low (<0.1 ng/ml), intermediate (0.1-1.0 ng/ml), or high (1.0-2.0 ng/ml). More OFCs were associated with intermediate (66%) than with either low (28%) or high (6%) concentrations of P4. In experiment 2, the fate of follicles (diameter, >/=10 mm) that formed in the presence of an OFC was determined and related to circulating concentrations of P4 during follicular development. Follicles (n = 59) that formed in the presence of an OFC ovulated (n = 19), formed a cyst (n = 30), or underwent normal growth and regression (NGR; n = 10). Endogenous P4 in the 7-day period during follicular development was classified as low (if P4 dropped to <0.1 ng/ml for 1 day or longer), intermediate (if P4 averaged between 0.1 and 1.0 ng/ml and never dropped to <0.1 ng/ml), or high (if P4 averaged >1.0 ng/ml and never dropped to <0.1 ng/ml). In the presence of intermediate P4, 75% of observed follicles formed cysts, compared with 10% that ovulated and 15% that experienced NGR. In the presence of low P4, 53%, 41%, and 6% of follicles ovulated, formed a follicular cyst, or experienced NGR, respectively. This study was conducted to determine relationships between the uterine microflora and reproductive characteristics of dairy cows. Uterine lumina were swabbed during estrus immediately prior to artificial insemination (A I). Swabbings were cultured for bacteria and accessed for cytologic evidence of inflammation. The time of sampling averaged 104 d post partum. Bacteria in the uterus were cultured from 65% of the cows (n = 85). The number of cows with any given microbial genus ranged from 1 to 35 and the number of different genera per cow ranged from 0 to 8. There were on significant correlations between amount and type of uterine microflora, days post partum, numbers of observed estrus, conception rate and uterine inflammation. The number of observed estrus periods was not correlated with the presence of aerobic bacteria. No significant relationships were found between microbes and uterine inflammation or conception to the A.I. service at the time of sampling. A negative correlation between uterine inflammation and conception approached significance (r=-0.24, P > 0.10). Acquisition of uterine lumen samples by the technique utilized had no effect on conception to A I service. 相似文献

The effects of work and diet supplementation on progesterone secretion and on incidence of short luteal phases and ovulations without estrus was investigated in 40 postpartum F(1) crossbred dairy cows. These cows were allocated to 1 of 4 treatment groups: Group SPNW, supplement-nonworking; Group SPW, supplement-working; Group NSNW, nonsupplement-nonworking; and Group NSW, nonsupplement-working. After calving, working cows pulled sledges with a load of 300 to 450 Newtons(N); 4 hours per day 4 days per week, for a total of 100 days over a 1-year period. All cows were fed natural grass hay ad libitum while the supplemented cows were fed 3 kg of concentrate per head per day. The proportion of cows which showed behavioral estrus by 1 year post partum was 100, 100, 60 and 20% for Group SPNW, SPW, NSNW and NSW cows, respectively. Based on plasma progesterone concentrations, ovulation started 62 days earlier than onset of behavioral estrus. A total of 73 ovulations occurred by 1 year post partum. Forty-nine (67.1%) and 26 (32.9%) ovulations occurred in the supplemented and nonsupplemented cows while 33 (45.2%) and 40 (54.8%) ovulations occurred in the working and nonworking cows, respectively. Of the total ovulations, 26 (35.6%) were not associated with behavioral signs of estrus and occurred in 13 (32.5%) cows. The incidence of ovulation without estrus was higher (P<0.05) in working (42.4%) than in nonworking (30%) cows and in nonsupplemented (41.7%) than in supplemented (32.7%) cows. Short luteal phases occurred in 32.5% of the cows before the establishment of normal estrous cycles. In working cows, diet supplementation off-set the negative effect of work on the onset of estrus and conception. However, a relatively higher number of cows in Group SPW had ovulations without estrus before a normal estrous cycle was established. The incidence of short luteal phases or ovulations without estrus did not influence the pregnancy rate in subsequent normal estrus periods. In conclusion, in the supplemented cows, work did not influence the proportion of cows showing estrus and conceiving, but it significantly delayed the postpartum anestrus interval. In the nonsupplemented cows, reproductive activity was impaired in both working and nonworking cows, but was pronounced in working cows. However, once pregnancy was established there was no effect of work on the maintenance of pregnancy. Our study shows that with appropriate feeding regimens lactating crossbred cows could be used for draught purposes without any detrimental effects on fertility, but calving intervals would be extended. Finally, the physiological mechanisms involved in anestrus and ovulations without estrus and the significance of such phenomena in affecting postpartum reproductive performance and fertility in working cows require further investigation. 相似文献

