Cucumber ( Cucumis sativus L.) cultivars Marketmore, Lama, XPH 1187, XPH 1484 and Sprint 440 (N) were grown in a greenhouse under two levels of biologically effective ultraviolet-B ( UV -B) radiation (daily dose: 0 and 11.6 kJ m−2 UV-BBE) for 31 days. Significant intraspecific differences were observed in plant height, number of leaves, leaf area and total dry weight. Based upon total biomass accumulation, Marketmore was found to be the most tolerant, and XPH 1484 the most sensitive to UV-B radiation. The dose response of accumulation of UV absorbing compounds (measured as absorbance of methanolic extracts) in leaf tissues showed an increase in UV absorbing compounds with UV-B dose in Marketmore, Sprint 440 (N) and XPH 1187. In Lama and XPH 1484, however, doses below 8.7 kJ m−2 UV-BBE produced no change in UV absorbing compounds. This study suggests that intraspecific differences in UV-B radiation sensitivity in cucumber may be related to inherent differences in the accumulation of UV absorbing compounds in leaves.  相似文献

UV-B-sensitive (Poinsett) and -insensitive (Ashley) cultivars of cucumber ( Cucumis sativus L.) were grown in growth chambers at 600 μmol m−2s−1 of photosynthetically active radiation provided by metal halide (MH) or high pressure sodium/deluxe (HPS/DX) lamps. Plants were irradiated 15 days from seeding for 6 h per day under 18. 2 kJ m−2 day−1 of biologically effective UV-B (UV-BBE) radiation. One of the most pronounced effects of UV-B was a 27 to 78% increase in phenylalanine ammonialyase (PAL) activity. UV-B also increased total polyamines. Catalase and superoxide dismutase varied greatly in their response to UV-B. There were no interactive effects on PAL or catalase activity, or total polyamines. There was a UV × PAR source interaction for superoxide dismutase activity. UV-B increased chlorosis and decreased height, dry weight and leaf area. Stem elongation, biomass production, leaf enlargement and chlorosis were greater under HPS/DX lamps than under MH lamps. Chlorosis was greater in Poinsett than in Ashley and in lower leaves than in upper ones. Aside from chlorosis, there were no interactive effects of UV-B, PAR source or cultivar on any of the growth parameters measured, suggesting that the growth response of cucumber seedlings to UV-B is unaffected by PAR source or cultivar. Similarly, except for SOD activity, the biochemical response to UV-B was also not influenced by PAR source or cultivar.  相似文献

Using quartz optical fibres, penetration of both monochromatic (310 nm) and polychromatic UV-B (280–320 nm) radiation in leaves of Brassica napus L. (cv. Ceres) was measured. Plants were grown under either visible light (750 μmol m−2 s−1 photosynthetically active radiation) or with the addition of 8. 9 KJ m−2 day−1 biologically effective UV-B (UV-BBE) radiation. Results showed that of the 310 nm radiation that penetreated the leaf, 90% was within the intial one third of the leaf with high attenuation in the leaf epidermis, especially in UV-treated plants. Polychromatic UV-B radiation, relative to incident radiation, showed a relatively uniform spectral distribution within the leaf, except for collimated radiation. Over 30% of the UV-screening pigments in the leaf, including flavonoids, were found in the adaxial epidermal layer, making this layer less transparent to UV-B radiation than the abaxial epidermis, which contained less than 12% of the UV-screening pigments. UV-screening pigments increased by 20% in UV-treated leaves relative to control leaves. Densely arranged epicuticular wax on the adaxial leaf surface of UV-treated plants may have further decreased penetration of UV-B radiation by reflectance. An increased leaf thickness, and decreases in leaf area and leaf dry weight were also found for UV-treated plants.  相似文献

