Synopsis Immunoreactive (ir) gonadotropin-releasing hormone (GnRH) is localized in many neurons of the terminal nerve (TN) and midbrain tegmentum, while few ir-cells are observed in the preoptic area and ventral hypothalamus. The paucity of preoptic ir-cells may relate to an unusual feature of the elasmobranch pituitary, i.e. a lack of portal control of gonadotropin-producing cells. TN and midbrain GnRH-ir neurons may be major sources of GnRH used to modulate or otherwise control both pituitary and brain cells via delivery through the systemic circulation. These ir-nuclei also appear to directly innervate CNS regions (the preoptic area, habenula and clasper control area of the spinal cord) involved in sexual functions. Important regulatory mechanisms, represented by interactions between GnRH pathways and sex-steroid concentrating neurons, are likely to occur in the preoptic area, habenula and midbrain tegmentum.  相似文献

Synopsis Cartilaginous fishes, the sharks, rays and chimaeras (class Chondrichthyes), are a very old and successful group of jawed fishes that currently contains between 900 and 1100 known living species. Chondrichthyians show a high morphological diversity during most of their evolutionary career from the Paleozoic to the present day. They are relatively large predators which have remained a major, competitive element of marine ecosystems despite the varied rivalry of numerous other marine vertebrate groups over at least 400 million years. Although restricted in their ecological roles by morphology, reproduction and other factors, the living cartilaginous fishes are highly diverse and show numerous alternative life-history styles which are multiple answers to exploiting available niches permitted by chondrichthyian limitations. Chondrichthyians living and fossil can be divided into at least eighteen ecomorphotypes, of which the littoral ecomorphotype is perhaps the most primitive and can serve as an evolutionary origin for numerous specialist ecomorphotypes with benthic, high-speed, superpredatory, deep-slope and oceanic components. Reproductive modes in cartilaginous fishes are of six types, ranging from primitive extended oviparity through retained oviparity and yolk-sac viviparity (previously ovoviviparity) to three derived forms of viviparity. Reproductive modes are not strongly correlated with ecomorphotypes and with the phylogeny of living elasmobranchs. The success and importance of cartilaginous fishes is largely underrated by marine biologists and by the public, and requires new and heretical emphasis to overcome the present inadequacies of chondrichthyian research and the problems of overexploitation that cartilaginous fishes face.  相似文献

The present study has examined the role of the colon in regulating ammonia and urea nitrogen balance in two species of chondrichthyans, the ratfish, Hydrolagus colliei (a holocephalan) and the spiny dogfish, Squalus acanthias (an elasmobranch). Stripped colonic tissue from both the dogfish and ratfish was mounted in an Ussing chamber and in both species bi-directional urea flux was found to be negligible. Urea uptake by the mucosa and serosa of the isolated colonic epithelium through accumulation of 14C-urea was determined to be 2.8 and 6.2 fold greater in the mucosa of the dogfish compared to the serosa of the dogfish and the mucosa of the ratfish respectively. Furthermore, there was no difference between serosal and mucosal accumulation of 14C-urea in the ratfish. Through the addition of 2 mM NH4Cl to the mucosal side of each preparation the potential for ammonia flux was also examined. This was again found to be negligible in both species suggesting that the colon is an extremely tight epithelium to the movement of both urea and ammonia. Plasma, chyme and bile fluid samples were also taken from the agastric ratfish and were compared with solute concentrations of equivalent body fluids in the dogfish. Finally molecular analysis revealed expression of 3 isoforms of the urea transport protein (UT) and an ammonia transport protein (Rhbg) in the gill, intestine, kidney and colon of the ratfish. Partial nucleotide sequences of the UT-1, 2 and 3 isoforms in the ratfish had 95, 95 and 92% identity to the equivalent UT isoforms recently identified in another holocephalan, the elephantfish, Callorhinchus milii. Finally, the nucleotide sequence of the Rhbg identified in the ratfish had 73% identity to the Rhbg protein recently identified in the little skate, Leucoraja erinacea.  相似文献

Summary Gonadotropin-releasing hormone (GnRH) neurons and their pathways in the rat brain were localized by immunocytochemistry in 6-to 18-day-old female animals, by use of thick frozen or vibratome sections, and silver-gold intensification of the diaminobenzidine reaction product. GnRH-immunoreactive perikarya were observed in the following regions: olfactory bulb and tubercle, vertical and horizontal limbs of the diagonal band of Broca, medial septum, medial preoptic and suprachiasmatic areas, anterior and lateral hypothalamus, and different regions of the hippocampus (indusium griseum, Ammon's horn). In addition to the known GnRH-pathways (preoptico-terminal, preoptico-infundibular, periventricular), we also observed GnRH-immunopositive processes in several major tracts and areas of the brain, including the medial and cortical amygdaloid complex, stria terminalis, stria medullaris thalami, fasciculus retroflexus, medial forebrain bundle, indusium griseum, stria longitudinalis medialis and lateralis, hippocampus, periaqueductal gray of the mesencephalon, and extracerebral regions, such as the lamina cribrosa, nervus terminalis and its associated ganglia. By use of the silver-gold intensification method we present Golgi-like images of GnRH perikarya and their pathways. The possible distribution of efferents from each GnRH cell group is discussed.  相似文献

