The process of sex differentiation in fishes is regulated by genetic and environmental factors. The sex of Patagonian pejerrey (Odontesthes hatcheri) appears to be under strong genotypic control (GSD) because the sex ratios are balanced (1:1) between 17°C and 23°C. However, sex ratios become female-biased at <15°C and male-biased at 25°C, which shows that this species also possesses some degree of temperature-dependent sex determination (TSD). Identification of the genetic sex of an individual will help elucidate the molecular basis of sex differentiation in this species. In this study, we used amplified fragment length polymorphism (AFLP) analysis to develop a genetic linkage map for both sexes and a sex-linked DNA marker for Patagonian pejerrey. The AFLP analysis of 23 male and 23 female progeny via 64 primer combinations produced a total of 153 bands. The genetic linkage map consisted of 79 markers in 20 linkage groups and 48 markers in 15 linkage groups for males and females, respectively. One AFLP marker tightly linked to the sex-determining locus was identified: the marker, ACG/CAA-217, amplified to the male-specific DNA fragment. Sequence analysis of this region revealed a single nucleotide polymorphism (SNP) between males and females, which was converted into a SNP marker. This marker provides genetic confirmation that the sex of Patagonian pejerrey is determined genetically and would be useful for the analysis of the molecular basis of GSD and TSD in this species.  相似文献

The inheritance of anthracnose resistance of the common bean ( Phaseolus vulgaris L.) differential cultivar G 2333 to Colletotrichum lindemuthianum races 73 and 89 was studied in crosses with the susceptible cultivar Rudá. The segregation ratios of 15 : 1 in the F2 and 3 : 1 in the backcrosses to Rudá indicate that for each of the races tested there are two independent resistance loci in G 2333. A random amplified polymorphic DNA (RAPD) molecular marker (OPH181200C) linked in resistance to race 73 was identified in a BC3F2:3 population derived from crosses between Rudá and G 2333. A RAPD molecular marker OPAS13950C, previously identified as linked to gene Co-42 , was also amplified in this population. Co-segregation analyses showed that these two markers are located at 5.6 (OPH181200C) and 11.2 (OPAS13950C) cM of the Co-42 gene. These markers were not present in BC1F2:3 plants resistant to race 89 indicating that this population carries a different resistance gene. DNA amplification of BC1F2:3 plants with RAPD molecular marker OPAB450C, previously identified as linked to gene Co-5 , indicated that this gene is present in this population.  相似文献

Incidence of sex chromosome aneuploidy in men is as high as 1:500. The predominant conditions are an additional Y chromosome (47,XYY) or an additional X chromosome (47,XXY). Behavioral studies using animal models of these conditions are rare. To assess the role of sex chromosome aneuploidy on sexual behavior, we used mice with a spontaneous mutation on the Y chromosome in which the testis-determining gene Sry is deleted (referred to as Y) and insertion of a Sry transgene on an autosome. Dams were aneuploid (XXY) and the sires had an inserted Sry transgene (XY Sry ). Litters contained six male genotypes, XY, XYY, XX Sry , XXY Sry , XY Sry and XYY Sry . In order to eliminate possible differences in levels of testosterone, all of the subjects were castrated and received testosterone implants prior to tests for male sex behavior. Mice with an additional copy of the Y chromosome (XYY) had shorter latencies to intromit and achieve ejaculations than XY males. In a comparison of the four genotypes bearing the Sry transgene, males with two copies of the X chromosome (XX Sry and XXY Sry ) had longer latencies to mount and thrust than males with only one copy of the X chromosome (XY Sry and XYY Sry ) and decreased frequencies of mounts and intromissions as compared with XY Sry males. The results implicate novel roles for sex chromosome genes in sexual behaviors.  相似文献