Experiments were conducted under greenhouse conditions to investigate the effects of enhanced UV-B radiation (280 to 320 nm) on height, fresh and dry weights, leaf chlorophyll and carotenoids, CO2 uptake rates, and Hill activity in soybean ( Glycine max L. cv. Bragg). Plants were exposed for 6 h continuously from midmorning to midafternoon each day to UV-B radiation which was provided by Westinghouse FS-40 sun lamps filtered with 0.127-mm cellulose acetate film (UV-B enhanced) or 0.127-mm Mylar S film (UV-B Mylar control). Three different UV-B enhanced radiation levels were tested: 1.09 (treatment T1), 1.36 (treatment T2), and 1.83 (treatment T3) UV-B sun equivalent units (UV-Bsec) where 1 UV-Bsec= 15.98 mW·m−2 of solar UV-B obtained by applying EXP -[(α-265)/21]2, a weighting function that simulates the DNA absorption spectrum, to the UV-B lamp systems. These UV-B levels correspond to a calculated decrease in stratospheric ozone content of 6%, 21%, and 36% for treatment T1, T2, and T3, respectively.
Daily exposure of soybean plants to UV-B radiation significantly decreased height, fresh and dry weights, leaf chlorophyll and carotenoid contents, and CO2 uptake rates. Leaf pigment extracted in 80% acetone from UV-B-treated soybean plants showed considerable increase in absorption in the wavelength region of 330 to 400 nm with increased UV-B radiation levels. Chloroplast preparations from leaves of T2 and T3 plants showed significant reductions in Hill reaction measurements.  相似文献

Soybeans Glycine max (L.) cv. Essex were hydroponically grown in a greenhouse at 2 levels of ultraviolet-B (UV-B) radiation (0 and 2 500 J m−1 day−1 biologically effective UV-B radiation) and 4 levels of P (6.5, 13, 26 and 52 μ M ). Plants were grown in each treatment combination to the complete expansion of the 4th trifoliolate leaf. UV-B radiation and reduced P supply generally decreased plant height, leaf area and total biomass, but increased specific leaf weight and flavonoid content (measured as absorbance of methanolic extracts). Although both UV-B radiation and low P supply produced deleterious effects on plant biomass, the effects were non-additive. The combination of UV-B and the lowest P level (6.5 μ M ) had no effect on total biomass or leaf area. This was at least partially due to the accumulation of flavonoids and leaf thickening. The results show that the sensitivity of soybean to UV-B radiation is dependent upon plant P supply. Plants experiencing P deficiency are less sensitive to UV-B than plants at optimum P levels.  相似文献

A growth analysis was made of ultraviolet-B (UV-B)-sensitive (Poinsett) and insensitive (Ashley) cultivars of Cucuumis satives L. grown in growth chambers at 600 μmol m−2 s−1 of photosynthetic photon flux (PPF) provided by red- and far-red-deficient metal halide (MH) or blue- and UV-A-deficient high pressure sodium/deluxe f HPS/DX) lamps. Plants were irradiated 6 h daiiy with 0.2 f-UV-B) or 18.2 C+UV-B) kJ m−2 day−1 of biologically effective UV-B for 8 or 15 days from time of seeding. In general, plants given supplemental UV-B for 15 days showed lower leaf area ratio (LARs, and higher specific leaf mass (SLM) mean relative growth rate (MRGR) and net assimilation rate (NAR) than that of control plants, but they showed no difference in leaf mass ratio (LMR), Plants grown under HPS/DX lamps vs MH lamps showed higher SLM and NAR. lower LAR and LMR. hut no difference in MRGR. LMR was the only growth parameter affected by cultivar: at 15 days, it was slightly greater in Poinsett than in Ashley. There were no interactive effects of UV-B. PPF source or cultivar on any of the growth parameters determined, indicating that the choice of either HPS/DX or MH lamps should not affect growth response to UV-B radiation. This was true even though leaves of UV-B-irradiated plants grown under HPS/DX lamps have been shown to have greater chlorosis than those grown under MH lamps.  相似文献

Pisum sativum cv. Guido grown under controlled environment conditions was exposed to either low or high UV-B radiation (2·2 or 9·9 kJ m–2 d–1 plant-weighted UV-B, respectively). Low or high UV-B was maintained throughout growth (LL and HH treatments, respectively) or plants were transferred between treatments when 22 d old (giving LH and HL treatments). High UV-B significantly reduced plant dry weight and significantly altered plant morphology. The growth and morphology of plants transferred from low to high UV-B were little affected, when compared with those of LL plants. By contrast, plants moved from high to low UV-B showed marked increases in growth when compared with HH plants. This contrast between HL and LH appeared to be related to the effect of UV-B on plant development. Exposure to high UV-B throughout development consistently reduced leaf areas. In fully expanded leaves there was no significant UV-B effect on cell area and reduced leaf area could be attributed to reduced cell number, suggesting effects on leaf primordia. Further reductions in the leaf area of younger leaves were the result of the slower development rate of plants grown at high UV-B, which also resulted in significant reductions in leaf number.  相似文献