Factors affecting walking activity at estrus during postpartum period and subsequent fertility in dairy cows 总被引:1,自引:0,他引:1
The postpartum period is critical for efficient reproduction, and certain features of the normal estrus cycle during this period have been associated with subsequent reproductive performance. The objectives of the present investigation were to evaluate the effects of environmental, production, and reproductive factors on walking activity at estrus as determined using pedometers during the first 50 days in milk (DIM) (study 1). In a second study, we tried to establish whether pedometer measurements recorded during this period could be used to predict subsequent fertility by considering the number of cows becoming pregnant before 90 DIM. We analyzed data derived from 995 parturitions in a single herd. Detection of estrus was performed using a pedometer system. Variables were screened for associations with walking activity by analysis of variance (ANOVA) through generalized linear model procedures (PROC GLM). Increased milk production and parity, and a mean relative humidity (RH) higher than 95% were associated with lower pedometer measurements. A higher number of animals simultaneously in estrus rendered higher pedometer readings. No significant effects of the year, season, DIM, number of previous estruses, and climatic data other than high mean RH were observed. Relationships between pedometer measurements and other variables recorded during the first 50 days postpartum, and subsequent fertility were assessed by applying logistic regression models. We detected no significant effects of year, milk production, season, and mean activity increase at estrus on high fertility. The likelihood of pregnancy before 90 DIM decreased for each additional lactation and for cows in anestrus between days 0 and 50 postpartum. The efficiency of a synthetic prostaglandin analog (ICI 80996) for the control of the estrus cycle in dairy cows has been tested with and without simultaneous use of progestagen implants (SC 21009, Searle). Synchronization of estrus is closer after combined progestagen-prostaglandin treatments than after prostaglandins alone (78.7 % in estrus in 48h vs 60.2 % ; P<0.001).The percentage of cows not observed in estrus during the first 96h after the end of treatment was also lower after combined treatments (20.7 % vs 31.8 % ; P<0.05). The best estrus synchronization was obtained by injecting prostaglandin analog two days before implant removal (86 % in estrus in 48h and 14.0 % not observed in estrus within 96h).Fertility was studied in a field trial after one or two inseminations at a predetermined time for each treatment. A higher pregnancy rate was obtained with two inseminations after combined progestagen-prostaglandin treatment (49.1 %). This pregnancy rate differed significantly (P<0.05) from those obtained with one A.I. after the same treatment or two A.I. after the prostaglandin treatment but did not differ from the pregnancy rate obtained with one A.I. 80h after the second prostaglandin injection. The most important problem in dairy cows is the fall in pregnancy rate between 21 days and 6 months (19.7 to 35.1 % depending on treatment ; P<0.05). 相似文献

