We investigated the direct utilization of fine particulate detritus (dried and homogenized plant material in the size range of bacteria) as a food source for protozoans using axenic cultures of the cryptomonad, heterotrophic flagellate, Chilomonas paramecium, and the hymenostome ciliate, Tetrahymena pyriformis. When fed media containing only particulate detritus, these species revealed growth rates similar to those reported for field populations. The growth rates of Chilomonas fed exclusively particulate detritus were similar to those obtained on a bacterial diet. Considering the high percentage of detritus particles in the size range of bacteria in lakes, our results imply that direct utilization of detritus by protozoans may form an additional pathway of carbon in aquatic food webs that has generally been overlooked.  相似文献

Filtration was studied in two Arctic clams, Hiatella arctica and Mya sp., collected in Young Sound, Northeast Greenland. Clearance rates were determined as a function of ambient temperature and algal cell concentration, using the clearance method and feeding with a unicellular flagellate. For both species, clearance rates increased with increasing temperature from <у up to 4-8°C. At higher temperatures, filtration ceased and the clams closed their valves. Clearance rates were also determined in temperate specimens of H. arctica collected on the west coast of Sweden. For these specimens, clearance rates increased with increasing temperature from 0 to 18-20°C. When weight-specific clearance rates were compared between the two populations and between species, there were no differences at 1°C. Clearance rates in Arctic H. arctica were maximal at algal cell concentrations corresponding to 2.5-8 µg chlorophyll a l-1. Temperature compensation in Arctic bivalves is discussed and it is concluded that adaptations to constant low temperatures consist of a lower minimum temperature, for active filtration. Low clearance rates due to low temperatures did not seem to limit growth, under the prevailing conditions in Young Sound.  相似文献

The biology, morphology, and ultrastructure of the freshwater rapacious flagellate Colponema aff. loxodes, which attacks bodonids and chrysomonads, are studied. The flagellate is characterized by three-membrane alveolar pellicle, vesicular nucleus, two heterodynamous flagella, two microtubule bands which armor the longitudinal groove, and mitochondria with tubular cristae. Toxicysts (thread-organelles) are found in the cytoplasm. The posterior flagellum is characterized by the proximal fold. Micropores are completely absent. After being caught, the prey is taken into the longitudinal groove. Vegetative swimming cells are present in the life cycle. No reproduction or latent cysts are found. The taxonomical position of Colponema aff. loxodes is discussed in comparison with other colponemids and protists.  相似文献

Ant-gardens represent a special type of association between ants and epiphytes. Frequently, two ant species can share the same nest in a phenomenon known as ‘parabiosis’, but the exact nature (i.e., mutualistic or parasitic) of this interaction is the subject of debate. We thus attempted to clarify the mutual costs and benefits for each partner (ants and plants) in the Crematogaster levior/Camponotus femoratus ant-garden parabiosis. The ants’ response to experimental foliar damage to the epiphytes and to the host tree as well as their behavior and interactions during prey capture were investigated to see if the purported parasitic status of Cr. levior could be demonstrated in either the ant-ant or in the ant-plant interactions. The results show that both species take part in protecting the epiphytes, refuting the role of Cr. levior as a parasite of the ant-garden mutualism. During capture of large prey Ca. femoratus took advantage from the ability of Cr. levior to discover prey; by following Cr. levior trails Ca. femoratus workers discover the prey in turn and usurp them during agonistic interactions. Nevertheless, the trade-off between the costs and benefits of this association seems then to be favorable to both species because it is known that Cr. levior benefits from Ca. femoratus building the common carton nests and furnishing protection from vertebrates. Consequently, parabiosis can then be defined as the only mutualistic association existing between ant species, at least in ant-gardens. With the discovery of targeted gene replacement, moss biology has been rapidly advancing over the last 10 years. This study demonstrates the usefulness of moss as a model organism for plant photosynthesis research. The two mosses examined in this study, Physcomitrella patens and Ceratodon purpureus, are easily cultured through vegetative propagation. Growth tests were conducted to determine carbon sources suitable for maintaining heterotrophic growth while photosynthesis was blocked. Photosynthetic parameters examined in these plants indicated that the photosynthetic activity of Ceratodon and Physcomitrella is more similar to vascular plants than cyanobacteria or green algae. Ceratodon plants grown heterotrophically appeared etiolated in that the plants were taller and plastids did not differentiate thylakoid membranes. After returning to the light, the plants developed green, photosynthetically active chloroplasts. Furthermore, UV-induced mutagenesis was used to show that photosynthesis-deficient mutant Ceratodon plants could be obtained. After screening approximately 1000 plants, we obtained a number of mutants, which could be arranged into the following categories: high fluorescence, low fluorescence, fast and slow fluorescence quenching, and fast and slow greening. Our results indicate that in vivo biophysical analysis of photosynthetic activity in the mosses can be carried out which makes both mosses useful for photosynthesis studies, and Ceratodon best sustains perturbations in photosynthetic activity.  相似文献