Plants of three commercial cultivars of Hordeum vulgaris L. (Barberousse, Gerbel, Panda) were continuously exposed to two concentrations of SO2 (40±5 and 117±20 ppb) against a control (charcoal-filtered air). Experiments were performed in fumigation chambers. Fumigation was started 9 days after seeding and plant material was harvested after 48 days. In none of the three cultivars did visible symptoms of injury appear on the leaves after exposure to SO2, while they all showed a reduction in plant height and dry matter of shoots and roots and a significant increase in the shoot/root dry weight ratios depending on the cultivars. The time course of conductivity, and K+ Ca2+ and sugar effluxes were determined for 24 h. Leaves from cvs Gerbel and Panda exposed to SO2 exhibited a progressive increase in conductivity, and in Ca2+ and sugar efflux (expressed as percentages of their total effluxes). Plusmn; efflux of the three cultivars and conductivity in cv. Barberousse increased only after 24 h incubation. A relative leakage ratio (RLR) was calculated from the UV absorption at 280 nm after 24 h incubation of the leaf strips and the total 280 nm UV absorption, obtained after liquid N2 killing of the tissues. The RLR was well correlated with the total amino acids and sugars found in the leachate and can be used as a test for determining the tissue response to SO2.

This is a study of the impact of increased ultraviolet-B (UV-B) radiation on the secondary chemistry of Salix myrsinifolia (dark-leaved willow). For nearly two decades, the loss of stratospheric ozone above the high latitudes of the Northern Hemisphere has increased UV-B radiation (280–320 nm) over the long-term mean. Willows (Salicaceae) are widely distributed in these northern regions. To determine the effects of increased UV-B radiation on willows, the plantlets of three clones of S. myrsinifolia were grown under ambient (3.6 kJ m−2 day−1) or enhanced (7.18 kJ m−2 day−1) UV-B irradiance. After the 2-week indoor experiment, the concentrations of UV-B-screening phenolics (flavonoids and phenolic acids) and low-UV-B-screening phenolics (salicylates and condensed tannins) in fresh leaves were investigated and the biomass of leaves, stems and roots was determined. As expected, the total amount of flavonoids in willow leaves clearly increased when plantlets were exposed to higher UV-B irradiation. However, the degree of increase of individual compounds varied: luteolin-7-glucoside, monomethyl-monocoumaryl-luteolin-7-glucoside and one myricetin derivative increased significantly, while the apigenin-7-glucuronide increased only slightly. The enhanced UV-B also increased the amount of p -hydroxycinnamic acid derivative. The UV-B effects on other phenolic acids and tannins were minor. In contrast to the other phenolics, the amounts of two salicylates, salicin and saligenin, decreased under enhanced UV-B irradiation. Our results indicate that the concentrations of both UV-B-screening and low-UV-B-screening phenolic compounds in leaves of S. myrsinifolia may vary in response to elevated UV-B radiation. However, while the UV-B protective flavonoids and phenolic acids accumulate during UV-B exposure, the concentrations of certain salicylates decrease.  相似文献

Growth and development of hydroponically grown pea seedlings ( Pisum sativum L. cv. Alaska) were measured using stem and root length as well as number of leaves and lateral roots. The growth was dependent on the presence of cotyledons and was modulated by the irradiance. All plants were grown in a full nutrient solution. If grown at low irradiance (73 μmol m-2s-1) they depended more and for a longer time on the cotyledons than plants grown at high irradiance (220 μmol m-2s-1). Low irradiance caused stem elongation but decreased root length and number of lateral roots as compared to plants grown at high irradiance. The dark respiration of the leaves was measured as oxygen uptake. In plants grown at the low irradiance, excision of the cotyledons caused the rate of oxygen uptake to increase by a factor of three, and the increase was sensitive to cyanide. Decotyledonized plants showed a high respiration rate and a diminished leaf growth for their entire life cycle. CO2 fixation also increased in decotyledonized pea seedlings grown at either irradiance. The mobilization of food reserves from the seeds was positively correlated to seed dry weight, but only if the plants were grown at 73 μmol m-2s-1. Increasing dry weight of the seed enhanced top growth, whereas root growth was depressed, so that top and root responds differently with regard to that part of growth which depends on mobilization of reserves from the seed.  相似文献