The present study examined ovarian changes preceding the resumption of the ovarian cycle in postpartum dairy cows with different parities under similar body nutritional conditions. In postpartum primi- (n=6), bi- (n=4), and multiparous (n=6) Holstein dairy cows, ovarian ultrasonographic observations starting at 7 days after calving were performed every other day and then daily after the confirmation of clinical signs of oestrus for the detection of postpartum first ovulation. Blood samples were collected at the same time as ultrasonography and analyzed for oestradiol and progesterone to monitor ovarian activity. To evaluate the nutritional condition of the cows, body weight and body condition score (BCS, 1=emaciated to 5=obese) were measured weekly and blood samples for the analysis of glucose, insulin, and non-esterified fatty acid (NEFA) were collected at the same time until postpartum second ovulation. Dominant follicles (>8mm in diameter) of the first follicular wave were detected at 7 days after calving in all cows. The first wave follicle ovulated in five of six multiparous cows, whereas no first wave follicle ovulated in any of the primiparous cows. The days to first ovulation after calving in primiparous cows (31.8+/-8.3 days) were significantly greater (p<0.05) than those in multiparous cows (17.3+/-6.3 days), but were not significantly different from biparous cows (28.8+/-8.6 days). There was a significant relationship between parity and days to first ovulation after calving (p<0.05). BCS was maintained at a level of more than 2.5 during the postpartum period in all cows and there was no influence of parity on postpartum changes in BCS, glucose, insulin, or NEFA throughout the experiment. The present study demonstrated a negative relationship between parity and number of days from calving to first ovulation in dairy cows under similar body nutritional conditions. It is possible that the influence of parity on the resumption of ovarian cycle is modulated by the factors different from the nutrition-related changes during the postpartum period in dairy cows. 相似文献

The relationships between the concentration of milk progesterone and early embryo survival on Days 4-8 inclusive and between the concentration of progesterone on different days from Days 0-8 inclusive following ovulation and insemination were examined in dairy cows. The relationships were examined following 77 randomly chosen artificial inseminations to cows in standing oestrus. There was a significant (P < 0.05) linear and quadratic relationship between the concentration of milk progesterone on each of Days 4-6 after ovulation and the probability of embryo survival. There was no association (P > 0.05) between milk progesterone concentration and probability of embryo survival on Days 7 and 8 after ovulation. There were no associations between milk progesterone concentration on Days 0-2 and the concentrations on Days 4-7, however, progesterone concentrations on Days 4 and 5 were highly predictive of the concentration on Days 6 and 7, respectively. Overall, the results indicate that suboptimal progesterone support during the early luteal phase is likely to deleteriously affect embryo viability and in addition, that it is possible to predict milk progesterone concentrations during the early luteal phase based on earlier stage concentrations and thus identify cows at risk of early embryo loss. 相似文献

Lactating Friesian dairy cows (2nd-4th parity) which calved in spring (N = 7) or autumn (N = 15) were used. Their ovaries were examined by ultrasound scanning and blood samples were obtained daily for progesterone and oestradiol concentrations from the 5th day after calving until the first post-partum ovulation occurred. Five autumn-calving cows selected at random were bled every 15 min over a 6-h period on 1 day each week for 4 weeks after calving to assess the patterns of LH secretion. Follicular development during the post-partum anoestrous period was characterized by the growth and regression of small (less than or equal to 4 mm) and medium-sized (5-9 mm) follicles, until a dominant follicle (greater than 10 mm) was detected. The first detected dominant follicle ovulated in 14 cows, became cystic in 4 cows (all in autumn), and failed to ovulate in 1 cow. It was not possible to detect a dominant follicle in 3 cows due to scanning difficulties. The post-partum interval to detection of the first dominant follicle (mean +/- s.d.) was shorter (P less than 0.05) in autumn (6.8 +/- 1.8 days) than in spring (20 +/- 10.1 days). However, there was no significant difference between the respective intervals to first ovulation (autumn 27.4 +/- 25.9 and spring 27.3 +/- 18.9 days). Autumn-calved cows which had cysts had longer (P less than 0.001) intervals to first ovulation (58.2 +/- 23.5 days) than did normal cows (12.0 +/- 2.5 days). All cows with cysts had twin ovulations at their first post-partum ovulation. A pulsatile pattern of LH secretion was detected in the first week post-partum and LH pulse frequency was 2-3 per 6-h period in Weeks 1 and 2 post partum and increased to 5-7 pulses per 6-h period in the presence of a dominant or cystic follicle. Concentrations of progesterone in plasma during post-partum anoestrus were usually low (less than 0.2 ng/ml); oestradiol concentrations were also low (less than 5 pg/ml), but higher values (5-110 pg/ml) were observed in cows that had a dominant or a cystic follicle. 相似文献