Summary In three species of teleosts — carp Cyprinus carpio; grass carp Ctenopharyngodon idella; and crucian carp Carassius auratus — the caudal neurosecretory system displays small, medium-sized and large neurons. Urotensin I (UI)-immunoreactive and UI-nonreactive neurons were found in all three groups; in general, the number of the latter neurons exceeded that of the former. Noteworthy are: (i) UI-immunoreactive fibers in the caudal spinal cord and (ii) dense accumulations of UI-immunoreactive product around the capillaries of the urophysis. In two species of elasmobranchs — cat shark Heterodontus japonicas and swell shark Cephaloscyllium umbratile — neurosecretory neurons decreased in size in rostro-caudal direction. Most of the neurosecretory perikarya, their axons and the corresponding neurohemal areas were UI-immunoreactive, but a small number of secretory neurons was devoid of immunoreaction. Oxytocin, arginine vasopressin, substance P, somatostatin, neurotensin, vasoactive intestinal polypeptide and gastrin-releasing peptide were not detected in the caudal neurosecretory system of the carp.  相似文献

Summary The distribution of immunoreactive neuropeptides was investigated in the retina of three species of skates (Raja clavata, R. radiata, R. oscellata), elasmobranch fish often used in electrophysiological work on the retina. Enkephalins, neuropeptide Y (NPY), substance P and glucagon were found in different types of amacrine cells.All four peptides appeared in cell bodies in the innermost part of the inner nuclear layer. Processes from the cells containing enkephalins were numerous and ramified throughout the inner plexiform layer. Processes from the cells containing glucagon were thick and rare, and were found throughout the inner plexiform layer, at times with a predominance in sublaminae 1 and 4. NPY-immunoreactive fibres appeared mainly in sublamina 1 but also in 2 or 3, and substance-P-immunoreactive fibres in sublaminae 1,4 and 5.Antisera against somatostatin, VIP or neurotensin did not show any immunoreactivity in the skate retina.  相似文献

Vertebrates originated in the lower Cambrian. Their diversification and morphological innovations have been attributed to large-scale gene or genome duplications at the origin of the group. These duplications are predicted to have occurred in two rounds, the "2R" hypothesis, or they may have occurred in one genome duplication plus many segmental duplications, although these hypotheses are disputed. Under such models, most genes that are duplicated in all vertebrates should have originated during the same period. Previous work has shown that indeed duplications started after the speciation between vertebrates and the closest invertebrate, amphioxus, but have not set a clear ending. Consideration of chordate phylogeny immediately shows the key position of cartilaginous vertebrates (Chondrichthyes) to answer this question. Did gene duplications occur as frequently during the 45 Myr between the cartilaginous/bony vertebrate split and the fish/tetrapode split as in the previous approximately 100 Myr? Although the time interval is relatively short, it is crucial to understanding the events at the origin of vertebrates. By a systematic appraisal of gene phylogenies, we show that significantly more duplications occurred before than after the cartilaginous/bony vertebrate split. Our results support rounds of gene or genome duplications during a limited period of early vertebrate evolution and allow a better characterization of these events.  相似文献

Validated age and growth estimates are important for constructing age-structured population dynamic models of chondrichthyan fishes, especially those which are exploited. We review age and growth studies of chondrichthyan fishes, using 28 recent studies to identify areas where improvements can be made in describing the characteristics of ageing structures (both traditional and novel) utilized to estimate ages of sharks, rays, and chimaeras. The topics identified that need consistency include the: (1) terminology used to describe growth features; (2) methods used to both verify and validate age estimates from chondrichthyan calcified structures, especially edge and marginal increment analyses; and (3) the functions used to produce and describe growth parameters, stressing the incorporation of size at birth (L 0) and multiple functions to characterize growth characteristics, age at maturity and longevity.  相似文献