We assessed the fertility (reproductive success, litter size, testis weight, spermatocyte-to-spermatid ratio) of F1s and backcrosses between different wild-derived outbred and inbred strains of two mouse subspecies, Mus musculus domesticus and M. m. musculus . A significant proportion of the F1 females between the outbred crosses did not reproduce, suggesting that female infertility was present. As the spermatocyte-to-spermatid ratio was correlated with testis weight, the latter was used to attribute a sterile vs. fertile phenotype to all males. Segregation proportions in the backcrosses of F1 females yielded 11 (inbred) to 17% (outbred) sterile males, suggesting the contribution of two to three major genetic factors to hybrid male sterility. Only one direction of cross between the inbred strains produced sterile F1 males, indicating that one factor was borne by the musculus X-chromosome. No such differences were observed between reciprocal crosses in the outbred strains. The involvement of the X chromosome in male sterility thus could not be assessed, but its contribution appears likely given the limited introgression of X-linked markers through the hybrid zone between the subspecies. I examined the intrinsic postzygotic incompatibilities between two pupfishes, Cyprinodon elegans and Cyprinodon variegatus. Laboratory hybridization experiments revealed evidence of strong postzygotic isolation. Male hybrids have very low fertility, and the survival of backcrosses into C. elegans was substantially reduced. In addition, several crosses produced female-biased sex ratios. Crosses involving C. elegans females and C. variegatus males produced only females, and in backcrosses involving hybrid females and C. elegans males, males made up approximately 25% of the offspring. All other crosses produced approximately 50% males. These sex ratios could be explained by genetic incompatibilities that occur, at least in part, on sex chromosomes. Thus, these results provide strong albeit indirect evidence that pupfish have XY chromosomal sex determination. The results of this study provide insight on the evolution of reproductive isolating mechanisms, particularly the role of Haldane's rule and the 'faster-male' theory in taxa lacking well-differentiated sex chromosomes.  相似文献

Age at length, growth and reproduction of 220 Mastacembelus mastacembelus specimens from the Atatürk Dam Lake were studied from July 2005 to July 2006. Total lengths and weights ranged from 7.0 to 85.0 cm and from 6 to 1100 g, respectively. Maximum age was 13 years. The regression model fitted to length and weight data was W  =   0.0228  L 2.43 for males and W  =   0.0029  L 2.95 for females. The von Bertalaffy growth equations for males and females were L t  = 99.2 [1−e−0.11 ( t −0.12)] and L t  = 69.2 [1−e−0.26 ( t −0.35)], respectively. Males dominated especially at an older age; the overall sex ratio was 1 : 0.63 (M:F). The breeding period was from May to July. Fecundity ranged from 2540 to 24 000 eggs per female.  相似文献

The dioecious character of Pistacia vera L (the pistachio tree) limits its breeding capacity. Thus, early stage selection of males can save time, labor, and land. This study aimed to develop sex-linked single nucleotide polymorphism (SNP) markers, together with expressed sequence tag-derived simple sequence repeats (EST-SSRs), to determine position of the sex locus in pistachio by constructing a linkage map of its sex chromosome for the first time. Nine novel sex-linked SNP markers were successfully identified by SNaPshot minisequencing analysis of 25 SNP loci from 17 restriction site-associated DNA (RAD) reads in 309 individuals. All nine markers were heterozygous in females and homozygous in males supporting a ZW/ZZ sex determination system in pistachio. A total of 105 segregating SSRs and sex-linked markers were used to identify the sex chromosome and the position of the sex locus through analysis of a Siirt × Ba?yolu F1 population with 122 progenies. Of these 105 markers, four common and four paternal SSRs were mapped onto the sex chromosome, along with the phenotypic sex locus and sex-linked markers. The resulting consensus map had a total length of 65.19 cM. The sex locus and sex-linked SNP markers were located in the center of the chromosome at a distance of 31.86 and 31.92 cM, respectively. This study presents valuable information about the sex chromosome and sex locus position as well as novel polymorphic EST-SSRs and nine sex-linked SNP markers in pistachio.  相似文献