An experiment was performed to compare the effects of 3 short-term treatments with progesterone and estradiol benzoate (EB) on follicular growth, synchrony of estrus and pregnancy rate after fixed-time insemination in lactating postpartum beef cows. In Treatment 1 (n = 46), each cow received a progesterone-containing intravaginal insert for 7 d with injection of EB (2 mg, i.m.) at the time of device insertion. In Treatment 2 (n = 46), the insert was used for only 5 d with injection of EB (2 mg, i.m.) at the time of insertion. Cows in Treatment 3 (n = 47) received an insert for 5 d with no EB at the time of insertion. Each cow in the 3 groups received PGF2 alpha (25 mg, i.m.) at the time of insert removal, followed by EB (1 mg, i.m.) 30 h later. The cows were then inseminated 28 to 30 h after treatment with EB (58 to 60 h after insert removal). Treatment with 2 mg EB terminated the growth of the largest ovarian follicle (> 5 mm in diameter) at device insertion in 16/16 and 14/15 cows in Treatments 1 and 2, respectively. Estrus was detected within an 8-h target period (48 to 56 h after insert removal) in 93, 87 and 81% of cows in Treatments 1, 2 and 3, respectively (P > 0.05). Pregnancy rates at 39 d post insemination were 60, 50 and 51% for Treatments 1, 2 and 3, respectively (P > 0.05). The pregnancy rates did not differ between cows that were anovulatory or those that had ovulated before the initiation of treatments (54%), or among cows that were 28 to 40, 41 to 60 or > 60 days post partum at insemination (43, 59 and 54%, respectively). Treatment with progesterone inserts for 5 or 7 d, PGF2 alpha at the time of insert removal and 1 mg EB 30 h later induced the high degree of synchrony of estrus and ovulation necessary for fixed-time insemination. 相似文献

The objective of this study was to evaluate the reproductive performance of lactating cows in seasonal dairy herds after estrus synchronization with PGF2alpha (PG) with or without supplementation with progesterone (P4). In Trial 1, synchronized cows (S1; n = 521) were compared with untreated control cows (C; n = 518) in 5 herds. Estrus of cows in the S1 group was synchronized with 2 treatments of PG (Lutalyse) 13 d apart. The breeding season started 2 d after the second PG. Cows were first bred by AI for 7 wk and then herd sires were used. Compared with C cows, estrus synchronization in the treated cows reduced the conception rate to first AI (61.1 vs 70.5%; P < 0.01) and the intervals from start of the breeding season to conception for cows conceiving to AI (11.0 vs 14.6 d; P < 0.05) or to both AI and natural mating (16.5 vs 18.4 d; P < 0.05). There was no effect on conception rate to second AI (68.8%), on pregnancy rate by Day 24 (72.3%) or Day 49 (86.3%) of the breeding season, or on the percentage of cows not pregnant at end of the breeding season (5.0%). In Trial 2, effects of P4 supplementation before the second PG on reproductive performance were evaluated in 4 herds. Estrus of each cyclic cow was synchronized with PG as in Trial 1. Half of the cows in each herd were treated with an intravaginal P4 device (CIDR) for 5 d before the second PG (S2+P4, n = 608), whereas the remaining half received no CIDR treatment (S2, n = 593). Compared with S2 cows, P4 treatment increased the estrous response rate to the second PG (89.6 vs 82.9%; P < 0.01), the conception rate to first AI (65.1 vs 59.7%; P = 0.07), the pregnancy rate by Day 6 of the breeding season (59.3 vs 49.0%; P < 0.001), and reduced the intervals from start of the breeding season to conception for cows conceiving to AI (8.6 vs 10.4 d; P < 0.10) or to both AI and natural mating (12.7 vs 16.4 d; P < 0.01). Treatment with a used CIDR from Days 16 to 21 after start of breeding to re-synchronize returns to service had no effect on conception rate to first or second AI but may decrease the conception rate to second AI in cows previously treated with CIDR. In conclusion, estrus synchronization with the double PG system can reduce fertility, while P4 supplementation for 5 d before the second PG can improve estrous response and overall reproductive performance. Postpartum dairy cows (n=35) were used to determine the effects of feeding propylene glycol (PG) on metabolic variables related to ovarian function and on oocyte developmental competence. Starting on Day 7 postpartum, each animal received an oral dose (500 ml) of either PG or water once daily. Blood samples were collected on Days 5, 15, 25 and 35 pp to measure insulin, non-esterified fatty acids (NEFAs), beta-hydroxybutyrate (BHB) and glucose concentrations. Oocytes were recovered by ultrasound guided follicular aspiration starting on approximately Day 30 postpartum and submitted to in vitro fertilization. Ovarian follicular activity was examined daily by ultrasonography from Day 7 until ovulation or Days 35-40 postpartum. Animals receiving PG had elevated insulin concentrations over the subsequent 90 min following dosing (P<0.05) compared to control animals. Glucose concentrations followed a similar pattern. Irrespective of treatment, concentrations of NEFA declined significantly from Days 15 to 35 postpartum. Administration of PG resulted in a decrease in NEFA (P<0.001) and BHB (P<0.001) over the subsequent 90 min compared to control animals. Treatment with PG had no effect on follicular dynamics, mean days to emergence of the first cohort of follicles postpartum, or days to dominance and duration of dominance for any follicular wave recorded postpartum. There was also no difference in mean days to first ovulation or in size of the preovulatory follicle between treatments. Oocyte quality as measured by blastocyst development after IVF was not affected by treatment. These results suggest that administration of PG has the ability to positively alter the systemic concentrations of a number of metabolic variables which have been related to fertility. However, we did not observe an effect of PG treatment on follicular dynamics or the length of the postpartum interval. An effect on oocyte developmental competence remains to be proven. 相似文献

