哺乳动物胚胎在体外培养中普遍存在早期发育阻滞的现象.对此,人们用形态学、生物化学、分子生物学、显微操作等手段开展了磷酸、葡萄糖、次黄嘌呤和细胞质因素对早期胚胎发育阻滞的影响的研究.本文综合分析了共培养系统的优缺点.说明了采用完全成分已知的培养液对进行有关研究的必要性.列出了有效运用于克服小鼠、大鼠、仓鼠、兔、猪、羊、牛、猴等动物早期胚胎阻滞的成分已知的培养液的名称。 Abstract:The early embryo developmental block is a common phenomenon in mammal when embryos are cultured in vitro.Many studies of phosphorus,glucose,hypoxanthine and cytoplasmic factors on early embryo developmental block carried out by different methods such as morphology,biochemistry,molecular biology and micromanipulation have been reviewed.The merit and shortcoming were analyzed and the necessity of using simple or components limited media overcoming early embryo developmental block were also reviewed.Media that have been shown effective in overcoming early embryo developmental block in mouse,rat,hamster,rabbit,pig,sheep,cattle and monkey were listed.  相似文献

本文对有关哺乳类(包括人)着床前早期胚胎发育阻滞的形成与突破的研究近况作了概述。早期胚胎发育阻滞一般都发生在最初四个细胞周期中的某个长G_2相阶段,该时期恰好是贮存在胚胎细胞中源自母系的mRNA失活或降解、胚胎细胞自身基因组应该被激活的临界期,胚胎在这时对环境压力非常敏感。过氧化氢类物质在胚胎细胞内高含量累积也是导致发育阻滞的因素之一。早期胚胎的正常发育必须依赖于源自生殖道上皮细胞的信号,这类外源信号被初步认为蛋白质性质。提供某些生长因子,或清除微环境中有害物质等都将有助于 胚胎的正常发育。早期胚胎间的协同作用在体外胚胎正常发育中的正效应已引起人们的重视。  相似文献

克服昆明小鼠体外受精卵发育阻滞方法的研究   总被引:2,自引:0,他引:2  
本研究应用CZB和WM培养液进行昆明小鼠体外受精胚胎的发育培养,建立了一个可行的胚胎体外培养的新方法,并通过改变培养液的成分及其含量,对胚胎发育的阻滞机理和突破方法进行了初步的探索。培养于WM中的受精卵发生阻滞,有48%停留于2细胞阶段;而CZB中的胚胎有81%发育为桑椹胚和囊胚。在WM中添加EDTA和谷氨酰胺得到了66%的囊胚;加大WM中乳酸钠和丙酮酸钠的比值未能克服发育的阻滞现象。实验结果表明,EDTA和谷氨酰胺在克服阻滞时具有协同作用。  相似文献

通过在CZB培养液中添加不同浓度葡萄糖及在胚胎发育的不同阶段加入葡萄糖,对小鼠胚胎进行体外培养,以探讨葡萄糖在小鼠早期胚胎体外发育中的作用。其结果表明,小鼠胚胎在含糖CZB与在无糖CZB中培养比较,4-细胞发育率无差异;各浓度葡萄糖组囊胚率显著高于无糖组,其中3.0mmol/L浓度组囊胚细胞数显著高于其余组;实验二:2-细胞至4-细胞、4-细胞至桑椹胚前添加葡萄糖囊胚率显著提高。上述结果证明,在小鼠胚胎体外培养中加入葡萄糖不会导致2-细胞阻滞;葡萄糖浓度增加至10mmol/L对小鼠胚胎无毒性作用,其最适浓度为3.0mmol/L;2-细胞至4-细胞、4-细胞至桑椹胚前添加葡萄糖是必要的。关键词 葡萄糖;小鼠;2-细胞阻滞;胚胎;体外发育  相似文献