The growth of two commercially important flatfish, turbot ( Psetta maxima ) (L.) and brill ( Scophthalmus rhombus ) (L.), was investigated in the Adriatic using whole otoliths (sagittae) and stained otolith sections. At variance with the pattern usually observed in temperate seas, the opaque zone was found to be laid down in autumn and winter, and the translucent zone in spring and summer. Growth rates differed according to sex, with the females attaining greater body lengths. The von Bertalanffy growth parameters were: L=66.2 cm, K=0.31 years–1, and t0=–0.14 years for turbot males, L=81.5 cm, K=0.21 years–1, and t0=–0.48 years for turbot females; L=40.2 cm, K=0.49 years–1, and t0=–1.03 years for brill males; L=50.1 cm, K=0.27 years–1, and t0=–1.75 years for brill females. Growth rates and maximum age recorded for turbot were comparable to those reported in the North Sea.  相似文献

An individual's sex depends upon its genes (genotypic sex determination or GSD) in birds and mammals, but reptiles are more complex: some species have GSD whereas in others, nest temperatures determine offspring sex (temperature-dependent sex determination). Previous studies suggested that montane scincid lizards (Bassiana duperreyi, Scincidae) possess both of these systems simultaneously: offspring sex is determined by heteromorphic sex chromosomes (XX-XY system) in most natural nests, but sex ratio shifts suggest that temperatures override chromosomal sex in cool nests to generate phenotypically male offspring even from XX eggs. We now provide direct evidence that incubation temperatures can sex-reverse genotypically female offspring, using a DNA sex marker. Application of exogenous hormone to eggs also can sex-reverse offspring (oestradiol application produces XY as well as XX females). In conjunction with recent work on a distantly related lizard taxon, our study challenges the notion of a fundamental dichotomy between genetic and thermally determined sex determination, and hence the validity of current classification schemes for sex-determining systems in reptiles.  相似文献

SUMMARY 1. Temporary ponds are inhabited by a variety of invertebrates, of which anostracans are an important group. We studied the lifetables of male and female anostracan Streptocephalus mackini at 3 algal concentrations (0.5 × 106, 1.0 × 106 and 1.5 × 106 cells mL−1).
2. Regardless of sex, S. mackini showed better survivorship at lower food levels. The longest average lifespan observed was 85 ± 2 days for males fed Chlorella at 0.5 × 106 cells mL−1.
3. Both net reproductive rate and generation time decreased with increasing food level. The highest net reproductive rate was about 120 cysts per female. The longest generation time of about 40 days, observed at 0.5 × 106 cells mL−1, was more than three times that at 1.5 × 106 cells mL−1.
4. The rate of population increase ( r ) was nearly the same (0.31 ± 0.06) at high (1.5 × 106 cells mL−1) and intermediate (1.0 × 106 cells mL−1) food levels. The r -value at low food level (0.5 × 106 cells mL−1 of Chlorella ) was 0.20 ± 0.01 per day.  相似文献

The survival, growth, gonadal development and sex ratios of gynogenetic diploid turbot Scophthalmus maximus between 9 and 36 months of age were investigated. Gynogenesis was induced by activation of freshly collected eggs with diluted (1 : 10 with Ringer) and UV‐irradiated (30000 erg mm−2) sperm, followed by a cold shock at −1 to 0° C for 25 min, starting at 6·5 min after fertilization. The exclusive maternal inheritance of the resulting gynogenetic diploids was confirmed in all individuals used in the experiment by microsatellite markers. Mean total length and mass throughout the experiment was higher in the controls than in the gynogenetics. The gonadal development in gynogenetic males proceeded normally, and in gynogenetic females it was delayed during the first sexual maturation but was normal during the second one. The sex ratio was 1 male (M) : 1 female (F) in the controls, as expected, but 1 M : 3 F in the gynogenetics. In a second, independent experiment, carried out with fish originating from different broodstock, however, the sex ratio was 1 M : 1 F in the controls and 0 M : 1 F ( i.e . 100% females) in the gynogenetics. Together, these results suggested that the main sex‐determining genetic component in turbot fits well to female homogamety, in accordance with data from other flatfishes. These results indicate that it will be possible to obtain near or all‐female turbot stocks through induced gynogenesis or by crosses involving hormonally sex‐reversed fish.  相似文献

