The mechanisms by which low [K(+)](o) induces spontaneous activity was studied in sheep Purkinje fibers. Purkinje strands were superfused in vitro and membrane potentials were recorded by means of a microelectrode technique. The results show that low [K(+)](o) increases the slope and amplitude of early diastolic depolarization, sharpens the transition between early and late diastolic depolarizations, induces an after-potential and large pre-potentials through a negative shift of an oscillatory zone. Pre-potentials occur progressively sooner during diastole and merge with the after-potential to induce uninterrupted spontaneous discharge. During recovery, when the rate slows, after- and pre-potentials separate once more, the slower discharge decreasing the after-potentials but not the pre-potentials. Low [K(+)](o) has little effect on the plateau, but markedly slows phase 3 repolarization and may altogether prevent it. At depolarized levels, voltage oscillations, slow responses, sinusoidal fluctuations or quiescence may be present depending on voltage. During the recovery, a train of either sub-threshold oscillations or spontaneous action potentials appear towards the end of phase 3 repolarization. The cessation of the action potentials unmasks large sub-threshold oscillations, that occur in the oscillatory zone. Drive, high [Ca(2+)](o) and norepinephrine increase slope and amplitude of early diastolic depolarization as low [K(+)](o) does. In low [K(+)](o), Cs(+) prevents spontaneous discharge at polarized levels, but not the decrease in resting potential nor the onset of slow responses at depolarized levels. Cs(+) blocks the train of oscillations and of action potentials occurring during recovery. We conclude that low [K(+)](o) steepens early diastolic depolarization and increases its amplitude through an after-potential that results from an increased Ca(2+) load; allows the attainment of the threshold through Cs(+)-sensitive voltage oscillations which develop when the oscillatory zone is entered either by diastolic depolarization or by phase 3 repolarization; and causes voltage oscillations also at depolarized levels, but through a Cs(+)-insensitive different mechanism.  相似文献

PATIENTS AND METHOD: During a period of 15 years we investigated 200 patients suspected of normal pressure hydrocephalus (NPH) by means of an intrathecal infusion test. In 107 patients (54%) the diagnosis of a normal pressure hydrocephalus could be confirmed. For the evaluation of the course of disease we used the Black-Grading-Scale for shunt assessment [1] and the clinical grading for chronic hydrocephalus (Kiefer-scale) [4] pre- and postoperatively as well as in a follow-up examination seven months after surgical treatment. The aim of our study was to find out a quick and easy-to-handle bed-side examination for the grading of NPH. DISCUSSION: The Black-Grading-Scale does not allow to distinguish between patients in an unchanged condition and those with a worsening of their symptoms. Therefore this scale is useful for patients with an obstructive hydrocephalus but not for those with a NPH. In our opinion the clinical grading of Kiefer [4] seems to be the most reliable scale for the grading and long term follow-up of a normal pressure hydrocephalus. CONCLUSION: Our own created NPH-Recovery-Rate is based on the clinical grading for chronic hydrocephalus (Kiefer-scale) [4]. It allows the interindividual comparison between the courses of disease in patients with normal pressure hydrocephalus.  相似文献

Eight Enterococcus faecium strains isolated from ewe milk and artisanal cheese from northwest Argentina were screened for biotechnological properties relevant to flavour development. The API ZYM test showed absence of proteases, presence of high amounts of peptidases, and high esterase-lipase activities. Low extracellular proteolytic activity was observed. Most strains produced diacetyl in milk, with E. faecium OvL 214 and OvL 254 being the best producers. Biomass and growth rate increased when citrate was added to the medium, suggesting that these strains could use citrate as a main energy source. After 24 h of incubation, citrate was completely consumed in complex medium supplemented with glucose and citrate. An average of 17% residual citrate was detected in complex media supplemented with citrate. For all strains, esterase activity was detected up to alpha-naphthyl-caproate. They hydrolyzed alpha-naphthyl derivatives of fatty acids in this order: C3 > C6 > C4 > C8 > C2. Post-electrophoretic detection of esterase activities revealed the presence of multiple esterases. Hydrolysis of tributyrin, tricaprylin, and milk fat was observed in cell-free extracts. Enterococcus faecium strains isolated from ewe milk and artisanal cheese from northwest Argentina present the metabolic potential to contribute to cheese flavour development.  相似文献