兔输卵管上皮细胞解除大鼠早期胚胎发育阻滞的研究   总被引:1,自引:0,他引:1  
本研究利用兔输卵管上皮细胞(ROEc), 与异种动物大鼠的受精卵共培养,结果大量出现突破2细胞发育阻滞的现象。体外受精卵和体内受精卵的2细胞发育阻滞突破率分别为62%和73%;利用ROEC 条件培养液培养大鼠的体外受精卵,2细胞发育阻滞的突破率达68%,且能顺利发育至桑椹胚和囊胚。将ROEC直接培养在含~(35)S-甲硫氨酸的CZB 培养液中,经SDS-聚丙烯酰胺凝胶电泳及放射自显影,发现在该条件培液中出现了分子量分别为135Kd,68Kd,55Kd,51Kd 和44Kd 由ROEC 分泌的多肽,其中68 Kd 区带最显著,而135 Kd区带非常弱。用(125)~I-酰化剂标记示踪发现,与ROEC 共培养24 h 的大鼠胚胎透明带上含有68Kd 蛋白,并发现55 Kd 蛋白与透明带结合的痕迹。这些蛋白很可能就是胚胎发育调控由母型向合子型过渡并突破早期发育阻滞的关键因子,在功能上似无种属特异性。  相似文献

人IVF(in vitro fertilization)培养的胚胎常易发生发育阻滞,这极大地降低了IVF的治疗效率。近年来发现,组蛋白精氨酸甲基化酶3(PRMT3)在早期胚胎发育过程中起着重要的作用,但是其对阻滞胚胎发育的作用及机制仍不清楚。该研究利用IVF废弃的胚胎为实验材料。采用Q-PCR和Confocal检测早期不同发育时期胚胎中PRMT3的核酸和蛋白表达情况,以及H4R3me2a甲基化水平的变化情况。另外,在阻滞胚胎中过表达PRMT3蛋白,观察阻滞胚胎的进一步发育。研究结果表明,与正常发育胚胎相比,阻滞胚胎中PRMT3核酸和蛋白的表达均呈现显著下降(P0.05);同时,阻滞胚胎中H4R3me2a的甲基化水平也明显降低(P0.05)。过表达PRMT3蛋白能够挽救部分阻滞胚胎的发育,甚至个别阻滞胚胎还能够发育到囊胚阶段。总之,早期发育胚胎中PRMT3表达的降低或缺失可能是导致胚胎发育阻滞产生的主要原因之一,PRMT3是早期胚胎发育过程中必需且非常重要的关键因子。  相似文献

在以往研究工作的基础上证明了通过添加2.5mmol/L的牛磺酸和0.1mmol/L的EDTA至M16培养液中,可支持昆明白小鼠的体外受精(IVF),并支持受精卵突破2-细胞阻滞发育至囊胚期。本研究进一步证明了牛磺酸和EDTA在昆明小鼠早期胚胎发育和克服2-细胞阻滞中起关键作用。 Abstract:The results of our early experiments show that the 2-cell block can be overcome by culturing zygote in modified M16 ,modified CZB and TE medium.Our research shows that the taurine and EDTA play key role in overcoming 2-cell block in Kunming mouse.The results show that the addition of taurine and EDTA to M16 medium can support the IVF and develoment to blastocyst in vitro in Kunming strain mice.This is the first report that M16 medium added with taurine and EDTA can be used in both IVF and culture medium to overcome the 2-cell block of embryo development in Kunming strain mouse.  相似文献

In order to investigate whether the kinetics and stage of embryo development affect cryosurvival of in vitro produced bovine embryos, cleaved embryos were categorized in six groups based on their developmental kinetics regarding the stage of embryonic block in bovine (8–16 cell stage): I and II – early (day 2) and late (day 3) 5–8 cell, III and IV – early (day 3) and late (day 4) 8–16 cell, and V and VI – early (day 4) and late (day 5) morula. The cryosurvival and developmental competence of these embryos were compared with each other and also with the corresponding control groups. The potential of 5–8 cell stage embryos to survive vitrification and further develop towards blastocyst stage was significantly lower than vitrified and un-vitrified 8–16 cell and morula stage embryos. These results suggest that, the survival rate and potential of embryos to develop towards blastocyst stage might be affected by the kinetic of the embryo development. Moreover, the results of this study indicated that the optimal stages of early embryo vitrification are post-embryonic block.  相似文献