Ovaries were obtained at slaughter from 12 Holstein dairy cows at 15, 25 or 35 days after their 4th calving. Non-atretic and atretic antral follicles were separated into 6 size classes according to size and the numbers in each class were expressed as a percentage of the total for each ovary. Non-atretic follicles of diameter 0.16-0.28 mm decreased from 27.5% at Day 15 to 1.5% at Day 35 whereas those of 0.29-0.67 mm and 0.68-1.57 mm diameter increased from 37.4 to 47.2% and from 11.5 to 17.3% respectively (all P less than 0.05). The proportions of follicles measuring 1.58-3.68, 3.69-8.56 and greater than 8.56 mm remained almost constant. The atretic follicles of 0.29-3.68 mm varied significantly in number according to the post-partum interval and to whether they were in the ovary containing the CL of pregnancy. It is concluded that the CL of pregnancy and/or the conceptus have a carry-over effect on the rate of growth of the antral follicles even after parturition. 相似文献

The objective of this study was to examine the effect of a chronic stressor, lameness, on reproductive parameters. Seventy cows 30-80 days post-partum were scored for lameness and follicular phases synchronized with GnRH followed seven days later by prostaglandin (PG). Fifteen Lame animals did not respond to GnRH ovarian stimulation. Milk progesterone for 5 days prior to PG was lower in the remaining Lame cows than Healthy herdmates. Fewer Lame cows ovulated (26/37 versus 17/18; P = 0.04) and the interval from PG to ovulation was shorter in Lame cows. In Subset 1 (20 animals), the LH pulse frequency was similar in ovulating animals (Lame and Healthy) but lower in Lame non-ovulators. An LH surge always preceded ovulation but lameness did not affect the interval from PG to LH surge onset or LH surge concentrations. Before the LH surge, estradiol was lower in non-ovulating cows compared to those that ovulated and estradiol concentrations were positively correlated with LH pulse frequency. In Subset 2 (45 cows), Lame ovulating cows had a less intense estrus than Healthy cows, although Lame cows began estrus and stood-to-be-mounted earlier than Healthy cows. In conclusion, we have identified several parameters to explain poor fertility in some chronically stressed animals. From 30 to 80 days post-partum, there was a graded effect that ranged from 29% Lame cows with absence of ovarian activity, whereas another 21% Lame cows failed to express estrus or ovulate a low estrogenic follicle; in 50% cows, many reproductive parameters were unaffected by lameness. 相似文献