In this review, I explore the effects of both social organization and the physical environment, specifically habitat complexity, on the brains and behavior of highly visual African cichlid fishes, drawing on examples from primates and birds where appropriate. In closely related fishes from the monophyletic Ectodinii clade of Lake Tanganyika, both forces influence cichlid brains and behavior. Considering social influences first, visual acuity differs with respect to social organization (monogamy versus polygyny). Both the telencephalon and amygdalar homologue, area Dm, are larger in monogamous species. Monogamous species are found to have more vasotocin-immunoreactive cells in the preoptic area of the brain. Habitat complexity also influences brain and behavior in these fishes. Total brain size, telencephalic and cerebellar size are positively correlated with habitat complexity. Visual acuity and spatial memory are enhanced in cichlids living in more complex environments. However habitat complexity and social forces affect cichlid brains differently. Taken together, our field data and plasticity data suggest that some of the species-specific neural effects of habitat complexity could be the consequence of the corresponding social correlates. Environmental forces, however, exert a broader effect on brain structures than social ones do, suggesting allometric expansion of the brain structures in concert with brain size and/or co-evolution of these structures [Current Zoology 56 (1): 144-156 2010].  相似文献

Synopsis Adult clearnose skates, Raja eglanteria, were captured during the winters of 1981 and 1983, and observed to mate in captivity. Mating and egg depositions take place on the central west coast of Florida from December through mid-May. During copulation the male holds the trailing edge of the female's right or left pectoral fin firmly in his mouth, swings his tail beneath hers and inserts one clasper into the distal end of her reproductive tract. Copulation may last one to four hours during which time sperm pass from the urogenital papilla of the male along the clasper groove to the female. Sperm move cranially to the upper portion of the shell gland where they are stored and remain viable for at least three months. The ovum is fertilized in the shell gland. The egg case bears a prominent projection or horn at each corner. The two posterior ones are shorter and bear tendrils which are covered with a sticky substance that insures attachment to the substrate when the egg is deposited. Fertilized eggs are laid in pairs at intervals ranging from 1 to 13 days (mean of 4.5 ± 2.2 days). As development proceeds within the egg case a plugged slit on the lateral side of each horn opens and permits seawater to wash the developing embryo. Incubation periods for eggs maintained between 20–22°C decrease in duration throughout the egg laying season, ranging from 94 days initially to 77 days for eggs laid later in the spring. At hatching, the anterior end of the egg case ruptures, and the skate emerges abruptly with its pectoral fins rolled dorsally.  相似文献

There are two types of superactive agonists of gonadotropin-releasing hormone (GnRHa-I: (D-amino acid)6-GnRH and GnRHa-II: (D-amino acid)6-(desGly)10-GnRH- ethylamide) the high hormonal activity of which is understood to be due to their higher receptor affinity and their higher proteolytic stability as compared with the native GnRH sequence. Using the soluble fractions of various rat tissues in studies on the inactivation of GnRH peptides, we confirmed the higher proteolytic resistance of GnRHa-II, but not of D-Phe6-GnRH (GnRHa-I) and of another analog, D-Trp3-D-Phe6-GnRH, as compared with GnRH. The exact behaviour of the peptides during degradation was found to be dependent on the peptide concentrations used, showing the importance of using conditions as near to the physiological ones a possible. Towards the membrane fractions, however, the order of degradability was found to be GnRH much greater than D-Phe6-GnRH much greater than D-Trp3-D-Phe6-GnRH. The pharmacokinetic consequences of the different proteolytic degradabilities of the GnRH peptides, observed in rats, were a moderate increase in the biological half-life of D-Phe6-GnRH by 2.5-fold, as compared with GnRH, and a small increase in half-life of D-Trp3-D-Phe6-GnRH by 1.4-fold when compared with D-Phe6-GnRH. Whereas no intact GnRH was recovered in rat urine, small amounts of D-Phe6-GnRH (about 1% of dose) and high amounts of D-Trp3-D-Phe6-GnRH (25.5%) were excreted into urine. Combining the biochemical and pharmacokinetic data, it is concluded that proteolytic stability of GnRH analogs in pharmacological terms means stability towards membrane enzymes (pharmacologically-related stability) and that designing analogs with further increased proteolytic stability will be of only limited consequences with respect to their biological half-lives, the glomerular filtration rate of the kidney becoming the determining factor in the peptide clearance.  相似文献

Summary The presence of peptides in the gastrointestinal tract of the rainbow trout, Salmo gairdneri, was investigated immunocytochemically. VIP-like immunoreactivity was demonstrated in nerves in all layers of the stomach and the intestine, whereas substance P-like immunoreactivity was localized to endocrine cells, predominantly in the mucosa of the stomach, and to nerves mainly concentrated in the myenteric plexus throughout the gut. Endocrine cells reactive to gastrin/CCK antiserum were demonstrated in the intestinal mucosa, while no immunoreactivity was found in the stomach. Bombesin-immunoreactive and somatostatin-immunoreactive cells were localized in the stomach mucosa, and cells reactive to glucagon antiserum in the intestinal mucosa. Radioimmunoassay of stomach mucosa and muscle confirmed the presence of VIP-like and substance P-like immunoreactivity in these tissues, while gastrin/CCK-like immunoreactivity was low and bombesin-like immuno-reactivity was insignificant. In conclusion, molecules resembling the mammalian brain-gut peptides may be involved in the neuronal and hormonal control of gut function in fish.  相似文献