The combined effects of ultraviolet-B (UV-B, 280–320 nm) radiation and water stress were investigated on the water relations of greenhouse grown soybean [ Glycine max (L.) Merr. cv. Essex]. On a weighted (Caldwell 1971), total daily dose basis, plants received either 0 or 3 000 effective J m2 UV-BBE supplied by filtered FS-40 sunlamps. The latter dose simulated the solar UV-B radiation anticipated at College Park, Maryland, U.S.A. (39°N latitude) in the event that the global stratospheric ozone column is reduced by 25%. Plants were either well-watered or preconditioned by drought stress cycles. Diurnal measurements of water potential and stomatal conductance were made on the youngest fully expanded leaf. Various internal water relations parameters were determined for detached leaves. Plants were monitored before, during and after water stress. There were no significant differences in leaf water potential or stomatal conductance between treatments before plants were preconditioned to water stress. However, drought stress resulted in significantly lower midday and afternoon leaf water potentials and lower leaf conductances as compared to well-watered plants. UV-B radiation had no additional effect on leaf water potential; however, UV did result in lower leaf conductances in plants preconditioned to water stress. Turgid weight:dry weight ratio, elastic modulus, bound water and relative water content were unaffected by UV-B radiation. Osmotic potentials at full and zero turgor were significantly lower in the drought stressed treatments as compared to well-watered plants.  相似文献

The chlorophyll content and partitioning of assimilate of bean ( Phaseolus vulgaris L. 'Pinto') plants were determined 6 days after treatment of the second internode (I2 with 5 μg of brassinosteroid (BR), a growth-promoting steroidal lactone. Plants were grown for 6 days under equal levels (90 μmol s-1 m-2) of photosynthetic photon flux density (PPFD) provided by cool white fluorescent (CWF) or incandescent (INC) lamps and equal levels of far-red (28 W m-2, 700–800 nm) radiation provided by the same INC or far-red (FR) fluorescent lamps. Brassinosteroid treatment had no appreciable effect on total biomass production but caused a decrease of 15–20% dry matter distribution in the upper portion of the shoot, a small (4%) but constant increase in dry matter in l2 and a large (11–16%) increase in dry matter in the lower portion of the shoot (especially I1). Treatment with BR increased assimilate accumulation in the primary leaves, especially under INC and FR lamps, and reduced dry matter in the trifoliate leaves. BR also caused a 16–21% reduction in total leaf area and even a greater reduction in area of the trifoliate leaves, but significantly increased specific leaf weight of the primary leaves and the first trifoliate leaf and the amount of dry matter in the lateral shoots under all radiation sources. In comparison to controls, BR treatment increased dry matter accumulation in the treated internode 3.3x under CWF and 1.6x under INC or FR. BR treatment also increased chlorophyll content in the primary leaves under all radiation sources and in the trifoliate leaves under CWF and INC lamps. These findings suggest a possible mobilization role of BR and establish the importance of adequate PPFD (and photosynthate) for maximum swelling and splitting response to brassinosteroid.  相似文献