This study examined the changes in sex ratios and sex reversal rates in pejerrey Odontesthes bonariensis that occur with the progression of the spawning season in a seminatural setting. Four groups of hatchery-produced pejerrey larvae were stocked in floating cages in La Salada de Monasterio lake (Pampas region), a natural habitat of this species, and reared from hatching beyond gonadal sex determination with minimum human interference. Cage 1 was stocked at the beginning of the spring spawning season and the other cages were stocked with monthly delays until cage 4 in early summer. The genotypic (amhy+, XY/YY; amhy−, XX) and phenotypic (testis, male; ovary, female) sex ratios and proportions of genotype/phenotype mismatched individuals were estimated and their relation to water temperature and daylength during the experiment was analysed by generalized linear modelling. Water temperature varied between 11 and 30.5°C, and daylength duration between 11 h 22 min and 14 h 35 min. Sex genotyping revealed nearly balanced sex ratios of XY/YY (46%–49.1%) and XX (50.9%–54%) fish in cages 2–4 whereas the genotypic sex ratio in cage 1 was clearly biased towards XY/YY fish (60.6%). Phenotypic males ranged from 42% to 54.4% in cages 1–3. Cage 4, in turn, had significantly more phenotypic males (66%). The percentage of XX males (phenotypic male/genotypic female) was 23.1% in cage 1, decreased to a minimum of 5.4% in cage 2 and gradually increased in cages 3 and 4 to a maximum of 40.7% in the latter. The percentages of XY/YY females (phenotypic female/genotypic male) were highest in cage 1 (30%) and decreased progressively in the other cages to a significantly lower value (4.3%) in cage 4. These results generally support the findings of laboratory studies on the effect of temperature on the sex determination of this species and also provide novel evidence of a XX genotype-specific masculinizing effect of short daylength.  相似文献

Abstract:  Diapause induction and photoperiodic response curves were determined for 33 strains of Tetranychus pueraricola derived from kudzu vine at three constant temperatures (15, 18 and 20°C) under a short-day condition (10 : 14 h; light : dark). Females of all but one of the strains entered diapause at all three temperatures with little variation in diapause percentages among the strains. The exception was the southernmost strain, which was found to be a non-diapause (ND) strain. The critical photoperiod gradually decreased towards the south at a rate of about 1 h for each 5 degrees of latitude. The diapause strains (D1 and D2) exhibited 100% diapause, whereas the ND strain exhibited 0% diapause. By crossing these strains, we determined that 'non-diapause' was a dominant character over 'diapause' and the character was controlled by simple Mendelian inheritance. To clarify why the female progeny from the crosses between the D1 and ND strains did not segregate into the diapause and non-diapause phenotypes in a 1:1 ratio in the B1 generation, round-robin crosses were carried out among the three strains. The results showed that the F1 generation was reproductively compatible and showed high egg hatchability with a female-biased sex ratio. In the B1 generation, the crosses between the D1 and ND strains and between the D1 and D2 strains exhibited extremely low egg hatchability and produced mostly female progeny, whereas offspring from the crosses between the D2 and ND strains showed more than 50% hatchability for B1 eggs and a female-biased sex ratio. Thus, the absence of segregation observed in the crosses between the D1 and ND strains appears to be due to the severe hybrid breakdown that occurred in the B1 generation.  相似文献

The reproductive ecology of river lamprey Lampetra fluviatilis was investigated during the spawning period 2003 in the River Derwent, north‐east England. Over this period 1199 ± 104 individuals day−1(mean ±  s . d .) were counted on one spawning site (area c . 450 m2), but mark‐recapture estimates suggested that >5000 river lamprey used this site over the same period and egg deposition was estimated as 168 000 eggs m−2. The operational sex ratio of river lamprey in spawning clusters changed between spawning phases, from domination by females during the nest‐building phase (male : female ratio, 1 : 3·46), to a preponderance of males during the spawning phase (male : female, 1 : 0·37), followed by a return to a majority of females after spawning (male : female, 1 : 3·74). Recapture data showed that >97% of recaptured, tagged males were recorded at two or more nests, whereas almost 50% of recaptured, tagged females were recorded at the same nest, suggesting a promiscuous mating system, with a tendency towards polygyny within the population. Within the lower 80 km of the River Derwent and its tributaries, evidence of river lamprey spawning was found at only six sites, and most spawning (>80% of the observed spawning population) was at one site.  相似文献

