Understanding the genetic basis of photosynthetic efficiency (PE) contributing to enhanced seed yield per plant (SYP) is vital for genomics‐assisted crop improvement of chickpea. The current study employed an integrated genomic strategy involving photosynthesis pathway gene‐based association mapping, genome‐wide association study, quantitative trait loci (QTL) mapping, and expression profiling. This identified 16 potential single nucleotide polymorphism loci linked to major QTLs underlying 16 candidate genes significantly associated with PE and SYP traits in chickpea. The allelic variants were tightly linked to positively interacting QTLs regulating both enhanced PE and SYP traits as exemplified by a chlorophyll A‐B binding protein‐coding gene. The leaf tissue‐specific pronounced up‐regulated expression of 16 associated genes in germplasm accessions and homozygous individuals of mapping population was evident. Such combinatorial genomic strategy coupled with gene haplotype‐specific association and in silico protein–protein interaction study delineated natural alleles and superior haplotypes from a chlorophyll A‐B binding (CAB) protein‐coding gene and its interacting gene, Timing of CAB Expression 1 (TOC1), which appear to be most promising candidates in modulating chickpea PE and SYP traits. These functionally pertinent molecular signatures identified have efficacy to drive marker‐assisted selection for developing PE‐enriched cultivars with high seed yield in chickpea.  相似文献

The plant cell wall is a complex structure consisting of a variety of polymers including cellulose, xyloglucan, xylan and polygalacturonan. Biochemical and genetic analysis has made it possible to clone genes encoding cellulose synthases (CesA). A comparison of the predicted protein sequences in the Arabidopsis genome indicates that 30 divergent genes with similarity to CesAs exist. It is possible that these cellulose synthase-like (Csl) proteins do not contribute to cellulose synthesis, but rather to the synthesis of other wall polymers. A major challenge is, therefore, to assign biological function to these genes. In an effort to address this issue we have systematically identified T-DNA or transposon insertions in 17 Arabidopsis Csls. Phenotypic characterization of "knock-out" mutants includes the determination of spectroscopic profile differences in mutant cell walls from wild-type plants by Fourier-transform IR microscopy. A more precise characterization includes cell wall fractionation followed by neutral sugar composition analysis by anionic exchange chromatography.  相似文献

Mesorhizobium strain RC3, isolated from chickpea nodules, tolerated chromium up to 500 μg/ml and reduced it by 90% at pH 7 after 120 h. It produced plant growth-promoting substances, both in the presence and absence of chromium. Strain RC3 produced 35 μg indole acetic acid/ml in Luria Bertani broth with 100 mg tryptophan/ml, which decreased with an increase in chromium concentration. Chromium application to soil at 136 mg/kg was toxic to chickpea plants but when RC3 at 136 mg/kg was also added, it increased the dry matter accumulation, number of nodules, seed yield and grain protein by 71, 86, 36 and 16%, respectively, compared to non-inoculated plants. Nitrogen in roots and shoots were increased by 46 and 40%, respectively, at 136 mg Cr/kg. The bio-inoculant decreased the uptake of chromium by 14, 34 and 29% in roots, shoots and grains, respectively.  相似文献

The pigment composition of leaves from a number of different plant species collected from field sites in the region of Sheffield, UK, have been compared using high-performance liquid chromatography. Expression of pigment content per unit leaf area was dominated by variation in the total leaf chlorophyll. Neither chlorophyll per unit area nor the chlorophyll a/b ratio were found to be correlated with the habitat from which the plants originated. When the amounts of different carotenoids were expressed relative to the total carotenoid pool, it was found that whilst neither total carotene (α- +β-carotene) nor neoxanthin correlated with ability to grow in shade, the leaf content of both lutein and the total xanthophyll cycle carotenoids (zeaxanthin, anther-axanthin and violaxanthin) did, with lutein content being high in shade species and xanthophyll cycle intermediates low. There was a strong negative correlation between the relative amounts of each of these groups of carotenoids. The ratio of lutein to xanthophyll cycle carotenoids was strongly correlated to an index of shade tolerance.  相似文献

Summary RFLP mapping of chromosome 5R in the F3 generation of a rye (Secale cereale L.) cross segregating for gibberellic acid (GA3)-insensitive dwarfness (Ct2/ct2) and spring growth habit (Sp1/sp1) identified RFLP loci close to each of these agronomically important genes. The level of RFLP in the segregating population was high, and thus allowed more than half of the RFLP loci to be mapped, despite partial homozygosity in the parental F2 plant. Eight further loci were mapped in an unrelated F2 rye population, and a further two were placed by inference from equivalent genetic maps of related wheat chromosomes, allowing a consensus map of rye chromosome 5R, consisting of 29 points and spanning 129 cM, to be constructed. The location of the ct2 dwarfing gene was shown to be separated from the segment of the primitive 4RL translocated to 5RL, and thus the gene is probably genetically unrelated to the major GA-insensitive Rht genes of wheat located on chromosome arms 4BS and 4DS. The map position of Sp1 is consistent both with those of wheat Vrn1 and Vrn3, present on chromosome arms 5AL and 5DL, respectively, and with barley Sh2 which is distally located on chromosome arm 7L (= 5HL).  相似文献