The aim of this work was to study the effect of ultraviolet-B (UV-B) exposure on oxidative status in chloroplasts isolated from soybean ( Glycine max cv . Hood). Chloroplasts were isolated from soybean leaves excised from either control seedlings or those exposed to 30 and 60 kJ m−2 day−1 of UV-B radiation for 4 days. Chloroplastic oxidative conditions were assessed as carbon-centered radical, carbonyl groups and ascorbyl radical content. Treatment with UV-B increased the carbon-centered radical-dependent EPR signal significantly by 55 and 100% in chloroplasts from leaves exposed to 30 and 60 kJ m−2 day−1 UV-B, respectively, compared to radical content in chloroplasts from control leaves. The content of carbonyl groups increased by 37 and 62% in chloroplasts isolated from soybean leaves irradiated for 4 days with 30 and 60 kJ m−2 day−1 UV-B, respectively. The content of soluble metabolites in isolated chloroplasts should not be taken as absolute in vivo values; however, these data are valuable for comparative studies. UV-B exposure did not significantly affect ascorbyl radical content compared to controls. The content of ascorbic acid and thiols in chloroplasts isolated from leaves exposed to 60 kJ m−2 day−1 UV-B was increased by 117 and 20.8%, respectively, compared to controls. Neither the content of total carotene nor that of β -carotene or α -tocopherol was affected by the irradiation. The results presented here suggest that the increased content of lipid radicals and oxidized proteins in the chloroplasts isolated from leaves exposed to UV-B could be ascribed to both the lack of antioxidant response in the lipid soluble fraction and the modest increase in the soluble antioxidant content.  相似文献

The long-term response of citrus rootstock seedlings to CO2 enrichment was examined in Carrizo estrange ( Poncirua trifoliata (L.) Raf. x Citrus sinensis (L.) Osbeck] and Swingle citrumelo ( P. trifoliate x C. parodist Macf.]. Plaotlets 14 weeks old were transferred to outdoor controlled-environment chambers and maintained for 5 months from Feb. 14 to July 21. During this period, new growth (cm) of citrange and citrumelo shoots at 660 μl1−1 was 94 and 69% greater, respectively, than at 330 μ1 1−1. Total dry weight of both rootstock shoots had increased by over 100%. Growth of few species is affected this markedly by elevated CO2 levels.
More carbon was partitioned to above-ground organs in CO2-enriched citrus seedlings. Stem dry matter per unit length was also 32 and 44% greater in citrange and citrumelo, respectively. Total leaf area was increased by 124% in citrange and 85% in citrumelo due to greater leaf number and size. Variations in overall relative growth rate appeared to be related to the rapid, sequential, flush-type growth in citrus, in which an entire shoot segment with its associated leaves remains an active sink until fully expanded. RuBP carboxylase (EC 4.1.1.39) activity in leaves of recently-expanded flushes was higher in citrumelo plants grown at 660 vs 330 μ1 1−1 CO2 and changed diurnally for citrange (but not citrumelo) leaves at both CO2 levels. The results are consistent with the hypothesis that positive long-term effects of CO2 enrichment may be greater in species or during growth periods where sink capacity for carbon utilization is high.  相似文献

In vitro shoots of cv. Doyenne ďHiver pear ( Pyrus communis L.) were irradiated under controlled environments for 6 h per day at 5 different levels of biologically effective UV-B radiation (UV-BBE). UV-B exposure caused a progressive increase in apical necrosis above background levels and stimulated leaf abscission. Shoots grown for 2 weeks at 7. 8 mol m−2 day −1 of photosynthetic photon flux (PPF) and treated with 8. 4 or 12. 0 kJ m−2 day −1 UV-BBE produced up to 4 times more ethylene than those given 2. 2 or 5. 1 kJ m−2 day−1 UV-BBE or untreated controls. Exposure of shoots to 12 kJ m−2 day −1 of UV-BBE caused an increase in free putreseine content after 4 to 14 days of irradiation. Shoots showed a decrease in CO2 uptake after 3 days of UV-B: thereafter, they appeared to recover their photosynthetic capacity. Under typical PPF conditions used in micropropagation (90 μmol m−2 S−1). 8. 4 kJ m−2 day −1 of UV-B radiation was injurious to realatively tender tissues of in vitro pear shoots: increasing the level of UV-BBE to 12 kJ m−2 day−1 produced even more adverse effects.  相似文献

Abstract. Plantago maritima L. was grown at three levels of salinity, 50, 200, 350 mol m−3 NaCl, and the effects on growth, ion content and photosynthetic capacity were studied. Shoot and root dry weight, leaf production and leaf length were all substantially reduced in plants grown at high salinity. Total leaf area of plants grown at 350 mol m−3 NaCl was only 20% of that in plants at low salinity. Both the Na+ and K+ content of leaves and roots increased with external salinity. There was no change in the Na+/K+ ratio of leaves or roots at different salinity levels. Despite the large reductions in growth and high accumulation of Na+ ions, leaf photosynthetic rate was only slightly reduced by salinity stress. The reduction in photosynthesis was not caused by reduced biochemical capacity as judged by photosynthetic response to intercellular CO2 and by ribulose-1,5-bisphosphate carboxylase activity, but was due to reduced leaf conductance and low intercellular CO2 concentration. The increased stomatal limitation of photosynthesis resulted in higher water-use efficiency of plants grown at high salinity.  相似文献