The salmon louse (Lepeophtheirus salmonis (Krøyer, 1837)) is a parasitic copepod that can, if untreated, cause considerable damage to Atlantic salmon (Salmo salar Linnaeus, 1758) and incurs significant costs to the Atlantic salmon mariculture industry. Salmon lice are gonochoristic and normally show sex ratios close to 1:1. While this observation suggests that sex determination in salmon lice is genetic, with only minor environmental influences, the mechanism of sex determination in the salmon louse is unknown. This paper describes the identification of a sex-linked Single Nucleotide Polymorphism (SNP) marker, providing the first evidence for a genetic mechanism of sex determination in the salmon louse. Restriction site-associated DNA sequencing (RAD-seq) was used to isolate SNP markers in a laboratory-maintained salmon louse strain. A total of 85 million raw Illumina 100 base paired-end reads produced 281,838 unique RAD-tags across 24 unrelated individuals. RAD marker Lsa101901 showed complete association with phenotypic sex for all individuals analysed, being heterozygous in females and homozygous in males. Using an allele-specific PCR assay for genotyping, this SNP association pattern was further confirmed for three unrelated salmon louse strains, displaying complete association with phenotypic sex in a total of 96 genotyped individuals. The marker Lsa101901 was located in the coding region of the prohibitin-2 gene, which showed a sex-dependent differential expression, with mRNA levels determined by RT-qPCR about 1.8-fold higher in adult female than adult male salmon lice. This study’s observations of a novel sex-linked SNP marker are consistent with sex determination in the salmon louse being genetic and following a female heterozygous system. Marker Lsa101901 provides a tool to determine the genetic sex of salmon lice, and could be useful in the development of control strategies.  相似文献

Significant correlations between leaf carbon isotope discrimination (Δ) and altitude and between gas exchange and altitude have been reported in previous studies, raising the question of whether the altitudinal variations in discrimination and gas exchange can be attributed to genetic differences among populations from different altitudes. Studies that focus on in situ analysis cannot distinguish the effects of genetic variation from environmental variation. This article describes an experiment in which seeds of Plantago depressa (C3 species) and Setaria viridis (C4 species) collected from a wide altitudinal range were grown in the same environment. Carbon isotopic ratios (δ13C) and gas exchange of the seedlings were measured. The progenies of P.   depressa and S.   viridis no longer display any significant Δ decreases with the altitude of origin as seen in situ. Furthermore, photosynthetic rate, stomatal conductance, the ratio of intercellular to ambient CO2 and intrinsic water use efficiency for P.   depressa and S.   viridis grown in the greenhouse are also not significantly related to the altitude of origin. The observations suggest that altitudinal variations in Δ and gas exchange are not because of genotypic differences, independent of photosynthetic type.  相似文献

Backcalculated lengths at the end of the first growth season in wild Atlantic salmon Salmo salar differed significantly between parr smolting at age 1, 2 and 3 years over a period of 11 years (i.e. 1983–1993). Mean body lengths of the respective age groups at the end of the first growth period were 11·1, 6·2 and 4·7 cm, respectively. The mean percentage distribution of fish smolting at age 1, 2 and 3 was 14, 78 and 7%, and the mean smolt age was 1·95 years. Mean lengths at smolting of age groups 1, 2 and 3 were 13·6, 15·8 and 17·5 cm, respectively. Females outnumbered males among the downstream migrating smolts with a mean sex ratio (females/ males) estimated at 1·61, with a significant female surplus in 7 of the 11 years sampled. Of the smolts sampled, 14% exhibited enlarged gonads indicative of parr maturation, and all were males (37% of the parr males sampled). Mean annual smolt density from 1975 to 1996 was 13·4 individuals 100 m−2 ranging between 0·3–31 smolts 100 m−2. Mean densities (100 m−2) of the smolts aged 1, 2 and 3 years were 1·5, 9·3 and 0·9 fish, respectively. Mean annual biomass for the 22-year period (1975–1996) was estimated at 437 g 100 m−2, with a range of variation from 136 to 683 g 100 m−2. Smolt age 2 made up 81% of the mean annual biomass (355 g 100 m−2) and smolt age 1 and 3, 8% (35 g 100 m−2) and 11% (47 g 100 m−2), respectively.  相似文献