A genetic transformation system has been developed for callus cells of Crataegus aronia using Agrobacterium tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with 5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this is the first time to report an Agrobacterium-mediated transformation system in Crataegus aronia.  相似文献

This study examined the efficacy of the predatory mite Typhlodromus athiasae Porath and Swirski (Acari: Phytoseiidae) as a biological control agent of the citrus red mite Panonychus citri (McGregor) (Acari: Tetranychidae) on seedlings of Washington and Valencia citrus cultivars at 1:10, 1:20 and 1:40 predator:prey release ratios under greenhouse conditions. At predator:prey ratios of 1:10, significant reductions on P. citri populations were observed one week after release of T. athiasae, and populations remained at low levels thereafter. The highest mean numbers of P. citri were found in the third week on the Washington cultivar and in the fourth week on Valencia, in a control group with no predators. This study demonstrates the potential of T. athiasae to effectively control P. citri on Washington and Valencia cultivars under greenhouse conditions at predator:prey ratios of 1:10. However, T. athiasae was unable to control the citrus red mite populations when the predator:prey ratio was reduced to 1:40. We therefore recommend a release ratio of 1:10 for effective control of P. citri in greenhouses on seedlings of Washington and Valencia citrus.  相似文献

The energy flow in aquatic food webs and their structures are largely determined by food utilisation of predators. Mysid shrimps are important predators in various aquatic ecosystems. We studied the stomach contents of three common littoral mysids from the Baltic Sea. The aim was to study whether the diets differ between species and size classes inhabiting shallow coastal areas. The effects of season (spring, summer, autumn) and habitat were also explored. Results showed that all species were highly omnivorous, utilising various phyto- and zooplankton prey, algal filaments and dead organic material through the growing season. No ontogenetic diet shifts were observed although different size classes preferred slightly different prey. The amount of detritus increased in the diets during growth. In addition, large mysids ate more macro- and less microzooplankton compared with the small ones. There were also species-specific differences in the food utilisation. Neomysis integer ate more benthic material, Praunus flexuosus more macrozooplankton and P. inermis more phytoplankton compared with the others. These differences reflect microhabitat differences and probably also size differences of the studied species. Seasonal variation was also observed in the diets. Food utilisation followed the changes in the food availability, e.g. phytoplankton spring bloom and zooplankton peak abundances in late summer. Results confirm the omnivorous nature of mysids showing the importance of a diversity of prey as energy sources during growth.  相似文献

Variability in the feeding ecology of young fishes over short and long time scales in estuaries is likely to affect population dynamics. We studied 14 years of early stage Striped Bass feeding ecology in the Hudson River Estuary over a 25-year time span, including years in which invasive zebra mussels markedly altered energy flow within the estuary. We predicted that feeding success would be low and that diet composition would be altered during years of high zebra mussel impact, particularly in upriver locations where mussels occur. Feeding success in the short term was indicated by volume of gut contents and in the long term by dry mass at length, i.e. condition; these measures were positively intercorrelated and varied significantly year to year. We tested for associations between condition and multiple biotic and abiotic environmental variables. There was a strong negative effect of zebra mussel grazing rate on condition in upriver locations and a weak positive effect in downriver locations. In upriver locations, condition was 33% higher when local salinity was high and zebra mussel grazing rates were low, whereas in downriver locations, condition was 35% higher when zebra mussel grazing rates and copepod abundance were high and local dissolved oxygen was low. Copepods, amphipods, mysids, and Leptodora constituted the highest prey-specific index of relative importance throughout the estuary. There was no evident effect of the zebra mussel invasion on diet composition. This long-term study corroborates the inferences of earlier studies that zebra mussels reduced early-stage striped bass growth rate.  相似文献