Work in model species has identified a considerable number of genes that regulate different aspects of organ growth. This has led to an increasingly detailed knowledge about how the basic cellular processes underlying organ growth are controlled, and which factors determine when proliferation gives way to expansion, with this transition emerging as a critical decision point during primordium growth. Progress has been made in elucidating the genetic basis of allometric growth and the role of tissue polarity in shaping organs. We are also beginning to understand how the mechanisms that determine organ identity influence local growth behaviour to generate organs with characteristic sizes and shapes. Lastly, growth needs to be coordinated at several levels, for example between different cell layers and different regions within one organ, and the genetic basis for such coordination is being elucidated. However, despite these impressive advances, a number of basic questions are still not fully answered, for example, whether and how a growing primordium keeps track of its size. Answering these questions will likely depend on including additional approaches that are gaining in power and popularity, such as combined live imaging and modelling.  相似文献

Summary Twenty-eight diallel trials over 8 years and two locations were analysed to estimate genetic variances for agronomic characters of chickpea (Cicer arietinum L.). The data were analysed according to Method 4 and Model I of Griffing (1956). Days to flowering, plant height, and seed size were found to be predominantly under additive inheritance and were highly predictable. Both additive and non-additive genetic components were important for seed yield, number of branches, pods per plant, and seeds per pod. Although both general combining ability (gca) and specific combining ability (sca) varied significantly with generation, components of gca mean squares were invariably much larger than gca x generation interaction components, indicating that either the F1 or the F2 generation can be used to estimate the gca components effectively. Combined diallel analysis of F2s over locations revealed the importance of combining ability x location interactions and emphasized the need for testing over more than one location for the precise estimation of combining ability. The implications of these findings and those reported earlier in the literature on the breeding strategies/methods for the genetic improvement of agronomic characters in chickpea are discussed.ICRISAT journal article no. 1199  相似文献

Abstract

Developing fast growing sorghum seedlings is an important breeding goal for temperate climates since low springtime temperatures are resulting in a prolonged juvenile development. The adaptation of sorghum to tropical and subtropical highlands gives hint for certain genetic variation. The goals of the present study were to detect marker-trait associations for leaf and dry matter growth rate and for chlorophyll fluorescence and content (SPAD) in relation to temperature. A diversity set comprising 194 genotypes was tested in eight controlled environments with temperatures ranging from 9.4 to 20.8 °C. Significant marker-trait associations (p < 0.05) were identified for each individual temperature regime and on the parameters of regression analyses describing the responses of growth or chlorophyll related traits to temperatures. The diversity set was fingerprinted with 171 diversity array technology (DArT) and 31 simple-sequence repeat (SSR) markers. SSRs were used to analyze the population structure while association studies were performed on DArT markers. Promising marker-trait associations for growth rates in relation to temperature were detected on chromosomes SBI-01, SBI-03, SBI-07, and SBI-10. Many promising loci were also significantly associated to the results obtained in individual low-temperature environments. Marker-trait associations for chlorophyll content and fluorescence did occasionally co-locate to those for growth during juvenile development but there was no evidence supporting our hypothesis that seedling growth at low temperatures is largely influenced by SPAD or fluorescence.  相似文献

Quantitative trait loci (QTLs) for chlorophyll content were studied using a doubled haploid (DH) population with 168 progeny lines, derived from a cross between two elite Chinese wheat cultivars Huapei 3 × Yumai 57. Chlorophyll content was evaluated at the maximum tillering stage (MS), the heading stage (HS), and the grain filling stage (GS), at three different environments in 2005 and 2006 cropping seasons. QTL analyses were performed using a mixed linear model approach. A total of 17 additive QTLs and nine pairs of epistatic QTLs were detected. Ten of 17 additive QTLs for chlorophyll content were persistently expressed at more than two growth stages, which suggest developmentally regulated loci controlling genetics for chlorophyll content in different growth stages in wheat. One novel major QTL for chlorophyll content was closely linked with the PCR marker Xwmc215 and was persistently expressed at three growth stages.  相似文献