It has been suggested that climate change at the Cretaceous-Palaeogene (K-Pg) boundary, initiated by a bolide impact or volcanic eruptions, caused species with temperature-dependent sex determination (TSD), including dinosaurs, to go extinct because of a skewed sex ratio towards all males. To test this hypothesis, the sex-determining mechanisms (SDMs) of Cretaceous tetrapods of the Hell Creek Formation (Montana, USA) were inferred using parsimony optimizations of SDMs on a tree, including Hell Creek species and their extant relatives. Although the SDMs of non-avian dinosaurs could not be inferred, we were able to determine the SDMs of 62 species; 46 had genotypic sex determination (GSD) and 16 had TSD. The TSD hypothesis for extinctions performed poorly, predicting between 32 and 34 per cent of survivals and extinctions. Most surprisingly, of the 16 species with TSD, 14 of them survived into the Early Palaeocene. In contrast, 61 per cent of species with GSD went extinct. Possible explanations include minimal climate change at the K-Pg, or if climate change did occur, TSD species that survived had egg-laying behaviour that prevented the skewing of sex ratios, or had a sex ratio skewed towards female rather than male preponderance. Application of molecular clocks may allow the SDMs of non-avian dinosaurs to be inferred, which would be an important test of the pattern discovered here.  相似文献

Southern elephant seals breed at Península Valdés (PV, Argentina) along 200 km of coastline. Annual pup counts at peak breeding season for the entire colony increased from 12,113 in 1995 to 14,350 in 2006. Two demographic subunits were identified in the North and South of PV with different trends in births numbers, sex ratios and harem sizes. Birth numbers increased in the South, but decreased sharply in the North. To explain the trends in the colony and subunits, a population model was proposed that integrates social structure (harem size and sex ratio) in a fertility function that quantifies the effects of the social structure on the number of births. We found that a better fit to census data results from our model compared to a linear one  (χ12= 4.027, P = 0.045)  . The model was then used to test alternative hypotheses about the role of recruitment and migration on the dynamic of the two subunits. Results indicated the relevance of considering social structure in population models of gregarious and polygynous species, and is an additional tool for comparative studies between populations of elephant seals where long term census are available.  相似文献

Datisca glomerata is an androdioecious plant species containing male and hermaphroditic individuals. Molecular markers and crossing data suggest that, in both D. glomerata and its dioecious sister species D. cannabina, sex is determined by a single nuclear locus, at which maleness is dominant. Supporting this conclusion, an amplified fragment length polymorphism (AFLP) is heterozygous in males and homozygous recessive in hermaphrodites in three populations of the androdioecious species. Additionally, hermaphrodite x male crosses produced 1:1 sex ratios, while hermaphrodite x hermaphrodite crosses produced almost entirely hermaphroditic offspring. No perfectly sex-linked marker was found in the dioecious species, but all markers associated with sex mapped to a single linkage group and were heterozygous in the male parent. There was no sex-ratio heterogeneity among crosses within D. cannabina collections, but males from one collection produced highly biased sex ratios (94% females), suggesting that there may be sex-linked meiotic drive or a cytoplasmic sex-ratio factor. Interspecific crosses produced only male and female offspring, but no hermaphrodites, suggesting that hermaphroditism is recessive to femaleness. This comparative approach suggests that the hermaphrodite form arose in a dioecious population from a recessive mutation that allowed females to produce pollen.  相似文献