The purpose of the present study was to hasten the resumption of ovarian activity early postpartum in lactating dairy cows, using equine chorionic gonadotropin (eCG), to enhance follicular growth, followed by hCG, to induce ovulation. Primiparous Holstein dairy cows (n=21) were assigned equally into eCG, eCG-hCG and Control groups. Cows in the eCG and eCG-hCG groups received an i.m. injection of eCG (500 IU Folligon?) on Day 6 postpartum. Cows in the eCG-hCG group were also given an i.m. injection of hCG (500 IU Chorulon?), once dominant follicle reached the diameter of 13-16 mm following eCG injection. Cows in Control group did not receive any treatment. Daily blood sampling and ultrasound examination were conducted, starting at Day 6 postpartum until confirming the third ovulation. Follicles ≥10 mm in diameter were detected on Day 11.5±1.48, 10.1±0.52 and 11.1±1.36 after calving in Control, eCG and eCG-hCG groups, respectively (P>0.05). The first wave dominant follicle ovulated in 71.4% of cows treated with eCG and eCG-hCG. In contrast, none of the first wave dominant follicles ovulated in Control cows. By Day 20 postpartum, all cows in eCG group, 6/7 cows in eCG-hCG group and none of the cows in Control group ovulated (P<0.05). Short estrous cycles (≤16 days) were detected in 2/7, 1/7 and 6/7 cows in eCG, eCG-hCG and control groups, respectively (P<0.05). In conclusion, injection of eCG on Day 6 postpartum could assist the early resumption of ovarian activity by enhancing ovarian follicle growth and early ovulation in postpartum cows. The aim of this study was to determine if the duration of treatment with an intravaginal progesterone releasing insert (IVP(4)) after treatment with oestradiol benzoate (ODB) at the time of insertion and 24 h after removal would affect selected variables including: size of ovarian follicles at the time of removal of inserts, diameter of ovulatory follicles, plasma concentrations of progesterone following ovulation, and duration of the following oestrous cycle. Characteristics of oestrus at a synchronised and spontaneous oestrus were also monitored. Non-lactating Holstein cows were synchronised with an IVP(4) for 5 (n = 10), 7 (n = 10), 8 (n = 9) or 9 (n = 9) days together with injections of ODB at device insertion (2 mg) and 24 h after removal (1 mg). Ultrasonography showed no significant effect of treatment on the day of emergence of preovulatory follicles relative to the day of removal of inserts (overall mean = -4.22 +/- 0.58; P = 0.15) for cows that ovulated within 120 h insert removal (n = 36). Treatment with ODB and an IVP(4) for 5 days reduced the diameter of preovulatory follicles at the time of removal of inserts and for the following 2 days compared to cows treated for 7-9 days (mean difference 2.56 +/- 1.15 mm; P = 0.033) but did not reduce the diameter of the ovulatory follicle (P = 0.21). Day of emergence relative to removal of inserts was associated with the diameter of the ovulatory follicle (R2 = 0.69; P < 0.001). Concentrations of progesterone and the diameter of the corpus luteum following ovulation were not affected by treatment (P > 0.20), but were affected by the diameter of the ovulatory follicle (P < 0.01). Diameter of the ovulatory follicle did not affect interoestrous and interovulatory intervals (P > 0.40). We conclude that treatment with an IVP(4) for 5 compared to 7-9 days with ODB administered at device insertion, and 24 h after removal reduced the diameter of preovulatory follicles at the time of removal of the insert but did not reduce the diameter of the ovulatory follicle or concentrations of progesterone in plasma. Emergence of preovulatory follicles closer to the time of removal of inserts reduced the diameter of the ovulatory follicle when oestrus was induced with ODB. Ovulation of smaller follicles reduced concentrations of progesterone in plasma following ovulation but did not affect oestrous cycle duration. 相似文献