Immunohistochemical studies were conducted on sections of cat hypothalamus in order to determine the distribution of neurons containing alpha-melanocyte stimulating hormone and beta-endorphin immunoreactivity. A large number of neurons in the arcuate nucleus were stained after incubation of sections with antisera to either substance. Analysis of serial sections suggested that each neuron revealed with one antiserum was also revealed with the other antiserum, indicating the co-existence of alpha-melanocyte stimulating hormone and beta-endorphin immunoreactivity within these arcuate neurons. In contrast, a more diffuse group of lateral hypothalamic neurons which extended from the retrochiasmatic level to the posterior hypothalamus were stained only with the antiserum directed against alpha-melanocyte stimulating hormone. The present results largely confirm findings in the rat hypothalamus, although the lateral hypothalamic group of alpha-melanocyte stimulating hormone immunoreactive neurons appears to be more extensive in the cat.  相似文献

Based on direct observations of free‐ranging specimens, the giant manta ray Manta birostris is reported from the Marquesas Islands, the first sighting in French Polynesia. Sightings of its sister species, the reef manta ray Manta alfredi, are also reported at the most easterly location in the Pacific Ocean. Preliminary individual identification as well as notes on their distribution are also reported.  相似文献

The mature spermatozoa of two Taiwan protandrous hermaphrodite Sparidae Acanthopagrus berda and Acanthopagrus australis are investigated and compared with those of other two Sparidae (Lagodon rhomboids and Archosargus probatocephus) from the Western hemisphere. Ultrastructurally the spermatozoon of these four species has a spherical, homogeneously electron-dense nucleus with an axial nuclear fossa. The midpiece contains one to four spherical mitochondria and encircles the basal body of the flagellum. The mature spermatozoa of the four species are of the primitive or ect-aquasperm form and conform to the teleostean type I spermatozoon with the flagellar axis inserts perpendicular and medial to the nuclear fossa. Variation in the depths of the nuclear fossa and mitochondria number is substantial in these four Sparidae species. This study provide useful systematic characters to the existing knowledge of comparative spermatology of Sparidae.  相似文献

Summary In the four species of elasmobranchs examined (Triakis scyllia, Heterodontus japonicus, Scyliorhinus torazame, Dasyatis akajei), all identifiable caudal neurosecretory cells and their corresponding neurohemal areas showed urotensin II (UII)-immunoreactivity with varied intensity. To localize urotensin I (UI) in the caudal neurosecretory system of the dogfish, Triakis scyllia, h-CRF (1–20) antiserum that cross-reacts with UI was used in place of UI antiserum. CRF/UI-immunoreactivity was demonstrated in the neurosecretory cells and neurohemal areas. A considerable number of neurons showed both UII- and CRF/UI-immunoreactivities, suggesting that UII and UI are produced in the same neurosecretory cells. However, some neurons exhibited UII-immunoreactivity, but no CRF/UI-immunoreactivity. Cells immunoreactive only to CRF antiserum were not detected. At least two populations of neurons exist in the dogfish caudal neurosecretory system: (i) cells immunoreactive for both CRF/UI and UII, and (ii) cells immunoreactive for UII. The dorsal cells of the lamprey, Lampetra japonica, did not react with either UII or CRF antiserum.  相似文献

In the cow, a specific glycoprotein-bovine oviduct-specific glycoprotein (BOGP)-is secreted by the epithelial cells of the oviduct at the follicular stage of the estrous cycle. In this study, we examined the effects of purified BOGP on the viability and motility of bovine spermatozoa in culture in vitro. Frozen-thawed bovine spermatozoa were incubated in modified Tyrode's solution (TALP) that contained purified BOGP (TALP-BOGP). In TALP-BOGP, both the viability and motility of bovine spermatozoa were more effectively maintained than in the control medium without any added protein. The increases in both the viability and motility of spermatozoa were dose-dependent. Spermatozoa were also incubated in TALP medium supplemented with bovine serum albumin, egg albumin, lactalbumin, or gastric mucin, and their viability and motility in these media were compared with that in TALP-BOGP. Both the viability and motility of spermatozoa were more effectively maintained in TALP-BOGP throughout a 12-hr incubation than in other media tested. An immunolabeling study demonstrated that a monoclonal antibody specific for BOGP reacted with the posterior region of the head, the middle portion, and the tail of spermatozoa that had been incubated with TALP-BOGP, suggesting that BOGP becomes specifically associated with particular regions of the spermatozoon. These results suggest that BOGP is a potent factor for maintenance of the viability and motility of sperm. On the basis of the present results, we also propose that BOGP may play an important role in sperm functions during the reproductive process. © 1995 wiley-Liss, Inc.  相似文献