The abnormalities of early post‐implantation embryos can lead to early pregnancy loss and many other syndromes. However, it is hard to study embryos after implantation due to the limited accessibility. The success of embryo culture in vitro can avoid the challenges of embryonic development in vivo and provide a powerful research platform for research in developmental biology. The biophysical and chemical cues of the microenvironments impart significant spatiotemporal effects on embryonic development. Here, we summarize the main strategies which enable researchers to grow embryos outside of the body while overcoming the implantation barrier, highlight the roles of engineered microenvironments in regulating early embryonic development, and finally discuss the future challenges and new insights of early embryo culture.  相似文献

Bovine in vitro embryo production is an inefficient process; while maturation and fertilization proceed apparently normally, the proportion of embryos reaching the transferable (blastocyst) stage is rarely over 40% and those that do reach this stage are often compromised in quality and competence. There is considerable evidence of a significant influence of follicular origin on oocyte developmental potential and it appears that once the oocyte is removed from the follicle its developmental capacity is capped. Evidence suggests that while culture conditions during bovine in vitro embryo production can impact somewhat the developmental potential of the early embryo, the intrinsic quality of the oocyte is the key factor determining the proportion of oocytes developing to the blastocyst stage. This paper highlights some of the problems associated with in vitro production of embryos and discusses some of the ways of overcoming these problems.  相似文献

In recent years, progress has been achieved in the production of pig embryos through IVM and IVF techniques. Cytoplasmic maturation of oocytes has been improved by modifications to IVM procedures. However, the historical problem of polyspermic penetration still remains a major issue to be solved. Recent studies indicate that the type of IVF medium and certain modifications to that medium can reduce polyspermy. Efforts should be directed to increase the developmental competence and quality of embryos. At present, many embryo culture (EC) media are available that can overcome the historical 4-cell block and support development of early in vivo derived embryos to the blastocyst stage. In contrast, blastocyst development of in vitro produced embryos in these culture media varies significantly. Furthermore, morphology and cell numbers in in vitro produced blastocysts are inferior to their in vivo counterparts. However, several modifications to EC techniques have improved embryo quality and developmental competence. Testing embryo viability through surgical transfer to recipient animals has resulted in acceptable pregnancy rates with moderate litter sizes. Although reliable in vitro systems are available for the generation of pig embryos, the problem of polyspermy and poor embryo development hamper their large-scale implementation. Further research efforts should be directed to improve oocyte/embryo quality and the methods to minimize polyspermy through development of novel IVM, IVF, and EC techniques.  相似文献

The process that leads to embryo formation appears to follow a defined pattern, whose sequential developmental steps—under strict genetic control—can be analysed through the study of mutants affecting embryogenesis. We present the analysis of four embryo-specific (emb) mutants of maize, characterised by abnormal development not overcoming the proembryo or early transition stage, that define three separate genes on the basis of their chromosomal location and complementation pattern. A common feature emerging from histological analysis is that suppression of morphogenesis is accompanied by an uncontrolled pattern of cell division. The block in embryo development is associated with abnormal suspensor proliferation, possibly due to the absence of a signal elaborated by the embryo proper and required for suspensor cell identity maintenance. Mutant endosperm morphogenesis is not impaired, as shown by the formation of the expected domains, i.e. aleurone, starchy endosperm, embryo-surrounding region and basal endosperm transfer layer. The program of cell death appears impaired in the mutants, as expected if this process is essential in determining the shape and morphology of the developing organs. An unexpected result is obtained when mutant embryo rescue is attempted. Immature embryos transferred to a basal medium germinated, yielding small but otherwise normal seedlings, an observation not consistent with the histological evidence of a complete absence of morphogenetic potential. The analysis of emb mutants appears a promising tool to elucidate crucial points of embryo development such as the coupling of cell division with morphogenesis, cell-to-cell interactions, the relationship between embryo and endosperm development, and the interaction between embryo proper and suspensor.  相似文献