OBJECTIVE: To determine whether the frequency of nuclear grooves in intermediate squamous cells in cervical smears is related to inflammatory or neoplastic events. STUDY DESIGN: Sixty benign and 40 neoplastic, nonatrophic cervical smears, collected by conventional methods and stained by Papanicolaou stain, were selected for this study. Twenty smears of the benign cohort showed evidence of inflammation. The neoplastic cohort comprised 20 smears representative of low grade and 20 of high grade squamous intraepithelial lesion (LSIL and HSIL, respectively), with 50% in each group showing evidence of inflammation. The patients, of mixed ethnic backgrounds, were between 18 and 45 years of age. The frequency of nuclear grooves in 100 morphologically benign intermediate squamous cells were determined in each case. The results were evaluated by statistical analysis. RESULTS: The study established that the presence of inflammation had no impact on the frequency of nuclear grooves in benign intermediate squamous cells in either benign or neoplastic smears. When compared with benign smears, there was no increase in the frequency of nuclear grooves in LSIL. Smears of HSIL showed the highest frequency of nuclear grooves. The difference between HSIL and other groups was statistically significant (P < .01). CONCLUSION: The frequency of nuclear grooves in either normal or neoplastic smears is unrelated to inflammation. In smears with neoplastic changes, an increase in grooved nuclei occurred in HSIL. The mechanism and significance of this observation are unknown and warrant further study.  相似文献

The aim of this study was to prepare alginate-immobilized freeze-dried cultures of Streptococcus thermophilus and to compare the acidifying activities of these rehydrated cultures with classical free cell liquid inoculants. Streptococcus thermophilus BT1 grew in alginate beads and the population reached 10(10) cfu g(-1) after 6 h incubation. Re-inoculation of the beads in fresh medium with a further 6 h incubation did not improve the biomass level, but extending the incubation at 42 degrees C to 24 h caused significant death. The rehydrated immobilized cell technology (ICT) starter contained 13% free cells. In acidifying activity tests, the ICT culture had a similar acidification curve to that of a classical milk-grown free cell culture, except that it reached lower final pH values. Although the differences between the ICT and liquid cultures were not important, there were significant effects of inoculation level on lag time, maximum acidification rate and on the pH and time at which the acidification rate was at its highest.  相似文献

1. The tributyrinases in milk are mainly associated with casein micelles. Dilution or addition of sodium chloride increases the enzyme activity, probably by dissociating the micelle–tributyrinase complexes. 2. Tributyrinase activities of milks activated by dilution and sodium chloride addition were in the range 0·2–1·7μequiv. of acid liberated/ml. of milk/min. from tributyrin emulsion at pH8·5 and 25°. The enzymes have a bivalent-cation requirement for full activity and are rather unstable when separated from casein. 3. Ultracentrifugation of skim milks containing sodium chloride (0·75m) gave preparations low in casein but containing about 70% of the milk tributyrinases. The tributyrinases in such preparations appear to be bound in complexes of molecular weight about 350000. Dilution may result in dissociation to give the free enzymes. 4. Pancreatic lipase also formed complexes with casein micelles, but wheat-germ esterase, xanthine oxidase, milk alkaline phosphatase and other enzymes did not.  相似文献

Iodination at pH 9.5 has converted three of the six tyrosyl residues of ribonuclease to 3,5-diiodotyrosyl residues. The iodinated ribonuclease is still enzymatically active toward ribonucleic acid. The diiodotyrosyl residues have approximately normal pK's, while the tyrosyl residues remaining have abnormally high pK's. Hence, iodination has taken place on the normal tyrosyl residues.Two of the iodinated tyrosyl residues have been shown to be No. 115 and No.92; hence, these appear to be normal tyrosyl residues. One of the non-iodinated tyrosyl residues is No. 97, and hence is one of the abnormal tyrosyl residues. The location of the other normal tyrosyl residue (or the other two abnormal ones) remains uncertain.  相似文献