The influence of solar UV-A and UV-B radiation at Beltsville, Maryland, on growth and flavonoid content in four cultivars of Cucumis sativus L. (Ashley, Poinsett, Marketmore, and Salad Bush cucumber) was examined during the summers of 1994 and 1995. Plants were grown from seed in UV exclusion chambers consisting of UV-transmitting Plexiglas, lined with Llumar to exclude UV-A and UV-B, polyester to exclude UV-B, or cellulose acetate to transmit UV-A and UV-B. Despite previously determined differences in sensitivity to supplemental UV-B radiation, all four cultivars responded similarly to UV-B exclusion treatment. After 19–21 days, the four cultivars grown in the absence of solar UV-B (polyester) had an average of 34, 55, and 40% greater biomass of leaves, stems, and roots, respectively, 27% greater stem height, and 35% greater leaf area than those grown under ambient UV-B (cellulose acetate). Plants protected from UV-A radiation as well (Llumar) showed an additional 14 and 22% average increase, respectively, in biomass of leaves and stems, and a 22 and 19% average increase, respectively, in stem elongation and leaf area over those grown under polyester. These findings demonstrate the extreme sensitivity of cucumber not only to present levels of UV-B but also to UV-A and suggest that even small changes in ozone depletion may have important biological consequences for certain plant species.  相似文献

The effects of decreased irradiance on fresh and dry weight, root respiration, levels of carbohydrates and N-compounds, and extractable activities of enzymes involved in C and N metabolism were evaluated in maize ( Zea mays L. cv. Plauto) seedlings during the 7 days following transfer from 450 to 200 μmol m−2 s−1 PAR. The fresh weight of roots and stems, the initiation of new leaves, root respiration rate, and the accumulation of dry matter, soluble sugars, starch, malate and amino acids in both leaves and roots were strongly reduced at low irradiance. In contrast, the level of nitrate was increased in leaves and only marginally affected in roots. Leaf phosphoenolpyruvate carboxylase (EC 4.1.1.31) activity started to decrease after 24–34 h, whereas ribulose-1,5-bisphosphate carboxylase (EC 4.1.1.39) activity and chlorophyll content were unaffected or only slightly reduced. In both leaves and roots, the adjustment of N metabolism to low irradiance occurred through a relatively rapid (30% after 10 h) and large (60% after 3 days) decrease of nitrate reductase (NR; EC 1.6.6.1) activity, followed by slower and smaller changes in the activity of nitrite reductase (EC 1.7.7.1), glutamine synthetase (EC 6.3.1.2) and NAD-dependent glutamate dehydrogenase (EC 1.4.1.2). We suggest that the preferential decrease of NR activity relative to other N-assimilating enzymes may be important for preventing the accumulation of toxic N-compounds like ammonia in both leaf and root tissues.  相似文献

Light effects on in vitro adventitious root formation in axillary shoots of a 95-year-old black cherry ( Prunus serotina Ehrh.) were examined using microcuttings derived from cultured vegetative buds. Three studies were performed: 1) complete darkness and 4 levels of continuous white light irradiance were tested at 70, 278, 555 and 833 μmol m−2 s−1; 2) white, red, yellow and blue light were tested to assess the importance of spectral quality; and 3) the effect of blue light at intensities of 7,15, 22 and 30 μmol m−2 s−1 was also studied, Measurements included rooting percentage, total number of roots per shoot, and shoot and root dry weight. There was a strong negative effect of white light intensity upon root formation. Blue light between 15 and 22 μmol m−2: s−1 significantly retarded root formation and completely inhibited it at 36 μmol m−2 s−1. Shoots treated with yellow light exhibited the highest rooting percentage, mean number of roots per shoot, and root dry weight.  相似文献