Acyloxyacyl hydrolase (AOAH) is an enzyme of bovine polymorphonuclear neutrophil leukocytes (PMN) that is capable of detoxifying endotoxin (25). The activity of AOAH in PMN isolated from the blood was investigated in dairy cows that expelled the fetal membranes normally (Group NFM) and in cows with retained fetal membranes (Group RFM) to obtain better insight into the role of the AOAH enzyme of neutrophils in endotoxin-related diseases, which occur frequently in dairy cows during the early postpartum period, especially in RFM cows. Twenty early postpartum dairy cows were used in the study: 13 NFM cows and 7 RFM cows. In the RFM cows, the percentage of PMN in blood (29+/-4%) was significantly (P<0.05) lower than in NFM cows (43+/-4%). The average AOAH activity in RFM cows (mean +/- SEM = 89+/-13 pmol fatty acid/10(7) PMN/h) was lower than in NFM cows (107+/-6 pmol fatty acid/10(7) PMN/h), but the difference in neutrophil AOAH activity between the 2 groups was not significant. There was also a higher percentage of immature neutrophils in isolated leukocyte suspensions from RFM cows (22+/-8%) than from NFM cows (15+/-4%), so that impairment of AOAH activity in early postpartum cows could be explained, in part, by immaturity of the neutrophils. These results suggest that the decreased AOAH activity of PMN could play a role in the pathogenesis of endotoxin-related diseases in dairy cows during the early postpartum period. 相似文献

The effect of ketone body status on occurrence of first ovulation during early lactation was assessed in 84 multiparous dairy cows under field conditions. Animals were equally distributed across 8 farms and were controlled by the same herd fertility monitoring program. Cows were visited twice antepartum and 6 times postpartum at weekly intervals between 5:30 and 8:30 AM. On these occasions, body condition scores and milk yields were measured, blood and milk samples were taken, cows were gynecologically examined, and parameters of reproduction were determined. The onset of first ovulation was specified by milk progesterone determination and rectal palpation. Cows starting postpartum ovarian cyclicity within or after 30 d were classified as early and late responders (ER and LR, respectively). Resumption of the estrous cycle within 30 d postpartum is considered optimal under practical conditions, and classification based on this threshold value resulted in groups of equal size and equal distribution of ER + LR cows within farms. Ketone bodies measured were beta-hydroxybutyrate in serum and acetoacetate and acetone in serum and milk. Blood serum and milk ketone body concentrations during the first 6 wk of lactation were higher in LR than in ER, whereas plasma glucose and nonesterified fatty acid and milk fat, protein and urea concentrations did not differ between groups. Maximal concentrations of ketone bodies from parturition to first ovulation were better predictors of the onset of the estrous cycle than mean or minimal concentrations over the same period. Milk acetone and serum beta-hydroxybutyrate concentrations provided the most reliable information with regard to resumption of ovarian activity of all ketone bodies. 相似文献

In 25, 3-to-13 year old, dairy cows (Braunvieh and Hoehenfleckyieh) FSH, LH and progesterone plasma profiles were determined by RIA. Blood was sampled at 6-hour intervals from parturition to 40–78 days postpartum, and the results correlated with the commencement of cyclic functions. For FSH, generally basic values were recorded, without characteristic features associating any values with the onset of cyclic ovarian activities or the occurrence of the first heat. LH profiles varied greatly between individuals with regard to the onset of elevations, regularity of patterns and peak values. The first preovulatory LH peak was recorded 17.3±9.8 days (range 4–46) postpartum. The first heat occurred on day 28.4±16 (range 6–55) postpartum, indicating that 13/23 cows ovulated without behavioral estrus, as reproductive cycles were re-established. Peak LH values increased with progressive cycles (1st peak 5.7±4.8 ng/ml; 2nd peak 11.8±8.7 ng/ml; 3rd peak 13.5±9.9 ng/ml plasma). Progesterone values also showed great variations in the profile of their first postpartum elevation. In 13/25 cows the first cycle was shortened (13.1±2.9 days), prolonged in 3 animals (34±4 days) and normal in 7 cows (20.4±1.9 days). Heat, preovulatory LH peak and progesterone profile were normal in all animals on subsequent cycles. Two animals did not start cycling. 相似文献