The biological and logical meaning of trees, which are one of the important woody plants of our ecosystem, are reviewed in this article. Trees are mostly used for timber purposes, but in the present article the utility of trees with respect to their importance in restoring, reclaiming and rejuvenating denuded and disturbed soils, their ecological, ecodevelopmental and environmental use, and their educational and recreational value in gardening, landscaping and bioesthetic planning is described. In addition, the importance of trees is discussed with reference to their value as a source of sustenance: food, sugars, starches, spices and condiments, beverages, fumitories, masticatories and narcotics, medicines, essential oils, fatty oils and vegetable fats, waxes, soap substitutes, vegetable ivory, fodder, fuel, bioenergy or biofuel, fertilizers, fiber, pulp and paper, tannins, dyes, rubber and other latex products, gums, resins and cork. Lastly, the food plants of mulberry and non-mulberry silkworms, which feed on the leaves of many forest trees, are mentioned.  相似文献

Over 4,000 cells from 105 normal and 96 abnormal uterine cervical scrapes were prepared according to the UCLA monolayer procedure, stained by a routine Papanicolaou method and visually classified by two cytopathologists and a technologist into seven classes: parabasal, metaplastic, mild dysplasia, moderate dysplasia, severe dysplasia, carcinoma in situ and invasive carcinoma. Canonical analysis was used to correlate effects-coded class membership variables with 23 cell features derived from digital image analysis. In general, nuclear texture measures derived from linear combinations of run-length correlations along with features derived from a Markov transitional probability matrix provided the best predictors of cell class. After cells were divided into benign (moderate dysplasia or less) and malignant (severe dysplasia or worse) groups, discriminant analysis correctly classified 84% of the benign cells and 91% of the malignant cells.  相似文献

Research about mare’s milk is mainly focused on quality and information about quantity is incomplete partly due to the lack of a consensus on the method of measuring milk yield. The live weight, body condition at foaling and age of mares are factors influencing milk yield. The influence of mare parity, however, remains unclear. Over a period of 2 years (2018–2019), milk yield was evaluated on 65 mares (51 multiparous and 13 primiparous). Mares and foals were kept in a group at pasture. One method of milk yield measurement and one proxy method were applied; milking and weight-suckle-weight (WSW), respectively. The procedure was performed at five timepoints during the lactation period (3–30–60–90 and 180 days) without repetition. The relevance of WSW was addressed by studying the correlation between the two methods on 23 individuals. Factors influencing milk yield, through milking data, were studied on 57 individuals. Data was divided into two subsets. The first was an explanatory matrix containing the live weight of mares 24 h after parturition, parity, age, year of lactation and foal gender. The second was a response matrix containing data from milking at the five timepoints of the lactation. A correlation was found (RV = 0.41) between milking and WSW at day 3, however no correlation was found for other timepoints (RV ≤ 0.15). The live weight of the mare 24 h after foaling, age and parity appeared to have a significant impact on milk production (P < 0.05). Thus, older or multiparous mares showed a higher milk yield than younger or primiparous mares. In addition, mares with a higher live weight after foaling produced more milk than those with a lower live weight. Overall, results can lead us to two main conclusions. First, the WSW method performed at five different timepoints of the lactation, but without repeated measurements, is not an efficient way to estimate the milk yield of mares. Secondly, results concerning the live weight and age of mares were in accordance with previous studies. The influence of parity was also highlighted, confirming trends showed by other authors. Age and parity are closely related in our population, making it difficult to differentially assess their effects. Being able to identify the impact of both factors independently would benefit several sectors of the horse industry from sport to mare milk producers.  相似文献