Rhizomes are prostrate subterranean stems that provide primitive mechanisms of vegetative dispersal, survival, and regrowth of perennial grasses and other monocots. The extent of rhizome proliferation varies greatly among grasses, being absent in cereals and other annuals, strictly confined in caespitose perennials, or highly invasive in some perennial weeds. However, genetic studies of rhizome proliferation are limited and genes controlling rhizomatous growth habit have not been elucidated. Quantitative trait loci (QTLs) controlling rhizome spreading were compared in reciprocal backcross populations derived from hybrids of rhizomatous creeping wildrye (Leymus triticoides) and caespitose basin wildrye (L. cinereus), which are perennial relatives of wheat. Two recessive QTLs were unique to the creeping wildrye backcross, one dominant QTL was unique to the basin wildrye backcross, and one additive QTL was detectable in reciprocal backcrosses with high log odds (LOD = 31.6) in the basin wildrye background. The dominant QTL located on linkage group (LG)-2a was aligned to a dominant rhizome orthogene (Rhz3) of perennial rice (Oryza longistamina) and perennial sorghum (Sorghum propinquum). Nonparametric 99 % confidence bounds of the 31.6-LOD QTL were localized to a distal 3.8-centiMorgan region of LG-6a, which corresponds to a 0.7-Mb region of Brachypodium Chromosome 3 containing 106 genes. An Aux/IAA auxin signal factor gene was located at the 31.6-LOD peak, which could explain the gravitropic and aphototropic behavior of rhizomes. Findings elucidate genetic mechanisms controlling rhizome development and architectural growth habit differences among plant species. Results have possible applications to improve perennial forage and turf grasses, extend the vegetative life cycle of annual cereals, such as wheat, or control the invasiveness of highly rhizomatous weeds such as quackgrass (Elymus repens).  相似文献

Wilt and root rot are the major constraints in chickpea production and very difficult to manage through agrochemicals. Hence, for an ecofriendly and biological management, 240 strains of Bacillus and Bacillus derived genera were isolated from chickpea rhizosphere, further narrowed down to 14 strains on the basis of in vitro production of indole acetic acid, siderophore, phosphate solubilization, hydrolytic enzymes and were evaluated for antagonism against chickpea pathogens (Fusarium oxysporum f. sp. ciceri race 1, F. solani and Macrophomina phaseolina). The strains were identified on the basis of physiological characters and 16S RNA gene sequencing. The genotypic comparisons of strains were determined by BOX-polymerase chain reaction profiles and amplified rDNA restriction analysis. These isolates were evaluated in greenhouse assay in which B. subtilis (B-CM191, B-CV235, B-CL-122) proved to be effective in reducing wilt incidence and significant enhancement in growth (root and shoot length) and dry matter of chickpea plants. PCR amplification of bacillomycin (bmyB) and β-glucanase genes suggests that amplified genes from the Bacillus could have a role to further define the diversity, ecology, and biocontrol activities in the suppression of soil-borne pathogens.  相似文献

Shoot tips, cotyledonary nodes and hypocotyls of chickpea (Cicer arietinum L.) were grown on 3 media: plant induction medium (PIM), callus induction medium (CIM), and shoot induction medium (SIM). Maximum growth and differentiation was seen in PIM, whereas minimum was observed in CIM. Shoot tips which differentiated to multiple shoots evolved negligible amounts of ethylene. Maximum ethylene evolution was recorded by hypocotyls in PIM. Ethylene appears to have stimulatory effect on shoot bud differentiation in cotyledonary nodes. But in hypocotyls, increased ethylene inhibited growth and differentiation. Calli on media containing only auxin (PIM) evolved significantly more ethylene, whereas those on media with cytokinin (SIM) evolved more methane. Callus forming explants like cotyledonary nodes and hypocotyls evolve more ethylene than shoot tips. Random Amplified Polymorphic DNAs (RAPDs) were used to investigate quantitative trait loci (QTL) for traits related to height growth on 126 F2 seedlings of maritime pine (Pinuspinaster Ait). The haploid megagametophyte was used to determine the maternal genotype of each F2 individual. The seedlings were raised for 2 years in a greenhouse under accelerated growth conditions consisting of intense fertilization combined with continuous light treatments. Total height was measured at different developmental stages, and height growth components were measured after the second growth period. QTLs were identified for each trait. For total height, QTLs of different developmental stages were located on distinct linkage groups. However, rather than a complete temporal change in QTL expression, our results showed that maturation may induce a progressive shift of the genetic control of height growth. This may provide an explanation for a low juvenile-mature phenotypic correlation previously reported for height. Height growth components related to the initiation (controlled by the apical meristem) and elongation of shoot cycles (controlled by the subapical meristem) were mapped to different chromosomes, suggesting that the activity of these meristems is controlled by separate genetic mechanisms.  相似文献