Many butterfly species are declining in range and abundance, sometimes to the point of becoming vulnerable to extinction. Several traits increase a species’ vulnerability to population decline through stochastic processes, including high larval specificity and poor dispersal rate. The Duke of Burgundy Hamearis lucina relies on Primula as its sole larval host plant. This monophagus dependency, coupled with susceptibility to environmental stochasticity, low dispersal and poor recolonization potential, means it is vital that sites supporting this rapidly-declining species are managed optimally. Here, we use two calcareous grassland sites in the UK with different grazing systems to identify optimal grazing management for Primula abundance and, for the first time, Primula characteristics linked previously to Duke oviposting preference and success: size, condition, succulence and surrounding sward height. We find that autumn and winter grazing intensity are both positively associated with Primula abundance, but there is a trade-off for winter grazing with negative effects on plant size, condition and succulence. Winter grazing also decreased the sward height below the optimum. Plants were bigger and better at the site managed using continuous (free-roaming) grazing versus the site managed using rotational (paddock-based) grazing. We recommend moderately high grazing intensity during autumn using a free-roaming system where possible to attain abundant Primula, with a reduction in grazing intensity or grazing removal during winter to ensure suitability of individual plants. This management would also benefit other host plants, such as bird’s foot trefoil Lotus corniculatus, which are vital for other declining butterflies that frequently co-occur with Dukes.  相似文献

Prey stage preference of female Kampimodromus aberrans (Oudemans) (Phytoseiidae) at constant densities of different stages of Tetranychus urticae Koch (Tetranychidae), functional response types and parameters of the predator females to the varying densities of eggs, larvae, protonymphs and deutonymps of T. urticae were determined in order to establish its potential for the mite biological control. Experiments were conducted at 25 ± 1°C, 65 ± 10% RH and 16:8 (L:D) photoperiod. Our results indicated that the predator consumed significantly more prey larvae than other prey stages. Functional response type of predator was determined by a logistic regression model. The predator exhibited a Type II response on all prey stages. The attack rate (α) and handling time (T h ) coefficients of a Type II response were estimated by fitting a “random-predator” equation to the data. The lowest estimated value α and the highest value of T h (including digestion) were obtanined for the predator feeding on deutonmph. The lowest value of T h were obtained for the predator feeding on prey larvae, but the attack rate value obtained on larva wasn’t different than that obtained on egg and protonymph. According to our results, K. aberrans could be an efficient biological control agent of T. urticae at least at low prey densities. However, further field based studies are needed to draw firm conclusions.  相似文献

The predation rate of fifth instars and female adults of Orius laevigatus (Fieber) fed honeybee pollen, Ephestia kuehniella Zeller eggs or an egg yolk based artificial diet on second instars of the western flower thrips Frankliniella occidentalis (Pergande) was examined in the laboratory. Predation rate of both fifth instars and female adults was not influenced by their diet. Despite a lower body weight, O. laevigatus reared on artificial diet or pollen killed as many prey as their peers reared on E. kuehniella eggs, suggesting that body weight is not a reliable predictor of predation rate. The use of non-prey foods for O. laevigatus for mass production or as a supplementary food to sustain its populations in the field when prey are scarce is discussed.  相似文献

A mixed trophic state production process for algal lipids for use as feedstock for renewable biofuel production was developed and deployed at subpilot scale using a green microalga, Auxenochlorella (Chlorella) protothecoides. The process is composed of two separate stages: (1) the photoautotrophic stage, focused on biomass production in open ponds, and (2) the heterotrophic stage focused on lipid production and accumulation in aerobic bioreactors using fixed carbon substrates (e.g., sugar). The process achieved biomass and lipid productivities of 0.5 and 0.27 g/L/h that were, respectively, over 250 and 670 times higher than those obtained from the photoautotrophic cultivation stage. The biomass oil content (over 60 % w/DCW) following the two-stage process was predominantly monounsaturated fatty acids (~82 %) and largely free of contaminating pigments that is more suitable for biodiesel production than photosynthetically generated lipid. Similar process performances were obtained using cassava hydrolysate as an alternative feedstock to glucose.  相似文献

ABSTRACT. We investigated the feeding of the small heterotrophic dinoflagellates (HTDs) Oxyrrhis marina , Gyrodinium cf. guttula , Gyrodinium sp., Pfiesteria piscicida , and Protoperidinium bipes on marine heterotrophic bacteria. To investigate whether they are able to feed on bacteria, we observed the protoplasm of target heterotrophic dinoflagellate cells under an epifluorescence microscope and transmission electron microscope. In addition, we measured ingestion rates of the dominant heterotrophic dinoflagellate, Gyrodinium spp., on natural populations of marine bacteria (mostly heterotrophic bacteria) in Masan Bay, Korea in 2006–2007. Furthermore, we measured the ingestion rates of O. marina , G . cf. guttula , and P. piscicida on bacteria as a function of bacterial concentration under laboratory conditions. All HTDs tested were able to feed on a single bacterium. Oxyrrhis marina and Gyrodinium spp. intercepted and then ingested a single bacterial cell in feeding currents that were generated by the flagella of the predators. During the field experiments, the ingestion rates and grazing coefficients of Gyrodinium spp. on natural populations of bacteria were 14–61 bacteria/dinoflagellate/h and 0.003–0.972 day−1, respectively. With increasing prey concentration, the ingestion rates of O. marina , G . cf. guttula , and P. piscicida on bacteria increased rapidly at prey concentrations of ca 0.7–2.2 × 106 cells/ml, but increased only slowly or became saturated at higher prey concentrations. The maximum ingestion rate of O. marina on bacteria was much higher than those of G . cf. guttula and P. piscicida . Bacteria alone supported the growth of O. marina . The results of the present study suggest that some HTDs may sometimes have a considerable grazing impact on populations of marine bacteria, and that bacteria may be important prey.  相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes.  相似文献

Several predatory mites have been found in association with the coconut mite, Aceria guerreronis Keifer, in northeast Brazil. However, the latter still causes damage to coconut in that region. The objectives of this work were to compare the frequencies of occurrence of Neoseiulus (Phytoseiidae) and Proctolaelaps (Melicharidae) species on standing and aborted coconuts in coastal Pernambuco State, northeast Brazil and to analyze their possible limitations as control agents of the coconut mite, based on evaluations of the restrictions they may have to access the microhabitat inhabited by the pest and their functional and reproductive responses to increasing densities of the latter. Neoseiulus baraki (Athias-Henriot) was found mostly on standing coconuts whereas Proctolaelaps bickleyi (Bram) was found mostly on aborted coconuts. Measurements of the entrance to the microhabitat occupied by the coconut mite, between the bracts and the subjacent fruit surface, showed that this different pattern of predator prevalence could be related to predator sizes, although other environmental factors could not be disregarded. Progressively higher predation rate of N. baraki was observed up to an experimental density that corresponded to 1,200 coconut mites per fruit, which is close to the average number determined in northeast Brazil, reducing slightly afterwards. Predation rate of P. bickleyi reduced consistently but slightly with increasing prey densities, but in absolute values, rates were always much higher than determined for N. baraki. The excessively high killing capacity of P. bickleyi, probably related to its high feeding requirement, may be detrimental in terms of stability. In fact, such high requirement for food suggests that P. bickleyi might not have a strong relation with the coconut mite and that the latter may not be its main food source under natural conditions. It is concluded that body sizes of both predators and the exceedingly high feeding requirement of P. bickleyi may limit their performance as control agents of the coconut mite.  相似文献

A bacterial strain, KM1S, was isolated from a Malaysian rainforest soil sample by using a defined enrichment medium that specifically facilitates selection of quorum quenching bacteria. KM1S was clustered closely to Bacillus cereus by 16S ribosomal DNA sequence analysis. It degraded N-3-oxo-hexanoyl homoserine lactone and N-3-oxo-octanoyl homoserine lactone in vitro rapidly at 4.98 and 6.56 μg AHL h−1 per 109 CFU/ml, respectively, as determined by the Rapid Resolution Liquid Chromatography. The aiiA homologue, encoding an autoinducer inactivation enzyme catalyzing the degradation of N-acylhomoserine lactones, of KM1S was amplified and cloned. Sequence analysis indicated the presence of the motif 106HXDH-59 amino acids-H169-21 amino acids-D191 for N-acylhomoserine lactone lactonases.  相似文献

Auxin receptors TIR1/AFBs play an essential role in a series of signaling network cascades. These F-box proteins have also been identified to participate in different stress responses via the auxin signaling pathway in Arabidopsis. Cucumber (Cucumis sativus L.) is one of the most important crops worldwide, which is also a model plant for research. In the study herein, two cucumber homologous auxin receptor F-box genes CsTIR and CsAFB were cloned and studied for the first time. The deduced amino acid sequences showed a 78% identity between CsTIR and AtTIR1 and 76% between CsAFB and AtAFB2. All these proteins share similar characteristics of an F-box domain near the N-terminus, and several Leucine-rich repeat regions in the middle. Arabidopsis plants ectopically overexpressing CsTIR or CsAFB were obtained and verified. Shorter primary roots and more lateral roots were found in these transgenic lines with auxin signaling amplified. Results showed that expression of CsTIR/AFB genes in Arabidopsis could lead to higher seeds germination rates and plant survival rates than wild-type under salt stress. The enhanced salt tolerance in transgenic plants is probably caused by maintaining root growth and controlling water loss in seedlings, and by stabilizing life-sustaining substances as well as accumulating endogenous osmoregulation substances. We proposed that CsTIR/AFB-involved auxin signal regulation might trigger auxin mediated stress adaptation response and enhance the plant salt stress resistance by osmoregulation.  相似文献