Higher plants have evolved specific mechanisms for partitioning the cytoplasm of dividing cells. In the predominant mode of phragmoplast-assisted cytokinesis, a cell wall and flanking plasma membranes are made de novo from a transient membrane compartment, the cell plate, which in turn forms by vesicle fusion from the centre to the periphery of the dividing cell. Other modes of cytokinesis appear to occur in meiotic cells and developing gametophytes. Here we review recent progress in the analysis of plant cytokinesis, focusing on genetic studies in Arabidopsis which are beginning to identify structural and regulatory components of phragmoplast-assisted cytokinesis. Two classes of mutations have been described. In one class, the defects appear to be confined to cell plate formation, suggesting that the execution of cytokinesis is specifically affected. Mutations in the other class display more general defects in cell division. We also discuss possible roles of proteins that have been localised in cytokinetic cells but not characterised genetically. Finally, mutations affecting meiotic or gametophytic cell divisions suggest that mechanistically different modes of cytokinesis occur in higher plants.  相似文献

The inheritance of the spring growth habit was studied in 63 old local cultivars and landraces of common wheat from Eastern and Western Siberia and the Tyva Republic. Minimal polymorphism was observed for the dominant Vrn genes, controlling the spring growth habit in landraces of these regions. The control was digenic and involved the Vrn1 and Vrn2 dominant genes in the majority (95%) of cultivars and was monogenic in three cultivars. None of the cultivars had the Vrn3 dominant gene, characteristic of the neighboring regions of China and Central Asia. Among 137 old local cultivars and landraces of Siberia, only one (cultivar Sibirskaya (K-23347) from Irkutsk oblast, was comparable in the response to the natural short day (photoperiod) to Chinese cultivars. Comparison of the results and the data reported for commercial cultivars revealed that the genotype frequencies of the dominant Vrn genes in Siberian landraces and commercial cultivars of common wheat remained essentially unchanged at least for the past 100 years. At the same time, Siberian landraces significantly differed in Vrn dominant gene frequencies from cultivars of the adjacent regions. It was assumed that the control of the spring growth habit by the two Vrn dominant genes is optimal for the climatic conditions of Siberia.  相似文献

In field experiments in India and Niger runner and bunch groundnut cultivars were compared for their pod distribution pattern and its relevance to the calcium (Ca) supply for pod development. Bunch cultivars produced sixty to eighty percent of their pods within 5 cm of the tap root. Runner cultivars explored a radius of up to 30 cm for pod production and exploited the soil area in a more homogeneous manner than bunch types. The available soil volume per pod was 19 to 27 cm3 for bunch types and 43 to 46 cm3 for runner types, varying the potential for Ca competition between pods. Computation of the soil Ca content needed to satisfy pod Ca requirements showed that much higher concentrations were needed for the bunch cultivars than for the runners. No significant differences in Ca content of pods existed between bunch and runner cultivars. However, in the runner cultivars, the Ca content of the more widely dispersed pods in outer zones was greater than that of the more densely populated inner pod zones. Regression analysis of shelling percentage across a range of environments showed that the shelling percentage of runners declined less rapidly than did the shelling percent of bunch types, indicating that runners were more efficient in exploiting Ca at lower soil Ca availability than the bunch types.Approved as ICRISAT Journal Article No. 1372.  相似文献

Plant development is controlled by complex endogenous genetic programs and responses to environmental cues. Proteome analyses have recently been introduced to plant biology to identify proteins instrumental in these developmental processes. To date most plant proteome studies have been employed to generate reference maps of the most abundant soluble proteins of plant organs at a defined developmental stage. However, proteomics is now also utilized for genetic studies comparing the proteomes of different plant genotypes, for physiological studies analyzing the influences of exogenous signals on a particular plant organ, and developmental studies investigating proteome changes during development. Technical advances are now beginning to allow a proteomic dissection of individual cell types, thus greatly increasing the information revealed by proteome analyses.  相似文献

A biomechanical method to distinguish self-supporting and non self-supporting growth habits is applied to exceptionally preserved “twigs” ofPitus dayi Gordon. The analysis investigates whether these isolated stem segments are consistent with a self-supporting tree-like habit as suggested by the stumps, trunks and branches of the genusPitus Witham preserved more commonly in the fossil record. Because of difficulties in accurately identifying certain fossil tissues, three centrisymmetrical models were constructed to test a range of possible tissue combinations over five ontogenetic stages. The results suggest a self-supporting habit with trends in mechanical parameters during ontogeny similar to those of extant, self-supporting plants. Less explicitly constrained to the analysis of habit, the investigation also examines the structural significance of specific tissues during ontogeny as observed from contributions of individual tissues to cross-sectional area, axial second moment of area and flexural stiffness.Pitus dayl produced a physiologically “cheap” primary cauline cortex which was rapidly replaced by the development of a rhytidome. A mechanically significant, cauline hypoderm comprising thickwalled sclerenchymatous tissue is absent. This arrangement differs from other tested Palaeozoic pteridosperms interpreted as semi-self-supporting such asLyginopteris oldhamia andCalamopitys sp. in which the primary cortex is mechanically significant and secondary growth of the wood does not reach mechanically significant thresholds within the primary body.  相似文献
